A series of nanofibrous scaffolds were prepared by electrospinning of poly(vinyl alcohol) (PVA)/gelatin aqueous solution. PVA and gelatin was dissolved in pure water and blended in full range, then being electrospun to prepared nanofibers, followed by being crosslinked with glutaraldehyde vapor and heat treatment to form nanofibrous scaffold. Field emission scanning electron microscope (FESEM) images of the nanofibers manifested that the fiber average diameters decreased from 290 to 90 nm with the increasing of gelatin. In vitro degradation rates of the nanofibers were also correlated with the composition and physical properties of electrospinning solutions. Cytocompatibility of the scaffolds was evaluated by cells morphology and MTT assay. The FESEM images revealed that NIH 3T3 fibroblasts spread and elongated actively on the scaffolds with spindle‐like and star‐type shape. The results of cell attachment and proliferation on the nanofibrous scaffolds suggested that the cytotoxicity of all samples are grade 1 or grade 0, indicating that the material had sound biosafety as biomaterials. Compared with pure PVA and gelatin scaffolds, the hybrid ones possess improved biocompatibility and controllability. A fibrous scaffold is required to provide three‐dimensional (3D) cell growth microenvironments and appropriate synergistic cell guidance cues. In this study, porous scaffolds with different mass ratio of poly(lactic acid) to poly(3‐hydroxybutyrate‐co‐4‐hydroxybutyrate) (P(3HB‐co‐4HB)) for tissue engineering were prepared by a modified particle leaching method. The effect of the addition of P(3HB‐co‐4HB) on microstructural morphology, compression property, swelling behavior, and enzymatic degradation of hybrid scaffolds was systematically investigated. The results indicated that this method was simple but efficient to prepare highly interconnected biomimetic 3D hybrid scaffolds (PP50/50 and PP33/67) with fibrous pore walls. The cytocompatibility of hybrid scaffolds was evaluated by in vitro culture of mesenchymal stem cells. The cell‐cultured hybrid scaffolds presented a complete 3D porous structure, thus allowing cell proliferation on the surface and infiltration into the inner part of scaffolds. The obtained hybrid scaffolds with pore size ranging from 200 to 450 µm, over 90% porosity, adjustable biodegradability, and water‐uptake capability will be promising for cartilage tissue engineering applications. Poly(?‐caprolactone) (PCL)/gelatin (GE) nanofiber scaffolds with varying concentrations of lanthanum chloride (LaCl3, from 0 to 25 mM) were fabricated by electrospinning. The scaffolds were characterized by scanning electron microscopy, contact angle and porosity measurements, mechanical strength tests, and in vitro degradation studies. In vitro cytotoxicity and cell adhesion and proliferation studies were performed to assess the biocompatibility of the scaffolds, and in vivo wound healing studies were conducted to assess scaffold applications in the clinic. All prepared scaffolds were noncytotoxic, and the growth of adipose tissue–derived stem cells on LaCl3‐containing scaffolds was better than on the pure PCL/GE scaffold. Cell proliferation studies showed the greatest cell growth in the PCL/GE/LaCl3 scaffolds. Further, in vivo studies proved that the PCL/GE/LaCl3 scaffolds can promote wound healing. The results suggest that nanofiber scaffolds containing LaCl3 promote cell proliferation and have good biocompatibility, and thus potential for application in the treatment of skin wounds. © 2018 Wiley Periodicals, Inc. Electrospinning is significantly one of the simple and versatile methods for producing micro- and nanofibrous scaffolds. Its assembly can be modified in different ways to combine material properties with different morphological structures for biomedical applications. In this process, collector design plays an important role to determine the nanofiber orientation in electrospun nanoweb. In this work, 3D patterned scaffolds were produced by electrospinning of polyamide-66 solution on different 3D collectors that have been obtained. The aim of this work is to investigate the attachment of the chondrocyte cells on the prepared electrospun scaffolds that have different types of nanofiber orientations that could be used in tissue engineering applications.  相似文献

ZnO nanocrystallites have been in situ embedded in cellulose nanofibers by a novel method that combines electrospinning and solvothermal techniques. Zn(OAc)2/cellulose acetate (CA) precursor hybrid nanofibers with diameter in the range of 160–330 nm were first fabricated via the electrospinning technique using zinc acetate as precursor, CA as the carrier, and dimethylformamide (DMF)/acetone(2 : 1) mixture as cosolvent. The precursor nanofibers were transformed into ZnO/cellulose hybrid fibers by hydrolysis in 0.1 mol/L NaOH aqueous solution. Subsequently, these hybrid fibers were further solvothermally treated in 180°C glycerol oil bath to improve the crystallite structure of the ZnO nanoparticles containing in the nanofibers. The structure and morphology of nanofibers were characterized by scanning electron microscopy, transmission electron microscopy, and X‐ray diffraction. It was found that hexagonal structured ZnO nanocrystallites with the size of ~ 30 nm were dispersed on the nanofiber surfaces and within the nanofibers with diameter of about 80 nm. The photocatalytic property of the ZnO/cellulose hybrid nanofibers toward Rhodamine (RhB) was tested under the irradiation of visible light. As a catalyst, it inherits not only the photocatalytic ability of nano‐ZnO, but also the thermal stability, good mechanical property, and solvent‐resistibility of cellulose nanofibers. The key advantages of this hybrid nanofiber over neat ZnO nanoparticles are its elasticity, dimensional stability, durability, and easy recyclability. © 2011 Wiley Periodicals, Inc. Engineered polymer scaffolds play an important role in tissue engineering. An ideal scaffold should have good mechanical properties and provide a biologically functional implant site. Considering their large surface area and high porosity, nanofibers have good potential as biomimetic scaffolds. However, the main shortcomings of scaffolds consisting of nanofibers are their mechanical inability to sustain a stress environment for neotissues and shape‐ability to form a variety of shapes and sizes. In this study, we produced design‐based poly (ε‐carprolactone) (PCL) nanofiber mats using an electrospinning method with various auxiliary electrodes and an xy moving system. To achieve stable initial solution at a nozzle tip of the electrospinning, various types of auxiliary electrodes were introduced. To characterize the effect of the electrodes in the electric‐field distribution near the nozzle tip, we calculated the electric field concentration factor and compared it with the experimental results. The nanofiber mat produced using the moving xy target system demonstrated orthotropic mechanical properties due to the fiber orientation, and human dermal fibroblasts seeded on the structure tended to grow according to nanofiber orientation. Novel three-dimensional (3-D) nano-/microfibrous poly(lactic-co-glycolic acid) (PLGA) scaffolds were fabricated by hybrid electrospinning, involving a combination of solution electrospinning and melt electrospinning. The scaffolds consisted of a randomly oriented structure of PLGA microfibers (average fiber diameter = 28 μm) and PLGA nanofibers (average fiber diameter = 530 nm). From mercury porosimetry, the PLGA nano-/microfiber (10/90) scaffolds were found to have similar pore parameters to the PLGA microfiber scaffolds. PLGA nano-/microfibrous scaffolds were examined and compared with the PLGA microfiber scaffolds in terms of the attachment, spreading and infiltration of normal human epidermal keratinocytes (NHEK) and fibroblasts (NHEF). The cell attachment and spreading of both cell types were several times higher in the nano-/microfiber composite scaffolds than in the microfibrous scaffolds without nanofibers. This shows that the presence of nanofibers enhanced the attachment and spreading of the cells on the nano-/microfiber composite scaffolds. Moreover, the nanofibers helped the cells infiltrate easily into the scaffolds. Overall, this novel nano-/microfiber structures has great potential for the 3-D organization and guidance of cells provided for tissue engineering.  相似文献

Three dimensional (3D) biodegradable porous scaffolds play a crucial role in bone tissue repair. In this study, four types of 3D polymer/hydroxyapatite (HAp) composite scaffolds were prepared by freeze drying technique in order to mimic the organic/inorganic nature of the bone. Chitosan (CH) and poly(lactic acid‐co‐glycolic acid) (PLGA) were used as the polymeric part and HAp as the inorganic component. Properties of the resultant scaffolds, such as morphology, porosity, degradation, water uptake, mechanical and thermal stabilities were examined. 3D scaffolds having interconnected macroporous structure and 77–89% porosity were produced. The pore diameters were in the range of 6 and 200 µm. PLGA and HAp containing scaffolds had the highest compressive modulus. PLGA maintained the strength by decreasing water uptake but increased the degradation rate. Scaffolds seeded with SaOs‐2 osteoblast cells showed that all scaffolds were capable of encouraging cell adhesion and proliferation. The presence of HAp particles caused an increase in cell number on CH‐HAp scaffolds compared to CH scaffolds, while cell number decreased when PLGA was incorporated in the structure. CH‐PLGA scaffolds showed highest cell number on days 7 and 14 compared to others. Based on the properties such as interconnected porosity, high mechanical strength, and in vitro cell proliferation, blend scaffolds have the potential to be applied in hard tissue treatments. POLYM. Organic–inorganic hybrid materials, composed of phases that interact on a nanoscale and a microstructure that mimics the extracellular matrix, can potentially provide attractive scaffolds for bone regeneration. In the present study, hybrid scaffolds of gelatin and bioactive glass (BG) with a fibrous microstructure were prepared by a combined sol–gel and electrospinning technique and evaluated in vitro. Structural and chemical analyses showed that the fibers consisted of gelatin and BG that were covalently linked by 3‐glycidoxypropyltrimethoxysilane to form a homogeneous phase. Immersion of the gelatin–BG hybrid scaffolds in a simulated body fluid (SBF) at 37°C resulted in the formation of a hydroxyapatite (HA)‐like material on the surface of the fibers within 12 h, showing the bioactivity of the scaffolds. After 5 days in SBF, the surface of the hybrid scaffolds was completely covered with an HA‐like layer. The gelatin–BG hybrid scaffolds had a tensile strength of 4.3 ± 1.2 MPa and an elongation to failure of 168 ± 14%, compared to values of 0.5 ± 0.2 MPa and 63 ± 2% for gelatin scaffolds with a similar microstructure. The hybrid scaffolds supported the proliferation of osteoblastic MC3T3‐E1 cells, alkaline phosphatase activity, and mineralization during in vitro culture, showing their biocompatibility. The results indicate that these gelatin–BG hybrid scaffolds prepared by a combination of sol–gel processing and electrospinning have potential for application in bone regeneration. © 2012 Wiley Periodicals, Inc. This study was designed to determine whether the surface modifications of the various poly(3‐hydroxybutyrate‐co‐4‐hydroxybutyrate) [P(3HB‐co‐4HB)] copolymer scaffolds fabricated would enhance mouse fibroblast cells (L929) attachment and proliferation. The P(3HB‐co‐4HB) copolymer with a wide range of 4HB monomer composition (16–91 mol %) was synthesized by a local isolate Cupriavidus sp. USMAA1020 by employing the modified two‐stage cultivation and by varying the concentrations of 4HB precursors, namely γ‐butyrolactone and 1,4‐butanediol. Five different processing techniques were used in fabricating the P(3HB‐co‐4HB) copolymer scaffolds such as solvent casting, salt‐leaching, enzyme degradation, combining salt‐leaching with enzyme degradation, and electrospinning. The increase in 4HB composition lowered melting temperatures (Tm) but increased elongation to break. P(3HB‐co‐91 mol % 4HB) exhibited a melting point of 46°C and elongation to break of 380%. The atomic force analysis showed an increase in the average surface roughness as the 4HB monomer composition increased. The mouse fibroblasts (L929) cell attachment was found to increase with high 4HB monomer composition in copolymer scaffolds. These results illustrate the importance of a detailed characterization of surface architecture of scaffolds to provoke specific cellular responses. © 2011 Wiley Periodicals, Inc. The objective of this work is generation of propolis/polyvinyl alcohol (PVA) scaffold by electrospinning for 3D cell culture. Here, PVA used as co-spinning agent since propolis alone cannot be easily processed by electrospinning methodology. Propolis takes charge in maximizing biological aspect of scaffold to facilitate cell attachment and proliferation. Morphological analysis showed size of the electrospun nanofibers varied between 172–523 nm and 345–687 nm in diameter, for non-crosslinked and crosslinked scaffolds, respectively. Incorporation of propolis resulted in desired surface properties of hybrid matrix, where hybrid scaffolds highly favored protein adsorption. To examine cell compatibility, NIH-3T3 and HeLa cells were seeded on propolis/PVA hybrid scaffold. Results confirmed that integration of propolis supported cell adhesion and cell proliferation. Also, results indicated electrospun propolis/PVA hybrid scaffold provide suitable microenvironment for cell culturing. Therefore, developed hybrid scaffold could be considered as potential candidate for 3D cell culture and tissue engineering.  相似文献

In this study, starch‐based hybrid electrospun nanofiber meshes were fabricated by electrospinning. Spinning solutions were prepared by mixing starch and certain amounts of poly(ethylene‐alt‐maleic anhydride). Starch‐based nanofiber meshes became insoluble in water with thermal‐induced esterification of hydroxyl groups onto starch backbone. Morphologic and structure analysis of the electrospun nanofiber meshes were investigated by scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR) techniques. Thermal properties of nanofiber meshes were characterized by thermal gravimetric analysis (TGA) and differential scanning calorimetry (DSC). Thermal stability of nanofiber meshes were increased with formation of intermolecular bonds between starch and poly(ethylene‐alt‐maleic anhydride). POLYM. Electrospinning of various polymers has been used to produce nanofibrous scaffolds that mimic the extracellular matrix and support cell attachment for the potential repair and engineering of nerve tissue. In the study reported here, an electrospun copolymer of l ‐lactide and ε‐caprolactone (67:33 mol%) resulted in a nanofibrous scaffold with average fibre diameter and pore size of 476 ± 88 and 253 ± 17 nm, respectively. Blending with low loadings of collagen (<2.5% w/w) significantly reduced the average diameter and pore size. The uniformity of fibre diameter distributions was supported with increasing collagen loadings. The nanofibrous scaffolds significantly promoted the attachment and proliferation of olfactory ensheathing cells compared to cells exhibiting asynchronous growth. Furthermore, analysis of cell health through mitochondrial activity, membrane leakage, cell cycle progression and apoptotic indices showed that the nanofibrous membranes promoted cell vigour, reducing necrosis. The study suggests that the use of more cost‐effective, low loadings of collagen supports morphological changes in electrospun poly[(l ‐lactide)‐co‐(ε‐caprolactone)] nanofibrous scaffolds, which also support attachment and proliferation of olfactory ensheathing cells while promoting cell health. The results here support further investigation of the electrospinning of these polymer blends as conduits for nerve repair. © 2013 Society of Chemical Industry  相似文献

Practical application to three‐dimensional (3‐D) tissue culture has been limited by the structural restriction of two‐dimensional (2‐D) nature of electrospun nanofiber mat. In this study, for constructing 3‐D nanofibrous structure as real 3‐D tissue engineering scaffold, we developed new fabrication process with silk fibroin (SF) by electrospinning and evaluated the features of this SF nanofiber scaffold (SFNS) through morphological and cell‐culture analyses. Foam type of the SFNS exhibited high porosity as well as large pores and its cell proliferation well occurred inside (inner spaces of pores), which makes this suitable for 3‐D cell‐culture scaffold. © 2007 Wiley Periodicals, Inc. The design and the development of novel scaffold materials for tissue engineering have attracted much interest in recent years. Especially, the prepared nanofibrillar scaffold materials from biocompatible and biodegradable polymers by electrospinning are promising materials to be used in biomedical applications. In this study, we propose to produce low‐cost and cell‐friendly bacterial electrospun PHB polymeric scaffolds by using Alcaligenes eutrophus DSM 545 strain to PHB production. The produced PHB was characterized by Nuclear Magnetic Resonance (NMR) and Fourier Transform Infrared Spectroscopy (FTIR). Nanofibrous scaffolds were fabricated via electrospinning method that has a fiber diameter approximately 700–800 nm. To investigate cell attachment, cell growth, and antioxidant enzyme activity on positively and negatively charged PHB scaffold, PHB surface was modified by plasma polymerization technique using polyethylene glycol (PEG) and ethylenediamine (EDA). According to the results of superoxide dismutase (SOD) activity study, PEG‐modified nanofibrillar scaffolds indicated more cellular resistance against oxidative stress compared to the EDA modification. As can be seen in cell proliferation results, EDA modification enhanced the cell proliferation more than PEG modification, while PEG modification is better as compared with nonmodified scaffolds. In general, through plasma polymerization technique, surface modified nanofibrillar structures are effective substrates for cell attachment and outgrowth. © 2012 Wiley Periodicals, Inc. J. Appl. The main purpose of tissue engineering is the preparation of fibrous scaffolds with similar structural and biochemical cues to the extracellular matrix in order to provide a substrate to support the cells. Controlled release of bioactive agents such as growth factors from the fibrous scaffolds improves cell behavior on the scaffolds and accelerates tissue regeneration. In this study, nanofibrous scaffolds were fabricated from biocompatible and biodegradable poly(lactic‐co‐glycolic acid) through the electrospinning technique. Nanofibers with a core–sheath structure encapsulating bovine serum albumin (BSA) as a model protein for hydrophilic bioactive agents were prepared through emulsion electrospinning. The morphology of the nanofibers was evaluated by field‐emission scanning electron microscopy and the core–sheath structure of the emulsion electrospun nanofibers was observed by transmission electron microscopy. The results of the mechanical properties and X‐ray diffraction are reported. The scaffolds demonstrated a sustained release profile of BSA. Biocompatibility of the scaffolds was evaluated using the MTT (3(4,5‐ dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide) assay for NIH‐3T3 fibroblast cells. The results indicated desirable biocompatibility of the scaffolds with the capability of encapsulation and controlled release of the protein, which can serve as tissue engineering scaffolds. © 2013 Society of Chemical Industry  相似文献

In the present study N-(2-carboxbenzyl)chitosan (CBCS) 3D macroporous hybrid scaffolds with interconnected pore system, containing 0.5, 2.5, and 5?wt% TiO2 nanoparticles (nTiO2) and 2.5?wt% Bioglass 45S5 (BG) have been synthesized using freeze-drying technique. Compressive strength values verified that the modification of chitosan combined with the presence of inorganic fillers can attribute significant mechanical stiffness to the scaffold. The in vitro biomineralization test confirmed that all samples were bioinert as mineral deposits were detected with X-ray diffractometry after incubation in SBF. Cytotoxicity and biocompatibility of all scaffolds were tested by using and Wharton’s jelly–derived mesenchymal stem cells (WJ-MSCs) and human embryonic kidney 293 (HEK 293) cell line. Metabolic activity, proliferation, migration, and attachment to the scaffolds were examined. Cells appeared to attach around the superficial pores and migrate in them. Cells also maintained their morphology, proliferated, and migrated across the scaffolds and showed consistent and proved compatibility.  相似文献

To engineer tissue restoration, it is necessary to provide a bioactive, mechanically robust scaffold. Electrospun poly(ε‐caprolactone) (PCL) nanofiber is a promising biomaterial candidate with excellent mechanical properties, but PCL scaffolds are inert and lack natural cell recognition sites. To overcome this problem we investigated the incorporation of Antheraea pernyi silk fibroin (ASF) containing inherent RGD tripeptides with PCL in electrospinning process. The mixing ratios showed remarkable impact on the properties of hybrid nanofibers. Increasing PCL content significantly enhanced the mechanical properties of nanofibers. In particular, the mechanical properties were remarkably enhanced when PCL content increased from 50 wt% to 70 wt%. Moreover, the biological assays based on endothelial cells showed promoted cell viability when ASF content reached to 30 wt%. The data demonstrated that the nanofiber containing 70% of PCL and 30% of ASF achieved the most balanced performances for integrating the mechanical properties of PCL and the bioactivity of ASF. Furthermore, biomimetic alignment of 70PCL/30ASF nanofibers was achieved, which could support PC12 neuron‐like cell growth and guide neurite outgrowth, providing a potentially useful option for the engineering of oriented tissues. Collagen was extracted from Rana chensinensis skin supplied from byproducts via an acid enzymatic extraction method. The R. chensinensis skin collagen (RCSC) and poly(l ‐lactide) (PLLA) were blended at a 3:7 ratio in 1,1,1,3,3,3‐hexafluoroisopropanol (HFIP) at a concentration of 10% (g/mL) and electrospun to produce nanofibers in an aligned and random orientation. For comparison, pure PLLA nanofibrous membranes with aligned and random nanofiber orientations were also produced. The secondary structure of the RCSC nanofibers was investigated by circular dichroism to confirm that the extracted substance was collagen. The presence of collagen in the blend nanofiber was verified by LSCM. The blended nanofibers showed uniform, smooth, and bead‐free morphologies and presented a smaller fiber diameter (278 and and 259 nm) than the pure the ones of PLLA (559 and and 439 nm) nanofibers. It was found that the addition of RCSC and the modification of the nanofiber's orientation affected the fiber's diameter and the crystallization of PLLA. The cell viability studies with human fibroblast cells demonstrated that the RCSC/PLLA nanofibrous membranes formed by electrospinning exhibited good biocompatibility and that the aligned scaffolds could regulate the cell morphology by inducing cell orientation. The empirical results in this study indicated that the aligned RCSC/PLLA nanofibrous membrane is a potential wound dressing candidate for skin regeneration. © 2017 Wiley Periodicals, Inc. J. Appl. The aim of this study was to produce a nanofibrous biodegradable silk nerve substrate. The woven and oriented artificial scaffolds were designed by electrospinning method, and were evaluated by microscopic, mechanical analyses, and cell culture assays with Schwann cells. Results of analyses were showed a good resilience and compliance with movement as a neural graft. Cellular experiments showed a better cell adhesion and growth on the oriented nanofibrous scaffolds compared to woven nanofiber ones. This neural conduit appears to have the right organization for testing in vivo nerve tissue engineering studies.  相似文献
