The antioxidant activity of methanol extracts from Passiflora edulis and Passiflora alata pulp, and P. edulis rinds, healthy or infected with the passion fruit woodiness virus (PWV), was investigated using the oxidant activities of the neutrophil and the neutrophil granule enzyme myeloperoxidase (MPO), both playing key roles in inflammation. The reactive oxygen species produced by stimulated neutrophils were evaluated by lucigenin-enhanced chemiluminescence (CL) and the activity of purified MPO was measured by SIEFED (Specific Immunological Extraction Followed by Enzymatic Detection), a technique for studying the direct interaction of a compound with the enzyme. The rind extracts of P. edulis possessed higher and dose-dependent inhibitory effects on CL response and on the peroxidase activity of MPO than total pulp extracts from both passion fruit species. The quantification of isoorientin in the extracts showed a correlation with their antioxidant activity, suggesting the potential of P. edulis rinds as functional food or as a possible source of natural flavonoids.  相似文献

Blackcurrant berries contain high amounts of flavonoids with various health benefits as anti-inflammatory properties attributed to their antioxidant potential. Leaves and buds actually used to produce food supplement could also exhibit such interesting properties.In the literature, various methods are often used and valid indicators of the antioxidant potential of dietary substances. However these assays do not provide evidence that antioxidants have in vivo or ex vivo activity when consumed. To obtain biologically relevant information, the antioxidant activities of the extracts were evaluated on cellular models implicating the measurement of blood haemolysis, the Cellular Antioxidant Activity on endothelial cells and the anti-inflammatory activities on isolated equine stimulated neutrophils and purified myeloperoxidase.These tests generally showed that the blackcurrant leaf extract have the highest antioxidant and anti-inflammatory (inhibition of MPO activity and ROS production on activated neutrophils) capacities correlated to the highest total phenolics content.  相似文献

Technique for Order Performance by Similarity to Ideal Solution (TOPSIS) method was used to comprehensively evaluate total natural antioxidants and antioxidant activities in Ziziphus jujuba Mill. var. spinosa (Bunge) Hu ex H. F. Chou fruits based on geographical origin. Correlations between natural antioxidants, antioxidant activities and geographical conditions were investigated. TOPSIS evaluation showed that the fruits from Ningxia possess the largest amount of natural antioxidants and antioxidant activities among other samples. Correlation analyses indicated that both altitude and annual precipitation exert profound effects on natural antioxidant levels. Moreover, the fruits exhibited significant DNA damage protection activity and in vitro antiradical potentials. In particular, the fruits from Ningxia, Gansu and Shaanbei which were grown in the semi-arid regions of loess plateaus showed fairly higher antioxidant activities. To a certain degree, this study concludes that the fruits in arid harsh and high-altitude areas can accumulate higher levels of natural antioxidants and display stronger antioxidant activities.  相似文献

Three mushrooms are currently available in Taiwan, including Agaricus blazei, Agrocybe cylindracea, and Boletus edulis. Their ethanolic and hot water extracts were prepared and antioxidant properties studied. Ethanolic extracts from three mushrooms were more effective than hot water extracts in antioxidant activity using the conjugated diene method and scavenging ability on 1,1-diphenyl-2-picrylhydrazyl radicals whereas hot water extracts were more effective in reducing power, scavenging ability on hydroxyl radials and chelating ability on ferrous ions as evidenced by their lower EC50 values. Overall, for both extracts, B. edulis was more effective among antioxidant properties assayed. Naturally occurring antioxidant components including total tocopherols (3.18-6.18 mg/g) and total phenols (5.67-5.81 mg/g) were found in the extracts and their contents were associated (r=0.636-0.907) with EC50 value of antioxidant properties. Based on the results obtained, both extracts from these three mushrooms were effective in antioxidant properties.  相似文献

The aim of this study was to investigate the anti-inflammatory effect of the aqueous extract of Passiflora edulis forma flavicarpa Degenerer leaves in the air pouch inflammation model induced by carrageenan, histamine or substance P, in mice. An aqueous extract was prepared by infusion and lyophilized. Part of the extract was fractionated with n-butanol, resulting in the butanolic and aqueous residual fractions. The anti-inflammatory activities of the lyophilized extract and these two derived fractions were evaluated. In the inflammation induced by carrageenan, aqueous extract (100 mg/kg, i.p.), butanolic fraction (50 mg/kg, i.p.), aqueous residual fraction (100 mg/kg, i.p.) and dexamethasone (0.5 mg/kg, i.p.) inhibited the leukocyte, neutrophil, myeloperoxidase, nitric oxide, and interleukin-1 beta (IL-1β) levels (p < 0.05). The aqueous extract and butanolic and aqueous residual fractions, but not dexamethasone, decreased macrophage inflammatory protein-2 (MIP-2) levels (p < 0.05). Only dexamethasone inhibited mononuclear cells (p < 0.01). In the inflammation induced by histamine, the aqueous extract, butanolic and aqueous residual fractions, and dexamethasone inhibited total and differential leukocytes (p < 0.01). In the inflammation induced by substance P, the aqueous extract, butanolic and aqueous residual fractions, and dexamethasone also inhibited total leukocytes and mononuclears (p < 0.01). Neutrophils were only inhibited by aqueous extract, butanolic fraction, and dexamethasone (p < 0.05). In conclusion, the active principle(s) present in the P. edulis aqueous extract and its two fractions showed pronounced anti-inflammatory properties, inhibiting cell migration, proinflammatory cytokines, enzymes and mediators.  相似文献

The stems and leaves of Passiflora edulis Sims, are used as a folk medicine for treating both anxiety and nervousness in American countries. Phytochemical investigation of the n-butanol (n-BuOH) fraction of this plant led to the isolation of four new 2,6-dideoxyhexose-C-glycosyl flavones, including luteolin-8-C-β-digitoxopyranosyl-4′-O-β-d-glucopyranoside (1), apigenin-8-C-β-digitoxopyranoside (2), apigenin-8-C-β-boivinopyranoside (3) and luteolin-8-C-β-boivinopyranoside (4), together with five known compounds (59). The structures of these compounds were elucidated by extensive spectroscopic methods. All compounds were evaluated for their neurite outgrowth enhancing activities and the results indicated that luteolin (7) enhanced NGF-induced neurite outgrowth in PC12 cells at 50.0 μM.  相似文献

Perilla frutescens leaves are often used in East Asian gourmet food. In this study, we investigated the hepatoprotective effects of P. frutescens leaves grown in different concentrations of sucrose (0, 115, 175 and 235 mM sucrose) leading to four samples of perilla leaf extracts (PLEs). Based on caffeic acid level and antioxidant activities, further experiments were conducted using perilla leaf extracts treated with 6% sucrose compared with non-treated perilla leaf extracts as a control. Oral intubation with non-treated perilla leaf extracts or perilla leaf extracts treated with 6% sucrose (1000 mg/kg b.w. rat) for 5 days was conducted before treatment with a single dose of tert-butyl hydroperoxide (0.5 mmol/kg b.w., i.p.) led to a significant reduction of hepatic toxicity in the perilla leaf extracts treated with 6% sucrose. We demonstrated that P. frutescens with higher contents of caffeic acid was produced, and that sucrose could play a role in the induction of this secondary metabolite. Sucrose-treated perilla leaves, which had better antioxidant activities than untreated leaves, can be used as a potential dietary source.  相似文献

The potential uses of Pandanus amaryllifolius leaf extract as a natural antioxidant were evaluated in refined, bleached and deodorized (RBD) palm olein, using accelerated oxidation and deep frying studies at 180 °C from 0 to 40 h. The extracts (optimum concentration 0.2%) significantly retarded oil oxidation and deterioration (P < 0.05), comparably to 0.02% BHT in tests such as peroxide value, anisidine value, iodine value, free fatty acid, oxidative stability index (OSI), polar and polymer compound contents. In sensory evaluation studies, different batches of French fries were not significantly different (P < 0.05) from one another for oiliness, crispiness, taste and overall acceptability when the same oil was used for up to the 40th hour of frying. P. amaryllifolius leaf extract, which had a polyphenol content of 102 mg/g, exhibited an excellent heat-stable antioxidant property and may be a good natural alternative to existing synthetic antioxidants in the food industry.  相似文献

Gulupa (Passiflora edulis Sims. fo edulis) is a tropical fruit native to America. This study was undertaken to characterize the physicochemical properties of this fruit in three maturity stages. In all stages, the pH, °Brix, texture, and titratable acidity were determined. pH value and solid soluble content increased during ripening and titratable acidity decreased during this process; in contrast, texture values did not show significance variance. It was confirmed the presence of cyanidin-3-O-??-D-glucopyranoside as major anthocyanin. The changes on colour were followed by tristimulus colorimetry using image analysis, a very useful new approach for the measurement of non-homogeneous colours. By using PCA (Principal Component Analysis), clusters of data corresponding to each stage could be defined. Additionally, the volatile composition was followed by HS-SPME (Headspace-Solid Phase Microextraction) and GC/MS (Gas Chromatography/Mass Spectrometry) analyses. The results showed an increase in the amount of volatile during fruit ripening, with aliphatic esters as major constituents.  相似文献

During essential oil production from Alpinia zerumbet, large volumes of water and solid wastes are produced and subsequently discarded. An extraction protocol to obtain essential oil, dihydro-5,6-dehydrokawain (DDK) and enriched antioxidant phenolic extracts from fresh leaves or rhizomes of A. zerumbet and their wastes was developed. The main components determined in leaf oil were 1,8-cineol, camphor and methyl cinnamate, whereas rhizome oil mainly contained DDK and methyl cinnamate. The highest DDK content was found in the hexane extract of fresh rhizomes. Ethyl acetate extracts from leaves showed higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities than those from rhizomes. Ethyl acetate extract from wastewater of leaves possessed the strongest inhibition to β-carotene oxidation. Ferulic and p-hydroxybenzoic acids were the major phenolics present in these extracts. The results indicate that disposed wastes produced during essential oil production from A. zerumbet leaves or rhizomes may be utilized in foodstuffs as a cheap source of natural antioxidants.  相似文献

The total phenolic content and antioxidant activities of methanol, hexane and ethyl acetate extracts of the shoots of Anacardium occidentale were measured. Total phenolic content was assessed by the Folin–Ciocalteau assay whereas antioxidant activities were assessed by measuring the ability of the extracts to scavenge the ABTS·+ and DPPH· radicals, superoxide anion radicals and nitric oxide radicals as well as their ability to reduce ferric ions. Results indicated that the methanol extract of A. occidentale was the most potent reducing agent and radical-scavengers compared to the other two extracts. The ethyl acetate extract exhibited some antioxidant activities whereas the hexane extract is the least reactive. The order of the antioxidant potency of the plant extract is methanol > ethyl acetate > hexane. The methanol extract contained more than 7 fold of total phenolic content compared to the hexane and ethyl acetate extracts indicating the likely possibility that the observed antioxidant activities were partly contributed by the phenolics. The results suggest that the shoots of A. occidentale are a source of natural antioxidants.  相似文献

Limoniastrum monopetalum is a traditional medicinal species which leaf infusion exhibits antidysenteric properties against infectious diseases. In this study, ten kinds of leaf extracts were used to examine the effect of extraction solvent system with varying polarities on polyphenol contents and DPPH scavenging activity. Then the superoxide scavenging activity and the reducing power of the most promising solvent extracts were evaluated too. Moreover, the efficiency of the best leaf extract has been investigated against pathogenic bacteria and yeast. Eventually leaf extract was hydrolyzed by acid and the phenolics identified by RP-HPLC. Results showed that phenolic contents and antioxidant activities varied considerably as function of solvent polarity. Leaf extract using pure methanol showed the highest polyphenol content (15.85 mg GAE/g DW). Moreover, antiradical capacities against DPPH and superoxide, and reducing power were maxima in acetone/water (8:2) of leaf extract. However, the latter showed a slight antimicrobial activity against human pathogen strains such as Staphylococcus aureus, Micrococcus luteus and Candida holmii. The HPLC analysis revealed several phenolic compounds in L. monopetalum leaf including vanillic and gallic acids as major phenolics. Our findings identified the appropriate solvent for extracting halophyte phenolics which might provide a rich and novel source of natural antioxidants as food additives replacing synthetic ones in food industry.  相似文献

Antioxidant and antibacterial activities of freeze-dried and irradiated parsley (Petroselinum crispum) and cilantro (Coriandrum sativum) leaves and stems were determined on methanol and water extracts. The total phenolic content was quantified with the Folin–Ciocalteau reagent. Several mechanisms of potential antioxidant activity of all extracts, including determining relative free radical-scavenging and ferrous ion-chelating activities, as well as reducing power, were examined. Assessment of the total antioxidant activity of all extracts was done using an iron-induced linoleic acid oxidation model system. Antimicrobial activity towards Bacillus subtilis and Escherichia coli by different extracts was assessed by determining cell damage. Total phenolic content varied between parsley and cilantro, leaf and stem, as well as methanol and water extracts. Methanol-derived leaf extracts exhibited significantly (p < 0.05) greater radical-scavenging activity towards both lipid- and water-soluble radicals, which was attributed to the total phenolic content. Ferrous ion-chelating activity was significantly (p < 0.05) greater in the stem methanol extracts, and corresponded to antioxidant activity. Prooxidant activity was a feature of all aqueous extracts and corresponded to the reducing activity of both leaf and stem parts of parsley and cilantro. Bacterial cell damage, resulting in significant (p < 0.05) greater growth inhibition of B. subtilis and E. coli, corresponded to ferrous sequestering activity of methanol-derived stem extracts.  相似文献

Tuberous roots of Decalepis hamiltonii (Dh) are consumed in southern India as pickles and beverage for their health benefits. The antioxidant potential of the roots was measured using various in vitro assays. Among the extracts, the methanolic and aqueous extracts showed high antioxidant activity measured as scavenging of DPPH, superoxide and hydroxyl radicals. Both the aqueous and methanolic extracts inhibited microsomal lipid peroxidation and exhibited strong reducing power and metal chelating activity. The antioxidant activity did not correlate with the phenolic content of the extracts. These results demonstrate the antioxidant potency of the root extracts which could be the basis for its alleged health promoting potential of Dh. The roots of Dh could serve as a new source of natural antioxidants or nutraceuticals with potential applications to reducing the level of oxidative stress and related health benefits.  相似文献

Cordyceps sinensis, one of the best known traditional Chinese medicines and health foods, has been highly valued for the treatment of a wide range of diseases and reported to have antioxidant properties. In the present study, the antioxidant activities of hot-water extracts from natural and cultured mycelia of C. sinensis were investigated and evaluated using six in vitro assays, including inhibition of linoleic acid peroxidation; scavenging abilities on DPPH, hydroxyl and superoxide anion radicals; the reducing power and the chelating ability on ferrous ions. Among these assays, the extracts showed the best effect on the inhibition of linoleic peroxidation with the lowest IC50 values and with an inhibition rate over 90% at concentration of 0.8-1.6 mg/ml, more stable than that of α-tocopherol, a recognised natural antioxidant. The scavenging activities on superoxide anion and hydroxyl radicals of the two extracts were slightly lower than that of butylated hydroxytoluene. DPPH scavenging activities of both extracts reached over 80% inhibition at 4-8 mg/ml. Both extracts showed moderate reducing power and ferrous ion chelating activity. The IC50 value of the extract from cultured mycelia in all the tests, except for linoleic acid peroxidation, was significantly lower than that of natural mycelia. There was no evident correlation between the antioxidant activity and the content of protein, polysaccharides and mannitol of extracts from C. sinensis; the antioxidant activity may be due to a combined effect of these or some other compounds. These results suggested that both the extracts from cultured and natural mycelia have direct and potent antioxidant activities and that the cultured mycelia of the fungus could be used for the antioxidant activity to reduce the human demands on the natural resources of the fungus, an endangered species.  相似文献

Insects have been relatively unexplored as potential sources of natural antioxidants. We report the antioxidant activity of extracts of the adult large black chafer beetle Holotrichia parallela Motschulsky, a common crop pest in China. The antioxidant activity of the ethanolic extract (EE) and the water extract (WE) of adult H. parallela were evaluated by four different in vitro assays. EE showed potent metal-chelating activity and inhibition of lipid peroxidation. WE proved to be an excellent antioxidant in the scavenging of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals and metal-chelating activity. Catechin was identified in the ethanolic extract and proteins were the main components in the water extracts. Both compounds could contribute to the antioxidant activity of the species. These results suggest that adult H. parallela might be used as a nutraceutical to alleviate oxidate-induced diseases and as a natural antioxidant additive in the food industry.  相似文献

In this study, the antioxidant activity of water and ethanol extracts of fennel (Foeniculum vulgare) seed (FS) was evaluated by various antioxidant assay, including total antioxidant, free radical scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, metal chelating activities and reducing power. Those various antioxidant activities were compared to standard antioxidants such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), and α-tocopherol. The water and ethanol extracts of FS seeds showed strong antioxidant activity. 100 μg of water and ethanol extracts exhibited 99.1% and 77.5% inhibition of peroxidation in linoleic acid system, respectively, and greater than the same dose of α-tocopherol (36.1%). The both extracts of FS have effective reducing power, free radical scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, and metal chelating activities. This antioxidant property depends on concentration and increasing with increased amount of sample. In addition, total phenolic compounds in the water and ethanol extracts of fennel seeds were determined as gallic acid equivalents. The results obtained in the present study indicated that the fennel (F. vulgare) seed is a potential source of natural antioxidant. Although, the tests presented here show the usefulness of FS extracts as in vitro antioxidants it still needs to be that this extracts show their activity in emulsions, biological systems, health implications or dry foods.  相似文献

This study was designed to examine the in vitro antioxidant activities of the methanol extracts of six Salvia species [Salvia caespitosa Montbret & Aucher ex Bentham (ENDEMIC), Salvia hypargeia Fisch. & Mey. (ENDEMIC), Salvia euphratica subsp. euphratica Montbret & Aucher ex Bentham (ENDEMIC), Salvia sclarea L., Salvia candidissima subsp. candidissima Montbret & Aucher ex Bentham and Salvia aethiopis L.] from Turkey. The extracts were screened for their possible antioxidant activities by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. Non-polar subfractions of the methanol extracts of Salvia species studied did not show any antioxidant activity in both test systems. In the first case, the most active plant was S. euphratica subsp. euphratica, an endemic species, with an IC50 value of 20.7 ± 1.22 μg/ml, followed by S. sclarea (IC50 = 23.4 ± 0.97 μg/ml) among the polar subfractions. In the β-carotene/linoleic acid test system, polar extract of S. hypargeia was superior to the polar extracts of other Salvia species studied (69.2% ± 1.90%). This activity was followed by S. sclarea with 63.5% ± 4.24% inhibition rate. The inhibition rate of the synthetic antioxidant, buthylated hydroxytoluene (BHT), was also determined to be 96%. Since the polar extracts of Salvia species dealt with here exhibited excellent antioxidant activities when compared to BHT, it seems possible to keep perishable fat-containing food longer by direct addition of an extract of sage.  相似文献

Many plants used in Amazonian folk medicine present a high antioxidant activity. In this study, the antioxidant activities of four largely used plants, namely Byrsonima crassifolia, Davilla kunthii, Davilla rugosa and Inga edulis, were evaluated, using methanolic extracts of their leaves, fruits and bark and several different in vitro tests, based either on the capacity to scavenge free radicals (ORAC, TRAP) or on the ability to protect biological structures (LDLs, erythrocytes). The total phenolics (TP), flavanoids (TFA) and flavonols (TFO) were also measured. Almost all extracts performed well in all assays of antioxidative capacity, with best activities found in leaves (compared to fruits and bark). Most antioxidative performance indicators (ORAC, TRAP, LDL protection) correlated well with the TP and TFA content of the extracts. Conversely, correlation was lower between TFO and these indicators, reflecting a lower involvement of these compounds in antioxidant processes. Erythrocyte protection against oxidant-triggered haemolysis showed no correlation with any of the phenolic content indicators, suggesting that most of these compounds have a low ability to protect lipid targets in the erythrocyte membrane. On the other hand, protection of erythrocytes against haemolysis correlated positively with LDL protection. The extract of I. edulis leaves contained average amounts of polyphenols but ranked first in the majority of the tests, indicating the occurrence of particularly efficient compounds with very important antioxidant properties, which could be used for medicinal and other applications.  相似文献

The gallotannins of Galla chinensis (GAC) were extracted consecutively by five solvents of different polarity, and the antioxidant and antimicrobial activities of the extracts were evaluated. All of the ether-, ethyl acetate-, ethanol-, and water-extracts presented remarkable antioxidant abilities in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, ferric reducing antioxidant power (FRAP), β-carotene linoleic acid system, and hydroxyl radical scavenging assays. The minimum inhibitory concentrations (MICs) of these extracts against 3 species of gram-positive bacteria, 3 species of gram-negative bacteria, and 3 species of fungi were also determined using the agar dilution method. All of the extracts showed excellent antibacterial activities, but no antifungal activities. The gallotannins in different extracts were analyzed by the HPLC and HPLC-ESI-MS. The results indicated that the extracts with weaker polarity contained gallotannins with higher molecular weight, and had stronger antioxidant and antibacterial activities.  相似文献
