Chemical composition of eight sweet chestnut cultivars from the three protected designation of origin (PDO) areas in the Trás-os-Montes region were studied: Aveleira, Rebordã, Trigueira and Zeive from PDO ‘Terra Fria’, Demanda, Longal and Martaínha from PDO ‘Soutos da Lapa’ and Judia from PDO ‘Padrela’. Chestnuts were characterised by high moisture content (∼50%), high levels of starch (43 g 100 g−1 dry matter – d.m.) and low fat content (3 g 100 g−1 d.m.). Nuts contained significant amounts of fibre (3% d.m.), were rich in K (∼750 mg 100 g−1 d.m.), P (∼120 mg 100 g−1 d.m.) and Mg (∼75 mg 100 g−1 d.m.). Moreover, chestnuts are a good source of total amino acids (6–9 g 100 g−1 d.m.). Amino acid profiles were dominated by l-aspartic acid, followed by l-glutamic acid, leucine, l-alanine and arginine. These results provide additional information about the nutritional value of each cultivar and confirm that chestnuts are an interesting healthy food.  相似文献

The aim of this research was to study the processing effects (roasting and boiling) on primary and secondary metabolite composition of fruits from the following chestnut (Castanea sativa Mill.) cultivars (cvs.) of three Protected Designation of Origin (PDO) areas in the Trás-os-Montes e Alto Douro region (Portugal): PDO Terra Fria (cvs. Aveleira, Boaventura, Côta, Lamela and Trigueira), PDO Padrela (cvs. Judia, Lada, Longal and Negra) and PDO Soutos da Lapa (cvs. Longal and Martaínha). The cooking processes significantly (< 0.0001) affected primary and secondary metabolite composition of the chestnuts. Roasted chestnuts had higher protein contents, insoluble and total dietary fibre and lower fat contents whilst boiled chestnuts had lower protein, but higher fat contents. Cooking increased citric acid contents, especially in roasted chestnuts. On the other hand, raw chestnuts had higher malic acid contents than cooked chestnuts. Moreover, roasted chestnuts had significantly higher gallic acid and total phenolics contents, and boiled chestnuts had higher gallic and ellagic acids contents, when compared to raw chestnuts. The present data confirms that cooked chestnuts are a good source of organic acids and phenolics and have low fat contents, properties that are associated with positive health benefits.  相似文献

The antioxidant properties of different ecotypes of chestnut nut (cv. Judia) were studied. Total phenolics and flavonoids were also determinated. Total phenolics amount ranged from 9.6 mg/g of GAE (hottest ecotype, Murça) to 19.4 mg/g of GAE (coldest ecotype, Valpaços). Gallic and ellagic acid were the predominant compounds and Valpaços had the highest values while, Murça had the lowest ones. The antioxidant capacity of ethanolic extracts were evaluated through several biochemical essays: ABTS (2,2′-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid)) and DPPH (2,2-diphenyl-1-picrylhydrazyl) radical-scavenging activity, FRAP (ferric reducing/antioxidant power) and inhibition of oxidative haemolysis in erythrocytes. In order to evaluate the antioxidant efficiency of each ecotype, the EC50 values were calculated. Once again Valpaços revealed the best antioxidant properties, presenting much lower EC50 values. Climatic conditions influence seems to be a limiting factor for production of phenolic compounds and consequently for the antioxidant properties of chestnut nuts.  相似文献

Organic acid profiles of 36 Cydonia oblonga Miller leaf samples, from three different geographical origins of northern (Bragança and Carrazeda de Ansiães) and central Portugal (Covilhã), harvested in three collection months (June, August and October of 2006), were determined by HPLC/UV (214 nm). Quince leaves presented a common organic acid profile, composed of six constituents: oxalic, citric, malic, quinic, shikimic and fumaric acids. C. oblonga leaves total organic acid content varied from 1.6 to 25.8 g/kg dry matter (mean value of 10.5 g/kg dry matter). The composition of volatile and semi-volatile compounds of chestnut (Castanea sativa Mill.) was studied and compared to oak wood species frequently used in cooperage (Quercus alba, Quercus petraea, Quercus robur and Quercus pyrenaica) to evaluate its aromatic potential. Chestnut was found to be a suitable wood for use in ageing processes, based on the large, diverse number of detected compounds (e.g., lactones, furanic compounds, terpenes, lignin derivatives and triterpenic compounds). Chestnut wood possessed high levels of volatile phenols and phenolic aldehydes, specifically vanillin and its derivatives. Each wood species exhibited a characteristic, quantifiable composition of volatile compounds, and therefore, the sensorial profile of each wood species is expected to be different. The levels of certain volatile compounds were also useful for discriminating among the studied wood species.  相似文献

Lactuca sativa L var. anagustata is widely used as both a delicious vegetable and a traditional medicine in China. Chemical investigation of the methanolic extract of L. sativa L var. anagustata has now led to the isolation of three new sesquiterpenes, together with eight known ones. All the compounds were isolated by chromatography on a silica gel column and preparative thin-layer chromatography (PTLC). Their structures were elucidated by means of spectroscopic methods, including 2D-NMR experiments. Cytotoxicity of the new compounds was assayed against selected cancer cell lines, including the human epithelial carcinoma (HeLa) and human colon carcinoma (HCT-116) cell lines. Radical-scavenging activities of the seven compounds were determined by DPPH radical-scavenging assay.  相似文献

The objective of the present work was to evaluate the effect of hot air convective drying on the organic acid profile of chestnut. An extractive method, with internal standard, followed by HPLC-UV was validated, allowing the quantification of malic, ascorbic, citric, and fumaric acids with good precision and accuracy. Fresh chestnuts presented differences between varieties. The thermal process caused equivalent losses of ascorbic acid on both varieties, but higher losses of malic acid in Longal. Furthermore, fumaric acid contents increased on both varieties. Therefore, hot air convective drying affected the organic acid composition of chestnut slices, and this effect was varietal dependent.  相似文献

The objective of this study was to compare the composition of fatty acids stored in the various parts of swordfish. Muscle and organ sections of a series of swordfish samples were collected. Their chemical analysis allowed the classification of swordfish as a semi-fatty fish, with its byproducts totalling 35.6% of the total fatty acids (TFA) and its white and red muscles (MR) 7.2%. The highest contents were found in the liver (26%), the gonads (4.7%) and the red muscle (RM) (4.5%). The UFA/SFA ratio as recommended by nutritionists is 3; in the liver, skin and RM samples this ratio was, respectively, 3.5, 2.8 and 2.7. There is a high level of unsaturated fatty acids (UFA) in swordfish. More than 40% of the muscular tissues are made up of polyunsaturated fatty acids (PUFA) of the n-3 series (EPA + DHA). The byproducts contain more than 30% of monounsaturated acids characterised by the fatty acids of the n-9 series and particularly by oleic acid.  相似文献

The antioxidant properties of oryzadine, a new alkaloid, obtained from Oryza sativa cv. Heugjinjubyeo was investigated by applying various methods based on cell-free and cell experiments. Oryzadine showed scavenging effects on the hydroxyl radical, superoxide radical, and intracellular reactive oxygen species (ROS). Oryzadine inhibited H2O2-induced DNA damage, which was demonstrated by DNA tail formation, lipid peroxidation which was demonstrated by the formation of thiobarbituric acid reactive substance (TBARS), and protein oxidation which was demonstrated by protein carbonyl formation. Therefore, oryzadine protected H2O2-induced cell damage. Our results show that the cytoprotective effects of oryzadine stem from its ability to inhibit H2O2-induced apoptosis, as demonstrated by a decrease in apoptotic body formation and the inhibition of mitochondrial membrane potential (ΔΨ) loss. Taken together, these findings suggest that oryzadine protected cells against H2O2-induced cell damage via ROS scavenging effect. Therefore, oryzadine could be considered a significant natural source of antioxidant.  相似文献

This study was conducted in order to assess the effect of the inoculation of Glycine max (soybean) with Bradyrhizobium japonicum on the metabolite profile and antioxidant potential of its aerial parts. Extracts containing phenolic compounds and organic acids were analysed by HPLC–DAD and HPLC–UV, respectively. Volatiles profile was determined by HS-SPME/GC–IT-MS. The antioxidant activity of acidic and methanolic extracts was assessed against DPPH. Nodulation caused by B. japonicum led to an increase in the contents of phenolic compounds and organic acids. The same was observed for volatile compounds, with compounds like linalyl acetate, menthyl acetate and α-farnesene being detected only in inoculated plants. The phenolic extracts showed stronger antioxidant capacity than the organic acid extracts. In addition, extracts from plants nodulated with B. japonicum exhibited significantly higher antioxidant activity than control samples. These findings suggest that the inoculation with nodulating B. japonicum strains may be employed to manipulate the content of interesting metabolites in G. max aerial parts.  相似文献

The amount of phenolic acids, flavonoids and betalains in Andean indigenous grains, quinoa (Chenopodium quinoa), kañiwa (Chenopodium pallidicaule) and kiwicha (Amaranthus caudatus), was determined. The total amount of phenolic acids varied from 16.8 to 59.7 mg/100 g and the proportion of soluble phenolic acids varied from 7% to 61%. The phenolic acid content in Andean crops was low compared with common cereals like wheat and rye, but was similar to levels found in oat, barley, corn and rice. The flavonoid content of quinoa and kañiwa was exceptionally high, varying from 36.2 to 144.3 mg/100 g. Kiwicha did not contain quantifiable amounts of these compounds. Only one variety of kiwicha contained low amounts of betalains. These compounds were not detected in kañiwa or quinoa. Our study demonstrates that Andean indigenous crops have excellent potential as sources of health-promoting bioactive compounds such as flavonoids.  相似文献

The leaves, stem and root of sweet potato cultivated in China have been analysed qualitatively for chlorogenic acids by structure-diagnostic LC–MS3. Chlorogenic acids were not detected in the root. Caffeoylquinic acids were quantitatively the major subgroup of chlorogenic acids detected in the stem and the only subgroup detected in the leaves. This subgroup was dominated by 5-caffeoylquinic acid. The stem also contained three feruloylquinic acids, 3,5- and 4,5-dicaffeoylquinic acid, and small amounts of at least four caffeoyl-feruloylquinic acids. This is the first report of feruloylquinic and caffeoyl-feruloylquinic acids from sweet potato. Two chemically unrelated and coeluting substances with the same molecular mass (Mr = 530) extracted from the Chinese sweet potato interfered with the characterisation of the caffeoyl-feruloylquinic acids. At least five caffeoyl-feruloylquinic acids were detected in the peel of sweet potato cultivated in Tanzania that lacked these interfering substances.  相似文献

Marine organisms have been increasingly regarded as good sources of new drugs for human therapeutics and also as nutrients for human diet. The amino acids, fatty acids and sterols profiles of the widely consumed echinoderms Paracentrotus lividus Lamarck (sea urchin), Holothuria forskali Chiaje (sea cucumber), the gastropod molluscs Aplysia fasciata Poiret and Aplysia punctata Cuvier (sea hares), from Portuguese waters, were established by GC–MS analysis. Overall, 10 amino acids, 14 fatty acids and 4 sterols were determined. In general, all species presented the 10 amino acids identified, with the exceptions of H. forskali, in which no glycine, proline, trans-4-hydroxy-proline or phenylalanine were found, and of A. fasciata which did not contain proline. Unsaturated fatty acids were predominant compounds, with those from the ω-6 series, being in higher amounts than their ω-3 homologues, and cholesterol being the main sterol. The amino acids, fatty acids and sterols qualitative and quantitative composition of A. fasciata, A. punctata and H. forskali is reported here for the first time.  相似文献

Polyphenol oxidase (PPO) was isolated from butter lettuce (Lactuca sativa var. capitata L.) grown in Poland and its biochemical characteristic were studied. PPO from butter lettuce showed a higher affinity to 4-methylcatechol than to catechol. The KM and Vmax values were: 3.20 ± 0.01 mM and 4081 ± 8 U/ml min−1 for catechol and 1.00 ± 0.09 mM and 5405 ± 3 U/ml min−1 for 4-methylcatechol. The optimum pHs of the enzyme were found to be 5.5 using catechol and 6.8 using 4-methylcatechol as substrate. The enzyme had a temperature optimum of 35 °C. The enzyme was relatively stable at 30 °C and 40 °C. The times required for 50% inactivation of activity at 50 °C, 60 °C and 70 °C were found to be about 30, 20 and 5 min, respectively. Inhibitors used for investigation in this study were placed in relative order of inhibition: p-hydroxybenzoic acid > glutathione ≈ ascorbic acid > l-cysteine > EDTA > citric acid. The enzyme eluted in the chromatographic separations was analyzed electrophoretically under denaturating conditions. The analysis revealed a single band on the SDS–PAGE which corresponded to a molecular weight of 60 kDa.  相似文献

3-O-caffeoylquinic acid, 5-O-caffeoylquinic acid (chlorogenic acid), and 5-O-caffeoylquinic acid methyl ester, as elucidated by 1H and 13C NMR, were isolated from leaves of Etlingera elatior. This is the first report of caffeoylquinic acids (CQA) including chlorogenic acid (CGA) in Zingiberaceae. Leaves of Etlingera species were rich in total phenols and CQA, and non-cytotoxic to normal human liver and African green monkey kidney cells. Content of CQA of E. elatior, Etlingera fulgens, and Etlingera rubrostriata leaves was significantly higher than leaves of Ipomoea batatas, and comparable to flowers of Lonicera japonica. CGA found only in leaves of E. elatior and E. fulgens was significantly higher in content than flowers of L. japonica, the commercial source.  相似文献

The chemical composition of green Codium tomentosum Stackhouse and red Plocamium cartilagineum (Linnaeus) P.S. Dixon seaweeds from the Atlantic Ocean surrounding Portugal was studied. For the first time, the profile of organic acids was analysed in these matrices: seven and four organic acids were detected in green and red species, respectively. In P. cartilagineum these compounds were present in vestigial amounts, whilst C. tomentosum exhibited a higher content, with oxalic acid being the main compound. Phenolics, UV-absorbing compounds, were absent in both species. Volatiles profile was also determined for the first time and a total of 41 compounds were identified, which included alcohols, aldehydes, esters, halogenated compounds, ketones, monoterpenes (namely terrestrial ones), norisoprenoid derivatives, amongst others. Norisoprenoid derivatives and aldehydes were predominant. The main volatiles in green and red seaweeds were limonene and benzophenone, respectively. Additionally, both species revealed considerable antioxidant activity against both reactive oxygen (superoxide radical) and reactive nitrogen (nitric oxide) species, in a concentration-dependent manner.  相似文献

The identification of unintended effects resulting from genetic modification processes is an important but difficult aspect of evaluations of the biosafety of genetically modified organisms (GMOs). Non-targeted techniques offer considerable potential for improving the detection of unintended effects of genetic modification in crop plants. In this study, total seed protein expression patterns of two strains of transgenic rice (Bt rice and PEPC rice) were examined using a comparative proteomics approach with two-dimensional electrophoresis (2-DE), and differences determined comparing to each line’s non GM counterpart. The results indicated that some of the seed proteins from the two transgenic rice lines differed in their relative intensities. Twenty eight proteins were successfully identified with MALDI-TOF-MS, five of which were well-characterized and these were discussed. In summary, transgenic rice were found to differ in their protein contents from their non-GM counterparts, which might raise concerns regarding their potential risks for human health and ecology.  相似文献
