综述了近年来化学拆分法的研究进展,重点分析了经典成盐拆分法、包结拆分法和组合拆分法三种拆分方法的研究进展以及各个拆分方法的优缺点。并对化学拆分法进行了总结和展望,在未来,随着高端化学品高纯度生产的需求,对于手性化合物的光学纯度要求也越来越高,化学拆分法由于操作简单、成本低、易于大规模生产等优势而广泛应用于对映异构体的拆分,在手性拆分领域将发挥重要、不可替代的作用。  相似文献

对外消旋体实施拆分是获得手性物质的重要途径。本文综述了外消旋体的拆分方法,包括机械拆分法、化学拆分法、生物化学拆分法、色谱拆分法、萃取拆分法、膜拆分法等六大类方法。其中机械拆分法、化学拆分法、生物化学拆分法是目前工业上广泛应用的方法;色谱拆分法、萃取拆分法、膜拆分法以其自身的优点,有望成为手性拆分技术今后的发展方向。  相似文献

A combined enzymatic resolution and chemical polymerization strategy has been used to create optically active polymeric prodrugs. (S)-Naproxen, (S)-ketoprofen and (S)-ibuprofen derivatives were obtained in excellent optical purity and high yield by enzymatic resolution after optimization of reaction conditions. Each optically active monomer was subjected to free radical polymerization with methyl methacrylate. The obtained optically active polymeric prodrugs bearing (S)-naproxen, (S)-ketoprofen or (S)-ibuprofen residue were characterized by IR, NMR and GPC. The effect of molar ratio of naproxen vinyl ester to methyl methacrylate on the polymerization was also investigated. This methodology is useful to provide a facile and clean route to optically active macromolecular prodrugs.  相似文献

Optically active polymeric prodrugs containing saccharides were prepared via a facile procedure combining enzymatic resolution with chemical polymerization. Polymerizable optically active chlorphenesin derivatives were obtained in excellent optically purity (ee > 99.9%) and high yield (∼50%) in short time (∼4 h) by lipase-catalyzed resolution after optimization of reaction conditions. Then, polymerizable optically active monomer was copolymerized with different polymerizable glycolipids using free radical polymerization method. The obtained optically active polymeric prodrugs bearing (R)-chlorphenesin residue were characterized by IR, NMR and GPC. In vitro released studies showed that the cumulative released optically pure chlorphenesin (ee = 88-92%) from the polymeric prodrug (VADG) was from 19.3% to 34.3% in pH 7.4, pH 5.4 and pH 1.2 after 7 days. The cumulative liberation rate of optically active polymeric prodrugs (ee = ∼88%) with different glycolipids was from 26.4% to 38.5% in pH 1.2.  相似文献

综述了扁桃酸拆分和手性扁桃酸合成的研究进展,对非对映体盐结晶法、萃取法、色谱法、电泳法、酶催化法拆分消旋扁桃酸的原理、工艺和特点进行了评述,并介绍了生物转化法、电化学法和直接化学法合成手性扁桃酸的研究状况。其中非对映体盐结晶法是目前工业生产手性扁桃酸的重要方法,而生物转化法合成手性扁桃酸具有良好的工业应用前景。  相似文献

This review provides a critical analysis of catalytic kinetic resolution reactions from a practical perspective, asking the question of when, if ever, is kinetic resolution the best option for the synthesis of an optically active target. A series of crucial conditions are identified, and it is postulated that if all of them are met, then indeed kinetic resolution can be highly practical. A variety of catalytic kinetic resolution processes are evaluated in the context of these criteria, with particular emphasis on catalyst availability, substrate scope, availability of the racemic substrate and of alternative methods for accessing enantiopure substrate or product, and key experimental considerations. It is found that several catalytic systems have been developed that offer almost unbeatable methods for the preparation of useful chiral building blocks.  相似文献

Enantiomerically pure 4‐(dimethylamino)‐3‐(1‐hydroxyalkyl)pyridines and 4‐(dimethylamino)‐3‐[hydroxy(phenyl)methyl]pyridine have been prepared through efficient chemoenzymatic routes. For this purpose different lipases and oxidoreductases have been tested in the preparation of optically active 4‐chloro derivatives and baker’s yeast was found to be an excellent catalyst for the bioreductions of the corresponding ketones. Their applications as enantioselective nucleophilic catalysts have been studied, important catalytic properties were observed in the stereoselective construction of quaternary centers.  相似文献

Novel optically active methacrylates polymer with controlled molecular weight have been prepared simply by opening ring reaction of poly(glycidyl methacrylate) and S-(−)-α-methylbenzylamine. The structure of the novel optically active methacrylates polymer was characterized by FT-IR, and chiroptical properties of the polymer were characterized by polarimeter and circular dichroism spectrum. Thermogravimetry analysis and differential scanning calorimetry were employed to examine the thermal properties of the polymer. This preparation process was simple and the racemization of optically active monomers in preparation can be avoided. The molecular weight of optically active methacrylates polymer could be controlled by molecular weight of poly(glycidyl methacrylate) prepared by atom transfer radical polymerization.  相似文献
