Singlet oxygen, as the main member of reactive oxygen species, plays a significant role in cancer photodynamic therapy. However, the in vivo real‐time detection of singlet oxygen remains challenging. In this work, a Förster resonance energy transfer (FRET)‐based upconversion nanoplatform for monitoring the singlet oxygen in living systems is developed, with the ability to evaluate the in vivo dose–effect relationship between singlet oxygen and photodynamic therapy (PDT) efficacy. In details, this nanoplatform is composed of core–shell upconversion nanoparticles (UCNPs), photosensitizer MC540, NIR dye IR‐820, and poly(acryl amine) PAA‐octylamine, where the UCNPs serve as an energy donor while IR‐820 serves as an energy acceptor. The nanoparticles are found to sensitively reflect the singlet oxygen levels generated in the tumor tissues during PDT, by luminescence intensity changes of UNCPs at 800 nm emission. Furthermore, it could also enable tumor treatment with satisfactory biocompatibility. To the best knowledge, this is the first report of a theranostic nanoplatform with the ability to formulate the in vivo dose–effect relationship between singlet oxygen and PDT efficacy and to achieve tumor treatment at the same time. This work might also provide an executable strategy to evaluate photodynamic therapeutic efficacy based on singlet oxygen pathway.  相似文献

Combining high‐frequency alternating magnetic fields (AMF) and magnetic nanoparticles (MNPs) is an efficient way to induce biological responses through several approaches: magnetic hyperthermia, drug release, controls of gene expression and neurons, or activation of chemical reactions. So far, these experiments cannot be analyzed in real‐time during the AMF application. A miniaturized electromagnet fitting under a confocal microscope is built, which produces an AMF of frequency and amplitude similar to the ones used in magnetic hyperthermia. AMF application induces massive damages to tumoral cells having incorporated nanoparticles into their lysosomes without affecting the others. Using this setup, real‐time analyses of molecular events occurring during AMF application are performed. Lysosome membrane permeabilization and reactive oxygen species production are detected after only 30 min of AMF application, demonstrating they occur at an early stage in the cascade of events leading eventually to cell death. Additionally, lysosomes self‐assembling into needle‐shaped organization under the influence of AMF is observed in real‐time. This experimental approach will permit to get a deeper insight into the physical, molecular, and biological process occurring in several innovative techniques used in nanomedecine based on the combined use of MNPs and high‐frequency magnetic fields.  相似文献

Stem cells have attracted increasing research interest in the field of regenerative medicine because of their unique ability to differentiate into multiple cell lineages. However, controlling stem cell differentiation efficiently and improving the current destructive characterization methods for monitoring stem cell differentiation are the critical issues. To this end, multifunctional graphene–gold (Au) hybrid nanoelectrode arrays (NEAs) to: (i) investigate the effects of combinatorial physicochemical cues on stem cell differentiation, (ii) enhance stem cell differentiation efficiency through biophysical cues, and (iii) characterize stem cell differentiation in a nondestructive real‐time manner are developed. Through the synergistic effects of physiochemical properties of graphene and biophysical cues from nanoarrays, the graphene‐Au hybrid NEAs facilitate highly enhanced cell adhesion and spreading behaviors. In addition, by varying the dimensions of the graphene‐Au hybrid NEAs, improved stem cell differentiation efficiency, resulting from the increased focal adhesion signal, is shown. Furthermore, graphene‐Au hybrid NEAs are utilized to monitor osteogenic differentiation of stem cells electrochemically in a nondestructive real‐time manner. Collectively, it is believed the unique multifunctional graphene‐Au hybrid NEAs can significantly advance stem‐cell‐based biomedical applications.  相似文献

Obtaining bifunctional electrocatalysts with high activity for the oxygen evolution reaction (OER) and oxygen reduction reaction (ORR) is a main hurdle in the application of rechargeable metal‐air batteries. Earth‐abundant 3d transition metal‐based catalysts have been developed for the OER and ORR; however, most of these are based on oxides, whose insulating nature strongly restricts their catalytic performance. This study describes a metallic Ni‐Fe nitride/nitrogen‐doped graphene hybrid in which 2D Ni‐Fe nitride nanoplates are strongly coupled with the graphene support. Electronic structure of the Ni‐Fe nitride is changed by hybridizing with the nitrogen‐doped graphene. The unique heterostructure of this hybrid catalyst results in very high OER activity with the lowest onset overpotential (150 mV) reported, and good ORR activity comparable to that for commercial Pt/C. The high activity and durability of this bifunctional catalyst are also confirmed in rechargeable zinc‐air batteries that are stable for 180 cycles with an overall overpotential of only 0.77 V at 10 mA?2.  相似文献

Oxygen inhibition remains a challenge in photo‐curing technology despite the expenditure of considerable effort in developing a convenient, efficient, and low‐cost prevention method. Here, a novel strategy to prevent oxygen inhibition is presented; it is based on the self‐assembly of multifunctional nano‐photo‐initiators (F2‐POSS‐(SH)4‐TX/EDB) at the interface of air and the liquid monomer. These nano‐photo‐initiators consist of a thiol‐containing polyhedral oligomeric silsesquioxane (POSS) skeleton onto which fluorocarbon chains and thioxanthone and dimethylaminobenzoate (TX/EDB) photo‐initiator moieties are grafted. Real‐time Fourier‐transform infrared spectroscopy (FT‐IR) is used to investigate the photo‐polymerization of various acrylate monomers that are initiated by F2‐POSS‐(SH)4‐TX/EDB and its model analogues in air and in N2. FT‐IR results show that F2‐POSS‐(SH)4‐TX/EDB decreases the effects of oxygen inhibition. X‐ray photo‐electron spectroscopy and atomic force microscopy reveal that the self‐assembly of F2‐POSS‐(SH)4‐TX/EDB at the air/(liquid monomer) interface forms a cross‐linked top layer via thiol–ene polymerization; this layer acts as a physical barrier against the diffusion of oxygen from the surface into the bulk layer. A mismatch in the shrinkage between the top and bulk layers arise as a result of the different types of photo‐cross‐linking reactions. Subsequently, the surface develops a wrinkled pattern with a low surface energy. This strategy exhibits considerable potential for preventing oxygen inhibition, and the wrinkled pattern may prove very useful in photo‐curing technology.  相似文献

Fluorescence imaging in the spectral region beyond the conventional near‐infrared biological window (700–900 nm) can theoretically afford high resolution and deep tissue penetration. Although some efforts have been devoted to developing a short‐wave infrared (SWIR; 900–1700 nm) imaging modality in the past decade, long‐wavelength biomedical imaging is still suboptimal owing to the unsatisfactory materials properties of SWIR fluorophores. Taking advantage of organic dots based on an aggregation‐induced emission luminogen (AIEgen), herein microscopic vasculature imaging of brain and tumor is reported in living mice in the SWIR spectral region. The long‐wavelength emission of AIE dots with certain brightness facilitates resolving brain capillaries with high spatial resolution (≈3 µm) and deep penetration (800 µm). Owning to the deep penetration depth and real‐time imaging capability, in vivo SWIR microscopic angiography exhibits superior resolution in monitoring blood–brain barrier damage in mouse brain, and visualizing enhanced permeability and retention effect in tumor sites. Furthermore, the AIE dots show good biocompatibility, and no noticeable abnormalities, inflammations or lesions are observed in the main organs of the mice. This work will inspire new insights on development of advanced SWIR techniques for biomedical imaging.  相似文献

Sluggish kinetics of the multielectron transfer process is still a bottleneck for efficient oxygen evolution reaction (OER) activity, and the reduction of reaction overpotential is crucial to boost reaction kinetics. Herein, a correlation between the OER overpotential and the cobalt‐based electrode composition in a “Microparticles‐in‐Spider Web” (MSW) superstructure electrode is revealed. The overpotential is dramatically decreased first and then slightly increased with the continuous increase ratio of Co/Co3O4 in the cobalt‐based composite electrode, corresponding to the dynamic change of electrochemically active surface area and charge‐transfer resistance with the electrode composition. As a proof‐of‐concept, the optimized electrode displays a low overpotential of 260 mV at 10.0 mA cm?2 in alkaline conditions with a long‐time stability. This electrochemical performance is comparable and even superior to the most currently reported Co‐based OER electrocatalysts. The remarkable electrocatalytic activity is attributed to the optimization of the electrochemically active sites and electron transfer in the MSW superstructure. Theoretical calculations identify that the metallic Co and Co3O4 surface catalytic sites play a vital role in improving electron transport and reaction Gibbs free energies for reducing overpotential, respectively. A general way of boosting OER kinetics via optimizing the electrode configurations to mitigate reaction overpotential is offered in this study.  相似文献

Metal halide perovskite solar cells (PSCs) have advanced to the forefront of solution‐processed photovoltaic techniques and made stunning progress in power conversion efficiency (PCE). Further improvements in device performances rely on perfecting the structure and morphology of perovskite films. However, undesirable defects such as pinholes and grain boundaries are often created in film preparations due to lack of knowledge of the precise reaction mechanism. Here, in situ grazing‐incidence X‐ray diffraction (GI‐XRD) investigations are performed, facilitated by other techniques, on the formation of the widely adopted MAPbI3 (MA = methylammonium) perovskite films from their intermediate adduct (IA) phases. The influences of solvent vapor atmospheres on MAPbI3 films are also systematically investigated, where the dynamic conversion processes between different phases are visualized in real time. Further in situ GI‐XRD and infrared spectroscopy measurements reveal that the IA phases contain both N,N‐dimethylformamide and dimethyl sulfoxide (DMSO) as coordinating molecules. By tuning the DMSO concentration in perovskite precursors, the ideal perovskite film is formed and the best PCE is achieved for the planar MAPbI3‐based PSCs. These findings highlight the role of IA phases and the effect of solvent atmospheres on the quality of perovskite films, providing direct insights into their growth mechanism.  相似文献

Inflammation is a common cause of many acute and chronic inflammatory diseases. A major limitation of existing anti‐inflammatory therapeutics is that they cannot simultaneously regulate pro‐inflammatory cytokine production, oxidative stress, and recruitment of neutrophils and macrophages. To overcome this limitation, nanoparticles (NPs) with multiple pharmacological activities are synthesized, using a chemically modified cyclic oligosaccharide. The manufacture of this type of bioactive, saccharide material‐based NPs (defined as LCD NP) is straightforward, cost‐effective, and scalable. Functionally, LCD NP effectively inhibits inflammatory response, oxidative stress, and cell migration for both neutrophils and macrophages, two major players of inflammation. Therapeutically, LCD NP shows desirable efficacies for the treatment of acute and chronic inflammatory diseases in mouse models of peritonitis, acute lung injury, and atherosclerosis. Mechanistically, the therapeutic benefits of LCD NP are achieved by inhibiting neutrophil‐mediated inflammatory macrophage recruitment and by preventing subsequent pro‐inflammatory events. In addition, LCD NP shows good safety profile in a mouse model. Thus, LCD NP can serve as an effective anti‐inflammatory nanotherapy for the treatment of inflammatory diseases mainly associated with neutrophil and macrophage infiltration.  相似文献

Oxygen vacancies play crucial roles in defining physical and chemical properties of materials to enhance the performances in electronics, solar cells, catalysis, sensors, and energy conversion and storage. Conventional approaches to incorporate oxygen defects mainly rely on reducing the oxygen partial pressure for the removal of product to change the equilibrium position. However, directly affecting reactants to shift the reaction toward generating oxygen vacancies is lacking and to fill this blank in synthetic methodology is very challenging. Here, a strategy is demonstrated to create oxygen vacancies through making the reaction energetically more favorable via applying interfacial strain on reactants by coating, using TiO2(B) as a model system. Geometrical phase analysis and density functional theory simulations verify that the formation energy of oxygen vacancies is largely decreased under external strain. Benefiting from these, the obtained oxygen‐deficient TiO2(B) exhibits impressively high level of capacitive charge storage, e.g., ≈53% at 0.5 mV s?1, far surpassing the ≈31% of the unmodified counterpart. Meanwhile, the modified electrode shows significantly enhanced rate capability delivering a capacity of 112 mAh g?1 at 20 C (≈6.7 A g?1), ≈30% higher than air‐annealed TiO2 and comparable to vacuum‐calcined TiO2. This work heralds a new paradigm of mechanical manipulation of materials through interfacial control for rational defect engineering.  相似文献

Development of alternative energy sources is crucial to tackle challenges encountered by the growing global energy demand. Hydrogen fuel, a promising way to store energy produced from renewable power sources, can be converted into electrical energy at high efficiency via direct electrochemical conversion in fuel cells, releasing water as the sole byproduct. One important drawback to current fuel‐cell technology is the high content of platinum‐group‐metal (PGM) electrocatalysts required to perform the sluggish oxygen reduction reaction (ORR). Addressing this challenge, remarkable progress has been made in the development of low‐cost PGM‐free electrocatalysts synthesized from inexpensive, earth‐abundant, and easily sourced materials such as iron, nitrogen, and carbon (Fe–N–C). PGM‐free Fe–N–C electrocatalysts now exhibit ORR activities approaching that of PGM electrocatalysts but at a fraction of the cost, promising to significantly reduce overall fuel‐cell technology costs. Herein, recent developments in PGM‐free electrocatalysis, demonstrating increased fuel‐cell performance, as well as efforts aimed at understanding the key limiting factor, i.e., the nature of the PGM‐free active site, are summarized. Further improvements will be accomplished through the controlled and/or rationally designed synthesis of materials with higher active‐site densities, while at the same time establishing methods to mitigate catalyst degradation.  相似文献
