Skin Compatibility of External Products Beside dermatotherapeutics, cosmetics and skin cleaners a great amount of further products gets more or less intensely into contact with the human skin. To give an example domestic chemicals and many professional substances can be mentioned. As always new substances are developed or new compositions are applied to produce such products, tests of skin compatibility at a large scale are necessary, since it is required by the law of food and requisites that there is no objection with respect to health and because the chemical law, too, includes a consideration of health criteria. The great number of new substances and newly developed finished products requires a most rational use of test capacities which are not at all inexhaustible not only because of the law for prevention of the cruelty to animals but also because of the limited number of suitable test persons and experienced scientists. Therefore the procedure for testing skin compatibility of new substances or substance combinations is discussed basing on an algorithmic presentation. Way and extent of the expected contact with the skin is a significantly more important selection criterion than the future application of the product which is to be judged.  相似文献

Bacterial infections of skin and soft tissue represent a major health threat, especially if they are caused by multidrug-resistant strains such as MRSA. Novel treatment options for topical application are urgently needed, and even if new drug candidates are identified, their properties must match the specific physical requirements of the skin in order to penetrate and reach the site of bacterial infection. β-Lactones have been shown to eliminate bacterial virulence, but knowledge about their potential in topical treatments had thus far been lacking. Herein we present the synthesis and in-depth investigations of skin permeation and skin distribution of a radioactive (14) C-labeled tool compound that was formulated with various basic ointments and applied to pig skin samples. Our results show that skin penetration depends highly on the nature of the formulation base. Vaseline is the most efficient ointment and is best suited for effecting maximal drug delivery within and through the stratum corneum, an important entrance barrier, and meets the required quantities for eliciting anti-virulence effects. We are confident that the nature of these new β-lactone virulence inhibitors confers applicability and potency for topical treatment, and will translate into a new formulation of this highly potent drug candidate for the treatment of skin and soft tissue infections.  相似文献

To simulate temperature rise of human skin and to predict burn injury during radiant heat exposure, the traditional method is to use a sensor to simulate skin surface and use a numerical model to simulate heat transfer in inner skin. However, the numerical models of skin burns are based on few experimental data of nude skin and some simplifications of human skin characteristics. In this study, a new multi‐layered skin simulant is presented for low radiant heat exposures up to 15 kW/m2. The skin simulant has implanted thermocouples into layered polydimethylsiloxane (PDMS) materials with controlled thermal properties and thicknesses of the skin layers. The multi‐layered skin simulant developed in this study has a good reproducibility for temperature measurements with similar temperature rise profiles compared with human skin except at skin surface. For burn injury prediction, the results of our PDMS skin model can be linearly corrected using ASTM model as a reference. Our developed skin simulant provides an advanced method to directly simulate the heat transfer inside human skin in a multi‐layered structure rather than using the combined physical sensor‐numerical model in the traditional way.  相似文献

Composite membranes with a porous support layer and a dense skin layer have been extensively used in gas separation processes. A new approach, a mesoscale Lattice Boltzmann Simulation approach, is proposed and used to model the pore‐scale gas flow and mass transfer in the inhomogeneous membrane matrixes studied. Only physical forces are considered. Chemical forces are equivalently converted to physical forces through the relaxation time. Selective permeation of moisture through a composite membrane is modeled. The overall permeability is evaluated. It is found that mass transfer inhomogeneity exists not only in the porous media but also in the seemingly uniform dense skin layer. Increasing the diffusivity in the skin layer is more effective than decreasing the skin layer thickness in optimizing the overall membrane performance. The new approach gives more detailed insights into the directions for future design of composite membranes for gas separations like air dehumidification. © 2014 American Institute of Chemical Engineers AIChE J, 60: 3925–3938, 2014  相似文献
