采用γ射线(Co60)及亚硝基胍复合诱变的方法对产柠檬酸黑曲霉菌株进行诱变。γ射线(Co60)诱变剂量为1400Gy,亚硝基胍的浓度为1mg/mL,作用时间为4min,在此条件进行复合诱变。经多轮筛选最终获得到一株产酸为15.5g/dL且遗传稳定性高的菌株。并对黑曲霉发酵产柠檬酸的工艺进行优化。确定种子液的最适条件:氮源选取豆饼粉,糖浓度为10%;最适发酵条件:培养温度为35℃,初始pH值5~6,氮源选取豆饼粉且氮源用量为0.8%,在50L发酵罐中进行中试试验,试验结果为周期63h,产酸17.94g/100mL,转化率为99.7%,比出发菌株发酵周期缩短3h,产酸提高10%,转化率提高5%。 相似文献

为了选育高产柠檬酸菌种,以前期分离到的一株黑曲霉(A spergillusniger)Y Z-35为出发菌株,经紫外线(U V)与硫酸二乙酯(D ES)复合诱变后,进行了产柠檬酸发酵条件优化试验。结果表明,选育出黑曲霉U V -D E-3是一株优良高产菌株,该菌株的产酸率达到11.54%,比初始菌株(Y Z-35)产酸率提高了69.70%;以红薯为发酵原料,在初始糖含量12%、温度35℃、起始pH 值为6.5,转速160 r/m in、发酵时间72 h的优化条件下,摇瓶发酵平均产酸率达到14.24%。 相似文献

选择计量罐式连续流加补料系统,采用新选育的黑曲霉(Aspergillus niger)Wml-016菌株在50 L自动发酵罐上研究间歇补料对发酵的影响。利用中心复合设计和响应面分析方法,实验得出木薯原料柠檬酸补料发酵的优化条件:发酵到第36 h开始进行4次流加补料,间隔时间为1.5 h,补入DE值为80的木薯水解液和质量浓度为0.04%的(NH4)2SO4。采用上述工艺发酵产酸率达到18.5%,残糖为0.2%,周期仅为64 h,结果明显优于分批发酵。 相似文献

A new strain of Aspergillus niger isolated from soil and its mutant were used for citric acid production from carob under solid-state fermentation conditions. The parental strain produced 30 g/kg citric acid, while the mutant G4, selected after four rounds of gamma ray irradiation, produced 60 g/kg. Maximum citric acid production was obtained after 7 days of incubation, as the acid production was 34 and 64 g/kg for parental and mutant strains, respectively. The addition of 2% methanol increased citric acid production from the parental strain to 42 and the mutant G4 to 65 g/kg. Trace elements, namely Cu, Fe, and Zn, promoted the production of citric acid as the acid production from the parental strain increased to 46 g/kg and for mutant G4 increased to 73 g/kg after their addition. The optimum spore inoculum concentration for acid production was 107 ml?1, and the optimum pH was 5 for both parental and mutant strains. 相似文献

研究了SHAM (水杨酸氧肟酸 )对产酸的影响。 5mg/ 6 0mL的SHAM基本抑制柠檬酸的产生。当W 3菌株在加有 10mg/ 10mLSHAM(L1)的平板上生长时 ,只能着生 10 %的孢子 ,在加有 2 0mg/ 10mLSHAM (L2 )的平板上生长时不能着生孢子。据此选育出不能够在L1平板上着生孢子的SHAM敏感株和在L2平板上能够着生孢子的SHAM抗性株。通过发酵试验 ,SHAMS- 2菌株产柠檬酸比出发菌株W 3高 4 0 % ,转化率达到 91 7% 相似文献

本文提出了一种利用离子交换树脂萃取发酵法生产柠檬酸的新方法。该方法将离子交换树脂填充柱与发酵反应器相连接,发酵一定时间后,发酵液流经离子交换柱,分离出产物柠檬酸,再返回发酵反应器循环使用。与传统的发酵方法相比,萃取发酵使发酵周期由9天缩短到6天,糖转化率由原来的88.4%提高到96.6%,柠檬酸生产速率由0.447g/I·h提高到0.753g/l·h。 相似文献
