Sorption of iodine by polyurethane and melamine-formaldehyde foams using iodine sublimation and iodine solutions

Iodine sorption by polyurethane (PU) and melamine-formaldehyde (MF) foams was studied using both iodine sublimation and iodine solutions with hexanes and toluene. In the sublimation process, the diffusion kinetics was investigated and the interaction between iodine and PU foams was characterized by DSC, TGA, Raman spectroscopy and electrical conductivity measurements. In the solution process, the equilibrium absorption followed the distribution law and the distribution coefficients varied depending on the solvent used. MF foam achieved no iodine absorption in both processes which can be attributed to the lack of charge-transfer interactions.

多价EZH2肿瘤相关抗原肽的制备及其抗原性

目的人工设计制备多价EZH2肿瘤相关抗原肽,并鉴定其抗原特性。方法设计合成编码多价EZH2肿瘤相关抗原肽的DNA序列,克隆入原核表达载体p ET-30a(+)中,构建含4个EZH2肿瘤相关抗原肽段(Aa120-128、Aa165-174、Aa291-299、Aa735-742)的重组多价抗原肽原核表达质粒p ET-30a(+)-EZH2-antigen,转化E.coli BL21(DE3),经IPTG诱导表达后,用Ni-NTA树脂在变性条件下对重组多肽进行亲和层析纯化,Western blot分析重组多价EZH2肿瘤相关抗原肽的抗原特性。用重组多价EZH2肿瘤相关抗原肽免疫大耳白兔,制备多克隆抗体,间接ELISA法检测血清中抗体效价,Western bolt分析多抗的特异性。结果重组原核表达质粒p ET-30a(+)-EZH2-antigen经酶切及测序证实构建正确;表达的重组多价EZH2肿瘤相关抗原肽表观相对分子质量约4 300,主要以包涵体形式存在;纯化的多肽纯度不低于90%,浓度为0.5 mg/ml,且能被兔抗EZH2多抗和兔抗His标签多抗有效识别和结合;纯化的多价EZH2肿瘤相关抗原肽能在动物体内高效诱导抗体生成,抗血清效价为1∶64×104,且能特异性识别和结合PC3人前列腺癌细胞中表达的EZH2蛋白。结论成功制备了一种多价EZH2肿瘤相关抗原肽,该多价抗原肽相对分子质量虽小,但抗原肽密度高,可能具有更强的肿瘤相关抗原的活性,具有重要的应用前景。

Anti-carbohydrate antibodies elicited by polyvalent display on a viral scaffold

Tetra- and hexasaccharides were arrayed on the exterior surface of cowpea mosaic virus by using a copper-catalyzed azide-alkyne cycloaddition reaction. Inoculation of chickens with these virus conjugates gave rise to large quantities of polyclonal anti-glycan IgY antibodies that displayed excellent avidity and specificity on analysis with printed glycan microarrays. Avian IgY antibodies are produced in significantly higher yield than is possible for mouse or rabbit IgG, and exhibit reduced cross reactivity with native mammalian proteins. For a tri-LacNAc antigen, affinity purification against immobilized mono-LacNAc was necessary to provide a set of antibodies with specific binding properties. Comparable performance was observed for the virus-based polyclonal versus a commercial monoclonal antibody raised against the globo-H tetrasaccharide; this highlights the utility of the glycan microarray for both quality control and rapid in-depth analysis. Virus-carbohydrate conjugates are promising candidates for development in diagnostic and immunotherapeutic applications.     

湿法磷酸中碘含量的测定

采用碘离子选择电极法分析测定湿法磷酸中碘含量。测定前需将碘用N a2SO3还原为I形式。该方法操作简便,结果准确,加标回收率为99.5%~101.8%。

近年来我国多地连续出现大面积的雾霾天气,迫使人们去思考如何从源头上去防治环境污染。绿色化学是指化学反应以"原子经济性"为基本原则,过程和终端均为零排放和零污染,是一门从源头阻止污染的化学。为此,我们尝试在大学化学教学中渗透绿色化学理念,对教材进行优化重组、拓展,从理论教学和实验教学入手,把绿色化学理念融入到具体教学内容中,使学生在潜移默化中受到绿色化学教育,增加学生的环保意识,效果颇佳。     

Anomalous high iodine value of squalene and the impact on iodine values of shark liver oils

Squalene has six ethylenic bonds, but the experimental iodine values in two different solvent systems—chloroform and cyclohexane/acetic acid—were 25% higher than the theoretical values. We propose that this results from an additional halogen adding at each of the two terminal ethylenic bonds carrying two methyl groups. In the solvent system of cyclohexane alone, the excess is only 3–4% greater than the theoretical. Mixtures of squalene in seal oil confirmed the additivity of the experimental squalene high iodine value and the seal oil fatty acid iodine value with reasonable accuracy but depended on the skill of the operator in obtaining the titration end point for cyclohexane/acetic acid. This observation has particular relevance for shark liver oils and olive oils.
