This paper describes the isolation of Enterococcus host strains, for potential use as simple bacteriophage (phage)-based microbial source tracking (MST) tools. Presumptive Enterococcus host strains were isolated from cattle feces, raw municipal wastewater, agricultural runoff, and waters impacted by farms or wastewater treatment works (WWTW) in southern England, United Kingdom (UK). All enterococcal host strains (n = 390) were first screened for their ability to detect phage in samples of raw municipal wastewater and fecal material from cattle, pigs, and sheep. Host strains that detected phage (n = 147) were ranked according to both their specificity to a particular fecal source and also the number of phages (expressed as plaque-forming units, PFU) that they detected per milliliter of sample. Host strains that demonstrated host specificity and which detected phages at levels greater than 100 PFU/mL (n = 29) were further tested using additional fecal samples of human and nonhuman origin. The specificity and sensitivity of the enterococcal host strains were found to vary, ranging from 44 to 100% and from 17 to 83%, respectively. Most notably, seven strains exhibited 100% specificity to either cattle, human, or pig samples. Isolates exhibiting specificity to cattle were identified as belonging to the species Enterococcus casseliflavus , Enterococcus mundtii , or Enterococcus gallinarum , while human and pig isolates were members of either Enterococcus faecium or Enterococcus faecalis . The high specificity of phages infecting Enterococcus hosts and the simplicity and relatively low cost of the approach collectively indicate a strong potential for using this method as a tool in MST.  相似文献

Differential scanning calorimetry (DSC) data are increasingly used to substantiate product claims of hair repair. Decreasing peak temperatures may indicate structural changes and chemical damage. Increasing the DSC, wet peak temperature is, therefore, often considered as proof of hair repair. A detailed understanding of the technique and hair structure indicates that this may not be a sound approach. Surveying the rich literature on the use of dynamic thermal analysis (DTA) and differential scanning calorimetry (DSC) for the analyses of human hair and the effect of cosmetic treatments, we underline some of the problems of hair structure and data interpretation. To overcome some of the difficulties of data interpretation, we advise that DSC acquired data should be supported by other techniques when used for claim substantiation. In this way, one can provide meaningful interpretation of the hair science and robust data for product claims support.  相似文献

Current fecal indicator bacteria (FIB) and emerging microbial source tracking (MST) methods may indicate the presence and even the likely source of water contamination, but they are less effective at determining the potential risk to health from human enteric viruses. This paper investigates the presence of human-specific phages (detected using a low-cost MST method) in municipal wastewaters (MW) and assesses whether they may be used effectively to screen for the likely presence of human adenovirus (HAdV) and norovirus (NoV). The findings demonstrated that all samples positive for HAdV and/or NoV also contained phages infecting Bacteroides GB-124 (mean = 4.36 log(10) PFU/100 mL) and that GB-124 phages, HAdV, and NoV were absent from samples of nonhuman origin. HAdV and NoV were detected more frequently in MW samples containing higher levels of phages (e.g., >10(2)) and FIB (e.g., >10(3)). Interestingly, at one sewage treatment works (STW), the levels of GB-124 phages present in treated MW were not significantly lower (p = 0.001) than those in untreated MW. There was a positive correlation (R = 0.42) between the size of STW and the number of GB-124 phages present in the final treated effluent. Therefore, the detection of GB-124 phages by a simple phage-lysis method may have considerable potential as a low-cost surrogate for the detection of certain human pathogenic viruses in MW and receiving waters.  相似文献

In this regard, irradiation is an interesting alternative to be considered to satisfy quarantine requirements, to control severe losses during transportation and commercialization, and to insure food safety. Bamboo shoot processing produces substantial quantities of wastes in China and other parts of the world, with estimates of up to 70% of the harvested bamboo shoots being discarded as waste biomass. The solid waste residues that are generated as processing by-products consist of the outer sheath and basal section of the bamboo shoot, and wastewater is also produced, all of which pose a significant environmental burden. In comparison to the edible bamboo shoot tips, data on the composition of bamboo shoot processing residue (BSPR) are comparatively scarce. The tender shoots are rich in nutrients and bioactive compounds that are associated with health benefits against many chronic and degenerative diseases, and there is potential for these compounds to also be present in BSPR. Therefore, this paper reviews the current literature available on BSPR composition from both English and Chinese journals, for comparison with the tender bamboo shoot. From this information, the potential of using BSPR as a cheap source of valuable nutrients and bioactive compounds for value-added products (e.g., food additives, functional foods, and pharmaceuticals) is discussed. To maximize the value from BSPR and reduce its environmental impact as a waste, a systematic biorefinery approach for waste valorization is recommended.  相似文献

Accurate cow-side blood β-hydroxybutyrate (BHB) detection meters are valuable tools for rapid diagnosis of hyperketonemia. The main objective of this study was to compare the blood BHB measured in whole blood by the BHBCheck meter (PortaCheck, Moorestown, NJ) to a previously validated meter, Precision Xtra meter (Abbott Laboratories, Abbott Park, IL) and a colorimetric laboratory assay. Samples (n = 426) were collected from postpartum primiparous and multiparous Holstein cows (n = 79 cows) enrolled in 1 of 2 experiments (Exp) with different sampling schedules (Exp 1: n = 39 cows, 58 samples; Exp 2: n = 40 cows, 368 samples). In both Exp, whole-blood samples were collected from the coccygeal vessels after morning milking, before morning feeding. Blood samples were used immediately for BHB quantification via the BHBCheck meter and the Precision Xtra meter. Blood was also collected into evacuated tubes containing no additive (Exp 1) or potassium oxalate/sodium fluoride (Exp 2), which were centrifuged for serum or plasma separation and stored at ?20°C for subsequent analysis. Laboratory quantification of BHB concentration was done by the BHB LiquiColor Assay (EKF Diagnostics-Stanbio, Boerne, TX; certified for serum and plasma). Data were analyzed by UNIVARIATE, CORR, FREQ, REG, and LOGISTIC procedures of SAS 9.4 (SAS Institute Inc., Cary, NC). Within this sample set, average parity was 3.3 lactations and DIM was 14 d. The proportion of samples classified as hyperketonemia (BHB ≥1.2 mmol/L) was 25, 28, and 31% as determined by the colorimetric assay, BHBCheck meter, and Precision Xtra meter, respectively. The correlation for BHBCheck meter BHB concentration compared with the colorimetric assay concentrations was r = 0.96, with a sensitivity of 91% and specificity of 93%. Correlation, sensitivity, and specificity of the Precision Xtra meter concentrations were 0.97, 98%, and 92%, respectively. Bland-Altman plots demonstrated minimal bias for both meters. Area under the receiver operator characteristic curve suggests adequate diagnostic accuracy of both meters. Overall, accuracy, sensitivity, and specificity of the BHBCheck meter was similar to the Precision Xtra meter and laboratory assay, indicating the BHBCheck meter is appropriate for use as a cow-side diagnostic test for hyperketonemia in dairy cows.  相似文献

The phytohormone ethylene exerts numerous beneficial and detrimental effects on postharvest quality and storage life of fruit and vegetables. In view of the current global challenge of reducing postharvest losses and waste of fruit and vegetables, the importance of ethylene management in the supply chain is paramount. For this purpose, various methods have been applied along the supply chain over the years; however, effective management of ethylene under real-time storage and transport conditions still remains a challenging task. This review explores the potential of photocatalytic and photochemical oxidation of ethylene for effective removal of this gas along the value chain of fruit and vegetables. These techniques involve the use of ultraviolet (UV) radiation with or without a catalyst. In photocatalytic oxidation, a semiconductor such as titanium dioxide is essential which acts as a photocatalyst on irradiation with UV light and thus facilitates the oxidation of ethylene at its surface, whereas in photochemical oxidation, extreme short wave (i.e. below 200 nm) vacuum ultraviolet radiation (VUV) consisting of high-energy photons eliminates ethylene in the gaseous state. This review gives a brief overview of current commercial techniques used in ethylene removal and then focuses on the photocatalytic and the photochemical oxidation of ethylene and the combination of both methods. The various factors affecting these processes are also discussed including the advantages and the drawbacks associated with them, and current applications of these methods in fruit and vegetable storage systems are highlighted. In addition, a future outlook on the application of these methods in postharvest storage of fresh produce is given.  相似文献

Production Yield Analysis (PYA) is a structured system approach to optimize the production yield of production processes. The paper outlines the developed method and the 10 basic steps of the PYA. The PYA-method makes it possible to calculate the Yield Index of a process. Thermal treatment is a traditional method for food processing, which can kill microorganisms but also lead to physicochemical and sensory quality damage, especially to temperature-sensitive foods. Nowadays consumers’ increasing interest in microbial safety products with premium appearance, flavor, great nutritional value and extended shelf-life has promoted the development of emerging non-thermal food processing technologies as alternative or substitution to traditional thermal methods. Fish is an important and world-favored food but has a short shelf-life due to its extremely perishable characteristic, and the microbial spoilage and oxidative process happen rapidly just from the moment of capture, making it dependent heavily on post-harvest preservation. The applications of novel non-thermal food processing technologies, including high pressure processing (HPP), ultrasound (US), pulsed electric fields (PEF), pulsed light (PL), cold plasma (CP) and ozone can extend the shelf-life by microbial inactivation and also keep good sensory quality attributes of fish, which is of high interest for the fish industry. This review presents the principles, developments of emerging non-thermal food processing technologies, and also their applications in fish industry, with the main focus on microbial inactivation and sensory quality. The promising results showed great potential to keep microbial safety while maintaining organoleptic attributes of fish products. What’s more, the strengths and weaknesses of these technologies are also discussed. The combination of different food processing technologies or with advanced packaging methods can improve antimicrobial efficacy while not significantly affect other quality properties under optimized treatment.  相似文献

To assess the economic importance of breeding traits, economic values (EV) were derived for 3 German dairy cattle breeds: German Holstein (HOL), Angler (ANG), and Red and White Dual-Purpose (RDN). For that purpose, the stochastic bio-economic model SimHerd (SimHerd A/S, Viborg, Denmark) was used, which simulates the expected monetary gain in dairy herds. The EV was calculated as the alteration in average net return of the herd responding to a marginal change in the trait of interest. When deriving EV using SimHerd, economic consequences resulting from changes in the age structure of a dairy herd (i.e., structural herd effects) are considered. However, this requires the simulation of relationships between traits in the bio-economic model. To avoid double counting, the EV of a trait was corrected for effects from alterations in correlated traits using multiple regression analysis. The EV were derived for 23 traits in terms of production, conformation and workability, dairy health, calf survival, and reproduction performance. Furthermore, the relative economic importance of the breeding traits was calculated. Relative emphasis on production was between 39.9 and 44.4% in the breeds studied. Total costs per case of ketosis and metritis ranged from €167 to €196 and €173 to €182, respectively. Highest marginal EV of direct health traits were found for mastitis (€257 to €271 per case) and lameness (€270 to €310 per case). Consequently, relative emphasis on direct health traits was between 15.7 and 17.9%. The EV of reproduction performance showed largest differences among the cattle breeds. Overall relative emphasis on reproduction was 10.5% in HOL, 10.8% in ANG, and 6.5% in RDN. The relative economic importance of cow mortality ranged from 15.5 to 16.0% across the breeds. Collectively, the study showed the high economic importance of functional traits in the cattle breeds studied.  相似文献

This work deals with the investigation of β-lactoglobulin (β-LG) aggregation by granulomorphometry. In the first part of this study, we showed that the binding interactions involved in aggregate structure could be identified by their appearance in granulomorphometric pictures. The reliability of this analytical approach was demonstrated by comparing the appearance of β-LG aggregates in the presence and absence of a thiol-blocking agent (N-ethylmaleimide). The translucency of the aggregates was associated with hydrophobic interactions and their opacity was associated with disulfide bonds. We state, based on the morphology of the aggregates, along with the color of protein aggregates and insoluble materials, that hydrophobic interactions had a better water-holding capacity than disulfide bonds. Additionally, our results suggest that disulfide and hydrophobic bonds compete for β-LG aggregate shaping. In the second part of this work, interesting features of granulomorphometry useful for identifying aggregate binding interactions were highlighted to clarify the effect of temperature on the aggregation mechanisms occurring in a β-LG concentrate with a moderate calcium content (6.6mmol·L(-1)). Heat treatment experiments were performed between 70 and 95°C, and granulomorphometric measurements (aggregate size, aggregate number, and gray level of the picture) were conducted at different sampling times up to 4h. Results, which were interpreted in light of calculated β-LG denaturation levels, revealed that the aggregation mechanism could be split into 2 steps. Initially, β-LG denatured quickly, leading to fast β-LG aggregation by disulfide bonds. The denaturation rate then declined, which drastically slowed the disulfide aggregation mechanism. From that point on, a second aggregation path became preponderant. It consisted of the agglomeration of small aggregates by hydrophobic interactions and resulted in the formation of large aggregates containing both interaction types. This second aggregation mechanism was clearly favored at high temperatures because it was not detected in our experiments at temperatures below 85°C.  相似文献

Children with phenylketonuria (PKU) and severe cow’s milk protein allergy (CMPA) consume prescribed, specially formulated, foods for special medical purposes (FSMPs) in addition to having restricted intake of normal foods. These vulnerable patients are exposed to artificial sweeteners from the consumption of a combination of both free and prescribed foods. Young patients with PKU and CMPA aged from 1 to 3 years have a higher risk of exceeding the acceptable daily intake (ADI) for sweeteners than age-matched healthy children. A probabilistic modelling approach has been adapted successfully to assess the exposure of young patients with PKU and CMPA to low-calorie sweeteners. To assist professionals in the screening and formulation of foods containing food additives for such patients, a simplified exposure method/tool has been developed. The tool is intended to ensure that total dietary exposure can be considered. The simplified tool is not intended to replace the probabilistic model but may be used as a screening tool to determine if further investigation on exposure is warranted. The aim of this study was to develop and validate this simplified exposure tool to support those currently used by healthcare professionals (HCPs) using data available from the probabilistic modelling of exposure in young children with PKU and CMPA. The probabilistic model does not allow for swift screening of exposure scenarios nor is the present EFSA Food Additive Intake Assessment Model (FAIM) fully suitable for application to medical foods. The simplified exposure tool in medical nutrition (SETIM) reported here is both reliable and consistent and provides additive usage levels which minimise regular exposure above the ADI in patients. In addition to the usefulness of SETIM for the medical nutrition industry, the tool has the potential to enhance the practice of evidence-based medical nutrition by official risk assessment bodies, registration authorities and healthcare professionals.  相似文献
