This study reports a comparative screening of four species of Origanum in Turkey, based on their essential oil composition, total phenolic content, antioxidant and antibiofilm activities. The major components of essential oils were p-cymene, linalool, and thymol. The total phenolic contents differed from 3.81 to 47.54 mg of GAE/g of extract. The concentrations of flavonoids varied from 12.74 to 58.39 mg of Ru/g of extract. Antioxidant activity was determined in vitro using DPPH reagent and expressed as concentration of each extract required to inhibit radical by 50% (IC50) values that ranged from 16.03 to 48.94 μg/ml. Our results indicated that chloroform extracts of species O. majorana and O. onites, with a total content of polyphenols (47.54 mg of GAE/g and 45.17 mg of GAE/g, respectively) and an IC50 of 16.03 μg/ml and 16.89 μg/ml, respectively were more antioxidant. Among the essential oil concentrations tested, maximum antibiofilm activity was found as 92.24% against M. luteus NRRL-B 1013 by O. majorana essential oil at 50 mg/ml.  相似文献

BACKGROUND: To find natural compounds with insecticidal properties which could be an alternative to synthetic insecticides, oregano essential oil was tested against the insect Rhizopertha dominica devastating stored cereals. RESULTS: Oregano oils were isolated by steam distillation, analysed by gas chromatography and gas chromatography–mass spectrometry and investigated for their insecticidal activities against Rhizopertha dominica. Contact and fumigant toxicities of global oil and three of its fractions recovered respectively after 2 h and after 0–2.5, 2.5–5 and 5–10 min of extraction were tested using the filter paper method. Eighteen components, representing 92.6% of the oil, were identified. The major components were thymol (38.8%), carvacrol (32.9%), p‐cymene (7.9%) and γ‐terpinene (5.1%). Toxicity tests showed that insect mortality increases with the concentration used (0.39%, 0.78%, 1.56%, 3.12% and 6.24%V). For all concentrations, the contact effect of global oil and fraction F2 was better than the fumigant effect. Whereas fractions F1 and F3 were more toxic by fumigation for concentrations lower or equal to 1.56%, their contact effect was more significant for concentrations higher than 1.56%. CONCLUSION: These results suggest that oregano oil has important insecticidal properties and may be useful as a natural grain protectant against Rhizopertha dominica to replace synthetic insecticides. In this study the antioxidant activity of methanolic extracts of oregano and sage samples was tested. Samples of oregano belonged to Origanum onites and O indercedens species, whilst samples of sage belonged to Salvia officinalis and S fruticosa species. Two methods were used to evaluate the antioxidant activity of sage and oregano extracts: the crocin test and the Rancimat test. The methanolic extracts were also analysed by HPLC for the qualitative/quantitative determination of phenolic compounds. The total phenolic compound content of oregano samples showed no significant differences between the two species, but rosmarinic acid was present in higher amount in O indercedens. Carvacrol content sharply differentiated flowers from leaves. Samples of O indercedens had a higher antioxidant activity evaluated by the crocin test, whereas no differences were evidenced by the Rancimat test. For sage samples, carnosic acid and methyl carnosate showed a significant difference between the two species, with S fruticosa samples having a higher content than S officinalis samples. Samples of S fruticosa also had a higher antioxidant activity evaluated by the crocin test. The antioxidant activities of sage samples were, on average, higher than those of oregano samples. Some samples of sage had a very high antioxidant activity, with induction times more than 10‐fold higher than that of lard used as the reference sample. © 2002 Society of Chemical Industry  相似文献

BACKGROUND: Two varieties of Origanum majorana (Canadian and Tunisian) were evaluated for their phenolic, flavonoid and tannin contents, individual phenolic compounds and antioxidant activities under NaCl constraint. RESULTS: The results showed a significant variability in phenolic composition and antioxidant behavior between the two varieties under salt stress. The phenolic composition of methanolic extracts was determined by reversed‐phase high‐performance liquid chromatography. Amentoflavone was the predominant flavonoid compound; in addition, trans‐2‐hydrocinnamic acid became the major phenolic acid with salt treatment of the Tunisian variety. In the control, Canadian variety extract was characterized by high levels of gallic acid and amentoflavone. However, under 75 mmol L?1 NaCl, gallic acid content doubled, whereas amentoflavone content was maintained in the Canadian variety. Stimulation of phenolic acid biosynthesis was observed in these two varieties under salt treatment despite the fact that shoots of the Tunisian variety showed higher antioxidant activities compared to those from the Canadian variety. Tunisian O. majorana might have developed tolerance to salinity and avoided tissue damage by activating enzymes involved in the galactosylation of quercetin into quercetin‐3‐galactoside and quercetin‐3‐rhamnoside. BACKGROUND: Characterisation of the essential oils from O. glandulosum collected in three locations of Tunisia, chemical composition and the evaluation of their antioxidant activities were carried out. RESULTS: The essential oils from Origanum vulgare L. subsp. glandulosum (Desf.) Ietswaart collected from three localities of north Tunisia—Krib, Bargou and Nefza—were obtained in yields of 2.5, 3.0 and 4.6% (v/w), respectively. The essential oils were analysed by GC and GC/MS and assayed for their total phenolics content, by the Folin–Ciocalteu method, and antioxidant effectiveness, using the 2,2‐diphenyl‐1‐picrylhydrazil (DPPH) radical scavenging assay. The main components of these essential oils, from Nefza, Bargou and Krib, were p‐cymene (36%, 40% and 46%), thymol (32%, 39% and 18%), γ‐terpinene (24%, 12% and 16%) and carvacrol (2%, 2% and 15%), respectively). The ability to scavenge the DPPH radicals, expressed by IC50, ranged from 59 to 80 mg L?1. The total phenolic content, expressed in gallic acid equivalent (GAE) g kg?1 dry weight, varied from 9.37 to 17.70 g kg?1 dw. CONCLUSIONS: A correlation was identified between the total phenolic content of the essential oils and DPPH radical scavenger capacity. Consumers’ concerns about the environment and health have led to the development of new food packaging materials avoiding petroleum-based matrices and synthetic additives. The present study has developed polylactic acid (PLA) films containing different concentrations of essential oil from Origanum vulgare L. virens (OEO). The effectiveness of this new active packaging was checked for use in ready-to-eat salads. A plasticising effect was observed when OEO was incorporated in PLA films. The rest of the mechanical and physical properties of developed films did not show much change when OEO was included in the film. An antioxidant effect was recorded only for films containing the highest percentages of the active agent (5% and 10%). In addition, films exhibited in vitro antibacterial activity against Staphylococcus aureus, Yersinia enterocolitica, Listeria monocytogenes, Enterococcus faecalis and Staphylococcus carnosus. Moreover, in ready-to-eat salads, antimicrobial activity was only observed against yeast and moulds, where 5% and 10% of OEO was the most effective.  相似文献

The present work examines the in vitro antioxidant and antimicrobial properties of the essential oil and various extracts from the herbal parts of Origanum syriacum L var bevanii. Polar subfractions of methanol extracts from both deodorised and non‐deodorised materials showed the highest DPPH (2,2‐diphenyl‐l‐picrylhydrazyl) radical‐scavenging activity, with IC50 values of 21.40 and 26.98 µg ml?1 respectively, whereas the IC50 of the essential oil was 134.00 µg ml?1. The antioxidant potential of the extracts appeared to be closely related to the presence of polar phenolics. However, the inhibitive effect on linoleic acid oxidation might be promoted by the presence of non‐polar phenolics, as both hexane and dichloromethane extracts showed high antioxidant activities. The antimicrobial activity of the essential oil was superior to those of the other extracts. Nineteen compounds representing 962 g kg?1 of the essential oil were identified; carvacrol (669 g kg?1) was the main component. Overall, the results suggest that the essential oil and extracts from the herbal parts of O syriacum could be used as natural preservative ingredients in the food industry. BACKGROUND: The effects of salt on the essential oil yield and fatty acid composition of aerial parts of two marjoram varieties were investigated. Plants with 6 leaves were treated with NaCl (75mM). RESULTS: Salt treatment led to a reduction in aerial part growth. Salinity increased the fatty acid content more significantly in Tunisian variety (TV) than in Canadian variety (CV). CV showed an increase in double‐bond index (DBI) and a decrease in malondialdehyde content under salt stress, while the opposite was observed in TV. The DBI was mainly affected by a strong reduction in oleic and linoleic acids in TV, whereas a strong stimulation of linoleic acid in CV was observed. Salt decreased and increased the essential oil yield in TV and CV respectively. The main constituents of the essential oil of TV were trans‐hydrate sabinene and terpinen‐4‐ol, which showed a significant decrease under salt stress. In contrast, the main constituents of the essential oil of CV were sabinene and trans‐hydrate sabinene, which showed a significant decrease and increase respectively under salt stress. CONCLUSION: Marjoram oil is a rich source of many compounds such as essential oils and fatty acids, but the distribution of these compounds differed significantly between the two varieties studied. 姜黄为姜科姜黄属的多年生草本植物,是一种药食两用材料。采用水蒸汽蒸馏法提取了姜黄精油,研究了其对ABTS+自由基、羟自由基的清除率及其抑制亚油酸脂质过氧化的能力,通过MTS法研究了其对HepG2细胞的细胞毒作用,采用流式细胞技术评估了其在细胞水平的抗氧化能力。结果表明:姜黄精油对ABTS+自由基具有良好的清除效果,但其清除能力低于VE;对羟自由基的清除效果非常明显,与VE的清除能力相当;并且能有效抑制亚油酸脂质过氧化。姜黄精油在0.02%(v/v)浓度范围内没有明显的细胞毒作用,在此浓度范围内能有效清除HepG2细胞内的活性氧。由此表明,姜黄精油具有开发为安全天然的食品抗氧化剂的潜力。   相似文献

The essential oil obtained by hydrodistillation from the aerial parts of Origanum minutiflorum O Schwarz & PH Davis, an endemic species in Turkey, was analysed for its antimicrobial activity in vitro. Gas chromatography/mass spectrometry analysis of the essential oil resulted in the identification of 34 constituents accounting for 961.5 mL L?1 of the oil, the major compounds present being carvacrol (793.4 mL L?1), p‐cymene (32.6 mL L?1) and γ‐terpinene (21.4 mL L?1). The in vitro antimicrobial activity of the essential oil was investigated in order to evaluate its efficacy against 16 bacteria and two yeasts, using disc diffusion and minimum inhibitory concentration methods. The essential oil showed strong antimicrobial activity against all test micro‐organisms except Pseudomonas aeruginosa. Its main components carvacrol and p‐cymene were also assayed for their antimicrobial activities. Carvacrol exhibited comparable activity to the crude oil, proving it to be the main component responsible for the biological activity observed. This study demonstrates the in vitro antimicrobial activity of the essential oil of this endemic remedy against a wide spectrum of clinically important micro‐organisms, including pathogenic yeasts, being the first report on the anticandidal properties of the essential oil of O. minutiflorum. Strawberries are a good source of micronutrients, especially antioxidant phenolics. More information is needed to better exploit the health-promoting effect of such fruits. Several studies focused on the effects of genotype, cultural practices, and seasonal variation on the antioxidant potential of strawberries, but often yielding contradictory results and/or focusing on only 1 source of variability. In the present study, we showed that total phenolic compounds, ascorbic acid, and antioxidant capacity strongly differed between genotypes of strawberry. The precise results revealed the importance of genetic background for the antioxidant capacity and for the content of total phenolics (with up to 3.3-fold variations). Other parameters may also influence the antioxidant capacity in strawberry such as harvest time, culture conditions, and environmental factors. Moreover, in this study, the harvesting time (at the same ripening stage) appeared to be very important, more important than genotype. Variations of the antioxidant capacity of up to 4.1-folds were observed following the harvesting time of "Elsanta" cultivar. PRACTICAL APPLICATION: This article compares the antioxidant capacity and the content in ascorbic acid and phenolic compounds of strawberries of different varieties and of fruits harvested from April to December at the same ripening stage. The importance of strawberry antioxidant capacity resides in its benefits for human health.  相似文献

采用GC-MS法分析经水蒸气蒸馏法所提柠檬果皮精油的化学组分,并对其抗氧化与抑菌活性进行了研究。实验结果表明:鉴定出23种挥发性成分,占精油总量的94.57%,主要为dl-柠檬烯(42.93%)、γ-萜品烯(8.41%)、α-松油醇(6.39%)和E-柠檬醛(6.09%)等;柠檬精油对OH自由基有较好的清除效果,清除率可达95.14%;而对DPPH自由基的清除率在实验条件下最高为34.81%,远低于同浓度的PG和TBHQ。200mg/kg添加量的柠檬精油可延缓猪油和花生油的贮藏期,但略差于同浓度TBHQ和VE的抗氧化效果。抑菌活性实验结果表明柠檬精油对6种供试菌都有一定的抑菌作用,其中对大肠杆菌的抑菌效果最好,其最低抑菌浓度(MIC)为0.1μL/m L。   相似文献

Proximate, minerals, amino acids (AAs), phenolic composition, antioxidant and functional properties of flaxseed (FSC), sesame (SSC), mustard (MSC), nigella (NSC) and groundnut (GSC) seedcakes were studied. SSC exhibited higher ash and fat contents while GSC and FSC had more protein and mineral contents (P, Mg, Mn and Cu), respectively. The total flavonoids, phenolics and antioxidant activity were higher in NSC (18.60 mg QE g−1), MSC (5.12 mg GAE g−1) and FSC (12.28 µmol TE g−1), respectively. NSC exhibited the highest oil absorption capacity (2.10 g g−1) and emulsifying activity index (89.52 m2 g−1). Twenty-one AAs including citrulline and ɣ-aminobutyric acid were detected in oilseed cakes (OSC). MSC had more essential AAs (methionine, isoleucine, tryptophan, threonine and lysine) while GSC had more total hydrophobic, acidic and basic AAs. Syringic acid and rutin were identified as major phenolic compounds in GSC and MSC, respectively. The total free phenolic acids were more in GSC while FSC had high total bound phenolic acids and flavonoids. OSC can be utilised in food products as a supplement to improve nutritional properties.  相似文献

The chemical composition of essential oil from pine needles (Cedrus deodara) was determined, and its antioxidant and antimicrobial activities were evaluated. Twenty-three components, representing 95.79% of the oil, were identified by gas chromatography mass spectrometry. The main components include α-terpineol (30.2%), linalool (24.47%), limonene (17.01%), anethole (14.57%), caryophyllene (3.14%), and eugenol (2.14%). Pine needle essential oil showed remarkable antioxidant activity in scavenging free radicals, in lipid peroxidation, and in reducing power assays. Moreover, the essential oil revealed strong antimicrobial activity against typical food-borne microorganisms, with minimum inhibitory concentration and minimum bactericidal concentration values of 0.2 to 1.56 and 0.39 to 6.25 μg/mL, respectively. Transmission electron microscope observation ascertained that the bactericidal mechanism of pine needle essential oil may be the induction of cytoplasmic outflow and plasmolysis. These results suggest that the essential oil from pine needles has potential to be used as a natural antioxidant and antimicrobial agent in food processing. PRACTICAL APPLICATION: The present study provides a theoretical basis for the potential application of essential oil from pine needles (C. deodara) to be used as a natural resource of antioxidant and antimicrobial agents in food industry.  相似文献

采用水蒸气提取法对蒌叶挥发油进行提取,在单因素实验的基础上,以正交实验筛选最佳的提取工艺条件;同时考察蒌叶挥发油抗氧化和抑菌活性。结果表明,蒌叶挥发油提取的最佳工艺条件为:料液比m干蒌叶粉∶V=50∶1000(g/m L),超声时间为30 min,超声温度70℃,超声功率192 W,加热回流提取4 h,蒌叶挥发油的得率为2.96%。蒌叶挥发油清除DPPH·和·OH的IC50分别为33.19、71.11μg/m L;挥发油对金黄色葡萄球菌、枯草杆菌、大肠杆菌、蜡状芽孢杆菌、四联球菌、藤黄八叠球菌、白色葡萄球菌、黑曲霉、毛霉、青霉均有明显的抗菌活性,其MIC分别为0.625、1.25、0.625、0.625、0.625、2.50、1.25、0.313、0.625、0.313 mg/m L,故蒌叶挥发油具有较强的抗氧化和抑菌活性。   相似文献

BACKGROUND: The effect of foliar fertilization and Glomus intraradices inoculation on the growth, qualitative and quantitative pattern of essential oil in Salvia officinalis was determined. Sage plants were grown in a glass house on a soil/sand mixture (w/w = 3:1). Agroleaf® total, N:P:K = 20:20:20 + microelements, was used at the whole vegetative growth stage as a 0.3% solution. Inoculation with Glomus intraradices was done at the sowing stage. RESULTS: Application of foliar fertilization and/or mycorrhizal colonization improved dry biomass accumulation and increased the content of antioxidant metabolites (ascorbate and reduced glutathione). Applied treatments lowered the activities of the antioxidants enzymes catalase, ascorbate peroxidase and superoxide dismutase, while guaiacol peroxidase increased. The relative quantity of essential oil pattern was also altered as a result of the applied treatments. Combined application (FF + Gi) significantly promoted 1,8‐cineole and α‐thujone, mycorrhizal colonization enhanced bornyl acetate, 1,8‐cineole, α‐ and β‐thujones, while foliar fertilization increased bornyl acetate and camphor. The favorable effect of root colonization by Glomus intraradices was determined both on quantitative and qualitative pattern of sage essential oil. CONCLUSION: We conclude that inoculation with Glomus intraradices resulted in improved essential oil yield and quality, while combined application of foliar fertilizer and mycorrhizal fungi predominantly enhanced shoot biomass accumulation. 以正己烷为溶剂提取草果精油,利用气相-质谱联用技术(GC-MS)分析其化学成分,同时研究了草果精油的抗氧化和抑菌活性。结果表明,草果精油成分比较复杂,其中有23种化合物被鉴定,主要成分为香叶醇(29.42%)、橙花叔醇(9.87%)、十八碳烯酸(8.62%)等。草果精油对DPPH和ABTS自由基均显示出了较好的清除效果,EC50值分别为3.84、3.20mg/mL;不同浓度的草果精油对5种测试菌都有一定的抑菌作用,其中对大肠杆菌的抑菌效果最好,其最低抑菌浓度(MIC)为2.0mg/mL。   相似文献
