BACKGROUND: Soy milk is one of the common soy‐based foods in Asia. In this study the effects of soy milk fermented with selected probiotics on nitric oxide (NO)‐mediated vascular relaxation factors in cell model systems were investigated. RESULTS: Soy milk fermented with Lactobacillus plantarum TWK10 or Streptococcus thermophilus BCRC 14085 for 48 h showed a greater transformation of glucoside isoflavones to aglycone isoflavones (P < 0.05). An increase in aglycone isoflavones in ethanol extracts from fermented soy milk stimulated NO production and endothelial NO synthase (eNOS) activity in human umbilical vein endothelial cells. It also had a stimulating effect on superoxide anion scavenging and prostaglandin E2 production. In addition, it enhanced mRNA expression of the E‐prostanoid 4 receptor in rat thoracic aorta smooth muscle cells. Moreover, a small amount of O induced by water extracts from fermented soy milk at low concentration (1 mg mL?1) increased the content of calcium ions and activated eNOS, thereby promoting NO production and the coupling state of eNOS. CONCLUSION: Soy milk fermented with selected probiotics promotes the relaxation factors of vascular endothelial cells and can be applied in the development of functional foods. © 2012 Society of Chemical Industry  相似文献

It has been proposed that isoflavones and probiotics possess specific biological activities that enable them to affect lipid metabolism. The purpose of this study was to evaluate the effects of a soy product fermented with Enterococcus faecium and Lactobacillus jugurti and supplemented with isoflavones on the plasma lipid and glucose profiles, and on the retroperitoneal (RET) and epididymal (EPI) adipose tissue adipocyte areas in juvenile rats. Male Wistar rats were subjected to 6-week experimental diets: (1) cholesterol-enriched diet (rat chow) plus fermented soy product supplemented with isoflavones, (2) cholesterol-enriched diet plus unfermented placebo, (3) cholesterol-enriched diet plus placebo supplemented with isoflavones, (4) standard diet (rat chow), or (5) cholesterol-enriched diet. Soy products affected adipose tissue in a regional-specific manner and may be responsible for the observed increase in RET adipocyte area (μ2): (1) 12757.00 ± 286.25; (2) 13269.00 ± 326.87 or (3) 13050.00 ± 226.85 versus (4) 8714.92 ± 262.41 or (5) 8892.70 ± 131.79 or decrease in EPI adipocyte area (μ2): (1) 7759.00 ± 108.89 and (2) 7481.90 ± 111.21 compared with the control group (4) 8346.60 ± 158.89. The effect on epididymal adipose tissue was probably due to isoflavones. Probiotics significantly raised the plasma level of HDL cholesterol. Fermented soy product supplemented with isoflavones did not promote changes in the glucose and triglyceride plasma levels. This product may offer a new approach for improving or preventing lipid metabolism abnormalities by decreasing visceral adipocyte area, adjusting cholesterol-mediated loss of RET adipose tissue (lipoatrophy), and increasing the HDL cholesterol plasma level.  相似文献

旨在为大豆蛋白在冷冻发酵面团中的应用提供一定的理论依据,对添加大豆分离蛋白(SPI)、质构化大豆分离蛋白(TSP)、水解大豆分离蛋白(SPH)的冷冻发酵面团馒头及冷冻发酵面团进行研究,分析了添加不同种类大豆蛋白的冷冻发酵面团馒头的理化指标(比容、水分含量、pH),并通过测定冷冻发酵面团的二硫键含量、蛋白质二级结构和微观结构,探讨不同种类大豆蛋白对冷冻发酵馒头品质的影响机制。结果表明:添加SPH的发酵面团冷冻储藏前后所蒸制的馒头比容变化最小,在冷冻储藏40 d时蒸制的馒头比容相比冷冻储藏10 d增加了4.59%;同一冷冻储藏时间下添加SPH的发酵面团蒸制的馒头水分含量最低;添加大豆蛋白可以减小在冷冻储藏过程中发酵面团馒头pH的下降速率,至少可延长冷冻发酵面团10 d的保质期;通过傅里叶红外光谱和激光共聚焦对冷冻发酵面团蛋白质结构分析,得出添加3种大豆蛋白均会降低面筋蛋白网络结构的均匀性和连续性,SPI和TSP对面筋蛋白网络结构的影响小于SPH,但在冷冻储藏过程中添加SPH的发酵面团的面筋蛋白网络结构相对较为稳定。综上,大豆蛋白与面筋蛋白通过二硫键和非共价键相互作用,改变面筋蛋白网络结构的...  相似文献

BACKGROUND: In this study, lactic acid bacteria (LAB) were isolated, characterized and identified from jiang‐sun (fermented bamboo shoots; a traditional fermented food in Taiwan). Samples were collected at seven time intervals from a fixed fermenting bucket during the fermentation process of jiang‐sun and its initial ingredients, dochi (fermented soybeans) and bamboo shoots. RESULTS: A total of 57 LAB cultures were isolated; 42 cultures were isolated from jiang‐sun samples and 15 cultures were isolated from dochi and bamboo shoots. These isolates were characterized phenotypically and then divided into three groups (A–C) by restriction fragment length polymorphism analysis and sequencing of 16S ribosomal DNA. Alteration of microbial populations during the fermentation process was observed. While various LAB were found in the dochi and bamboo shoots, it was mostly replaced by Lactobacillus plantarum after 1 day of fermentation. Furthermore, the antibacterial activities of the isolates were determined, and one Enterococcus faecium strain showed inhibitory activity against all the indicator strains. CONCLUSION: Results suggest that L. plantarum is the main LAB present during the fermentation of jiang‐sun. 为探究点浆时豆乳温度、点浆后蹲脑温度和蹲脑时间对酸浆豆腐品质的影响,提高酸浆豆腐品质,本研究利用质构分析、SDS-PAGE和低场核磁等技术分析了豆腐品质、豆乳中蛋白凝固过程、大豆蛋白各亚基含量和水分分布的变化规律。结果表明,点浆时豆乳温度对豆乳沉淀中蛋白和大豆蛋白各亚基含量无显著影响,但对豆腐品质和水分分布有较大影响。当点浆时豆乳温度为75℃时,豆腐的硬度最小,而其弹性、得率、含水量、保水性、感官得分和T21弛豫时间最大。蹲脑温度和蹲脑时间对豆腐品质、豆乳沉淀中蛋白含量和大豆蛋白各亚基含量影响显著,当蹲脑温度75℃和蹲脑时间20 min时,豆腐硬度最小为15.80 N,而弹性、得率、含水量、保水性和T21弛豫时间最佳,分别为5.15 mm、241.3%、76.36%、65.68%、43.29 ms,且豆腐品质豆乳沉淀中蛋白含量最佳。当点浆时豆乳温度为75℃、蹲脑温度为75℃和蹲脑时间20 min时,豆腐品质和感官得分最优。  相似文献

As a nontargeted metabolomics approach, we investigated changes in the plasma metabolite levels in a mouse model of obesity induced by a high‐fat diet and fermented soybean product diet. We analyzed the plasma samples by using ultra‐performance liquid chromatography coupled with quadrupole time‐of‐flight mass spectrometry (UPLC‐Q‐TOF‐MS). In the present study, the animals were divided into four groups according to the diet type; normal fat diet control group (ND), high‐fat diet control group (HD), high‐fat diet plus 30% cooked soybean power (HD + S), and high‐fat diet plus 30% 72‐h fermented Cheonggukjang powder (HD + CGJ). To examine the changes in plasma metabolite levels because of high‐fat diet feeding, total cholesterol and triglyceride levels were measured. Total cholesterol and triglyceride levels were lower in the HD + S and HD + CGJ groups than in the ND group. According to partial least‐squares discriminant analysis (PLS‐DA), major metabolites contributing to the discrimination between each group were assigned as lipid metabolites in plasma, e.g., lyso‐phosphatidylcholines and phosphatidylcholines. Therefore, diets containing soy‐based food products, which are rich sources of isoflavonoids, might be helpful for controlling the lipid metabolism under high‐fat diet conditions.  相似文献

The objective of this study was to compare the influence of hesperidin and naringin, the main flavonones of orange and grapefruit, on plasma lipid profile and antioxidant activity in rats fed a cholesterol‐containing diet. Sixty male Wistar rats were randomly divided into six groups of 10, named Control, Hesperidin, Naringin, Chol, Chol/Hesperidin and Chol/Naringin. The Control group was fed a basal diet (BD) and 1–2 mL of distilled water. To the BD of the other five groups were added 0.1–0.2 mg of hesperidin dissolved in 1–2 mL of distilled water (Hesperidin group), 0.46–0.92 mg of naringin in 1–2 mL of water (Naringin group), 1% of non‐oxidised cholesterol (NOC) and 1–2 mL of water (Chol), 1% of NOC and 0.1–0.2 mg of hesperidin in 1–2 mL of water (Chol/Hesperidin), 1% of NOC and 0.46–0.92 mg of naringin in 1–2 mL of water (Chol/Naringin). After 30 days of the experiment it was found that the diets supplemented with hesperidin and naringin increased the plasma antioxidant activity. In conclusion, diets supplemented with hesperidin and naringin significantly hindered the increase in plasma lipid levels caused by cholesterol feeding. Hesperidin and naringin, bioactive compounds of citrus fruits, are powerful plasma lipid lowering and plasma antioxidant activity increasing flavonones. This study was designed to investigate the effect of soy protein inclusion in milk replacer diets for goat kids on protein, RNA, and DNA contents in small intestinal mucosa, on the importance of RNA biosynthesis from dietary RNA precursors for mucosal RNA synthesis, and on the activities of enzymes involved in nucleotide degradation in small intestinal mucosa. Diets were based on cow's milk. In the control group, 35% of the milk protein was replaced by casein (CN) protein, and in the soy group (SPAA), the same amount of milk protein was replaced by soy protein supplemented with essential AA known to be at lower concentrations in soy than in CN (Thr, Val, Ile, Leu, His, Lys, Met). Diets were isonitrogenous and isoenergetic. At 47 d of age, goats were harvested and samples of proximal, middle, and distal jejunal mucosa were collected 5 h after feeding 15N-labeled RNA from yeast (13 mg/kg of body weight). Growth and feed conversion did not differ between the control and SPAA kids. Mucosal protein concentrations were lower in the SPAA than the control kids. Concentrations of RNA and DNA did not differ between feeding groups, but in all kids mucosal RNA concentrations were higher in proximal than in middle and distal jejunum. Protein:RNA ratios were higher in the control than the SPAA kids and were lowest in proximal jejunum. Activities of alkaline phosphatase in enterocytes were higher in proximal than in middle and distal jejunum. Activities of mucosal xanthine oxidase were highest in distal jejunum and were higher in the SPAA than the control kids, especially in the middle and distal sites. The 15N-enrichment of mucosal RNA was higher in the control than the SPAA kids, especially in distal jejunum, and was lowest in distal jejunum. In contrast, 15N-enrichment of urea in plasma tended to be higher and Gly concentration in plasma was lower in the SPAA than the control kids. Data indicate that protein content and the protein:RNA ratio were lower in jejunal mucosa of goat kids fed milk replacer with partial replacement of CN protein by soy protein. These findings were accompanied by a lower level of reutilization of preformed dietary RNA precursors for RNA biosynthesis in jejunal mucosa and a higher activity of xanthine oxidase. Thus, feeding soy protein instead of CN protein reduced the incorporation of preformed dietary RNA precursors for RNA biosynthesis in the mucosa and activated key enzymes involved in nucleic acid breakdown.  相似文献

BACKGROUND: There are a number of antinutritional factors present in soybeans that exert a negative impact on the nutritional quality of the protein. Among those factors that are destroyed by heat treatment are protease inhibitors and lectins. Protease inhibitors show antinutritional effect and moreover the digestibility of the protein is limited by the presence of these antinutrients. The aims of the present study are (1) to study the effect of autoclaving on the trypsin inhibitor inactivation, nitrogen solubility and protein digestibility of defatted soy flour and (2) to study the effect of enzymatic modification on the functional properties of autoclaved soy flour. RESULTS: The solubility of the soy flour decreased with increase in autoclaving time. Partial hydrolysis of the autoclaved soy flour increased its acid solubility (pH 4.5) from 17% to 56% over a control value of 24% without affecting its functional properties. Inactivation of trypsin inhibitors improved the protein digestibility of soy flour from 25% to 95%. Particle size analysis of the autoclaved flour indicated the formation of soy protein aggregates, which resulted in poor solubility. The enzymatic modification of autoclaved soy flour resulted in its property as a good emulsifying agent with an emulsion capacity of 118 ± 4 mL. CONCLUSION: Enzymatic modification of the heat‐processed soy flour increased its solubility and other functional attributes. The increased acid solubility would be advantageous in the utilization of soy proteins in acidic foods. The effect of heat treatment on the properties of soy protein‐stabilised emulsions was investigated. Emulsions were prepared with unheated and heat‐treated soy protein (NSP and HSP) dispersions. Heating on soy protein dispersions at 95 °C for 30 min resulted in smaller average oil droplet size, lower tendency for oil droplet flocculation, higher protein adsorption and lower viscosity. The properties of emulsions were significantly influenced by the protein concentration. The sodium dodecyl sulphate–polyacrylamide gel electrophoresis (SDS–PAGE) profiles showed that the heat treatment on soy protein dispersions increased the protein adsorption at O/W interface. The viscosity of all samples at low shear rate was inversely proportional to the d32, suggesting a positive relation to the total interfacial area per unit volume. Emulsions showed shear‐thinning behaviour. The relaxation time was found to increase with aqueous phase viscosity determined by the Cross viscosity model.  相似文献

A low‐salt (4% NaCl, w/w) douchi supplemented with different concentration of fermented rice culture (FRC) of 0%, 5%, 10%, 15% and 20% (v/w) was produced, and the effect of FRC on microbiology, biochemistry and sensory quantity was investigated. Results indicated that FRC had a significant effect on low‐salt douchi. Compared with the control (added 0% FRC), in the other treatments, the counts of yeast and lactic acid bacteria increased by about 50%, whereas the moulds decreased by about 30%; the pH reduction was close to 4.7; total acid increased to about 1.300 g per 100 g; reducing sugar maintained a decrease to 21.07 mg per g; amino nitrogen increased to about 0.900 g per 100 g, but not above the control (1.013 g per 100 g). In general terms, total free amino acid contents and three organic acids were higher in low‐salt products than four commercial products. Sensory evaluation of products supplemented with 10% and 15% FRC revealed better overall acceptability compared with other treatments and a commercial product.  相似文献

The effect of inulin and/or okara flour on Lactobacillus acidophilus La-5 and Bifidobacterium animalis Bb-12 viability in a fermented soy product (FSP) and on probiotic survival under in vitro simulated gastrointestinal conditions were investigated throughout 28 days of storage at 4 °C. Employing a 22 design, four FSP trials were produced from soymilk fermented with ABT-4 culture (La-5, Bb-12, and Streptococcus thermophilus): FSP (control); FSP-I (with inulin, 3 g/100 mL of soymilk); FSP-O (with okara, 5 g/100 mL); FSP-IO (with inulin + okara, ratio 3:5 g/100 mL). Probiotic viabilities ranged from 8 to 9 log cfu/g during the 28 days of storage, and inulin and/or okara flour did not affect the viability of La-5 and Bb-12. Bb-12 resistance to the artificial gastrointestinal juices was higher than for La-5, since the Bb-12 and La-5 populations decreased approximately 0.6 log cfu/g and 3.8 log cfu/g, respectively, throughout storage period. Even though the protective effect of inulin and/or okara flour on probiotic microorganisms was not significant, when compared to a fresh culture, the FSP matrix improved Bb-12 survival on day 1 of storage and may be considered a good vehicle for Bb-12 and could play an important role in probiotic protection against gastrointestinal juices.  相似文献
