Glucose administered to fasted rats caused a marked stimulation in hepatic cholesterogenesis and cholesterol 7 alpha-hydroxylation, and an increase in biliary excretion of cholesterol and total bile acids. The excretion of cholic acid was not incluenced during the first few hr after glucose administration, but was significantly increased after 5 hr. Chenodeoxycholic acid showed a similar change, but the increase was only ca. one tenth of that of cholic acid. The excretion of deoxycholic acid was markedly increased by 1 hr, but gradually decreased thereafter. Pretreatment with neomycin abolished the increase in deoxycholic acid by fasting and glucose administration. Other bile acid components showed no significant change. It thus was presumed that cholesterol endogenously synthesized in the liver was metabolized mainly to cholic acid. In contrast, exogenous cholesterol was metabolized mainly to chenodeoxycholic acid. During the period of the acute enhancement of cholic acid formation from the endogenous cholesterol, biliary excretion of deoxycholic acid was increased. This probably occurred through the depression of 7 alpha-rehydroxylation of deoxycholic acid, or through the enhancement of microbial formation of deoxycholic acid in the lumen, and through the increase of intestinal absorption.  相似文献

The present study investigated the metabolic fate of dietary TAG and DAG and also their digestion products in the stomach and small intestine. A diet containing 10% TAG or DAG oil, enriched in 1,3-DAG, was fed to Wistar rats ad libitum for 9 d. After 18 h of fasting, each diet was re-fed ad libitum for 1 h. The weights of the contents of the stomach and small intestine were measured, and the acylglycerol and FFA levels were analyzed by GC at 0, 1, and 4 h after the 1-h re-feeding. The amounts of re-fed diet ingested and the gastric and small intestinal content were not different between the two diet groups. In the TAG diet group, the main products were TAG and DAG, especially 1(3),2-DAG. In addition, 1,3-DAG and 1(3)-MAG were present in the stomach, and the 1,3-DAG levels increased over time after the re-feeding period. In the DAG diet group, the main products in the stomach were DAG, MAG, FFA, and TAG. There were significantly greater amounts of 1,3-DAG, 1(3)-MAG, and FFA in the DAG diet group in the stomach compared with the TAG diet group. The amount of FFA in the stomach relative to the amount of ingested TAG plus DAG in the DAG diet group was higher than that in the TAG diet group. Acylglycerol and FFA levels were considerably lower in the small intestine than in the stomach. These results indicate that, in the stomach, where acyl migration might occur, the digestion products were already different between TAG and DAG oil ingestion, and that DAG might be more readily digested by lingual lipase compared with TAG. Furthermore, almost all of the dietary lipid was absorbed, irrespective of the structure of the acylglycerol present in the small intestine.  相似文献

The hypolipidemic properties of ethyl 6-chlorochroman-2-carboxylate (II), 6-phenylchroman-2-carboxylate (III) and 6-cyclohexylchroman-2-carboxylate (IV) were compared to clofibrate (I) in sucrose-fed fasted male Sprague-Dawley rats. All compounds were administered at doses of 0.2 and 0.4 mmol/kg, orally, twice daily for 7 consecutive days. In this model, II was a more effective hypocholesterolemic drug than clofibrate, whereas III and IV were inactive. Chlorochroman II, like clofibrate, decreased serum α-lipoprotein cholesterol and pre-β-lipoprotein triglyceride concentrations and concomitantly increased serum β-lipoprotein triglyceride concentrations. In clofibrate-treated rats, serum free cholesterol concentrations increased concurrent with a reduction in serum lecithin: cholesterol acyltransferase activity, but no such correlation was observed for II. Only II lowered liver cholesterol levels and increased liver triglyceride levels. No consistent inhibition of liver microsomal 3-hydroxy-3-methylglutaryl-CoA reductase activity was observed with these analogs. The observed changes in triglyceride and cholesterol concentrations among serum lipoproteins were of a greater magnitude after chlorochroman II and clofibrate administration to sucrose-fed rats than in our previous studies using chow-fed fasted rats. These data suggest that chloro-substitution at the 6-position of the phenylchroman ring is important for hypolipidemic activity of these cyclic clofibrate analogs.  相似文献

It has been shown that cocaine given in the diet is able to reduce fat accumulation in the liver of protein-malnourished rats (Arch. Latinoamer. Nutr. 19: 69-79, 1969). This study was, therefore, designed to approach the probable action of the drug upon the process (increased triglycerides synthesis and normal/decreased capacity for exporting triglycerides from the liver into the blood) which leads to an increased fat accumulation in the liver under this physiological condition. To accomplish this purpose, the total and fractioned lipids in the liver and total lipids as well as lipoproteins in serum were determined in female Wistar rats (120-130 g) fed either a 5% corn protein diet or a 20% casein diet, with and without cocaine (15 mg HCl cocaine/10 g of diet) for 18 days. The results, aside from confirming the reduction (p less than 0.001) of fat accumulation in the liver of rats fed on the 5% corn protein diet plus cocaine, revealed that this drug also reduced triglycerides concentration (significantly, (p less than 0.001, when results were calculated by difference, and slightly reduced them when results were determined) in this tissue. Nevertheless, it increases both total lipids (p less than 0.05) and triglycerides-rich pre-beta lipoprotein (p less than 0.10) levels in the serum of these animals. Otherwise, these lipidic parameters were not modified by cocaine in rats on the 20% casein diet, except for the total cholesterol level in liver and the cholesterol-rich beta lipoprotein level in serum. Respectively, these were slightly and significantly (p less than 0.001) reduced by the drug. These evidences and their statistical significance suggest that cocaine given chronically with the 5% corn-protein diet for 18 days, reduces at least partially (other biochemical event in the liver could have also accounted for its effect at this level) the liver fat accumulation, by increasing the triglycerides output from the liver into the blood. Elsewhere, cocaine appears to be able to induce some metabolic alterations in the hepatic cholesterol of well-nourished rats.  相似文献

Because China has put more emphasis on people with disabilities, their living conditions and protection have received increasing attention. In an emergency, the visually impaired people are limited in their ability to evacuate, especially along egress paths with obstacles. This paper studied the relationship between the travel time of visually impaired people and obstacle density in their path. Eight sighted people and 32 people with visual impairments were chosen to carry out the walking experiment. The results of the experiment showed that the travel time of sighted people was shorter than that of visually impaired people under the same obstacle and non‐obstacle situations. Based on the analysis of variance of travel time, it was clear from the results that the travel time of the visually impaired people rapidly increased when there were obstacles. When the obstacle density was kept to within a certain range, there was no obvious increase in travel time. Once the density exceeded this range, there was another obvious increase. Based on this result, it can be seen that keeping the obstacle density to within a certain range could be helpful for the visually impaired people during an evacuation when the egress paths cannot be made obstacle‐free.  相似文献

Polymer solution for oil displacement is mostly used in the middle and late stage of water flooding reservoir development, and reservoir groundwater conditions are often one of the main conditions restricting polymer application. Therefore, it is necessary to develop salt tolerance of polymer solutions with different aggregation behaviors, so as to facilitate the synthesis and optimization of suitable polymer systems. The differences in the micro-aggregation behavior of three polymers with different molecular structures were explored. On this basis, the effects of divalent metal cations on the properties of the polymer solutions were analyzed by assessing the micro-aggregation behavior, apparent viscosity, hydrodynamic size, and shear rheological characteristics. The results showed that the linear partially hydrolyzed polyacrylamide (HPAM) was seriously affected by divalent cations, and the viscosity decreased obviously. The aggregation behavior of the polymer changed by hydrophobic association can enhance the salt tolerance of the solution. The hydrophobically modified partially hydrolyzed polyacrylamide (HMPAM) with “chain beam” aggregation behavior has strong intermolecular connection, which enables it to withstand the content of calcium and magnesium ions of 1100 mg l−1. When the content of calcium and magnesium ions exceeds 600 mg l−1, dendritic hydrophobically associating polymer (DHAP) will destroy the interaction between molecular chains, resulting in the decrease of apparent viscosity and hydrodynamic size. For polymer flooding in high-salinity reservoir, salt tolerant polymer system can be constructed by optimizing molecular weight and hydrophobic group content.  相似文献

In this study, we examined whether the increased availability of lipids in blood resulting from two types of diet manipulation regulated metabolic gene expression in the skeletal muscle of rats. Feeding for 4 wk on an isocaloric-sucrose or a hypercaloric-fat diet increased plasma TAG in the fed condition by increments of 70 and 40%, respectively, and increased fasting insulinemia (approximately 3-fold) compared with a starch diet. The fat diet impaired glucose tolerance and caused obesity, whereas sucrose-fed rats maintained their normal weight. We analyzed the expression of genes that regulate the exogenous FA supply (LPL, FAT/CD36, FATP1), synthesis (ACC1), glucose (GLUT4, GLUT1, HK2, GRAT1, glycogen phosphorylase) or glycerol (glycerol kinase) provision, or substrate choice for oxidation (PDK4) in gastrocnemius and soleus muscles at the end of the glucose tolerance test. LPL, FAT/CD36, FATP1, PDK4, and GLUT4 mRNA as well as glycogen phosphorylase and glycerol kinase activity levels in both muscles were unchanged by the diets. Increased mRNA levels of GLUT1 (1.6- and 2.6-fold, respectively) and GFAT1 (about 1.7-fold) in gastrocnemius, and of ACC1 (about 1.5-fold) in soleus, were found in both the sucrose and fat groups. In the fat group, HK2 mRNA was also higher (1.8-fold) in the gastrocnemius. Both sucrose and saturated-fat diets prompted hyperinsulinemia and hyperlipemia in rats. These metabolic disturbances did not alter the expression of LPL, FAT/CD36, FATP1, PDK4, and GLUT4 genes or glycogen phosphorylase and glycerol kinase activity levels in either analyzed muscle. Instead, they were linked to the coordinated upregulation in gastrocnemius of genes that govern glucose uptake and the hexosamine pathway, namely, GLUT1 and GFAT1, which might contribute to insulin resistance.  相似文献

Reports in the literature concerning the relationship of protein nutrition to aflatoxicosis are contradictory. In an attempt to elucidate this relationship more clearly, we have examined the effects of low, normal, and high protein-containing diets on tumor incidence and development, as well as on several biochemical indices, in rats which have been exposed to low levels of aflatoxin in a “chronic” rather than “acute” situation. In our study, male weanling rats were place for 3 months on otherwise adequate diets containing either 8, 22, or 30% casein with and without aflatoxin B1 at 1.7 ppm. Half of the animals in each group received diets which were further supplemented with the amino acid, cystine, at 0.6% of the diet. (Sulfur-containing amino acids are the most limiting amino acids in casein, and the addition of cystine to the diet serves to improve the biological quality of the protein source.) After 3 months the animals were fed control diets without aflatoxin until they were killed at 1 year. Weight gain was markedly decreased and liver weights increased in response to aflatoxin in all groups except those on the low protein diets, where aflatoxin had no effect on these protein diets, where aflatoxin had no effect on these indices. No tumors were found in the livers of rats fed the low protein, aflatoxin-supplemented diet. In the other groups, the severity of the liver involvement increased progressively with increased protein levels in the diet. When cystine was included in the diet, tumors were observed also in the animals fed the low protein diet; furthermore, the livers of those animals on “normal” and high protein diets were much more severely involved than were the livers of animals on non-cystine supplemented diets. Plasma cholesterol levels were increased in response to aflatoxin when the diets containing 22 and 30% protein were fed and when cystine was included in the 8% protein diet. Liver cholesterol levels were increased in response to aflatoxin in all groups except in those receiving the low protein diets. Among these latter animals, aflatoxin administration had no effect on liver cholesterol values. Changes as a result of aflatoxin administration were also observed in the fatty acid composition of sterol esters, triglycerides, and phospholipids of liver and tumor tissue.  相似文献

目的观察不同剂量南极磷虾粉及其与钙联合对骨质疏松模型大鼠的作用。方法将大鼠随机分为10组,即虾粉低、中、高剂量组、钙组、联合低、中、高剂量组、氟化钠组、假手术组、模型对照组,每组10只,通过切除双侧卵巢建立骨质疏松模型,假手术组仅切除卵巢附近部分脂肪组织。各实验组分别灌胃低、中、高剂量虾粉、钙、低、中、高剂量虾粉+钙、氟化钠,假手术组与模型对照组正常饮食。12周后,经腹动脉取血,分离血清,检测钙离子浓度及碱性磷酸酶(Alkaline phosphatase,ALP)活性;取股骨观察骨密度及生物力学差异,并检测骨灰分含量;取胫骨检测骨氟含量。结果虾粉低、中、高剂量组及联合低、中、高剂量组大鼠血钙浓度随虾粉剂量的增加逐渐降低,虾粉高剂量组与模型对照组相比,差异有统计学意义(P<0.05),而联合低、中、高剂量组大鼠血钙浓度高于模型对照组;虾粉低、中、高剂量组ALP活性均高于模型对照组(P<0.05),且随着虾粉剂量的增加,ALP活性增强。联合低、中、高剂量组大鼠骨密度均高于模型对照组(P均<0.05),且优于低、中、高剂量虾粉组;与模型对照组相比,虾粉低剂量组及联合中、高剂量组均可明显增加骨最大力学载荷值(P均<0.05);与模型对照组相比,低、中、高剂量虾粉组随虾粉剂量增加骨氟含量增多,与钙联合应用后,降低了骨氟含量(P均<0.01),联合低、中、高剂量组与相同剂量的虾粉组相比,骨氟含量均降低;虾粉低、中、高剂量组与模型对照组相比,骨灰分含量升高(P均>0.05)。结论使用虾粉治疗骨质疏松时,低剂量效果最佳;若虾粉与钙联合治疗,中剂量虾粉加钙改善骨质疏松效果最佳。  相似文献

目的观察培哚普利对去势大鼠骨质疏松的影响。方法将大鼠随机分为5组,每组10只。经切除大鼠双侧卵巢建立骨质疏松动物模型(OVX组)后,低(LP组)、中(MP组)、高剂量培哚普利组(HP组)经灌胃分别按2、4、8 mg/(kg·d)给药(溶解于1 ml生理盐水),正常对照组(CON组)仅切除卵巢周围部分脂肪组织,CON与OVX组仅灌胃等体积(1 ml)的生理盐水,均仅给药1次。给药24周后经股动脉采血并处死大鼠,检测血清钙、磷及碱性磷酸酶(ALP)活性,取股骨检测骨密度及生物力学变化。结果与CON组比较,OVX组血清钙、磷、ALP活性及骨密度明显下降(P0.05)。与OVX组比较,MP及HP组大鼠的血清钙含量明显升高(P0.05);MP组大鼠的血清磷含量明显升高(P0.05);HP组大鼠的ALP活性明显升高(P0.05),MP及HP组大鼠的骨密度均明显升高(P0.05);MP组大鼠股骨的最大力学载荷明显升高(P0.05)。结论各剂量培哚普利对改善骨质疏松模型大鼠的骨密度及生物力学均有效果,以中剂量培哚普利改善效果最佳。  相似文献

Nicotine, a major component of cigarette smoke, plays an important role in the development of cardiovascular disease and lung cancer in smokers. The effect of nicotine on lipoprotein metabolism was studied using rats as the experimental animal. There was a significant increase in the total cholesterol, phospholipids, and triglycerides as well as the amount of lipids associated with very low density lipoprotein (VLDL) and low density lipoprotein (LDL) in sera of nicotine-treated rats. The incorporation of 3H labeled leucine into the apo B was found to be increased both in the medium and associated cells in the hepatocytes isolated from nicotine-treated rats indicating an increased synthesis and secretion of the apo B containing lipoproteins. This was further confirmed by the higher incorporation of 14C acetate into total and individual lipids of LDL and VLDL secreted into the medium as well as that associated with different lipids in the cell layer. The activity of lipoprotein lipase in extrahepatic tissues and plasma lecithin cholesterol acyltransferase activity were significantly lower in nicotine-treated rats. These results indicate that nicotine exerts hyperlipidemic effects particularly by increasing the synthesis and secretion of triglyceride-rich lipoproteins. Since nicotine is one of the major hazardous components present in cigarette smoke and tobacco, one can extrapolate that the deleterious effect exerted by nicotine on rats extends to cigarette smokers and those who use other forms of tobacco.  相似文献

The formation of monoacyl- and diacylglycerol 3-phosphate (P) by rabbit heart mitochondrial and microsomal fractions was studied by varying the concentration of acyl-CoA and that of bovine serum albumin in the assay system. The two subcellular fractions were prepared by the conventional differential centrifugation technique. The optimal concentration of acyl-CoA for both mitochondrial and microsomal acylation of glycerol 3-P was shifted to a higher range of acyl-CoA concentrations by greater amounts of albumin. A similar shift in the acyl-CoA concentration-enzyme activity relationship was observed in the acylation reaction of 1-palmitoylglycerol 3-P by the heart microsomes. The addition of albumin increased slightly the rate of diacylglycerol 3-P accumulation but increased greatly the rate of monoacylglycerol 3-P accumulation at any concentration of acyl-CoA; the effect was observed with mitochondrial or microsomal fraction as the crude enzyme source. Moreover, palmitoyl-CoA and linoleoyl-CoA served equally well as the acyl donor for the acylation reaction. However, relatively more monoacyl- than diacylglycerol 3-P was accumulated in the assays with rabbit heart mitochondrial fraction in the presence of albumin, whereas more diacylthan monoacyglycerol 3-P was formed by the microsomal fraction. As a result, the microsomal diacyl: monoacyl-glycerol 3-P ratio was invariably greater than the mitochondrial ratio at a given concentration of acyl-CoA and albumin. The biosynthesis of diacylglycerols from 2-monoacylglycerols and free fatty acids was examined in evert sacs of rat intestinal mucosa. By means of alternate labeling of the monoacylglycerols and fatty acids, and conventional stereo-specific analysis, it was shown that the main products of synthesis were thesn-1,2-diacylglycerols (53–63%), butsn-2,3-diacylglycerols (37–47%) were also formed in significant amounts. The total yield and proportions of the isomeric diacylglycerols recovered appeared to vary with the nature of the monoacylglycerol and the complexity of the free fatty acid mixture supplied.  相似文献

Indirect evidence has suggested that lipid peroxidation is associated with iron overload in vivo. As a measure of lipid peroxidation, pentane expired in the breath of rats loaded with an accumulated dose of either 100 mg or 186–200 mg of iron injected intraperitoneally as iron dextran was measured over a 7 to 8 week period, and the effect on pentane production of feeding antioxidant-supplemented diets was determined. By the seventh week of feeding the diets, rats fed 0.3% L-ascorbic acid produced 17% less (P=0.03) pentane than did rats fed the basal antioxidant-deficient diet, whereas rats fed 0.004% dl-α-tocopherol acetate produced 92% less (P<0.001). After being fed the basal diet for 7 weeks, iron-loaded rats produced 76±9 pmol pentane/100 g body wt/min. When synthetic antioxidants were added to the diet at a concentration of 0.25%, the order of effectiveness in decreasing pentane production after 1 week was: N,N′-diphenyl-p-phenylenediamine > ethoxyquin > butylated hydroxyanisole > butylated hydroxytoluene > propyl gallate ∼ no antioxidant. After removal of either ethoxyquin or N,N′-diphenyl-p-phenylenediamine from the diets for 1 week, pentane production increased to a high level. The total amount of lipid soluble fluorophores in individual spleens of rats fed N,N′-diphenyl-p-phenylenediamine, ethoxyquin, dl-α-tocopherol acetate, ascorbic acid and no antioxidant were correlated significantly with the corresponding total integrated amount of pentane produced by the individual rats over the 7 to 8 week period. This study has provided some of the most direct evidence to date that lipid peroxidation is associated with iron overload in vivo.  相似文献

In an attempt to clarify the mechanism of lipid accumulation inMortierella ramanniana var.angulispora, diacylglycerol acyltransferase (DGAT) in the membrane fraction from this fungus was characterized. The enzyme had an optimum pH of 7.0–7.5, and enzyme activity was blocked by SH-reagents. Metal ions were not essential for maintaining DGAT activity.n-Octyl-β-d-glucoside, 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate and Tween 80 were found to preserve activity, while Triton X-100 and sucrose monolaurate inhibited it. As the inhibition of DGAT activity by Triton X-100 was overcome by the addition of diacylglycerol (DG), the dependency of DGAT activity on exogenous DG was determined in the presence of 0.1% Triton X-100. DGAT activity in the membrane fraction was traced in fungi cultured for different time periods or in media at different carbon to nitrogen (C/N) ratios. Although the increase in total lipid content with culture time was accompanied by an increase in DGAT activity, total lipid changes related to changes in C/N ratio did not correlate with DGAT activity. Factors other than DGAT activity in the membrane fraction would appear to be involved in the regulation of total lipid content in this fungus.  相似文献

通过对酶催化部分水解反应和酶催化酯化反应的有效整合,提出发展了全酶法制备甘油二酯的专利工艺.在前期实验室研究的基础上,进行全酶法制备甘油二酯整套工艺的放大研究.多批次的小试、中试实验结果与实验室研究的结果比较相近,采用游离酶催化油脂部分水解反应及采用填充床酶反应器的酯化反应还具有反应效率较高、工艺操作简单、反应重现性好、生产成本低廉等优点,是制备中、高含量甘油二酯产品的适宜方法;反应后的半成品经过分子蒸馏的分离、提纯及进一步的精制,可以获得符合相关国家标准的甘油二酯产品.  相似文献

The effect of saccharides, such as glucose, on the photocatalytic decolorization of various dyes has been investigated using TiO2 as a photocatalyst in an aqueous solution saturated with nitrogen. An azo dye, such as Orangell, used as a typical dye, was reduced by cleavage of the azo linkage, as evidenced by the production of sulfanilic acid. All of the dyes tested were decomposed with first‐order kinetics. The rate constants of decolorization in the aqueous solution saturated with nitrogen increased with the addition of a saccharide such as glucose. Furthermore, the effect of the reductive saccharides on the photocatalytic decolorization of dyes was quantitatively correlated with their dispersion force. The reaction rate of the photocatalytic decolorization of dyes in the aqueous solution saturated with nitrogen, including glucose, is 7 times that in the solution saturated with oxygen.  相似文献

将代谢工程理论应用于动物细胞的流加培养过程。通过构建代谢网络模型,研究了杂交瘤细胞从动态到拟稳态过程中物质和能量代谢通量的时间分布图。结果表明:当葡萄糖浓度较高时,葡萄糖在乳酸生成途径和TCA循环的代谢通量分配比例分别为90%和10%,ATP生成的比例分别为20%和60%,说明葡萄糖大部分经糖酵解途径生成乳酸,而进入TCA循环的少量部分则提供大部分的能量生成。当葡萄糖浓度降至其限制细胞生长的浓度时,进入TCA循环的葡萄糖比例逐步提高到100%,葡萄糖的氧化程度从0. 6 mmol·mmol-1增至4. 52 mmol·mmol-1,说明葡萄糖代谢逐渐从乳酸生成途径迁移到TCA循环,从无氧代谢逐渐转移到有氧代谢。  相似文献
