Mixed starter of pure mould and yeast was designed to circument the problem of flavour characteristics in rice wine. The result underlined that the mixed starter (Q7) made by simultaneous inoculation of Rhizopus oryzae, Saccharomyces cerevisiae, Wickerhamomyces anomalus and Candida glabrata had the highest contents of α-amylase (8.02 ± 0.96 U/g) and sensory scoring (92.05 ± 2.24), presenting relatively strong flower-aroma, wine-aroma, Qu-aroma and rice-aroma. Meanwhile, the total concentration of flavour compounds in rice wine (RQ, 125.828 mg L−1) obtained from rice fermentation using Q7 were better than the commercially available rice wine (SS, 125.828 mg L−1). The E-nose analysis showed that the flavour components in RQ differed dramatically from SS. The E-tongue results indicated that the sourness, sweetness and umami aftertaste were greatly higher in RQ, and the bitterness and astringency are lower compared with SS. This study provided a method to improve the flavour and quality of rice wine.  相似文献

Yeast assimilable nitrogen (YAN) concentration and composition impact hydrogen sulphide (H2S) production and fermentation kinetics during wine fermentation, but this phenomenon has not been extensively studied in cider fermentation. Our hypothesis was that H2S production during cider fermentation could be decreased through pre‐fermentation modification of concentrations of individual amino acids. Apple juice (53 mg L−1 YAN) was supplemented with asparagine, arginine, methionine or ammonium and fermented with EC1118 and UCD522 yeast strains. No difference in H2S production among fermentations was observed with addition of asparagine, arginine or ammonium. Methionine addition of 5 mg L−1 decreased H2S production by yeast strain EC1118 at 53 mg L−1 YAN. With 153 mg L−1 initial YAN, only methionine addition of 50 mg L−1 decreased H2S production, and no tested methionine rates decreased H2S production with 253 mg L−1 initial YAN. Supplementation to 153 mg L−1 YAN resulted in increased H2S production at all methionine concentrations tested. Sensory differences in aroma were detected in samples supplemented with ammonium and methionine, and these differences were correlated with observed differences in H2S production. Water kefir microbiota was used to develop novel soy whey-based beverages that have antioxidant activity. In the present study, comparative phenolics, antioxidant and metagenomic composition of the soy whey beverages fermented using three different water kefir microbiota, named WKFS-A, WKFS-B and WKFS-C were investigated. WKFS-B beverage had the highest concentrations of isoflavone aglycones (208.73 ± 2.78 mg L−1) and phenolic acids (132.33 ± 3.41 mg L−1) compared with WKFS-A (193.88 ± 1.15 mg L−1) and (91.73 ± 2.34 mg L−1) and WKFS-C (160.63 ± 1.76 mg L−1) and (97.13 ± 2.63 mg L−1), respectively. The WKFS-B also showed higher DPPH and ABTS radical scavenging activity and ferric reducing antioxidant power compared with WKFS-A and WKFS-C beverages. Microbial species diversity index analysis showed that a higher concentration of isoflavone aglycones, phenolic acids and increased antioxidant activity in the WKFS-B beverage correlates with the higher relative abundance of Lactobacillus genus. This study thus revealed that Lactobacillus dominated water kefir microbiota produces soy whey beverages with high phenolic acids, isoflavone aglycones and antioxidant activity.  相似文献

Pomegranate juices (PJs) were clarified using hydrocolloids [albumin (A), casein (C), chitosan (CH) and xanthan gum (XG)] and in combination with each other. The lowest turbidity was observed in PJs clarified with ‘CH+XG’ and ‘A+C+CH’. Unlike ‘CH+XG’, effects of albumin and its combinations on clarity were mainly related with the contents of ellagic acid (= −0.861), α-punicalagin (= −0.761), rutin (= −0.734), β-punicalagin (= −0.730) and total punicalagin (= −0.713). ‘CH+XG’ caused the highest phenolic (602 mg L−1) and anthocyanin (449 mg L−1) contents, and the second highest Amax (0.811 ABS), resulting from copigmentation of delphinidin-3-glucoside and delphinidin-3,5-diglucoside with CH. Since A showed moderate copigmentation effect, A had lower hyperchromic effect than CH. Moreover, C reduced Amax, anthocyanin content and colour density when combined with other hydrocolloids. As a result, ‘CH+XG’ would be the best combination to obtain high clarity, maintain phenolics and anthocyanins and provide high copigmentation effect in PJ.  相似文献

Binding interactions of human serum albumin (HSA) with sunset yellow (SY) and allura red (AR), two food colorants, were investigated at the molecular level through fluorescence and UV absorption as well as molecular docking. The collective results of the study under the simulated physiological conditions proposed a static type of binding occurring between the two dyes and HSA. When compared with AR (293 K: Ksv = (4.21 ± 0.36) × 104 L·mol−1; Kb = (0.30 ± 0.23) × 106 L·mol−1), SY (293 K: Ksv = (6.80 ± 0.10) × 104 L·mol−1; Kb = (3.11 ± 2.01) × 106 L·mol−1) had stronger quenching ability and higher affinity for HSA due to less steric hindrance. It can be deduced that the energy transfer from HSA to the two dyes occurred with high probability based on the Förster resonance energy transfer theory (r < 7 nm, 0.5 R0 < r < 2.0 R0). The spectral analysis suggested that the formation of the dye-HSA complex resulted in the change in microenvironment around Tyr and Trp residues and in the secondary structure of the protein. According to molecular docking simulation, the two structural analogs almost bound to the same site of HSA, near Sudlow's Site I, but significant difference existed in the number and location of hydrogen bond (H-bond) formed between the dyes and HSA. From the molecular docking along with the thermodynamic parameters (AR: ΔHo = −(58.79 ± 15.24) kJ·mol−1, ΔSo = −(115.1 ± 31.10) J·mol−1·K−1; SY: ΔHo = −(52.24 ± 3.15) kJ·mol−1, ΔSo = −(50.07 ± 11.14) J·mol−1·K−1), it could be inferred that H-bond and van der Waals forces were the major binding forces involved in formation of the dye-HSA complexes.  相似文献

This study deals with the development of a method for total Hg, inorganic Hg and methylmercury (MeHg) determination in red wine by using flow injection-cold vapour generation–inductively coupled plasma mass spectrometry (FI-CVG-ICP-MS) and gas chromatography-ICP-MS (GC-ICP-MS). For Hg speciation analysis, a derivatization step was carried out using a 1% (m/v) sodium tetraphenylborate (NaBPh4) solution, followed by extraction of Hg species and their quantification by GC-ICP-MS. The main parameters evaluated were the make-up gas flow rate, volume of the NaBPh4 solution, time for derivatization reaction/analyte extraction and solvent used for Hg species extraction. Accuracy was evaluated by analyte recovery, whereas recoveries ranged from 99% to 104% for Hg(II) and MeHg. The limits of detection (LODs) for Hg(II) and MeHg were 0.77 and 0.80 μg L−1, respectively. Wine from Argentina, Brazil, Chile and Uruguay were analysed. The wine samples were also acid digested for total Hg determination by FI-CVG-ICP-MS. The LOD of the method used for total Hg determination was 0.01 μg L−1. The concentrations of Hg species in red wine measured by GC-ICP-MS were lower than the respective LODs. Only total Hg was detected in the analysed samples, where the highest concentration of Hg found was 0.55 ± 0.02 μg L−1.  相似文献

The present work discusses the efficiency of pulsed electrical treatments for the inactivation of yeasts. The application of pulsed electric fields (PEFs) and high voltage electrical discharges (HVEDs) as alternatives to sulfites, which are used as anti-microbial to stop the fermentation of sweet white wine, was investigated. The influence of sulfite concentration (from 0 mg·L 1 to 500 mg·L 1), PEF (from 4 kV·cm 1 to 20 kV·cm 1; from 0.25 ms to 6 ms) and HVED (40 kV/cm; 1 ms or 4 ms) treatments on the inactivation of total yeasts and non-Saccharomyces yeasts was determined. The addition of SO2 (250 mg·L 1) resulted in 8 log total yeast reduction. The maximum yeast inactivation obtained with PEF and HVED was respectively 3 and 4 logs. The use of SO2, HVED and PEF allows decreasing the non-Saccharomyces yeast level by 7, 5 and 4 logs respectively. However, the wine browning was less pronounced for the samples treated by PEF in comparison with HVED and SO2 treatments. PEF seems to be the most suitable alternative technique to sulfite addition.  相似文献

This study was conducted aiming at improving the quality of jujube juice by mixed fermentation of lactobacilli. Selection of favourable lactobacilli and addition of nitrogen sources were explored as an efficient method to improve the viability of probiotic in jujube juice. After fermentation, the viability increased to 9.15 ± 0.10 Log CFU mL−1, while the content of lactic acid increased to 5.61 ± 0.03 mg mL−1. The total phenolic and total flavonoid content were 2663.03 ± 11.95 μg mL−1 and 163.95 ± 0.47 μg mL−1 respectively. Moreover, the stability of fermented jujube juice during refrigeration was investigated, which showed that the viability dropped to 8.84 ± 0.6 Log CFU mL−1 and the concentration of lactic acid slowly increased to 6.51 ± 0.04 mg mL−1; the ABTS value showed a 4.26% reduction and FRAP value did not significantly (< 0.05) change during refrigerated storage. In addition to the existing knowledge, our data aid to the future applications of the jujube as a potential ingredient in novel probiotic foods formulation.  相似文献

The content of total polyphenolics, antioxidative capacity and antiproliferative activity were tested in wild and cultivated blackberry pomace. Wild blackberry pomace extract Tw2 showed the highest following contents: total polyphenolics (50.16 mg GAE g−1 dw), flavonoids (7.73 mg Qc g−1 dw), flavonols (6.63 mg Qc g−1 dw) and total monomeric anthocyanins (13.40 mg Cy g−1). Tw2 extract significantly inhibited free radicals: IC50DPPH = 127.76 μg mL−1, IC50ABTS = 26.53 μg mL−1 and IC50 ˙ OH = 168.62 μg mL−1, and the growth of breast adenocarcinoma IC50MCF7 = 306.68 μg mL−1 and cervix epitheloid carcinoma cell lines IC50HeLa = 315.49 μg mL−1. Wild blackberry varieties had higher extraction yields, higher total polyphenolic contents and possessed stronger biological effects compared to cultivated blackberries (P < 0.05). All blackberry extracts showed high biological potential that could be attributed to high total polyphenols and flavonoids content and could be utilised as value-added functional food.  相似文献

The free radical scavenging capacity of selected red, rosé and white Spanish wines from different grape varieties was determined by the 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH·) method using a new methodology developed at our laboratory. The amount of sample necessary to decrease by 50% the initial DPPH· concentration (EC50), the time needed to reach the steady state at EC50 concentration (TEC50) and the antiradical efficiency (AE = 1/EC50TEC50) were determined in the wine samples. Some differences between rosé wines made with Garnacha and Tempranillo grape varieties were observed in the UV‐vis spectra and in the free radical scavenging parameters, those from Garnacha variety having the highest antioxidant activity. The antiradical efficiency of the wines followed a decreasing order: red wines (22.44 × 10−6) > rosé wines (4.90 × 10−6) > white wines (1.88 × 10−6). In this study, the chemical composition, total phenolic content and antioxidant activity (DPPH, ORAC and FRAP assays) of A. subrufescens and P. ostreatus, cultivated with handmade and commercials supplements, were compared. Additionally, the compounds ergosterol, saccharopine, and hexitol were identified in A. subrufescens by HPLC-MS/MS. The antioxidant compound p-coumaric acid and dihexoses was found in both mushroom species. A. subrufescens presented higher total phenolic content (73.8 ± 0.6 mg GAE 100 g−1) and antioxidant activity than P. ostreatus (16.6 ± 0.5 mg GAE 100 g−1). The handmade supplement based on the waste of noble grains presented statistically similar phenolic content to the mushrooms cultivated with commercial ones Spawn Mate II SE (86.1 ± 1.4 and 92.9 ± 0.3 mg GAE 100 g−1, respectively). Therefore, the results support the use of handmade supplements based on agro-wastes as a viable alternative to the use of high-cost commercial ones.  相似文献

BACKGROUND: Mushroom polysaccharides play an important role in functional foods because they exhibit biological modulator properties such as antitumour, antiviral and antibacterial activities. The present study involved the production, purification and characterisation of intracellular and extracellular free and protein‐bound polysaccharides from Pleurotus ostreatus and the investigation of their growth‐inhibitory effect on human carcinoma cell lines. RESULTS: Several fermentation parameters were obtained: batch polysaccharide productivities of 0.013 ± 8.12 × 10?5 and 0.037 ± 0.0005 g L?1 day?1 for intracellular and extracellular polysaccharides respectively, a maximum biomass concentration of 9.35 ± 0.18 g L?1, Pmax = 0.935 ± 0.018 g L?1 day?1, µmax = 0.218 ± 0.02 day?1, YEP/X = 0.040 ± 0.0015 g g?1 and YIP/X = 0.014 ± 0.0003 g g?1. Some polysaccharides exhibited superoxide dismutase (SOD)‐like activity of 50‐200 units. Fourier transform infrared analysis of the polysaccharides revealed absorption bands characteristic of such biological macromolecules. Cytotoxicity assays showed that both intracellular and extracellular polysaccharides exhibited antitumour activity towards several tested human carcinoma cell lines in a dose‐dependent manner. CONCLUSION: The polysaccharides of P. ostreatus exhibited high SOD‐like activity, which strongly supports their biological effect on tumour cell lines. The extracellular polysaccharides presented the highest antitumour activity towards the RL95 carcinoma cell line and should be further investigated as an antitumour agent. This study demonstrates the development of a novel wine lees (WL) based integrated biorefinery for the production of several added-value products. WL were initially fractionated for the production of antioxidants, tartrate and ethanol and the remaining stream was converted into a fermentation nutrient supplement for poly(3-hydroxybutyrate) (PHB) production using the strain Cupriavidus necator DSM 7237. Hydrolysis of pretreated WL was carried out using crude enzyme consortia produced via solid state fermentation of Aspergillus oryzae. Optimization of hydrolysis was based on the enhancement of total Kjeldahl nitrogen to free amino nitrogen (FAN) conversion yield by evaluating the effect of the initial pH value, temperature, initial proteolytic activity and initial WL concentration. WL hydrolysates and crude glycerol were used as nutrient and carbon sources, respectively, in batch and fed-batch fermentations for the production of PHB. Bacterial growth and PHB production were influenced significantly by the FAN content of the WL derived hydrolysates and by the addition of trace elements. Using an initial FAN concentration of 700 mg L 1 and supplementation with trace elements led to the production of 30.1 g L 1 of PHB concentration with an intracellular content of 71.3% (w w-1) and a productivity of 0.56 g L 1 h 1 during fed-batch fermentation.  相似文献

The potential of peat hydrolysates for the production of astaxanthin from Phaffia rhodozyma yeast biomass in continuous fermentation was studied. Chemostat fermentations of the yeast were performed using peat hydrolysates as substrate. Kinetic parameters were calculated for the growth of the yeast and the biomass productivity was determined using mathematical models. A low maximum specific growth rate (μmax=0·08 h−1) and a high Monod constant (Ks=26·2×103) were obtained for this substrate. The low maintenance energy requirement found, 0·020 g (gh)−1, confirms the aerobic metabolism of the yeast with this substrate. Under selected conditions, a biomass productivity of 0·108 g (lh)−1 and an astaxanthin productivity of 0·046 mg (lh)−1 were obtained. The biomass produced had a high protein and astaxanthin content (490±8 g kg−1 and 0.43±0.07 g kg−1, respectively), indicating that it can be used as a feed additive. © 1998 SCI.  相似文献

The total content of phenolic compounds in purple sweet potato (PSP) was determined and the release of such compounds from PSP in gastrointestinal digestion was studied in vitro. The extraction conditions for the maximum recovery of free phenol (FP) and bound phenol (BP) from PSP were determined by response surface methodology (RSM). The maximum recovery of FPPSP was 14.16 ± 0.87 mg GAE per g short for dry weight (DW), which was obtained using 60% (v/v) ethanol maceration with a liquid–solid ratio of 57.21:1 (mL g−1) at 51.93 °C for 2.12 h. The maximum recovery for BPPSP was 7.54 mg GAE per g DW, which was obtained upon hydrolysis with 1.87 mol L−1 NaOH at a liquid–solid ratio of 35.93:1 (mL g−1) for 4.74 h. The maximum phenolic content was released after 1 and 2 h for the in vitro gastric and intestinal digestion respectively. The release of the phenolics was promoted by pepsin and gastric acid during gastric digestion, while it was further promoted by trypsin during intestinal digestion.  相似文献

A strain, Rahnella aquatilis L103, which was isolated from the soil around the roots of mushrooms, produced antibacterial protein though fermentation. This protein has broad antibacterial spectrum towards gram-positive and gram-negative bacteria. The protein has a significant antioxidant capacity on scavenging ABTS (82.5%, 120 μg mL−1), DPPH· (64.1%, 600 μg mL−1) and OH· (60.1%, 750 μg mL−1) as well. The protein was considered to be safe within concentration of 150 μg mL−1 as shown by MTT results and was applied to beef refrigerated preservation. The treatment group presented lower total viable counts (TVC), total volatile basic nitrogen (TVB-N), 2-thiobarbituric acid (TBA), meanwhile, higher sensory score and longer shelf life (2 days) compared with the control group (P < 0.05). The results indicated that the protein under safe concentration was beneficial to beef preservation.  相似文献

Rice bran, the industry byproduct, contains high levels of phenolic compounds. Based on glycerol, a green recovery of phenolics from rice bran was proposed. Extraction temperature (X1), glycerol concentration (X2) and liquid-to-solid ratio (X3) were optimised by response surface methodology. Basic physical properties, that is, viscosity, conductivity, density were analysed and UPLC-Triple-TOF/MS method was applied for phenolic identification. Results showed the optimum conditions were X1 = 66.76 °C, X2 = 19.47% and X3 = 32.92 mL g−1. Under these conditions, the recovery yield in total polyphenols was 708.58 ± 12.36 mg GAE/100 g DW. Moreover, the recovery capacity of phenolic compounds was significantly correlated with viscosity, conductivity and density of glycerol solution. Under the optimal conditions, totals of thirteen phenolic compounds were identified and phenolic acids (4-hydroxybenzoic acid, vanillic acid, syringic acid, 4-coumaric acid, ferulic acid and salicylic acid) were quantified. These results demonstrate glycerol can serve as an ideal green solvent to recover phenolic compounds from rice bran.  相似文献

Bioactive compounds from Brazilian hop (Humulus lupulus L.) cultivars were extracted by ultrasound and their phenolic profile compared with commercial hop from the USA. The most effective extraction conditions (solution of ethanol 49%, at 52 °C and a solid/liquid ratio of 1 g per 34 mL) for the total phenolic compounds (TPC) were determined using a Central Composite Rotatable Design. The Brazilian hop showed higher content of TPC (33.93 ± 0.67 mg GAE g−1), total flavonoids (54.47 ± 0.10 mg QE g−1) and higher antioxidant activity (ABTS: EC50 21.29 ± 1.36 μL mL−1; DPPH: EC50 3.91 ± 0.17 μL mL−1) when compared with the USA hop. The main phenolic compounds present in the extracts were the flavonoids isoquercitrin and quercetin. The antioxidant properties of the Brazilian hop extract had not been reported yet in the literature for this raw material, thus showing potential to be incorporated in polymeric films used as active packaging.  相似文献

Cyclopropane fatty acids, which can be simply converted to methylated fatty acids, are good unusual fatty acid candidates for long-term resistance to oxidization and low-temperature fluidity useful for oleochemistry and biofuels. Cyclopropane fatty acids are present in low amounts in plants or bacteria. In order to develop a process for large-scale biolipid production, we expressed 10 cyclopropane fatty acid synthases from various organisms in the oleaginous yeast Yarrowia lipolytica, a model yeast for lipid metabolism and naturally capable of producing large amounts of lipids. The Escherichia coli cyclopropane fatty acid synthase expression in Y. lipolytica allows the production of two classes of cyclopropane fatty acids, a C17:0 cyclopropanated form and a C19:0 cyclopropanated form, whereas others produce only the C17:0 form. Expression optimization and fed-batch fermentation set-up enable us to reach a specific productivity of 0.032 g·L−1·hr−1 with a genetically modified strain containing cyclopropane fatty acid up to 45% of the total lipid content corresponding to a titre of 2.3 ± 0.2 g/L and a yield of 56.2 ± 4.4 mg/g.  相似文献

Daqu, a saccharifying and fermenting agent for the production of Chinese vinegar and liquor, is manufactured through a spontaneous solid-state fermentation process (SSF). To investigate the influence of bioaugmentation with native microorganisms on Daqu SSF process, physicochemical properties, microbial biomass, volatile compounds and microbial communities were analysed in laboratory-scale Daqu. The results showed that the amylase activity of Daqu enhanced 6.35% ± 0.74% by inoculating Bacillus amyloliquefaciens, Saccharomycopsis fibuligera and Absidia corymbifera. Obviously higher microbial richness was found in the Daqu with bioaugmentation, although the microbial community structure remained relatively stable. Moreover, a total of twenty-two volatile compounds were detected in the mature Daqu. Slightly increase in the content of alcohols was found in the enhanced group by fungal inoculation, including ethanol (46.58 ± 1.36 mg kg−1), isobutanol (0.19 ± 0.04 mg kg−1) and isoamyl alcohol (1.55 ± 0.13 mg kg−1). This study demonstrated that bioaugmentation had a positive effect on the amylase activity, main volatile compounds and microbial community richness.  相似文献
