The free and bound phenolic compounds in 10 common Chinese edible flowers were investigated using reversed phase high‐performance liquid chromatography. Their antioxidant capacities were evaluated using 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical‐scavenging activity, 2,2'‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulphonic acid) (ABTS) radical‐scavenging activity, oxygen radical absorption capacity (ORAC), ferric reducing antioxidant power (FRAP), and cellular antioxidant activity (CAA). Free factions were more prominent in phenolic content and antioxidant capacity than bound fractions. Paeonia suffruticosa and Flos lonicerae showed the highest total phenolic content (TPC) 235.5 mg chlorogenic acid equivalents/g of dry weight and total flavonoid content 89.38 mg rutin equivalents/g of dry weight. The major phenolic compounds identified were gallic acid, chlorogenic acid, and rutin. P. suffruticosa had the highest antioxidant capacity in the DPPH, ABTS, and ORAC assays, which were 1028, 2065, 990 μmol Trolox equivalents/g of dry weight, respectively, whereas Rosa chinensis had the highest FRAP value (2645 μmol Fe2+ equivalents /g of dry weight). The P. suffruticosa soluble phenolics had the highest CAA, with the median effective dose (EC50) 26.7 and 153 μmol quercetin equivalents/100 g of dry weight in the phosphate buffered saline (PBS) and no PBS wash protocol, respectively. TPC was strongly correlated with antioxidant capacity (R = 0.8443 to 0.9978, P < 0.01), which indicated that phenolics were the major contributors to the antioxidant activity of the selected edible flowers.  相似文献

Apples (Malus domestica L.) are the most common source of phenolic compounds in northern European diet. Besides pectins, dietary fibers, vitamins, and oligosaccharides they contain phenolic compounds of different classes. Apple powders are convenient functional forms retaining significant amounts of phenolic antioxidants. In this study reducing and radical scavenging profiles of freeze‐dried powders of “Aldas,? “Auksis,? “Connel Red,? “Ligol,? “Lodel,? and “Rajka? were determined and phenolic constituents were identified using ultra high‐performance liquid chromatography coupled to quadrupole and time‐of‐flight mass spectrometers. A negative ionization mode was applied and seventeen compounds: phenolic acids (coumaroylquinic, chlorogenic), flavonoids (quercetin derivatives), and procyanidin derivatives (B1, B2, and C1) were identified in all tested apple samples. Total values of Trolox equivalents varied from 7.72 ± 0.32 up to 20.02 ± 0.52 and from 11.10 ± 0.57 up to 21.42 ± 0.75 μmol/g of dry weight of apple powder in FRAP (ferric reducing antioxidant power) and ABTS (2,2‐azinobis‐(3‐ethylbenzothiazoline‐6‐sulfonic acid) postcolumn assays, respectively. The greatest Trolox equivalent values were determined for apples of “Aldas? cultivar. Chlorogenic acid and procyanidin C1 were the most significant contributors to total reducing and radical scavenging activity in all apple cultivars tested, therefore they could be considered as markers of antioxidant activity.  相似文献

Eleven organically grown apple cultivars and 11 apple cultivars of integrated production from Austria and Slovenia were analyzed by HPLC for the content of phenolic compounds in peel and pulp. We identified chlorogenic acid, p‐coumaric acid, procyanidin B3, protocatechuic acid, (?)‐epicatechin, phloridzin, rutin and quercetin‐3‐rhamnoside in apple peel. In apple pulp, (+)‐catechin was also identified in all the cultivars. Some other phenols (procyanidin B3, rutin and quercetin‐3‐rhamnoside) could not be identified or were not properly separated. With regard to the phenolic content in the apple peel, there were no differences between organically grown apple cultivars and apple cultivars of integrated production. Organically grown apples, however, exhibited a higher content of phenolic substances in the apple pulp compared with the apple cultivars of integrated production. This may be due either to the different genotype source or to the growing technology. Higher concentrations of phenolic compounds in organically grown cultivars could be a result of plant response to stress. The apple peel contained higher concentrations of identified phenols than the pulp. The apple peel represents up to 10% of the whole fruit; therefore the phenolsic compounds in the pulp are of greater importance to the consumer than the phenolic compounds in the peel. BACKGROUND: Antioxidant and antimicrobial potentials of Serbian red wines produced from different international Vitis vinifera grape varieties and their correlation with contents of phenolic compounds were studied by spectrophotometric and chromatographic methods. The antioxidant activity of red wines was estimated through their ability to scavenge 2,2′‐diphenyl‐1‐picrylhydrazyl free radical (DPPH?). The red wines, gallic acid, (+)‐catechin and quercetin were screened in vitro for antimicrobial activity against Gram‐positive and Gram‐negative strains using microdilution and disc diffusion techniques. RESULTS: Excellent correlations between the contents of quercetin‐3‐glucoside (R2 = 0.9463) and quercetin (R2 = 0.9337) and DPPH?‐scavenging ability of the red wines were found. Serbian red wines exhibited significant activity against Staphylococcus aureus, Listeria inocua, Micrococcus flavus, Sarcina lutea, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Salmonella enteritidis and Shigella sonnei strains, which was in correlation with their phenolic composition and antioxidant activity. The compounds gallic acid, quercetin and (+)‐catechin showed high activity against B. subtilis, S. aureus, S. lutea and M. flavus Gram‐positive and S. enteritidis and P. aeruginosa Gram‐negative strains. CONCLUSION: The results show that quercetin‐3‐glucoside and quercetin concentrations can be used as markers for the determination of antioxidant and antimicrobial potentials of red wines. BACKGROUND: The phenolic profile, determined by the relative proportions of different phenolic compounds, is characteristic for each grape variety and its corresponding wine. The aim of this study was to determine the phenolic and hydroxycinnamate acid, flavan‐3‐ol and flavonol contents and free radical‐scavenging activity of single‐cultivar (Cabernet Sauvignon) wines from selected Balkan vineyard regions by spectrophotometric methods and high‐performance liquid chromatography with photodiode array and fluorescence detection. RESULTS: The contents of phenolic compounds varied depending on the agroclimatic factors and oenological practices of the vineyard region. The antioxidant activity of wine samples was estimated by their ability to scavenge the stable 2,2′‐diphenyl‐1‐picrylhydrazyl free radical (DPPH). All wines showed high DPPH‐scavenging activity (70.03–83.53%, mean 73.76%). Significant correlations between catechin (R2 = 0.8504) and quercetin (R2 = 0.8488) concentrations and DPPH‐scavenging ability of the wines were found. CONCLUSION: The concentrations of the main components catechin and quercetin can be used as biochemical markers for the authentication of red grape cultivars and their corresponding single‐cultivar wines. BACKGROUND: The presence of phenolics in fruit, red wine and vinegar has positive health effects due to their significant antioxidant activity. The aim of the study was to determine the effects of two different vinegar production methods on antioxidant activity and phenolic level of vinegars derived from Ulugbey Karasi grapes. Traditional surface and industrial submerge methods were used to make vinegar. Samples were taken from fresh red grape juice, maceration, wine, traditional vinegar and industrial vinegar. RESULTS: Total phenolic content of traditional and industrial vinegar samples were 2690 mg L?1 and 2461 mg L?1 GAE, respectively. ORAC values of traditional and industrial vinegar samples were 10.50 µmol mL?1and 8.84 µmol mL?1 TE, respectively. Antioxidant activity values of traditional and industrial vinegars were 13.50 mmol L?1 and 10.37 mmol L?1 TEAC, respectively. Gallic acid, catechin, epicatechin, chlorogenic acid, caffeic acid, syringic acid, p‐coumaric acid and ferulic acid were detected in grape juice, wine and vinegar samples. The content of catechin in industrial vinegar (27.50 mg L?1) was significantly higher than that of in traditional vinegar (13.76 mg L?1) (P < 0.05). Traditional vinegar had higher amounts of chlorogenic and syringic acids than the industrial vinegar (P > 0.05). Eight phenolic compounds including: p‐coumaric acid, vanillic acid, caffeic acid, chlorogenic acid, trolox, quercetin, curcumin, and resveratrol were treated with riboflavin (RF) photosensitization and in vitro antioxidant capacities of the mixtures were determined by 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH), 2,2’ azino bis(3‐ethylbenzothiazoline‐6‐sulphonic acid) (ABTS), and ferric reducing antioxidant power (FRAP) assays. Mixtures containing p‐coumaric acid and vanillic acid under RF photosensitization showed increases in ferric ion reducing ability and radical scavenging activity of DPPH, whereas mixtures of other compounds had decreases in both radical scavenging ability and ferric reducing antioxidant power. Hydroxycoumaric acid and conjugated hydroxycoumaric and coumaric acids were tentatively identified from RF photosensitized p‐coumaric acid, whereas dimmers of vanillic acid were tentatively identified from RF photosensitized vanillic acid. RF photosensitization may be a useful method to enhance antioxidant properties like ferric ion reducing abilities of some selected phenolic compounds.  相似文献

Nine commercial varieties of tomato (Rambo, Senior, Ramillete, Liso, Pera, Canario, Durina, Daniella and Remate) produced in Spain were analysed for their lycopene content, content of phenolic compounds and antioxidant capacity. The phenolic compounds were characterised as flavonoids (quercetin, kaempferol and naringenin) and hydroxycinnamic acids (caffeic, chlorogenic, ferulic and p‐coumaric acids). Antioxidant activity was measured using the DPPH and ABTS assays. The concentrations of lycopene and the various phenolic compounds as well as the antioxidant activity were significantly influenced by the tomato variety. Quercetin, the most abundant flavonoid, was found in concentrations ranging between 7.19 and 43.59 mg kg?1 fresh weight, while naringenin levels were lower than 12.55 mg kg?1. The most abundant hydroxycinnamic acid was chlorogenic acid, with values ranging from 14 to 32 mg kg?1 fresh weight, followed by caffeic acid, while p‐coumaric and ferulic acids showed similar concentrations lower than 5 mg kg?1. The highest content of lycopene was found in Ramillete, Pera and Durina (>50 mg kg?1 fresh weight), while the concentration in the other varieties was between 50 and 30 mg kg?1, with the exception of Liso (less than 20 mg kg?1). The antioxidant activity of tomato extracts varied with the tomato variety and the assay method used. Individual compounds found to be significantly related to antioxidant capacity were lycopene and ferulic and caffeic acids, but not quercetin and chlorogenic acid. © 2002 Society of Chemical Industry  相似文献

The high‐acid apple (Malus sieversii f. niedzwetzkyana (Dieck) Langenf) possessing deep red peel and light red flesh is widely distributed in China. To determine if a useful apple variety is being ignored, the monomeric phenolic profiles and antioxidant activities of peel, flesh, pomace, whole fruit and juice were evaluated. The results were compared to those of ‘Fuji’ and ‘Granny Smith’ apples. The high‐acid apple possessing the highest total phenolic and anthocyanin contents among three apple varieties contained the most chlorogenic acid, phloridzin, epicatechin, ferulic acid, catechin, vanillic acid, p‐coumaric acid and caffeic acid. The extraction yields of phenolics (64%) and anthocyanins (89%) from the high‐acid apple to juice were the highest. The high‐acid apple possessing the greatest antioxidant activity had the strongest antihaemolysis activity, indicating that it could be used to produce juice and value‐added ingredient to assist in the prevention of chronic disease.  相似文献

BACKGROUND: Two varieties of Origanum majorana (Canadian and Tunisian) were evaluated for their phenolic, flavonoid and tannin contents, individual phenolic compounds and antioxidant activities under NaCl constraint. RESULTS: The results showed a significant variability in phenolic composition and antioxidant behavior between the two varieties under salt stress. The phenolic composition of methanolic extracts was determined by reversed‐phase high‐performance liquid chromatography. Amentoflavone was the predominant flavonoid compound; in addition, trans‐2‐hydrocinnamic acid became the major phenolic acid with salt treatment of the Tunisian variety. In the control, Canadian variety extract was characterized by high levels of gallic acid and amentoflavone. However, under 75 mmol L?1 NaCl, gallic acid content doubled, whereas amentoflavone content was maintained in the Canadian variety. Stimulation of phenolic acid biosynthesis was observed in these two varieties under salt treatment despite the fact that shoots of the Tunisian variety showed higher antioxidant activities compared to those from the Canadian variety. Tunisian O. majorana might have developed tolerance to salinity and avoided tissue damage by activating enzymes involved in the galactosylation of quercetin into quercetin‐3‐galactoside and quercetin‐3‐rhamnoside. The phenolics in 20 apple cultivars comprising 19 English cider apple varieties and one dessert apple variety were analysed by high‐performance liquid chromatography/tandem mass spectrometry. The cider varieties contained higher levels than the dessert apple and the peel was richer in phenolics than the flesh. The phenolic concentrations ranged between 230 and 4920 mg kg?1 fresh weight in the flesh and between 546 and 6306 mg kg?1 fresh weight in the peel. Fifteen compounds from five different phenolic groups, flavan‐3‐ols, flavonols, anthocyanins, hydroxycinnamates and dihydrochalcones, were detected. The major components in the flesh were 5‐O‐caffeoylquinic acid, procyanidin B2 and (?)‐epicatechin, while (?)‐epicatechin and quercetin glycosides predominated in the peel. In this study, fifty‐one monofloral Sardinian honeys from ten various floral origins were screened for their phenolic content, antioxidant activity, colour and electrical conductivity. The total phenolic amounts have been evaluated by Folin–Ciocalteu method, whereas quantification of several phenolic compounds (phenolic acids and flavonoids) has been carried out by HPLC‐DAD technique. The richest sample in phenolic compounds resulted strawberry tree honey with about 40 mg GAE/100 g, as well FRAP test and DPPH˙ test confirm that antioxidant activity of strawberry tree honey extract exceed both honey extracts and synthetic antioxidants like BHA and BHT. Among the studied phenolic compounds a total of five phenolic acids (ferulic, syringic, trans‐cinnamic, chlorogenic and p‐hydroxycinnamic) and nine flavonoids (catechin, kaempferol, rutin, quercetin, luteolin, apigenin, galangin, pinocembrin and pinobanksin) were identified. Our results show good correlations between total polyphenol amount and antioxidant activity and between colour and electrical conductivity.  相似文献

Black rice bran contains phenolic compounds of a high antioxidant activity. In this study, the 40% acetone extract of black rice bran was sequentially fractionated to obtain 5 fractions. Out of the 5 fractions, ethyl acetate fraction was subfractionated using the Sephadex LH‐20 chromatography. The antioxidant activity of phenolic compounds in the extracts was investigated by 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical assay, 2,2‐azino‐bis‐(3‐ethylenebenzothiozoline‐6‐sulfonic acid) (ABTS) radical cation assay, reducing power. The subfraction 2 from ethyl acetate fraction had the highest total phenolic contents (TPC) (816.0 μg/mg) and the lowest EC50 values (47.8 μg/mL for DPPH radical assay, 112.8 μg/mL for ABTS radical cation assay, and 49.2 μg/mL for reducing power). These results were 3.1, 1.3, and 2.6 times lower than those of butylated hydroxytoluene (BHT), respectively. At a concentration of 100 μg/mL, the antioxidant activity and TPC of various extracts was closely correlated, with correlation coefficients (R2) higher than 0.86. The major phenolic acid in subfraction 2 was identified as ferulic acid (178.3 μg/mg) by HPLC and LC‐ESI/MS/MS analyses. Our finding identified ferulic acid as a major phenolic compound in black rice bran, and supports the potential use of black rice bran as a natural source of antioxidant.  相似文献

The effect of instant controlled pressure drop‐assisted hot air drying (AD‐DIC) on browning‐related components in apple slices was studied. Results showed the AD temperature had no significant effect on colour index of final apple slices. The aspartic acid, glutamic acid, lysine, histidine, arginine, sucrose and dehydroascorbic acid (DHAA) in apple slices increased firstly during the AD process and then decreased during DIC process. The fructose, glucose, reduced ascorbic acid and major phenolic compounds reduced with the abrupt increase of 5‐hydroxymethylfurfural (5‐HMF) during AD‐DIC. There were significant correlations (< 0.05) among moisture content (w.b.), water activity (aw), water state, reduced ascorbic acid, DHAA, chlorogenic acid, procyanidin B1, (+)‐catechin, (?)‐epicatechin and phlorizin during AD‐DIC. The colour difference showed that the major browning reaction of apple slices during the predrying process was enzymatic browning, while nonenzymatic browning played a crucial role on green‐red value (CIE a* value) during DIC.  相似文献

In this study, total phenolic contents and antioxidant activities of grape seed extracts obtained from twelve different grape seeds from common varieties grown in Turkey were determined. Grape seeds were extracted with 70% acetone and extraction yield of grape seed were calculated. The total phenolic content of grape seed extracts were determined by the Folin‐Ciocalteu procedure and ranged from 33 945 to 58 730 mg per 100 g extract as gallic acid equivalent. Antioxidant activities of grape seed extracts with two different free radical scavenging methods, ABTS [2,2/‐azinobis (3‐ethylbenzothiazoneline‐6‐sulfonic acid)] and DPPH (2,2‐diphenyl‐picrylhydrazyl) assays, using Trolox equivalent as standards, were investigated. Grape seed extracts exhibited antioxidant activities 2.46–4.14 and 3.55–5.76 [Trolox equivalent antioxidant capacities (TEAC) mg?1 extracts] in ABTS and DPPH assays, respectively. Compared with varieties, Muskule extracts exhibited the lowest total phenolic content, TEACABTS and TEACDPPH value while Narince extracts had the highest total phenolic content and TEACDPPH value, and Alphonse Lavalleé had the highest TEACABTS value. Total phenolic content showed that there is a significant correlation with TEACDPPH (r = 0.7974, P ≤ 0.001) and TEACABTS values (r = 0.4860, P ≤ 0.05).  相似文献

In this study, in order to evaluate the in vitro bioaccessibility of fresh and sun‐dried figs, total antioxidant capacity (TAC), total proanthocyanidin content (TPA) and the major phenolic compounds were determined at different phases of simulated gastrointestinal (GI) tract digestion for Sarilop and Bursa siyahi fig varieties. Four major phenolic compounds (chlorogenic acid, rutin, cyanidin‐3‐glucoside (C3G) and cyanidin‐3‐rutinoside (C3R)) were investigated for GI tract digestion. The results of in vitro GI tract digestion revealed that the dialysed fraction (IN) represented 9–26% and 1–22% of the initial TAC of the whole‐fresh yellow and purple figs, respectively. Moreover, in case of 2,2‐azinobis(3‐ethylbenzothiazoline)‐6‐sulphonic acid (ABTS), TPA and chlorogenic acid contents, drying caused an increase in the IN fraction of yellow figs (38, 140, 50%, respectively). The bioaccessibility of C3G and C3R were quite low for fresh figs (0–5% of the initial values), whereas for dried figs, anthocyanins were not detected at all in the IN fraction.  相似文献

用高效液相色谱仪测定了不同品种苹果果实绿原酸、儿茶素、表儿茶素、根皮素和槲皮素等主要酚类物质的含量。结果表明,不同品种苹果果肉酚类物质总量以富士和新红星最高,金冠最低。富士苹果果肉酚类物质以绿原酸为主,新红星和王林苹果果肉以表儿茶素为主。不同品种苹果果皮酚类物质总量以新红星最高,各品种苹果果皮酚类物质总量均高于果肉。果皮中酚类物质以黄烷-3-醇类为主。  相似文献

Mung bean grains exhibit high level of antioxidant activity due the presence of phenolic compounds. Near‐infrared spectroscopy (NIRS), in conjunction with chemometrics, was used to develop a rapid, nondestructive, chemical free and easy‐to‐use method to determine these compounds in sixty genotypes of mung bean. NIRS calibration curve with high‐performance liquid chromatography (HPLC) as reference method was used to determine phenolic compounds (catechin, chlorogenic acid, caffeic acid, p‐coumaric acid, t‐ferulic acid, vitexin, isovitexin, myricetin, quercetin and kaempferol). It was observed that partial least square regression (PLSR) model in the wavelength range of 1600–2500 nm with standard normal variate (SNV) and linear baseline correction (LBC) as preprocessing techniques can measure phenolic compound accurately (R2 > 0.987) with root‐mean‐square error less than 1.82%. This study shows that NIRS along with chemometrics is an accurate method to estimate the phenolic compounds rapidly and nondestructively.  相似文献

An apple puree formulated with red grape skins was developed on pilot scale as a new beverage with antiglycation properties. The addition level of grape skins was selected by a liking test with 70 consumers. The selected formulation was a fibre‐rich source and delivered grape anthocyanins, flavonols and flavanols resulting in ~ twofold higher antiglycation activity than the apple puree. Pasteurisation (3‐D in the target microorganism Alicyclobacillus acidoterrestris) did not affect the antiglycation activity, which decreased by 30% upon sterilisation. Storage for 1 month in the temperature range 15–35 °C affected the contents of anthocyanins, monomeric, dimeric and oligomeric flavanols, while chlorogenic acid, flavonols and dihydrochalcones were stable. About 90% antiglycation activity was retained after one‐month storage at 15 °C. The use of red grape skin as ingredient could represent an opportunity for the apple processing industry to develop a value‐added product.  相似文献

A process for the combined recovery of pectin and phenolic compounds from apple pomace, the primary by-product of apple juice production, was developed. The process includes extraction of dried apple pomace with diluted mineral acid and adsorption of phenolic constituents by a hydrophobic styrene–divinylbenzene copolymerisate. After elution with methanol, the polyphenolics were concentrated in vacuo, stabilised by lyophilisation, and characterised by high-performance liquid chromatography. The predominant compounds were phloridzin, chlorogenic acid and quercetin glycosides. Adsorptive removal especially of oxidised phenolic compounds led to a considerable decolourisation of the pomace extracts, as revealed by determination of L*a*b* values, hue angle and chroma. Gelling properties of pectin were not adversely affected. While the polyphenolics recovered from apple pomace may be used as natural antioxidants or as functional food ingredients, extended fields of application may be obtained for decolorised, refined apple pectins. Furthermore, investigations on the phenolic composition of several New Zealand apple cultivars, of apple seeds, and on the effects of pomace drying on the stability of polyphenolics were carried out.  相似文献
