This study was undertaken to examine the chemical compositions of essential oil and tested the efficacy of oil and organic extracts from seeds of Zizyphus jujuba against food-borne pathogens. The chemical compositions of the oil was analysed by Null. Twenty three compounds representing 91.59% of the total oil were identified. The oil (5 μl of 1:5 (v/v) dilution of oil with methanol) and extracts of hexane, chloroform, ethyl acetate and methanol (300 μg/disc) of Z. jujuba displayed a remarkable antibacterial activity against Staphylococcus aureus (ATCC 6538 and KCTC 1916), Listeria monocytogenes ATCC 19166, Bacillus subtilis ATCC 6633, Pseudomonas aeruginosa KCTC 2004, Salmonella typhimurium KCTC 2515 and Escherichia coli ATCC 8739. The scanning electron microscopic studies also demonstrated the effect of essential oil on the morphology of Staph. aureus ATCC 6538 at the MIC value, along with the potential effect on cell viabilities of the tested bacteria.  相似文献

This study determined the extent that irradiation of fresh beef surfaces with an absorbed dose of 1 kGy electron (e-) beam irradiation might reduce the viability of mixtures of O157 and non-O157 verotoxigenic Escherichia coli (VTEC) and Salmonella. These were grouped together based on similar resistances to irradiation and inoculated on beef surfaces (outside flat and inside round, top and bottom muscle cuts), and then e-beam irradiated. Salmonella serovars were most resistant to 1 kGy treatment, showing a reduction of ≤ 1.9 log CFU/g. This treatment reduced the viability of two groups of non-O157 E. coli mixtures by ≤ 4.5 and ≤ 3.9 log CFU/g. Log reductions of ≤ 4.0 log CFU/g were observed for E. coli O157:H7 cocktails. Since under normal processing conditions the levels of these pathogens on beef carcasses would be lower than the lethality caused by the treatment used, irradiation at 1 kGy would be expected to eliminate the hazard represented by VTEC E. coli.  相似文献

In the last two decades several foodborne disease outbreaks associated with produce were reported. Tomatoes, in particular, have been associated with several multi-state Salmonella outbreaks. Inactivation of inoculated Escherichia coli O157:H7, Listeria monocytogenes, Salmonella enterica and Shigella flexneri on whole Roma tomato surfaces by X-ray at 0.1, 0.5, 0.75, 1.0, and 1.5 kGy was studied. The main purpose of this study was to achieve a 5 log reduction in consistent with the recommendations of the National Advisory Committee on Microbiological Criteria for Foods. Moreover, the effect of X-ray on inherent microflora (mesophilic counts, psychrotrophic counts and yeast and mold counts) of untreated and treated Roma tomatoes, during storage at ambient temperature (22 °C) for 20 days was also determined. Mixtures of three or two strains of each tested organism was spot inoculated (100 μl) onto the surface of Roma tomatoes (approximately 7–9 log per tomato), separately, and air-dried, followed by treatment with X-ray doses at 22 °C and 55–60% relative humidity. Surviving bacterial populations on tomato surfaces were evaluated using a nonselective medium (tryptic soy agar) with a selective medium overlay for each bacteria; E. coli O157:H7 (CT-SMAC agar), L. monocytogenes (MOA), and S. enterica and S. flexneri (XLD). Treatment with X-ray significantly reduced the population of the tested pathogens on whole Roma tomato surfaces, compared with the control. Approximately 4.2, 2.3, 3.7 and 3.6 log CFU reduction of E. coli O157:H7, L. monocytogenes, S. enterica and S. flexneri per tomato were achieved by treatment with 0.75 kGy X-ray, respectively. More than a 5 log CFU reduction per tomato was achieved at 1.0 or 1.5 kGy X-ray for all tested pathogens. Furthermore, treatment with X-ray significantly reduced the inherent microflora on Roma tomatoes. The effectiveness of electron beam irradiation and high pressure treatment for the sanitation of cold-smoked salmon from two points of view, microbial safety and shelf-life extension, was compared. From the response of L. monocytogenes INIA H66a to irradiation, a D value of 0.51 kGy was calculated. For samples stored at 5 °C, 1.5 kGy would be sufficient to attain a Food Safety Objective (FSO) of 2 log10cfu/g L. monocytogenes for a 35-day shelf-life, whereas 3 kGy would be needed in the case of a temperature abuse (5 °C + 8 °C). Pressurization at 450 MPa for 5 min was considered to be an insufficient treatment, since the FSO of 2 log10cfu/g L. monocytogenes was only attained for a shelf-life of 21 days at 5 °C. However, treatment at 450 MPa for 10 min achieved this FSO for samples held during 35 days at 5 °C, or during 21 days under temperature abuse (5 °C + 8 °C) conditions. Irradiation at 2 kGy kept the microbial population of smoked salmon below 6 log10cfu/g after 35 days at 5 °C, with negligible or very light changes in its odor. Pressurization at 450 MPa for 5 min also kept the microbial population below 6 log10cfu/g after 35 days at 5 °C and did not alter odor, but affected negatively the visual aspect of smoked salmon.  相似文献

The behaviour of Listeria monocytogenes and Staphylococcus aureus in vacuum-packed cooked ham slices treated with an electron beam and stored at 4, 7 and 10 °C was investigated. Cooked ham slices were inoculated with L. monocytogenes and S. aureus and electron beam treated at 2 and 3 kGy. After treatment, a long temperature-dependent death phase was observed, followed by growth at a slower rate than in untreated samples. Assuming a hypothetical load of 10 cells/g or cm2 of L. monocytogenes and S. aureus as an original contamination in an industrial situation, an E-beam treatment of vacuum-packed cooked ham slices at 2 kGy guarantees the microbiological safety of the product along its shelf life, even if a noticeable temperature (10 °C) abuse occur during its storage period. Likewise, the E-beam treatment gave rise to a substantial increase of the RTE cooked ham shelf life off-sensory features associated to the spoilage only were detected in non-treated samples (controls) after 8 and 18 days of storage at 10 °C and 7 °C, respectively.  相似文献

Bdellovibrio-and-like organisms (BALOs) are a group of highly motile delta-proteobacteria that prey on other gram-negative bacteria. However, nothing is known of the application potential of marine BALOs in safeguarding seafood safety. Here, biological characterization of two marine BALOs strains and their application in the elimination of Vibrio parahaemolyticus in oyster (Crassostrea ariakensis) at the laboratory scale were investigated.BALOs strains BDH12 and BDHSH06 were isolated from sediment of Daya bay in Shenzhen of China, with Shewanella putrefaciens strain 12 and V. parahaemolyticus strain SH06 as preys, respectively, when using double layer agar technique. They were identified as BALOs morphologically by transmission electron microscopy, while partial 16S rDNA sequencing analysis revealed that they showed no close relationships with members of the known genera Bdellovibrio, Bacteriolyticum, Bacteriovorax, or Peredibacter.Biological characterizations revealed that both strains had the optimal pH, salinity and temperature at 7.2, 3% and 30 °C, correspondingly. They could not utilize autoclaved, dead cells as hosts. Prey range analysis revealed that individually, BDH12 and BDHSH06 lysed 82.5% (47 strains) and 84.2% (48 strains) of the total 57 preys tested respectively. In combination, they lysed 98.2% (56 of 57) strains. All strains of V. parahaemolyticus, Vibrio cholerae and Vibrio alginolyticus tested could be lysed by both strains.A 7-day laboratory-scale V. parahaemolyticus elimination experiment in oyster showed that in the control, the cell counts of total vibrios and V. parahaemolyticus strain Vp plus in water and in oyster intestines were on the rise, whereas in the BALOs treated groups, their numbers were down from 8.09 ± 0.05 log CFU/ml and 8.02 ± 0.04 log CFU/ml to 2.39 ± 0.01 log CFU/ml and 2.33 ± 0.01 log CFU/ml, respectively. The same patterns could also be observed in oyster intestines. Results of this study indicate the feasibility of using BALOs to biologically control or even eliminate V. parahaemolyticus in seafood oyster.  相似文献

The objectives of the present work were to assess the use of moderate doses of gamma irradiation (2 to 5 kGy) and to reduce the risk of pathogen presence without altering the quality attributes of bovine trimmings and of patties made of irradiated trimmings. Microbiological indicators (coliforms, Pseudomonas spp and mesophilic aerobic counts), physicochemical indicators (pH, color and tiobarbituric acid) and sensory changes were evaluated during storage. 5 kGy irradiation doses slightly increased off flavors in patties. Two pathogenic markers (Listeria monocytogenes and Escherichia coli O157:H7) were inoculated at high or low loads to trimming samples which were subsequently irradiated and lethality curves were obtained. Provided that using irradiation doses ≤ 2.5 kGy are used, reductions of 2 log CFU/g of L. monocytogenes and 5 log CFU/g of E. coli O157:H7 are expected. It seems reasonable to suppose that irradiation can be successfully employed to improve the safety of frozen trimmings when initial pathogenic bacteria burdens are not extremely high.  相似文献

The salted and dried product of tuna roe (bottarga) is a seafood characteristic of the Mediterranean area and exported all over the world. Samples of bottarga from bluefin tunas (Thunnus thynnus, L.) caught in the southwest Mediterranean sea were analysed. The samples were characterised by high content of marine wax esters (55–67 mol% of lipid classes), of docosahexaenoic (22:6 n-3, 25 w%) and oleic (18:1 n-9, 19 w%) fatty acids. Cholesterol was detected as 7–9 w% of lipids. Free fatty acids, index of lipid hydrolysis, represented 32–39 mol% over total fatty acids. Among metabolites, nutrients as taurine, nicotinamide and β-alanine, were found. The microflora comprised staphylococci, enterococci (2.2 log10 CFU/g) and lactic acid bacteria (3 log10 CFU/g). The food-borne pathogens Staphylococcus aureus, Listeria monocytogenes and Salmonella spp. were not detected. These findings indicate tuna bottarga as valuable source of nutrients.  相似文献

Feta, a white brine cheese, was produced and contaminated with Listeria monocytogenes. Contamination occurred either at the beginning (pre-process contamination) or at the end of Feta manufacturing (post-process contamination). In the first case the milk was contaminated with 103 cfu/ml, and 2 months later, in the final product, the L. monocytogenes population was approximately 105 cfu/g. In the second case, the brine (NaCl, 7% w/v), in which the Feta was packaged, was contaminated with 103 cfu/ml. Contaminated Feta samples were vacuum-packaged and exposed to irradiation doses of 1.0, 2.5 and 4.7 kGy and stored at 4 °C for a month. In the pre-process contaminated samples none of the irradiation doses eliminated L. monocytogenes; however the highest dose reduced the viable population to a level which is in compliance with EC regulations. In the post-process contamination, the 2.5 kGy and 4.7 kGy doses reduced L. monocytogenes counts below the detection limit. Irradiation had no effect on the texture of Feta. Irradiation at 4.7 kGy increased Feta's redness and decreased its yellowness and lightness. Sensorial analyses showed that at the 4.7 kGy dose, the aroma profile of Feta was temporarily affected, since it was restored after 30 days of cold storage.  相似文献

We evaluated the effects of ionizing radiation (1-3 kGy) and incorporation of antimicrobials on the functional properties of low-density polyethylene (LDPE)/polyamide films. We established the antimicrobial effectiveness of several coatings of FDA-approved antimicrobial compounds including sorbic acid, carvacrol, trans-cinnamaldehyde, thymol and rosemary oleoresin using selected food pathogen surrogates. The antimicrobial coatings were applied to one side of the LDPE films and dried. Films were irradiated using a 10 MeV linear electron beam accelerator at room temperature. All films showed inhibition zones in an agar diffusion test against Listeria innocua ATCC 33090 and Escherichia coli ATCC 884. In the liquid culture test, the antimicrobials significantly (p?0.05) reduced the specific growth rate of L. innocua by 3.8-8.5%, and decreased final cell concentration of both strains by 5.7-14.6% and 7.2-16.8%, respectively. All active compounds retained the antimicrobial activity when exposed to 1-3 kGy. Neither the presence of active compound nor dose affected the film's tensile strength and toughness. Additionally, films became more ductile and had improved moisture barrier functionality. Film's oxygen permeability was not affected by either treatment. Results are an initial step toward the development of self-sterile active packaging materials for use in combination with irradiation treatment of foods.  相似文献

Vibrio parahaemolyticus has been considered as one of the most important food-borne bacterial pathogens. Because of the safety concerns, detection and characterization of V. parahaemolyticus have attracted much attention. In this study, electrochemiluminescence polymerase chain reaction (ECL-PCR) method combined with universal probes hybridization technique was applied to rapid detection of V. parahaemolyticus, infected and uninfected sea foods for the first time. Whether the sea food samples were infected was discriminated by detecting the gyrase B (gyrB) gene. We detect V. parahaemolyticus both in artificially contaminated sea foods and natural samples. The experiment results show that the infected and uninfected sea food samples can be clearly identified and the detection limit for V. parahaemolyticus is 1.6 pg purified genomic DNA in the presence of 1 μg non-specific background DNA. The technique may provide a new means in V. parahaemolyticus detection due to its simplicity and high efficiency.  相似文献

The single and combined effects of enterocin AS-48 and high hydrostatic pressure (HHP) on Listeria monocytogenes, Salmonellaenterica, and Staphylococcus aureus was investigated in fuet (a low acid fermented sausage) during ripening and storage at 7 °C or at room temperature. AS-48 (148 AU g−1) caused a drastic 5.5 log cfu g−1 decrease in L. monocytogenes (P < 0.001) and a significant (P < 0.01) inhibition (1.79 logs) for Salmonella at the end of ripening (10 d). After pressurization (400 MPa) and storage Listeria counts remained below 5 cfu g−1 in all fuets containing AS-48 (pressurized or not). HHP alone had no anti-Listeria effect. HHP treatment significantly reduced Salmonella counts, with lowest levels in pressurized fuets with AS-48. S. aureus showed similar growth for all treatments and storage conditions. These results indicate that AS-48 can be applied alone to control L. monocytogenes and combined with HHP treatment to control Salmonella in fuets.  相似文献

Essential oils (EOs) of three Apiaceae species, including Bunium persicum, Cuminum cyminum and Carum copticum, extracted by hydrodistillation, were analyzed by gas chromatography (GC) and GC/mass spectrometry. The main components of EOs of B. persicum were γ-terpinene (44.2%), cuminaldehyde (16.9%), γ-terpinen-7-al (10.5%), and ρ-cymene (8%). The major constituents of Cu. cyminum were cuminaldehyde (30.2%), ρ-cymene (14.1%), γ-terpinene (12.8%), and safranal (9.4%), while those of Ca. copticum were thymol (48.4%), ρ-cymene (21.8%) and γ-terpinene (21.3%). The antibacterial effects of the EOs were assessed on several food-borne pathogens, namely Staphylococcus aureus, Bacillus cereus, Escherichia coli O157:H7, Salmonella enteritidis, and Listeria monocytogenes. The ranges of minimum inhibitory concentration (MIC) of the oils were 0.03–0.5, 0.18–3.0, and 0.37–3.0 mg/ml, respectively, for Ca. copticum, B. persicum and Cu. cyminum. Moreover, the combination of B. persicum and Cu. cyminum EOs confirmed synergistic and additive activities against the pathogens.  相似文献

The increase in reported food-borne outbreaks linked with consumption of raw fruits and vegetables has motivated new research focusing on prevention of pre-harvest produce contamination. This study evaluates and compares the effectiveness of three non-thermal technologies, chlorine dioxide gas, ozone gas and e-beam irradiation, for inactivation of Salmonella enterica and Escherichia coli O157:H7 on pre-inoculated tomato, lettuce and cantaloupe seeds, and also their corresponding effect on seeds germination percentage after treatments. Samples were treated with 10 mg/l ClO2 gas for 3 min at 75% relative humidity, with 4.3 mg/l ozone gas for 5 min and with a dose of 7 kGy electron beam for 1 min. Initial load of pathogenic bacteria on seeds was ~ 6 log CFU/g. Results demonstrate that all treatments significantly reduce the initial load of pathogenic bacteria on seeds (p < 0.05). In particular, after ozone gas treatments 4 log CFU/g reduction was always observed, despite the seeds and/or microorganisms treated. ClO2 and e-beam treatments were noticeably more effective against Salmonella on contaminated tomato seeds, where 5.3 and 4.4 log CFU/g reduction were respectively observed. Germination percentage was not affected, except for cantaloupe seeds, where the ratio was significantly lowered after ClO2 treatments. Overall, the results obtained show the great applicability of these non-thermal inactivation techniques to control and reduce pathogenic bacteria contamination of seeds.  相似文献

The influences of lactic acid bacteria and brine solutions on the biogenic amine formation by Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Enterococus faecalis, Pseudomonas aeruginosa, Listeria monocytogenes, Aeromonas hydrophila and Salmonella paratyphi A in fermented trout fillets were investigated. Fish fillets were divided into four groups, group 1 without any lactic acid bacteria inoculation, group 2 and group 3 with different salt concentration inoculated with lactic acid bacteria and food-borne pathogens, and group 4 inoculated with lactic acid bacteria and food-borne pathogens without a salt solution. The histamine content in trout fillets in group 4 was found to be more than 10 mg/100 g, while the other groups contained less than 7.5 mg/100 g. The highest tyramine production was found for group 1 and group 3, ranging from 3 to 18 mg/100 g. Lactic acid bacteria did not seem to play an important role on biogenic amine production by food borne pathogens, while adding brine solution on fillets has inhibitory effects on some of the biogenic amines.  相似文献

The morphological, thermal and pasting properties of starch separated from potatoes of three varieties (Kufri Chandramukhi, Kufri Jyoti and Kufri Chipsona-2), treated either with CIPC (isopropyl N-(3 chlorophenyl) carbamate) or γ-irradiation (Co60, 0.1 and 0.5 kGy) and subsequently stored at 8, 12 and 16 °C for 90 days, were studied. Scanning electron microscopy (SEM) showed the presence of oval and irregular shaped starch granules with a diameter range of 15–16 μm. Mean granule size of starch separated from potatoes stored at 12 °C ranged from 18–25 μm and irradiation treatment resulted in an increase in the proportion of small size granules. The irradiation of potatoes with 0.5 kGy resulted in starch with significantly lower peak-, trough- and breakdown-viscosity as compared to starch from potatoes treated with either CIPC or 0.1 kGy irradiation. The irradiation of potatoes with 0.5 kGy caused a significant increase in setback and pasting temperature. Pasting temperature of starch was observed to vary with the storage temperature. Starch separated from potatoes stored at higher temperature showed lower pasting temperature and vice versa. The starch from potatoes stored at 8 °C showed higher peak-, trough- and breakdown-viscosity and lower setback. Peak viscosity increased and swelling volume decreased with increase in storage temperature. FTIR spectra showed that the starch from irradiated potatoes displayed a significant decrease in the intensity of the C–H stretch region between 2800 and 3000 cm−1, which was observed to be irradiation dose-dependent, and higher with 0.5 than 0.1 kGy. However, a slight broadening of O–H stretch (3000–3600 cm−1) in starches from irradiated potatoes was observed. The spectral changes caused by γ-irradiation were apparent in the O–H stretch (3000–3600 cm−1), C–H stretch (2800–3000 cm−1) and bending mode of water (1600–1800 cm−1).  相似文献

The food-borne pathogens Listeria monocytogenes, Salmonella enterica, Staphylococcus aureus, Yersinia enterocolitica and Campylobacter jejuni, and the spoilage lactic acid bacteria (LAB), Escherichia coli and the yeast Debaryomyces hansenii were inoculated on slices of cooked ham, dry cured ham and marinated beef loin. During storage at 4 °C, L. monocytogenes and LAB increased up to 3.5 log units while the other species, unable to grow under refrigeration, continued at the spiking level. The application of a 600 MPa treatment effectively inactivated most of the microorganisms, the counts of which, except for LAB that increased in cooked ham and in beef loin, progressively decreased or maintained below the detection limit during the whole storage (120 days at 4 °C).  相似文献

Antibacterial effects of American cranberry (Vaccinium macrocarpon) concentrate on foodborne pathogens, Escherichia coli O157:H7, Listeria monocytogenes, Salmonella Typhimurium, and Staphylococcus aureus in vitro were investigated. Cranberry concentrate at various concentrations was prepared in distilled water (DW) or Brain Heart Infusion (BHI) broth. Pathogens were inoculated in each sample and incubated at 21 and 4 °C for 0, 1, 5, 7, and 24 h (DW samples) and 0, 1, 3, and 5 days (BHI samples). Transmission electron microscopy (TEM) was used to study the effects of cranberry concentrate on cellular structure of pathogens. DW results showed that S. Typhimurium and L. monocytogenes were reduced to non-detectable levels at 5 h in 100 μl/ml treatment at 21 and 4 °C. At 24 h, no target pathogens were detected from the 100 μl/ml treatment. BHI data indicated that the 100 μl/ml treatment reduced the four pathogens by 3-8 log CFU/ml compared with the control on Day 5 at 21 and 4 °C. TEM revealed damage to the bacterial cell walls and membranes. Cranberry concentrate has antibacterial effects on the four foodborne pathogens. Based on potential health benefits and proven antimicrobial effects, American cranberry concentrate may have dual applications as a food preservative.  相似文献

The objective of this study was to investigate the antioxidant and antibacterial activities of exopolysaccharide (EPS) from Bifidobacterium bifidum WBIN03 (B-EPS) and Lactobacillus plantarum R315 (L-EPS). The 1,1-diphenyl-2-picrylhydrazyl (DPPH)-radical scavenging, hydroxyl radical-scavenging, and superoxide radical-scavenging abilities were measured to evaluate antioxidant activity. Inhibition of erythrocyte hemolysis and lipid peroxidation was also measured. Both B-EPS and L-EPS had strong scavenging ability against DPPH and superoxide radicals at high concentration. The inhibitory effect of B-EPS on erythrocyte hemolysis was stronger than that of L-EPS in a concentration range from 0.30 to 1.00 mg/mL, whereas the hydroxyl scavenging ability of L-EPS (39.15 ± 0.58%) was significantly higher than that of 0.15 mg/mL ascorbic acid (24.33 ± 1.17%) and B-EPS (17.89 ± 3.30%) at 0.10 mg/mL. The inhibition of lipid peroxidation of 0.50 mg/mL B-EPS and L-EPS was 13.48 ± 1.74% and 12.43 ± 0.51%, respectively, values lower than that of ascorbic acid at the same concentration (23.20 ± 1.41%). Furthermore, all these abilities were enhanced in a concentration-dependent manner. Agar diffusion assay showed that both EPS exhibited antibacterial activities against tested pathogens such as Cronobacter sakazakii, Escherichia coli, Listeria monocytogenes, Staphyloccocus aureus, Candida albicans, Bacillus cereus, Salmonella typhimurium, and Shigella sonnei at 300 μg/mL. In conclusion, both EPS have antimicrobial and antioxidant activities and could have applications in the food industry.  相似文献
