The methodology of three-dimensional quantitative structure-activity relationships (3D-QSAR) has been employed to develop a quantitative model for the relationship between single-cell activities for a large set of analogs of (Z)-5-decenyl acetate, a pheromone component of the turnip moth, Agrotis segetum, and the structural characteristics of the analogs. The method of partial least squares (PLS) projections to latent structures has been used to delineate the relationships between measured activities and chemical structures. The training set includes 42 compounds, and a test set of seven molecules has been employed to assess the predictivity of the model. The model derived is able to calculate the activities of the seven test compounds with an average unsigned error of 0.3, close to the experimental error. Contour maps of regression-like coefficients from transformed PLS components display electronic and nonbonded features of high importance for the observed variation in the activities of compounds in the training set. The contours for the nonbonded (van der Waals) field support previous conclusions that the interaction between the pheromone component and its receptor is characterized by a high steric complementarity.  相似文献

The female sex pheromone composition and the male electro-physiological response with respect to the three main sex pheromone components, (Z)-5-decenyl acetate, (Z)-7-dodecenyl acetate, and (Z)-9-tetradecenyl acetate, were investigated in populations ofAgrotis segetum from Armenia and Bulgaria. The percentage composition of the female-produced pheromone was 15247 and 14257 for the respective populations. Corresponding male receptor frequencies were 9901 and 6922. EAG response profiles of the male antennae were similar for the two populations. The populations from Armenia and Bulgaria differed from the earlier investigated French and Swedish populations, which have larger amounts of (Z)-5-decenyl acetate in gland extracts and have a majority of (Z)-5-decenyl acetate-sensitive receptors. Investigation of receptor frequencies on antennae of male Hungarian moths showed that individuals could be classified as either Swedish or Armenian/Bulgarian type. Males of the Swedish type were preferentially attracted to the three-component pheromone blend, whereas blends of (Z)-7-dodecenyl and (Z)-9-tetradecenyl acetate, and (Z)-7-dodecenol [pure or in mixture with (Z)-5-decenol] attracted the Armenian/Bulgarian type. Analogs of (Z)-5-decenyl acetate, a pheromone component of the turnip moth,Agrotis segetum, with a chloro, bromo, or iodo substituent in place of the terminal methyl group have been synthesized and tested by electrophysiological single-sensillum recordings. The electrophysiological results have been interpreted in terms of substituent size and hydrophobicity. Interaction energies based on receptor interaction models have been calculated using the molecular mechanics [MM2(85)] method. The results support our previously suggested receptor interaction model in which the terminal alkyl chain interact with a complementary hydrophobic receptor pocket with very limited flexibility.  相似文献

In order to test a previous conclusion that chain-elongated analogs of (Z)-5-decenyl acetate(1), a pheromone component of the turnip moth,Agrotis segetum, adopt a loop conformation of the terminal alkyl chain in the bioactive conformation, a series of alkyl ether and enol ether analogs of1 and (Z)-5-dodecenyl acetate(2) have been synthesized and tested using singlecell electrophysiology. In these analogs a methylene group in positions 7 and 9 of1 and in positions 7 and 11 in2 have been replaced by an oxygen atom in order to energetically facilitate the formation of a loop conformation in the chain-elongated analogs. The electrophysiological results in combination with molecular mechanics (MM2 and MM3) calculated conformational energies show that the activity decreases of the chain-elongated ether analogs are significantly smaller than that for2 and that these activity decreases parallel the conformational energies for a loop formation of the terminal chains in the analogs. The results support our previous conclusion that the terminal chain of chain-elongated analogs of1 adopts a loop conformation in their bioactive conformations.  相似文献

Methyl-substituted analogs of (Z)-5-decenyl acetate, a pheromone component of the turnip moth,Agrotis segetum, have been synthesized and studied by electrophysiological single-sensillum recordings and molecular mechanics calculations [MM2(85)]. The analogs are monomethyl substituted in the 2, 3, 4, and 5 positions and geminally dimethyl substituted in the 2, 3, and 4 positions. The methyl groups have been employed as space probes to study the degree of steric complementarity between the acetate-substituted alkyl chain of the pheromone component and its receptor. The electrophysiological activities, interpreted in terms of a receptor interaction model, indicate significant steric repulsive interactions between the introduced methyl groups and the receptor. The sex pheromone of the female almond moth, Cadra cautella, constitutes a highly variable signal. Age and period of the L/D cycle had significant effects on the variation in titers of (Z,E)-9,12-tetradecadienyl acetate (Z9,E12-14:Ac) and (Z)-9-tetradecenyl acetate (Z9-14:Ac) and the ratio of Z9,E12-14:Ac to Z9-14:Ac. Across age classes, minimum and maximum mean titers (ng +/- SE) of Z9,E12-14:Ac and Z9-14:Ac ranged from 8.7 +/- 0.46 to 21 +/- 1.0 and from 2.0 +/- 0.14 to 2.9 +/- 0.15, respectively; the mean ratio of Z9,E12-14:Ac to Z9-14:Ac ranged from 5.2 +/- 0.21 to 11 +/- 0.40. The titers of Z9,E12-14:Ac and Z9-14:Ac and the ratio of Z9,E12-14:Ac to Z9-14:Ac were highest at the onset of the scotophase and lowest at the onset of the photophase. Similarly, the titer of Z9,E12-14:Ac and the ratio of Z9,E12-14:Ac to Z9-14:Ac were highest in females sampled 1 d postemergence and declined over all age classes. The titer of Z9-14:Ac increased from day 1 to day 2, and then declined to levels equivalent to day 1. Analysis of pupal and adult mass demonstrated a positive correlation (r = 0.874, P < 0.001); however, the titer of neither Z9,E12-14:Ac or Z9-14:Ac nor the ratio of Z9,E12-14:Ac to Z9-14:Ac were significantly correlated with pupal mass. Age-related variations in pheromone titer and ratios were heritable. The narrow-sense heritability of the observed variation in 1-d-old females, 1 hr into the scotophase is 1.2 +/- 0.32, 0.75 +/- 0.24, and 0.46 +/- 0.17 for the titer of Z9,E12-14:Ac and Z9-14:Ac and the ratio of Z9,E12-14:Ac to Z9-14:Ac, respectively. A significant additive genetic correlation was observed between that of Z9,E12-14:Ac and Z9-14:Ac, but not between that of either Z9,E12-14:Ac or Z9-14:Ac and the ratio of Z9,E12-14:Ac to Z9-14:Ac. The results of the genetic analyses suggest that the traits of titer of Z9,E12-14:Ac and Z9-14:Ac have a greater potential to respond to selection than the trait of ratio of Z9,E12-14:Ac to Z9-14:Ac.  相似文献

Structure-activity relationships for 6-, 7-, 8-, and 9-alkyl substituted analogs of (Z)-5-decenyl acetate, a pheromone component of the turnip moth,Agrotis segetum, have been studied by electrophysiological single-sensillum recordings, and interpreted in terms of a receptor-interaction model. The compounds were prepared by alkenyl cuprate reactions with,-unsaturated carbonyl derivatives or alkyl halides. Selected combinations of (Z)-5-decenyl, (Z)-7-dodecenyl, and (Z)-9-tetradecenyl acetates, the pheromone components of the turnip mothAgrotis Segetum were tested for field attractancy at six, two, and three sites in Europe, Asia, and Africa, respectively. At all of the sites in Eurasia and in northern Africa the ternary mixture of the acetates captured most males, while at the sites south of the Sahara in Africa, (Z)-5-decenyl acetate alone was responsible for attraction. Differences in male attraction among the populations studied confirm the existence of significant population variation in the pheromone ofA. segetum. Interpretation of the present results together with earlier studies suggests that this variation is more or less continuous in Eurasia and north Africa, while a clearly distinct pheromone type is present in the areas south of the Sahara desert.  相似文献

Structure-activity relationships for chain-shortened analogs of (Z)-5-decenyl acetate, a pheromone component of the turnip moth,Agrotis segetum, have been studied by electrophysiological single-sensillum technique and interpreted in terms of a previously reported receptor-interaction model. The results indicate that the terminal methyl group, as well as the acetate group, interacts with highly complementary receptor sites. The terminal alkyl chain is suggested to interact with a hydrophobic pocket extending over the two methylene groups closest to the terminal methyl group. The amounts of stimulus actually released from the odor source have been studied. The results demonstrate the necessity to take differences of volatility into account in comparisons of electrophysiological data for compounds of different chain lengths. Female turnip moths (Agrotis segetum) from a laboratory culture inbred for more than 30 generations, and the offspring (first and third generation) from field-collected insects were analyzed individually for acetates and alcohols in the pheromone gland. Quantitative analysis of individual components was performed at the subnanogram level by gas chromatographymass spectrometry (selected ion monitoring). The titer of the pheromone, i.e., the sum of the homologous acetates (Z)-5-decenyl acetate, (Z)-7-dodecenyl acetate, and (Z)-9-tetradecenyl acetate was 2.0 ± 0.3 ng in the laboratory culture and 3.2 ± 0.6 ng in the wild strain. There was no correlation between pheromone titer and female weight. The relative proportion of the pheromone components varied substantially between individuals, but there was no statistically significant difference between the two populations. The percentages of the respective compounds (¯X ± coefficient of variation) were 14.8 ± 127% for Z5-10:OAc, 55.6 ± 32% for Z7-12:OAc, and 29.6 ± 59% forZ9-14:OAc. The pheromone composition varied more in the wild strain than in the laboratory culture. The significance of the pheromone variation to the attraction of males was tested in a field experiment. The apple leafminer moth, Phyllonorycter ringoniella, is becoming a more serious insect pest on apple trees with four to five generations a year in Korea. In order to devise a forecasting method for more accurate estimation of their numbers and development timing, the sex attractant was studied. Various ratios, from 10:0 to 0:10, of the two components, (Z)-10-tetradecenyl acetate (Z10–14:Ac) and (E,Z)-4,10-tetradecadienyl acetate (E4,Z10–14:Ac), identified from the sex pheromone gland (Jung and Boo, 1997), were tested for attractivity in terms of behavioral response (taxis, approach, and landing) against P. ringoniella males in a wind tunnel. The lure with Z10–14:Ac/E4,Z10–14:Ac in a ratio of 4:6 elicited the highest response in two (taxis and approach) measurement categories. For eliciting landing behavior, the two blends of 5:5 and 4:6 were best. The single component, Z10–14:Ac, elicited taxis behavior, but a combination of two chemicals was needed for eliciting all three behaviors. In the field, male attraction to various lure mixtures in Pherocon IC traps was usually greater than attraction to virgin females. The best field activity was in the lure baited with a 4:6 ratio of Z10–14:Ac and E4,Z10–14:Ac. Similar results were obtained from tests conducted in a net house. This optimum ratio for attracting P. ringoniella males in Korea is different from those reported in Japan (10:3) or China (7:3 to 6:4). The isomer E10–14:Ac neither improved nor depressed the number of catches when added at up to 10% of the total mixture to lures of the two components in the 4:6 ratio. The attractivity of the lures increased with higher amounts of the pheromones, up to 10 g in the wind-tunnel experiment and 5 mg in the apple orchard. The number of males captured was not significantly different among traps installed at 0.3, 1.5, or 2 m above the ground, or among wing, delta, or water traps. A rubber septum dispenser impregnated with 1 mg of the 4:6 mixture maintained its field attractivity for up to eight weeks.  相似文献

The response of individual male turnip mothsAgrotis segetum was observed in a sustained flight tunnel to a mixture of decyl acetate, (Z)-5-decenyl acetate, (Z)-7-dodecenyl acetate, and (Z)-9-tetradecenyl acetate in proportions similar to those found in gland extracts from virgin females (0.6: 1:5:2.5). Lures containing 3–30 g (Z)-5-decenyl acetate proved to be maximally attractive, with approximately 60% of the males completing all behavioral steps from activation to copulation efforts. A 300-g dosage caused significant arrestment of upwind flight. Peak response to synthetics, however, was significantly lower than to female glands. Omitting decyl acetate from the blend did not affect the activity, while omission of any of the three monounsaturated acetates caused a dramatic decrease in response. In the field maximum trap catches were achieved with 1- to 30-g lures. The subtractive assay carried out in the field confirmed the neutrality of decyl acetate and the importance of the three monoenes. Adding 1% of (Z)-8-dodecenyl acetate (earlier reported as an inhibitor) to the four-component mixture decreased the trap catch to about 50%, and increasing the amount of (Z)-8-dodecenyl acetate to 27% decreased the activity further to about 10%. The enantiomers of analogs of (Z)-5-decenyl acetate, a pheromone component ofAgrotis segetum, substituted by a methyl group in the 2, 3, 4, 7, and 8 positions and dimethyl substituted in the 4,7 positions, have been synthesized and studied by an electrophysiological single-cell technique and by molecular mechanics calculations. The results demonstrate that the electrophysiological activity as well as the ability of the (Z)-5-decenyl acetate receptor to differentiate between enantiomers depends on the position of the methyl substituent. For analogs methyl substituted in the 2, 4, or 8 position, no differences in the activities of the enantiomers could be observed. In contrast, the enantiomers of the 3- and 7-methyl analogs display a significant difference in the activities, theR-enantiomers being more active than theS-enantiomers. The female-produced sex attractant pheromone of Coloradia velda has been identified by a combination of analytical methods and field tests as a blend of (10E,12Z)-hexadecadien-1-yl acetate (10E,12Z–16:Ac), (10E,12E)-hexadecadien-1-yl acetate (10E,12E–16:Ac), and (10E,12Z)-hexadecadien-1-ol (10E,12Z–16:OH) in a 10:1:0.33 ratio, respectively. The corresponding aldehyde (10E,12Z–16:Ald) was also found in extracts of female pheromone glands, but in blends with 10E,12Z–16:Ac and 10E,12E–16:Ac at low doses did not significantly increase the attractiveness of baits to male moths.  相似文献

Partially fluorinated analogs of the European corn borer (Ostrinia nubilalis) female sex pheromone, 11-tetradecenyl acetate (97:3Z:E), having mono- and trifluorsubstitutions at the terminal carbon of the pheromone chain, mimicked the biological activity of the pheromone, while analogs with fluorine at either side of the double bond and a pentafluoro analog were essentially inactive. Comparison of the pheromonal activity of these analogs with the previously reported activity of similarly fluorinated pheromones in five other species of moths revealed an unpredictable relationship between fluorine substitution pattern and pheromone-mimicking activity. Fluorine substitution patterns that rendered pheromonal analogs biologically inactive in the European corn borer had no detrimental influence upon pheromonal activity in other species and the converse was also true. This is evidence that the relative importance of electronic qualities of sites within a pheromone molecule differ from species to species. Furthermore, it indicates that the biochemical components (pheromone receptor proteins, binding proteins, and enzymes) that make up moth olfactory chemosensory systems must also vary structurally from species to species, despite the fact that they are involved in olfactory sensing of compounds having very similar chemical structure.  相似文献

This study describes the identification of a sex pheromone component of a cossid moth, Cossus insularis. Coupled gas chromatographic–electroantennographic detection (GC–EAD) analysis of solid-phase microextraction (SPME) collections of volatiles released by live female moths showed that two compounds elicited EAG responses from the antennae of male moths. These compounds were identified as (E)-3-tetradecenyl acetate (E3-14:Ac) and (Z)-3-tetradecenyl acetate (Z3-14:Ac) by mass spectral analysis and retention index comparisons with synthetic standards. The ratio of E3-14:Ac and Z3-14:Ac was 95:5 in the effluvia of a female. In field bioassays, sticky traps baited with blends of E3-14:Ac and Z3-14:Ac showed that E3-14:Ac is an essential component of the pheromone. However, the role of Z3-14:Ac is unclear, because E3-14:Ac as a single component was as attractive to male moths as blends of E3-14:Ac and Z3-14:Ac, including the 95:5 blend released by live female moths.  相似文献

The major component of the female-produced sex pheromone of Scoliopteryx libatrix has been characterized by chemical analysis, synthesis, electrophysiological studies and field tests as (6Z,13)-methylheneicosene, probably the 13S-isomer. This is the first example of a branched chain alkene as a sex pheromone in the Noctuidae and is markedly different from the pheromones of other members of the family. The systematic position of S. libatrix, belonging to a monotypic genus of a one-member subfamily within the Noctuidae, may reflect the unusual structure of the sex pheromone.  相似文献

Anadevidia peponis and Macdunnoughia confusa are defoliators of plants in the family Cucurbitaceae and Compositae, respectively, in Japan. GC-MS analyses of crude pheromone gland extracts treated with or without dimethyl disulfide indicated that females of A. peponis produced six monoene acetates and two monoene alcohols and that M. confusa females produced five monoene acetates. These components include (Z)-7-dodecenyl acetate as a major common constituent and three other acetates as minor common constituents. The minor constituents are quite different in blend composition. In addition, with (Z)-7-dodecenyl acetate, an indispensable component for male attraction is (Z)-5-decenyl acetate for A. peponis and (Z)-9-tetradecenyl acetate is essential for M. confusa. Field tests with synthetic lures showed synergistic effects of some other minor components and male attraction of three additional Plusiinae species, Macdunnoughia purissima, Ctenoplusia albostriata, and Chrysodeixis eriosoma, suggesting their reproductive isolation is based in part on pheromonal communication.  相似文献

In 1996, the exotic white-spotted tussock moth (WSTM), Orgyia thyellina (Lepidoptera: Lymantriidae), was discovered in Auckland, New Zealand. Because establishment of WSTM would threaten New Zealand's orchard industry and international trade, eradication of WSTM with microbial insecticide was initiated. To monitor and complement eradication of WSTM by capture of male moths in pheromone-baited traps, pheromone components of female WSTM needed to be identified. Coupled gas chromatographic–electroantennographic detection analysis of pheromone gland extract revealed several compounds that elicited responses from male moth antennae. Mass spectra of the two most EAD-active compounds suggested, and comparative GC-MS of authentic standards confirmed, that they were (Z)-6-heneicosen-11-one (Z6–11-one) and (Z)-6-heneicosen-9-one, the latter termed here thyellinone. In field experiments in Japan, Z6–11-one plus thyellinone at a 100:5 ratio attracted WSTM males, whereas either ketone alone failed to attract a single male moth. Addition of further candidate pheromone components did not enhance attractiveness of the binary blend. Through the 1997–1998 summer, 45,000 commercial trap lures baited with 2000 g of Z6–11-one and 100 g of thyellinone were deployed in Auckland towards eradication of the residual WSTM population.Dedicated to my dear mother in honor of her 75th birthday  相似文献

(Z)-10,(Z)-12-Tetradecadienyl acetate (Z10,Z12–14:OAc) and (E)-10,(E)-12-tetradecadienyl acetate (E10,E12–14:OAc) are sex pheromone components of the apple blotch leafminer (ABLM), Phyllonorycter crataegella. Compounds extracted from female pheromone glands were identified by coupled gas chromatographic–electroantennographic detection (GC-EAD) analyses, retention index calculations of EAD-active compounds, and by comparative GC-EAD analyses of female ABLM-produced and authentic (synthetic) compounds. In field experiments in apple Malus domestica orchards in Connecticut, Z10,Z12–14:OAc alone attracted ABLM males. Addition of E10,E12–14:OAc to Z10,Z12–14:OAc at 0.1:10 or 1:10 ratios enhanced attractiveness of the lure. Geometrical isomers Z10,E12- or E10,Z12–14:OAc at equivalent ratios were behaviorally benign and slightly inhibitory, respectively. In field experiments in British Columbia, Z10,Z12–14:OAc plus E10,E12–14:OAc did not attract Phyllonorycter moths, supporting the contention that ABLM is not present in the fruit growing regions of British Columbia. Z10,Z12–14:OAc added to P. mespilella pheromone, (E)-4,(E)-10-dodecadienyl acetate, strongly inhibited response by P. mespilella males. Recognition of the ABLM pheromone blend by allopatric P. mespilella males suggests a phylogenetic relationship and previous sympatry of these two Phyllonorycter spp. If pheromonal attraction of ABLM males were reciprocally inhibited by P. mespilella pheromone, a generic Phyllonorycter pheromone blend could be tested for pheromone-based mating disruption of the apple leaf-mining Phyllonorycter guild in North America.  相似文献
