A low vitamin E/selenium status has been associated with increased vulnerability of dairy cattle to mastitis. Since polymorphonuclear leucocytes (PMN) provide the major cellular defence mechanism within the mammary gland, the effect of in vitro supplementation with vitamin E and selenium on the function of these cells was investigated. Both vitamin E and selenium enhanced the chemotactic and random migration of PMN and increased the production of superoxide following stimulation with phorbol myristate acetate. Vitamin E, but not sodium selenite, was also found to enhance the phagocytosis of opsonised Staphylococcus aureus by PMN. No synergistic effects of the two nutrients were observed. These results obtained in vitro may indicate the potential benefits of in vivo supplementation of dairy cows with vitamin E and selenium in terms of enhancing their natural resistance to mastitis.  相似文献

OBJECTIVE: To determine the association between intramammary infections caused by various bacteria and hepatic damage, as measured by serum sorbitol dehydrogenase (SDH) activity, WBC counts, and PCV and the association of clinical variables with milk production after a case of clinical mastitis. DESIGN: Prospective, cohort study. ANIMALS: 82 cows with clinical mastitis. PROCEDURE: Information on milk production, mastitis status, and selected physiologic variables was collected during a 1-year period. Milk samples for bacteriologic evaluation were collected on day 1 of an episode of clinical mastitis. Physical examination was performed and blood samples for laboratory evaluation were collected on days 1, 5, and 9. Primary outcome was mature equivalent 305-day (ME305) milk production. Correlations were assessed using a multiple regression model. RESULTS: Higher WBC counts were associated with higher ME305 values. For cows with coliform mastitis, increases in SDH values were associated with higher ME305 values. For cows with coliform and streptococci/staphylococci mastitis, PCV was associated with ME305 values. Higher PCV values were associated with lower ME305 values for streptococci/ staphylococci and coliform infections. CLINICAL IMPLICATIONS: The association between milk production and WBC count indicated that cows mobilizing WBC were better able to neutralize mammary gland infections, which may result in better milk production. The association between milk production and PCV suggested that maintaining hydration in cows with clinical mastitis may be a critical aspect of treating all cows with mastitis.  相似文献

Three groups of dairy cows were immunized by subcutaneous (s.c.) administration of a preparation of live Streptococcus uberis (strain 0140J) and an intramammary infusion of a soluble surface extract derived from same the bacteria. Animals in Groups 1 and 2 received two s.c. vaccinations plus an intramammary inoculation. Animals in Group 3 received two s.c. vaccinations but did not receive the intramammary infusion. In addition to the vaccinated animals, each group also contained two non-vaccinated (control) animals. All animals were challenged experimentally by intramammary infusion (in two quarters per animal) of ca 100 c.f.u. of S. uberis (strain 0140J or C221) and monitored for clinical signs of disease, bacterial numbers in milk, somatic cell count in milk, and daily milk yield for the following 10 days. Animals in Group I were challenged with strain 0140J. Only one out of six challenged quarters of three vaccinated cows developed clinical disease compared to all (four out of four) quarters of non-vaccinated cows. Animals in Group 2 were challenged with strain C221. All challenged quarters of three vaccinated (six out of six) and two non-vaccinated (four out of four) cows developed clinical mastitis. Animals in Group 3 were challenged with strain 0140J. Five out of eight quarters on four vaccinated cows developed clinical mastitis but the onset was delayed in comparison with that in both non-vaccinated cows in which four out of four challenged quarters developed clinical mastitis. These results indicated that vaccination with live S. uberis protects against challenge with the homologous strain but was less effective against a heterologous strain. Reduced protection was also seen when the intramammary booster was omitted.  相似文献

We investigated the activity of glutathione peroxidase in whole blood; concentrations of vitamin E, vitamin A, and beta-carotene in serum; SCC; udder bacterial infections and the incidence of clinical mastitis; fertility treatments; and the success of first AI of 511 dairy cows for 1 yr. The mean Se content in whole blood and the concentrations of vitamin E, vitamin A, and beta-carotene concentrations in serum were 191 micrograms/L, 5.9 mg/L, 0.39 mg/L, and 12.9 mg/L, respectively. An increase in Se concentration in whole blood was associated with a decrease in all infections, including infections by Staphylococcus aureus, Actinomyces pyogenes, and Corynebacterium spp. (-17.7, -31.7, and -70.6%, respectively). There was no association among the different infections or SCC and concentrations of vitamin E, vitamin A, or beta-carotene, but an association existed between vitamin A concentration and SCC. The lower Se concentration in whole blood did not increase incidence of clinical mastitis. The Se concentration in whole blood (200 micrograms/L) was accepted as a target value to optimize udder health. The incidence of fertility disorders (anestrus, subestrus, cystic ovaries, or delayed ovulation) was 34.4%. The pregnancy rate following first insemination was 48.6%. No significant association was observed among Se in whole blood; concentrations of total vitamin E, vitamin A, or beta-carotene in serum; and fertility disorders or success of first AI.  相似文献

Results of a 20 month split-udder trial on the effect of discontinuation of postmilking teat disinfection on intramammary infections (IMI) with major and minor pathogens in seven dairy herds with a low somatic cell count are described. The incidence of Escherichia coli IMI was found to be significantly lower, whereas the incidence of IMI with Staphylococcus aureus and minor pathogens was significantly higher in quarters for which postmilking teat disinfection was discontinued than in disinfected quarters. It was concluded that discontinuation of postmilking teat disinfection decreased the incidence of E. coli IMI, accompanied by a, from a practical point of view, acceptable rise in somatic cell count. However, the possible increase in the incidence of S. aureus IMI calls for careful monitoring of the dynamics of IMI with contagious pathogens, when postmilking teat disinfection is discontinued in an attempt to reduce E. coli mastitis.  相似文献

The identification of cows that are positive for mastitis caused by Staphylococcus aureus is difficult under field conditions. The frequency of isolation of S. aureus from quarter milk samples was compared with the frequency of recovery of S. aureus from sediment after centrifugation of those same samples. Overall, 776 quarter milk samples from 194 cows were studied. Cultures that were positive for S. aureus were obtained from 82 samples; 153 sediments from quarter milk samples were also positive for S. aureus. The results of this investigation showed that cultures of the sediment of quarter milk samples increased the number of positive outcomes up to 145.5%, depending on the herd. Using a different group of samples, including samples taken 1 to 5 d or 7 to 10 d after calving and samples taken after intramammary therapy, a 94% increase in cultures that were positive for S. aureus after centrifugation was found compared with cultures of the same quarter milk samples that were not centrifuged. Sedimented cultures may be useful in S. aureus control programs that require the segregation, selective treatment, or culling of cows that are positive for S. aureus.  相似文献

Via special media, Serratia marcescens isolates were found in 3 bedding pack samples and in 2 milking parlor floor samples, and in milk samples from 19 cows during an episode of mastitis in a dairy cow herd. Chromosomal digest patterns of isolated S marcescens were indistinguishable for 18 of the milk samples and all bedding pack samples. Our findings provide strong evidence that the bedding pack was the reservoir of S marcescens associated with the outbreak of intramammary infections. Additionally, our ability to match digest patterns of isolates in the bedding pack and milk confirms the theory that S marcescens is an environmental pathogen capable of causing mastitis.  相似文献

101 cows with 197 udder quarters with subclinical mastitis from 23 dairy farms were selected for different antibiotic treatments under field conditions. Group 1 consisting of 27 animals and 50 infected udder quarters was treated twice intramammaryly with 250 mg Cefacetril. Group 2 (26 animals/50 quarters) was treated twice intramuscular with 10 Mio IU Penethamathydrojodid and with 5 Mio IU respectively. Group 3 was treated twice with the combination of the intramammary and the intramuscular therapies in the above mentioned groups. Group 4 served as control. 23 animals with 36 subclinical infected udder quarters were treated twice intramuscular with 2.0 ml of sterile isotonic sodium-solution. Therapeutic success was controlled with bacteriological and cytological examinations of quarter milk probes one week, two weeks and four weeks after the end of treatments. Concerning bacteriological healing both the intramammary and the combined therapy had an even success with a 72.9% elimination rate of pathogenic bacteria whereas the intramuscular therapy led to bacteriological healing in 36.7%. In comparison cytological healing rates (< 100 x 10(3) cell counts per ml milk) with 29.8% in group 1, 32.3% in group 3 and 8.2% in group 2 were not satisfying. S. aureus was the predominating isolated bacteria. With respect to bacterial species found in the subclinically infected milk probes, elimination rates in Streptococcaceae and in Enterococcaceae was evident higher than in Staphylococcaceae. The objective of this study was to determine the relationship between body-condition score and postpartum reproductive efficiency in dairy cattle. Data on body-condition score, reproduction, diseases, and production from 1404 lactations of 639 cows, calving between January 1984 and November 1996 on a commercial farm, were used. The Cox proportional-hazards model with time-dependent covariates was used to qualify the relationship between body-condition score on a scale of 1 to 5 and risks of first insemination and pregnancy. Cows with body condition at calving < 3 had lower first-insemination risks. Loss of body-condition score between calving and 45 days after calving was associated with increased days open and days-to-first insemination. Cows with body-condition scores < 2 after Day 45 and before first insemination were less likely to be inseminated and become pregnant compared with cows that had higher body condition. Milk yield, mastitis, lameness, milk fever and genital infection were associated with a reduction in reproductive efficiency. The results indicated that loss of body-condition and actual body-condition scores before conception or first insemination or both can be used as a prognosis for days open and days-to-first insemination in dairy herds.  相似文献

Milk samples were collected from 108,312 dairy cows during 1601 farm visits made between January 1991 and June 1995. The herd visits were made by personnel from the Central Laboratory of the Quality Milk Promotion Services at Cornell University (Ithaca, NY) to farms located in central New York and northern Pennsylvania. Dairy Herd Improvement Association records were available for 32,978 cows in 327 herds. Intramammary infections, as defined by positive milk cultures, were present in 48.5% of all cows and in 36.3% of cows in herds enrolled in the Dairy Herd Improvement Association. Over 75% of the intramammary infections were caused by Streptococcus agalactiae, Streptococcus spp. other than Strep. agalactiae, Staphylococcus aureus, and coagulase-negative staphylococci. Mean days in milk at the time of diagnosis, linear score of the somatic cell count, cost of milk loss per lactation, and milk production effects were calculated for 24 etiologic agents of bovine mastitis.  相似文献

A randomized clinical trial including 1109 cows from 12 Australian dairy herds was used to evaluate the effects of monensin on the health (n = > 686 cows), production (n = 915 cows), and reproduction (n = > 908 cows) of dairy cows. Cows were allocated to a treatment group receiving a slow-release intraruminal bolus containing 32 g of sodium monensin that was administered 40 d before and 50 d following the anticipated calving date or to a control group. Treatment did not significantly alter any reproductive outcome; 54.5% of cows treated with monensin and 58.2% of control cows were pregnant at first service, and days to conception were lower for cows treated with monensin. The hazard rate (0.95) was not significant for these cows. The percentage of cows pregnant was 83.8 for control cows, and days to first estrus (hazard rate = 1.04) and first service (hazard rate = 1.04) were not significantly higher for treated cows. Treatment with monensin did not significantly alter the risk of any disease. The incidence of retained fetal membranes, pyometra, lameness, abortion, and infectious diseases was not significantly lower for cows in the treatment group, and the incidence of mastitis was not significantly higher for cows in the treatment group. Monensin significantly increased milk production by 0.75 L/d per cow and tended to increase milk fat and protein yields but had no significant effect on milk fat or milk protein percentages. Changes in the production of milk and milk constituents were consistent throughout lactation.  相似文献

A double-blinded randomized prospective clinical trial was designed to evaluate the effectiveness of flunixin meglumine and phenylbutazone for treatment of acute toxic mastitis in dairy cows. All cows were treated 4 times at 12-hour intervals by intramammary infusion of gentamicin (150 mg). A total of 45 dairy cows with toxic mastitis were randomly assigned to 1 of 3 treatment groups: group 1 (control), saline solution, IV; group 2, 1 g of flunixin meglumine, IV; or group 3, 4 g of phenylbutazone, IV. Physical examination and udder variables were assessed at initial examination and 24 hours later. Milk production was recorded at regular intervals from 1 week before until 10 weeks after development of mastitis. Rear quarters (34/45) were more commonly affected than front quarters. Thirty-five cows returned to the herd, 9 cows were culled, and 1 cow died. There were no significant differences among treatment groups in the need for further treatment or outcome. Klebsiella spp (18/45) and Escherichia coli (16/45) were the most common pathogens isolated by culture of milk from affected quarters. The overall bacteriologic cure rate on days 7 and 14 was 64 and 75%, respectively. At the time of initial examination, cows of the control group had higher rectal temperature than did cows of the flunixin group. At the examination 24 hours later, the rectal temperature of cows in all treatment groups was lower than the temperature at initial examination; at that time (24 hours), however, there were no significant differences in temperature among the treatment groups.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The objective of this study was to evaluate the efficacy of a premilking teat disinfectant for the prevention of mastitis in dairy cows under natural exposure conditions. Predipping was compared with a negative control using a split udder experimental design. All teats were dipped after milking with the same teat dip. Percentage of quarters newly infected by major mastitis pathogens was 34% lower in quarters with teats predipped and postdipped than in quarters with teats postdipped only. New IMI by Streptococcus uberis and Staphylococcus aureus were significantly lower in quarters with teats predipped and postdipped than in quarters with teats postdipped only. Differences in incidence of clinical mastitis between treatment groups approached significance. Predipping and postdipping were no more effective against Gram-negative bacteria, coagulase-negative Staphylococcus species, and Corynebacterium bovis than postdipping only. No chapping or irritation of teats was observed, and no adverse effects were noted using the test product as a premilking and postmilking teat disinfectant. Results of this study suggest that predipping and then postdipping with the test product was a more effective procedure against major mastitis pathogens than postdipping only.  相似文献

Microbiological, cytological, histopathological, and immunohistochemical investigations were carried out on four dairy cows affected by Serratia marcescens mastitis. The animals under study were from a herd of 120 lactating cows bred in the province of Rome. In the above herd, S. marcescens mastitis showed a prevalence of 20.8%. S. marcescens was the only bacterial agent isolated, prior to and after slaughter, from the teat milk, the mammary gland and the supramammary lymph nodes of the four cows under study. Cytologically, the four subjects exhibited high cell counts in their milk, with an average of up to 5,570,000 cells/ml in S.marcescens-infected quarters. Macroscopically, nodular lesions were apparent scattered throughout the mammary parenchyma, with enlargement of the regional lymph nodes. Histologically, a chronic, non-purulent mastitis, characterized by a marked fibrous tissue proliferation and the coexistence of corpora amylacea within the glandular alveoli, was observed in association with chronic hyperplastic lymphadenitis involving the supramammary lymph nodes of the four cows. Immunohistochemically, S. marcescens was demonstrated, by means of monoclonal antibodies, both in the mammary gland and in the supramammary lymph nodes from these four animals.  相似文献

The relationship between somatic cell counts (SCCs) up to 400,000 cells/ml in individual cows and clinical mastitis was studied by collecting monthly records for a year from 101 Holstein herds in the west of France. Monthly records obtained after any case of clinical mastitis occurring within the lactation were excluded. The resulting dataset consisted of 29,700 records from 4677 cows. The data were analysed by the logistic regression method. Herd, lactation number, calendar month, milk production, stage of lactation on the day of test, and SCC on previous test days were assumed to affect the likelihood of clinical mastitis. There was no significant relationship between SCC and the subsequent occurrence of clinical mastitis for an SCC up to 100,000 cells/ml, but the risk of clinical mastitis increased as the SCC increased up to 400,000 cells/ml. These results suggest that in individual cows, a low SCC recorded after five days in milk does not lead to an increased risk of clinical mastitis.  相似文献

Beginning at hatching, chicks were fed a Basal diet, without vitamin E or selenium (Se) or the same diet supplemented with vitamin E (100 IU/kg) and Se (0.2 ppm). The effect of these treatments on the expression of cell surface markers (CT-1a, CD3, CD4, CD8, sIgs, and Ia) defining specific thymocyte and peripheral blood leukocyte (PBL) subpopulations were examined using flow cytometric analyses. In parallel studies the effect of the dietary deficiencies on splenocyte proliferative responses to ConA or PHA stimulation was examined. The mean expression of CD3 and CT-1a per cell was increased while CD8 and CD4 expression was decreased on thymocytes from chicks fed the Basal diet. The proportion of double negative (CD4-, CD8-) thymocytes and single positive CD8+ thymocytes was significantly decreased while single positive CD4+ and double positive (CD4+, CD8+) thymocytes were significantly increased by the dietary vitamin E and Se deficiencies. The dietary deficiencies resulted in a decreased proportion of peripheral T cells and specifically decreased the number of CD4+ PBL. The proliferative response to both ConA and PHA was impaired by the vitamin E and Se dietary deficiencies. The proliferative response could be fully reconstituted but only after vitamin E and Se supplementation for periods longer than 1 week. Plasma SeGSHpx and alpha-tocopherol levels paralleled the mitogen responsiveness observed. These results support the conclusion that vitamin E and Se deficiencies may affect both the maturation of specific lymphocyte subpopulations and the functional and proliferative capabilities of the peripheral lymphocytes.  相似文献

Management practices associated with bulk milk somatic cell counts (SCC) were studied for 201 dairy herds grouped into three categories according to bulk milk SCC. The cumulative production of fat-corrected milk over 305 d of lactation and category for bulk milk SCC were highly correlated; herds within the low category had the highest milk production. Differences in bulk milk SCC among the categories were well explained by the management practices studied. This correlation was not only true for the difference between the high (250,000 to 400,000) and low (< or = 150,000) categories for bulk milk SCC but also for the difference between the medium (150,000 to 250,000) and low categories and the high and medium categories. Management practices that are known to be important for herds in the high category for bulk milk SCC, such as dry cow treatment, milking technique, postmilking teat disinfection, and antibiotic treatment of clinical mastitis, were also found to be important in the explanation of the difference between herds in the medium and low categories for bulk milk SCC. More attention was paid to hygiene for herds in the low category than for herds in the medium or high category. Supplementation of the diet with minerals occurred more frequently for cows in the low category for bulk milk SCC than for cows in the medium and high categories.  相似文献

The efficacies of two regimens for the treatment of acute clinical mastitis were compared in a randomised multi-centre field trial in Norway, using 657 cows. The purpose was to determine whether repeated intramuscular injections of penicillin G for three days were more effective than a single injection, when given in combination with intramammary treatment for five days. The results were evaluated on the basis of clinical and microbiological examinations and cell count determinations of quarter milk samples taken at the initial visit and four weeks later. There were no significant differences between the effects of the treatments, either for all the cows, or for subgroups of the cows based on age, stage of lactation, and systemic reaction, or the type of causal bacteria.  相似文献

OBJECTIVE: To evaluate use of an artificial vascular graft as treatment for obstructions of the teat and mammary gland cisternae in lactating cattle. DESIGN: Prospective clinical study. ANIMALS: 14 lactating dairy cows. PROCEDURE: After physical examination that included palpation, ultrasonography, evaluation of milk flow, and California mastitis test, each cow underwent surgical excision of obstructive tissue and implantation of a reinforced polytetrafluoroethylene vascular graft. Milk drained passively for 10 to 14 days after surgery. Follow-up evaluation was performed by telephone questionnaire of owners 1, 6, and 12 months after surgery regarding somatic cell count, time needed to milk affected quarter, compared with that of the contralateral quarter, and frequency of mastitis. After the subsequent nonlactation period and calving, milk flow was tested, using a quarter milking machine, and ultrasonographic examination of the affected teat was performed. RESULTS: Milk could be obtained from affected quarters from all cows 14 days after surgery, from 13 (93%) cows 1 month after surgery, from 10 (71%) cows 6 months after surgery, and from 3 (21%) cows 12 months after surgery, but milk flow in these 3 cows was considerably reduced. This technique failed because of collapse of the implant (4 cows), chronic mastitis (1), migration of the implant (4), and ingrowth of obstructive tissue between the distal aspect of the implant and the ridges of mucous membrane that radiate from the internal orifice of the streak canal (4), or a combination of these. CLINICAL IMPLICATIONS: Implantation of this vascular graft is a useful technique to restore teat patency for 6 months. A longer period of passive drainage of milk appears necessary for sufficient ingrowth of the graft. Lesions that extend to the distal most aspect of the teat cistern have a poor prognosis, because this area cannot be sufficiently overlapped by the graft.  相似文献

Supplementation of selenium and vitamin E to enhance disease resistance in dairy cattle has become common, particularly to prevent periparturient reproductive disorders and mastitis. To establish reference values for serum vitamin E and selenium concentrations in postparturient dairy cattle and to determine whether serum concentrations of these micronutrients varied with season and stage of lactation, cows from a stratified random sample of 50 herds were studied for 1 year. Blood samples were collected from each of the 50 study herds twice, from the 10 most recently parturient cows or from 10% of the herd, whichever was greatest. Mean concentration of vitamin E and selenium was 2.55 micrograms/ml and 78.12 ng/ml, respectively. Vitamin E concentrations were significantly (P < 0.05) higher during the summer and fall than during the winter and spring. Selenium concentrations were significantly (P < 0.05) lower during the summer and fall than during the winter and spring. Herd, season of blood sample collection, and time since parturition were significant (P < 0.02) in explaining variation in vitamin E and selenium concentrations.  相似文献
