Health effects associated with chronic, low-level exposures to arsenic in drinking water (<100 microg/L) remain unclear, in part due to uncertainties in assessing exposure. Drinking water concentrations have been used to assess past exposure to arsenic in epidemiological studies, under the assumption that a single measurement can be used to estimate historical exposure. This study aims to better understand (1) temporal variability in arsenic concentrations in drinking water and (2) the impact of point-of-use (POU) treatment devices on arsenic exposure measurements, and on reliability of the exposure measurement for population-level studies. Multiple drinking water samples were collected at two points in time (an average of fourteen months apart) for 261 individuals enrolled in a case-control study of arsenic exposure and bladder cancer in Michigan. Sources of drinking water included private wells (n = 221), public water supplies (n = 33), and bottled water (n = 7); mean arsenic concentration was highest in private wells (7.28 microg/L) and lowest in bottled water samples (0.28 microg/L). Arsenic concentrations in primary drinking water samples were highly correlated (r = 0.88, p < 0.0001, n = 196), with 3% of the water sources exceeding the United States Environmental Protection Agency's Maximum Contaminant Level (MCL) in one sample but not in the other sample. Measurement reproducibility did not vary by type of POU device (e.g., softener, filter, reverse osmosis system). Arsenic concentrations did differ, however, between samples treated with POU devices and untreated samples taken on the same day. Substantial differences in arsenic concentrations were consistently observed for reverse osmosis systems; other POU devices had variable effects on arsenic concentrations. These results indicate that while a single residential arsenic measurement may be used to represent exposure in this region, researchers must obtain information on changes in water source and POU treatment devices to better characterize population exposures over time.  相似文献

Laboratory studies were performed to determine if the addition of iron oxides throughout biosand filter (BSF) media would increase virus removal due to adsorption. The proposed mechanism is electrostatic adsorption of negatively charged virion particles to positively charged iron oxides formed during the corrosion of zerovalent iron. Initial tests conducted using continuous flow, small-scale glass columns showed high MS2 bacteriophage removal in an iron-amended sand column (5log10) compared to a sand-only column (0.5log10) over 20 pore volumes. Additionally, two experiments with a column containing iron particles revealed 4log10 and 5log10 removal of rotavirus in the presence of 20 mg/L total organic carbon. Full-scale BSFs with iron particles removed >4log10 MS2 for the duration of the experiment (287 days), while BSF with steel wool removed >4log10 MS2 for the first 160 days. Plug flow for the BSF was shown to depend on uniformity between the iron oxide material and sand media grains. The results suggest that the duration of effective virus removal by iron-amended biosand filtration depends on source water conditions and the quantity and composition of iron material added. Overall, this study provides evidence that iron-amended BSFs may advance the field of point-of-use technologies and bring relief to millions of people suffering from waterborne diseases.  相似文献

This study was designed to examine the potential downstream benefits of sequential disinfection to control the persistence of Escherichia coli under conditions relevant to drinking water distribution systems. Eight annular reactors (four polycarbonate and four cast iron) were setup in parallel to address various factors that could influence biofilm growth in distribution systems. Eight reactors were treated with chlorine, chlorine dioxide and monochloramine alone or in combination with UV to examine the effects on Escherichia coli growth and persistence in both the effluent and biofilm. In general, UV-treated systems in combination with chlorine or chlorine dioxide and monochloramine achieved greater log reductions in both effluent and biofilm than systems treated with chlorine-based disinfectants alone. However, during UV-low chlorine disinfection, E. coli was found to persist at low levels, suggesting that the UV treatment had instigated an adaptive mutation. During UV-chlorine-dioxide treatment, the E. coli that was initially below the detection limit reappeared during a low level of disinfection (0.2 mg/L) in the cast iron systems. Chloramine was shown to be effective in disinfecting suspended E. coli in the effluent but was unable to reduce biofilm counts to below the detection limit. Issues such as repair mechanism of E. coli and nitrification could help explain some of these aberrations. Improved understanding of the ability of chlorine-based disinfectant in combination with UV to provide sufficient disinfection will ultimately effect in improved management and safety of drinking water.  相似文献

Tap water quality was examined in six sampling events over a 2-year period in a residential community of Greater Cairo, Egypt. The sampling sites chosen had a granular activated carbon (GAC), reverse osmosis (RO) or distillation point-of-use (POU) treatment device so that the performance of these units for standard water quality parameters could be evaluated. Tap water satisfied Egyptian and international drinking water standards and guidelines for the majority of samples analysed. Exceptions were the violation of international limits for lead in 10 samples, and total trihalomethane (TTHM) levels that occasionally exceeded drinking water limits. Only the RO and distillation units achieved removal of total dissolved solids (TDS), alkalinity and hardness. However, GAC units are considerably less costly in the local market. All three POU units exhibited potential reduction of chemical contaminants such as trihalomethanes (THMs), improved taste via chlorine removal and improvement in clarity. Realization of these benefits, however, is conditioned upon proper operation and maintenance of the device.  相似文献

The spatial and temporal distributions of indicator bacteria in a small, multiple-use source drinking water reservoir in Southern California, USA were quantified over the period August 2001-July 2002. High levels of total and fecal coliform bacteria were present in Canyon Lake (annual geometric mean concentrations+/-SEM of 3.93+/-0.02 and 3.02+/-0.03 log cfu/100mL, respectively), while comparatively low levels of enterococci and E. coli were found (1.16+/-0.02 log cfu/100mL and 0.30+/-0.03 log MPN/100mL, respectively). As a result, these different indicator bacteria yielded quite divergent indices of water quality, with 72.1% of all surface samples (n=294) exceeding the USEPA single-sample limit of 400 cfu/100mL fecal coliform bacteria, while none (0%) of the samples exceeded the single-sample limit for E. coli (n=194). Regression analyses found a positive correlation between total and fecal coliform bacteria (R=0.50, significant at p<0.001) and between enterococci and E. coli (R=0.51, significant at p<0.001), but no correlation or inverse correlations were found between coliform concentrations and enterococci and E. coli levels. External sources responsible for the high total and fecal coliform bacteria were not identified, although laboratory studies demonstrated growth of the coliform bacteria in lake water samples. Enterococci and E. coli were not observed to grow, however. Bacteria concentrations varied relatively little laterally across the lake, although strong vertical gradients in fecal coliform and enterococcus bacteria concentrations were present during summer stratification, with concentrations about 10x higher above the thermocline when compared with surface concentrations. In contrast, total bacteria, total virus and total coliform bacteria levels were unchanged with depth. Seasonal trends in bacteria concentrations were also present. This study shows that the choice of indicator bacteria and sampling depth can both strongly affect the apparent microbial water quality of a lake or reservoir.  相似文献

Biological filters combining microbial activity and rapid sand filtration are used in drinking water treatment plants for enhanced biodegradable organic matters (BOM) removal. Biofilms formed on filter media comprised of bacteria enclosed in a polymeric matrix are responsible for the adsorption of BOM and attachment of planktonic microorganisms. This study investigated the removal of Escherichia coli cells injected into laboratory-scale biofilters and the role of biofilm in retaining the injected E. coli. Green fluorescent protein was used as a specific marker to detect and quantify E. coli in the biofilms. About 35% of the total injected E. coli cells were observed in the filter effluents, when initial cell concentrations were measured at 7.4 x 10(6) CFU/mL and 1.6 x 10(7) CFU/mL in two separate experiments. The results from real-time PCR and plate count analysis indicated that 95% of the E. coli retained inside the filters were either non-viable or could not be recovered by colony counting techniques. Injected cells were unevenly distributed inside the filter with more than 70% located at the top 1/5 of the filter. Images obtained from an epifluorescent microscope showed that E. coli cells were embedded inside the biofilm matrix and presented mainly as microcolonies intertwined with other microorganisms, which was consistent with findings from standard plate count methods and qPCR.  相似文献

Drinking water supply companies monitor the presence of Escherichia coli in drinking water to verify the effectiveness of measures that prevent faecal contamination of drinking water. Data are lacking, however, on the sensitivity of the monitoring programmes, as designed under the EU Drinking Water Directive. In this study, the sensitivity of such a monitoring programme was evaluated by hydraulic model simulations of contamination events and calculations of the detection probability of the actual sampling programme of 2002. In the hydraulic model simulations of 16-h periods of 1l h(-1) ingress of untreated domestic sewage, the spread of the contamination through the network and the E. coli concentration dynamics were calculated. The results show that when large parts of the sewage reach reservoirs, e.g. when they originate from the treatment plant or a trunk main, mean detection probabilities are 55-65%. When the contamination does not reach any of the reservoirs, however, the detection probability varies from 0% (when no sampling site is reached) to 13% (when multiple sites are reached). Mean detection probabilities of nine simulated ingress incidents in mains are 5.5% with an SD of 6.5%. In reality, these detection probabilities are probably lower as the study assumed no inactivation or clustering of E. coli, 100% recovery efficiency of the E. coli detection methods and immediate mixing of contaminations in mains and reservoirs. The described method provides a starting point for automated evaluations and optimisations of sampling programmes.  相似文献

The performance of a rotating biological contactor (RBC) for the post-treatment of the effluent of an up-flow anaerobic sludge blanket (UASB) was the subject of this study. Different hydraulic and organic loading rates have been investigated. The removal efficiencies of COD(total), COD(suspended), COD(colloidal) and COD(soluble) increased at a higher hydraulic retention time (HRT) and a lower influent organic loading rate. The results obtained indicated that a two-stage RBC reactor at an HRT of 10 h and an organic loading rate of 6.4g COD m(-2) d(-1) represents an effective post-treatment process. Most COD(suspended) and COD(colloidal) were removed in the first stage while nitrification proceeded in the second stage. The overall nitrification efficiency was 92% at an ammonia loading rate of 1.1 gm(-2) d(-1). Total E. coli removal at HRTs of 10, 5 and 2.5h were 99.5%, 99.0% and 89.0%. respectively. The major part of suspended E. coli ( >4.4 microm) was removed by sedimentation or by adsorption in the biofilm of the first stage of RBC (99.66%). However, E. coli in the colloidal fraction (<4.4 to >0.45 microm) was eliminated in the second stage of RBC (99.78%). A comparison of the performance of a one-stage versus two-stage RBC system, operated at the same total loading rate, revealed an improvement in the effluent quality of the two-stage effluent as compared to the one-stage effluent. The two stages RBC were used to examine the effect of hydraulic shock loads on reactor performance in terms of COD, nitrification and E. coli removal.  相似文献

The effects of sample storage on enumeration of Escherichia coli in marine bathing water and culturable bacteria in drinking water were evaluated. Results showed that overnight storage at 0-5 degrees C significantly reduced the counts of E. coli in bathing water (p = 0.0001) with a mean reduction of 25%. A similar effect of sample storage was observed for the enumeration of culturable bacteria in drinking water at 22 +/- 2 degrees C for 66 +/- 4 h (p = 0.0074; mean reduction = 25%) or at 36 +/- 2 degrees C for 44 +/- 4h (p = 0.0353; mean reduction = 6%). The use of a delayed incubation method, i.e. overnight storage at 0-5 degrees C of inoculated agar plates prior to incubation, did not significantly affect the counts of culturable bacteria when plates were incubated at 22 + 2 degrees C for 66 +/- 4 h, whereas it resulted in a significant increase of the bacterial numbers when plates were incubated at 36 +/- 2 degrees C for 44 +/- 4 h (p = 0.0002; mean increase = 32%). Based on these results, it is suggested to avoid the use of overnight or longer sample storage for the enumeration of E. coli in bathing water samples, as well as for the enumeration of culturable bacteria in drinking water. The delayed incubation method appears to be a reliable procedure for the enumeration of culturable bacteria and could represent a valid alternative to sample storage in order to overcome problems associated with the performance of bacteriological counts during weekends or statutory holidays. However, a multi-laboratory study is needed to evaluate the reproducibility of the delayed incubation method for the enumeration of culturable bacteria and its possible use for the enumeration of E. coli by membrane filtration.  相似文献

A significant consideration in forward planning for water treatment works design and operation concerns the effectiveness of a filtration plant in providing a barrier to particulates in the low micrometre size range, including Cryptosporidium oocysts. The performance of rapid gravity filtration plants is believed to be dependent on backwash and start-up regimes. It was the aim of this study to optimize direct sand filtration by identifying optimum filter backwash and start-up conditions which minimized the passage of particulates into the filtrate. The filter ripening period has long been identified as a cause for concern with respect to particulate passage into the filtrate; this work has shown that up to 40% of all particles that pass into supply during a 48 h run, do so in the first hour of operation. Optimum combined air water “collapse-pulsing” backwash durations were identified that reduced the number of 2–5 μm particles entering the filtrate, especially during the ripening period. Slow start-up was also found to reduce the number of 2–5 μm particles in the filtrate during the ripening period. The reductions in particulate passage resulting from a slow start was found to be media dependent, with smaller media requiring a longer slow start duration than coarser media.  相似文献

A rapid DNA extraction and quantitative, real time polymerase chain reaction (QRTPCR) analysis method targeting the ureA gene of Helicobacter pylori was evaluated for the measurement of these organisms on membrane filters at levels that might be expected to be found in drinking water samples. No interference was seen from high levels of background organisms and related, non-target species were detected at approximately 4-5 log(10) lower levels of sensitivity than H. pylori by this assay. A standard curve was generated for the method from analyses of filters containing known numbers of added H. pylori cells. Cell numbers on these filters were determined by staining with a species-specific fluorescent antibody and solid phase cytometry analyses. The mean detection sensitivity of the method was 10 H. pylori cells per filter with a 95% confidence sensitivity of 40 cells and a 95% confidence precision interval of +/-0.57 log(10) based on duplicate analyses of the samples. One liter drinking water samples from several locations in the US were inoculated with the same H. pylori cell suspensions used to generate the standard curve and gave measurements that were consistent with the standard curve suggesting that these sample matrices produced no interference in the method. This method may be useful for the rapid screening of drinking water for H. pylori.  相似文献

The biosand filter (BSF) is a household slow sand filter that is operated intermittently such that an idle time of typically 18-22 h occurs in between daily charges of water. Virus attenuation during the idle time was investigated over repeated daily filtration cycles to capture the effect of media aging that encompasses processes occurring throughout the filter depth rather than restricted to the schmutzdecke at the media surface. A threshold aging period of about one to two weeks was required before virus attenuation began. The observed rates of MS2 and PRD-1 reduction were first-order and reached maxima of 0.061- and 0.053-log per hr, respectively, over seven-to-ten weeks. Suppression of microbial activity by sodium azide eliminated virus reduction during the idle time thus indicating that the operative media aging process was microbially mediated. The mechanism of virus reduction was not modification of media surfaces by physical/chemical or microbial processes. Instead, it appears that the activity of the microbial community within the filter is responsible. The most likely biological pathways are production of microbial exoproducts such as proteolytic enzymes or grazing of bacteria and higher microorganisms on virus particles. Implications of these findings for BSF design and operation and their relevance to other biological filtration technologies are discussed.  相似文献

A two-factor three-block experimental design was developed to permit rigorous evaluation and modeling of the main effects and interactions of sand size (d10 of 0.17 and 0.52 mm) and hydraulic head (10, 20, and 30 cm) on removal of fecal coliform (FC) bacteria, MS2 bacteriophage virus, and turbidity, under two batch operating modes (‘long’ and ‘short’) in intermittent slow sand filters (ISSFs). Long operation involved an overnight pause time between feeding of two successive 20 L batches (16 h average batch residence time (RT)). Short operation involved no pause between two 20 L batch feeds (5 h average batch RT). Conditions tested were representative of those encountered in developing country field settings. Over a ten week period, the 18 experimental filters were fed river water augmented with wastewater (influent turbidity of 5.4-58.6 NTU) and maintained with the wet harrowing method. Linear mixed modeling allowed systematic estimates of the independent marginal effects of each independent variable on each performance outcome of interest while controlling for the effects of variations in a batch’s actual residence time, days since maintenance, and influent turbidity. This is the first study in which simultaneous measurement of bacteria, viruses and turbidity removal at the batch level over an extended duration has been undertaken with a large number of replicate units to permit rigorous modeling of ISSF performance variability within and across a range of likely filter design configurations and operating conditions.On average, the experimental filters removed 1.40 log fecal coliform CFU (SD 0.40 log, N = 249), 0.54 log MS2 PFU (SD 0.42 log, N = 245) and 89.0 percent turbidity (SD 6.9 percent, N = 263). Effluent turbidity averaged 1.24 NTU (SD 0.53 NTU, N = 263) and always remained below 3 NTU. Under the best performing design configuration and operating mode (fine sand, 10 cm head, long operation, initial HLR of 0.01-0.03 m/h), mean 1.82 log removal of bacteria (98.5%) and mean 0.94 log removal of MS2 viruses (88.5%) were achieved.Results point to new recommendations regarding filter design, manufacture, and operation for implementing ISSFs in local settings in developing countries. Sand size emerged as a critical design factor on performance. A single layer of river sand used in this investigation demonstrated removals comparable to those reported for 2 layers of crushed sand. Pause time and increased residence time each emerged as highly beneficial for improving removal performance on all four outcomes. A relatively large and significant negative effect of influent turbidity on MS2 viral removal in the ISSF was measured in parallel with a much smaller weaker positive effect of influent turbidity on FC bacterial removal. Disturbance of the schmutzdecke by wet harrowing showed no effect on virus removal and a modest reductive effect on the bacterial and turbidity removal as measured 7 days or more after the disturbance. For existing coarse sand ISSFs, this research indicates that a reduction in batch feed volume, effectively reducing the operating head and increasing the pore:batch volume ratio, could improve their removal performance by increasing batch residence time.  相似文献

Laboratory experiments were conducted on six point-of-use (POU) ceramic water filters that were manufactured in Nicaragua; two filters were used by families for ca. 4 years and the other filters had limited prior use in our lab. Water spiked with ca. 106 CFU/mL of Escherichia coli was dosed to the filters. Initial disinfection efficiencies ranged from 3 - 4.5 log, but the treatment efficiency decreased with subsequent batches of spiked water. Silver concentrations in the effluent water ranged from 0.04 - 1.75 ppb. Subsequent experiments that utilized feed water without a bacterial spike yielded 103-105 CFU/mL bacteria in the effluent. Immediately after recoating four of the filters with a colloidal silver solution, the effluent silver concentrations increased to 36 - 45 ppb and bacterial disinfection efficiencies were 3.8-4.5 log. The treatment effectiveness decreased to 0.2 - 2.5 log after loading multiple batches of highly contaminated water. In subsequent loading of clean water, the effluent water contained <20-41 CFU/mL in two of the filters. This indicates that the silver had some benefit to reducing bacterial contamination by the filter. In general these POU filters were found to be effective, but showed loss of effectiveness with time and indicated a release of microbes into subsequent volumes of water passed through the system.  相似文献

Filtration of drinking water by point-of-use (POU) or point-of-entry (POE) systems is becoming increasingly popular in the United States. Drinking water is filtered to remove both organic and inorganic contaminants. The objective of this study was to evaluate the use of granular activated carbon from nutshells (almond, English walnut, pecan) in a POU water filtration system to determine its effectiveness in removing select, potentially toxic metal ions, namely, copper (Cu2+), lead (Pb2+) or zinc (Zn2+) found in drinking water. The nutshell-based carbon system was designated "Envirofilter" and was compared to four commercial POU systems with brand names of BRITA, Omni Filter, PUR and Teledyne Water Pik. Eight prototype "Envirofilters", consisting of individual or binary mixtures of carbons made from acid-activated almond or pecan shells and steam-activated pecan or walnut shells were constructed and evaluated for adsorption of the three metal ions. The results indicated that a binary mixture of carbons from acid-activated almond and either steam-activated pecan or walnut shells were the most effective in removing these metals from drinking water of all the POU systems evaluated. Binary mixtures of acid-activated almond shell-based carbon with either steam-activated pecan shell- or walnut shell-based carbon removed nearly 100% of lead ion, 90-95% of copper ion and 80-90% of zinc ion. Overall the performance data on the "Envirofilters" suggest that these prototypes require less carbon than commercial filters to achieve the same metal adsorption efficiency and may also be a less expensive product.  相似文献

Survival of Escherichia coli O157:H7 in surface waters may increase the potential for dissipation of the organism to facilitate cycles of livestock re-infection and lead to human infection. Although previous studies have monitored survival of the organism in a range of surface waters, there is limited information on the influence of physico-chemical characteristics on persistence. Microcosms of four different surface water types (n=31) from the UK were inoculated with E. coli O157:H7 and incubated at 10 degrees C. The water types studied were: lake, puddle, river, and animal-drinking trough waters. Numbers of E. coli O157:H7 declined over time in all waters, although cells were still detected in 45% of non-sterile samples after 2 months. Persistence of E. coli O157:H7 was enhanced by water aeration and by prior sterilisation; however there was no correlation between water chemistry and mean E. coli O157:H7 die-off times or rates in any water type. Survival of the pathogen was better in lake and puddle waters than in river or drinking trough waters. Further studies are needed to establish the key water quality factors that regulate pathogen survival.  相似文献

Indigenous bacteria are essential for the performance of drinking water biofilters, yet this biological component remains poorly characterized. In the present study we followed biofilm formation and development in a granular activated carbon (GAC) filter on pilot-scale during the first six months of operation. GAC particles were sampled from four different depths (10, 45, 80 and 115 cm) and attached biomass was measured with adenosine tri-phosphate (ATP) analysis. The attached biomass accumulated rapidly on the GAC particles throughout all levels in the filter during the first 90 days of operation and maintained a steady state afterward. Vertical gradients of biomass density and growth rates were observed during start-up and also in steady state. During steady state, biomass concentrations ranged between 0.8-1.83 x 10−6 g ATP/g GAC in the filter, and 22% of the influent dissolved organic carbon (DOC) was removed. Concomitant biomass production was about 1.8 × 1012 cells/m2h, which represents a yield of 1.26 × 106 cells/μg. The bacteria assimilated only about 3% of the removed carbon as biomass. At one point during the operational period, a natural 5-fold increase in the influent phytoplankton concentration occurred. As a result, influent assimilable organic carbon concentrations increased and suspended bacteria in the filter effluent increased 3-fold as the direct consequence of increased growth in the biofilter. This study shows that the combination of different analytical methods allows detailed quantification of the microbiological activity in drinking water biofilters.  相似文献

The culturability of Escherichia coli in undersaturated drinking water with respect to CaCO3 (corrosive water) or in oversaturated water (non-corrosive water) was tested in different reactors: glass flasks (batch, "non-reactive" wall); glass reactors (chemostat, "non-reactive" wall) versus a corroded cast iron Propella reactor (chemostat, "reactive" wall) and a 15-year-old distribution system pilot (chemostat, "reactive" wall with 1% corroded cast iron and 99% cement-lined cast iron). The E. coli in E. coli-spiked drinking water was not able to maintain its culturability and colonize the experimental systems. It appears from our results that the optimal pH for maintaining E. coli culturability was around 8.2 or higher. However, in reactors with a reactive wall (corroded cast iron), the decline in E. coli culturability was slower when the pH was adjusted to 7.9 or 7.7 (i.e. a reactor fed with corrosive water; pHpHs). We tentatively deduce that corrosion products coming from chemical reactions driven by corrosive waters on the pipe wall improve E. coli culturability.  相似文献

The microbiological quality of drinking water from 144 private water supplies in the Netherlands was tested and additionally the occurrence of Escherichia coli O157 was examined. Faecal indicators were enumerated by using standard membrane filtration methods. The presence of E. coli O157 was determined using a specific enrichment method. Eleven percent of the samples contained faecal indicators whereas E. coli O157:H7 was isolated from 2.7% of the samples that otherwise met the drinking water standards. The E. coli O157 positive water supplies were located on camp-sites in agricultural areas with large grazer densities. Pulsed field gel electrophoresis (PFGE) analysis suggested that cattle might have been the cause of contamination. Our results indicate that compliance with microbiological quality standards obtained in routine monitoring does not always guarantee the absence of pathogens. The presence of pathogens such as E. coli O157 may suggest possible health consequences; however, a risk assessment process should be performed as the monitoring of both faecal indicator parameters and pathogens do not predict the effect of microbial contamination of drinking water on a population.  相似文献
