Seeds from two commercial bean varieties were cooked and stored for different times and analysed for chemical composition and in vitro starch digestibility. Parallel portions of cooked seeds were dried at 55 °C, milled and stored as flours. In general, protein and ash contents in both samples did not change with storage time, but statistical differences were shown between the two varieties (p < 0.05). Available starch (AS) contents in flours from the ‘negro’ variety did not change (p < 0.05) with storage time and, in general, were higher than in ‘flor de mayo’ samples, whose AS levels decreased during storage. The lower AS in ‘flor de mayo’ flour could be the consequence of formation of resistant starch due to retrogradation. Samples of whole ‘negro’ seeds did not show differences in AS content at 0, 24 and 48 h of storage compared with the corresponding flours, but at 72 and 96 h the AS increased in the whole samples. ‘Flor de mayo’ showed a similar pattern in flour and whole samples, with slightly higher values in the whole seeds. In general, total resistant starch (RS) content in the two varieties was higher in the flours than in ‘whole’ seeds, a fact that is not easy to explain at present. ‘Negro’ flour presented an RS content around 65.0 g kg?1, and approximately 55.0 g kg?1 was recorded in ‘flor de mayo’, with slight changes when storage time increased. Whole ‘flor de mayo’ showed significant levels of the retrograded portion of resistant starch (RRS), which did not change with storage time (p < 0.05). However, values were lower than in the flours. A pattern similar to that of the ‘negro’ variety was obtained for ‘flor de mayo’, since the flour exhibited higher amounts of RRS; however, in this variety, the RRS content in ‘whole’ samples decreased after prolonged storage. The effects of different soaking and cooking methods were investigated on available carbohydrate content and starch digestibility of red and white kidney beans. Total soluble sugars, reducing sugars, non-reducing sugars and starch contents of red and white kidney beans were 9.95 and 11.3%, 0.82 and 0.96%, 9.13 and 10.3%, and 44.4 and 47.8%, respectively. All these available carbohydrate components decreased to various extents as a result of soaking and cooking. From 2.51 to 13.6% and 7.03 to 28.0% of total soluble sugars were lost on soaking kidney beans in tap water and sodium bicarbonate solution, respectively. However, losses in total soluble sugars were maximum (19.9–60.9%) on cooking pre-soaked kidney beans. Losses in starch contents were 4.27 to 24.7% and 30.4 to 70.7% as a result of the soaking and cooking processes, respectively. Besides these losses, starch digestibility of kidney beans was also markedly improved as a result of cooking. However, no appreciable improvement in starch digestibility was observed after soaking kidney beans in water or alkaline solution.  相似文献

BACKGROUND: Beans are rich and inexpensive sources of proteins and carbohydrates around the world, but particularly in developing countries. However, many legume varieties are still underutilized. In this study, physical characteristics of the seeds of three Phaseolus lunatus cultivars were characterized. Also, the chemical composition and starch digestibility in the cooked beans were assessed. RESULTS: ‘Comba floja’ variety exhibited the highest thousand‐kernel weight whereas the lowest was found in ‘comba violenta’. This agrees with seed dimensions: ‘comba floja’ had the Longest seeds (16.36 mm) and ‘comba violenta’ the shortest ones (13.98 mm). All samples exhibited high protein content, but levels in ‘comba blanca’ variety (216 g kg?1) were lower than the in other two cultivars. Total starch (370–380 g kg?1) and potentially available starch content (330–340 g kg?1) were similar in the three varieties. Resistant starch level in the cooked seeds ranged between 38 and 45 g kg?1. Low enzymatic hydrolysis indices (HI) were recorded (30.2–35%), indicating a low digestion rate for Phaseolus lunatus starch. HI‐based predicted glycemic indices ranged between 34% and 39%, which suggests a ‘slow carbohydrate’ feature for this legume. CONCLUSION: Phaseolus lunatus beans appear to be a good source of protein and slow‐release carbohydrates with potential benefits for human health. The study investigated the effect of traditional soaking and cooking, storage after cooking and freezing (? 18 °C, 21 days) and autoclaving of two varieties of runner bean on starch digestibility. Results achieved were compared with digestibility of isolated starch subjected to similar treatments. The digestibility of native starch from Nata var. seeds was lower after isolation than in raw flour. This starch was characterized by a higher content of fat and lower values of swelling power (SP) and amylose leaching (AML). After the thermal treatment, a significantly higher content of rapidly digestible starch (RDS) was observed both in seeds and starch. It was accompanied by reduced contents of resistant starch (RS) and slowly digestible starch (SDS). In flours from cooked seeds, the content of RDS was observed to be higher than in flours from autoclaved seeds, despite similar changes in contents of other constituents (ash and protein). It was probably due to better starch gelatinization owing to the long-lasting soaking of seeds. This resulted in a greater decrease of amylose content of starch compared to the other flours. Differences in SP, AML and thermal properties between starches isolated from two bean varieties had no influence on their digestibility after cooking. The storage of starch pastes at a temperature of ? 18 °C, unlike that of seeds, resulted in a significant increase in RS content, which shows the importance of other flour components in the process of starch retrogradation.  相似文献

RS from mung bean starch was prepared by autoclaving, pullulanase debranching, and retrogradation. Physicochemical properties, crystalline structure, and in vitro digestibility of selected RS samples with different RS content were investigated. Compared to native starch, AAM content of RS increased but MW decreased greatly. SEM clearly showed RS samples exhibited irregular shaped fragments with compact structure. XRD pattern indicated that RS samples had typical B‐type pattern with sharp peaks at 17.0°, 22.2°, and 23.9° 2θ. The relative crystallinity, gelatinization temperatures, and enthalpy increased with increasing RS content. The α‐amylase digestibility of RS was lower than that of native starch. The results suggested that the decrease in enzymatic digestion of RS might due to compact and ordered crystalline structures after debranching and recrystallization.  相似文献

Volatile extracts of moderately off-flavoured frozen peas, prepared by vacuum sublimation followed by concentration of the sublimate by low temperature vacuum distillation, were examined by gas chromatography, silica gel chromatography and combined gas chromatography–mass spectrometry. The constituents identified included alcohols (29), carbonyls (31), esters (12), hydrocarbons (19), terpenes (6) and 3-alkyl-2-methoxypyrazines (3). Olfactory monitoring of the gas chromatographic column effluent indicated that most of the strong odours were associated with alkanals, alk-2-enals, alka-2,4- and 2,6-dienals, octa-3,5-dien-2-ones, 3-alkyl-2-methoxypyrazines and hexanol. However, no compound which, by itself, could account for the “hay-like” off-flavour of peas was evident. The off-flavour is considered to be complex and composed largely of the more odorous unsaturated carbonyls derived from the enzymic breakdown of the unsaturated pea lipids. The volatiles of pea shells similarly examined were found to contain many of the compounds present in peas.  相似文献

The nutritive value of transgenic peas expressing an α‐amylase inhibitor (α‐Ai1) was evaluated with broiler chickens. The effects of feeding transgenic peas on the development of visceral organs associated with digestion and nutrient absorption were also examined. The chemical composition of the conventional and the transgenic peas used in this study were similar. In the two feeding trials, that were conducted normal and transgenic peas were incorporated into a maize–soybean diet at concentrations up to 500 g kg?1. The diets were balanced to contain similar levels of apparent metabolisable energy (AME) and amino acids. In the first trial, the birds were fed the diets from 3 to 17 days post‐hatching and with levels of transgenic peas at 250 g kg?1 or greater there was a significant reduction in body weight but an increase in feed intake resulting in deceased feed conversion efficiency. In the second trial, in which the birds were fed diets containing 300 g kg?1 transgenic peas until 40 days of age, growth performance was significantly reduced. It was also demonstrated that the ileal starch digestibility coefficient (0.80 vs 0.42) was significantly reduced in the birds fed transgenic peas. Determination of AME and ileal digestibility of amino acids in 5‐week‐old broilers demonstrated a significant reduction in AME (12.12 vs 5.08 MJ kg?1 DM) in the birds fed the transgenic peas. The AME value recorded for transgenic peas reflected the lower starch digestibility of this line. Ileal digestion of protein and amino acids was unaffected by treatment. Expression of α‐Ai1 in peas did not appear to affect bird health or the utilisation of dietary protein. However, the significant reduction in ileal digestion of starch in transgenic peas does reduce the utility of this feedstuff in monogastric diets where efficient energy utilisation is required. In a series of 10-day pair feeding experiments it was found that the nutritional value of diets containing beans was essentially the same for rats aged between 30 and 123 days. Thus net protein utilisation (NPU) values of 25–39 on diets containing Processor bean (35 g protein kg?1) + egg albumin (65g protein kg?1) were obtained. As food intakes were considerably reduced when rats were fed diets containing more than 35g protein kg?1 of Processor bean, the measurement of protein utilisation became increasingly more difficult. The severe disruption of the brush borders of duodenal and jejunal enterocytes, originally observed when bean-containing diets were fed to young (30-day-old) rats was also found with rats up to the age of 120 days on similar diets. Similarly, the development of circulating anti-lectin antibodies in the animals showed no age dependence within the age limits investigated. It was also shown that oral immunisation did not protect the rats from the effects of toxicity and that the immune response was a result of continuous absorption of lectin throughout the feeding period. Thus the extent and the mechanism of toxicity of Phaseolus vulgaris bean lectins were found not to be dependent on the age or maturity of the animal.  相似文献

Chemical composition, mineral analysis, in vitro starch digestibility, and amino acid analysis of a kidney bean (Phaseolus vulgaris. L.), grown in Chongqing, China, were carried out in 2011. The results showed that the bean flour contained low reducing sugar (0.79%) and fat (2.72%), but high protein (24.8%) and starch (44.84%). Mineral analysis indicated high Mg and Ca contents with low Na/K ratio (0.04). The proportions of rapidly digestible starch (RDS), slowly digestible starch (SDS) and resistant starch (RS) were 39.64%, 41.46% and 18.90% respectively. And the variation in their proportions, along with prolonged heating time, showed increase in RDS, and decrease in SDS and RS, but the SDS + RS amount can still account for major parts (51.63%) of the total starch after a 90‐min pre‐cooking. Comparing the amino acid contents of the bean sample with recommended profile FAO/WHO/UNU (1985), the results fulfil the standard except threonine and lysine, whose contents are slightly lower than the requirements (0.93 and 0.85).  相似文献

Field peas (Pisum sativum), an important pulse crop in Australia, are consumed as human food either as whole seeds or as splits after decortication. The yield of splits is an important economic factor for processors and the cooking quality is important for consumers. Effects of genotype, other physical characteristics and pretreatment in various solutions on both dehulling and cooking quality were studied for 23 known genotypes and are market sample of unknown genotype. Large variations were found for most characteristics. Seed size was positively correlated with yield of splits while husk content and broken seeds were negatively correlated with yield. Preconditioning seeds in salt solutions improved yield of splits. Variation in cooking quality among genotypes was reduced following splitting and cooking time was reduced by decortication and splitting and by presoaking in salt solutions (10 or 20 g kg−1), particularly with sodium tripolyphosphate. Loss of seed material into cooking water was correlated with cooking time. No characteristic was found that could be used to predict cooking time. © 1998 SCI.  相似文献

The objective of the present study was to investigate the effect of drought on the metabolism of proline, ornithine and polyamines, and the enzymes involved in their synthesis and degradation, in tepary and common bean seeds. Furthermore, the effect on seed yield, protein content and in vitro protein digestibility was also studied. Genotypes of tepary bean (Phaseolus acutifolius) used were: PI‐301‐801 from Nicaragua; G‐400‐35 from Oaxaca, Mexico; L‐242‐24 and L‐242‐36 from Sonora, Mexico; L‐172 from Arizona, USA; and the common bean (Phaseolus vulgaris) variety Flor de Mayo Bajío (FMB) from Mexico. The cultivars were grown in a greenhouse under controlled temperature, light and humidity. Water content in plants (WCP) decreased when irrigation was stopped (1 and 2 weeks after blooming). Common bean had the lowest WCP value (73.75%), while the best genotypes were PI‐301‐801 (91.62%), G‐400‐35 (88.70%) and L‐242‐24 (85.73%). Tepary bean plants had superior water absorption capacity, which helped to increase yield. De novo synthesis of drought proteins may explain the increased protein content present under drought conditions. The results suggest that water stress induced ornithine amino transferase activity and ornithine concentration. Proline also increased under drought conditions (p < 0.05). Genotypes L‐242‐24 and G‐400‐35, which had the highest yield, also had a major concentration of polyamines under water stress. The results also suggest that arginase activity participated in amino acid synthesis such as ornithine and, consequently, in the protein concentration under water stress. Genotypes G‐400‐35 and L‐242‐24 are good alternatives for cultivation in arid zones. Copyright © 2003 Society of Chemical Industry  相似文献

Protein content was used as an indicator of environmental conditions for a study on varietal and environmental variation in proximate composition, minerals, amino acids and certain antinutrients of field peas. Four field pea varieties, each with three levels of protein content, were selected. Crude protein content overall ranged from 20.2 to 26.7%. Analysis of variance showed that both variety and environmental conditions had a significant effect on starch, acid detergent fibre (ADF), neutral detergent fibre (NDF) and fat content, but ash content was only affected by variety. Significant varietal and environmental differences in potassium (K), manganese (Mn) and phosphorus (P) were noted. Calcium (Ca) and copper (Cu) showed significant varietal differences, while iron (Fe), magnesium (Mg) and zinc (Zn) had significant environmental differences. Environmental conditions showed significant effects on alanine, glycine, isoleucine, lysine and threonine content. Variety had a significant effect on sucrose, raffinose and phytic acid content, whereas environmental conditions had an influence on trypsin inhibitor activity (TIA). The major pea components protein and starch were inversely correlated. ADF, NDF, Fe, Mg, Zn and the amino acid arginine were positively correlated with protein content. The amino acids glycine, histidine, isoleucine, lysine and threonine were negatively correlated with protein content. It was found that tryptophan was the most deficient amino acid and the sulphur‐containing amino acids were the second limiting amino acids in peas. Raffinose was positively correlated with sucrose but negatively correlated with verbascose. There were significant correlations between mineral contents and some of the proximate components. Saline extract solutions from seeds of both Phaseolus vulgaris and Vicia faba varieties were prepared and tested for their total polyphenolic content and effect on intestinal D -Glucose transport across rat ileum in vivo. Intestinal D -glucose transport was significantly reduced (P < 0.001) by the presence of seed-extracts in the intestinal loop, regardless of the source of the extract. The order of potency (Phaseolus vulgaris, var. artropurpurea > Vicia faba, var. caballar > Vicia faba, var. minor > Phaseolus vulgaris, var. albus) is closely related to the polyphenolic content. Clear differences in the mode of action of the extracts were also observed. Extracts with appreciable polyphenolic content caused both a reversible and a lasting inhibitory effect on glucose transport, whilst extracts with negligible polyphenolic content only induced a reversible inhibition. Polyamide treatment of the extracts decreased their ability to inhibit glucose transport, but their inhibitory effect was not completely abolished by the treatment, further suggesting the presence of some other anti-absorptive factor(s) in the seeds.  相似文献

Glycoprotein II, the major storage protein of the seeds of kidney bean (Phaseolus vulgaris), was largely resistant to hydrolysis by pure gut endopeptidases. However, the protein was extensively digested in vitro and nearly 90% of it became soluble in 5% trichloroacetic acid by treatments first with extracts of rat stomach followed by intestinal contents or alternatively first with pepsin, followed by pancreatin. Heat denaturation (100°C for 10 min) had no appreciable effect on the digestion. A similarly high extent (between 80 and 90%) and rate of digestion of pure Glycoprotein II was observed in vivo. However, intubation of pure Glycoprotein II into the stomach of previously fasted rats increased the production of insoluble intestinal secretions containing relatively large amounts of nitrogen. As this N passed into the caecum, it reduced both the apparent digestibility of Glycoprotein II and the net gain of N for the animal. Moreover, in the presence of a mixture of raw bean proteins in the stomach of rats the time taken to empty its content into the intestine was increased appreciably. Additionally, the toxic effects of the lectins in these mixtures, particularly on continuous feeding, were observed.  相似文献

Weight changes, food intake, in-vivo intestinal absorption of D -galactose (2.77mm) and L -leucine (5 mM) and in-vitro intestinal absorption of d-galactose (2 mm) were studied in male growing chickens fed ad libitum from hatching for periods of 21 and 49 days on 210 g kg?1 protein diets containing either heated soya bean (HSB, control) with or without added tannic acid (25 and 30 g kg?1) or raw kidney bean (RKB, Phaseolus vulgaris L) as the main sources of protein. As compared with HSB-fed birds, weight gain was significantly smaller (P <0.05) in the HSB + tannic acid- and RKB-fed birds; these chickens ate significantly more (P <0.05) than HSB-fed birds. Regardless of the number of days of feeding, both HSB + tannic acid- and RKB-fed birds showed a significant reduction (P <0.05) in the rates of in-vivo intestinal absorption of galactose and leucine and in-vitro intestinal absorption of galactose, as compared with HSB-fed chickens, these effects being especially marked in the RKB-fed birds. The possible implications of these findings are discussed.  相似文献

‘Phospholipid acyl-hydrolase’ (PLAH), an enzymic activity releasing fatty acid from phosphatidylcholine (PC), has been identified and characterised in green peas. The Km value for PC dipalmitoyl ester was 0·167 mm. The enzymic activity possessed a pH optimum of 5·6 and was stable for 20 min only at that pH value. The optimum temperature was 45°C and thermal sensitivity was indicated by a 94% decrease in activity upon exposure of the enzyme to 55°C for 3 min, and by an exponential decrease in activity upon storage at 4°C for 1 week. The enzyme was optimally activated by 2·0 mm calcium chloride at pH 5·6, and the optimal concentration of sodium dodecyl sulphate was 0·75 mg ml?1. Pea PLAH was non-competitively inhibited by sodium cyanide, EDTA and p-chloromercuribenzoate, with no activity in the presence of mercuric chloride. The results from this study are related to those of other workers on lipid-degrading enzymes in peas, and a pathway is proposed for the enzymic degradation of endogenous lipids in fresh or unblanched frozen peas during post-harvest storage.  相似文献
