Tryptophan (Trp) is an amino acid and an essential component of the human diet. It plays a crucial role in many metabolic functions. Clinicians can use Trp levels in the course of diagnosing various metabolic disorders and the symptoms associated with those diseases. Furthermore, supplementation with this amino acid is considered in the treatment of depression and sleep disorders, mainly due to the Trp relationship with the synthesis of serotonin (5-HT) and melatonin. It is also used in helping to resolve cognitive disorders, anxiety, or neurodegenerative diseases. Reduced secretion of serotonin is associated with autism spectrum disorder, obesity, anorexia and bulimia nervosa, and other diseases presenting peripherals symptoms. The literature strongly suggests that Trp has a significant role in the correct functionality of the brain-gut axis and immunology. This information leads to the consideration of Trp as an essential dietary component due to its role in the serotonin pathway. A reduced availability of Trp in diet and nutraceutical supplementation should be considered with greater concern than one might expect. This paper constitutes a review of the more salient aspects gleaned from the current knowledge base about the role of Trp in diseases, associated nutritional disorders, and food science, in general.  相似文献

Foodborne viruses, especially noroviruses (NoV), are increasingly reported as the cause of foodborne outbreaks. NoV outbreaks have been reported linked to fresh soft red fruits and leafy greens. Belgium, Canada and France were the first countries to provide data about the prevalence of NoV on fresh produce. In total, 867 samples of leafy greens, 180 samples of fresh soft red fruits and 57 samples of other types of fresh produce (tomatoes, cucumber and fruit salads) were analyzed. Firstly, the NoV detection methodology, including virus and RNA extraction, real-time RT-PCR and quality controls were compared among the three countries. In addition, confirmation and genotyping of the NoV strains was attempted for a subset of NoV positive samples using conventional RT-PCR targeting an alternative region followed by sequencing. Analysis of the process control showed that 653, 179 and 18 samples of the leafy greens, soft red fruits and other fresh produce types were valid for analysis based on the recovery of the process control. NoV was detected by real-time RT-PCR in 28.2% (N = 641), 33.3% (N = 6) and 50% (N = 6) of leafy greens tested in Canada, Belgium and France, respectively. Soft red fruits were found positive by real-time RT-PCR in 34.5% (N = 29) and 6.7% (N = 150) of the samples tested in Belgium and France, respectively. 55.5% (N = 18) of the other fresh produce types, analyzed in Belgium, were found NoV positive by real-time RT-PCR. Conventional RT-PCR resulted in an amplicon of the expected size in 19.5% (52/266) of the NoV positive samples where this assay was attempted. Subsequent sequencing was only successful in 34.6% (18/52) of the suspected amplicons obtained by conventional RT-PCR. From this study, using the described methodology, NoV genomes were frequently detected in fresh produce however sequence confirmation was not successful for the majority of the samples tested. Infection or outbreaks were rarely or not known to be related to the NoV positive samples. With the increase in sensitivity of the detection methodology, there is an increasing concern about the interpretation of positive NoV results by real-time amplification. Strategies to confirm the results by real-time RT-PCR should be developed in analogy with the detection of microbial pathogens in foods. Detection might indicate contact with NoV in the fresh produce chain. Consequently, a potential risk for infection cannot be excluded but the actual risk from RT-PCR NoV positive produce is still unknown. Studies should be designed determining the probability of infection related to the presence or levels of NoV genomic copies.  相似文献

Protein oxidation has become a topic of great scientific interest in the field of food science and nutrition. Food proteins are known to be preferential targets of radical species, and protein oxidation has relevant consequences on protein functionality and food quality. Current trends in this field call attention to the nutritional and health dimensions of oxidized foods. Both lipid and protein oxidation products are accumulated in food during processing and storage and also upon food intake during the subsequent digestion phases. The gastrointestinal tract and internal organs are exposed to the cytotoxic and mutagenic potential of these species. While the molecular basis of the pathogenesis of particular dietary lipid oxidation products is well known, the impact of dietary oxidized proteins on human health has been largely ignored. The well-established association between in vivo protein oxidation and aging and age-related diseases urges scientists to investigate the contribution of dietary protein oxidation to particular pathological conditions. Recent reports indicate the involvement of dietary protein oxidation species on particular health disorders, which emphasizes the link between dietary and in vivo protein oxidation.  相似文献

The carotenoid pigment ‘canthaxanthin’ has been used in aquafeed for many years in order to impart the desired flesh colour in farmed salmonids. A side effect of extreme over-dosage of this carotenoid is the deposition of minute crystals in the eye, a fact leading to adverse media attention for canthaxanthin in the past, and some pressure to limit its use aquafeeds. A real appraisal of the possible amount of canthaxanthin ingested by humans through the consumption of notable quantities of highly pigmented salmon, reveals that it is extremely unlikely that consumers could be exposed to sufficient canthaxanthin to attain the established ADI for this pigment. Additionally, the ADI is 10 times lower than the reported no-effect level (NOEL) for canthaxanthin. Therefore, canthaxanthin is a safe source of carotenoids for salmonid feeds.  相似文献

The major allergen parvalbumin was purified from cod muscle tissues, and polyclonal antibodies were raised towards it. The antibodies were tested for specificity and an enzyme-linked immunosorbent assay (ELISA) was developed using these antibodies. The ELISA was applied to measure parvalbumin in cod skin, the starting material for fish gelatin made from deep sea, wild fish. The ELISA was sufficiently sensitive (LLOQ?=?0.8?ng?ml?1 in extracts, corresponding to 0.02?µg of parvalbumin per g of tissue), and did not cross-react with common food constituents. Fish gelatin, wine and beer, matrices for the potential use of this ELISA, did not cause disturbance of the assay performance. The data show that the parvalbumin content in cod muscle tissue is 6.25?mg?g?1, while the skins contained considerably less, 0.4?mg?g?1. Washing of the skins, a common industrial procedure during the manufacturing of fish gelatin, reduced the level of parvalbumin about 1000-fold to 0.5?µg?g?1, or 0.5?ppm. From 95 commercial lots of fish gelatin it is shown that 73 are below 0.02?µg?g?1 parvalbumin. From the other 22 lots, the one with the highest concentration contained 0.15?µg?g?1 of parvalbumin. Brominated flame retardants – polybrominated diphenyl ethers (PBDEs), polybrominated biphenyls (PBBs), hexabromocyclododecane (HBCD) and others – have been measured in 11 pooled breast milk samples from 109 first-time mothers in Ireland. Additionally, the study has measured levels of polybrominated dibenzo-p-dioxins and furans (PBDD/Fs), mixed halogenated dioxins (PXCC/Fs) and biphenyls (PXBs), polychlorinated naphthalenes (PCNs) and perfluoroalkylated substances (PFAS) in these samples. The mean sum of 19 PBDEs including BDE-209 was 4.85 ng g?1 fat, which is comparable with that found in other European countries. BDE-47, BDE-153, BDE-209, BDE-99 and BDE-100 were found at the highest concentrations. The only PBBs detected consistently were BB-77, BB-126 and BB-153, with highest concentrations being found for BB-153 (mean = 0.13 ng g?1 fat). The mean sum of HBCD enantiomers was 3.52 ng g?1 fat, with α-HBCD representing over 70% of the total. Of the other brominated flame retardants – tetrabromobisphenol-A (TBBP-A), hexabromobenzene (HBB), decabromodiphenylethane (DBDPE) and bis(2,4,6-tribromophenoxyethane) (BTBPE) – examined, only TBBP-A was detected above the limit of detection (LOD), in two of the 11 pools analysed. All measured PBDF congeners were observed (at 0.02–0.91 pg g?1 fat), but 2,3,7,8-tetrabromo-dibenzodioxin (TeBDD) was the only PBDD detected, with a mean concentration of 0.09 pg g?1 fat. The occurrence of the mixed chlorinated/brominated dibenzodioxins, dibenzofurans and biphenyls, 2-B-3,7,8-CDD, 2,3-B-7,8-CDF, 4-B-2,3,7,8-CDF, PXB 105, PXB 118, PXB 126 and PCB 156 in breast milk in the current study may indicate that levels of these contaminants are increasing in the environment. Polychlorinated naphthalenes were detected in all samples, but not perfluorooctane sulfonate (PFOS) and other PFAS. The pattern of occurrence of these brominated and fluorinated persistent organic pollutants (POPs) in Irish breast milk shows a general relationship to their occurrence in food, as reported in a number of surveillance studies carried out by the Food Safety Authority of Ireland.  相似文献

Seafood frequently contains high concentrations of arsenic (approximately 10-100 mg/kg dry weight). In marine algae (seaweed), this arsenic occurs predominantly as ribose derivatives known collectively as arsenosugars. Although it is clear that arsenosugars are not acutely toxic, there is a possibility of arsenosugars having slight chronic toxicity. In general, trivalent arsenicals are more toxic than their pentavalent counterparts, so in this work we examine the hypothesis that trivalent arsenosugars might be significantly more toxic than pentavalent arsenosugars in vitro. We compared the in vitro toxicity of (R)-2,3-dihydroxypropyl-5-deoxy-5-dimethylarsinoyl-beta-D-riboside, a pentavalent arsenosugar, to that of its trivalent counterpart, (R)-2,3-dihydroxypropyl-5-deoxy-5-dimethylarsino-beta-D-riboside. The trivalent arsenosugar nicked plasmid DNA, whereas the pentavalent arsenosugar did not. The trivalent arsenosugar was more cytotoxic (IC50 = 200 microM, 48 h exposure) than its pentavalent counterpart (IC50 > 6000 microM, 48 h exposure) in normal human epidermal keratinocytes in vitro as determined via the neutral red uptake assay. However, both the trivalent and the pentavalent arsenosugars were significantly less toxic than MMA(III), DMA(III), and arsenate. Neither the pentavalent arsenosugar nor the trivalent arsenosugar were mutagenic in Salmonella TA104. The trivalent arsenosugar was readily formed by reaction of the pentavalent arsenosugar with thiol compounds, including, cysteine, glutathione, and dithioerythritol. This work suggests that the reduction of pentavalent arsenosugars to trivalent arsenosugars in biology might have environmental consequences, especially because seaweed consumption is a significant environmental source for human exposure to arsenicals.  相似文献

In most present modeling of the sediment-water flux of hydrophobic organic chemicals (HOCs), the flux due to sediment erosion/deposition is described explicitly by means of a sediment transport model, while the remaining components of the flux (molecular diffusion, bioturbation, and groundwater flow) are lumped together and modeled as a "diffusive" flux. This diffusive flux is usually described by means of a mass transfer approximation with the implicit assumption of a well-mixed contaminant layer of thickness h in the sediments. On the basis of recent experiments and theoretical modeling, the justification forthis assumption and the quantification of this diffusive flux are discussed here. In particular, for HOCs with large partition coefficients, it is argued that a well-mixed layer often may not exist, and, when it does, it is slow to form; that h is difficult to define and even harder to quantify; and that, as far as long-term predictions (up to 100 years) of this diffusive flux are concerned, the exact value for h probably does not matter. What does matter are the magnitudes and time dependencies of each of the components of the flux and the interactions between the diffusive flux and the flux due to erosion/deposition.  相似文献

Livestock poisoning, primarily liver damage, caused by consumption of plants containing 1,2-dehydropyrrolizidine ester alkaloids (dehydroPAs), and the corresponding N-oxides, is a relatively common occurrence worldwide. Because of the economic impact, extensive investigations of such episodes have been performed, particularly in Australia, South Africa the United States and, more recently, South America. Plant species most commonly involved are members of the families Boraginaceae, Asteraceae and Leguminosae. These may be native species that periodically flourish under particular climatic conditions or introduced species that thrive in the absence of natural control factors such as herbivory and competition. Contamination of grain crops with dehydroPA-producing plants has resulted in large-scale incidents of food poisoning in humans, with high morbidity and mortality, especially in Africa and in central and south Asia, with recent episodes in Afghanistan and possibly Ethiopia. Attention has recently focused on the potential for low levels of dehydroPAs to contaminate many food products in developed countries, possibly leading to progressive, chronic diseases that may not include overt hepatotoxicity. This overview examines the potential for better control of exposure and means of monitoring dehydroPA intake by extrapolation of knowledge gained from animal studies to the human situation.  相似文献

In the UK agriculture is by far the largest source of nitrous oxide (N(2)O) emissions. Direct N(2)O emissions as a result of nitrogen (N) application to soils have been well documented in the UK, whereas indirect emissions produced in surface waters and groundwaters from leached N are much less understood with limited data to support IPCC emission factors. Indirect emissions were studied in surface waters in the Upper Thurne, a lowland drained arable catchment in eastern England. All surface waters were found to have dissolved N(2)O concentrations above that expected if in equilibrium with ambient concentrations, demonstrating all surface waters were acting as a source of N(2)O. The drainage channels represented 86% of the total indirect N(2)O flux, followed by wetland areas, 11%, and the river, 3%. The dense drainage network was found to have the highest dissolved N(2)O concentrations of all the water bodies studied with a combined N(2)O flux of 16 kg N(2)O-N per day in March 2007. Such indirect fluxes are comparable to direct fluxes per hectare and represent a significant proportion of the total N(2)O flux for this catchment. Separate emission factors were established for the three different surface water types within the same catchment, suggesting that the one emission factor used in the Intergovernmental Panel on Climate Change (IPCC) methodology for predicting all indirect N(2)O emissions is inappropriate.  相似文献

Oxyphytosterols are similar to oxycholesterols in structure, and they exhibit pro-atherogenic properties. Recently, more interests were focused on the metabolism of oxyphytosterols for their increasing intake from phytosterol-enriched food. In this review, we discussed the origin, absorption, distribution, and transport of oxyphytosterols in vivo and their biological effects in humans. The two dominant oxyphytosterols in human plasma are 7-keto-sitosterol and 7-keto-campesterol, but their origins are unclear. It is suggested that oxysitosterols are formed to eliminate sitosterol from tissue to the blood stream. Aside from the pro-atherogenic, oxyphytosterols also exhibit pro-inflammatory properties and antiviral activity against equine herpesvirus 1. Further research is needed to investigate the physiological and pathological role of oxyphytosterols in humans.  相似文献

Antioxidants, preferentially those of dietary origin, have for a long time been considered to help against diseases that are presumably aggravated by oxidative stress, such as cardiovascular diseases, cancer, and neurodegenerative disorders. The outcome of clinical trials undertaken to corroborate this hypothesis, however, remained largely inconclusive. Evidence is now emerging that some dietary "antioxidants" influence signaling pathways and the expression of genes relevant in atherosclerosis by mechanisms other than antioxidative ones. By concrete examples we show that (1) vitamin E has gene regulatory functions which might be more important than acting as an antioxidant in vivo, (2) selenium itself is not an antioxidant at all, and even not in general when incorporated into glutathione peroxidases, and (3) a moderate oxidative stress is beneficial rather than detrimental since it can induce defense mechanisms counteracting xenobiotic and oxidative stress. Thus, there is only a future for antioxidants in the prevention of any disease if their real mechanism of action is considered and suitable read-outs and biomarkers are established.  相似文献
