This study examined the effect of hydrolysis of casein by added plasmin (6 mg L−1) on the heat stability of raw, pre-heated, serum protein-free or concentrated skim milk. Plasmin activity markedly affected the heat stability–pH profile of skim milk and serum protein-free milk, apparently by altering the properties of the casein micelles. It is probable that changes in the surface charge of the micelles, as a result of the hydrolysis of caseins, contributed to this effect. Hydrolysis by plasmin reduced the zeta-potential of the casein micelles from ∼−19 to ∼−16 mV. The effect of hydrolysis of casein by plasmin on the heat stability of pre-heated milk was less pronounced, shifting the heat stability–pH profile to more alkaline values; the heat stability of concentrated milk was unaffected by plasmin. A very high (50 mg L−1) level of added plasmin resulted in clearing of the skim milk; the L* value decreased from ∼75 to ∼35 after 24 h incubation at 37 °C. Clearing was correlated with a change in casein micelle diameter from an initial value of ∼175 to ∼250 nm. It is suggested that plasmin-induced changes in zeta-potential may promote micellar aggregation or changes in micelle stucture.  相似文献

The impact of thermosonication (TS) and pulsed electric field (PEF), individually and combined, on the survival of Listeria innocua 11288 (NCTC) in milk was investigated. TS (400 W, 160 s) without pre-heating reduced L. innocua by 1.2 log10 cfu mL?1, while shorter treatment times produced negligible inactivation, suggesting TS to be a hurdle rather than an effective standalone treatment. PEF (30 and 40 kV cm?1, 50 μs) at 10 °C caused a reduction of L. innocua of 1.1 and 3.3 log cycles, respectively. The highest field strength (40 kV cm?1) combined with TS (80 s) led to 6.8 log10 cfu mL?1 inactivation. Milk pre-heated to 55 °C (over 60 s) prior to TS followed by PEF (30 and 40 kV cm?1) showed inactivation between 4.5 and 6.9 log10 cfu mL?1, the latter being comparable (P > 0.05) with thermal pasteurisation. The data indicate that TS followed by PEF represents a valid alternative for L. innocua inactivation in milk.  相似文献

The mechanism of high-pressure (HP)-induced changes in rennet coagulation properties of milk, particularly the role of whey protein-casein micelle associations, was studied. Treatment at 100 or 250 MPa reduced the rennet coagulation time (RCT) of raw skimmed bovine milk, compared with untreated milk. Treatment at 400 MPa had little effect, but at 600 MPa, RCT increased considerably. HP-induced increases in RCT did not occur in serum protein-free milk or milk treated with the sulphydryl-oxidising agent KIO3, which prevents association of denatured β-lactoglobulin with casein micelles. Treatment at 5 or 10 °C at 250–600 MPa resulted in shorter RCT than treatment at 20 °C. In milk without KIO3, coagulum strength was highest after treatment at 250 or 400 MPa, whereas in milk with KIO3 it was highest after treatment at 400 MPa. These results indicate the significance of HP-induced association of whey proteins with casein micelles for rennet coagulation properties of milk.  相似文献

The effects of pulsed electric field (PEF) treatments at field intensities of 25–37 kV cm 1 and final PEF treatment temperatures of 15 °C and 60 °C on the inactivation of alkaline phosphatase (ALP), Total Plate Count (TPC), Pseudomonas and Enterobacteriaceae counts were determined in raw skim milk. At 15 °C, PEF treatments of 28 to 37 kV cm 1 resulted in 24–42% inactivation in ALP activity and < 1 log reduction in TPC and Pseudomonas count, while the Enterobacteriaceae count was reduced by at least 2.1 log units to below the detection limit of 1 CFU mL 1. PEF treatments of 25 to 35 kV cm 1 at 60 °C resulted in 29–67% inactivation in ALP activity and up to 2.4 log reduction in TPC, while the Pseudomonas and Enterobacteriaceae counts were reduced by at least 5.9 and 2.1 logs, respectively, to below the detection limit of 1 CFU mL 1. Kinetic studies suggested that the effect of field intensity on ALP inactivation at the final PEF treatment temperature of 60 °C was more than twice that at 15 °C. A combined effect was observed between the field intensity and temperature in the inactivation of both ALP enzyme and the natural microbial flora in raw skim milk.Industrial relevanceMilk has been pasteurised to ensure its safety and extend its shelf life. However, the need for retaining heat-sensitive nutrient and sensory properties of milk has resulted in interest in the application of alternative technologies. The results of the current study suggest that PEF as a non-thermal process can be employed for the treatment of raw milk in mild temperature to achieve adequate safety and shelf life while preserving the heat-sensitive enzymes, nutrients and bioactive compounds.  相似文献

Production of goat milk cheese in North America has been growing rapidly during the past several years. However, information on chemical composition and its seasonal variation of year-round bulk-collected goat milk is limited. The objective of this study was to analyze the chemical composition of commercial goat milk shipments for an entire year to provide fundamental information for cheese making and milk cheese yielding potential and pricing. Samples were collected weekly from bulk milk shipments to a commercial cheese company over 12 mo, beginning in April, 1996, and analyzed for contents (%) of total solids (TS), fat (F), lactose, crude protein, casein, nonprotein nitrogen (NPN), ash, minerals, and specific gravity (G). Chemical composition of the goat milk varied widely during the year. The contents of fat and TS decreased over the first 20 wk from 3.6 and 12.7% to 3.0 and 11.3%, respectively, and then increased to peak values of 13.4 and 4.4% in January. Crude protein and casein contents also decreased over the first 20 wk, from 3.5 and 2.7% to 3.2 and 2.3%, respectively, before increasing gradually to 3.8 and 2.9% in February. The concentration of lactose seemed to decrease below mean levels during August and January. Ash content declined during the first 20 wk from 0.82 to 0.78%, and then increased sharply to 0.90% by wk 36 before decreasing sharply again toward the end of the study. Calcium content decreased steadily from about 0.16 to 0.14% by wk 20, before increasing to 0.16% by around wk 40. It was found that TS content could be estimated using the equation: TS = 0.13 G + 1.41 F + 4.28 (r2 = 0.94, P < 0.01).  相似文献

An experiment was undertaken to investigate the effect of milk fat level (0%, 2.5% and 5.0% w/w) and gel firmness level at cutting (5, 35 and 65 Pa) on indices of syneresis, while curd was undergoing stirring. The curd moisture content, yield of whey, fat in whey and casein fines in whey were measured at fixed intervals between 5 and 75 min after cutting the gel. The casein level in milk and clotting conditions was kept constant in all trials. The trials were carried out using recombined whole milk in an 11 L cheese vat. The fat level in milk had a large negative effect on the yield of whey. A clear effect of gel firmness on casein fines was observed. The best overall prediction, in terms of coefficient of determination, was for curd moisture content using milk fat concentration, time after gel cutting and set-to-cut time (R2 = 0.95).  相似文献

Whole raw milk was processed using a 15 L h−1 homogeniser with a high-pressure (HP) valve immediately followed by a cooling heat exchanger. The influence of homogenisation pressure (100–300 MPa) and milk inlet temperature Tin (4°C, 14°C or 24°C) on milk temperature T2 at the HP valve outlet, on fat globule size distribution and on the reduction of the endogenous flora were investigated. The Tin values of 4–24°C led to milk temperatures of 14–33°C before the HP valve, mainly because of compression heating. High Tin and/or homogenisation pressure decreased the fat globule size. At 200 MPa, the d4.3 diameter of fat globules decreased from 3.8±0.2 (control milk) to 0.80±0.08 μm, 0.65±0.10 or 0.37±0.07 μm at Tin=4, 14°C or 24°C, respectively. A second homogenisation pass at 200 MPa (Tin=4°C, 14°C or 24°C) further decreased d4.3 diameters to about 0.2 μm and narrowed the size distribution. At all Tin tested, an homogenisation pressure of 300 MPa induced clusters of fat globules, easily dissociated with SDS, and probably formed by sharing protein constituents adsorbed at the fat globule surface. The total endogenous flora of raw milk was reduced by more than 1 log cycle, provided homogenisation pressure was ⩾200 MPa at Tin=24°C (T2∼60°C), 250 MPa at Tin=14°C (T2∼62°C), or 300 MPa at Tin=4°C (T2∼65°C). At all Tin tested, a second pass through the HP valve (200 MPa) doubled the inactivation ratio of the total flora. Microbial patterns of raw milk were also affected; Gram-negative bacteria were less resistant than Gram-positive bacteria.  相似文献

Pulsed Electric Field (PEF) treatment of milk provides the opportunity to increase the shelf-life of fresh milk for distribution to distant markets. PEF treatments were evaluated in sterile (UHT) milk to determine the inactivation of added spoilage Pseudomonas isolates and the subsequent gains in microbial shelf-life (time taken to reach 107 CFU mL 1). Little inactivation of Pseudomonas was achieved at 15 or 40 °C compared with 50 or 55 °C. The greatest inactivation (> 5 logs) was achieved by processing at 55 °C with 31 kV cm 1 (139.4 kJ L 1). Heat treatment at the application temperature without PEF treatment caused minimal inactivation of Pseudomonas (only 0.2 logs), demonstrating that the inactivation of the Pseudomonas was due to the PEF treatment rather than the heat applied to the milk. At added Pseudomonas levels of 103 and 105 CFU mL 1, the microbial shelf-life of PEF-treated milk was extended by at least 8 days at 4 °C compared with untreated milk. The total microbial shelf-life of the PEF-treated milk was 13 and 11 days for inoculation levels of 103 and 105 CFU mL 1 respectively. The results indicate that PEF treatment is useful for the reduction of pseudomonads, the major spoilage bacteria of milk.Industrial relevancePseudomonads are the major psychrotrophic spoilage microflora of refrigerated, stored HTST pasteurised milk. Long-life (UHT) products are an important component of milk sales in South-East Asia, but in recent years there has been an increasing demand for less processed milk products with extended shelf-life. The recent practice of shipping fresh bulk milk from Australia to South-East Asian countries has necessitated additional heat treatment prior to export and on arrival, to achieve the required shelf-life. Pulsed electric field treatment of HTST milk, applied alone or in combination with mild heat under optimised conditions, offers the opportunity of shelf-life extension, while limiting the reduction in quality attributes of milk associated with more severe additional heat treatments.  相似文献

The anti-bacterial effect of high pressure homogenisation (HPH) on milk is widely reported but the shelf-life of HPH-treated milk, as reported in this communication has not been studied thus far. Raw whole milk was homogenised at 200 or 250 MPa at 55 or 70 °C and counts of total bacteria (TBC), psychrotrophs, pseudomonads, coliforms, lactobacilli, Bacillus cereus and Staphylococcus aureus were determined throughout subsequent storage for 14 days at 4 °C. Immediately after HPH treatment, counts of all bacteria were below the level of detection but after storage for 14 days at 4 °C, TBC, psychrotroph and pseudomonad counts had reached ∼108 cfu mL−1 in all samples treated with HPH. The limited shelf-life obtained indicates that HPH of milk at these processing parameters it is not a suitable alternative to pasteurisation for extending the shelf-life of milk.  相似文献

A set-type fermented milk manufactured from goat's milk was developed. Optimal curd tension was achieved by supplementation of milk with skim milk powder and whey protein concentrate (WPC). Milk was fermented employing a commercial probiotic starter culture (ABT-2), which contained Streptococcus thermophilus ST-20Y, Lactobacillus acidophilus LA-5, and Bifidobacterium BB-12. Supplementation of milk with 3% WPC reduced fermentation time by 2 h due to the increase in viable counts of S. thermophilus and Bifidobacterium by 0.3 and 0.7 log units, respectively. Addition of WPC increased the protein content (1%) as well as potassium and magnesium content (0.3 and 0.02 g kg−1, respectively). Increase of the protein content led to an increase in the apparent viscosity and gel firmness of the product, and at the same time whey syneresis was reduced. As a consequence, the product received a high score for appearance, taste, aroma, texture and overall acceptance.  相似文献

The influence of transglutaminase (TGase)-induced cross-linking on the ethanol stability of skimmed milk was investigated. The stability of milk against ethanol-induced coagulation increased in sigmoidal fashion with milk pH (5.0–7.5) for all samples; ethanol stability also increased upon incubation (0–24 h) with 0.05 g L−1 TGase at 30 °C. In untreated milk, addition of ethanol induced a collapse of the polyelectrolyte brush of κ-casein on the micelle surface, thereby facilitating micellar aggregation. Dynamic and static light scattering measurements indicated that in TGase-treated milk, the ethanol-induced collapse of the polyelectrolyte brush was far less than in untreated milk, suggesting that the increased ethanol stability of TGase-treated casein micelles is caused by the cross-linking of the polyelectrolyte brush on the micellar surface.  相似文献

Buffalo milk proteins (casein, co-precipitate or whey protein concentrate) were phosphorylated with phosphorus oxychloride (POCl3) at three different pH values (5.0, 7.0 and 9.0). The solubilities of phosphorylated milk proteins were examined over the pH range 3.0–9.0 in water and ionic (0.1 m NaCl or 10–70 mm Ca2+) systems. The solubilities of buffalo milk proteins decreased at pH 3.0, while there was an increase in the solubilities of casein and co-precipitate near their isoelectric points upon phosphorylation. Solubilities of these phosphorylated milk proteins were pH dependent in 0.1 m NaCl but there was a decrease in their solubilities with increase in calcium ion concentration. This alteration could be due to the shifting of isoionic points of phosphorylated buffalo milk proteins towards acidic pH.  相似文献

The combined effect of high-pressure (HP) treatment and bacteriocin-producing lactic acid bacteria (BP-LAB) on the survival of Listeria monocytogenes Scott A in cheeses made from raw milk that was inoculated with the pathogen at 4.80 log cfu mL−1, a commercial starter and one of seven strains of BP-LAB was investigated. On day 3, the counts of L. monocytogenes were 7.03 log cfu g−1 in a control cheese (without BP-LAB, not HP treated), 6.06–6.74 log cfu g−1 in cheeses with BP-LAB, 6.13 log cfu g−1 in a cheese without BP-LAB and treated on day 2 at 300 MPa, 2.01 log cfu g−1 in a cheese without BP-LAB and treated on day 2 at 500 MPa, 3.83–5.43 log cfu g−1 in cheeses with BP-LAB and treated on day 2 at 300 MPa, and 1.81 log cfu g−1 or less in cheeses with BP-LAB and treated on day 2 at 500 MPa. Listeria monocytogenes and Escherichia coli O157:H7 are major foodborne pathogens implicated in various outbreaks involving pasteurized or unpasteurized milk, and various dairy products. The objective of this study was to determine the antibacterial effect of caprylic acid (CA, C8:0) and its monoglyceride, monocaprylin (MC) on L. monocytogenes and E. coli O157:H7 in whole milk. A five-strain mixture of E. coli O157:H7 or L. monocytogenes was inoculated in autoclaved milk (106 CFU/ml) containing 0, 25, or 50 mM of CA or MC. At 37°C, all the treatments, excepting 25 mm CA, reduced the population of both pathogens by approximately 5.0 log CFU/ml in 6 h. At 24 h of storage at 8°C, MC at both levels and CA at 50 mM decreased L. monocytogenes and E. coli O157:H7, respectively by >5.0 log CFU/ml. At 48 h of 4°C storage, populations of L. monocytogenes and E. coli O157:H7 were decreased to below detection level (enrichment negative) by 50 mm of MC and CA, respectively. Results indicate that MC could potentially be used to inhibit L. monocytogenes and E. coli O157:H7 in milk and dairy products, but sensory studies need to be conducted before recommending their use.  相似文献

Cryoconcentration combined with a cascade effect was used to concentrate skim milk up to 25.12% total dry matter. Size, shape, and inter-micellar distance of casein micelles were characterized by ZetasizerNano-ZS, transmission electron microscopy, and ImageJ analyses. Flow properties of the cryoconcentrated skim milk were evaluated during 5 weeks of storage under refrigerated condition at 4 °C. Milk color was also evaluated according to the L*, a*, and b* system. The cryoconcentrated skim milk obtained after three cryoconcentration cycles was characterized by a monomodal distribution of its micelles with a tendency to smaller casein micelles. Approximately 60% of the total micellar volume was occupied by the casein micelles with a size of 100–200 nm, less than 18% of the volume with a size of 50–100 nm and only less than 1% was occupied by micelles with a size > 350 nm. This result shows that cryoconcentration changed the distribution of the mean size of the casein micelles to smaller units. No significant difference was observed on the inter-micellar distance. Cryoconcentration significantly improved the color of skim milk by increasing the L* value up to 67 which was similar to that of whole milk. Transition from a Newtonian to a non-Newtonian behavior was observed from the fourth week storage with a slight increase of casein micelle size.Industrial relevanceA concentration procedure of skim milk based on a complete block cryoconcentration technique was proposed. Application of this sub-zero technology permitted the concentration of skim milk total dry matter up to 25%. The casein micelle size was positively affected by moving the major part of the micelles toward the smaller size, whereas the inter-micellar distance was not affected. This new knowledge can be exploited in milk-based products to enhance the product stability. The cryoconcentrated skim milk color was positively affected since its L* value, which represents the milk whiteness, was significantly improved. The flow behavior of the cryoconcentrated milk was of Newtonian type up to 4 weeks of storage at 4 °C. The generated knowledge in this study can be easily used by the milk processing industry in order to make stable milk product with high dry matter content without adding milk powder, which negatively affects the product sensory properties (floury consistency).  相似文献

Studies of the formation of soluble κ-casein/whey protein (WP) complexes in heated (90 °C 10 min−1) milk and related mixtures of proteins have been made. The use of milk samples containing different genetic variants, and having different compositions, allowed the effects of changing the natural protein balance on the formation of particles to be investigated. In addition, studies were made of the effects of addition of WP or of purified κ-casein to the milk samples. The addition of WP caused an increase in the amount and the size of the complexes, but addition of κ-casein to the milk had little or no effect on the complex formation, nor did it seem that the added κ-casein could react with the WP in the milk. Conversely, in systems where the casein micelles were absent, the purified κ-casein reacted well with WP, suggesting that in milk heated at its normal pH the WP react preferentially with the κ-casein on the casein micelles.  相似文献

The present work discusses the efficiency of pulsed electrical treatments for the inactivation of yeasts. The application of pulsed electric fields (PEFs) and high voltage electrical discharges (HVEDs) as alternatives to sulfites, which are used as anti-microbial to stop the fermentation of sweet white wine, was investigated. The influence of sulfite concentration (from 0 mg·L 1 to 500 mg·L 1), PEF (from 4 kV·cm 1 to 20 kV·cm 1; from 0.25 ms to 6 ms) and HVED (40 kV/cm; 1 ms or 4 ms) treatments on the inactivation of total yeasts and non-Saccharomyces yeasts was determined. The addition of SO2 (250 mg·L 1) resulted in 8 log total yeast reduction. The maximum yeast inactivation obtained with PEF and HVED was respectively 3 and 4 logs. The use of SO2, HVED and PEF allows decreasing the non-Saccharomyces yeast level by 7, 5 and 4 logs respectively. However, the wine browning was less pronounced for the samples treated by PEF in comparison with HVED and SO2 treatments. PEF seems to be the most suitable alternative technique to sulfite addition.  相似文献

Lactobacillus plantarum ST8KF, isolated from kefir, produced a 3.5 kDa bacteriocin (bacST8KF) active against Lb. casei, Lb. salivarius, Lb. curvatus and Listeria innocua. BacST8KF was sensitive to proteolytic enzymes, but stable between pH 2.0 and 10.0, and heat resistant (20 min at 121 °C). BacST8KF did not adsorb to the surface of the producer cell. Maximum activity (25,600 AU mL−1) was recorded in MRS broth with glucose, in MRS broth with glucose replaced by sucrose, and in MRS broth with glucose, supplemented with KH2PO4 after 24 h at 30 °C. Tri-ammonium citrate and glycerol in excess of 5.0 g L−1 repressed bacST8KF production. Production of bacST8KF increased from 800 AU mL−1 after 3 h of fermentation in MRS broth at 30 °C to 12,800 AU mL−1 after 9 h and to 51,200 AU mL−1 after 27 h. These results suggest that bacST8KF may be a secondary metabolite and shows that its mode of activity is bacteriostatic.  相似文献

In this study, transglutaminase was used to produce cross-linked casein, gelatin and casein–gelatin blend (100:0, 75:25, 50:50, 25:75 and 0:100) edible films. Cross-linking was investigated by SDS–PAGE. Mechanical and water vapor barrier properties of the films were characterized using ASTM procedures, and the film morphology was evaluated using scanning electron microscopy. The casein–gelatin film showed significant greater elongation values (P < 0.05) with or without transglutaminase treatment, as compared to films made from gelatin or casein alone. Mixtures of casein and gelatin produced a synergistic effect only observed in the film elongation, while no improvement was detected for tensile strength and water vapor barrier properties, except for the casein:gelatin (75:25) formulation with transglutaminase, which showed the lowest water vapor permeability value (5.06 ± 0.31 g mm/m2 d kPa). Enzymatic cross-linking also induced a substantial increase in the high molecular weight protein components in the film forming solutions.  相似文献

The proteolysis of casein (CN) occurring in packaged pasteurized milk (PM) during refrigerated storage was studied with relation to hygienic and microbiological characteristics of starting raw milk. Six batches of raw milk having standard plate count (SPC) from 1.5×104 to 2.5×105 cfu mL−1 and somatic cell count (SCC) from 1.6×105 to 4.4×105 units mL−1 were pasteurized (73 °C for 15 s), packaged and stored at 4 °C for 12 days. Capillary zone electrophoresis of CN showed breakdown of β-CN in all PM samples during storage. An HPLC method for monitoring proteose peptones (PP) formation was developed. Level of PP in PM samples increased, with keeping time from 667–789 to 947–1383 mg L−1 and PP formation was significantly (P<0.05) related to SCC of starting raw milk. Electrospray ionization–mass spectrometry showed that PP were mainly represented by PP-5 from either A1 or A2 variants of β-CN. Five commercial samples of PM were analysed for PP formation during 14-day storage at 4 °C. Commercial samples prepared by microfiltration process or bactofugation combined with pasteurization showed the slowest formation of PP. The effect of storage temperature on PP formation was evaluated by keeping a conventional PM sample at either 8 or 12 °C for 12 days. Proteolysis of all major CNs upon action of plasmin and bacterial proteinases was observed under these conditions. PP level thus proves to be a reliable analytical index for evaluating the ageing of packaged PM during refrigerated storage.  相似文献
