Soymilk fermented with 2 Lactobacillus casei strains were stored at various temperatures (80 °C, 4 °C, 24.8 °C and 37 °C) for 8 weeks and isoflavone concentration analysed at weekly intervals using RP-HPLC. The degradation of each isoflavone compound at each storage temperature was found to fit first order kinetic model. Aglycone as well as glucosides generally appeared to be stable during storage (P < 0.01) at the 4 storage temperatures. Aglycone forms had smaller degradation constants compared to glucosides at all storage temperature and in the presence of both microorganisms. Specifically, aglycones showed a unique trend of smaller degradation at lower storage temperatures (−80 °C and 4 °C) than at higher temperatures (24.8 °C and 37 °C). Glucoside genistin was least stable at all storage temperatures compared to other isoflavones in the fermented soymilk with each strain while aglycone daidzein was the most stable. L. casei 2607 in fermented soymilk stored at 4 °C after 8 weeks gave the least degradation for daidzein of a mere 3.78% loss from 9.53 to 9.17 ng/μL. L. casei 2607 showed greater hydrolytic potential than L. casei ASCC 290 as denoted by higher degradation of isoflavone glucosides in fermented soymilk at lower storage temperatures.  相似文献

The aim of this work was to study how heat treatment, the type of starter culture, incubation temperature, and storage time can affect the physicochemical characteristics of stirred yoghurt. A four-factor experimental design was used for data analysis. Yoghurt milk was heated at 95 °C for 5 min or 130 °C for 80 s. Yoghurts were produced with three different starter cultures that had been incubated at 37, 42 or 45 °C and stored at 4 °C for 15 days. Visual roughness, number of grains, perimeter of grains, storage modulus, and yield stress all decreased when heating temperature was increased, when an exopolysaccharide-producing starter culture was used, or when incubation temperature was decreased. Storage time did not affect any of the physicochemical properties of yoghurt, except for the pH.  相似文献

The survival parameters of Escherichia coli O157:H7 during milk fermentation (carried out by the LIM or “longer incubation method” at 30 °C, or by the SIM or “short incubation method” at 43 °C) and storage of home-made yoghurt at refrigeration temperatures (2, 4, or 8 °C) were studied. The E. coli O157:H7 counts increased slightly during fermentation by the LIM, from 5.1 to 5.4 log cfu mL−1, and it was not found after 21 d of storage at 2 or 4 °C, and after 10 d at 8 °C. The microorganism counts increased from 4.8 to 5.4 log cfu mL−1 during the SIM, and it was not detected after 7 d stored at 8 °C. The microorganism grew faster at 43 °C (generation time=0.93 h) than at 30 °C (4.12 h) during the fermentation period. The death time decreased with the increase of the storage temperature (from 38.1 h at 2 °C to 30.1 h at 8 °C) in the yoghurt produced by fermentation at 30 °C; however, a clear relationship between death time and storage temperature was not evident at 43 °C. The pH values of the yoghurt ranged from 4.0 to 4.7.  相似文献

The effect of inulin addition (0–4%) upon texture and microstructure of set yoghurt with different levels of fat (0.2%–3.5%) was investigated. A two-factor experimental design with four treatments was used for data analysis. Skimmed milk with various inulin and cream concentrations was standardized to 4% protein content, homogenized, heated to 92 °C and fermented at 42 °C until a pH of 4.6 was reached. The chemical composition, pH, consistency and microstructure properties of the yoghurts were analysed after 6 days of storage at 5 °C. The statistical analysis showed that inulin and fat significantly affected the rheological and sensory results. Higher yield stress, “firmness” and “creaminess” values were observed in yoghurt produced with higher inulin additions, whereas the pH value was not affected. A significant correlation was found between yield stress and sensory determined firmness (r = 0.91). The microstructure examined by confocal laser scanning microscopy (CLSM) was only slightly affected by the concentrations of inulin in the range studied, possibly due to weak protein interactions between the inulin and the milk protein network.  相似文献

The aim of this work was to study how heat treatment and casein (CN) to whey protein (WP) ratio of skim milk affect physical characteristics of stirred yoghurt. Different heat treatments (95 °C/256 s, 110 °C/180 s and 130 °C/80 s) were applied to the yoghurt milk with the CN to WP ratios of 1.5:1, 2:1, 3:1 and 4:1. Physical properties, including graininess and roughness, of stirred yoghurt were determined during storage at 4 °C for 15 days. Visual roughness, number of grains, perimeter of grains, storage modulus, and yield stress decreased, when heating temperature or CN to WP ratio increased.  相似文献

The survival of probiotic microorganisms including Lactobacillus acidophilus, Lactobacillus casei, Lactobacillus rhamnosus and Bifidobacterium spp. was evaluated in yoghurt and freeze-dried yoghurt after processing and storage. The effectiveness of microencapsulating probiotic organisms as well as adding cryoprotectants and prebiotics in improving their viability was also investigated. The viability of Bifidobacterium infantis 17930 and L. rhamnosus GG was reduced by 0.07 log, while that of L. casei 1520 and Bifidobacterium longum 1941 was reduced by 0.28 and 0.39 log, respectively. There was a 7% improvement in the viability of L. casei 1520 when cryoprotectant ‘Unipectine™ RS 150’ was added at 2.5% (w/v). The prebiotic ‘Raftilose®P95’ when added at 1.5% w/v to yoghurt improved the viability of the combined selected probiotic organisms by 1.42 log during four weeks of storage at 4 °C. Microencapsulation with alginate improved viability of combined selected probiotic organisms by 0.31 log in freeze-dried yoghurt stored at 21 °C.  相似文献

Experimentally prepared Jerusalem artichoke inulins (JAI) were compared with two commercial chicory root inulins for their prebiotic potentials in media broth model and growth-sustaining ability in non-fat yoghurts. Experimental yoghurts were made with 12% reconstituted skim milk (RSM) supplemented with 4% inulin powders, inoculated with mixed cultures of Lactobacillus casei LC-01, Streptococcus thermophilus and Lactobacillus delbrueckii subsp. bulgaricus (1:0.5:0.5 based on supplier’s recommendation) and incubated overnight at 37 °C. Non-supplemented yoghurt was prepared from 16% RSM and used as control. The survival and acidifying activity of lactic and probiotic cultures in all yoghurts were investigated on weekly intervals during the shelf life of 28 days at 4 °C. Incorporation of JAI resulted in improved viability of LC-01, maintaining >7.0 log CFU/g during cold storage but did not affect the viability of yoghurt bacteria in comparison with the control.  相似文献

The mechanical properties and microstructure of 2.7% and 4.5% sodium caseinate gels chemically acidified by glucono-δ-lactone (GDL) and cross-linked by microbial transglutaminase (TG) were studied. The acidification was performed at different temperatures. According to SDS–PAGE TG clearly caused polymerisation of caseinate irrespective of the treatment temperature (4–50 °C), The cross-linking of the proteins was more extensive at temperatures 22–50 °C. Low amplitude viscoelastic measurements showed that 4.5% caseinate gels acidified at 50 °C were formed much faster than gels acidified at 22 °C. TG only slightly increased the time of gelling. Control gels prepared without TG at temperatures of 4, 22, 37 and 50 °C were mechanically weak. Examination of the control gels with a confocal laser scanning microscope showed that gels formed at 37 and 50 °C were coarse and porous with large cavities between particle aggregates, whereas those formed at 22 °C were much more homogeneous. The TG-treated and acidified sodium caseinate dispersions formed firm gels, indicating cross-linking of casein proteins. Interestingly, the strongest gels were formed at 22 and 37 °C. TG treatment improved the homogeneity of the gel structure at temperatures of 37 and 50 °C. The hardness of TG-treated gels acidified at 4 °C increased during 1 week of storage.  相似文献

The influence of temperature on the growth and biological activity of two probiotic strains (Bifidobacterium longum CRL 849 and Lactobacillus fermentum CRL 251) as pure and mixed cultures in soymilk (SM) were evaluated. Maximum growth was observed at 37°C in both mixed and pure cultures. In a product prepared with the mixed culture (1:1) at 37°C, the amount of lactic acid produced was approximately 55 mmol l−1 after 24 h with a slow production rate (2.8 mmol l−1 h−1); the formation of acetic acid was higher with respect to pure cultures (82.01 mmol l−1 after 24 h), and final pH (24 h) was 5.0. About 85% of the total amount of sugars in SM was reduced, mainly sucrose. Stachyose was reduced (71%) after 4 h of incubation. Maximum activity of alpha-galactosidase (alpha-gal) (13.2 U ml−1) was observed after 6 h. At 37°C the bifidobacterium strain was viable in mixed culture throughout the period assayed. At lower (30°C) or higher (42°C) temperatures, mixed culture showed slower growth and lower acid production in SM but the alpha-gal activity was stimulated at 30°C.  相似文献

Study on the gelatinization kinetics of rice showed that gelatinization process was divided into two steps: swelling of the amorphous region and disruption of the crystalline region. Higher temperature storage (37 °C) resulted in an increase in the breaking point temperature suggesting that energy for the disorder of these two regions of starch in rice stored at 37 °C was higher than the rice stored at 4 °C. Storage-induced changes in rice led to significant increases in DSC peak temperature (p < 0.05) and significantly broadened peak width (p < 0.01) for rice stored at 37 °C compared to rice stored at 4 °C. As the peak temperature of rice stored at 37 °C was not influenced by the “annealing” treatment in contrast with the increased peak temperature of rice stored at 4 °C after the “annealing” treatment, the results indicate that the ageing process (37 °C storage) has already re-ordered the rice grain structure and that the annealing process under these conditions has no further effect on starch thermal properties. Because starches isolated from rice grain stored at 4 °C and 37 °C had similar thermal properties, this implies that the effects of storage on thermal properties are associated with the interactions between starch and non-starch components following storage. The gelatinization endotherm shifted to a lower temperature (p < 0.01) and a narrowed peak width was achieved after cellulase and protease treatments of stored rice, which indicates that the changes in cell wall remnants and proteins are responsible for the changes in rice thermal properties during storage. Scanning electron microscopy was applied to visualize the treatments of cellulase and protease on rice.  相似文献

Lactobacillus fermentum Lf2, an autochthonous strain isolated as a non starter culture in Cremoso cheese, produces high EPS levels (~ 1 g/L) in optimized conditions (SDM broth, pH 6.0, 30 °C, 72 h). Technological (texture profile and rheological analysis) and sensory properties of non-fat yogurts with 300 and 600 mg EPS/L were studied at 3 and 25 days after manufacture. Yogurts with different EPS concentrations showed higher hardness values than the control group at both periods of time, being the only significant difference that remained stable during time. The consistency index was also higher for the treated samples at both times evaluated, being significantly different for samples with 300 mg/L of EPS extract, while the flow behavior index was lower for EPS-added yogurts. The thixotropic index was lower (P < 0.05) for samples with the highest EPS extract concentration at the end of the storage time. Regarding the sensory analysis, those yogurts with 600 mg/L of EPS extract presented the highest values of consistency at 3 days of storage. No considerable differences for defects (milk powder, acid, bitter and cooked milk flavors) were perceived between treated and control samples at both times evaluated. Syneresis was also studied and samples with 600 mg/L of EPS extract presented the lowest syneresis values at 25 days of storage, which considerably decreased with the time of storage.In conclusion, the EPS from L. fermentum Lf2, used as an additive, provided yogurt with creamy consistency and increased hardness, without the presence of unwanted defects and improving the water holding capacity of the product. All the analysis done showed the potential of this extract to be used as a technofunctional natural ingredient, and it should be considered its positive impact on health, according to previous studies.  相似文献

Physical properties of stirred yoghurt made from reconstituted skim milk that was high-pressure (HP)-treated at 100, 250 or 400 MPa, at 25, 70 or 90 °C, for 10 min, prior to inoculation with yoghurt cultures, were studied; portions of milk HP-treated at 25 °C were also heat-treated at 90 °C for 10 min before or after pressure treatment. Control yoghurts were made from skim milk given a heat treatment at 90 °C for 10 min. Fermentation time was not affected by treatment applied to the milk. HP treatment of skim milk at 25 °C, before or after heat treatment, gave stirred yoghurts of similar viscosities to that made from conventionally heat-treated milk. Lower viscosities were obtained when stirred yoghurts were made with milk HP-treated at elevated temperatures. A model is proposed to correlate properties of yoghurt with HP/heat-induced changes in interactions and structures of protein in the milk samples.Industrial relevanceTo meet end user expectations, the dairy industry needs to diversify its product range by tailoring specific functionalities. To meet these expectations, new processing methods such as high-pressure processing are of interest for their potential to achieve specific and/or novel functionalities and/or improve efficiencies, including reduced chemical and water use. Volatile carbonyl compounds, organic acids and yoghurt bacteria counts were investigated in yoghurts made from ultra-high pressure homogenized milk. Yoghurts were manufactured from milk treated using ultra-high pressure homogenization at 200 or 300 MPa and at 30 °C or 40 °C, and compared with those produced from heat-treated milk with 3% added skim milk powder. To study the evolution of these parameters, samples were analysed after days 1, 14 and 28 of storage. Yoghurts from milk heat-treated or treated at 300 MPa had very similar profiles of organic acids and volatile compounds, as well as similar bacterial counts of both starter cultures. In comparison, yoghurts from milk treated at 200 MPa at either 30 °C or 40 °C gave different profiles, together with a sharp decrease in counts of lactobacilli. During storage, only slight differences in flavour compounds and yoghurt bacteria counts were detected, except in those samples from milk treated at 200 MPa.  相似文献

The current work used fingerprinting-kinetics for the first time to monitor shelf-life changes in a low-pH, pasteurised, shelf-stable product, more particular in orange juice. Orange juice samples were stored as a function of time at four different storage temperatures (20, 28, 35 and 42 °C). To obtain insight into chemical changes in the volatile food fraction, samples were fingerprinted with headspace GC–MS. The objectives of this work were twofold: (i) to identify major chemical changes of pasteurised orange juice during shelf-life and (ii) to study the kinetics of selected shelf-life compounds in the context of accelerated shelf-life testing (ASLT). At 20 °C, changes in terpenes and a decrease in aldehydes were observed. Oxides and sulphur compounds increased and esters decreased at increased storage temperatures (at 28 °C and above). Concerning ASLT, four volatile compounds had clear temperature and time dependent kinetics within the investigated temperature range.  相似文献

In the present study we investigated the influence of heat treatment of lupin-based (LB) milk alternatives and different exopolysaccharide (EPS)-producing lactic acid bacteria on the physical characteristics of set-type LB yogurt alternatives. LB milk alternatives, obtained from protein isolate of Lupinus angustifolius cv. Boregine, were either pasteurized at 80 °C for 60 s or ultra-high temperature (UHT) heated at 140 °C for 10 s and was fermented with Lactobacillus plantarum TMW 1.460 and 1.1468, Pediococcus pentosaceus BGT B34 and Lactobacillus brevis BGT L150. Fermentation duration was strongly affected by heat treatment: different strains needed between 25 to 35 h in UHT LB milk alternative to reach a pH of 4.5 compared to 14 to 24 h in pasteurized LB milk alternative. EPS extraction revealed slightly higher amounts of EPS for UHT LB yogurt alternatives (~ 0.5–0.9 g/l; pasteurized: ~ 0.4–0.7 g/l). The more intensive heat treatment (UHT) resulted also in better rheological (apparent viscosity, hysteresis loop area, flow point, elastic, viscous and complex modulus) and textural properties (firmness, consistency, cohesiveness and index of viscosity) of the investigated LB yogurt alternatives. Furthermore, LB yogurt alternatives out of UHT milk alternative revealed a lower tendency to syneresis, measured with siphon and centrifugation method. This work contributes to the fundamental knowledge of the textural properties of LB yogurt alternatives.  相似文献

The rheological properties of stirred yoghurt were studied as a function of the delay between milk heat-treatment and inoculation (0, 1 and 2 days), of pH in the acid gel on stirring (4.4, 4.7, and 5.0), of the storage temperature (4, 12, and 20 °C) for 24 h following stirring and of over-acidification (allowed or inhibited). At low pH values, the gels exhibited higher elastic modulus (G′) and fracture strength. They yielded stirred yoghurts with higher G′ and viscosity, and higher increase in G′ and viscosity during storage (“rebodying”). Rebodying was only partially explained by over-acidification and cooling. Changing the storage temperature had no impact on the evolution of G′ after stirring; hydrophobic interactions were therefore probably not involved in rebodying. Electrostatic interactions seemed to play a major role in rebodying, as pH on stirring was the significant factor.  相似文献

The effects of high pressure on the physicochemical, chemical, microbiological and sensory characteristics of stirred low-fat yoghurt were studied. Laboratory-made yoghurts were treated at high pressure (100–400 MPa) for 15 min at 20°C. No significant changes in pH and total organic acids were observed after pressuring the yoghurt. Pressures over 200 MPa prevented post-acidification of the yoghurt during chilled storage. Pressurized yoghurts exhibited higher viscosity and amino acid contents than did the untreated controls, and the differences were maintained after chilled storage. High-pressure treatments at 300 and 400 MPa reduced the number of viable cells of lactobacilli to below the legal minimum permitted in many countries. Significant differences in sensory characteristics between untreated and pressurized yoghurts (200–300 MPa) were detected after chilled storage.  相似文献

The effect of deaeration, cysteine addition and electroreduction of milk on the viability of various Bifidobacterium strains in pasteurized milk during refrigerated storage at +7 °C for 4 weeks was assessed. Preliminary assays in deaerated milks showed considerable variability to oxygen sensitivity among the eight strains examined during refrigerated storage. Three strains with different sensitivity were selected for subsequent assays. Assays on the effect of oxygen during growth at 37 °C were included in the screening and results suggest that the growth inhibition observed could serve as an indicator of the negative effect of oxygen on stability of bifidobacteria during refrigerated storage in similar oxygen conditions. The electroreduction treatment had a positive impact on the survival of two of the three cultures tested during the 4 week storage period at 7 °C. Addition of cysteine and deaeration alone had similar effects on cell viability during storage, suggesting that the benefit of electroreduction was mainly linked to its action of lowering the level of dissolved oxygen in the samples. Data from this study suggest that electrochemical reduction of milk, as well as deaeration or addition of reducing agents could be applied to enhance the survival of bifidobacteria during extended storage at 7 °C in milk.  相似文献

Astaxanthin is a powerful antioxidant with various health benefits such as prevention of age-related macular degeneration and improvement of the immune system, liver and heart function. To improve the post-harvesting stability of astaxanthin used in food, feed and nutraceutical industries, the biomass of the high astaxanthin producing alga Haematococcus pluvialis was dried by spray- or freeze-drying and under vacuum or air at − 20 °C to 37 °C for 20 weeks. Freeze-drying led to 41% higher astaxanthin recovery compared to commonly-used spray-drying. Low storage temperature (− 20 °C, 4 °C) and vacuum-packing also showed higher astaxanthin stability with as little as 12.3 ± 3.1% degradation during 20 weeks of storage. Cost-benefit analysis showed that freeze-drying followed by vacuum-packed storage at − 20 °C can generate AUD$600 higher profit compared to spray-drying from 100 kg H. pluvialis powder. Therefore, freeze-drying can be suggested as a mild and more profitable method for ensuring longer shelf life of astaxanthin from H. pluvialis.  相似文献

The rheological behaviour of tarhana, a popular Turkish fermented wheat flour–yoghurt soup, was studied at different temperatures (30 to 70 °C). Results showed that an increase in shear rate resulted in a decrease in apparent viscosity, suggesting pseudoplastic behaviour at all temperatures studied. A power law equation was used to calculate the flow behaviour indices, n, ranging from 0.52 to 0.87. The n values were higher for the soups made from wholemeal flour than obtained for the soups made from white wheat flour. Consistency indices decreased with temperature and were described by the Arrhenius equation with activation energies from 48.6 to 28.6 kJ/mol.  相似文献
