AIM: To express recombinant S6K2 in baculovirus expression system; to purify large quantities of recombinant S6K2 for biochemical studies; to generate and characterise specific MABs against recombinant S6K2; to study the patterns S6K1 and S6K2 expression and subcellular localization in normal, benign and malignant breast tissues. METHODS: Recombinant baculovirus, expressing wild type S6K2 was generated using Bac-to-Bac system (Invitrogen); recombinant S6K was purified from infected Sf9 cells using affinity purification approach; monoclonal antibodies against recombinant S6K2 were generated; the specificity of generated MABs towards recombinant and endogenous S6K2 were examined by ELISA, Western blotting, immunoprecipitation and immuhohistochemical staining; immunohistochemical detection of S6K1 and S6K2 in human breast tissues was performed using specific monoclonal antibodies towards S6K1 and S6K2. RESULTS: Large amounts of enzymatically active S6K2 were purified using baculovirus expression system; highly purified preparations of S6K2 were used to generate and characterize anti-S6K2 MABs; elevated levels of S6K1 and S6K2 were found in breast tumors when compared to normal breast tissues; S6K2 is frequently localized in the nuclei of adenocarcinoma tissues, but rarely in fibroadenoma or "normal" breast tissues. CONCLUSION: Production of recombinant S6K2 in large amount and generation of specific monoclonal antibodies towards S6K2 has provided us with excellent tools to study the function and regulation of this important signalling molecule in normal and cancer cells. Immunnohistochemical analysis of S6K1 and S6K2 expression in normal and malignant breast clearly indicates that both kinases are overexpressed in breast tumors, when compared to "normal" tissues. The retention of S6K2 in the nuclei of malignant cells may be caused by disregulation of nucleocytoplasmic shuttling and could subsequently affect cell growth and proliferation.  相似文献

Herein, asymmetric isomer effects are systematically explored by designing and synthesizing two benzo[c][1,2,5]thiadiazole (BT)-fused nonacyclic electron acceptors. By changing from BP6T-4F to asymmetric ABP6T-4F, significantly enhanced dielectric constant and inhibited excessive molecular aggregation and unfavorable edge-on orientation could be achieved. The reduced exciton binding energy also facilitates a more efficient dissociation process in PM6:ABP6T-4F compared to PM6:BP6T-4F with the same energy offset. Moreover, the weaker crystallization behavior enables a significantly enhanced miscibility between PM6 and ABP6T-4F than that between PM6 and BP6T-4F, which leads to an optimized micromorphology with smooth surface, suitable domain size, and ordered π–π stacking. Organic solar cells (OSCs) based on PM6:ABP6T-4F achieve a 15.8% power conversion efficiency (PCE), which is remarkably higher than that of PM6:BP6T-4F-based OSCs (6.4%). Furthermore, ternary devices are also fabricated considering good compatibility between ABP6T-4F and CH1007 to deliver a PCE over 17%. This study reveals the effectiveness and great potential of asymmetric isomerization strategy in regulating molecular properties, which will provide guidance for the future design of non-fullerene acceptors.  相似文献

There has been a rapid growth in the need to support mobile nodes in IPv6-based networks. IETF has completed to standardize Mobile IPv6 (MIPv6) and Hierarchical Mobile IPv6 (HMIPv6) for supporting IPv6 mobility. Even though existing literatures have asserted that HMIPv6 generally improves MIPv6 in terms of handover speed, they do not carefully consider the details of the whole handover procedures. In this paper, based on the current IETF standards of both MIPv6 and HMIPv6, we conduct a comprehensive study of all IP-level handover procedures: movement detection, duplicate address detection, and location registration. Based on this study, we provide a mathematical analysis on MIPv6 and HMIPv6 performance in terms of handover speed. From the analysis, we reveal that the average HMIPv6 handover latency is not always lower than the average MIPv6 handover latency. Furthermore, even the intra-domain handover latency of HMIPv6 is not reduced much compared with MIPv6 handover latency. A finding of our analysis is that optimization techniques for movement detection and duplicate address detection are essential to shortening HMIPv6 handover latency and increasing the benefit of HMIPv6.
