ILLLI对银屑病患者外周血CD4+T细胞Fas蛋白表达的影响   总被引:2,自引:0,他引:2  
目的:探索ILLLI治疗对银屑病患者外周血CD4+T细胞Fas蛋白表达的影响。方法:取患者及正常对照组静脉血10ml,肝素抗凝,等份分别加入4支试管中,1支为未照射组,3支照射组分为30、60和120分钟,以输出功率5mW的氦氖激光照射,并进行荧光染色和流式细胞分析。结果:照射前银屑病组表达Fas蛋白的CD4+T细胞百分比为21.4±3.1%,较正常对照组的16.8±2.1%显著增加,差异有显著性,p<0.05。正常对照组在激光照射30分钟、60分钟和120分钟,表达Fas蛋白的CD4+T细胞百分比分别为18.0±5.3%、16.4±5.2%和19.8±6.7%,与照射前差异无显著性,p>0.05;而患者组在激光照射30分钟、60分钟和120分钟时,表达Fas蛋白的CD4+T细胞百分比分别为24.3±2.8%、28.3±5.3%和30.5±7.9%,可以看出,随着照射时间的延长,表达Fas蛋白的CD4+T细胞的数量不断增加,60分钟和120分钟,表达Fas蛋白的CD4+T细胞百分比较照射前增加显著,有统计学差异,p<0.05。结论:银屑病的发病机制可能与细胞凋亡失控等多种因素有关。激光治疗银屑病的机制可能与它下调IL—8水平、上调TNF—(表达、纠正患者的凋亡失控状态有关。  相似文献

目的:探讨慢性扁桃体炎患者经Nd:YAG激光治疗前后T淋巴细胞亚群的变化特点及临床意义.方法:采用流式细胞仪检测正常健康人及慢性扁桃体炎患者激光治疗前后外周血中T淋巴细胞亚群(CD3、CD4、CD8)水平.结果:慢性扁桃体炎患者外周血中CD 3、CD 4及CD 4/CD 8均显著低于正常对照组(P<0.01),CD 8细胞则明显高于正常对照组(P<0.05).经激光治疗后,CD 3、CD 4及CD 4/CD 8水平较治疗前明显升高(P<0.01),达到正常对照组水平(P>0.05),CD 8水平则较治疗前下降(P<0.01),接近正常对照组水平(P>0.05).结论:慢性扁桃体炎患者机体细胞免疫功能受到抑制,导致机体免疫力下降.而通过Nd:YAG激光治疗提高了机体的免疫力,而且对机体损伤小,同时减少了复发次数,避免了手术摘除.  相似文献

目的探索ILLLI治疗对银屑病员细胞免疫功能的影响.方法ILLLI照射时间1h,每天1次,10次为一疗程,观察治疗前后银屑病患者的外周血T淋巴细胞亚群及NK细胞的变化.结果(1)CD+3治疗后均值有明显增高,P值<0.05,治疗前后有显著差异.(2)CD+4均值治疗后略有下降,CD+4/CD+8、NK细胞均值治疗后略有上升,接近正常人均值.(3)CD+4、CD+8、NK细胞均值在30%以下治疗后均有增高;均值30%以上治疗后均有下降,前者P值≤0.05、后者P值<0.01,治疗前后有显著及非常显著差异.结论ILLLI治疗对银屑病病员外周血T细胞亚群的功能有双向调控作用.提示ILLLI治疗对银屑病病员的细胞免疫功能有双向调控作用.  相似文献

本文探讨了δ-氨基酮戊酸—光动力疗法 (ALA -PDT)对活动期红斑狼疮 (SLE)患者外周血中激活T淋巴细胞的影响。实验采用免疫荧光单标记—流式细胞仪检测 12例活动期SLE患者外周血中CD71阳性淋巴细胞百分比 ,观察其在ALA -PDT前后的变化。发现ALA -PDT能显著降低CD71阳性淋巴细胞百分比 ,ALA -PDT前CD71阳性细胞百分比为 12 31±1 40 ,ALA -PDT后继续培养 18小时下降至 6 5 9± 0 95 ,有非常显著的差异 (P <0 0 1)。本试验证实ALA -PDT能降低活动期SLE外周血中激活T淋巴细胞百分比 ,即可能抑制活动期SLE外周血中激活的T淋巴细胞的活性  相似文献

目的:观察低氧、复氧对心肌细胞内Ca2+浓度([Ca2+]i)的影响,以及牛磺酸模拟心肌缺血/再灌注(I/R)过程中减轻钙超载的作用。方法:采用SD大鼠乳鼠进行心肌细胞培养,建立模拟I/R模型。实验分4组:①正常对照组;②模拟缺血/缺氧组:细胞低氧180min;③模拟I/R组:细胞低氧180min,复氧180min;③牛磺酸+I/R组:先加入终浓度为20mmol.L-1的牛磺酸,再行低氧180min,复氧180min。以Fluo-4/AM荧光指示剂负载,应用激光共聚焦显微镜技术检测心肌细胞[Ca2+]i变化。结果:对照组心肌细胞[Ca2+]i荧光强度(23.71±2.37)U和荧光光密度较低;低氧180min后复氧即刻,[Ca2+]i荧光强度开始增加(57.52±8.31),复氧180min后[Ca2+]i荧光强度(71.13±4.74)U和光密度显著增高(P<0.01vs对照组)。而牛磺酸组细胞内荧光强度和光密度较模拟I/R组显著降低[(42.42±4.17)U vs(71.13±4.74)U,P<0.01]。结论:心肌细胞缺血/缺氧导致Ca2+超载;模拟I/R Ca2+超载加剧,而牛磺酸有明显减轻心肌细胞模拟I/R时Ca2+超载的作用。  相似文献

目的:对神经病理性疼痛患者进行抑郁评估,并检测外周血免疫球蛋白、补体及循环免疫复合物水平,外周血T细胞的表面标志物如CD3+、CD4+、CD8+等,比较伴抑郁状态的神经病理性疼痛患者体液免疫和细胞免疫的变化,观察抑郁状态对神经病理性疼痛患者的免疫是否有影响。方法:选择我院2006至2007年符合神经病理性疼痛纳入标准的疼痛门诊及住院患者,通过Zung’S抑郁自评量表了解病人抑郁情况;采集外周静脉血测定血常规、IgA、IgG、IgM、IgE、C3、C4、循环免疫复合物(CIC)、CD3+、CD4+、CD8+T细胞。结果:入选的86位神经病理性疼痛患者中一半以上(52名占60.47%)的患者合并有不同程度的抑郁状态,其中轻度抑郁的患者29名占总抑郁患者的55.77%。白细胞总数及淋巴细胞百分比和绝对值在有无抑郁情况的神经病理性疼痛患者中有显著差异(P〈0.05),抑郁的患者较无抑郁的患者显著降低,但均在正常范围内。其他白细胞种类、IgG、IgA、IgM、IgE、C3、C4、CIC、CD3+、CD4+和CD8+等,在有无抑郁情况的神经病理性疼痛患者中没有显著差异(P〉0.05)。结论:神经病理性疼痛患者容易合并以轻度为主的不同程度的抑郁状态;神经病理性疼痛患者合并抑郁状态对免疫功能有轻微影响。  相似文献

目的:观察光动力治疗(PDT)肿瘤留置对残存肿瘤局部淋巴细胞表型的影响。方法:采用双侧小鼠荷H22移植型肝癌模型。将30只双侧荷瘤小鼠分为3组:PDT组,切除组,对照组。光动力治疗一侧肿瘤后,定量观察残存肿瘤和脾脏内CD4+和CD8+细胞的变化。使用免疫组织化学及图像分析技术。结果:在原位肿瘤内,有极少量的CD4+细胞。在残存肿瘤内,未见CD4+细胞,CD8+细胞呈明显的带状分布,在光动力治疗组,CD8+细胞的密度明显增高,但仍不足以杀死残存肿瘤,其活性也有待进一步研究。  相似文献

Monoclonal antibody (MAb) 1B4 of IgG3 isotype reacting with 55 +/- 10% peripheral blood lymphocytes was obtained. MAb 1B4 is bound with 100% of B and NK cells and 50-70% of T cells in blood. The most part of CD8+ cells (60-100%) and approximately 50% of CD4+ cells belong to 1B4+ population. T-ALL cells do not express 1B4 antigen. Analysis of reactivity of MAb 1B4 with transformed malignant cells has shown that 1B4 antigen appeared in the process of differentiation of B lymphocytes at the state of pre-B cells and disappeared with activation of B lymphocytes and transition to plasma cells.  相似文献

目的:探讨三氧化二砷(As2O3)联合小鼠肝癌(H22)源exosome对小鼠皮下种植性肝癌(H22)模型的抗瘤作用。方法:超速离心及密度梯度离心法提取exosome,Bradford法蛋白定量;建立BALB/c小鼠皮下荷瘤模型.实验分4组:空白成瘤对照组、As2O3组、As2O3联合exoeome组和exosome组。观察As2O3、exosome、及其联合应用的体内抗瘤效应;监测在体肿瘤的生长情况;计算抑瘤率、生存率:免疫组织化学法检测肿瘤组织中浸润的CD4^+、CD8^+T淋巴细胞。结果:与空白成瘤对照组比较,As2O3组、及其与exosome联合组的肿瘤生长明显放缓(P〈0.05)。其抑瘤率分别为44.58%、66.27%。As2O3组及As2O3联合exosome组肿瘤组织中浸润的CD4^+、CD8^+T淋巴细胞明显增加(P〈0.05)。结论:As2O3与exosome联合应用对小鼠皮下种植性实体性肝癌(H22)有协同治疗效应。  相似文献

Monoclonal antibodies (McAb) LT1, LT2, LT4, LT6, LT7, LT8, LB21 and LDR to surface antigens of human lymphocytes have been obtained. Basing on the data concerning the distribution of McAb reactivity with cells from different organs and tissues, and with tumour cell lines, on experiments on the mutual blockage and comodulation of antigens, on two-colour immunofluorescence and determination of the molecular weight of the detected antigens a conclusion is made that the McAb obtained react with CD5, CD2, CD4, CD6, CD7, CD8, CD21 and HLA-DR antigens, respectively. McAb LT1 and Leu1, as well as LT2 and Leu5b in pairs blocked each other completely. The determinant recognized by McAb LT8 is more completely overlapped with the epitope, reacting with McAb OKT8. The McAb LT4 binding site is only partially overlapped with the McAb Leu3 site of recognition and practically is not bound to the epitope recognized by McAb OKT4.  相似文献

Monoclonal antibodies ICO and OKT, OKM5, C-ALLA have been used to investigate surface lymphocyte antigens in normal (9), reactive (27) lymph nodes and in lymphogranulomatosis (9) in children. The study has shown that the number of lymphocytes with T and B markers is 45% in normal lymph nodes; OKT4/OKT8 is 0.9. In reactive lymph nodes all lymphocytes have T and B markers; OKT4/OKT8 is, 1.5. T-cell (OKT3+, OKT11+) or B-cell (sIg+, ICO-1+) type differentiation is preferable. B-cell type proliferation with a high number of ICO-1+ lymphocytes appeared in lymph nodes of 4 patients with lymphogranulomatosis. Polyclonal proliferation and differentiation were observed in other 5 children. No correlation between immunology and histologic types of the disease was revealed.  相似文献

Ultrastructure of NKH1+ and CD8+ subpopulations of large granular lymphocytes (LGL) obtained by the flow cytometry method using corresponding monoclonal antibodies was studied in patients with Hodgkin's disease. Electron microscopy has identified different changes in ultrastructure of LGL, especially of osmiophilic granules. The results indicate significant changes in the functional activity and defect of LGL in blood at early stages of the disease.  相似文献

Vaccine adjuvants that can induce robust protective immunity are highly sought after for the development of safer and more effective vaccines. Vaccine formulation parameters that govern efficacy are still far from clear, such as the diverse impacts of codelivering agonist molecules for innate cell receptors (e.g., pattern recognition receptors). In this study, a mesoporous silica‐templating approach is used to fabricate protein antigen (ovalbumin) particles covalently functionalized with agonists for NOD‐like receptor 2 (NOD2) and Toll‐like receptor 9 (TLR9). Particle‐induced combinatorial NOD2/TLR9 signaling results in synergistic inflammatory cytokine secretion by mouse macrophages (RAW 264.7). Administration of NOD2/TLR9 particles in mice results in adaptive immune responses that are both quantitatively and qualitatively different than those resulting from administration of particles conjugated with either NOD2 or TLR9 agonists alone. While delivery of NOD2 agonists alone activates T helper 2 (Th2)‐type responses (and no CD8+ T cell activation) and delivery of TLR9 agonists alone activates CD8+ T cell and T helper 1 (Th1)‐type responses, codelivery of NOD2 and TLR9 agonists enhances Th1‐type responses and abrogates CD8+ T cell activation. The results illustrate that in the particle‐based system, NOD2 activation plays different roles in polarizing adaptive immune responses depending on coactivation of TLR9.  相似文献

Comparative investigation of subpopulations and functional properties of lymphocytes isolated from the tumour tissue (TTL) as well as of lymphocytes of the peripheral blood (PBL) of 11 lung cancer patients and 18 breast cancer patients has shown that an increase in the quantity of theophylline-sensitive cells (suppressors and killers) is characteristic of TTL. After 20 hours of incubation of the mixture of the studied lymphocytes and sheep erythrocytes at 4 degrees C an increase in the number of E-RFC proved to be more intensive for TTL than for PBL. The result obtained may suggest the blocking of the superficial receptors of TTL to sheep erythrocytes.  相似文献
