The efficacy of five daily treatments with 7.5 mg fenbendazole/kg bodyweight against mucosal cyathostome larvae was evaluated in 20 12- and 24-month-old ponies with naturally acquired cyathostome infections. After three weeks communal grazing on infected pasture and six weeks indoors, one group of 10 ponies were treated. Six weeks later, both groups of ponies were humanely destroyed and their burdens of large intestinal cyathostome worms, including luminal parasites and mucosal larvae, were assessed. In the control animals approximately 7 per cent of the total worm burden was present in the gut lumen and 93 per cent was present as larvae in the large intestinal mucosa. The efficacy of fenbendazole against the luminal cyathostomes was 90.7 per cent (P < 0.01). The total numbers of mucosal larvae, recovered after digestion, were reduced by 95.3 per cent (P < 0.0005); mucosal late third and fourth stage larvae were reduced by 99.4 per cent (P < 0.0001), and early inhibited third stage larvae by 91.5 per cent (P < 0.005).  相似文献

Three fecal egg count reduction assays (FECR) and one critical trial were performed to determine the efficacy of pyrantel pamoate (PP) at 6.6 mg base kg-1 on a well managed stud farm in Louisiana where a loss of efficacy was suspected. Efficacy of PP based on FECR varied from 25% in mares to 83% in yearlings. Second treatments with PP 2 weeks following an initial treatment failed to reduce eggs per gram (EPG). A critical trial was performed to determine the cyathostome species resistant to PP. Three strongyle-naive ponies which acquired infections on the farm were used for this purpose. Following treatment with PP at the recommended dose, 11 species of cyathostomes remained in the intestine of the tracer ponies. Reduced efficacies (62%-88%) were noted for seven species. Resistance to oxibendazole (OBZ), which was > 90% effective on this farm in 1982, was also evaluated by FECR and found to exist. The results of one experiment indicate that dual resistance of parasites to PP and OBZ also exists.  相似文献

Several compounds (n = 13 single or combinations; most at therapeutic dosages) were evaluated between 1977 and 1992 in critical tests (n = 91) against benzimidazole (BZ) resistant small strongyles (Population S) and several other species of internal parasites in Shetland ponies, mostly under 1 year old. The closed breeding herd, from which the test ponies were selected, had been treated every 8 weeks with cambendazole (CBZ) for 4 years (1974-1978) and oxibendazole (OBZ) for 14 years (1978-1992). Published field test data (1974-1992) on older ponies in the herd showed BZ resistance of small strongyles. Average efficacies in the present critical tests against small strongyles for OBZ (n = 59 animals) were high in early years (95% or higher), but gradually declined to a low of 1% in 1991. Side-resistance of small strongyles was evident in critical tests (n = 1-6/single drug or combination) for several other BZs and a pro-BZ; ivermectin and piperazine were highly active, but pyrantel pamoate exhibited weak activity. BZ resistance was evident for six small strongyle species (Cyathostomum catinatum, Cyathostomum coronatum, Cylicocylus nassatus, Cylicostephanus calicatus, Cylicostephanus goldi, and Cylicostephanus longibursatus). Activity on bots, ascarids, large strongyles, and pinworms was essentially as expected, indicating no drug resistance.  相似文献

The long-term efficacy of an irradiation attenuated larval (L3) vaccine against Strongylus vulgaris was tested in ponies which were reared on pasture. Prior to foaling, mares were divided into two groups. One group of mares and foals received regular (eight weekly) treatment with ivermectin and the second group remained untreated. Half the foals in each pasture group were vaccinated at eight to ten weeks of age. Foals were weaned at three to four months of age and maintained on separate pastures. At eight to ten months of age, ponies were placed in box stalls and half of each treatment group were challenged with S. vulgaris (5 x 1000 L3). Clinical signs and lesions typical of acute verminous arteritis were found at necropsy in the ivermectin treated non-vaccinated challenged yearlings. Ivermectin treated vaccinated challenged yearlings did not show these clinical signs, had markedly reduced to absent arterial lesions and showed an 89 per cent reduction in arterial larval burdens post mortem. Significant differences in clinical signs, arterial lesions or arterial larval burdens were not seen between vaccinated and non-vaccinated foals reared without benefit of ivermectin treatment.  相似文献

A mathematical model was developed to describe the dynamics of the parasitic stages of Oesophagostomum dentatum in pigs. An immigration-death model with constant establishment, development and death rates was fitted to L3, L4 and adult worm burdens observed in a single-infection experiment. Female worm length was modelled by a function of worm age and total worm burden, while worm egg production (eggs per gram faeces per female worm) was modelled by a function of worm age and worm length. The model was then used to predict worm burdens observed in a trickle-infection experiment. The predicted worm burdens were much higher than those observed, suggesting that worm death rates were higher during the trickle infection. After increasing worm death rates to fit the observed worm burdens, female worm lengths and egg production in the trickle infection were predicted. At the medium- and high-dose rates, predicted worm lengths and, thus, egg production were lower than observed, while at the low-dose rate predicted egg production was too high. It appeared that in the trickle infections, total worm burden had less influence on observed female worm length and egg production than in the single infections. The results suggest that the demography of O. dentatum in pigs differs between single and trickle infections.  相似文献

Trials were conducted in ponies to evaluate the efficacy of pyrantel pamoate (Strongid-T(R)) and two newer anthelmintics not yet commercially available, nitramisole and avermectin B(1)a, against migrating Strongylus vulgaris larvae. Ponies were removed from their mares within 24-48 hr after birth and reared in isolation, worm free. Between six and 14 weeks of age they were infected with 2000 or 2500 infective S. vulgaris larvae. Subsequently, they were monitored daily for clinical signs until the experiment terminated at 28 days postinfection. All ponies showed increased body temperature and reduced appetite within the first week of infection. All anthelmintics were administered on day 7 and in addition pyrantel pamoate was given on day 8 postinfection. The anthelmintics were in liquid formulation. Nitramisole and pyrantel pamoate were given by stomach tube and avermectin B(1)a by subcutaneous injection.Following administration of these compounds toxic reactions were not observed. All anthelmintics caused a reduction in body temperature and increased appetite and effected a clinical cure. In ponies which were not treated with an anthelmintic, temperatures remained elevated and appetites never returned completely to normal. These ponies also showed variable degrees of lethargy, depression, recumbency and colic and the majority died between two and three weeks postinfection. At necropsy, these control ponies showed variable degrees of adhesions involving the abdominal organs, necrosis of the ileum and cecum and severe arteritis and thrombosis of the major abdominal arteries and their branches.Although pyrantel pamoate, used at eight times the therapeutic dose for intestinal nematodes in the horse, effected a clinical cure it did not produce a radical cure. At necropsy, ponies treated with pyrantel pamoate had arteritis and thrombosis of the cranial mesenteric artery and its major branches. Nitramisole and avermectin B(1)a were able to effect both a clinical and radical cure.  相似文献

L3 and L4 stages of H.polygyrus were prevented from developing further and were probably killed within 24 h of treatment with ivermectin although total parasite burdens, particularly when treatment was given 4-6 days after infection, declined over a longer period lasting several days. Strong resistance to challenge infection was expressed by infected mice dosed with ivermectin during the tissue phase of larval development. Even immunizing infections as brief as 12-36 h (when only L3 larvae would have been present in the mucosa) elicited strong acquired immunity. When infections were terminated 4-6 days after infection, acquired resistance was 95-100%. The stronger resistance of mice exposed to both L3 and L4 stages was associated with the recognition of low molecular weight polypeptides in adult worm homogenate and there was a highly significant correlation between percentage protection and anti-L4/anti-adult worm serum IgG1 antibodies.  相似文献

One hundred and four post mortem examinations of randomly selected sheep (52) and goats (52) were carried out weekly from March 1990 to February 1991. Gastrointestinal parasites were identified and counted. A large spectrum of 16 helminth species was found in 101 (97%) infected animals. The nematodes infecting small ruminants were, in order of predominance: Trichostrongylus colubriformis (96% of all animals), Oesophagostomum columbianum (82%), Haemonchus contortus (67%), Strongyloides papillosus (55%), Gaigeria pachyscelis (38%), Cooperia spp. (49%) and Trichuris ovis (12%). Forty-five (43%) necropsies revealed an infection with Moniezia benedeni, Avitellina centripunctata or Stilesia globulosa. Infections with Schistosoma bovis and Paramphistomum spp. were rarely seen. Goats carried significantly lower worm burdens than sheep and were less often infected by tapeworms. Nematode worm burdens followed a similar seasonal pattern in both host species. While Trichostrongylus colubriformis was also recovered in high numbers in the mid dry season, other species such as H. contortus, Strongyloides papillosus and Oesophagostomum columbianum reached distinct peaks in the rainy season (July-October). Different survival strategies for the prolonged dry season were observed. Haemonchus contortus outlived the unfavourable climatic conditions as inhibited larvae in the abomasal mucosa, while the other nematode species survived as adults with a reduced fecundity. Egg production per adult worm specimen was found to depend heavily on the season and egg counts in the dry season (November-June) did not correspond to the size of worm burdens. Unexpectedly high worm burdens were recovered from adult and old animals. Investigations of young animals indicated that no reinfection can occur during the dry season. The findings are discussed with regard to their relevance for strategic control of gastrointestinal nematodes in small ruminants.  相似文献

Studied the effects of 2 types of play experiences—constructive and dramatic—on preschool children's perspective-taking (PT) performance. Pretests and posttests for 3 types of PT (perceptual, cognitive, and affective) were administered to 51 preschoolers, (mean age 4 yrs 10 mo) in 3 groups. Group 1 received 10 constructive play sessions, Group 2 received 10 dramatic play sessions, and Group 3 received no play treatment. Both play treatments produced substantial and roughly equivalent improvements in PT. These effects were general across the 3 types of PT, and they were not constrained by the Ss' pretraining performance on the PT tests. In view of recent evidence that some PT measures may not be reliable, a follow-up reliability study was conducted with 48 preschool children, mean age 4 yrs 8 mo. Twenty-three well-conditioned ponies were evaluated for insulin and glucose response following oral glucose administration (1 g/kg bodyweight [bwt] as a 20 per cent solution). Ponies were defined as normal if total insulin secretion (TIS) was less than 149 mu iu/ml h and the glucose concentration was below 11.1 +/- 0.11 mmol/litre (200 +/- 2 mg/dl) at all times following oral glucose administration. When glucose concentrations were maintained below 11.1 +/- 0.11 mmol/litre, the area under the glucose curve (TG) was less than 17.4 mmol/litre/h (314 mg/dl/h). The ponies were assigned to four groups based on insulin and glucose response: Group 1 (n = 7), normal; Group 2 (n = 5), high insulin, normal glucose; Group 3 (n = 8), high insulin, high glucose and Group 4 (n = 3), high glucose, normal insulin. This classification is an initial attempt to define normal insulin and glucose response in ponies. Additional data need to be accumulated to define further insulin resistance and diabetes in ponies.  相似文献

Studies on effects of subclinical parasitism (low worm burden) in full-fed and maintenance-fed steers indicated that over a short interval low worm burdens do not have a statistically significant effect on the efficiency of energy utilization. In full-fed steers, energy retention tended to be higher in steers with lower worm burdens (mean 95.8 Ostertagia, 12.5 Cooperia) than in those with higher worm burdens (mean 677.5 Ostertagia, 201.8 Cooperia). Maintenance-fed steers generally had larger numbers of Ostertagia ostertagi and Cooperia spp than did the full-fed steers.  相似文献

For a study on the occurrence of resistance to reinfection with porcine nodular worm species, pigs were infected twice weekly with 1,000 infective larvae (L3) of Oesophagostomum quadrispinulatum for 8 weeks. All pigs, including noninfected controls, were then treated with fenbendazole. At 10 days after treatment, all pigs received a single challenge inoculation of 5,000 L3 of either O. dentatum or O. quadrispinulatum, respectively. Pigs were slaughtered at 6 weeks after the challenge infection for determination of their worm burdens. The pigs trickle- and challenge-infected with O. quadrispinulatum had significantly lower egg excretion levels (P < 0.01) and worm burdens (P < 0.05) than challenge control pigs, thus indicating some degree of host immunity against the homologous challenge infection. No resistance to reinfection was evident for the heterologous challenge infection. This study elucidates further aspects of the interaction between nodular worm species in the pig.  相似文献

The objective of this study was to determine the effect of treatment with additional progesterone (P4) or 17 beta-oestradiol (E2) at the end of a period of P4 treatment on ovarian follicular development, ovulation time, and plasma gonadotrophin and steroid hormone concentrations of Bos indicus cows. Initially, two injections of PGF2 alpha were given 14 days apart, and at the time of the second injection (Day 0) all cows received a single P4-releasing controlled internal drug release (CIDR) device that was removed 10 days later. Control cows (Group 1, n = 8) received no other treatment. On Day 8, cows in Group 2 (n = 8) and Group 3 (n = 8) received either a s.c. implant containing E2, or a second CIDR device, respectively. All CIDR devices and E2 implants were removed at a similar time on Day 10. Treatment with E2 or P4 delayed mean (+/- SD) time of ovulation (113.1 +/- 25.6 h, 153.4 +/- 44.5 h and 150.8 +/- 25.1 h for Groups 1, 2 and 3, respectively; P < 0.05) and the mean time (+/- SD) of the luteinising hormone (LH) peak (87.4 +/- 24.5 h, 124.3 +/- 45.0 h and 122.3 +/- 25.04 h for Groups 1, 2 and 3, respectively; P < 0.05). Both treatments delayed the mean (+/- SD) day of emergence of the ovulatory follicle (7.7 +/- 3.6 days, 11.3 +/- 1.7 days and 11.1 +/- 1.5 days for Groups 1, 2 and 3, respectively; P < 0.05), and reduced the variability in the day of emergence of the ovulatory follicle (P < 0.05) compared with the control cows. Variability in age and duration of dominance of the ovulatory follicle was greater in control animals compared with treated animals (P < 0.05). Treatment with E2 on Days 9 and 10 did not alter mean concentrations of gonadotrophins in the cows in Group 2 compared with control cows (P > 0.05), whereas treatment of cows with an additional CIDR device resulted in greater mean concentrations of FSH and lesser concentrations of LH on Day 9 (P < 0.05) compared with cows in Groups 1 and 2. By Day 10 mean concentrations of gonadotrophins were similar among cows in all three groups. Concentrations of E2 were less in cows in Group 3 compared with cows in Groups 1 and 2 from Day 9 to Day 11 (P < 0.05). We conclude that treatment with either E2 or P4 can influence the pattern of ovarian follicular development and ovulation in cattle; however, the mechanism of action of the two treatments may differ. Atretogenic treatments for ovarian follicles applied at the end of a period of progesterone treatment did not improve synchrony of ovulation.  相似文献

An attempt was made to immunise lambs by the intra-muscular injection of concentrated metabolites obtained from the in vitro culture of Ostertagia circumcincta, by the oral administration of lyophilised metabolites or by the subcutaneous injection of living third, fourth and fifth stage worms grown in vitro. Protection appeared to have been elicited by each treatment as the worm burdens, the lengths of the worms and the faecal worm egg counts in the immunised lambs were less than those of the control lambs.  相似文献

In 2 experiments, 265 Swiss albino Rb-3 9-wk-old untreated mice, when placed in a novel environment composed of a central cage surrounded by 4 dark boxes in which different sounds were emitted, explored the boxes for 1 or 2 hrs. In most cases, Ss settled into 1 of the 4 boxes and built their nest. Most of the controls chose the silent compartment, whatever the sound delivered in the other boxes. An acoustic choice test was then carried out. Four groups of mice were exposed to a particular sound for 10 days: Group 1: Gestation Day 10 to birth; Group 2: birth to Day 10; Group 3: Day 10 to Day 20; Group 4: Day 20 to Day 30. Ss were then tested for box preference on Day 63 in the 4-box choice, one of which contained the sound of the earlier exposure. Ss stimulated before the 10th day after birth behaved in the same way as controls, whereas Ss stimulated between the 10th and the 20th days after birth exhibited a strong preference for the box delivering the sound of the previous treatment. This choice was significant, whatever the harmonic (Exp I) or impulsive (Exp II) nature of the sound. Ss stimulated between the 20th and the 30th days showed a preference for the silent box, but among the others, 75% chose the sound received during infancy. This study examined the role of CD8+ and WC1+ T-cells and of interferon (IFN)-gamma in the development of protective immunity against infection with the enteric nematode parasite Trichostrongylus colubriformis in sheep. Monoclonal antibodies (mAb) were administered during induction of the immune response to deplete or neutralise these components. Protection against the primary and challenge infections was assessed by faecal egg count and total worm count. Prolonged administration of mAb recognising IFN-gamma and CD8 resulted in significantly increased protection during the 6 week primary infection and following challenge. CD8+ cells were depleted from blood but not from intestinal mucosa. After injection of mAb (CC15) recognising the surface antigen WC1, WC1+ and Tcr gamma delta + cells were depleted from blood but not markedly from enteric mucosa, and protection against challenge, although variable, was increased by up to 88%. It appears that CD8+ and WC1+/gamma delta+ cells and IFN-gamma all retard the potential development of naturally acquired immunity against the parasite.  相似文献

Horse conceptuses collected between Day 11 and Day 18 of pregnancy float in isotonic media. To investigate this phenomenon, blastocyst fluids from 30 conceptuses from 13 mares were analysed for osmolality and for concentrations of Na+, Cl-, K+, glucose, urea and creatinine. In conceptuses from Group A, samples from Day 11 to Day 16 yielded the following results (mean +/- s.e.m.): osmolality, 121.4 +/- 1.5 mOsm kg-1; Na+, 11.0 +/- 2.2 mM; Cl-, 29.3 +/- 2.5 mM; K+, 26.2 +/- 2.6 mM; glucose, 0.6 +/- 0.1 mM; urea, 6.0 +/- 0.6 mM; creatinine, 9.6 +/- 1.1 microM. Between Day 16 and Day 25, the osmolalities and Na+ concentrations increased gradually with age but the former never exceeded 255 mOsm kg-1. Fluids from Group B were obtained from eight conceptuses exposed to saline solutions of different osmolalities for various periods of time. An increased perivitelline space of 1-3 mm became evident at the lower pole of the floating conceptus after 45 min of exposure to solutions with osmolalities of > or = 300 mOsm kg-1, suggesting that Na+ and Cl- diffuse freely through the capsule but not through the trophoblast. The significance of the hypo-osmolality of equine blastocyst fluid is discussed but remains unclear.  相似文献

The efficacy of moxidectin was evaluated in young ponies naturally infected with gastrointestinal parasites. Eight animals were treated orally with moxidectin at 0.4 mg/kg bodyweight and eight received only the vehicle. They were all necropsied two weeks later. Faecal samples were examined daily for egg counts and larval cultures. Parasites were recovered from total faecal samples collected daily and from the gastrointestinal tracts at necropsy. Moxidectin reduced the strongyle egg counts by > 99 per cent from three days after treatment but some individuals remained positive for 10 days. The drug had little or no ovicidal activity. As evaluated in the critical-controlled test, moxidectin was 99 to 100 per cent effective (P < 0.05) against luminal stages of parascaris equorum, Strongylus vulgaris, Triodontophorus species, Craterostomum acuticaudatum, 19 cyathostome species and Oxyuris equi. Adult S edentatus were also completely removed. Its efficacy against third stage larvae of Gasterophilus intestinalis was 95 per cent (P < 0.05). Luminal nematode stages were removed within a few days, and bots continued to be eliminated for at least two weeks after treatment. No activity was observed against Anoplocephala perforliata. As evaluated in the controlled test, moxidectin was 100 per cent effective against Habronema muscae (P < 0.05) and had a 76 per cent but not significant efficacy against encysted small strongyle larvae.  相似文献

Determined the factors that enhanced milk intake during deprivation in albino rats 15, 20, and 25 days of age. Ss were subjected, for 8 hrs, to 1 of 6 regimens. Results show that intake at Day 15 was reduced by the opportunity to suckle, independent of receiving a milk load. This same trend was apparent, although not as strong, among Day 20 Ss. By Day 25, nonnutritive suckling during the privation period no longer attenuated milk intake, although preloads did, whether or not they were paired with nonnutritive suckling. Between 1988 and 1993, 82 patients with a median age (iq range) of 81 (73-87) years underwent thrombo-embolectomy (TE) and thrombolysis for acute lower limb ischaemia at the Royal Hallamshire Hospital, Sheffield. 28 patients (Group 1) underwent TE prior to the introduction of thrombolysis in 1991. TE with intraoperative thrombolysis (IT) as an adjunct was performed in 34 patients (Group 2) and 20 patients (Group 3) were treated using percutaneous thrombolysis (PT) in the first instance. 41 of these patients were in AF at presentation. Although 24 were on digoxin, only 4 were on warfarin. Group 3 patients were younger (p < 0.05; ANOVA) with a longer duration of ischaemia (p < 0.05; ANOVA) and had less limbs suffering a sensorimotor deficit (p = 0.007; chi 2) compared with Groups 1 and 2 which were similar in these respects. Overall mortality was 17%. Successful revascularisation was achieved in 18 (64%), 28 (82%) and 15 (75%) patients in Groups 1, 2 and 3 respectively. 14 of the 15 patients successfully treated initially by PT required further intervention in order to maintain revascularisation. Revascularisation failure was associated with the presence of a sensorimotor deficit but not associated with patient age or duration of ischaemia.  相似文献
