Cyanobacteria are prolific producers of bioactive natural products that mostly belong to the nonribosomal peptide and polyketide classes. We show here how a linear precursor peptide of microviridin K, a new member of the microviridin class of peptidase inhibitors, is processed to become the mature tricyclic peptidase inhibitor. The microviridin (mvd) biosynthetic gene cluster of P. agardhii comprises six genes encoding microviridin K, an apparently unexpressed second microviridin, two RimK homologues, an acetyltransferase, and an ABC transporter. We have over‐expressed three enzymes of this pathway and have demonstrated their biochemical function in vitro through chemical degradation and mass spectrometry. We show that a prepeptide undergoes post‐translational modification through cross‐linking by ester and amide bond formation by the RimK homologues MvdD and MvdC, respectively. In silico analysis of the mvd gene cluster suggests the potential for widespread occurrence of microviridin‐like compounds in a broad range of bacteria.  相似文献

The citrus leafminer is an important vector of citrus canker in many of the major citrus production areas of the world. (7Z,11Z)-Hexadecadienal was reported as a sex attractant for this insect in the 1980s, based on trap catches during pheromone screening trials in Japan. However, attempts to reproduce this work in other areas of the world have not been successful. We report here that (7Z,11Z)-hexadecadienal is only one component of the pheromone, with the other critical component being the analogous trienal, (7Z,11Z,13E)-hexadecatrienal. Both compounds were identified in the effluvia from live female moths by coupled gas chromatography (GC)-electroantennography using nonpolar and polar GC columns, and the identifications were confirmed by comparisons of mass spectra with those of authentic standards. Stereoisomers of the two compounds, and a number of analogs, were synthesized to confirm the identifications. In field trials, neither compound alone was attractive to male moths, but blends of the two were highly attractive, with thousands of insects being caught per trial. Addition of the isomeric (7Z,11Z,13Z)-hexadecatrienal inhibited attraction to the two-component blend. Biotic and abiotic environmental factors affect plant nutritional quality and defensive compounds that confer plant resistance to herbivory. Influence of leaf age, light availability, and girdling on foliar nutrition and defense of green ash (Fraxinus pennsylvanica Marsh) was examined in this study. Longevity of the emerald ash borer, Agrilus planipennis Fairmaire (Coleoptera: Buprestidae), adults reared on green ash foliage subjected to these factors was assayed. Mature leaves generally were more nutritious with greater amino acids and a greater ratio of protein to non-structural carbohydrate (P:C) than young leaves, in particular when trees were grown in shade. On the other hand, mature leaves had lower amounts of trypsin and chymotrypsin inhibitors, and total phenolics compared to young leaves. Lower defense of mature leaves alone, or along with higher nutritional quality may lead to increased survival and longevity of emerald ash borer feeding on mature leaves. Sunlight reduced amino acids and P:C ratio, irrespective of leaf age and girdling, and elevated total protein of young foliage, but not protein of mature leaves. Sunlight also dramatically increased all investigated defensive compounds of young, but not mature leaves. Girdling reduced green ash foliar nutrition, especially, of young leaves grown in shade and of mature leaves grown in sun. However emerald ash borer performance did not differ when fed leaves from trees grown in sun or shade, or from girdled or control trees. One explanation is that emerald ash borer reared on lower nutritional quality food may compensate for nutrient deficiency by increasing its consumption rate. The strong interactions among leaf age, light intensity, and girdling on nutrition and defense highlight the need for caution when interpreting data without considering possible interactions.  相似文献

We examined genetic variation in inducibility and in constitutive and herbivore-induced levels of glucosinolates, trypsin inhibitors, and resistance to herbivory in families of Brassica rapa originating from a wild population. We also examined phenotypic and genetic correlations among absolute levels of these traits in control and induced plants. We grew seedlings of 10 half-sib families in pairs in pots, and exposed one plant per pair to folivory by Trichoplusia ni larvae. Two days later, we sampled all plants for total glucosinolate and trypsin inhibitor levels and examined the preference and consumption by T. ni larvae of previously damaged (induced) and undamaged (control) plants. There was no significant variation among sire families in the induction of glucosinolates or trypsin inhibitors by T. ni feeding. Total glucosinolate levels in either control or induced plants did not vary by family. In contrast, trypsin inhibitor levels in both control and induced plants varied significantly by family. Trichoplusia ni fed less on induced plants than on control plants in the bioassay, but neither the induction of resistance by prior T. ni feeding nor absolute levels of damage done to control and induced plants varied significantly by sire family. Temporal blocking strongly affected trypsin inhibitor levels and the response of some families in the bioassays. There were no significant phenotypic or genetic correlations of levels of glucosinolates or trypsin inhibitors with each other or with damage in either control or induced plants. Overall, these results suggest that in the B. rapa population that we studied, both total glucosinolate content and biological resistance to herbivory by T. ni was nonvariable and almost universally inducible by prior T. ni feeding. In contrast, control and induced levels of trypsin inhibitors varied genetically and have the capacity to respond to future selection imposed by herbivores. However, the role of these defenses in constitutive or induced resistance to T. ni in this species remains unclear.  相似文献

Five species of green algae isolated from the Cleveland County, Oklahoma, area and a commercially obtained yellow-green alga were tested for allelopathic interactions. Axenic cultures ofPandorina morum Bory were stimulated in three sterile filtrates from nonaxenic cultures ofScenedesmus incrassatulus var.mononae G.M. Smith, with the oldest filtrate showing the least stimulation. By measuring the growth of axenic cultures inoculated into sterile filtrates of old cultures, instances of inhibition and stimulation were recorded in three screening experiments.Cosmarium vexatum West filtrate was inhibitory to the five other species, a characteristic probably important in its role of producing waterblooms in ponds and swamps. The allelopathic effects of filtrates of the five other species onBolrydium becherianum Vischer could be a key to its restriction to terrestrial sites. Instances of heat lability of the active substances were noted. These interactions, both allelopathic and stimulatory, probably play a significant role in the succession of algal blooms in nature.  相似文献

It was previously reported that females of the currant stem girdler, Janus integer Norton (Hymenoptera: Cephidae), produce a compound, (Z)-9-octadecen-4-olide (1), that is sensitively detected by the antennae of males only. These characteristics suggested a pheromonal function, and this has now been confirmed with behavioral tests. Field tests conducted during two seasons in a commercial red currant field in Washington State showed that synthetic racemic 1 is attractive to male J. integer under natural conditions. A clear dose-response was evident, with greatest numbers of girdlers caught in sticky traps baited with 10 mg of the pheromone (in rubber septa) and least in traps baited with 1 mg or less. During May 2002, 10, 5, 3, and 1 mg baited traps caught means of 41.4, 26.6, 6.7, and 2.7 males/trap/visit (3–5 day intervals), respectively, with a maximum of 229 males caught in a single trap baited with 5 mg. A new synthetic method for racemic 1 is presented. The absolute configuration of natural 1 from the male sawflies was determined to be (R). The potential for using the sex pheromone of J. integer to improve management of this currant and gooseberry pest is discussed.  相似文献

Multispecies sex pheromone trapping (trapping of more than one species in the same trap) for the bollworm,Helicoverpa (Heliothis) zea (Boddie); fall armyworm,Spodoptera frugiperda (J.E. Smith); armyworm,Pseudaletia unipuncta (Haworth); and the black cutworm,Agrotis ipsilon (Hufnagel) was evaluated. Baiting of individual traps with all possible combinations of the four species taken two at a time caused a reduction in the catches of at least one of any two species involved when compared to traps baited for only one species. These results demonstrated the species specificity of sex pheromones identified for these species. Although the effects were reduced in some cases, the interaction between species also was observed when adjacent traps were baited with the sex pheromone dispensers of the different species. These results indicate that the spacing between pheromone traps when more than one species are being trapped at the same site affects trap capture and should be evaluated further.This paper reports the results of research only. The sex pheromone of the fir coneworm moth consists of a blend of (3Z,6Z,9Z,12Z,15Z)-pentacosapentaene and (9Z,11E)-tetradecadienyl acetate. Analogous blends of polyunsaturated, long-chain hydrocarbons with much shorter chain aldehydes or alcohols recently have been discovered in three other moth species in the superfamily Pyraloidea. These combinations of components from two distinct structural classes may represent an important and widespread new pheromone blend motif within the Lepidoptera.  相似文献

In feeding experiments with insects reared in the laboratory, the presence of the dihydropyrrolizines hydroxydanaidal and danaidal in the male scent organs (coremata) of the arctiids,Estigmene acrea (Drury),Phragmatobia fuliginosa (L.), andPyrrharctia isabella (J.E. Smith), was shown to depend on the presence of a source of pyrrolizidine alkaloids (PAs) in the larval diet.Phragmatobia males given an artificial diet supplemented with the powdered roots of the PA-containing plantSymphytum officinale L. (comfrey) produced more hydroxydanaidal than danaidal, whereas males given an artificial diet supplemented with dried whole plants of another PA-containing species,Senecio vulgaris L., produced more danaidal than hydroxydanaidal.Pyrrharctia males produced hydroxydanaidal with little if any danaidal, whether the source of PAs was comfrey orS. vulgaris. A behavioral bioassay showed that the coremata of PA-deniedPyrrharctia male progeny of PA-denied parents were pheromonally inactive, whereas those of PA-denied male progeny of PA-supplied parents (male and/or female) were often active. This indicates that a small amount of pheromone is made from PAs transferred from the female to her eggs and that males effect copulatory transfers of PAs that are, in turn, passed to the eggs by the mated female. Field observations ofPhragmatobia andPyrrharctia larvae feeding on sources of PAs were reported. The PA monocrotaline was shown to be a feeding stimulant forPyrrharctia larvae.  相似文献

Using male antenna as the sensing element, three electroantennographic detection (EAD)-active peaks were detected from pheromone gland extracts of the citrus leafminer, Phyllocnistis citrella. Based on gas chromatography (GC)-mass spectrometry and GC-infrared data, the semiochemicals were tentatively identified as a novel pheromone, (Z,Z,E)-7,11,13-hexadecatrienal, a previously identified attractant, (Z,Z)-7,11-hexadecadienal, and (Z)-7-hexadecenal in a ratio of 30:10:1, respectively. Identification was confirmed with synthetic compounds, which gave retention times identical to those of the natural products on three capillary columns with polar and nonpolar phases. While traps baited only with the previously identified attractant alone did not catch any males in Brazil, binary and tertiary mixtures with the major constituents caught significantly more male moths than traps baited with five virgin females. This paper and the following paper (Moreira et al.) were submitted within a few days of each other. The editors and the authors agreed that they should be published in tandem.  相似文献

Honeybees (Apis mellifera) use odors to identify and discriminate among flowers during foraging. This series of experiments examined the ability of bees to detect and discriminate among the floral odors of different varieties of two species of canola (Brassica rapa and Brassica napus) and also among three varieties of snapdragons (Antirhinnum majus). Individual worker honeybees were trained using a proboscis extension assay. The ability of bees to distinguish a floral odor from an air stimulus during training increased as the number of flowers used during training increased. Bees conditioned to the odor of one variety of flower were asked to discriminate it from the odors of other flowers in two different training assays. Bees were unable to discriminate among flowers at the level of variety in a randomized presentation of a reinforced floral odor and an unreinforced floral odor. In the second type of assay, bees were trained with one floral variety for 40 trials without reinforcement and then tested with the same variety or with other varieties and species. If a bee had been trained with a variety of canola, it was unable to differentiate the odor of one canola flower from the odor of other canola flowers, but it could differentiate canola from the odor of a snapdragon flower. Bees trained with the odor of snapdragon flowers readily differentiated the odor of one variety of a snapdragon from the odor of other varieties of snapdragons and also canola flowers. Our study suggests that both intensity and odor quality affect the ability of honeybees to differentiate among floral perfumes.  相似文献

Trail-following bioassays show that trails of the two subterranean termites, Reticulitermes lucifugus grassei and R. santonensis, which are sympatric in some areas of southwestern France, are not species specific. Even when tested just above threshold level, trails from pentane extracts of whole workers of R. santonensis are always preferred by both species. If the R. santonensis extract is progressively diluted, the preference is lost. In purified extracts of workers of R. lucifugus grassei that elicited a trail-following response, only one compound is active. It was identified by GC-MS as the same major compound of the trail-following pheromone of R. santonensis: (Z,Z,E)-3,6,8-dodecatrien-1-ol (DTE-OH). The threshold concentration of activity of synthetic DTE-OH was determined to be 10–3 ng/cm of trail; optimal activity was obtained at 10–2 ng/cm of trail. The increase of trail-following activity of worker extracts of R. lucifugus grassei after hydrolysis by potassium hydroxide suggests that DTE-OH also is bound to other components in sternal gland secretions. DTE-OH was also identified in alates of R. lucifugus grassei, suggesting that the compound functions both as a trail-following and a sex pheromone, as has been shown to be the case in R. santonensis. This demonstrates the high economy developed by termites in their strategies of chemical communication.  相似文献

Analyses of extracts of pheromone glands and of volatiles from calling female fall armyworm moths,Spodoptera frugiperda (J.E. Smith), revealed the presence of the following compounds: dodecan-1-ol acetate, (Z)-7-dodecen-1-ol acetate, 11-dodecen-1-ol acetate, (Z)-9-tetradecenal, (Z)-9-tetradecen-1-ol acetate, (Z)-11-hexadecenal, and (Z)-11-hexadecen-1-ol acetate. The volatiles emitted by calling females differed from the gland extract in that the two aldehydes were absent. Field tests were conducted with sticky traps baited with rubber septa formulated to release blends with the same component ratios as those emitted by calling females. These tests demonstrated that both (Z)-7-dodecen-1-ol acetate and (Z)-9-tetradecen-1-ol acetate are required for optimum activity and that this blend is a significantly better lure than either virgin females or 25 mg of (Z)-9-dodecen-1-ol acetate in a polyethylene vial, the previously used standard. Addition of the other three acetates found in the volatiles did not significantly increase the effectiveness of the two-component blend as a bait for Pherocon 1C or International Pheromones moth traps.Mention of a commercial or proprietary product does not constitute an endorsement by the USDA.  相似文献

Ponderosa pine logs infested withIps paraconfusus males inhibited the attraction ofDendroctonus brevicomis in the field to either attractive logs cut from a ponderosa pine tree under attack byD. brevicomis or to their synthetic pheromones,exo-brevicomin, frontalin, and myrcene. Logs cut from trees under attack byD. brevicomis inhibited the response ofI. paraconfusus to logs infested with maleI. paraconfusus.Exo-brevicomin, frontalin, and myrcene did not inhibit their response but verbenone did. Verbenone was found in maleD. brevicomis dissected from attractive logs under attack during the same time the response ofI. paraconfusus was inhibited by these logs.Trans-verbenol andexo-brevicomin were found in femaleD. brevicomis while verbenone,trans-verbenol, and frontalin were found in maleD. brevicomis in relatively large amounts near the beginning of the aggregation phase of host colonization. All of these compounds had decreased at a similar rate 5 days later. This gradual decrease inexo-brevicomin and frontalin probably caused the observed reduction in attraction. The ecological significance of these compounds in relation to termination of the aggregation phase ofD. brevicomis and reduction of interspecific competition is discussed.Coleoptera: Scolytidae. Individuals of two species of crayfish (Orconectes virilis andO. propinquus) were tested in the laboratory for responses to chemicals released from physically damaged conspecifics. Individuals ofO. propinquus did not show an alarm response to crushed conspecifics. Individuals ofO. virilis responded to a water-borne substance released from crushed conspecifics by assuming an intermediate posture and ceasing movement. Similar alarm responses were shown by individuals ofO. virilis to crushed congeneric individuals (O. propinquus), and these responses were not eliminated by either freeze-thawing the crayfish used to prepare the signal or by treating freshly crushed crayfish with the enzyme trypsin. Individuals ofO. virilis showed strong feeding responses to solutions prepared from frozen fish flesh but showed a mixture of alarm and feeding responses to freshly killed fish. These results indicate that the alarm substance used byO. virilis is widespread.  相似文献

In nature, Papilio polytes utilizes a limited range of rutaceous plants as hosts. We isolated and identified oviposition stimulants for the butterfly from the foliage of its primary host plant Toddalia asiatica. Females readily deposited eggs in response to a methanolic extract of the plant. Partition of the extract with organic solvents revealed that chemicals responsible for eliciting egg-laying resided in a water-soluble fraction. Further bioassay-guided fractionation of the active fraction by column chromatography, preparative TLC, and HPLC led to the isolation of two oviposition stimulants. One was isolated from an amphoteric fraction and identified as trans-4-hydroxy-N-methyl-L-proline [(–)-(2S,4R)-4-hydroxy-1-methyl pyrrolidine-2-carboxylic acid; HMP]. The other, isolated from an acidic fraction, was identified as 2-C-methyl-D-erythronic acid [(–)-(2R,3R)-2-methyl-2,3,4-trihydroxybutanoic acid; MEA]. HMP alone evoked significant oviposition-stimulatory activity, although this was much lower than that of the original water-soluble fraction. MEA, on the other hand, alone did not elicit positive responses from females. However, HMP, when assayed in combination with MEA, markedly enhanced the female response, and the mixture was as active in stimulating oviposition as were the original water-soluble fraction and the plant foliage. We conclude that HMP is a substance crucial for host recognition by females, while MEA is a synergistic stimulant involved in host recognition and/or preference.  相似文献

A brief survey is given about recent results in the identification of semiochemicals in bark beetles: Males ofIps sexdentatus (Boern.), stressed by the attack on resinous trees produce large amounts of 3(S)-1-methyl-5-(1-hydroxyl-1-methylethyl)-cyclohexa-1,3-diene. The compound appears to be derived from 3-carene and acts as a repellent. Males ofIps typographus (L.), stressed through the attack on unsuitable host material release 3-methyl-7-methylene-1,3(E), 8-nonatriene, which seems to act as a repellent. The odor bouquet of three species ofPityogenes is described. The occurrence of (+)-grandisol and other compounds related to weevil pheromones points to a close relation between Scolytidae and Curculionidae. Females ofDendroctonus simplex (Le Conte) use (–)-frontalin as the main pheromone. 6-Methyl-6-hepten-2-one, a minor component among the volatile compounds released by the females, is regarded as a possible precursor of frontalin. Similarly, (2R,5S)-2(1-hydroxyl-1-methylethyl)-5-methyltetrahydrofuran, pityol, a pheromone ofPityophthorus spp., is regarded to at least share a common biogenetic precursor with 6-methyl-5-hepten-2-ol, sulcatol. A new bicylic acetal, 2-ethyl-1,5-dimethyl-6,8-dioxabicyclo[3.2.1]octane, is described as an aggregation pheromone of the beech bark beetle,Taphrorychus bicolor (Herbst). Structural relationships between bark beetle pheromones and plant volatiles are discussed.  相似文献

Gas chromatography–electroantennographic detection analysis of sex pheromone gland extracts of the common forest looper Pseudocoremia suavis (Lepidoptera: Geometridae), a polyphagous defoliator of introduced Pinaceae and many New Zealand trees, revealed four compounds that elicited antennal responses. The two major active compounds (6Z)-cis-9,10-epoxynonadec-6-ene and (3Z,6Z)-cis-9,10-epoxynonadeca-3,6-diene were identified by comparison with known standards. Of the two minor active compounds, one was tentatively identified as (3Z,6Z)-cis-9,10-epoxyhenicosa-3,6-diene, whereas the other could not be identified because of insufficient amounts in extracts. (6Z)-cis-9,10-Epoxynonadec-6-ene, (3Z,6Z)-cis-9,10-epoxynonadeca-3,6-diene, and (3Z,6Z)-cis-9,10-epoxyhenicosa-3,6-diene were present in P. suavis gland extracts from Eyrewell Forest, a Pinus radiata plantation in the South Island of New Zealand, in a ratio of 35:65:5, respectively. Trapping trials in Eyrewell Forest established that (6Z)-cis-9,10-epoxynonadec-6-ene attracted male P. suavis. However, addition of (3Z,6Z)-cis-9,10-epoxyhenicosa-3,6-diene to the lure at <10% of (6Z)-cis-9,10-epoxynonadec-6-ene reduced capture of male moths, suggesting that one of its enantiomers was acting as a behavioral antagonist. During January–March of 2005, a blend trial involving single, binary, and ternary mixtures of the three components at Eyrewell Forest and at three other sites (two in the South Island and one in the North Island) revealed the existence of a second taxon of P. suavis at the three additional sites that was attracted to lures containing (3Z,6Z)-cis-9,10-epoxynonadeca-3,6-diene, either singly or in binary and ternary mixtures with (6Z)-cis-9,10-epoxynonadec-6-ene and (3Z,6Z)-cis-9,10-epoxyhenicosa-3,6-diene. This second taxon was not attracted to lures loaded solely with (6Z)-cis-9,10-epoxynonadec-6-ene.  相似文献

A bioassay was developed to test the hypothesis that secondary metabolites from marine algae affect feeding by sea urchins. During experiments measuring chemoreception and gustation, feeding by the regular sea urchin,Lytechinus variegatus (Lamarck), was inhibited by extracts from the green marine alga,Caulerpa prolifera (Forsskal) Lamouroux andCymopolia barbata (Linneaus) Lamouroux. Cymopol, a monoterpene-bromohydroquinone component ofC. barbata, was partially responsible for the inhibited feeding observed in tests of theCymopolia crude extract. Caulerpenyne, an oxygenated sesquiterpene fromC. prolifera, was responsible for all of the urchin feeding inhibition observed in tests of theC. prolifera crude extract. Feeding was not affected by: (1) extracts from several otherCaulerpa species includingC. mexicana (Sonder) J. Agardh,C. ashmaedii Harvey,C. racemosa v.macrophysa (Kutzing) Taylor, andC. racemosa v.laetevirens (Montagne) Weber-van Bosse; (2) caulerpin, an indole-containing pigment isolated from all of theCaulerpa species exceptC. mexicana; and (3) an extract from the red marine alga,Gracilaria foliifera v.angustissima (Harvey) Taylor, which has no known secondary metabolites. Feeding inhibition was independent of the test diameter which correlated with the reproductive state of the urchins. Feeding inhibition was also independent of the starvation periods between experiments, and the temperature and salinity in ranges tolerated byL. variegatus obtained from the Florida Gulf Coast. The data strongly suggest that at least one alga,Caulerpa prolifera, is chemically defended against a dominant omnivore in its community.  相似文献

In the framework of an evolutionary study, trail pheromones have been studied in the most basal extant termite, Mastotermes darwiniensis (Mastotermitidae), and two other basal termites, the Termopsidae Porotermes adamsoni (Porotermitinae) and Stolotermes victoriensis (Stolotermitinae). Although workers of M. darwiniensis do not walk in single file while exploring a new environment under experimental conditions and are unable to follow artificial trails in ‘open field’ experiments, they do secrete a trail-following pheromone from their sternal glands. This unique behavior might reflect a primitive function of communication of the sternal gland. The major component of the pheromone appears to be the same in the three basal species: the norsesquiterpene alcohol (E)-2,6,10-trimethyl-5,9-undecadien-1-ol. This represents a new chemical category of trail-following pheromones for termites. The quantity of pheromone was estimated as 20 pg/individual in M. darwiniensis, 700 pg/individual in P. adamsoni, and 4 pg/individual in S. victoriensis. The activity threshold was 1 ng/cm in M. darwiniensis and 10 pg/cm in P. adamsoni. In M. darwiniensis, the trail pheromone was secreted by sternal gland 4 and to a lesser degree by sternal gland 3, sternal gland 5 being almost inactive. This study highlighted phylogenetic relationships between the Mastotermitidae and two subfamilies of the Termopsidae, the Porotermitinae and the Stolotermitinae.
