The clinical prospect of radioiodianted SP-4 as an atherosclerotic plaque imaging agent was studied,The SP-4 was synthesized by a solied phase method and identified by an amino acid analysis fater purification with PLC .SP-4 was labeled with ^131I and ^125I by the Chloramine-T method and purified throuth Sephadex G25 column,Tewlve New Zealand rabbits were divided into an atherosclerotic group (n=7,AR) and a control group (n=5,NR) ,All of the atherosclerotic rabbits were intravenous administrated with bovine serum albumin,then fed with high cholesterol and fat died,^125I-SP-4 was intravenous administrated to the rabbits of both groups,The biodistribution of ^125I-SP-4 in rabbits was investigated,The uptakes (%ID/g) in blood and thoracic aorta and abdominal aorta were calculated 4 hous postinjection.Macroautoradiography and microautoradiography were performed in 2 AR atherosclerotc adbominal aortas,The clearance of radionativity from plasma was very rapid,^125I-SP-4 was mainly excreted through kidneys ,The radioactive uptakes of abdominal aorta and thoracic aorta of AR at 4 hous postinjection wae significantly higher than that of NR.The films of macroautoradiography showed focal accumulation of the radioactivity in the areas of a newly formed edges of atheroscleotic plaques,On the slices of microautoradiography,the obvious radioactive accumulation could be found in the atherosclerotic plaques,THus it was seen that the SP-4 rematined its biological activity after radioiodination and was located at atherosclerotic lsions,it is potentially useful as an atherosclerotic plaque imaging agent.  相似文献

A novel histidine derivative containing 4-nitroimidazole, (S)-2-(4-((4-nitro-lH-imidazol-l-yl) methyl) benzamido) -3- (1H- imidazol-4-yl)propanoic acid (His-NI), was synthesized and labeled with [99mTc(CO)3(H2O)3]+. The tricarbonyl technetium complex, the 99mTc(CO)3-His-NI, showed a 99% yield under mild conditions at a low His-NI ligand concentration of 10-4mol·L-1, and its biodistribution in mice bearing S180 tumor had a selective accumulation in tumor (2.01±0.40 %ID/g at 1 h postinjection) and a slow clearance. The tumor/muscle ratio was 1.64 at 1 h, 3.10 at 4 h, and 3.88 at 24 h, indicating that the 99mTc(CO)3-His-NI has a potential to image tumor hypoxia.  相似文献

Ionic bifunctional membranes have been synthesized by grafting binary mixture of methacrylic acid (MAAc) and 4-vinyl pyridine (4-VP) onto Teflon-FEP film by pre-irradiation method. Optimum conditions pertaining to maximum percentage of grafting were evaluated as a function of different reaction parameters. Maximum percentage of grafting of binary mixture (MAAc-co-4-VP) (71.29%) was obtained at an optimum total dose of 54.48 kGy and the total concentration was 9.49 mol/L ([4-VP] = 0.07 mol/L and [MAAc ] = 9.42 mol/L) in 5 ml of water. The effect of alcohols as additives to the reaction medium on percent grafting of the binary mixture has also been studied. The membranes were characterized by FTIR spectroscopy, scanning electron microscopy and thermogravimetric analysis. Swelling studies of the membranes were performed in different solvents such as water, benzene, carbon tetrachloride and dimethyl formamide (DMF). Maximum swelling was observed in DMF with minimum swelling in benzene. Metal ion (Cu2+, Ni2+ and Fe2+) uptake studies show better affinity for Fe2+ ions. Conductance measurements in different aqueous salt solution showed that these membranes have affinity for Na+/K+ ions and Cl ions and hence can be used in desalination/separation processes for the separation of both type of cationic and anionic ions.  相似文献
