Use of antibiotics as feed additives in poultry production has been linked to the presence of antibiotic resistant bacteria in farm workers, consumer poultry products and the environs of confined poultry operations. There are concerns that these resistant bacteria may be transferred to communities near these operations; however, environmental pathways of exposure are not well documented. We assessed the prevalence of antibiotic resistant enterococci and staphylococci in stored poultry litter and flies collected near broiler chicken houses. Drug resistant enterococci and staphylococci were isolated from flies caught near confined poultry feeding operations in the summer of 2006. Susceptibility testing was conducted on isolates using antibiotics selected on the basis of their importance to human medicine and use in poultry production. Resistant isolates were then screened for genetic determinants of antibiotic resistance. A total of 142 enterococcal isolates and 144 staphylococcal isolates from both fly and poultry litter samples were identified. Resistance genes erm(B), erm(A), msr(C), msr(A/B) and mobile genetic elements associated with the conjugative transposon Tn916, were found in isolates recovered from both poultry litter and flies. Erm(B) was the most common resistance gene in enterococci, while erm(A) was the most common in staphylococci. We report that flies collected near broiler poultry operations may be involved in the spread of drug resistant bacteria from these operations and may increase the potential for human exposure to drug resistant bacteria.  相似文献

The application of poultry litter to agricultural fields can provide plant nutrients for crops and forage production, but fecal bacteria and the sex hormones estradiol and testosterone are components of litter that can be detrimental to the environment. Our objective was to determine if applications of poultry litter to small watersheds would contribute to the load of fecal bacteria and sex hormones to soil and runoff. We, therefore, investigated the fate and transport of fecal bacteria, estradiol and testosterone from surface applied poultry litter to four small cropped watersheds. Poultry litter was applied to meet the nitrogen requirements of pearl millet (Pennisetum glaucum L.) in 2000 and grain sorghum [Sorghum bicolor (L.) Moench] in 2001. Neither Salmonella nor Campylobacter were detected in the litter but the fecal indicator bacteria were. The average load of total coliforms, Escherichia coli, and fecal enterococci applied with the litter was 12.2, 11.9, and 12.7 log(10) cells ha(-1), respectively. The average load of estradiol and testosterone was 3.1 and 0.09 mg ha(-1), respectively. Runoff events first occurred 7 months after the first litter application in 2000, and 3 weeks after the second application in 2001. Only for the 25 July 2001 runoff event 3 weeks after the second litter application were the concentrations of total coliforms, E. coli, and fecal enterococci in runoff greater than background concentrations which were on average 5.2, 2.9, and 1.1 log(10) MPN 100 ml(-1), respectively. Average background levels of total coliforms, fecal enterococci, and E. coli in surface soil were 8.2, 7.9, and 3.5 log(10) cells kg(-1) soil. At the rate of litter application the concentrations of estradiol and testosterone in the litter did not appear to impact the background levels in the soil and runoff. Because concentrations of sex hormones in litter from other broiler operations are known to be greater than in the litter we applied, further study on the connection between concentrations of sex hormones in poultry litter and operational practices is recommended.  相似文献

Antibiotic and metal resistance is a global occurring phenomenon and a universal threat. The release of antibiotics and metals into the environment like natural water bodies stimulates resistant microbes that could become a reservoir for a resistant gene pool for other pathogenic bacteria. The aim of the study was to determine microbial diversity, distribution of antibiotic and metal resistance bacteria from five rivers. The resistance of 168 Gram negative isolates was tested against 10 antibiotics and five metals. The study revealed a high incidence of bacterial isolates resistant to ampicillin (44.7%), streptomycin (37.3%) and chloramphenicol (33.9%). The highest level of antibiotic resistance was detected in the River Ravi. The multiple antibiotic resistance (MAR) index of the five rivers ranged from <0.2 to 0.6. Isolates displayed tolerance to different concentrations of heavy metals. High antibiotic and metal‐resistances in samples from the Rivers Ravi and Soan suggest co‐resistance amongst the bacterial populations.  相似文献

The aim of the present study was to determine the level of antibiotic resistance patterns and distribution of heavy metal resistance of bacterial isolates from seawater, sediment and shrimps, and to determine if there is a relationship between antibiotic and heavy metal resistance. We undertook studies in 2007 in the industrially polluted Iskenderun Bay, on the south coast of Turkey. The resistance of 236 Gram-negative bacterial isolates (49 from seawater, 90 from sediment and 97 from shrimp) to 16 different antibiotics, and to 5 heavy metals, was investigated by agar diffusion and agar dilution methods, respectively. A total of 31 species of bacteria were isolated: the most common strains isolated from all samples were Escherichia coli (11.4%), Aeromonas hydrophila (9.7%) and Stenotrophomonas maltophilia (9.3%). There was a high incidence of resistance to ampicillin (93.2%), streptomycin (90.2%) and cefazolin (81.3%), and a low incidence of resistance to imipenem (16.5%), meropenem (13.9%) and cefepime (8.0%). Some 56.8% of all bacteria isolated from seawater, sediment and shrimp were resistant to 7 or more antibiotics. Most isolates showed tolerance to different concentrations of heavy metals, and minimal inhibition concentrations ranged from 12.5 μg/ml to > 3200 μg/ml. The bacteria from seawater, sediment and shrimp showed high resistance to cadmium of 69.4%, 88.9%, and 81.1% respectively, and low resistance to manganese of 2%, 6.7% and 11.3% respectively. The seawater and sediment isolates which were metal resistant also showed a high resistance to three antibiotics: streptomycin, ampicillin and trimethoprim-sulphamethoxazole. In contrast, the shrimp isolates which were metal resistant were resistant to four antibiotics: cefazolin, nitrofurantoin, cefuroxime and ampicillin. Our results show that Iskenderun Bay has a significant proportion of antibiotic and heavy metal resistant Gram-negative bacteria, and these bacteria constitute a potential risk for public health.  相似文献

Shrimp farming is a sufficiently large and mature industry to have an effective range of antimicrobial agents for most bacterial diseases in shrimp culture. However, at present, there exists great concern over the widespread use of antibiotics in aquaculture, which may result in residue of antibiotics in water and mud, and subsequently, the development of antibiotic resistance in bacteria in the environment. There is limited understanding about the effect of antibiotic residues on bacteria resistance in shrimp farming environment. Therefore, a study was conducted to investigate bacterial resistance to Norfloxacin (NFXC), Oxolinic Acid (OXLA), Trimethoprim (TMP) and Sulfamethoxazole (SMX), which were found in four shrimp farming locations in mangrove areas in Vietnam. Findings indicate that there is a relatively high incidence of bacteria resistance to these antibiotics observed in most of the studied sites, particularly to antibiotics with concentration of 0.1 microg/ml. Yet the relation between concentration of antibiotic residues and incidence of antibiotic resistance is not clearly defined. Among individual antibiotics, the incidence of resistance to TMP and SMX was higher than the others. Identification of bacteria isolated from mud samples by DNA analyzer shows that Bacillus and Vibrio are predominant among bacteria resistant to the antibiotics. The result of the study also indicates that these antibiotics in media degraded more rapidly due to the presence of resistant bacteria.  相似文献

In addition to mold, indoor bioaerosols also contain bacterial components that may have implications for human health. Endotoxin is a cell wall component in Gram-negative bacteria present at varying levels indoors that has been found to have respiratory health implications. Streptomyces is a large genus of Gram-positive bacteria, and some species have been shown to produce inflammatory reactions in vitro and in vivo. The aim of this study was to determine predictors of streptomycetes levels in house dust and to compare the variation in streptomycetes levels with that in endotoxin levels. Dust was collected by floor vacuuming from 178 homes in the Cincinnati metropolitan area. Streptomycetes levels were measured by quantitative PCR, and endotoxin was assayed by the Limulus amebocyte lysate method. Associations between home characteristics and bacterial contaminants, expressed as concentration and load, were investigated through multiple regression analyses. The presence of two or more dogs was a strong predictor of both streptomycetes and endotoxin levels. Season of dust collection and levels of outdoor molds were predictors of streptomycetes but not endotoxin levels. In contrast, number of inhabitants was a significant predictor of endotoxin load only. Neither streptomycetes nor endotoxin levels were associated with metrics of moisture damage.  相似文献

Increasing concentration of heavy metals and antibiotics within bacterial microenvironments contribute to medical problems. This research confirmed the presence of enteropathogenic bacteria in the sediment and water of Kahwa River. Vibrio cholerae, Shigella dysenteriae, Salmonella typhi and Salmonella paratyphi showed resistance to antibiotics and tolerance to heavy metals. The bacteria, particularly Vibrio, were extensively resistant to Ampicillin; less so to Chloramphenicol, Co-trimoxazole and Tetracycline with a relatively reduced resistance shown to Ciprofloxacin. Higher levels of antibiotic resistance were noticed in bacteria isolated from river sediment when compared to bacteria isolated from river water (p < 0.05). Tolerance to cadmium and lead was also more obvious in bacteria isolated from river sediment. An increasing association was observed between multiple antibiotic resistance and tolerance to heavy metals. Bacterial growth in the presence of cadmium was restricted when compared to lead.  相似文献

The organic water quality throughout a poultry processing wastewater treatment and renovation system, which included several points of chlorination, was investigated. The main classes of organic compounds which were identified were substituted phthalates and adipates and (methylated) fatty acids and their amide derivatives. Chloroform concentrations were lower than 8 μg l−1 throughout the renovation system. Total organic halide, total organic carbon and endotoxin concentrations in the final renovated water were slightly high when compared to typical levels occurring in finished waters from other systems. It was also determined that as treatment progressed, the contribution of compounds greater in molecular size than 400 Daltons to organic carbon content decreased. Based on these and previous results, it was concluded that there would not be an undue health risk if the renovated water would be recycled and reused for a trial period.  相似文献

Veterinary antibiotics are widely used at concentrated animal feeding operations (CAFOs) to prevent disease and promote growth of livestock. However, the majority of antibiotics are excreted from animals in urine, feces, and manure. Consequently, the lagoons used to store these wastes can act as reservoirs of antibiotics and antibiotic-resistant bacteria. There is currently no regulation or control of these systems to prevent the spread of these bacteria and their genes for antibiotic resistance into other environments. This study was conducted to determine the disinfection potential of chlorine, ultraviolet light and ozone against swine lagoon bacteria. Results indicate that a chlorine dose of 30 mg/L could achieve a 2.2-3.4 log bacteria reduction in lagoon samples. However, increasing the dose of chlorine did not significantly enhance the disinfection activity due to the presence of chlorine-resistant bacteria. The chlorine resistant bacteria were identified to be closely related to Bacillus subtilis and Bacillus licheniformis. A significant percentage of lagoon bacteria were not susceptible to the four selected antibiotics: chlortetracycline, lincomycin, sulfamethazine and tetracycline (TET). However, the presence of both chlorine and TET could inactivate all bacteria in one lagoon sample. The disinfection potential of UV irradiation and ozone was also examined. Ultraviolet light was an effective bacterial disinfectant, but was unlikely to be economically viable due to its high energy requirements. At an ozone dose of 100 mg/L, the bacteria inactivation efficiency could reach 3.3-3.9 log.  相似文献

The occurrence and spread of multi-drug resistant bacteria is a pressing public health problem. The emergence of bacterial resistance to antibiotics is common in areas where antibiotics are heavily used, and antibiotic-resistant bacteria also increasingly occur in aquatic environments. The purpose of the present study was to evaluate the impact of the wastewater treatment process on the prevalence of antibiotic resistance in Acinetobacter spp. in the wastewater and its receiving water. During two different events (high-temperature, high-flow, 31 °C; and low-temperature, low-flow, 8 °C), 366 strains of Acinetobacter spp. were isolated from five different sites, three in a wastewater treatment plant (raw influent, second effluent, and final effluent) and two in the receiving body (upstream and downstream of the treated wastewater discharge point). The antibiotic susceptibility phenotypes were determined by the disc-diffusion method for 8 antibiotics, amoxicillin/clavulanic acid (AMC), chloramphenicol (CHL), ciprofloxacin (CIP), colistin (CL), gentamicin (GM), rifampin (RA), sulfisoxazole (SU), and trimethoprim (TMP). The prevalence of antibiotic resistance in Acinetobacter isolates to AMC, CHL, RA, and multi-drug (three antibiotics or more) significantly increased (p < 0.01) from the raw influent samples (AMC, 8.7%; CHL, 25.2%; RA, 63.1%; multi-drug, 33.0%) to the final effluent samples (AMC, 37.9%; CHL, 69.0%; RA, 84.5%; multi-drug, 72.4%), and was significantly higher (p < 0.05) in the downstream samples (AMC, 25.8%; CHL, 48.4%; RA, 85.5%; multi-drug, 56.5%) than in the upstream samples (AMC, 9.5%; CHL, 27.0%; RA, 65.1%; multi-drug, 28.6%). These results suggest that wastewater treatment process contributes to the selective increase of antibiotic resistant bacteria and the occurrence of multi-drug resistant bacteria in aquatic environments.  相似文献

ABSTRACT

Automated Teller Machines (ATM) are likely to be contaminated with various microorganisms because of their contact with the hands of many users daily. The main objective of this study was to investigate ATMs as a potential source of bacterial contamination. This study was conducted in the Metropolitan Area of Porto, in which 50 swab samples were cultured on selective media. Some isolates were identified based on colonial, morphological and biochemical characteristics. Susceptibility to several antibiotics was also evaluated for each isolated organism. Most contaminated ATMs had high numbers of Enterobacteriaceae and Enterococci. Staphylococci and Enterobacteriaceae were the most prevalent bacteria found. The recovered bacterial isolates varied in their antibiotic resistance pattern; Staphylococcus spp. demonstrated the most resistant profiles. With this pioneering study in Portugal, it was demonstrated that although ATMs in metropolitan area of Porto were not highly contaminated, some potentially pathogenic bacteria were present and resistant to some commonly used antibiotics.  相似文献

Antibiotic resistance in clinical settings has been studied from last few decades but the possibility of development of antibiotic resistance at polluted environmental sites is also of concern. In developing countries, the major source of antibiotic contamination in surface water is improper disposal of effluents from industries, hospitals and domestic waste water treatment plants. The antibiotic pollution combined with other environmental pollution factors exerts selective pressure on environmental microbes, driving evolution and resulting in the spread of antibiotic resistance in a local to global scale. Present study aimed to determine the proliferation of ciprofloxacin resistant bacteria in aquatic environment of a river which is heavily impacted by industrial effluents. Correlation analyses were performed to evaluate the impact of anthropogenic factors. Statistical analysis evaluated the effect of fluoroquinolones, heavy metals, total organic carbon and total nitrogen on the levels of resistant culture in samples from the natural settings.  相似文献

Antimicrobial resistance of enterococci was investigated in 42 samples of crude inflow, treated effluent and sludge collected in 14 municipal sewage treatment plants of Portugal. A total of 983 enterococci were recovered and tested, using the diffusion agar method, regarding their sensitivity to 10 different antimicrobial drugs. Multidrug resistance was present in 49.4% of the isolates. Only 3.3% and 0.6% of the investigated strains were resistant to ampicillin and vancomycin, respectively. Resistances found against rifampicin (51.5%), tetracycline (34.6%), erythromycin (24.8%) and nitrofurantoin (22.5%), are causes for substantial concern. Almost 14% of isolates were resistant to ciprofloxacin. Wastewater treatment resulted in enterococci decrease between 0.5 and 4log; nevertheless, more than 4.4 x 10(5)CFU/100ml were present in the outflow of the plants. Our data indicate that the use of antimicrobials had created a large pool of resistance genes and that sewage treatment processes are unable to avoid the dissemination of resistant enterococci into the environment.  相似文献

The quality of harvested rainwater used for toilet flushing in a private house in the south-west of France was assessed over a one-year period. Temperature, pH, conductivity, colour, turbidity, anions, cations, alkalinity, total hardness and total organic carbon were screened using standard analytical techniques. Total flora at 22 °C and 36 °C, total coliforms, Escherichia coli and enterococci were analysed. Overall, the collected rainwater had good physicochemical quality but did not meet the requirements for drinking water. The stored rainwater is characterised by low conductivity, hardness and alkalinity compared to mains water. Three widely used bacterial indicators - total coliforms, E. coli and enterococci - were detected in the majority of samples, indicating microbiological contamination of the water. To elucidate factors affecting the rainwater composition, principal component analysis and cluster analysis were applied to the complete data set of 50 observations. Chemical and microbiological parameters fluctuated during the course of the study, with the highest levels of microbiological contamination observed in roof runoffs collected during the summer. E. coli and enterococci occurred simultaneously, and their presence was linked to precipitation. Runoff quality is also unpredictable because it is sensitive to the weather. Cluster analysis differentiated three clusters: ionic composition, parameters linked with the microbiological load and indicators of faecal contamination. In future surveys, parameters from these three groups will be simultaneously monitored to more accurately characterise roof-collected rainwater.  相似文献

The intensive use of antimicrobial agents, mainly oxytetracycline, to prevent and control bacterial pathologies in Chilean salmon culture is a frequent practice. A total of 103 gram-negative oxytetracycline-resistant bacteria recovered from various sources of 4 Chilean freshwater salmon farms were identified and investigated for their susceptibility patterns to various antibacterial agents, by using an agar disk diffusion method. Antibacterial resistance patterns of isolates were not correlated with bacterial species or strain source. A high number of bacteria resistant to amoxicillin, ampicillin. erythromycin, and furazolidone, as well as an important frequency of bacterial resistance to florfenicol, chloramphenicol, cefotaxime and trimethoprim-sulfamethoxazole was found. On the contrary, the proportion of bacteria resistant to gentamicin, kanamycin, flumequine and enrofloxacin was rather low. Resistant microflora showed a high taxonomic variability and mainly consisted of non-fermenting bacteria (77.7%). These strains mainly belonged to the species Pseudomonas fluorescens (29), Aeromonas hydrophila (10), Stenotrophomonas maltophilia (6), isolated from salmon fingerlings, and Acinetobacter lwoffii (5) isolated from pelletized feed. The occurrence of simultaneous resistance to various antibacterials was frequent. We observe a high frequency of bacteria resistant to 6-10 antibacterials (74 strains), and antibiotic resistance index (ARI) values ranging from 0.38 to 0.48 for the four salmon farms studied. These results suggest that Chilean salmon farms might play a role as reservoirs of antibacterial multiresistant bacteria, thus prompting the necessity for a more restrictive attitude towards the intensive use of antibacterials in salmon farming.  相似文献

Arsenic compounds have been used extensively in agriculture in the US for applications ranging from cotton herbicides to animal feed supplements. Roxarsone (3-nitro-4-hydroxyphenylarsonic acid), in particular, is used widely in poultry production to control coccidial intestinal parasites. It is excreted unchanged in the manure and introduced into the environment when litter is applied to farmland as fertilizer. Although the toxicity of roxarsone is less than that of inorganic arsenic, roxarsone can degrade, biotically and abiotically, to produce more toxic inorganic forms of arsenic, such as arsenite and arsenate. Experiments were conducted on aqueous litter leachates to test the stability of roxarsone under different conditions. Laboratory experiments have shown that arsenite can be cleaved photolytically from the roxarsone moiety at pH 4-8 and that the degradation rate increases with increasing pH. Furthermore, the rate of photodegradation increases with nitrate and natural organic matter concentration, reactants that are commonly found in poultry-litter-water leachates. Additional photochemical reactions rapidly oxidize the cleaved arsenite to arsenate. The formation of arsenate is not entirely undesirable, because it is less mobile in soil systems and less toxic than arsenite. A possible mechanism for the degradation of roxarsone in poultry litter leachates is proposed. The results suggest that poultry litter storage and field application practices could affect the degradation of roxarsone and subsequent mobilization of inorganic arsenic species.  相似文献

Heavy metal pollution of river water has health implications. This study was aimed to test if bacteria that are heavy metal resistant can resist antibiotics. Heavy metal-resistant bacteria were isolated using Nutrient agar plates amended with 100?μg?ml?1 of metal salts. Isolates were further screened to determine minimal inhibitory concentration against each metal. The isolates belonged to the genera of Neisseria, Bacillus, Pseudomonas, Staphylococcus, Streptococcus, Micrococcus, Corynebacterium and Proteus. Minimum inhibitory concentration of the isolates for heavy metals ranged from 200 to 2300?μg?ml?1. Isolates of Pseudomonas, Neisseria, Bacillus, Protues, Streptococcus, Staphylococcus and Micrococcus spp. exhibited multiple antibiotic resistance. Statistically significant correlation between metal resistance and antibiotic resistance was observed among bacterial isolates. The present work suggests that some of the isolates could be hazardous to human beings as a result of natural selection of dual resistant mechanisms.  相似文献

When Enterococcus faecalis is isolated from fresh feces, its host range appears to be limited to humans and birds. Although E. faecalis is found in human sewage, the extent to which the bacterium is found in broiler litter and in the feces of wild birds is unclear. These results have implications for bacterial source tracking. We determined if media designed for the isolation of fecal enterococci affected this host range, and if E. faecalis was routinely found in broiler litter and in the feces of wild birds. Of five different isolation media, none affected the isolation of E. faecalis. Enterococcus faecalis was routinely found in fresh broiler feces (522 of 1092 isolates; 48%), but rarely in broiler litter (12 of 1452 isolates; <2%). Therefore, broiler litter selects against this bacterium, and broiler litter is an unlikely environmental source of this bacterium. The presence of E. faecalis in eight wild bird species was highly variable. Unless the fecal loading rate from migratory or resident wild birds is high, water samples collected during baseflow conditions with high numbers of E. faecalis may indicate human fecal contamination.  相似文献

Abstract Residential microbial exposure has been suggested to be involved in the development of asthma. This paper describes bacterial endotoxin and mold β(1→3)-glucan levels in house dust and the relationship to selected home characteristics. Dust was sampled from mattresses and living room and bedroom floors of 25 houses in Germany. Endotoxin and β(1→3)-glucan levels ranged from 200-48,600 EU/g dust (100-32,900 EU/m2 sampled surface) and 182-3,507 μg/g (157-3,652 μg/m2), respectively. Bio-contaminant levels were highest on living room floors and lowest in mattresses. Dust, endotoxin and β(1→3)-glucan levels were 2–3 times higher on living room floors of centrally heated houses built after 1970 compared to older individually heated houses. This was not found for mattresses and bedroom floors. No associations between biocontaminant levels and other selected home characteristics (temperature, relative humidity, damp spots and insulation of windows) were found. β(1→3)-glucan levels were associated with total culturable fungi (per m2) in house dust, as well as with the fungal genus Alternaria (per g dust and per m2). In conclusion endotoxin and β(1→3)-glucan were readily detectable in house dust and significantly associated with heating system and/or age of the home.  相似文献

Little is known about the microbial selectivity of UV treatment for antibiotic resistant bacteria, and the results of limited studies are conflicting. To understand the effect of UV disinfection on antibiotic resistant bacteria, both total heterotrophic bacteria and antibiotic resistant bacteria (including cephalexin-, ciprofloxacin-, erythromycin-, gentamicin-, vancomycin-, sulfadiazine-, rifampicin-, tetracycline- and chloramphenicol-resistant bacteria) were examined in secondary effluent samples from a municipal wastewater treatment plant. Bacteria resistant to both erythromycin and tetracycline were chosen as the representative of multiple-antibiotic-resistant bacteria and their characteristics after UV treatment were also investigated.  相似文献
