A temperature- and pH-responsive drug delivery system was studied by using interpenetrating polymer network (IPN) hydrogels constructed with poly(acrylic acid) (PAAc) and poly(vinyl alcohol) (PVA). The release of indomethacin incorporated into these hydrogels showed pulsatile patterns in response to both pH and temperature. Indomethacin diffused from the polymer matrices through the swelling and deswelling mechanism. The release amount increased at higher temperature because of the swelling caused by the dissociation of hydrogen bonding. The drastic change of drug release was achieved by alternating pH of the buffer solution and was attributed to the change of states of ionic groups within IPN hydrogels. The free water contents were calculated by using differential scanning calorimetry (DSC), and were proved to be the main factor in the swelling. These results demonstrated that the drug release could be controlled by the swelling/deswelling degree of IPN hydrogels as functions of pH and/or temperature. © 1997 John Wiley & Sons, Inc. The electrically modulated properties of interpenetrating polymer networks (IPN) composed of poly(vinyl alcohol) (PVA) and poly(acrylic acid) (PAAc) under electric field were investigated for drug delivery systems. PVA/PAAc IPNs with various compositions were synthesized by a sequential method, that is, ultraviolet polymerization of AAc in the mixture of PVA and aqueous AAc monomer solution, followed by a freeze‐thawing process to prepare elastic hydrogels. The amount of loaded drug significantly increased with the content of PAAc containing ionizable groups in IPN. The amount of introduced ionic drug (cefazoline) was greater than that of the nonionic drug (theophylline). Release behaviors of drug molecules from negatively charged PVA/PAAc IPN were switched on and off in a pulsatile pattern depending on the applied electric stimulus. The released amount and the release rate of drug were influenced significantly by the applied voltage, ionic group contents in IPN, ionic properties of drug solute, and the ionic strength of release medium. In addition, the ionic properties of drug molecules dramatically affected release behaviors, thus the release of ionic drug was much more enhanced than that of the nonionic drug. © 1999 John Wiley & Sons, Inc. Hydrogels based on pH‐sensitive polymers are of great interest as potential biomaterials for the controlled delivery of drug molecules. In this study, a novel, pH‐sensitive hydrogel was synthesized by poly(aspartic acid) (PASP) crosslinked with 1,6‐hexanediamine and reinforced with ethylcellulose (EC). The loading and release characteristics of naproxen sodium (NS) were studied. The PASP–EC blend hydrogels had pH‐sensitive characteristics and were strongly dependent on the pH value. The release kinetics for NS from the PASP–EC blend hydrogels and PASP hydrogel were evaluated in simulated gastric fluid (pH = 1.05) and simulated intestinal fluid (pH = 6.8) at 37°C. The results showed that the drug‐loaded hydrogels were resistant to simulated gastric fluid, and hence, they could be useful for oral drug delivery. Compared with the PASP hydrogel, the PASP–EC blend hydrogels showed a lower release rate of NS in the same pH conditions. It was evident that the presence of hydrophobic groups (EC) retarded the release of NS and led to sustained release. The kinetics of NS release from the drug‐loaded hydrogels conformed to the Korsmeyer–Peppas model. The release exponent of the model was 0.7291, which indicated multiple drug release. The PASP–EC blend hydrogels were biodegradable and pH sensitive; there would be a wide range of applications for them in controlled drug‐delivery systems. © 2009 Wiley Periodicals, Inc. Copolymers composed of poly(vinyl alcohol) (PVA) and poly(dimethylsiloxane) (PDMS) were crosslinked with chitosan to prepare semi‐interpenetrating polymer network (IPN) hydrogels by an ultraviolet (UV) irradiation method for application as potential biomedical materials. PVA/PDMS copolymer and chitosan was cast to prepare hydrogel films, followed by a subsequent crosslinking with 2,2‐dimethoxy‐2‐phenylacetophenone as a nontoxic photoinitiator by UV irradiation. Various semi‐interpenetrating polymer networks (semi‐IPNs) were prepared from different weight ratios of chitosan and the copolymer of PVA/PDMS. Photocrosslinked hydrogels exhibited an equilibrium water content (EWC) in the range of 65–95%. Swelling behaviors of these hydrogels were studied by immersion of the gels in various buffer solutions. Particularly, the PCN13 as the highest chitosan weight ratio in semi‐IPN hydrogels showed the highest EWC in time‐dependent and pH‐dependent swelling. © 2002 Wiley Periodicals, Inc. pH‐sensitive anionic hydrogels composed of poly(vinyl alcohol) (PVA) and poly(γ‐glutamic acid) (γ‐PGA) were prepared by the freeze drying method and thermally crosslinked to suppress hydrogel deformation in water. The physical properties, swelling, and drug‐diffusion behaviors were characterized for the hydrogels. In the equilibrium swelling study, PVA/γ‐PGA hydrogels shrunk in pH regions below the pKa (2.27) of γ‐PGA, whereas they swelled above the pKa. In the drug‐diffusion study, the drug permeation rates of the PVA/γ‐PGA hydrogels were directly proportional to their swelling behaviors. The cytocompatibility test showed no cytotoxicity of the PVA/γ‐PGA hydrogels for the 3T3 fibroblast cell lines. The results of these studies suggest that hydrogels prepared from PVA and γ‐PGA could be used as orally administrable drug‐delivery systems. © 2008 Wiley Periodicals, Inc. Novel thermo-sensitive N,N-diethylacrylamide (DEAAm) based copolymer hydrogels were prepared via UV-induced free radical bulk polymerisation. UV polymerisation was employed to avoid the use of potentially toxic solvents; solution polymerisation has been the most common means for the preparation of PDEAAm-based hydrogels in the literature to date. The resultant hydrogels were analysed using nuclear magnetic resonance, Fourier transform infrared spectroscopy and modulated differential scanning calorimetry. Parameters such as the crosslinking degree and the nature of the incorporated hydrophilic component, N-vinyl-2-pyrrolidone (NVP) or N,N-dimethylacrylamide (DMAAm) were found to impact hydrogel structure, mechanical properties and swelling kinetics. Pulsatile swelling studies indicated that the hydrogels had thermo-reversible properties which were greatly affected by test temperature, nature of hydrophilic monomer used and crosslinker content. Aminophylline was selected as a model solute for drug loading and release studies by thermal deswelling in HCl buffer (pH 1.4) and phosphate buffer media (pH 6.8). The observed lag time prior to significant drug release from the more crosslinked P(DEAAm-NVP) hydrogels could make them suitable for delayed specific release in the intestine and potential alternatives to layers or membranes in time-specific and site-specific swelling-controlled drug delivery systems.  相似文献

Semi‐ and full‐interpenetrating polymer network (IPN) hydrogels composed of poly(vinyl alcohol) and polyethyleneimine (PEI) were prepared to investigate the bending behavior under the electric response. To find out the characteristics of the hydrogel in the medium, swelling ratio, and rate and water state of the hydrogels were measured. The swelling ratio of the semi‐IPN hydrogels increased with PEI content in the matrix, whereas that of full‐IPN hydrogels dramatically decrease with increase of PEI contents in the hydrogels. In the water state of hydrogel, the bound water and free water of semi‐IPN hydrogels increased with PEI weight ratio. The full‐IPN hydrogels show the lower free water content in comparison with the semi‐IPN hydrogel. The IPN hydrogels exhibited bending angle change in response to external stimulus such as voltage, the bending angle increased with PEI concentration. In addition, the repeated bending behaviors according to the magnitude of the applied electric field revealed that the bending angle is reversible without collapse of formation of hydrogel in all samples. Poly(vinyl alcohol) (PVA) and poly(N-vinyl pyrrolidone) (PVP) composite hydrogel with interpenetrating polymer networks (IPNs) was prepared by in situ polymerization and compared with pure PVA hydrogel. The prepared IPN hydrogel was characterized by infrared spectroscopy (IR), differential scanning calorimetry (DSC), dynamic mechanical analysis (DMA), X-ray photoelectron spectroscopy (XPS), and scanning electron microscopy. The mechanical property and cell culture were also tested. The results show that PVP can chemically bond with PVA and form uniform blend hydrogel. The content of PVP can affect the structure, crystallinity, glass transition temperature (Tg), and mechanical property of the hydrogel. The Tg of the PVA hydrogel is 2.7°C while the Tg of the IPN hydrogel is −37°C. The IPN hydrogel has lower glass transition temperature, corresponding to better elastic properties, and has better mechanical performance on stretch and compression than PVA hydrogel. The crystallinity (Xc) of PVA hydrogel and IPN hydrogel is 65.3 and 26.3%, respectively. The DMA curves and XPS analysis suggest that PVA and PVP are well miscible on a molecular level in the IPN hydrogel. The cell proliferation trend demonstrates that the addition of PVP has a positive influence on the cell growth and the IPN hydrogel may be used as a promising biomaterial for artificial cartilage substitute. © 2012 Wiley Periodicals, Inc. Temperature and pH‐responsive interpenetrating polymer network (IPN) hydrogels, constructed with poly(methacrylic acid) (PMAA) and poly(vinyl alcohol) (PVA), by a sequential IPN method, were studied. The characterization of IPN hydrogels was investigated by Fourier‐transform infrared spectroscopy, differential scanning calorimetry (DSC) and swelling under various conditions. The IPN hydrogels exhibited relatively high swelling ratios, in the range 230–380 %, at 25 °C. The swelling ratios of the PMAA/PVA IPN hydrogels were pH and temperature dependent. DSC was used for the quantitative determination of the amounts of freezing and non‐freezing water. The amount of free water increased with increasing PMAA content in the IPN hydrogels. BACKGROUND: Blended hydrogels are widely applied in medical fields. They can provide many advantages, such as biocompatibility and biodegradability. Many materials and methods are used to obtain blended hydrogels. In this work, carboxymethyl chitosan (CMCS) and poly(vinyl alcohol) (PVA) blended hydrogels were prepared using the freezing and thawing technique. The properties of the hydrogels prepared, i.e. gel fraction, swelling and pH‐responsive behaviors, were investigated. RESULTS: The gel fraction increased with increasing time of freezing and thawing as determined through gravimetric analysis. It was also found that the equilibrium degree of swelling improved obviously due to the addition of CMCS compared to pure PVA hydrogel. The blended hydrogel with composition CMCS/PVA 80/20 (by weight) possessed the highest swelling ratio. The results of the influence of pH values on the swelling behavior showed that minimum swelling ratios of the hydrogels occurred near the isoelectric point of CMCS. Protein release studies were performed under various pH conditions: the release was much slower under acid than under basic conditions. The release showed a burst in the first 15 h and then steadily increased. CONCLUSION: The addition of CMCS can improve the physical properties of pure PVA hydrogels and provide pH sensitivity. Temperature‐sensitive poly(N‐isopropylacrylamide) hydrogels were successfully synthesized by using poly(ethylene oxide) as the interpenetrating agent. The newly prepared semi‐interpenetrating polymer network (semi‐IPN) hydrogels exhibited much better properties as temperature‐sensitive polymers than they did in the past. Characterizations of the IPN hydrogels were investigated using a swelling experiment, FTIR spectroscopy, and differential scanning calorimetry (DSC). Semi‐IPN hydrogels exhibited a relatively high temperature dependent swelling ratio in the range of 23–28 at room temperature. A novel biodegradable sodium poly(aspartic acid) (PASP) hydrogel with microporous structure was manufactured using electrospun polysuccinimide (PSI) nanofibers. PSI is the intermediate of sodium PASP and could be electospun into nanofibers easily. Firstly, PSI nanofibers were prepared from PSI/N, N-dimethylformamide solution. Then the PSI nanofibrous mats were crosslinked and hydrolyzed to obtain biodegradable microporous sodium PASP nanofibrous hydrogels. The chemical structures, morphologies and pore sizes of PSI nanofibrous mats and microporous sodium PASP nanofibrous hydrogels were investigated. Moreover, the properties of PSI electrospinning solutions, and the swelling ratio and biodegradability of sodium PASP hydrogels were also examined. The results showed that the swelling ratio of microporous sodium PASP nanofibrous hydrogels achieved to 21.0–24.3 g/g and were obviously higher than that of the sodium PASP casting film, reporting a swelling ratio of only 4.6 g/g. When the microporous sodium PASP nanofibrous hydrogel was immersed in water, it exhibited quick absorption and morphological robustness. The microporous sodium PASP nanofibrous hydrogel showed 83 wt% weight loss after 7 days of trypsin catalyzed biodegradation, and the SEM analysis demonstrated the significant morphology change of the microporous sodium PASP nanofibrous hydrogel during the biodegradation.  相似文献

pH‐ and temperature‐responsive interpenetrating polymer network (IPN) hydrogels based on soy protein and poly(N‐isopropylacrylamide‐co‐sodium acrylate) were successfully prepared. The structure and properties of the hydrogels were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, differential scanning calorimetry, and thermogravimetric analyzer. The equilibrium and dynamic swelling/deswelling behaviors and the drug release properties of the hydrogels responding to pH and/or temperature were also studied in detail. The hydrogels have the porous honeycomb structures, good miscibility and thermal stability, and good pH‐ and temperature‐responsivity. The volume phase transition temperature of the hydrogels is ca. 40°C. Changing the soy protein or crosslinker content could be used to control the swelling behavior and water retention, and the hydrogels have the fastest deswelling rate in pH 1.2 buffer solutions at 45°C. Bovine serum albumin release from the hydrogels has the good pH and temperature dependence. A series of interpenetrating polymer networks (IPN) hydrogels with different compositions that based on xanthan gum (XG) and poly(aspartic acid) (PASP) were synthesized. The effects of various external surrounding stimuli, including pH, temperature, and ionic strength on XG–PASP hydrogels swelling properties were investigated. Chemical structural changes of the IPN hydrogels were characterized by Fourier transform infrared spectroscopy (FT‐IR), differential scanning calorimetry (DSC), transmission electron microscopy (TEM), and swelling ratio measurement. The swelling process was found to be a Fickian diffusion and reached swelling equilibrium quickly. Poly(vinyl alcohol) (PVA) hydrogels with tissue-like viscoelasticity, excellent biocompatibility, and hydrophilicity have been considered as promising cartilage replacement materials. However, the low mechanical properties of pure PVA hydrogels limit their applications for bearing complicated loads. Herein, we report silicon dioxide (SiO2)/PVA composite hydrogels fabricated by fabricated cyclically freezing/thawing the aqueous mixture of PVA and methyltrimethoxysilane (MTMS). MTMS hydrolyzes and forms SiO2 particles in situ to reinforce PVA hydrogel. Meanwhile, silanol group condenses with hydroxyl groups of PVA and chemically bonds with PVA. The resulting SiO2/PVA hydrogels exhibit much better mechanical properties than bare PVA hydrogel. In addition, the composite hydrogels keep very low swellable property. The structures and properties of poly(viny1 alcohol) (PVA) hydrogel prepared by the freezing and thawing method and poly(ethy1ene glycol) and polyacrylamide hydrogels prepared by polymerization were compared in view of immobilizing microorganisms. Observation in a scanning electron microscope and measurement of the physicochemical properties revealed that the frozen PVA gel is superior in water content and oxygen permeability than the other gels. The PVA hydrogel has a high tensile and folding strength and good handleability and processability. The durability against abrasion and chemical resistance of PVA gel were also studied. Activated sludge was immobilized and fixed on PVA gel particles and incubated. This gel had a treating capacity for synthetic sewage of about two to three times that with the standard activated sludge process. © 1995 John Wiley & Sons, Inc.  相似文献

To reach sustained drug release, a new composite drug‐delivery system consisting of poly(d,l ‐lactide‐co‐glycolide) (PLGA) nanoparticles (NPs) embedded in thermosensitive poly(N‐isopropyl acrylamide) (PNIPAAm) hydrogels was developed. The PNIPAAm hydrogels were synthesized by free‐radical polymerization and were crosslinked with poly(ethylene glycol) diacrylate, and the PLGA NPs were prepared by a water‐in‐oil‐in‐water double‐emulsion solvent‐evaporation method. The release behavior of the composite hydrogels loaded with albumin–fluorescein isothiocyanate conjugate was studied and compared with that of the drug‐loaded neat hydrogel and PLGA NPs. The results indicate that we could best control the release rate of the drug by loading it to the PLGA NPs and then embedding the whole system in the PNIPAAm hydrogels. The developed composite hydrogel systems showed near zero‐order drug‐release kinetics along with a reduction or omission of initial burst release. The differential scanning calorimetry results reveal that the lower critical solution temperature of the developed composite systems remained almost unchanged (<1°C increase only). Such a characteristic indicated that the thermosensitivity of the PNIPAAm hydrogel was not distinctively affected by the addition of PLGA NPs. In conclusion, an approach was demonstrated for the successful preparation of a new hybrid hydrogel system having improved drug‐release behavior with retained thermosensitivity. An interpenetrating polymer network (IPN) hydrogel composed of poly(vinyl alcohol) (PVA) and chitosan exhibited electric‐sensitive behavior. The PVA/chitosan IPN hydrogel was synthesized by an ultraviolet (UV) irradiation method that is used in several biomedical and industrial fields. The swelling behavior of the PVA/chitosan IPN hydrogel was studied by immersion of the gel in NaCl aqueous solutions at various concentrations. The swelling ratio decreased with increasing concentration of NaCl solution. The stimuli response of the IPN hydrogel in electric fields was also investigated. When a swollen PVA/chitosan IPN was placed between a pair of electrodes, the IPN exhibited bending behavior in response to the applied electric field. The bending angle and the bending speed of the PVA/chitosan IPN increased with increasing applied voltage and concentration of NaCl aqueous solution. The PVA/chitosan IPN also showed stepwise bending behavior depending on the electric stimulus. Temperature‐sensitive interpenetrating polymer network (IPN) hydrogels based on soy protein and poly(N‐isopropylacrylamide) were successfully prepared. The structure and properties were systematically characterized using Fourier transform infrared spectroscopy, scanning electron microscopy, differential scanning calorimetry and thermogravimetric analysis, and the swelling and deswelling behavior was also investigated. It was found that the hydrogels had good miscibility, thermal stability and temperature sensitivity, and the lower critical solution temperature was ca 32 °C. Changing the content of soy protein or crosslinker could be used to control the swelling behavior, water retention and network structure of the IPN hydrogels.
