Well‐defined poly(l ‐lactide‐b‐ethylene brassylate‐b‐l ‐lactide) (PLLA‐b‐PEB‐b‐PLLA) triblock copolymer was synthesized by using double hydroxyl‐terminated PEBs with different molecular weights. Gel permeation chromatography and NMR characterization were employed to confirm the structure and composition of the triblock copolymers. DSC, wide‐angle X‐ray diffraction, TGA and polarized optical microscopy were also employed to demonstrate the relationship between the composition and properties. According to the DSC curves, the cold crystallization peak vanished gradually with decrease of the PLLA block, illustrating that the relatively smaller content of PLLA may lead to the formation of a deficient PLLA type crystal, leading to a decrease of melting enthalpy and melting temperature. Multi‐step thermal decompositions were determined by TGA, and the PEB unit exhibited much better thermal stability than the PLLA unit. Polarized optical microscopy images of all the triblock samples showed that spherulites which develop radially and with an extinction pattern in the form of a Maltese cross exhibit no ring bond. The growth rate of the spherulites of all triblock samples was investigated. The crystallization capacity of PLLA improved with incorporation of PLLA, which accords with the DSC and wide‐angle X‐ray diffraction results. © 2019 Society of Chemical Industry  相似文献

The poly(l ‐lactide)‐b‐poly(ethylene glycol)‐b‐poly(l ‐lactide) block copolymers (PLLA‐b‐PEG‐b‐PLLA) were synthesized in a toluene solution by the ring‐opening polymerization of 3,6‐dimethyl‐1,4‐dioxan‐2,5‐dione (LLA) with PEG as a macroinitiator or by transterification from the homopolymers [polylactide and PEG]. Two polymerization conditions were adopted: method A, which used an equimolar catalyst/initiator molar ratio (1–5 wt %), and method B, which used a catalyst content commonly reported in the literature (<0.05 wt %). Method A was more efficient in producing copolymers with a higher yield and monomer conversion, whereas method B resulted in a mixture of the copolymer and homopolymers. The copolymers achieved high molar masses and even presenting similar global compositions, the molar mass distribution and thermal properties depends on the polymerization method. For instance, the suppression of the PEG block crystallization was more noticeable for copolymer A. An experimental design was used to qualify the influence of the catalyst and homopolymer amounts on the transreactions. The catalyst concentration was shown to be the most important factor. The triblock energetic copolymer poly(lactic acid)‐block‐(glycidylazide polymer)‐block‐polystyrene (PLA‐b‐GAP‐b‐PS) was synthesized successfully through atom‐transfer radical polymerization (ATRP) of styrene and ring‐opening polymerization of d,l ‐lactide. The energetic macroinitiator GAP‐Br, which was made from reacting equimolar GAP with α‐bromoisobutyryl bromide, firstly triggered the ATRP of styrene with its bromide group, and then the hydroxyl group on the GAP end of the resulting diblock copolymer participated in the polymerization of lactide in the presence of stannous octoate. The triblock copolymer PLA‐b‐GAP‐b‐PS had a narrow distribution of molecular weight. In the copolymer, the PS block was solvophilic in toluene and improved the stability of the structure, the PLA block was solvophobic in toluene and served as the sacrificial component for the preparation of porous materials, and GAP was the basic and energetic material. The three blocks of the copolymer were fundamentally thermodynamically immiscible, which led to the self‐assembly of the block copolymer in solution. Further studies showed that the concentration and solubility of the copolymer and the polarity of the solvent affected the morphology and size of the micelles generated from the self‐assembly of PLA‐b‐GAP‐b‐PS. The micelles generated in organic solvents at 10 mg mL?1 copolymer concentration were spherical but became irregular when water was used as a co‐solvent. The spherical micelles self‐assembled in toluene had three distinct layers, with the diameter of the micelles increasing from 60 to 250 nm as the concentration of the copolymer increased from 5 to 15 mg L?1. © 2017 Society of Chemical Industry  相似文献

A novel A2BA2‐type thermosensitive four‐armed star block copolymer, poly(N‐isopropyl acrylamide)2b‐poly(lactic acid)‐b‐poly(N‐isopropyl acrylamide)2, was synthesized by atom transfer radical polymerization and characterized by 1H‐NMR, Fourier transform infrared spectroscopy, and size exclusion chromatography. The copolymers can self‐assemble into nanoscale spherical core–shell micelles. Dynamic light scattering, surface tension, and ultraviolet–visible determination revealed that the micelles had hydrodynamic diameters (Dh) below 200 nm, critical micelle concentrations from 50 to 55 mg/L, ζ potentials from ?7 to ?19 mV, and cloud points (CPs) of 34–36°C, depending on the [Monomer]/[Macroinitiator] ratios. The CPs and ζ potential absolute values were slightly decreased in simulated physiological media, whereas Dh increased somewhat. The hydrophobic camptothecin (CPT) was entrapped in polymer micelles to investigate the thermo‐induced drug release. The stability of the CPT‐loaded micelles was evaluated by changes in the CPT contents loaded in the micelles and micellar sizes. A novel approach to self‐assembled and shell‐crosslinked (SCL) micelles from the diblock copolymer poly(L ‐lactide)‐block‐poly(L ‐cysteine) to be used as drug and protein delivery carriers is described. Rifampicin was used as a model drug. The drug‐loaded SCL micelles were obtained by self‐assembly of the copolymer in the presence of the drug in aqueous media. Their morphology and size were studied with dynamic light scattering and field emission scanning electron microscopy. The rifampicin loading capacity and encapsulation efficiency were studied with ultraviolet–visible spectrophotometry. The drug‐release rate in vitro depended on the oxidizing and reducing environment. Moreover, a straightforward approach to the conjugation of the copolymer with bovine serum albumin (BSA) was developed, and a gel electrophoresis test demonstrated that this conjugated BSA could be reversibly released from the copolymer substrate under reducing conditions. Surfactant‐free nanoparticles of poly(DL ‐lactide‐co‐glycolide) (PLGA) nanoparticles were prepared with or without poly(L ‐lactide)‐poly(ethylene oxide) (LE) diblock copolymer (abbreviated as PLGA/LE and PLGA nanoparticles) by dialysis method. LE diblock copolymer was used to make PLGA nanoparticles to alternate conventional surfactant. The size of PLGA and PLGA/LE nanoparticles was 295.3 ± 171.3 and 307.6 ± 27.2 nm, respectively, suggesting LE diblock copolymer might be coated onto the surface of nanoparticles. Observation of scanning electron microscope (SEM) showed that PLGA/LE nanoparticles have spherical shapes ranging ~ 200–500 nm. In 1H‐NMR study, characteristic peaks of the methyl protons of PLGA disappeared in D2O, whereas characteristic peaks of the methyl proton of both PEG and PLGA were shown in both CDCl3 and D2O, indicating that LE diblock copolymer coated on the surface of the PLGA nanoparticles. The higher the initial content of drug, the higher the drug contents and the lower the loading efficiency. PLGA/LE nanoparticles at higher drug contents resulted in slower adriamycin·HCl (ADR) release rate than that of lower drug contents. Also, slower release rate of ADR was achieved by entrapped into the PLGA/LE nanoparticles, whereas LE polymeric micelles showed rapid ADR release. © 2003 Wiley Periodicals, Inc. BACKGROUND: Biodegradable block copolymers have attracted particular attention in both fundamental and applied research because of their unique chain architecture, biodegradability and biocompatibility. Hence, biodegradable poly[((R )‐3 ‐hydroxybutyrate)‐block‐(D ,L ‐lactide)‐block‐(ε‐caprolactone)] (PHB‐PLA‐PCL) triblock copolymers were synthesized, characterized and evaluated for their biocompatibility. RESULTS: The results from nuclear magnetic resonance spectroscopy, gel permeation chromatography and thermogravimetric analysis showed that the novel triblock copolymers were successfully synthesized. Differential scanning calorimetry and wide‐angle X‐ray diffraction showed that the crystallinity of PHB in the copolymers decreased compared with methyl‐PHB (LMPHB) oligomer precursor. Blood compatibility experiments showed that the blood coagulation time became longer accompanied by a reduced number of platelets adhering to films of the copolymers with decreasing PHB content in the triblocks. Murine osteoblast MC3T3‐E1 cells cultured on the triblock copolymer films spread and proliferated significantly better compared with their growth on homopolymers of PHB, PLA and PCL, respectively. CONCLUSION: For the first time, PHB‐PLA‐PCL triblock copolymers were synthesized using low molecular weight LMPHB oligomer as the macroinitiator through ring‐opening polymerization with D ,L ‐lactide and ε‐caprolactone. The triblock copolymers exhibited flexible properties with good biocompatibility; they could be developed into promising biomedical materials for in vivo applications. BACKGROUND: The aim of the work presented was to synthesize a series of amphiphilic hyperbranched poly[(amine‐ester)‐co‐(D ,L ‐lactide)] (HPAE‐co‐PLA) copolymers and study the formation of copolymeric micelles. These copolymeric micelle systems are expected to be potential candidates for applications in protein drug delivery. RESULTS: The chemical structures of the copolymers were confirmed by Fourier transform infrared spectroscopy, 13C NMR and thermogravimetric analysis. Fluorescence spectroscopy and dynamic light scattering confirmed the formation of copolymeric micelles of the HPAE‐co‐PLA copolymers. The maintenance of stability of bovine serum albumin (BSA) during release from micelles in vitro was also measured using circular dichroism and fluorescence spectrometry. CONCLUSION: Novel hyperbranched HPAE‐co‐PLA copolymers have been synthesized. Conjugation of PLA to HPAE was proved to be an available method for the preparation of micelles for protein delivery. The BSA‐loaded micelles showed enhanced encapsulation efficiency and the structural stability of BSA was retained during the release process. Thermoresponsive poly(N‐isopropyl acrylamide) (PNIPAM)‐block‐hydroxy‐terminated polybutadine‐block‐PNIPAM triblock copolymers were synthesized by atom transfer radical polymerization; this was followed by the in situ epoxidation reaction of peracetic acid. The copolymers were characterized by 1H‐NMR, Fourier transform infrared spectroscopy, and size exclusion chromatography measurements, and their physicochemical properties in aqueous solution were investigated by surface tension measurement, fluorescent spectrometry, ultraviolet–visible transmittance, transmission electron microscopy observations, dynamic light scattering, and so on. The experimental results indicate that the epoxidized copolymer micelle aggregates retained a spherical core–shell micelle structure similar to the control sample. However, they possessed a decreased critical aggregate concentration (CAC), increased hydrodynamic diameters, and a high aggregation number and cloud point because of the incorporation of epoxy groups and so on. In particular, the epoxidized copolymer micelles assumed an improved loading capacity and entrapment efficiency of the drug, a preferable drug‐release profiles without an initial burst release, and a low cytotoxicity. Random copolyester namely, poly(ethylene terephthalate‐co‐sebacate) (PETS), with relatively lower molecular weight was first synthesized, and then it was used as a macromonomer to initiate ring‐opening polymerization of l ‐lactide. 1H NMR quantified composition and structure of triblock copolyesters [poly(l ‐lactic acid)‐b‐poly(ethylene terephthalate‐co‐sebacate)‐b‐poly(l ‐lactic acid)] (PLLA‐PETS‐PLLA). Molecular weights of copolyesters were also estimated from NMR spectra, and confirmed by GPC. Copolyesters exhibited different solubilities according to the actual content of PLLA units in the main chain. Copolymerization effected melting behaviors significantly because of the incorporation of PETS and PLLA blocks. Crystalline morphology showed a special pattern for specimen with certain composition. It was obvious that copolyesters with more content of aromatic units of PET exhibited increased values in both of stress and modulus in tensile test. © 2007 Wiley Periodicals, Inc. Biodegradable poly[(2‐methacryloyloxyethyl phosphorylcholine)‐block‐(D ,L ‐lactide)] (PMPC‐b‐PLA) diblock copolymers with various hydrophilic PMPC weight fractions (fPC) will spontaneously self‐assemble into well‐defined vesicles and large compound micelles (LCMs) in water. Transmission electron microscopy, scanning electron microscopy, dynamic light scattering and fluorescence microscopy were used to observe their aggregate morphologies. The degradation of the LCMs was investigated and the loss of molecular weight of PLA blocks was confirmed using 1H NMR analysis. The hydrolysis of PLA increases fPC and consequently shifts the preferred morphology from LCMs to vesicles. Such degradation‐induced morphological transitions mean that the biocompatible and biodegradable LCMs have great application potential in drug delivery. To reach sustained drug release, a new composite drug‐delivery system consisting of poly(d,l ‐lactide‐co‐glycolide) (PLGA) nanoparticles (NPs) embedded in thermosensitive poly(N‐isopropyl acrylamide) (PNIPAAm) hydrogels was developed. The PNIPAAm hydrogels were synthesized by free‐radical polymerization and were crosslinked with poly(ethylene glycol) diacrylate, and the PLGA NPs were prepared by a water‐in‐oil‐in‐water double‐emulsion solvent‐evaporation method. The release behavior of the composite hydrogels loaded with albumin–fluorescein isothiocyanate conjugate was studied and compared with that of the drug‐loaded neat hydrogel and PLGA NPs. The results indicate that we could best control the release rate of the drug by loading it to the PLGA NPs and then embedding the whole system in the PNIPAAm hydrogels. The developed composite hydrogel systems showed near zero‐order drug‐release kinetics along with a reduction or omission of initial burst release. The differential scanning calorimetry results reveal that the lower critical solution temperature of the developed composite systems remained almost unchanged (<1°C increase only). Such a characteristic indicated that the thermosensitivity of the PNIPAAm hydrogel was not distinctively affected by the addition of PLGA NPs. In conclusion, an approach was demonstrated for the successful preparation of a new hybrid hydrogel system having improved drug‐release behavior with retained thermosensitivity. Thermoresponsive amphiphilic copolymer, poly[N‐isopropyl acrylamide‐co‐3‐(trimethoxysilyl)propylmethacrylate]‐b‐poly{N‐[3‐(dimethylamino)propyl]methacrylamide} with a branched structure was designed and synthesized by consecutive reversible addition–fragmentation chain‐transfer polymerization. The further hydrolysis of trimethoxysilyl functions in 3‐(trimethoxysilyl) propyl methacrylate units led to the fabrication of core‐crosslinked (CCL) micelles with silica crosslinks at temperatures above the lower critical solution temperature of the poly(N‐isopropyl acrylamide) block. The thermally induced structural and morphological changes of the CCL micelles in aqueous solution were investigated by transmission electron microscopy and 1H‐NMR analyses. The resulting CCL micelles were further explored as nanocarriers for the codelivery of an anticancer drug and nucleic acid for enhanced therapeutic efficacy. The CCL micelles effectively condensed the nucleic acid and mediated higher gene transfer in the presence of serum than in serum‐free transduction. A cytotoxicity study revealed that whereas the pure CCL micelles exhibited unapparent cytotoxicity, the codelivery of p53 and doxorubicin with the CCL micelle formulation resulted in better treatment efficiency than sole chemotherapy. © 2014 Wiley Periodicals, Inc. Polystyrene terminated with benzyl alcohol units was employed as a macroinitiator for ring‐opening polymerization of ε‐caprolactone and L ‐lactide to yield AB‐ and ABC‐type block copolymers. Even though there are many reports on the diblock copolymers of poly(styrene‐block‐lactide) and poly(styrene‐block‐lactone), this is the first report on the poly(styrene‐block‐lactone‐block‐lactide) triblock copolymer consisting of two semicrystalline and degradable segments. The triblock copolymers exhibited twin melting behavior in differential scanning calorimetry (DSC) analysis with thermal transitions corresponding to each of the lactone and lactide blocks. The block derived from ε‐caprolactone also showed crystallization transitions upon cooling from the melt. In the DSC analysis, one of the triblock copolymers showed an exothermic transition well above the melting temperature upon cooling. Thermogravimetric analysis of these block copolymers showed a two‐step degradation curve for the diblock copolymer and a three‐step degradation for the triblock copolymer with each of the degradation steps associated with each segment of the block copolymers. The present study shows that it is possible to make pure triblock copolymers with two semicrystalline segments which also consist of degradable blocks. This article investigates the effects of electron beam (EB) radiation on poly(D ,L ‐lactic acid)‐b‐poly (ethylene glycol) copolymer (PLA‐b‐PEG‐b‐PLA). The copolymer films were EB irradiated at doses from 0 to 100 kGy. The degradation of these films was studied by measuring the changes in their molecular weight, mechanical and thermal properties. The dominant effect of EB radiation on PLA‐b‐PEG‐b‐PLA is chain‐scission. With increasing irradiation dose, recombination reactions or partial crosslinking may occur in addition to chain scission. The degree of chain scission Gs and crosslinking Gx of sample are calculated to be 0.213 and 0.043, respectively. A linear relationship is also established between the decreases in molecular weight with increasing irradiation dose. Elongation at break of the irradiated sample decreases significantly, whereas its tensile strength decreases slightly. The glass transition temperature (Tg) is basically invariant as a function of irradiation dose. A highly branched hybrid copolymer based on polyhedral oligomeric silsesquioxane (POSS) was designed to improve the brittleness of poly(d,l‐lactide) (PDLLA). The toughening material was synthesized using POSS‐OH as the core, which initiated the ring‐opening polymerization of ε‐caprolactone and d,l‐lactide sequentially to form the highly branched POSS‐g‐poly (ε‐caprolactone)‐b‐poly(d,l‐lactide) (POSS‐g‐PCL‐b‐PLA) copolymer with eight PCL‐b‐PLA arms. The POSS‐g‐PCL‐b‐PLA copolymer had a very good dispersion in the PDLLA matrix with the size of microdomains smaller than 1 µm when added at a low content below 10 wt %. In related to the nano‐scale size of microdomains in the blends, the crystallinity of PCL blocks was significantly suppressed. Thus, the addition of POSS‐g‐PCL‐b‐PLA is very effective to improve the roughness of the matrix polymer when added at a low content. © 2014 Wiley Periodicals, Inc. J. Appl. Polym. Supramolecular poly(?‐capolactone)/poly(lactide) alternating multiblock copolymers were prepared by UPy‐functionalized poly(lactide)‐b‐ poly(?‐capolactone)‐b‐ poly(lactide) copolymers. The prepared supramolecular polymers (SMPs) exhibit the characteristic properties of thermoplastic elastomers. The stereo multiblock SMPs (sc‐SMPs) were formed by blending UPy‐functionalized poly(l ‐lactide)‐b‐ PCL‐b‐ poly(l ‐lactide) (l ‐SMPs) and UPy‐functionalized poly(d ‐lactide)‐b‐ PCL‐b‐ poly(d ‐lactide) (d ‐SMPs) due to stereocomplexation of the PLLA and PDLA blocks. Sc‐SMPs with low content of d ‐SMPs (≤20%) are transparent, elastic solids, while those having high d ‐SMPs content are opaque, brittle solids. The effects of l ‐SMPs/d ‐SMPs mixing ratios on thermal, crystallization behaviors, crystal structure, mechanical and hydrophilic properties of sc‐SMPs were deeply investigated. The incorporation of UPy groups depresses the crystallization of polymer, and the stereocomplex formation accelerates the crystallization rate. The used initiator functionalized polyhedral oligomeric silsesquioxanes causes a different effect on the crystallization of PLA and PCL blocks. The tensile strength and elongation at break of l d /d d ‐SMPs (d represents the initiator diethylene glycol) are significantly larger than that of l p /d p ‐SMPs (p represents the initiator polyhedral oligomeric silsesquioxanes), and their heat resistance and hydrophilicity can be also modulated by the l ‐SMPs/d ‐SMPs mixing ratios and the different initiators. © 2017 Wiley Periodicals, Inc. Modification of poly(l ‐lactide) (PLLA) via blending with two or more polymers is one of the effective approaches used to overcome the brittleness of PLLA, which often requires the addition of a compatibilizer and generate opaque materials. To solve this problem, multiarmed polycaprolactone‐block‐PLLA (PCL‐b‐PLLA) was synthesized by consecutive ring‐opening polymerization of ε‐caprolactone and l ‐lactide using multihydroxyl alcohols as initiator. The structure and composition was confirmed by proton nuclear magnetic resonance and gel permeation chromatography. PLLA/multiarmed PCL‐b‐PLLA blends with various blend ratios were prepared via melt mixing. The presence of multiarmed PCL‐b‐PLLA (30%) in the PLLA matrix exhibited more than 80 times improvement in the elongation at break, as compared to unmodified PLLA. The addition of multiarmed PCL‐b‐PLLA in the PLLA contributed to the enhancement of the storage modulus in the low frequency, which was related to the entanglement of the PLLA and multiarmed PCL‐b‐PLLA. The blend interface had no obvious phase separation, and showed good adhesion between dispersed block copolymer phases within the continuous PLLA phase. The compatibilization mechanism and toughing mechanism were proposed. The resulting PLLA blends also showed good transparency. A series of triblock co‐polymers, consisting of a poly(ethylene glycol) (PEG) central block joined to two blocks of random p‐dioxanone‐co‐L ‐lactide copolymers were synthesized by ring‐opening polymerization of p‐dioxanone (PDO) and L ‐lactide (LLA) initiated by PEG in the presence of stannous 2‐ethylhexanoate catalyst. The resulting copolymers were characterized by various techniques including 1H and 13C NMR and FTIR spectroscopies, gel permeation chromatography, inherent viscosity, wide‐angle X‐ray diffractometry (WAXD) and differential scanning calorimetry (DSC). The conversion of PDO and L ‐lactide into the polymer was studied various mole ratios and at different polymerization temperature from 1H NMR spectra. Results of WAXD and DSC showed that the crystallinity of PEG macroinitiator was greatly influenced by the composition of PDO and L ‐lactide in the copolymer. The poly(2‐methacryloyloxyethyl phosphorylcholine)‐block‐poly(D ,L ‐lactide) (PMPC‐b‐PLA) was specially designed to develop biomimetic giant vesicles (GVs) and giant large compound vesicles via a simple spontaneous assemble in aqueous solution. The weight fraction of the hydrophilic PMPC block (fPC) was proved to play an important role in the size and morphology control of the self‐assembled aggregates.
