采用酒石酸铁法测定金丝小枣枣皮、枣核中的多酚类物质含量。首次利用化学发光分析和紫外-可见分光光度分析相结合的方法测定金丝小枣枣皮、枣核中间苯三酚和邻苯二酚的体外抗氧化活性及其清除自由基保护DNA等能力的变化规律。研究结果表明,间苯三酚和邻苯二酚对·OH、NO2-、DPPH具有较明显的抑制作用,对由·OH引起的DNA损伤也具有较好的保护作用。当间苯三酚和邻苯二酚质量浓度为60μg/mL时,体外抗氧化活性及清除自由基保护DNA作用最佳。本研究可为开发枣副产品的保健功能提供新的研究方向和理论基础。 相似文献

采用多个化学发光体系系统地研究了花生多肽的体外抗氧化活性及其对DNA损伤的保护作用。运用邻苯三酚-鲁米诺化学发光体系测定了花生多肽对超氧阴离子的清除作用,硫酸铜-邻菲哕啉-抗坏血酸-双氧水、硫酸亚铁-鲁米诺-双氧水和硫酸亚铁-鲁米诺3个体系测定了花生多肽对羟基自由基的清除作用,双氧水-鲁米诺体系测定了花生多肽对体外双氧水的清除作用,采用硫酸铜-邻菲哆啉-抗坏血酸-双氧水-脱氧核糖核酸测定了花生多肽对体外DNA损伤的保护作用。试验结果表明花生多肽具有较好的体外清除活性氧和保护DNA损伤的活性。花生多肽可作为开发抗氧化产品的新资源。 相似文献

采用化学发光法研究了樟树果实红色素(RPCF)的抗氧化活性及其对DNA损伤的保护作用。运用Pyrogallol-Luminol化学发光体系测定了RPCF对超氧阴离子的清除作用,CuSO4-Phen-VC-H2O2体系测定了RPCF对羟基自由基的清除作用,H2O2-Luminol体系测定了RPCF对双氧水的清除作用,CuSO4-Phen-VC-H2O2-DNA测定了RPCF对体外DNA损伤的保护作用。试验结果表明RPCF有较好的清除活性氧和保护DNA损伤的活性,其清除.O2-、.OH、H2O2能力及对DNA损伤的保护作用的半抑制浓度(IC50)分别为0.207,0.383,0.227和0.401 mg/mL。 相似文献

为研究商洛绿茶多糖体外抗氧化和抗肿瘤活性,采用DEAE-52纤维素柱色谱法对水提醇沉法制备的茶多糖进行纯化,继而在体外抗氧化体系上测定其对DPPH自由基、ABTS自由基、羟基自由基、超氧阴离子的清除能力,同时检测其对乳腺癌MCF-7细胞的生长抑制作用。结果表明,经DEAE-52纤维素柱层析纯化后得到SGTPs-1,SGTPs-2,SGTPs-3三个组分,多糖含量分别为83.5%,76.8%,89.7%,这3个组分对不同的自由基清除能力均呈现良好的量效关系,其中SGTPs-3的作用最强,其次为SGTPs-1,SGTPs-2最弱;SGTPs三个组分对MCF-7细胞均有一定的生长抑制作用,并且抑制率与浓度呈正相关。商洛绿茶多糖具有较好的体外抗氧化和抗肿瘤活性。 相似文献

分别采用邻苯三酚- 鲁米诺化学发光体系、硫酸亚铁- 鲁米诺- 过氧化氢化学发光体系和过氧化氢- 鲁米诺化学发光体系测定白腊、妃子笑、糯米糍和怀枝4 个品种的荔枝皮原花青素(procyanidins of litchi pericarp,LPPC)体外清除超氧阴离子自由基、羟自由基和过氧化氢的能力。同时运用硫酸铜- 邻菲啰啉- 抗坏血酸- 过氧化氢-DNA体系测定不同LPPC 对诱导体系DNA 损伤的保护作用。实验结果表明:白腊LPPC 具有较好的体外清除超氧阴离子自由基和过氧化氢的能力;妃子笑呈现出较显著的羟自由基清除能力和保护DNA 损伤的效果。不同品种的LPPC在不同化学发光体系中的抗氧化活性呈现较大差异。 相似文献

将米曲霉接种于鱼鳔匀浆液中对其进行直接发酵,利用菌产蛋白酶的作用水解鱼鳔蛋白。随着发酵时间的延长,发酵液水解度不断上升,发酵液中的多肽含量不断增加。对发酵所得鱼鳔多肽进行超滤,采用产O2·和·OH 化学模型研究其清除自由基的活性,采用亚油酸自氧化体系研究其抗氧化活性。结果显示,经发酵40h,水解度(22 ± 0.8)% 所得鱼鳔多肽体外抗氧化能力较强;超滤片段中,平均肽链长度为12 个氨基酸残基的多肽超滤液体外抗氧化能力较强;分子量分布在1000D 左右。发酵法所得鱼鳔多肽不仅对自由基具有强烈的清除作用,还能明显抑制脂质的氧化。 相似文献

虾青素是一种氧化型类胡萝卜素,主要来自微生物和海洋生物,因其特殊的化学结构而具有较强的抗氧化活性,从而被广泛应用于保健食品、医疗卫生等领域。本文从虾青素清除自由基能力、限制氧化剂渗透进细胞内、增加抗氧化酶活性、降低DNA的氧化损伤、抑制脂质过氧化5个方面阐述了虾青素的抗氧化机制,从体外清除自由基活性、油脂体系中抑制脂质氧化降解以及虾青素抑制促氧化酶、增强抗氧化酶活性3个方面阐述了虾青素的抗氧化能力,且综述了虾青素与其他天然抗氧化剂如番茄红素、叶黄素、β-胡萝卜素、维生素C和维生素E的抗氧化能力对比进展,为虾青素抗氧化活性的进一步研究提供参考。 相似文献

以牛蒡根为材料,采用常规方法测定牛蒡根总黄酮的DPPH自由基清除能力、羟基自由基清除能力、总抗氧化能力、还原力、金属螯合能力、抗脂质过氧化能力和对DNA氧化损失的保护作用,同时测定了牛蒡根总黄酮的含量。结果表明,牛蒡根总黄酮的自由基清除能力、总抗氧化能力、金属螯合能力和还原力随浓度的增加而增强,且呈剂量依赖性相关,DPPH自由基清除能力的半数抑制浓度(IC50)为15.7mg/mL;对大鼠肝脂质过氧化产生的丙二醛(MDA)有较好的清除效果,IC50为34μg/mL;对K562细胞DNA氧化损伤具有保护作用。由此说明,牛蒡根为一种很好的天然抗氧化剂。 相似文献

The aim of this study was to develop a combined method for measuring the total antioxidant activity, the reductive and the radical scavenging activity. Linoleic acid was used as the substrate for an iron-initiated lipid peroxidation to measure the total antioxidant activity. In addition, methyl esters of linoleic acid hydroperoxides were used as substrates to measure the reductive antioxidant activity. The radical scavenging antioxidant activity was calculated by subtracting the reductive antioxidative activity from the total antioxidative activity. As representative examples, the antioxidants alpha-tocopherol, ascorbic acid, trans-resveratrol and L-glutathione as well as commonly used food additives such as 2(3)-tert-butyl-4-hydroxyanisole (BHA) and 2,6-bis(1,1-dimethylethyl)-4-methylphenol (BHT) were analyzed. The results for the novel antioxidation test showed that alpha-tocopherol, BHA and BHT are primarily acting as radical scavengers, whereas ascorbic acid and L-glutathione show a strong reductive capacity. As linoleic acid as well as its hydroperoxides both are present in foods and in the organism, the test presented here can be considered representative of radical reactions occurring in food matrixes and in vivo. 选用新鲜干酪乳清为原料,研究碱性蛋白酶对乳清水解产物的抗氧化活性。以水解产物的亚铁还原能力、对卵磷脂脂质氧化体系的过氧化抑制作用、羟自由基清除能力和超氧阴离子自由基的清除能力为指标评价乳清水解产物的抗氧化能力。结果表明,乳清在水解前经过预热处理并不能增加其水解产物的抗氧化活性,1 h水解物的亚铁还原能力最高,2 h水解产物对卵磷脂脂质氧化体系的过氧化抑制作用最高,抑制率达到24.82%;2 h水解产物羟自由基清除率最高,达到70.28%;2 h水解产物超氧阴离子自由基清除率最高,达到21.4%。但是乳清水解产物的抗氧化能力与水解度没有线性关系。 相似文献

以新鲜猪皮为原料,基于凝胶特性以羟脯氨酸含量为指标,对影响胃蛋白酶酶解过程的各个因素进行研究,通过Box-Behnken设计优化酶解工艺;以清除O2-•、•OH、H2O2能力和DNA损伤保护作用为指标研究酶解产物的抗氧化功能。结果表明:最佳酶解条件为加酶量0.25%、料液比1∶2(g/mL)、酶解时间1.65 h、酶解温度41.15 ℃,在此酶解条件下其产物清除O2-•的IC50值为20.19 mg/mL;清除•OH的IC50值为6.36 mg/mL;清除H2O2的IC50值为0.65 mg/mL;对DNA损伤保护作用的IC50值为1.86 mg/mL。 相似文献

In India, many flowers are consumed as food. During the present study, 37 flowers have been analyzed for their 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity and total antioxidant capacity. The flowers were found to have different levels of antioxidant properties in the systems tested. On the basis of the activity and availability, a few flowers were further studied for hydroxyl radical scavenging activity, superoxide radical scavenging activity, lipid peroxidation prevention, and DNA damage prevention properties. The correlations suggest that the total phenol contents of the flowers were responsible for the antioxidant properties of the aqueous extracts. The unweighted-pair group average dendrogram showing interrelationships between the investigated species grouped them into two high level clusters based on activity. 相似文献

Yellow wine (YW) has been claimed to have beneficial effects on the prevention of cancer and cardiovascular disease, which are related to the oxidation activity in vivo. In this study, the antioxidant activity of YW and its concentrate and volatile compounds (VC) was determined in terms of total antioxidant, reducing capacity, free radical scavenging, metal chelating and superoxide anion radical scavenging activities. These various antioxidant activities were compared with synthetic antioxidant, such as butylated hydroxyanisole (BHA) and butylated hydroxytoluene (BHT). The total phenolic compounds were also determined. Results indicated that YW and its concentrate exhibited similar strong total antioxidant activity. The inhibition percentage of 100 μl of YW and its concentrate on peroxidation of 2.5 ml of linoleic acid emulsion was 97.99% and 95.74%, and both of them also had effective reducing capacity, 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging and superoxide anion radical scavenging activities, but had very low metal chelating activity on ferrous. However, the VC in YW had little antioxidant activities. Results indicated that active antioxidants were nonvolatile and resided in the concentrate after evaporation. Phenolic compounds may play a very important role in the antioxidant function. However, identification of specific antioxidant components should be further investigated. 相似文献
