In dairy cows, an increase in plasma concentration of glucose-dependent insulinotropic polypeptide (GIP) is associated with an increase in metabolizable energy intake, but the role of GIP in energy partitioning of dairy cattle is not certain. The objective of this study was to examine the relationship between plasma GIP concentrations and energy partitioning toward milk production. Four mid-lactation, primiparous, rumen-fistulated Holstein-Friesian cows were fed a control diet of 55% forage and 45% concentrate [dry matter (DM) basis] in a 4 × 4 Latin square design with 4-wk periods. The 4 treatments were (1) control diet fed at 1000 and 1600 h, and (2) once-daily (1000 h) feeding, (3) twice-daily (1000 and 1600 h) feeding, and (4) 4 times/d (1000, 1600, 2200 and 0400 h) feeding of the control diet plus 1 dose (1.75 kg on a DM basis at 0955 h) into the rumen of supplemental vegetable proteins (Amino Green; SCA NuTec Ltd., Thirsk, UK). Measurements of respiratory exchange and energy balance were obtained over 4 d during the last week of each period while cows were housed in open-circuit respiration chambers. Blood was collected from the jugular vein every 30 min for 12 h, using indwelling catheters, starting at 0800 h on d 20 of each period. Plasma GIP concentration was measured in samples pooled over each 5 consecutive blood samplings. The relationships between plasma GIP, DM intake, heat production, respiratory quotient (RQ), milk yield, and milk energy output were analyzed using linear correlation procedures, with metabolizable intake as a partial variant. Plasma GIP concentration was not correlated with heat production, or milk yield, but was positively correlated with milk energy yield (correlation coefficient = 0.67) and negatively correlated with RQ (correlation coefficient = −0.72). The correlations between GIP with RQ and milk energy output do not imply causality, but support a role for GIP in the regulation of energy metabolism in dairy cows. 相似文献

Activity of stearoyl-CoA desaturase (SCD) in the mammary gland is important for determining the relative proportions of saturated and monounsaturated fatty acids in milk and the concentration of the conjugated linoleic acid isomer rumenic acid (RA; cis-9,trans-11 18:2). Previous studies identified a large degree of between-cow variation in SCD activity, which was consistent across diets and suggests a genetic influence. The objectives of this study were to quantify genetic and phenotypic variations in fatty acid concentrations and SCD indices in milk fat and to estimate their heritabilities in a population of United Kingdom dairy cows. Milk samples were collected from 2,408 daughters of 597 Holstein-Friesian sires on 325 commercial farms for determination of fatty acid profiles. Indices of SCD activity were calculated by expressing each SCD product (cis-9 14:1, cis-9 16:1, cis-9 18:1, and RA) as a proportion of the precursor plus product [e.g., SCDI14 = cis-9 14:1/(14:0 + cis-9 14:1)]. For individual fatty acids, phenotypic variance was considerably greater than additive genetic variance, resulting in small and nonsignificant heritability estimates (± standard error) for all except 6:0 (h2 = 0.27 ± 0.10), 8:0 (h2 = 0.27 ± 0.09), 12:0 (h2 = 0.13 ± 0.07), cis-9 14:1 (h2 = 0.28 ± 0.10), and cis-9 18:1 (h2 = 0.12 ± 0.07). Heritability estimates of desaturase indices were significant for SCDI14 (h2 = 0.38 ± 0.11), SCDI18 (h2 = 0.19 ± 0.09), and SCDIRA (h2 = 0.21 ± 0.09), but not for SCDI16 (h2 = 0.05 ± 0.06). This study provides evidence that additive effects are responsible for a significant proportion of the phenotypic variation in SCD activity in dairy cows. It is concluded that because heritability of desaturase indices is moderate and significant in many cases, these indices could be investigated further for use in future breeding programs to increase concentrations of monounsaturated fatty acids and RA while decreasing concentrations of saturated fatty acids in milk fat. 相似文献

Although taking body temperature by rectal thermometer is the method most commonly used to identify sick cows in the postpartum period, no data on the repeatability of this measure are available. The overall objective of this study was to evaluate variability of rectal temperatures in dairy cows considering different factors (intra- and interinvestigator repeatability, different thermometers, penetration depth into the rectum, and defecation). High coefficients of correlation (r = 0.98) and small differences between values of rectal temperatures (observer A = 39.3 ± 1.0°C and observer B = 39.4 ± 1.0°C) provide evidence that rectal temperature was a repeatable measure in dairy cows. Testing was carried out using 4 different digital thermometers: GLA M750 (GLA Agricultural Electronics, San Luis Obispo, CA), MTI8101 (SES Scala Electronics, Stahnsdorf, Germany), MT1831 (Microlife AG, Widnau, Switzerland) and Domotherm TH1 (Uebe Medical GmbH, Wertheim, Germany). Thermometers were inserted into the rectum to a certain depth (GLA M750 and MTI8101 = 11.5 cm; MT1831 = 8.4 cm; Domotherm TH1 = 7.7 cm) and a measure was finished when a visual or acoustic signal was emitted by the thermometer. The measures could be influenced by the procedure itself (up to 0.5°C), type of thermometer (up to 0.3°C), and the penetration depth (11.5 cm or 6.0 cm in one of the experiments) into the rectum (up to 0.4°C difference between a penetration depth of 11.5 cm and 6.0 cm in one of the experiments). Differences in rectal temperature before and after defecation were minor (<0.1°C). These results indicate that some care is required in generalizing rectal measures of body temperature. 相似文献

The present study investigated the flooring preference during the 30 h before parturition in Holstein dairy cows housed individually in a maternity pen. Seventeen multiparous cows were moved, on average, 2 d before expected calving date into an individual maternity pen with 3 different flooring surfaces: 10 cm of sand, pebble-top rubber mats, or concrete flooring, each covered with 15 cm of straw. Calving location, lying time, and total time and number of lying bouts on each of the floor types were recorded during 2 periods: precalving (24 to 29 h before calving) and at calving (0 to 5 h before calving). Ten cows calved on sand, 6 on concrete, and 1 on the rubber mat. Lying bouts increased during the hours closest to calving, regardless of flooring. The number of lying bouts did not differ between flooring types precalving but cows had more lying bouts on sand and concrete compared with rubber at calving. Cows spent more time lying down on sand and concrete compared with rubber precalving, but lying times did not differ between treatments at calving. Cows that calved on sand spent more time lying on sand at calving compared with the other 2 flooring types. Cows that calved on concrete did not show a flooring preference at calving. These results indicate that rubber mats are the least preferred by dairy cows in the maternity pens, even when covered with a deep layer of straw. 相似文献

Heart rate variability (HRV), as a physiological measure of animal welfare, was investigated in 36 cows milked in a parallel milking parlor with nonvoluntary exit. Heart rate variability parameters measured during the morning resting (baseline period) were compared with those measured during different stages of the entire milking process. No differences were found in HRV parameters between the baseline period, preparation, and main milking. A considerable reduction in vagal activity was detected during the movement of the cows to the milking parlor (driving) and while cows were in the holding area. The parasympathetic measures of HRV decreased whereas the sympatho-vagal balance increased compared with baseline. The same pattern was observed regarding the stage between removing the teat cups and leaving the milking parlor (waiting). No differences in any sympathetic measures were observed between the baseline period and any of the milking stages. These findings indicate that the milking process itself (preparation and main milking) is not stressful for cows. Decreased parasympathetic activity during driving might be the result of the physical activity of the cows, whereas waiting in the holding area and in the milking stall after milking caused stress for animals. 相似文献

Twelve Holstein cows were used in a replicated Latin square experiment to determine the effect of adding dried molasses to high-alfalfa silage diets on dairy cow performance. Three isonitrogenous diets were formulated with a 68:32 forage:concentrate ratio, with alfalfa silage as the only forage source. Dietary treatments were a control diet with no added molasses and 3 and 6% dried molasses diets. Three lactating Holstein cows fitted with ruminal cannulas were used to determine the effects of dietary treatments on ruminal fermentation. Dietary treatments had no effect on dry matter (average 23.3 kg/d), crude protein (average 4.4 kg/d), or neutral detergent fiber (average 7.4 kg/d) intake. Milk yield, energy-corrected milk (average 35.4 kg/d), and 4% fat-corrected milk (average 33.8 kg/d) were not influenced by dietary treatments. Cows fed the control diet produced milk with less milk urea nitrogen concentration than those fed molasses-supplemented diets. Ruminal pH, NH3-N concentration, and total volatile fatty acids were not different among dietary treatments. The molar proportion of acetate linearly increased, whereas the molar proportion of propionate linearly decreased as the level of dried molasses increased. It was concluded that addition of dried molasses to high-alfalfa silage diets at 6% of the diet (dry matter basis) increased milk urea nitrogen but had no effect on animal performance. 相似文献

The objectives of this study were to develop and test a system for automatic washing of the hooves of dairy cows and to evaluate the effect of frequent automatic washing on the prevalence of digital dermatitis (DD). An automatic hoof washer was developed in an experimental dairy herd and tested in 6 commercial dairy herds in 2 experiments (1 and 2). In the experimental herd, automatic hoof washing resulted in cleaner hooves. In experiments 1 and 2, cows were washed after each milking on the left side only, leaving the right side unwashed as a within-cow control. In experiment 1, hooves were washed with a water and 0.4% soap solution. In experiment 2, hooves were washed with water only. In each experiment, DD was scored in a hoof-trimming chute approximately 60 d after the start of hoof washing. Data were analyzed using a generalized linear mixed model. The outcome was the DD status of each leg (DD positive or DD negative). Herd and cow within herd were included as random effects, and treatment (washing or control) was included as a fixed effect. The statistical analyses showed that the odds ratio of having DD was 1.48 in the control leg compared with the washed leg in experiment 1. In experiment 2, the odds ratio of having DD was 1.27 in the control leg compared with the washed leg. Disparities between nutrient intake and demand often result in a state of negative energy balance (EB) in the early-lactation dairy cow. Reducing dietary crude protein (CP) content and providing glucogenic nutrients may overcome this issue. This study evaluates whether or not offering a diet lower in CP and higher in nonfiber carbohydrates (LP-NFC) can improve EB and the metabolic status of the early-lactation dairy cow compared with a diet higher in CP and fibrous carbohydrates (HP-FC). Twenty Holstein-Friesian dairy cows were assigned to 1 of 2 dietary treatments in a randomized block design. Diets were isoenergetic (6.57 MJ of net energy for lactation) and formulated to contain 15% CP and 6% starch (HP-FC), or 12% CP and 28% starch (LP-NFC) and were offered for the first 63 d of lactation. Intake and milk yield were determined daily, whereas milk and blood samples, weights, and body condition scores were collected weekly. Intakes (mean ± standard errors of the mean, SEM) of dry matter (17.4 ± 0.6 kg/d) and energy (113.0 ± 4.6 MJ of net energy for lactation) were not different between treatments. However, the HP-FC group had a higher milk yield (31.8 vs. 28.9 ± 1.4 kg/d) and a lower EB compared with the LP-NFC group. Blood urea N concentration (3.5 vs. 1.8 ± 0.2 mmol/L) was higher, whereas bilirubin (6.0 vs. 6.7 ± 0.2 mmol/L) and β-hydroxybutyrate concentrations (0.7 vs. 0.8 ± 0.05 mmol/L) were lower in the HP-FC group compared with the LP-NFC group. These data suggest that EB can be improved during early lactation through the manipulation of milk output by offering a lower CP, higher NFC diet. 相似文献

Staphylococcus aureus is a major cause of mastitis in dairy cattle. This study estimated the herd prevalence of methicillin-resistant Staph. aureus (MRSA) among US dairy herds by testing bulk tank milk (BTM) samples using genotypic and phenotypic methods. A nationally representative sample of 542 operations had BTM cultured for Staph. aureus, and 218 BTM samples were positive upon initial culture. After 4 wk to 4 mo of frozen storage, 87% of 218 samples (n = 190) were still culture positive for Staph. aureus on blood agar, but none were positive for MRSA on the selective indicator medium CHROMagar MRSA. A duplex PCR was used to detect the Staph. aureus-specific nuc gene and the methicillin resistance gene, mecA, in mixed staphylococcal isolates from the 190 BTM samples that were positive for Staph. aureus after storage. Seven samples tested positive for nuc and mecA, and 2 samples tested positive for mecA only. MecA-positive Staphylococcus spp., but not MRSA, were subsequently isolated from 5 samples, whereas neither mecA-positive Staphylococcus spp. nor MRSA was isolated from the remaining 4 samples. Presence of methicillin-resistant, coagulase-negative Staphylococcus spp. may complicate the detection of MRSA by means of PCR on BTM. Bulk tank milk in the United States is not a common source of MRSA. 相似文献

The working hypothesis was that postpartum anovulatory intervals (PPAI) of grazing dairy cows are shortened by inclusion of concentrates that increase the nonstructural carbohydrate content of the transition diet. Dietary treatments were arranged as a 2 × 2 factorial, with 68 multiparous cows assigned to isoenergetic diets (114 MJ of metabolizable energy/cow per day) of pasture and pasture silage (PreP) or pasture and pasture silage supplemented with 3 kg of dry matter/cow per day a corn- and barley-based concentrate for 36 d prepartum (PreC). After calving, cows within each prepartum diet group were managed on isoenergetic diets (179 MJ of metabolizable energy/cow per day) of either pasture and pasture silage (PostP) or pasture and pasture silage supplemented with 5 kg of dry matter/cow per day a corn- and barley-based concentrate (PostC) for at least 35 d and until reestablishment of ovulatory cycles. Relative to day of calving (d 0), blood samples were collected at least weekly from d −28 to 35 and milk samples were collected twice weekly for progesterone determination to diagnose ovulatory status. The main variable of interest was PPAI, defined as the interval between calving and the first detected increase in milk progesterone (>3 ng/mL), followed by a pattern of progesterone concentrations consistent with onset of an ovulatory cycle. Subsequent mating records, pregnancy testing, and recalving data were also examined. Prepartum diet did not affect reproduction. The PPAI was 8 d shorter and the 6-wk pregnancy rate was 17% greater in PostC cows compared with PostP cows. Measured indicators of metabolic state and energy balance were poorly related to PPAI. The results support the existence of nutritional signals associated with nonstructural carbohydrates in the postpartum diet, independent of energy balance; these signals benefit the physiological mechanisms underlying the timing of first ovulation and possibly subsequent breeding performance. 相似文献

This study was conducted to determine whether feeding cinnamaldehyde (main component of cinnamon bark essential oil; Cinnamon cassia), condensed tannins from quebracho trees (Schinopsis balansae), or saponins from Yucca schidigera altered the milk fatty acid profile of dairy cows. For this purpose, 4 lactating cows were used in 4 × 4 Latin square design (28-d period) and fed a total mixed ration containing no additive (control), or supplemented with cinnamaldehyde (1 g/d; CIN), quebracho condensed tannin extract (150 g/d; 70% of tannins; QCT), or Yucca schidigera saponin extract (60 g/d; 10% of saponins; YSE). Results revealed no effects of feeding CIN or QCT on milk fatty acid profile. Supplementation with YSE resulted in some modifications of milk fatty acid profile as suggested by the reduced proportions of C6:0 (2.71 vs. 2.95%), C8:0 (1.66 vs. 1.89%), and trans-11 C18:1 (0.92 vs. 1.01%). Results show low potential of cinnamaldehyde, condensed tannins, and saponins to alter ruminal biohydrogenation process and modify the fatty acid profile of milk fat at the feeding rates used in this study. Further investigations are needed to determine the factors that limit the effects of these secondary metabolites on ruminal microbial populations involved in the biohydrogenation processes of unsaturated fatty acids. 相似文献

Cortisol has long been used as a marker of the stress response in animals. Cortisol can be analyzed from different media, most notably from the blood, saliva, and feces; however, the collection of cortisol from some of these media requires invasive procedures or excessive handling of the animals. Furthermore, it is not possible to capture long-term increases in circulating concentrations of cortisol from the blood, saliva, or feces. Hair cortisol has been found to be a reliable alternative for measuring chronic stress. With this emerging measure, appropriate sampling methodology must be developed and validated. The aim of this study was to determine the effects of hair color, sampling location, and processing method on cortisol concentrations in hair from lactating black and white Holstein cows (n = 18). Furthermore, we aimed to measure the hair growth rates at different body locations (n = 12) and test hair cortisol levels when resampled over short intervals (n = 37). Both black- and white-colored hair was collected from the shoulder, top line, hip, and tail switch of Holsteins; due to breed characteristics only white hair was harvested from the tail switch. All samples were cleaned with water and isopropanol, and then ground in a ball mill or finely cut with scissors once dry. Cortisol was extracted with methanol before being measured using a commercially available ELISA kit. Concentrations of cortisol were greater in white than in black hair (7.8 ± 1.1 vs. 3.8 ± 1.1 pg/mg). When only white samples were analyzed, hair from the tail switch had more cortisol than hair from the shoulder (11.0 ± 1.2 vs. 6.2 ± 1.2 pg/mg), whereas no difference was found when compared with the hip and top line. Samples ground with a ball mill had greater concentrations of cortisol extracted than those minced with scissors (10.4 ± 1.2 vs. 4.7 ± 1.2 pg/mg). The growth rate of hair was significantly greater at the tail switch compared with the hip and shoulder (0.51 ± 0.05 vs. 0.04 ± 0.05 vs. 0.03 ± 0.05 mm/d). When hair was collected every 3 wk after calving, a tendency was detected for multiparous cows to have greater concentrations of hair cortisol and significantly greater concentrations of cortisol on d 0 and 21 after calving compared with d 42, 84, and 126. In Holsteins, the hair on the tail switch is always white, grows more rapidly than other sites, and is sensitive enough to capture changes in cortisol over intervals as short as 3 wk, making it the ideal location for measuring hair cortisol. 相似文献

Feed management is one of the principal levers by which the production and composition of milk by dairy cows can be modulated in the short term. The response of milk yield and milk composition to variations in either energy or protein supplies is well known. However, in practice, dietary supplies of energy and protein vary simultaneously, and their interaction is still not well understood. The objective of this trial was to determine whether energy and protein interacted in their effects on milk production and milk composition and whether the response to changes in the diets depended on the parity and potential production of cows. From the results, a model was built to predict the response of milk yield and milk composition to simultaneous variations in energy and protein supplies relative to requirements of cows. Nine treatments, defined by their energy and protein supplies, were applied to 48 cows divided into 4 homogeneous groups (primiparous or multiparous × high or low milk potential) over three 4-wk periods. The control treatment was calculated to cover the predicted requirements of the group of cows in the middle of the trial and was applied to each cow. The other 8 treatments corresponded to fixed supplies of energy and protein, higher or lower than those of the control treatment. The results highlighted a significant energy × protein interaction not only on milk yield but also on protein content and yield. The response of milk yield to energy supply was zero with a negative protein balance and increased with protein supply equal to or higher than requirements. The response of milk yield to changes in the diet was greater for cows with high production potential than for those with low production potential, and the response of milk protein content was higher for primiparous cows than for multiparous cows. The model for the response of milk yield, protein yield, and protein content obtained in this trial made it possible to predict more accurately the variations in production and composition of milk relative to the potential of the cow because of changes in diet composition. The objective of the current study was to quantify the change in the prediction of dry matter intake (DMI) resulting from the inclusion of rumination time (RT) in the 2001 National Research Council (NRC) DMI prediction model. Forty-one Holstein cows fed the same total mixed ration were involved in a 10-wk study. Individual DMI were measured daily. The accuracy and precision of the original NRC prediction model, based on body weight, fat-corrected milk, and week of lactation as independent variables, was compared with the accuracy and precision of the same model with RT as an additional independent variable. The RT estimate was significant in the model developed but had a low value (0.031 kg/h). Root mean square prediction errors were very similar in the 2 models (1.70 and 1.68 kg/d) as were the other indicators (R2, linear bias, random error, and concordance correlation coefficient) selected to compare the models in this study. These results indicate no gain in DMI prediction precision or accuracy when RT is included in the NRC model. 相似文献

The objective of the present study was to estimate heritabilities of milk fatty acids (FA) and genetic and phenotypic correlations among milk FA and milk production traits in Canadian Holsteins. One morning milk sample was collected from each of 3,185 dairy cows between February and June 2010 from 52 commercial herds enrolled in Valacta (Ste-Anne-de-Bellevue, Quebec, Canada). Individual FA percentages (g/100 g of total FA) were determined for each sample by gas chromatography. After editing the data, 2,573 cows from 46 herds remained. Genetic parameters were estimated using multitrait animal models fitted under REML. The model included fixed effects of age at calving and stage of lactation each nested within parity and random effects of herd-year-season of calving, animal, and residual. The pedigree of animals with data was traced back 5 generations on both the male and female sides to account for relationships among animals. The estimates of heritability for individual FA ranged from 0.01 to 0.39, with standard errors ranging from 0.01 to 0.06. Generally, monounsaturated FA (MUFA) and saturated FA (SFA) showed higher heritability estimates than polyunsaturated FA (PUFA). Overall, SFA were negatively genetically correlated with MUFA and PUFA, whereas genetic correlations between MUFA and PUFA were positive. The SFA showed positive associations with fat yield and fat percentage, whereas unsaturated FA were negatively associated with fat yield and fat percentage. Bovine milk FA composition could be improved through genetic selection, and selection for MUFA or against SFA could alter the bovine milk fat profile in a desirable direction. 相似文献

The objective of the present study was to evaluate the relationship between the efficiency of conversion of feed N into milk N [N-use efficiency (NUE)] and 15N enrichment of milk casein from lactating cows fed corn silage-based diets. Samples of feeds and milk were obtained from 3 experiments with lactating dairy cows. All diets were based on corn silage and were designed to evaluate the effects of (1) diets with different ratios of effective rumen-degradable protein to fermentable metabolizable energy (experiment 1), (2) different proportions of quickly and slowly rumen-degradable protein (experiment 2), and (3) synchronizing the availability of fermentable metabolizable energy and effective rumen-degradable protein in the rumen (experiment 3). Although no significant effect of diet on casein δ15N values was detected, casein was more enriched than the diet in each of the experiments. Nitrogen-use efficiency was negatively related to adjusted Δ15N (casein δ15N − diet δ15N) for experiments 1 and 2 individually and when combining data from all 3 experiments. The relatively low values for Δ15N suggest that these productive animals were using dietary N efficiently, with a high proportion of N going to milk protein and less to urea. The weak, although significant, relationship between NUE and adjusted Δ15N, is consistent with relatively little variation in hepatic deamination and transamination, with variation in rumen efficiency having the predominant effect on NUE. The present study confirms the lower 15N enrichment in protein when NUE is high and the potential to use N-isotope fractionation as a marker of NUE. 相似文献

Milk composition varies with energy status and was proposed for measuring energy balance on-farm, but the accuracy of prediction using monthly samples is not high. With automated sampling and inline milk analysis, a much higher measurement frequency is possible, and thus improved accuracy of energy balance determination may be expected. Energy balance was evaluated using data in which milk composition was measured at each milking. Three breeds (Danish Holstein, Danish Red, and Jerseys) of cows (623 lactations from 299 cows) in parities 1, 2, and 3+ were used. Data were smoothed using a rolling local regression. Energy balance (EBal) was calculated from changes in body reserves (body weight and body condition score). The relationship between EBal and milk measures was quantified by partial least squares regression (PLS) using group means data. For each day in lactation, the within-breed and parity mean EBal and mean milk measures were used. Further PLS was done using the individual cow data. The initial PLS models included 25 combinations of milk measures allowing a range of nonlinear effects. These combinations were as follows: days in milk (DIM); DIM raised to the powers 2, 3, and 4; milk yield; fat content; protein content; lactose content; fat yield; protein yield; lactose yield; fat:protein ratio; fat:lactose ratio; protein:lactose ratio; and milk yield:lactose ratio, together with 10 “diff()” variables. These variables are the current minus the previous value of the milk measure in question. Using group means data, a very high proportion (96%) of the variability in EBal was explained by the PLS model. A reduced model with only 6 variables explained 94% of the variation in EBal. This model had a prediction error of 3.82 MJ/d; the 25-variable model had a prediction error of 3.11 MJ/d. When using individual rather than group means data, the PLS prediction error was 17.3 MJ/d. In conclusion, the mean Ebal of different parities of Holstein, Danish Red, and Jersey cows can be predicted throughout lactation using 1 common equation based on DIM, milk yield, milk fat, and milk protein measures. 相似文献

Two experiments were conducted to study changes induced by stage of lactation and milk ejection in the cisternal compartment of the udder in dairy cows. In experiment 1, 18 cows grouped according to stage of lactation were used 12 h after milking for measuring alveolar and cisternal milk volumes (by cannula) and cisternal area (by ultrasonography) in the front quarters. Cisternal milk and cisternal area were correlated (r = 0.74 to 0.82) for all stages of lactation. As lactation advanced, volumes of alveolar and cisternal milk and cisternal area decreased. Proportion of cisternal milk varied between stages (early, 33.2%; mid, 23.1%; and late, 42.6%). In experiment 2, 7 cows were used to show return of milk from cisternal to alveolar compartments when milk ejection was induced without milking. Cisternal area was measured before (0 min) and after (3, 15, 30, and 60 min) an i.v. oxytocin (OT) injection administered immediately before normal a.m. and p.m. milking times. Cisternal area increased dramatically from 0 to 3 min (98%) and decreased slowly thereafter. The 0- and 3-min data provide clear evidence of milk ejection, and their difference indicated cistern elasticity. Maximum cisternal area in each cow was similar for the 8- and 16-h milking intervals, indicating that in both cases the cistern was completely full of milk. Decrease in cisternal area after 3 min was significant at 15, 30, and 60 min. Decreased cisternal area was interpreted as the reflux of cisternal milk to the alveolar compartment. We termed this 'cisternal recoil.' In conclusion, ultrasonography was a useful method to evaluate dynamic changes in cisternal milk throughout lactation and after udder stimulation in dairy cows. The relationships between dairy cow milk-based energy status (ES) indicators and fertility traits were studied during periods 8 to 21, 22 to 35, 36 to 49, and 50 to 63 d in milk. Commencement of luteal activity (C-LA) and interval from calving to the first heat (CFH), based on frequent measurements of progesterone by the management tool Herd Navigator (DeLaval), were used as fertility traits. Energy status indicator traits were milk β-hydroxybutyrate (BHB) concentration provided by Herd Navigator and milk fat:protein ratio, concentration of C18:1 cis-9, the ratio of fatty acids (FA) C18:1 cis-9 and C10:0 in test-day milk samples, and predicted plasma concentration of nonesterified fatty acids (NEFA) on test days. Plasma NEFA predictions were based either directly on milk mid-infrared spectra (MIR) or on milk fatty acids based on MIR spectra (NEFAmir and NEFAfa, respectively). The average (standard deviation) C-LA was 39.3 (±16.6) days, and the average CFH was 50.7 (±17.2) days. The correlations between fertility traits and ES indicators tended to be higher for multiparous (r < 0.28) than for primiparous (r < 0.16) cows. All correlations were lower in the last period than in the other periods. In period 1, correlations of C-LA with NEFAfa and BHB, respectively, were 0.15 and 0.14 for primiparous and 0.26 and 0.22 for multiparous cows. The associations between fertility traits and ES indicators indicated that negative ES during the first weeks postpartum may delay the onset of luteal activity. Milk FPR was not as good an indicator for cow ES as other indicators. According to these findings, predictions of plasma NEFA and milk FA based on milk MIR spectra of routine test-day samples and the frequent measurement of milk BHB by Herd Navigator gave equally good predictions of cow ES during the first weeks of lactation.
