Polymorphisms MC4R c.1426 A>G (p.Asp298Asn) and LEPR c.2002 C>T (p.Leu663Phe) have been associated in pigs with feed intake, growth and fatness. Our aims were to analyze how these polymorphisms influence production and quality traits in Duroc × Iberian crossbred pigs, and moreover evaluate how the effects of gender and Duroc line were affecting these traits. Significant effects of gender, Duroc sire line and LEPR c.2002 C>T mutation, were detected on the main traits. LEPR c.2002 T allele increased body weight (2.5 ± 0.7 kg), backfat thickness (1.5 ± 0.4mm), loin intramuscular fat (0.7 ± 0.2 %), and lightness (1.0 ± 0.3); however it was decreasing premium cuts yield (-0.9 ± 0.2%). Allele c.2002 T increased SFA (0.60 ± 0.12 %), and decreased MUFA (-0.35 ± 0.12%) and PUFA (-0.25 ± 0.06%). No significant effects of MC4R c.1426 A>G SNP were identified. A genetic test based on the LEPR c.2002 C>T SNP may be useful to select Duroc sires to reduce undesirable heterogeneity of carcasses and premium cuts of crossbred animals.  相似文献

The complex and interrelated management components of dairy farming are associated with health, production, and profitability of the herd, yet there is limited objective data on current management practices of the far-off, close-up, and fresh periods across Canadian dairy farms. We aimed to describe management practices of Canadian dairy farms by using a pre-existing risk assessment tool and outline potential management opportunities. Upon veterinarians' or producers' request, a transition management risk assessment (The Vital 90, Elanco) was performed by trained observers (n = 10) during farm visits (n = 78) between August 2014 and March 2018. Most farms were in Ontario (n = 64), whereas the remaining were in Alberta (n = 5), British Columbia (n = 4), Manitoba (n = 1), Prince Edward Island (n = 2), Newfoundland (n = 1), and Saskatchewan (n = 1). The study included 79 questions about nutrition, pen management, and cow comfort of the dry (approximate ranges: far-off, ?60 to ?20 d in milk; close-up, ?20 to 0 d in milk) and fresh (0–30 d in milk) periods. The herds averaged 125 milking cows, and most had 2 defined dry groups (81%). Freestall (FS; 54%) and straw-bedded loose pack (BP; 81%) were the most common housing systems observed in the far-off and close-up periods, respectively. Heifers and cows were housed together in 56, 80, and 59% of the far-off, close-up, and fresh pens, respectively. A large proportion of the far-off (FS: >100% stocking density; BP: <9.3 m2/cow; 41%), close-up, and fresh pens (FS: >80% stocking density; BP: <13.9 m2/cow; 52 and 49%, respectively) were overstocked. Poor water access was observed across all periods (65, 58, and 24% of the far-off, close-up, and fresh, respectively). Only a few farms had proper heat abatement systems in place (absence of properly functioning soakers or fans; <10% in the dry and 15% in the fresh periods). Cows were able to sort their ration in 60% of the dry period pens and 31% of the fresh pens. In 73% of the farms, fresh cow health monitoring protocols were not in place. Colostrum cows and sick cows were housed together in 40% of the farms; 59% separated the newborn from the dam within 2 to 12 h of birth with colostrum harvested immediately thereafter. This work describes prevalent management practices in the dry and fresh periods and highlights areas for potential improvement. Future research should focus on the associations between management choices and health performance of dairy farms.  相似文献

Front-of-package (FOP) nutrition labels are designed to help consumers evaluate the healthiness of foods and to promote healthier food choices. In this study, an online experiment with Swiss consumers (N = 1313) was conducted to compare the effects of different nutrition label formats on consumers’ evaluations of snack food healthiness. Participants were asked to select the healthier option in 105 pairwise comparisons of 15 salty snacks. The participants were randomly assigned to one of five conditions: the FOP presented with (1) the nutrition facts table, (2) the multiple traffic light (MTL), (3) the Nutri-Score, (4) the Nutri-Score on half of the products, or (5) no nutrition information (control). The consumers’ evaluations of the snacks’ healthiness were fairly accurate, even without being given nutrition information on the packaging. The Nutri-Score led to the greatest accuracy in identifying the healthier of two snacks (when using the British FSA/Ofcom nutrient profiling score to determine product healthiness); however, this had only a minimal effect on the evaluation when only some of the products were labelled. Both FOP labels were superior to the FOP with and without the nutrition facts. This indicates that for maximum effectiveness, the labelling of all available products is needed. The perceived usefulness and public support of mandatory implementation were higher for the MTL than for the Nutri-Score label; however, for the latter, perceived usefulness and public acceptance were higher among the participants who became familiar with the label during the experiment than among those who did not.  相似文献

Mangos are a source of bioactive compounds with potential health promoting activity. Biological activities associated with mango fractions were assessed in cell-based assays to develop effective extraction and fractionation methodologies and to define sources of variability. Two techniques were developed for extraction and fractionation of mango fruit peel and flesh. Liquid chromatography-mass spectrometry (LC-MS) was used to assess compositional differences between mango fractions in flesh extracts. Many of the extracts were effective in inhibiting the proliferation of human breast cancer cells in vitro. All fractions showed bioactivity in PPAR activation assays, but quantitative responses showed marked fruit-to-fruit variability, highlighting the need to bulk fruit prior to extraction for activity-guided fractionation of bioactive components. This study also suggests that combinations of diverse molecular components may be responsible for cell-level bioactivities from mango fractions, and that purification and activity profiling of individual components may be difficult to relate to whole fruit effects. Practical Application: Although the health benefits of fruits are strongly indicated from studies of diet and disease, it is not known what role individual fruit types can play, particularly for tropical fruits. This study shows that there is a diversity of potentially beneficial bioactivities within the flesh and peel of mango fruit, although fruit-to-fruit variation can be large. The results add to the evidence that the food approach of eating all components of fruits is likely to be more beneficial to health than consuming refined extracts, as the purification process would inevitably remove components with beneficial bioactivities.  相似文献

Conjugated linoleic acid (CLA) has drawn much interest in last two decades in the area ranging from anticancer activity to obesity. A number of research papers have been published recently with regard to CLA's additional biological functions as reproductive benefits. However, not much is known how this mixture of isomeric compounds mediates its beneficial effects particularly on fertility. In this study, we demonstrated the cross talk between downstream signaling of CLA and important hormone regulators of endocrine system, i.e. FSH and IGF1, on buffalo granulosa cell function (proliferation and steroidogenesis). Experiments were performed in primary serum-free buffalo granulosa cell culture, where cells were incubated with CLA in combination with FSH (25?ng/ml) and IGF1 (50?ng/ml). Results showed that 10?μM CLA inhibits FSH- and IGF1-induced granulosa cell proliferation; aromatase, GATA4, and IGF1 mRNA; and estradiol-17β production. Western blot analysis of total cell lysates revealed that CLA intervenes the IGF1 signaling by decreasing p-Akt. In addition, CLA was found to upregulate peroxisome proliferator-activated receptor-gamma (PPARG) and phosphatase and tensin homolog (PTEN) level in granulosa cells. Further study using PPARG- and PTEN-specific inhibitors supports the potential role of CLA in granulosa cell proliferation and steroidogenesis involving PPARG, PTEN, and PI3K/Akt pathway.  相似文献
