There have been very few reports of the effect of pesticides on immediate allergy. In the previous report, the effect of pyrethroid pesticides and organophosphorus pesticides on immediate allergic reaction was investigated. Subsequently, 12 organonitrogen pesticides, 14 carbamate pesticides and 4 other pesticides were investigated for their effects on the enzyme activity of beta-hexosaminidase as an index of chemical mediator release from rat basophilic leukemia cells (RBL-2H3). Two organonitrogen pesticides, bitertanol and pyridaben, and two organotin pesticides, cyhexatin and fenbutatin oxide, were found to promote beta-hexosaminidase release. Bitertanol non-specifically caused the promotion of chemical mediator release, while the release-promotive action of pyridaben was related to IgE antibody and those of cyhexatin and fenbutatin oxide to cell injury. On the other hand, two organonitrogen pesticides, propiconazole and triadimenol, and imazalil showed release-inhibitory action. These data suggested that some pesticides can affect immediate allergy. 相似文献

The effects of foods and chemicals related to food hygiene on degranulation were evaluated using a method for assaying the enzyme activity of beta-hexosaminidase as an index of chemical mediator release from RBL-2H3 cells in vitro. Using a previously developed assay system, we had found a large number of inhibitors and promoters of degranulation of RBL-2H3 cells. In the present study, we examined the inhibitory effect of zinc chloride on the degranulation (beta-hexosaminidase release) from RBL-2H3 cells with or without antigen in the presence of the degranulation-promotive chemicals, namely, 4 food additives, 7 pesticides and 2 veterinary drugs. These promotive chemicals were classified into two types on the basis of inhibitory profile by zinc chloride: 1) those which showed marked degranulation-inhibitory action when the cells were stimulated with antigen, such as butylhydroxyanisole, dibutylhydroxytoluene, EPN, cis- and trans-permethrin, prothiofos, pyridaben, terbufos, 2) those which showed marked degranulation-inhibitory action whether the cells were stimulated with antigen or not, such as butyl p-hydroxybenzoate, o-phenylphenol, bitertanol, salinomycin. In conclusion, zinc had a dramatic inhibitory effect on enhanced degranulation induced by synthetic chemicals in vitro. 相似文献

We found that two distinct flavonoid glycosides isolated from the peel of Japanese persimmon (Diospyros kaki Fuyu), isoquercitrin (Isq) and hyperin (Hyp), are capable of inhibiting antigen-stimulated degranulation in rat basophilic leukaemia RBL-2H3 cells. In order to elucidate the underlying mechanisms, we examined effects of Isq and Hyp on cellular responses induced by antigen stimulation. Treatment with both Isq and Hyp markedly inhibited antigen-stimulated elevation of intracellular free Ca2+ concentration and reactive oxygen species (ROS). Isq and Hyp did not affect NADPH oxidase (NOX) activity, but they possessed DPPH radical-scavenging activity similar to that of epigallocatechin gallate, a potent anti-oxidant, Finally, Isq and Hyp showed little or no effects on Ag-stimulated Syk activation or phosphorylation of signalling molecules. These results indicate that inhibition of antigen-stimulated degranulation by Isq and Hyp is mainly due to suppression of intracellular Ca2+ elevation, which is caused by direct scavenging of ROS that are generated by NOX. Our findings suggest that Isq and Hyp, isolated from the peel of persimmon, would be beneficial for alleviating symptoms of type I allergy. 相似文献

Grapeseed extract (GSE) is a rich source of natural phenolic compounds and possesses various pharmacological activities, including antioxidant, anticarcinogenic and anti-inflammatory properties. However, effects of GSE on immunoglobulin (Ig) E-mediated allergic responses still remain elusive. In the present study, the effects of GSE on activation and degranulation of RBL-2H3 mast cells were investigated. GSE pretreatments (20-100 μg/ml) reduced IgE-antigen mediated release of β-hexosaminidase and histamine in RBL-2H3 cells. Additionally, GSE reversibly inhibited expression of FcεRI on RBL-2H3 cells. GSE treatments caused a significant elevation of intracellular cAMP levels, whereas the Ca2+ influx upon antigen stimulation was inhibited. Suppression on FcεRI expression together with decreased calcium uptake and increased cAMP level might be involved in attenuated degranulation of mast cells by GSE treatment. Our results suggest a possible pharmaceutical application of GSE in treating type I allergic diseases. 相似文献

The inhibitory effect of an aqueous extract from spinach on degranulation of RBL-2H3 cells is herein reported. The extract significantly suppressed antigen-induced degranulation in a dose-dependent manner without affecting cell viability. Active substances in the extract were heat-stable and trypsin-resistant with molecular weights ranging from 500 Da to 14 kDa. The extract inhibited elevation of the intracellular Ca2+ concentration caused by stimulation by antigen, while not suppressing degranulation induced by a calcium ionophore A23187. Immunoblot analysis revealed that the inhibitory effect results from downregulation of phosphorylation of both Syk kinase and phosphatidylinositol 3-kinase in the signalling pathways involved in degranulation caused by the antigen–antibody interaction. Taken together, these findings suggest that aqueous spinach extract has an anti-allergic activity that controls degranulation. 相似文献

目的 研究黄曲霉毒素B1 (aflatoxin B1, AFB1)暴露对鸡蛋过敏原卵白蛋白(ovalbumin, OVA)激发人外周血嗜碱性白血病细胞(KU812)脱颗粒的影响, 以阐明AFB1暴露与食物过敏的相关性。方法 构建AFB1暴露下OVA激发KU812细胞脱颗粒模型, 采用酶联免疫吸附法(enzyme linked immunosorbent assay, ELISA)检测AFB1对其生物活性介质β-氨基己糖苷酶(β-hexosaminidase, β-HEX)和组胺, 以及细胞因子白细胞介素-4 (interleukin-4, IL-4)、白细胞介素-6 (interleukin-6, IL-6)、白细胞介素-1β (interleukin-1β, IL-1β)释放量的影响; 然后采用实时定量荧光聚合酶链反应法分析对IL-4和IL-6的mRNA表达水平的影响。结果 OVA刺激KU812细胞后会使其释放更多生物活性介质, 包括β-HEX、组胺及细胞因子(IL-4、IL-6、IL-1β); 当有AFB1同时暴露, 则会不同程度地加重这些生物活性介质的释放量。其中, AFB1对β-HEX和组胺的释放量影响较小; 但能够显著提高细胞因子IL-4、IL-6以及IL-1β的分泌量(P<0.01), 尤其是对IL-1β的影响更显著, 相较于对照组其分泌量提高84%。同时IL-4和IL-6的mRNA基因的表达水平也被提高。结论 OVA激发KU812会导致其脱颗粒并释放与食物过敏相关的一些活性介质和细胞因子。而AFB1质量浓度为10000 ng/mL的同时暴露, 会提高这些相关活性介质和细胞因子的释放量, 从而加重OVA所引发的食物过敏反应。 相似文献

The anti-allergic effect of alcohol extracts from edible seaweeds was evaluated by the degranulation of a rat basophilic leukemia cell line (RBL-2H3). Extracts prepared from the brown algae Sargassum horneri, Ecklonia cava and Ecklonia kurome, inhibited the degranulation of RBL-2H3 cells, whereas those from green algae and red algae did not. The active substance in S. horneri was purified and identified as chlorophyll c2, a major pigment in brown algae. The purified chlorophyll c2 preparation also suppressed the degranulation of RBL-2H3 cells. In contrast, chlorophyll a and b did not affect cell degranulation. Therefore, chlorophyll c2 from brown algae might be useful for the prevention or mitigation of allergic symptoms. 相似文献

在埃切假丝酵母(Candida etchellsii)发酵过程中,通过盐度调控和氨基酸添加,强化目标产物HEMF(2(5)-乙基-4-羟基-5(2)-甲基-3(2H)-呋喃酮)的合成效率。分阶段调控发酵体系的盐度(初始阶段控制CaCl浓度为200 g/L,发酵40 h后提升至220 g/L),结合氨基酸添加(向发酵体系中添加丙氨酸、精氨酸和甘氨酸各1.67 g/L)。摇瓶结果表明:酵母C.etchellsii合成HEMF,其产量为110.74 mg/L。7 L发酵罐上罐验证,HEMF产量达到121.51 mg/L,相比空白(200 g/L CaCl浓度下且没有添加氨基酸)提高了21.2倍。分阶段盐度调控结合氨基酸添加策略显著强化了C.etchellsii对HEMF的合成。 相似文献

In this study we investigated the chromatographic behaviour of 40 veterinary drugs belonging to different classes (sulphonamides, nitrofurans, nitroimidazoles, antibiotics, anthelmintics, psychopharmacological substances, growth promoters) using the gel permeation media Sephadex LH-20, Bio Beads S-X 3 and Fractogel PGM 2,000. To allow the use of acetic acid in mobile phases all parts containing brass in the GPC equipment (Autoprep 1002 A) had to be exchanged for stainless steel components. The curves of elution with Sephadex LH-20 as stationary phase demonstrated good separation of the majority of drugs from co-extractives from liver, kidney, muscle, egg and milk by gravimetric measurement as well as by UV absorption. A clean-up of similar efficiency could however also be obtained with Sep-Pak silica cartridges, if final residue analysis was made by HPLC with UV detection. 相似文献

黄原胶(Xanthan Gum)是由野油菜黄单胞菌(Xanthomonas campestris)碳水化合物为主要底物,经发酵产生的‘种酸性胞外杂多糖。发酵中后期黏度增大,氧的传质能力下降,从而影响了黄原胶的产量。本课题研究黄原胶生物合成过程中,H2O2对供氧和提高产胶率的影响。根据添加H2O2实验得出,H2O2添加量为1ml即在发酵液中浓度为4.4mmol/L左右为最适的添加量,最佳添加时刻为48h左右。得出低浓度的H2O2能提高黄原胶的产量,产胶率平均提高了3.28%。 相似文献

4-Hydroxy-2,5-dimethyl-3(2#)-furanone (HDMF) was detected at concentrations of 598 μg/l and 427 μg/l, respectively, after 7 days of growth ofLactobacillus helveticus andL. delbrueckii on an aqueous suspension of whey powder. The uranium carbonate andersonite Na2Ca[UO2(CO3)3] x 6H2O was synthesized and identified with classical analytical and spectroscopic methods. The classical methods applied were powder X-ray diffraction (XRD), nitric acid digestion, and scanning electron microcopy combined with energy-dispersive spectroscopy (SEM/EDS). To characterize andersonite spectroscopically, time-resolved laser-induced fluorescence spectroscopy (TRLFS), X-ray photoelectron spectroscopy (XPS), and Fourier transform infrared spectroscopy (FT-IR) were used. Natural and synthetic andersonite samples were characterized with the nondestructive TRLFS by six fluorescence emission bands at 470.6, 486.1, 505.4, 526.7, 549.6, and 573.9 nm. In addition, andersonite was characterized by FT-IR measurements by the appearance of the asymmetric stretching vibration of the uranyl cation [v3(UO2(2+))] at 902 cm(-1) with a shoulder at 913 cm(-1). XPS measurements verified the composition of the synthetic andersonite sample. The measured intensity ratios of the XPS lines agree with the stoichiometry of Na2Ca[UO2(CO3)3] x 6H2O. The XPS features of the inner valence molecular orbitals are characteristic of the [UO2(CO3)3]4- structural moiety. These spectroscopic methods can be used to identify in a fingerprinting procedure secondary U(VI) phases in mixtures with other phases or as thin coatings on mineral and rock surfaces. 相似文献

Relative rate techniques were used to measure k(OH + HFE-7500) = (2.6+/-0.6) x 10(-14), k(Cl + HFE-7500) = (2.3+/-0.7) x 10(-12), k[Cl + n-C3F7CF(OC(O)H)CF(CF3)2] = (9.7+/-1.4) x 10(-15), and k[Cl + n-C3F7CF(OC(O)CH3)CF(CF3)2] < 6 x 10(-17) cm3 molecule(-1) s(-1) at 295 K [HFE-7500 = n-C3F7-CF(OC2H5)CF(CF3)2]. From the value of k(OH + HFE-7500) an estimate of 2.2 years for the atmospheric lifetime of HFE-7500 is obtained. Two competing loss mechanisms for n-C3F7-CF(OCHO.CH3)CF(CF3)2 radicals were identified in 700 Torr of N2/O2 diluent at 295 K; reaction with O2 and decomposition via C-C bond scission with kO2/k(decomp) = 0.013+/-0.006 Torr(-1). The Cl atom initiated oxidation of HFE-7500 in N2/O2 diluent gives n-C3F7CF(OC(O)CH3)CF(CF3)2 as the major product and n-C3F7CF(OC(O)H)CF(CF3)2 as a minor product. The atmospheric oxidation of HFE-7500 gives n-C3F7-CF(OC(O)CH3)CF(CF3)2 and n-C3F7CF(OC(O)H)CF(CF3)2 as oxidation products. A temperature-vacuum swing (TVS) cyclic process is applied to an amine-functionalized nanofibrilated cellulose sorbent to concurrently extract CO(2) and water vapor from ambient air. The promoting effect of the relative humidity on the CO(2) capture capacity and on the amount of coadsorbed water is quantified. The measured specific CO(2) capacities range from 0.32 to 0.65 mmol/g, and the corresponding specific H(2)O capacities range from 0.87 to 4.76 mmol/g for adsorption temperatures varying between 10 and 30 °C and relative humidities varying between 20 and 80%. Desorption of CO(2) is achieved at 95 °C and 50 mbar(abs) without dilution by a purge gas, yielding a purity exceeding 94.4%. Sorbent stability and a closed mass balance for both H(2)O and CO(2) are demonstrated for ten consecutive adsorption-desorption cycles. The specific energy requirements of the TVS process based on the measured H(2)O and CO(2) capacities are estimated to be 12.5 kJ/mol(CO2) of mechanical (pumping) work and between 493 and 640 kJ/mol(CO2) of heat at below 100 °C, depending on the air relative humidity. For a targeted CO(2) capacity of 2 mmol/g, the heat requirement would be reduced to between 272 and 530 kJ/mol(CO2), depending strongly on the amount of coadsorbed water. 相似文献
