为提高牡蛎蛋白的利用率,以新鲜牡蛎为原料,采用水解蛋白酶与风味蛋白酶对新鲜牡蛎进行复合酶解,并利用响应面法对牡蛎酶解工艺参数进行优化。结果表明:牡蛎蛋白的最优酶解条件为,复合酶的加酶量为0.70%、p H6.87,酶解温度58.6℃,酶解时间为3.4 h。在此条件下,新鲜牡蛎酶解后的水解度最高,为40.71%。  相似文献

以南美白对虾虾头为原料,对复合酶酶解的工艺条件进行优化,制备高水解度的虾头酶解液。首先确定最佳复合酶组合,然后通过单因素试验和响应面分析法优化并确定最佳的酶解条件。结果表明,采用风味酶与中性酶组合对虾头进行酶解(风味酶:中性酶为1:1,酶活比),加酶量2000U/g 蛋白、料液比2:1(g/ml)、pH7.33、温度54.27℃、时间3.97h。酶解后增味氨基酸含量明显增加,酶解蛋白液的风味良好,可以作为调味品基料使用。  相似文献

酶法改性处理玉米胚芽蛋白粉,并比较改性前后玉米胚芽蛋白粉的功能性质.以改性时间、底物浓度和加酶量为响应因素,玉米胚芽蛋白粉凝胶硬度为响应值,采用响应面分析方法,优化玉米胚芽蛋白粉酶改性工艺.结果表明:酶法改性玉米胚芽蛋白粉的最优工艺条件为改性时间3.18 h;底物质量分数10.88%;加酶量24.16 U/g,在此工艺条件下得到的凝胶硬度为98.13 g.改性后的玉米胚芽蛋白粉其功能性质有了较明显的改善,说明酶法改性玉米胚芽蛋白粉效果明显.  相似文献

The simultaneous effects of slurry concentration (25–35 % w/w), enzyme concentration (0.5–2.5 % w/w) and liquefaction time (30–90 min) on the yield, total solids and rheological parameters of oat milk developed were studied and optimised by the application of response surface methodology. The effect of independent and dependent variables have been studied using a central composite rotatable design of experiments. Power law model explains the flow behaviour of the developed oat milk samples with correlation coefficient (R 2) varying from 0.89 to 0.96. All the formulations exhibited pseudo plastic behaviour with the flow behaviour index (n) between 0.29 and 0.46 and consistency index varying from 1.033 to 10.22 Pa s n . Statistical analysis showed that yield, total solids and rheology of oat milk were significantly (p?<?0.05) correlated to slurry concentration, enzyme concentration and liquefaction time. The optimum conditions for making oat milk were: 27.1 % w/w slurry concentration, 2.1 % w/w enzyme concentration and liquefaction time of 49 min.  相似文献

为优化双酶水解技术生产婴幼儿米粉工艺,以α-淀粉酶添加量、β-淀粉酶添加量、调浆水温度为主要影响因素,结合实际生产中的其他水解条件,在单因素试验基础上,运用Box-Behnken试验设计原理,探讨α-淀粉酶添加量、β-淀粉酶添加量、调浆水温度的最佳组合。结果表明:α-淀粉酶添加量0.04‰(相当于0.15 U/g)、β-淀粉酶添加量0.26%(相当于1 820 U/g)、调浆水温度70.2 ℃时生产米粉的淀粉消化指数高达39.26%,与市售品牌米粉相比,淀粉消化指数提高10%以上。  相似文献

该研究利用响应面法优化一款富含益生元的代餐粉配方。利用高溶胀系数和具有饱腹感的魔芋精粉,添加具有改善肠道菌群的菊粉、低聚果糖、低聚异麦芽糖益生元,添加大豆分离蛋白、植物甾醇酯、木糖醇、香芋粉、三氯蔗糖成分,以代餐粉感官评分为指标,通过单因素试验、Plackett-Burman试验、最陡爬坡试验和Box-Behnken试验优化产品配方,并进行产品质量评价。结果显示,利用响应面方法优化得到最优配方为魔芋精粉添加量3.42 g、大豆分离蛋白0.80 g、植物甾醇酯0.23 g、益生元组合8.00 g(菊粉2.50 g、低聚果糖2.50 g、低聚异麦芽糖3.00 g)、木糖醇1.00 g、香芋粉1.20 g、三氯蔗糖0.005 g,实际感官评分为93.16。该配方代餐粉50 s即可充分溶解,冲调性较好,无颗粒,色泽均匀一致,口感细腻、黏度适中、味道纯正,气味协调,各理化指标及微生物检测按照国家食品相关标准进行了检测,结果均与添加量基本一致,符合国家标准。  相似文献

Optimum technology of spray-dried jujube powder was studied using a Box-Benhnken design. Effects of weight ratio of maltodextrin and the dry matter weight of jujube pulp (WR, 3–5), inlet air temperature (T, 140–160 °C), and shift of feed flow rate (FFR, 3–5) on the quality attributes (moisture content, lightness, the total color difference, vitamin C (VC) content and hygroscopicity) of jujube powder were systematically studied by response surface methodology and perturbation plot. Analysis of variance showed that a second-order polynomial equation could predict the experimental data well (R 2?>?0.92). Moisture content and the total color difference (?E) were significantly affected by WR, T, and FFR. The higher WR and lower FFR resulted in a decrease in moisture content and ?E. WR and FFR had extremely significant effects on VC content and hygroscopicity (P?<?0.01); with lower WR and FFR, higher values of VC content and hygroscopicity would be received. The effect of WR was the most significant variable among others. The optimum conditions that maximizing VC content and minimizing contents of moisture, ?E, and hygroscopicity of jujube powder were found as WR of 3, inlet air temperature of 140 °C, and FFR of 3 ( i.e., 11 ml/min). Under these conditions, the response values were moisture content of 3.36 %, lightness of 83.32, ?E of 9.92, VC content of 205.10 mg/100 g, and hygroscopicity of 18.59 %, respectively.  相似文献

以油茶粕蛋白为原料,研究了其最适水解酶,并对酶解工艺进行了优化。研究结果表明,油茶粕蛋白酶解的最适酶为2709碱性蛋白酶。以多肽得率为指标,运用Design-Expert7.1Trial统计分析软件,设计了4因素3水平试验,得到了油茶粕蛋白酶解的最优条件,pH值为8.55,酶用量为66.38 mg/g,温度为45.80℃,水解时间为5.24 h,在此条件下,预测多肽得率为39.72%。  相似文献

The main objective of this study was to understand the effects and relationship amongst four factors, which are reaction temperature, reaction time, enzyme load, and substrate mole ratio with the purpose of producing healthy functional cooking oil for long-term dietary treatment. Lipozyme RM IM lipase-catalyzed esterification of medium- and long-chain triacylglycerols (MLCT) from glycerol and mixtures of capric and oleic acid was optimized using response surface methodology (RSM) with a five-level, four-factorial design. Reaction temperature, reaction time, and substrate mole ratio strongly affected MLCT synthesis (P < 0.01). However, enzyme load did not have a significant (P > 0.01) effect on MLCT yield. Comparison between predicted and experimental value from central composite rotatable design optimization procedures revealed good correlation, implying that the reduced cubic polynomial model with backward elimination statistically expressed the percent MLCT yield obtained. The optimum MLCT yield was 59.76% by using 10 wt% enzyme load, reaction temperature of 70°C, reaction time of 14 h, and substrate mole ratio of 3.5:1. Experiments to confirm the predicted results using the optimal parameters showed an MLCT yield of 56.35% (n = 2). The choice on the types of fatty acids used in MLCT optimization work greatly influenced the physical and chemical properties of MLCT oil produced. The refined MLCT oil characteristics study showed this oil is suitable to be used for cooking/frying purposes as a high-value added product.  相似文献

为优化碱性蛋白酶酶解核桃蛋白制备抗氧化活性肽的工艺条件,考察酶解条件对酶解产物的抗氧化活性的影响。以羟基自由基的清除率、超氧阴离子自由基的清除率、还原能力为考察指标,使用响应面分析法,研究温度、pH值、底物浓度和酶添加量对制备抗氧化活性肽工艺的影响。经过优化得出最优酶解条件为:温度51℃、pH值8.13、底物浓度3.16%、酶添加量3.30%,在此最优条件下核桃多肽对羟基自由基的清除率为55.93%、对超氧阴离子自由基的清除率为47.85%、还原能力为55.34%。在10mg/mL的浓度下,核桃多肽的还原能力是VC的55.3%,对羟基自由基和超氧阴离子自由基的清除率分别是VC的93.8%和52.2%。  相似文献

研究木瓜蛋白酶和复合风味酶水解河蟹的工艺条件。在单因素试验基础上,通过SAS数据统计分析软件,运用四因素三水平的响应面设计方法,建立复合酶解蟹肉蛋白的二次多项数学模型,以水解度为响应值作响应面和等高线。结果表明:最适水解工艺为pH5、温度55℃、木瓜蛋白酶加酶量为5500U的条件下水解1.5h后再加入2LAPU的风味蛋白酶水解4.5h(酶解时间6h),此时的水解度预测值为31.19%、实测值30.08%,相对偏差为3.69%。最后,探索将水解液初步制成海鲜调味品。在上述条件下模拟出的酶解液鲜味明显且最后成品风味浓郁、色泽鲜黄、便于调味。  相似文献

Starch hydrolysates containing glucose can be obtained from potato (Solanum tuberosum). Acid hydrolysis generates a solution that must be neutralized to be used for fermentative purposes while an enzymatic hydrolysis generates a glucose solution which can be used without further treatments. This work deals with the modelling of the enzymatic hydrolysis of dried potatoes with α‐amylase and glucoamylase with temperature, time, activity units/substrate ratio and α‐amylase activity fraction as operational variables and released glucose concentration as dependent variable. The model obtained showed that the optimal conditions were 70ºC, 60 min; 74 activity units/g substrate and 0.13 α‐amylase activity U/total U fraction. Using these conditions, a glucose concentration of 38.9 g/L was predicted which corresponded well with an experimental value of 38.0 ± 0.5 g/L. The model obtained can be used in the development of processes for the hydrolysis‐fermentation of potato for the production of food additives or ethanol.  相似文献

研究微波结合酶解法组合技术对米糠多肽制备过程中黄曲霉毒素B_1(Aflatoxin B_1,AFB_1)的影响,为染毒米糠的脱毒提供新途径。在单因素试验的基础上,将料液比(X1)、微波时间(X2)、碱性蛋白酶酶解时间(X3)、酶解p H(X4)因素进行组合,应用Design Expert软件和Box-Behnken中心组合试验方法对AFB1降解条件进行了优化,测定米糠多肽AFB1的降解量。初始米糠中AFB1浓度为100.14μg/kg,运用响应面分析确定了其最佳工艺条件:微波功率750 W、微波时间9.56 min;碱性蛋白酶酶解温度50℃、酶解p H 10.74、酶解时间1.57 h、加酶量0.4%;料液比1:9.96,在此条件下,米糠中AFB1的降解率为98.68%,所制备的米糠多肽残留浓度为1.85μg/kg,符合国家标准(≤10μg/kg)。通过响应面优化微波结合酶法制备多肽工艺发挥协同作用,能有效降低米糠多肽AFB1含量满足国家标准限量要求。微波结合酶解处理条件温和,降解效率高、操作方便,可推广应用于受黄曲霉毒素的降解的粮油制备中。  相似文献

以琯溪红肉蜜柚为原料研制果酱,对其中添加剂的配比进行优化。采用单因素试验研究白砂糖、柠檬酸、琼脂和β-环糊精的添加量对红肉蜜柚果酱感官评价和质构特性的影响,在此基础上通过Box-Behnken中心组合试验设计和响应面分析法,探究果酱中各项配料添加量及其交互作用对产品感官品质的影响。结果表明:红肉蜜柚果酱中各配料的最优配比为:以100 g鲜果浆为计算基准,白砂糖添加量60%、柠檬酸添加量0.2%、琼脂添加量0.3%、β-环糊精添加量0.7%,采用此配比制得的果酱,感官评分为89分,具有良好的胶凝性和涂抹性,口感细腻,酸甜适中,且具有红肉蜜柚特有的风味和色泽。  相似文献
