Rosehip extracts of 18 samples representing six taxa in the genus Rosa were evaluated for antioxidant activities by use of different test systems. The ferric‐reducing antioxidant power (FRAP) and Trolox‐equivalent antioxidant capacity (TEAC) of the crude extracts ranged from 983.4 to 2187.1 µmol FRAP g−1 dry matter and from 457.2 to 626.2 µmol TEAC g−1 dry matter. The high antioxidant capacity was related to high contents of phytonutrients. The overall mean of antioxidants was 23.23 mg g−1 total carotenoids and 76.26 mg g−1 total phenolics. The phenolic component made a major contribution to the total antioxidant activities in both assays (overall mean was 90.5% and 75.7%), whereas the ascorbate made a minor contribution (8.6% and 16.9%) and the lipophilic component made an even smaller one (0.9% and 7.3%). However, the lipophilic component was the most effective when the comparison was based on the ratio of antioxidant activity to content of antioxidants. The crude extracts exhibited 50.9% (46.6–60.3%) inhibitory effect against the lipid peroxidation induced by 2,2′‐azobis(2,4‐dimethylvaleronitrile) (AMVN) and 85.0% (80.1–90.2%) inhibition in 2,2′‐azobis(2‐amidinopropane)hydrochloride (AAPH) assay at a concentration of 250 µg ml −1. Ascorbate acted as an antioxidant in both peroxy radical‐induced lipid peroxidations, but as a pro‐oxidant in the metal ion‐induced lipid peroxidation. The crude extracts showed a large inhibitory effect in the ferric ion‐induced lipid peroxidation and caused 83.7% inhibition at a concentration of 25 µg ml −1 dried rosehip powder. © 2000 Society of Chemical Industry  相似文献

Edible mushrooms are a good source of antioxidants. Methanol extracts of mushrooms such as Pleurotus sp., Agaricus bisporus, Morchella esculenta, Boletus edulis (approx. 2 mg mL?1) showed a high 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) scavenging activity close to 90%. Water extracts showed even higher antioxidant activity. In this case, B. edulis, Lentinus edodes and Amanita cesarea showed the highest 2,2′‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulfonic acid (ABTS) scavenging activity at approx. 0.14 mg mL?1. Other mushrooms such as Lactarius deliciosus and Cantharellus cibarius showed lower antioxidant activity in both extracts. Oxidative enzymes (peroxidases and polyphenol oxidases) present in the water fractions reduced their antioxidant activity by different extents since the phenols responsible for the antioxidant activity were not only those substrates of the oxidative enzymes. Other phenolic compounds and low‐molecular‐weight compounds were also involved in the antioxidant activity and differed depending on mushroom species. Total phenolic content (PC) was ~12 mg g?1 in lentils, 2.2 mg g?1 in chickpeas, 2.3 mg g?1 in soybeans, 2.5 mg g?1 in yellow peas and 1.2 mg g?1 in green peas. Total antioxidant activity (AA) determined by ABTS (2,2′‐azinobis‐3‐ethyl‐benzthiazoline‐6‐sulfonic acid) assay was highest in lentils at around 14 μmol Trolox equivalent antioxidant capacity (TEAC) g?1 and lowest in green peas at 1.9 μmol TEAC g?1. Bound phytochemicals contributed to 82–85% total AA in lentils. Free phytochemicals contributed more to total AA in chickpeas, yellow peas, green peas and soybeans than bound phytochemicals. AA and PC was reduced by ~80% in lentils and <30% in yellow peas by decortication, by 16–41% in lentils, chickpeas and peas by cooking, and by 22–42% in lentils by soaking. Total AA was significantly correlated with total PC. Soybeans had the greatest ability to scavenge free radicals, inhibit lipid peroxidation and chelate metals among the legumes tested. In an effort to discover new antioxidant natural compounds, wormwood (Artemisia absinthium L) an aromatic‐bitter herb, was screened. The sequential extraction was realized with five solvents of different polarities (70% methanol, petroleum ether, chloroform, ethyl acetate, n‐butanol). The antioxidative activity was tested by measuring their ability to scavenge stable 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) free radical and reactive hydroxyl radical during the Fenton reaction trapped by 5,5‐dimethyl‐1‐pyrroline‐N‐oxide (DMPO), using electron spin resonance (ESR) spectroscopy. Results demonstrated that the antiradical and antioxidative activity depend on the type and concentration of applied extracts and increased in the order ethyl acetate > methanol > n‐butanol > chloroform > petroleum ether > remaining water extracts. The investigation showed that the antiradical activity increased with increasing concentration of all extracts. The high contents of total phenolic compounds (25.6 mg g?1) and total flavonoids (13.06 mg g?1) indicated that these compounds contribute to the antiradical and antioxidative activity. In a model system, the formation of o‐semiquinone radicals from quercetin and chlorogenic acid was obtained to prove the mechanism (hydrogen donating and/or one‐electron reduction) of free‐radical scavenging activity. This study was conducted to obtain annatto extracts with both high antioxidant capacity and colour potential using solvents of different polarities (water, ethanol/water, ethanol, ethanol/ethyl acetate and ethyl acetate). The highest levels of total phenolic compounds were found in the water, ethanol/water and ethanol extracts (0.5 mg GAE mL?1), and the highest level of bixin was found in the ethanol/ethyl acetate extract (5.2 mg mL?1), which was characterised as the reddest and the most vivid one (a* = 40.5, = 46.1, C* = 58.4). The ethanol/ethyl acetate extract also showed the highest antioxidant activity (4.7 μm TEAC mL?1) and the highest percentage of tryptophan protection against singlet oxygen (63.6%). On the other hand, ethyl acetate and ethanol/water were the least effective solvents for the extraction of phenolic compounds and bixin, respectively. According to the multivariate statistical analysis, ethanol/ethyl acetate and ethyl acetate were the most promising solvents to obtain annatto extracts with both antioxidant and colour properties.  相似文献

Methanol extracts of olive pomace (two‐phase olive oil extraction) and olive pulp were analysed by reverse phase HPLC and the eluted fractions were characterised by electrospray ionisation mass spectrometry. This technique allowed the identification of some common phenolic compounds, namely, verbascoside, rutin, caffeoyl‐quinic acid, luteolin‐4‐glucoside and 11‐methyl‐oleoside. Hydroxytyrosol‐1′‐β‐glucoside, luteolin‐7‐rutinoside and oleoside were also detected. Moreover, this technique enabled the identification, for the first time in Olea europaea tissues, of two oleoside derivatives, 6′‐β‐glucopyranosyl‐oleoside and 6′‐β‐rhamnopyranosyl‐oleoside, and of 10‐hydroxy‐oleuropein. Also, an oleuropein glucoside that had previously been identified in olive leaves was now detected in olive fruit, both in olive pulp and olive pomace. With the exception of oleoside and oleuropein, the majority of phenolic compounds were found to occur in equivalent amounts in olive pulp and olive pomace. Oleoside was the main phenolic compound in olive pulp (31.6 mg g?1) but was reduced to 3.6 mg g?1 in olive pomace, and oleuropein (2.7 mg g?1 in the pulp) almost disappeared (<0.1 mg g?1 in the pomace). This study reported the chemical composition, phenolic content, antioxidant and anti‐lipase activity of oregano and Lippia essential oils. The major compounds found in oregano essential oil were γ‐terpinene (32.10%), α‐terpinene (15.10%), p‐cymene (8.00%) and thymol (8.00%). In Lippia essential oil, α‐limonene (76.80%) and 1,8‐cineole (4.95%) represented the major compounds. Oregano essential oil had higher phenolic content (12.47 mg gallic acid mL?1) and DPPH scavenging activity (IC50 0.357 μg mL?1) than Lippia essential oil (7.94 mg gallic acid mL?1 and IC50 0.400 μg mL?1, respectively). Both essential oils had similar antioxidant indexes (about 1.2) determined by Rancimat. Moreover, oregano essential oil had also higher anti‐lipase activity (IC50 5.09 and 7.26 μg mL?1). Higher phenolic content in the essential oils was related with higher scavenging and anti‐lipase activities. Oregano and Lippia essential oils could be used as natural antioxidants on food products.  相似文献

Two anthocyanin extracts from purple sweet potato (PSP) were prepared by means of microwave baking (MB) and acidified electrolysed water (AEW) or 95% ethanol. The extraction yield in AEW (pH 3.0) was up to 35.0% nearly 2.5 times higher than in ethanol. When pH ≤ 3.0, the lower the pH values of the extracts in solution were, the darker the red extracts were. Total flavonoids, phenolic and monomeric anthocyanin contents in AEW extract were 132.13, 64.52 and 102.31 mg g?1, respectively, whose values were the similar to or slightly lower than those in ethanol extract. On the contrary, its total sugar content (61.31 mg g?1) was nearly five times higher than that of the ethanol extract. In vitro assay indicated that the scavenging capability of DPPH free radicals of AEW extracts (IC50 = 12.0 μg mL?1) was stronger than that of the ethanolic ones. The reducing power and inhibiting lipid peroxidation of the two extracts were similar. Thus, the new extraction of MB‐AEW described here was not only simple and low in cost, but also had much higher extraction yield. The anthocyanin extracts with a strong antioxidation and a stable red colour could be widely used as food colouring additives and anti‐ageing health foods.  相似文献

Six bean cultivars grown in southern Manitoba for 2 years were evaluated for variability in yield of millstreams and phenolic constituents. The ethanolic extract of bean cultivars and millstreams was screened for antioxidant activity using the β‐carotene‐linoleate and the 1, 1‐diphenyl‐2‐picrylhydrazyl (DPPH) in vitro model systems. Cultivar was the main source of variation for yield of millstreams, content of phenolic compounds and antioxidant activities. Phenolic compounds in cultivars varied from 3.3 to 16.6 mg catechin equivalent and from 0.15 to 0.32 mg cyanidin‐3‐glucoside equivalent g?1 bean for total phenolic and anthocyanin contents, respectively. The bean cultivars exhibited antioxidant activity (AA) of 10–46% inhibition of lipid peroxidation in the linoleate and 0.4–1.3 trolox equivalent antioxidant capacities (TEAC) in the DPPH model systems. The hull millstream with maximum concentration of phenolic compounds exhibited the strongest antioxidant activity of 383 µM trolox equivalent g?1 hull. Total phenolic content, alone or in combination with other phenolic constituents, is a potential candidate as a selection criterion for antioxidant activity in beans. The effective utilisation of Moringa oleiferia mature leaves (MOL) extract as an antioxidant in cooked goat meat patties during refrigerated storage was investigated, and its efficiency was evaluated against butylated hydroxytoluene (BHT). The extract exhibited high phenolic content (48.36 mg of gallic acid equivalent per g), flavonoid (31.42 mg g?1 of sample) being the major component. Moringa oleiferia mature leaves extract showed excellent antioxidant activity as determined by radical‐scavenging activity of 1, 1‐diphenyl 2 picrylhydrazyl (DPPH). The IC50 value of MOL extract for 2, 2‐diphenyl‐1‐picrylhydrazyl radical scavenging was 18.54 μg mL?1. Total phenolic content (as gallic acid equivalent) significantly (P < 0.05) increased from 285.56 in control to 379.45 in patties with MOL extract. MOL extract (0.1%) when added to meat was found to retard lipid peroxidation of cooked goat meat patties as measured by TBARS number during refrigerated storage. The increase in TBARS number in MOL extract–treated samples was very slow and remained lowest (0.53 mg malonaldehyde per kg sample) up to 15 days. The antioxidant activity of MOL extract was found to be comparable to BHT. Addition of MOL extract did not affect any of the sensory attributes of patties. The MOL extract at a level of 100 mg/100 g meat was sufficient to protect goat meat patties against oxidative rancidity for periods longer than the most commonly used synthetic antioxidant like BHT.  相似文献

BACKGROUND: Pinus halepensis (Aleppo pine) is a widespread tree that can be found in both natural and urban environments. A discrimination study based on the antioxidant compounds, antioxidant capacity and fatty acid (FA) profile of P. halepensis cones (PHC) and seeds (PHS) was performed. RESULTS: The total amount of phenols was about 72‐fold higher in PHC extract than in PHS extract (P < 0.001). Anthocyanin and carotenoid contents were 10‐ and 12‐fold higher respectively in PHC extract. PHC and PHS extracts at a concentration of 1 mg mL?1 differed significantly in free radical‐scavenging activity on 2,2‐diphenyl‐1‐picrylhydrazyl radical (DPPH?) (86.65 vs 16.97%). PHC had higher antioxidant ability on 2,2′‐azino‐bis(3‐ethylbenzothialozine‐6‐sulfonic acid) radical cation (ABTS?+) than PHS (EC50 0.368 vs 2.345 mg mL?1). The FA profile of PHC oil revealed its richness in saturated FAs (41.5%) and high levels of trans FA isomers, with a predominance of trans,trans ‐linoleic acid (4.74%). However, polyunsaturated FAs in PHS oil represented more than 64% of total FAs. CONCLUSION: PHC showed important antioxidant activities as well as high levels of bioactive compounds. Thus PHC is a potential source of natural antioxidants that may afford several health benefits. However, the lipid extract of PHS seems to have more nutritional value as a polyunsaturated oil than that of PHC, which is high in saturated and trans FAs. BACKGROUND: Hydrolysates prepared from sea urchin (Strongylocentrotus nudus) gonad by enzymatic treatment showed strong 1,1‐diphenyl‐2‐picrylhydrazyl radical scavenging activity and reducing power. RESULTS: Hydrolysis of S. nudus gonad by the commercial protease papain was optimised for maximum degree of hydrolysis (DH) and trichloroacetic acid‐soluble peptide index (TCA‐SPI) using response surface methodology. Results showed that the optimal conditions were the following: temperature of 48.83 °C, pH of 6.92, enzyme‐to‐substrate ratio of 3143 U g?1, and substrate concentration of 83.5 g L?1. Under these conditions, a DH of 27.96 ± 0.54% and a TCA‐SPI of 57.32 ± 0.63% were obtained. The hydrolysate prepared in the optimal conditions was fractionated by an ultra‐filtration system and the resultant fraction below 10 kDa was found to effectively scavenge hydroxyl radical (EC50 = 13.29 ± 0.33 mg mL?1) and hydrogen peroxide (EC50 = 16.40 ± 0.37 mg mL?1), inhibit lipid peroxidation (EC50 = 11.05 ± 0.62 mg mL?1), chelate Fe2+ (EC50 = 7.26 ± 0.44 mg mL?1), and protect mice macrophages against death induced by tert‐butyl hydroperoxide. The study presents changes in the phenolic levels, antioxidant and anti‐inflammatory potential of purple basil leaves caused by different chemical elicitors: arachidonic acid (AA), jasmonic acid (JA) and β‐aminobutyric acid (BABA). The application of the all tested elicitors increased the concentration of phenolic compounds, including flavonoids and phenolic acids; especially, in comparison with control (457.62 μg g?1 FW), the rosmarinic acid level significantly increased after AA and JA treatment ‐ 705.0 and 596.5 μg g?1 FW, respectively. Phenolics from AA‐elicited plants showed the highest anti‐inflammatory activities designated as lipoxygenase (EC50 = 1.67 mg FW mL?1) and cyclooxygenase inhibition (EC50 = 0.31 mg FW mL?1). Elicitors' treatments (especially AA and JA) may be a very useful biochemical tool for improving the production of phenolic compounds in purple basil leaves.  相似文献

Black rice bran was fermented with Bacillus subtilis KU3 isolated from Korean traditional food, Kimchi. Antioxidant and cytotoxic activities of the fermented black rice bran were investigated. Total phenolic and anthocyanin contents decreased from 171.54 mg GAE g?1 and 2.31 mg g?1 to 139.13 mg GAE g?1 and 2.12 mg g?1, respectively, after fermentation. Antioxidant activities determined by 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging, β‐carotene bleaching and ferric thiocyanate assay were correlated with total phenolic and anthocyanin contents. Non‐fermented black rice bran extract (NFBE) showed greater antioxidant activities than fermented black rice bran extract (FBE). Cytotoxic activities measured by MTT assay showed that both NFBE and FBE had over 50% activities. The cytotoxic activities of FBE against MCF‐7 and HeLa cells were 71.65% and 68.07%, respectively, at 8.0 mg mL?1, but those of NFBE were lower than 50%. These results suggested that the cytotoxic activity of black rice bran improved through fermentation, while antioxidant activity reduced.  相似文献

The potential anti‐adipogenic effects of gluten‐free soy breads made from germinated soybean (GS), steamed soybean (SS) or roasted soybean (RS) were evaluated in an in vitro adipocyte cell model. GS and RS increased the total phenolic (TP) and total flavonoid (TF) contents of flours compared with the raw soybean (NS) flour. RS and GS had the highest TP (1.04 GAE mg g?1) and TF (0.92 CAE mg g?1) contents. Baking increased the TP content of breads, 0.09–0.26 GAE mg g?1, compared with the flours. Fermentation during breadmaking increased the DPPH scavenging activities compared with the flours. The ABTS scavenging exhibited similar patterns to those of DPPH. Lipid accumulation in 3T3‐L1 cells shows that the alcoholic extracts (100 μg mL?1) of SS flour and bread decreased adipocyte differentiation by 1.6‐ and 2.1‐fold, respectively, compared with control. SS bread extract substantially downregulated the adipogenesis‐related genes such as acetyl‐CoA carboxylase and glycerol‐3‐phosphate dehydrogenase.  相似文献

The objective of this work was to investigate the antioxidant and antibacterial activities of different extracts from Rabdosia rubescens and to further evaluate the antibacterial mechanism of extracts. The results showed that 80% acetone extracts had the highest contents of total polyphenols (8.09 mg GAE g?1) and flavonoids (5.69 mg RE g?1) and exhibited the strongest antioxidant activities, followed by 80% methanol and 80% ethanol, and the lowest for hexane extracts. Others except for hexane extracts showed different antibacterial activities against Gram‐positive strains, while no inhibitory effects were found on tested Gram‐negative bacterial strains. Among these extracts, 80% acetone and ethanol extracts had relatively higher antibacterial activities with the lowest minimum inhibitory concentration and minimum bactericidal concentration values of 5 and 10 mg mL?1. The antibacterial mechanism of ethanol extracts against Staphylococcus aureus might be described as it disrupts cell wall, increases cell membrane permeability and then leads to the leakage of cell constituents.  相似文献

Organic wastes as well as their ethanolic, water and heptane extracts and fermented products derived from herbs, fruit and vegetables obtained worldwide from food and pharmaceutical industries were investigated regarding their antioxidant activities using different methods [1.1‐diphenyl‐2‐picrylhydrazyl, FeIII‐reduction method and photochemiluminescence (PCL)]. The highest antioxidative activities were exhibited by different products of grape seed wastes (350–830 mg rosmarinic acid equivalent g?1 dried material = rae g?1 dm) followed by ethanolic extracts of blueberry (475 mg rae g?1 dm), larch (219 mg rae g?1 dm), willow (193 mg rae g?1 dm) and sunflower presscake (154 mg rae g?1 dm). Ethanolic extracts of the residues showed comparatively higher activity than water extracts. The least antioxidative capacities were found in heptane extracts of serenoa products (0.4; 0.6 mg rae g?1 dm) and fermented waste of carrots (1.6 mg rae g?1 dm). Correlating different assay methods, the strongest correlation was found between FeIII‐reduction and DPPH‐method (correlation coefficient r2 = 0.99**) and the weakest between FeIII‐reduction and PCL method (r2 = 0.57**) for raw materials.  相似文献

BACKGROUND: Recently much attention has been paid to biologically active plants because of their low production cost and fewer adverse effects compared with chemical drugs. In the present investigation the bioactivity of Phyllanthus niruri ethanol and aqueous extracts was evaluated in vitro. RESULTS: The ethanol extract of P. niruri showed a high level of flavonoid content (123.9 ± 0.002 mg g?1), while the aqueous extract showed the highest 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH; IC506.85 ± 1.80 µmol L?1) and 2,2′‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulfonic acid) (ABTS; 46.44 ± 0.53 µmol L?1) free radical scavenging activities with high phenol content (376 ± 0.02 mg g?1) and elevated levels of ferric reducing antioxidant power (FRAP; 23 883 ± 0.019 mmol g?1) with excellent antibacterial activity against Staphylococcus aureus (20 mm inhibition zone) and Streptococcus agalactiae (12 mm inhibition zone), respectively, in addition to the best immune activation potential of human peripheral blood mononuclear cells (450.5%). CONCLUSIONS: It is clear from our results that both extracts of P. niruri has excellent bioactivity roles via elevated levels of antibacterial, antioxidant and percentage of peripheral blood mononuclear cell proliferation, which could lead to the development of medications for clinical use. BACKGROUND: Total phenolics content, antioxidant activity and cytotoxicity of the methanol extracts from leaf parts of 13 Korean traditional salad plants were investigated in order to determine their properties. RESULTS: The highest phenolics content (mg ferulic acid equivalents kg?1 dry weight (d.w.), omit one) was found in methanol extracts from Polygonum aviculare, at 293.7 ± 6.0, followed by Euonymus alatus, at 250.7 ± 3.3, Saxifraga stolonifera, at 125.0 ± 8.1 and Ligularia fischeri, at 122.5 ± 5.9. The methanol plant extracts dose‐dependently increased free radical scavenging activity. Methanol extracts of Polygonum aviculare, Euonymus alatus and Saxifraga stolonifera, at 31 mg kg?1, exhibited the highest 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging activity (%) by 90.8 ± 4.2, 85.7 ± 3.9 and 64.1 ± 3.2, respectively. According to 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) assay, the methanol extracts from Portulaca oleracea (IC50 < 25.0 µg mL?1) showed the highest cytotoxicity against Calu‐6, followed by Plantago asiatica (49.2 µg mL?1) and Osmunda japonica (89.6 µg mL?1). CONCLUSION: Total phenolics content of the tested plant extracts was correlated with the DPPH radical scavenging activity, suggesting the phenolics compounds are contributing to the antioxidant properties of Korean salad plants. The leaf parts of the 13 Korean traditional salad plants described here that are currently used as foods may also provide some benefit to human health, and research into their potential benefits as preventative and/or therapeutic agents is warranted. The radical scavenging activity, reducing power and phenolic composition of pomegranate rind powder extract (RP) were determined and antioxidant properties of RP was evaluated in cooked chicken patties compared with vitamin C (VC) during refrigerated storage. Freshly minced chicken meat were assigned to one of the following six treatments: control (meat without any antioxidant); RP 5, RP 10, RP 15 and RP 20 (5, 10, 15 and 20 mg equivalent RP phenolics 100 g?1 meat, respectively) and VC 50 (50 mg VC 100 g?1 meat). The RP exhibited significantly (P < 0.05) higher reducing power and 1,1‐diphenyl‐2‐picrylhydrazyl radical scavenging activity. Incorporation of RP into chicken patties significantly (P < 0.05) reduced the HunterLab L* values compared with control and VC patties. Total phenolic content (as tannic acid equivalent) significantly (P < 0.05) increased from 308 in control to 441 μg g?1 in RP 20 patties. Addition of RP to chicken patties did not affect any of the sensory attributes. The values of thiobarbituric acid reactive substances were significantly (P < 0.05) reduced from 1.530 in control patties to 0.135 mg malonaldehyde kg?1 samples in RP patties. Pomegranate rind powder extract treatment (RP 10, RP 15 and RP 20) substantially inhibited (P < 0.05) lipid oxidation in cooked chicken patties to a much greater extent than VC treatment. Therefore, pomegranate rind powder can be utilized as an excellent natural antioxidant source.  相似文献
