Dihydroartemisinin (DHA)‐loaded methoxy poly(ethylene glycol)/poly(l ‐lactic acid) (mPEG5000—PLLA3200) amphiphilic block copolymeric micelles (DHA‐CM) have been prepared using modified solvent evaporation method. Physicochemical properties of DHA‐CM were investigated by using dynamic light scattering, transmission electron microscopy, high‐performance liquid chromatography, and Fourier transform infrared. Polymers formed stable, spherical, and worm‐like micelles with mean sizes smaller than 130 nm. In vitro release experiments revealed that DHA‐CM provided a more solubilizing effect than DHA suspension; in addition, it was showed that drug release profiles highly depended on pH values of dissolution media. Various types of lyoprotectants were tested to improve the redispersion performance of the freeze‐dried products. 3‐(4, 5‐dimethyl‐ thiazol‐2‐yl)‐2, 5‐diphenyl tetrazolium bromide assay was used to evaluate the cytotoxicity of micellar solutions of freeze‐dried DHA‐CM. The results showed that the IC50 values of DHA‐CM and DHA suspension for KB cell lines were 18.70 and 24.55 μM, respectively. In recent years, much attention has been given to the development of specialty polymers from useful materials. In this context, amphiphilic block copolymers were prepared by atom transfer radical polymerization (ATRP) of N‐phenylmaleimide (N‐PhMI) or styrene using a poly(2‐hydroxyethylmethacrylate)‐Cl macroinitiator/CuBr/bipyridine initiating system. The macroinitiator P(HEMA)‐Cl was directly prepared in toluene by reverse ATRP using BPO/FeCl3 6 H2O/PPh3 as initiating system. The microstructure of the block copolymers were characterized using FTIR, 1H‐NMR, 13C‐NMR spectroscopic techniques and scanning electron microscopy (SEM). The thermal behavior was studied by differential scanning calorimetry (DSC), and thermogravimetry (TG). The theoretical number average molecular weight (Mn,th) was calculated from the feed capacity. The microphotographs of the film's surfaces show that the film's top surfaces were generally smooth. The TDT of the block copolymer P(HEMA)80b‐P(N‐PhMI)20 and P(HEMA)90b‐P(St)10 of about 290°C was also lower than that found for the macroi′nitiator poly(HEMA)‐Cl. The block copolymers exhibited only one Tg before thermal decomposition, which could be attributed to the low molar content of the N‐PhMI or St blocks respectively. This result also indicates that the phase behavior of the copolymers is predominately determined by the HEMA block. In this article, we describe the synthesis and solution properties of PEG‐b‐PTMC star block copolymers via ring‐opening polymerization (ROP) of trimethylene carbonate (TMC) monomer initiated at the hydroxyl end group of the core PEG using HCl Et2O as a monomer activator. The ROP of TMC was performed to synthesize PEG‐b‐PTMC star block copolymers with one, two, four, and eight arms. The PEG‐b‐PTMC star block copolymers with same ratio of between hydrophobic PTMC and hydrophilic PEG segments were obtained in quantitative yield and exhibited monomodal GPC curves. The amphiphilic PEG‐b‐PTMC star block copolymers formed spherical micelles with a core–shell structure in an aqueous phase. The mean hydrodynamic diameters of the micelles increased from 17 to 194 nm with increasing arm number. As arm number increased, the critical micelle concentration (CMC) of the PEG‐b‐PTMC star block copolymers increased from 3.1 × 10?3 to 21.1 × 10?3 mg/mL but the partition equilibrium constant, which is an indicator of the hydrophobicity of the micelles of the PEG‐b‐PTMC star block copolymers in aqueous media, decreased from 4.44 × 104 to 1.34 × 104. In conclusion, we confirmed that the PEG‐b‐PTMC star block copolymers form micelles and, hence, may be potential hydrophobic drug delivery vehicles. © 2008 Wiley Periodicals, Inc. A new series of amphiphilic graft‐copolymers, composed of poly(vinyl chloride) (PVC) backbones and poly(ethylene oxide) side chains, was synthesized by chemical modification of PVC. The synthesis was based on the reaction between chlorine in PVC (polymerization degree 700) and sodium salt of polyethylene glycol (PEG). PEGs with molecular weights of 200 and 600 were used. The graft polymers were characterized by IR and gel permeation chromatography and the molecular parameters such as the average numbers of grafting chains on the PVC backbones were calculated as well as the grafting percent. The molecular weights of PEG were found to influence the rate of the grafting reaction and the final degree of conversion. The maximum grafting percentage of the resulted polymers after the purification was ca. 34%, regardless of the molecular weight of PEG. The graft reaction of succinic anhydride onto poly(vinyl alcohol) (PVA) was catalyzed by p‐toluenesulfonic acid monohydrate in solid state. The infrared spectra and 1H‐NMR spectra confirmed that succinic anhydride was successfully grafted onto PVA backbone. The influences of reaction temperature, reaction time, the amount of succinic anhydride, and the amount of catalyst on the graft reaction were studied. Uncrosslinked PVA graft copolymer with grafting degree up to about 6.5% could be obtained under low reaction temperature, short reaction time, and low amount of catalyst, whereas crosslinked PVA with high gel content could be obtained under high reaction temperature, long reaction time, and high amount of catalyst. © 2006 Wiley Periodicals, Inc. Amphiphilic graft copolymers consisting of poly(phthalazinone ether sulfone ketone) (PPESK) backbones and poly(ethylene glycol) (PEG) side chains were synthesized via reaction of chloromethylated PPESK (CMPPESK) with a sodium alkoxide of methoxyl PEG (PEG‐ ONa). The reactive precursor, CMPPESK, was prepared by the chloromethylation of PPESK with chloromethylether (CME) using concentrated H2SO4 as reaction medium. FTIR spectroscopy, 1H‐NMR and Solid‐state 13C CP‐MAS NMR analysis confirmed the covalent linking of PEG with PPESK backbones. The PEG content in the graft copolymers from 1H‐NMR analysis varied from 21.0 to 37.2 wt %, which was approximately in agreement with that calculated from TGA tests. The graft products have good solubility in many aprotic polar solvents and can be slightly swelled by water and ethanol, but water insoluble. Contact angle measurements revealed that the hydrophilicity of PPESK was significantly improved by the introduction of PEG graft chains, indicating the graft copolymer is a potential hydrophilic additive for PPESK membranes. © 2007 Wiley Periodicals, Inc. Dimethyl terephthalate (DMT) and ethylene glycol (EG) were used for the preparation of poly(ethylene terephthalate) (PET), and poly(ethylene glycol) (PEG) was added as a soft segment to prepare a PET–PEG copolymer with a shape‐memory function. MWs of the PEG used were 200, 400, 600, and 1000 g/mol, and various molar ratios of EG and PEG were tried. Their tensile and shape‐memory properties were compared at various points. The glass‐transition and melting temperatures of PET–PEG copolymers decreased with increasing PEG molecular weight and content. A tensile test showed that the most ideal mechanical properties were obtained when the molar ratio of EG and PEG was set to 80:20 with 200 g/mol of PEG. The shape memory of the copolymer with maleic anhydride (MAH) as a crosslinking agent was also tested in terms of shape retention and shape recovery rate. The amount of MAH added was between 0.5 and 2.5 mol % with respect to DMT, and tensile properties and shape retention and recovery rate generally improved with increasing MAH. © 2002 Wiley Periodicals, Inc. The objective of this study was to prepare high molecular weight poly(ester‐anhydride)s by melt polycondensation. The polymerization procedure consisted of the preparation of carboxylic acid terminated poly(?‐caprolactone) prepolymers that were melt polymerized to poly(?‐caprolactone)s containing anhydride functions along the polymer backbone. Poly(?‐caprolactone) prepolymers were prepared using either 1,4‐butanediol or 4‐(hydroxymethyl)benzoic acid as initiators, yielding hydroxyl‐terminated intermediates that were then converted to carboxylic acid‐terminated prepolymers by reaction with succinic anhydride. Prepolymers were then allowed to react with an excess of acetic anhydride, followed by subsequent polycondensation to resulting high molecular weight poly(ester‐anhydride)s. Upon coupling of prepolymers, size exclusion chromatography analyses showed an increase from 3600 to 70,000 g/mol in number‐average molecular weight (Mn) for the 1,4‐butanediol initiated polymer, and an increase from 7200 to 68,000 g/mol for the 4‐(hydroxymethyl)benzoic acid‐initiated polymer. 4‐Hydroxybenzoic acid and adipic acid were also used as initiators in the preparation of poly(?‐caprolactone) prepolymers. The reaction mechanism of decomposition process and the kinetic parameters of the poly(n‐butyl methacrylate‐b‐styrene), poly(nButMA‐b‐St), diblock copolymer synthesized by atom transfer radical polymerization (ATRP) were investigated by thermogravimetric analysis (TGA) at different heating rates. TGA curves showed that the thermal decomposition occurred in one stage. The apparent activation energies of thermal decomposition for copolymer, as determined by the Kissinger's, Flynn–Wall–Ozawa and Tang methods, which does not require knowledge of the reaction mechanism (RM), were 112.52, 116.54, and 113.41 kJ/mol, respectively. The experimental results were compared with master plots, in the range of the Doyle approximation. Analysis of experimental results suggests that in the conversion range studied, 3–18%, the actual RM is an A2 sigmoidal type. © 2009 Wiley Periodicals, Inc. J Appl Polym Sci, 2009  相似文献

Asymmetric block copolymer based on regioregular poly(3‐hexyl thiophene) (P3HT) and poly(ethylene oxide) (PEO) was synthesized through Heck reactions. The addition of PEO block has no influence in the effective conjugation length of P3HT block and apparently provides colloidal stability for the formation of stable nanostructures. Introduction of poor solvent to good solvent containing P3HT‐b‐PEO will induce the crystallization‐driven assembly of the P3HT into cylindrical micelles with a P3HT core, owing to π–π stacking of the conjugated backbone of P3HT. The absorption spectra of the cylindrical micelles reveal a red shift as compared to the polymer in good solvent, indicating the extension of conjugation length with an improved π–π stacking of the polymer chains within the cylindrical micelles. Our results indicated that cylindrical micelles with varied diameter and length can be obtained when solvent properties were varied using several different binary solvent mixtures. More interestingly, we demonstrate that ultrasonic processing can fragment the cylindrical micelles only when the ratio of poor solvent increases. This provides a facile and effective way to fabricate cylindrical micelles for applications in the area of polymer solar cell as well as organic optoelectronics device. © 2014 Wiley Periodicals, Inc. Methoxy poly (ethylene glycol) -poly (D, L-lactide) block copolymers (PEG-PLA) were prepared through ring-opening polymerization. The oil in water suspension method was used to prepare block copolymer micelles. The critical micelle concentration (CMC) by fluorescence spectroscopy was 0. 0056 mg· ml^- 1. The physical state of the inner core region of micelles was characterized with ^1 HNMR. The size of indomethacin (IMC) loaded micelles measured by dynamic light scattering (DLS) showed narrow monodisperse size distribution and the average diameters were less than 50 nm. In addition, the nanoparticles with relatively high drug loading content (DLC) were obtained.  相似文献

The spherulitic growth rates of a series poly (?‐caprolactone) homopolymers and poly(?‐caprolactone)‐b‐ poly(ethylene glycol) (PCL‐b‐PEG) block copolymers with different molecular weights but narrow polydispersity were studied. The results show that for both PCL homopolymers and PCL‐b‐PEG block copolymers, the spherulitic growth rate first increases with molecular weight and reaches a maximum, then decreases as molecular weight increases. Crystallization temperature has greater influence on the spherulitic growth rate of polymers with higher molecular weight. Hoffman–Lauritzen theory was used to analyze spherulitic growth kinetics and the free energy of the folding surface (σe) was derived. It is found that the values of σe decrease with molecular weight at low molecular weight level and become constant for high molecular weight polymers. Block copolymers, polystyrene‐b‐poly(styrene‐co‐maleic anhydride), have been prepared by reversible addition‐fragmentation chain transfer (RAFT) polymerization technique using three different approaches: 1‐phenylethyl phenyldithioacetate (PEPDTA) directly as RAFT agent, mediated polystyrene (PS) block as the macromolecular PS‐RAFT agent and mediated poly(styrene‐maleic anhydride) (SMA) block with alternating sequence as the macromolecular SMA‐RAFT agent. Copolymers synthesized in the one‐step method using PEPDTA as RAFT agent possess one PS block and one SMA block with gradient structure. When the macromolecular RAFT agents are employed, copolymers with one PS block and one alternating SMA block can be produced. However, block copolymers with narrow molecular weight distribution (MWD) can only be obtained using the PS‐RAFT agent. A series of copolymers of poly(2‐methacryloyloxyethyl phosphorylcholine)‐b‐poly(butylene succinate)‐b‐poly(2‐methacryloyloxyethyl phosphorylcholine) (PMPC‐b‐PBS‐b‐PMPC) were synthesized by atom transfer radical polymerization. The structure of the polymers was characterized by 1H NMR and infrared spectroscopy, and their thermal properties were described using TGA and DSC. In aqueous solutions, the PMPC‐b‐PBS‐b‐PMPC could form micelles with sizes ranging from 108 to 170 nm. In vitro release studies showed that acidic media and a longer PMPC chain benefited doxorubicin (DOX) release. 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) assays indicated that the micelles had low cytotoxicity to HeLa and L929 cells. DOX‐loaded micelles exhibited high cytotoxicity to HeLa cells. Flow cytometry results demonstrated that DOX‐loaded micelles could be internalized by HeLa cells. The in vitro phagocytosis results showed 3.9‐fold and 5.5‐fold reductions compared with poly(lactic acid) (PLA) nanoparticles and PDS55 micelles. These results demonstrate that PMPC‐b‐PBS‐b‐PMPC block copolymer micelles have great promise for cancer therapy. © 2017 Society of Chemical Industry  相似文献

Well‐defined poly(l ‐lactide‐b‐ethylene brassylate‐b‐l ‐lactide) (PLLA‐b‐PEB‐b‐PLLA) triblock copolymer was synthesized by using double hydroxyl‐terminated PEBs with different molecular weights. Gel permeation chromatography and NMR characterization were employed to confirm the structure and composition of the triblock copolymers. DSC, wide‐angle X‐ray diffraction, TGA and polarized optical microscopy were also employed to demonstrate the relationship between the composition and properties. According to the DSC curves, the cold crystallization peak vanished gradually with decrease of the PLLA block, illustrating that the relatively smaller content of PLLA may lead to the formation of a deficient PLLA type crystal, leading to a decrease of melting enthalpy and melting temperature. Multi‐step thermal decompositions were determined by TGA, and the PEB unit exhibited much better thermal stability than the PLLA unit. Polarized optical microscopy images of all the triblock samples showed that spherulites which develop radially and with an extinction pattern in the form of a Maltese cross exhibit no ring bond. The growth rate of the spherulites of all triblock samples was investigated. The crystallization capacity of PLLA improved with incorporation of PLLA, which accords with the DSC and wide‐angle X‐ray diffraction results. © 2019 Society of Chemical Industry  相似文献

We synthesized a methoxy poly(ethylene glycol) grafted chitosan (ChitoPEG) copolymer to prepare a retinoic acid (RA) encapsulated polymeric micelle. The RA‐encapsulated polymeric micelle of the ChitoPEG copolymer had a particle size of 100–500 nm and a spherical shape when observed by transmission electron microscopy. In a 1H‐NMR study, the specific peaks of RA and chitosan as a drug‐carrying inner core disappeared in deuterium oxide, and only the specific peak of methoxy poly (ethylene glycol) (MPEG) was observed, whereas specific peaks of MPEG, RA, and chitosan appeared in dimethyl sulfoxide. This indicated that the RA/ChitoPEG ion complexes were composed of a polymeric micelle with a core–shell structure and that free drug did not exist in the polymeric micelle formulations. Other evidence of drug incorporation into the polymeric micelle was witnessed in a differential scanning calorimetry analysis. The melting peaks of RA and chitosan were 182 and 220°C, respectively. The melting peak of the polymeric micelle was 200°C, whereas the melting peaks of the physical mixtures were those of both RA and the ChitoPEG copolymer. Functional poly(carbonate‐b‐ester)s were synthesized in buck by ring‐opening polymerization of the carbonate (TMC, MBC, or BMC) with tert‐butyl N‐(2‐hydroxyethyl) carbamate as an initiator, and then with ε‐CL (or ε‐BCL) comonomer. Subsequently, the PMMC‐b‐PCL with pendent carboxyl groups and the PTMC‐b‐PHCL with pendent hydroxyl groups were obtained by catalytic debenzylation. DSC analysis indicated that only one Tg at an intermediate temperature the Tgs of the two polymer blocks. A decrease Tg was observed when an increase contents of ε‐CL incorporated into the copolymers. In contrast, two increased Tms were observed with increasing PCL content. The block copolymers formed micelle in aqueous phase with critical micelle concentrations (cmcs) in the range of 2.23–14.6 mg/L and with the mean hydrodynamic diameters in the range of 100–280 nm, depending on the composition of copolymers. A polyblend of poly(ortho esters)–poly(ethylene glycol) (POE–PEG) was prepared. The release behavior of the acetanilide‐loaded film of the POE–PEG polyblend was studied. Blending POE with water‐soluble PEG can promote the release of drug in pH 7.4 PBS buffer at 37°C, while POE has plasticizing effect on PEG. Infrared and X‐ray diffraction studies reveal that there is some interaction between POE and acetanilide. Novel diblock copolymers designed for the preparation of micellar drug delivery systems, consisting of hydrophobic poly(allyl glycidyl ether) (PAGE) and hydrophilic poly(ethylene oxide) (PEO), were prepared, and their self‐assembly into micellar structures was studied. Copolymers differing in the length of the polymer blocks were purified and characterized. These amphiphilic copolymers with narrow molecular weight distributions were prepared through the anionic polymerization of allyl glycidyl ether with PEO monomethyl ether sodium salt as the macroinitiator. The PAGE–PEO copolymer readily formed small micelles with narrow size distributions via simple dissolution in water. The addition of pendant double bonds to the hydrophobic part of the chain was intended for further covalent modifications. Catalytic hydrogenation, the radical crosslinking of the micelle core, and the addition of thiol to double bonds of the copolymer were examples of such modifications that were proved to proceed with a quantitative yield for this copolymer. © 2004 Wiley Periodicals, Inc. Physical blends of poly(ethylene terephthalate) (PET) and poly(ethylene isophthalate) (PEI), abbreviated PET/PEI (80/20) blends, and of PET and a random poly(ethylene terephthalate‐co‐isophthalate) copolymer containing 40% ethylene isophthalate (PET60I40), abbreviated PET/PET60I40 (50/50) blends, were melt‐mixed at 270°C for different reactive blending times to give a series of copolymers containing 20 mol % of ethylene isophthalic units with different degrees of randomness. 13C‐NMR spectroscopy precisely determined the microstructure of the blends. The thermal and mechanical properties of the blends were evaluated by DSC and tensile assays, and the obtained results were compared with those obtained for PET and a statistically random PETI copolymer with the same composition. The microstructure of the blends gradually changed from a physical blend into a block copolymer, and finally into a random copolymer with the advance of transreaction time. The melting temperature and enthalpy of the blends decreased with the progress of melt‐mixing. Isothermal crystallization studies carried out on molten samples revealed the same trend for the crystallization rate. The effect of reaction time on crystallizability was more pronounced in the case of the PET/PET60I40 (50/50) blends. The Young's modulus of the melt‐mixed blends was comparable to that of PET, whereas the maximum tensile stress decreased with respect to that of PET.
