A new method was developed for the determination of quinoxaline-2-carboxylic acid, the marker residue of carbadox, in the edible tissues of food-producing animals using a colloidal gold probe-based immunochromatographic assay. The highly specific polyclonal antibody (PcAb), which was very sensitive to N-butylquinoxaline-2-carboxylic acid (BQCA) with an IC(50) value of 2.38 ng ml(-1), was selected for the development of an immunochromatographic assay (ICA). Only 5 min were required to perform this assay; it had a visual detection limit of 25 ng g(-1) for quinoxaline-2-carboxylic acid. The results of the analysis of quinoxaline-2-carboxylic acid in animal tissues using the immunochromatographic assay showed good agreement with those obtained by HPLC. In conclusion, the method was rapid and accurate for screening residues of carbadox in the edible tissues of food-producing animals.  相似文献

The feed drug additive carbadox is a suspected carcinogen and mutagen. To monitor effectively residues of carbadox in the edible tissues of food-producing animals, an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) to detect quinoxaline-2-carboxylic acid, the marker residue of carbadox, was developed. Several haptens were synthesised and conjugated to the carrier protein. Nine female New Zealand white rabbits were immunised with the immunising conjugates to produce polyclonal antibodies according to the designed schemes of immunisation. The highly specific antibody that was very sensitive to N-butylquinoxaline-2-carboxamide with an IC50 value of 7.75?µg?l?1 was selected for the development of an ic-ELISA. The standard curves based on the N-butylquinoxaline-2-carboxamide matrix calibration ranged from 0.2 to 51.2?µg?l?1. The decision limit and detection capability of the ic-ELISA were 0.60 and 0.83?µg?kg?1 for liver and 0.68 and 0.79?µg?kg?1 for muscle of swine, respectively. The recoveries were 57–108% with coefficients of variation of less than 20% when the quinoxaline-2-carboxylic acid was spiked into liver and muscle with the concentrations of 1.0–20.0?µg?kg?1. Excellent correlations between the results of the ic-ELISA and an HPLC method (r?=?0.9956???0.9969) were observed for incurred tissues. These results suggest that the ic-ELISA is a sensitive, accurate and low-cost method that would be a useful tool for screening residues of carbadox in the edible tissues of food-producing animals.  相似文献

For the treatment of rabbit dysentery and bacterial enteritis, veterinary practitioners often adopt veterinary medicinal products authorised for other food-producing species, but in some cases non-authorised drugs frequently used in the past, such as carbadox and olaquindox, might be illegally adopted. To verify the carbadox and olaquindox distribution and persistence in rabbit tissues, two independent in vivo studies were carried out. In the first study, 24 healthy rabbits received water medicated with carbadox at 100 mg l?1 over a period 28 days, whereas in the second one, 24 healthy rabbits were administered water containing olaquindox at 100 mg l?1. In each study rabbits were randomly assigned to four groups to be sacrificed respectively at 0, 5, 10 and 20 days from treatment withdrawal, for depletion studies. A control group of six animals was adopted for control and as a reservoir of blank tissues. Muscle and liver samples collected from each treated animal were stored at ?20°C pending the analysis. Sensitive and robust liquid chromatography-tandem mass spectrometry analytical methods were set up for the parent compounds and their main metabolites quinoxaline-2-carboxylic acid, desoxycarbadox and 3-methylquinoxaline-2-carboxylic acid to verify their residual. Data collected demonstrate that the combination of liver as target matrix, quinoxaline-2-carboxylic acid and 3-methylquinoxaline-2-carboxylic acid as marker residue and enzymatic digestion is strategic to evidence carbadox and/or olaquindox illegal treatments in rabbits, even 20 days after treatment withdrawal at concentration levels higher than 0.5 µg kg?1. This findings suggests that liver should be proposed as target matrix for official control in national monitoring plan.  相似文献

The main objective of this paper was to assess the suitability of an enzyme-linked immunosorbent assay (ELISA) as a rapid and simple screening method for the detection of neonicotinoid insecticide dinotefuran residues in various vegetable samples including carrot, cabbage, green pepper, leek, Japanese mustard spinach (komatsuna), and spinach. Simple dilution of crude sample extracts was effective to circumvent or reduce matrix interference derived from samples. Consequently, ELISA was useful to determine dinotefuran accurately and directly, only requiring methanol extraction and dilution with water. The theoretically estimated limit of detection of dinotefuran for tested samples ranged from 0.06 to 0.12?mg?kg?1. The ELISA gave recovery values close to 100% for all samples except komatsuna, which contained lower concentration levels, and the results of the ELISA agreed well with those obtained with conventional HPLC (r?>?0.99). Results show that the ELISA evaluated in this study is suitable for the rapid and simple screening method of dinotefuran residue in agricultural samples even without sample pre-treatment.  相似文献

Due to their potential adverse effects on human health, the use of carbadox and olaquindox in feedingstuffs was prohibited by the European Union since 1998. In this work, highly luminescent quantum dot beads (QBs) were synthesized by encapsulating CdSe/ZnS and used as novel fluorescent probes in the immunochromatographic assay (ICA) for simultaneous and quantitative determination of metabolites of olaquindox (3-methylquinoxaline-2-carboxylic acid, MQCA) and carbadox (quinoxaline-2-carboxylic, QCA). The fluorescence intensities of the test lines were recorded using a fluorescence strip reader. The 50% of inhibition for MQCA and QCA was shown to be 8.1 and 10.6 μg L?1, respectively. The whole assay process can be accomplished within 10 min. The immunosensor was used to analyze spiked samples, and analyte intra- and inter-assay recovery rates ranged from 78.7 to 92.2% for MQCA and 80.6 to 95.8% for QCA, and coefficients of variation were all below 12%. The incurred tissues samples were assayed using both QB-based ICA and commercial ELISA kit and were further confirmed with LC-MS/MS. The QB-based ICA results exhibited good agreement with both commercial ELISA and LC-MS/MS methods.  相似文献

Deoxynivalenol (DON) is a toxin produced by certain species of Fusarium fungi that can infest wheat, barley and corn. The fungi cause diseases in crops worldwide and some of the secondary metabolites, such as DON, can adversely affect animal health and food safety. To monitor DON in wheat rapidly, a biosensor using the principle of biolayer interferometry (BLI) was developed. The signal from the sensor was substantially amplified through the use of a primary antibody–colloidal gold conjugate. The amplification was much greater in the presence of wheat matrix than in buffered solution, suggesting matrix components may have contributed to the enhancement. The improved signal provided by the amplification allowed for the development of rapid qualitative and quantitative assays. The limit of detection of the method was 0.09?mg?kg?1; the limit of quantitation was 0.35?mg?kg?1. Recovery from wheat spiked over the range from 0.2 to 5?mg?kg?1 averaged 103% (RSD?=?12%). The quantitative assay compared favourably (r 2?=?0.9698) with a reference chromatographic method for 40 naturally contaminated wheats. The qualitative assay was able to classify accurately the same group of 40 samples as either above or below a 0.5?mg?kg?1 threshold. These results suggest that the BLI technique can be used to measure DON in wheat rapidly.  相似文献

An immunochromatographic (ICG) strip was developed for the simultaneous quantitative determination of five quinoxaline-1,4-dioxides in animal feed. For this purpose, polyclonal antibodies (PcAb) with group-specific quinoxaline-1,4-dioxides were conjugated to colloidal gold particles as the detection reagent for ICG strips to test for quinoxaline-1,4-dioxides. This method achieved semi-quantitative detection of quinoxaline-1,4-dioxides within 5–10 min. The visual lower detection limits of the strip for quinocetone, cyadox, carbadox, mequindox and olaquindox were 10, 15, 15, 20 and 20 ng ml?1, respectively. Using an ICG strip reader, the 50% inhibitions (IC50 values) were calculated to be 9.1, 13.5, 16.6, 20.2 and 21.3 ng ml?1 for quinocetone, cyadox, carbadox, mequindox and olaquindox, respectively. When used to analyse samples of animal feed, acceptable recovery rates of 77.5–99.5% and coefficients of variation (CVs) of 4.3–10.7% were obtained. Levels measured with the ICG strip for 10 spiked samples were confirmed by HPLC with a high correlation coefficient of 0.9965 (n = 10). In conclusion, the method was rapid and accurate for simultaneous determination of five quinoxaline-1,4-dioxides antibiotics in animal feed.  相似文献

Zilpaterol is a β-adrenergic agonist feed additive approved in the United States to increase weight gain and improve feed efficiency of cattle. A zilpaterol immunochromatographic assay was developed as an economical and user-friendly rapid detection method for zilpaterol and validated using urine and tissue samples derived from animal studies. The assay sensitivity was 1.7–23.2 ng g?1 or mL?1 across a variety of feed and animal matrices and did not cross-react with clenbuterol or ractopamine. No sample pre-treatment of cattle and sheep urine was needed, but horse urine and feed required dilution; skeletal muscle required solvent extraction prior to testing. Of 32 incurred sheep urine samples tested, zilpaterol content was correctly identified in all but 2 samples. Horse urine containing >10 ng mL?1 of incurred zilpaterol residue (n = 48) was correctly identified as zilpaterol positive. The assay correctly identified 0-day withdrawal sheep muscle samples as zilpaterol positive and the control and longer withdrawal day sheep muscle samples as negative. Zilpaterol was demonstrated to be stable in horse urine when stored at ?20°C for 7 years.  相似文献

Mass spectrometric methods were developed and validated for the analysis in chicken muscle of a range of antibiotic growth promoters: spiramycin, tylosin, virginiamycin and bacitracin, and separately for two marker metabolites of carbadox (quinoxaline-2-carboxylic acid and 1,4-bisdesoxycarbadox), and a marker metabolite of olaquindox (3-methyl-quinoxaline-2-carboxylic acid). The use of these compounds as antibiotic growth promoters has been banned by the European Commission. This study aimed to develop methods to detect their residues in muscle samples as a means of checking for the use of these drugs during the rearing of broiler chickens. When fed growth-promoting doses for 6 days, spiramycin (31.4?µg?kg?1), tylosin (1.0?µg?kg?1), QCA (6.5?µg?kg?1), DCBX (71.2?µg?kg?1) and MQCA (0.2?µg?kg?1) could be detected in the muscle 0 days after the withdrawal of fortified feed. Only spiramycin could consistently be detected beyond a withdrawal period of 1?day. All analytes showed stability to a commercial cooking process, therefore raw or cooked muscle could be used for monitoring purposes.  相似文献

The monoclonal antibodies (mAbs) against ractopamine (Rac) were prepared and their properties identified by indirect competitive enzyme-linked immunoabsorbant assay (ELISA). The IC50 of mAbs was 2.7 ng ml?1 towards Rac or 9.3 ng ml?1 towards Rac-glucuronides and no cross-reactivity (CR) towards other competitors except dobutamine (CR: 3.76%). Based on the mAbs, the Rac-kit (kit) and Rac-strip (strip) were developed to detect Rac residues in swine urine. The strip and kit assay could be performed within 5–10 min and 2 h, respectively, allowing the analysis of urine samples without the need for sample clean-up. The detection limits were 1 ng ml?1 for kit and 3 ng ml?1 with the unaided eye, and 0.2 ng ml?1 with the Strip Reader for strip. The correlation coefficients (R 2) were 0.988 for kit in the range 0–128.0 ng ml?1, and 0.987 for strip in the range 0–10.8 ng ml?1. Comparing the gas chromatography-mass spectrometry (GC-MS) with the kit or strip in swine urine spiked with Rac standards, the differences ranged from 1.4% to 4.5% for kit and 1.0% to 4.7% for strip. However, the differences were greater than 54% for the kit and 55% for the strip test for the analysis of urine from swine treated with Rac. The results obtained from GC-MS using hydrolysed urine samples were generally in good agreement with those obtained from strip or kit using non-hydrolysed urine samples.  相似文献

Carbadox cannot be used in food-producing animals within the European Union following the adoption of Commission Regulation EC 2788/98/EC. Monitoring of the longest remaining residue—quinoxaline-2-carboxylic acid (QCA)—is the most effective way of enforcing the prohibition on its use. The study was under taken to determine if QCA could be passed from pig to pig following the exposure of unmedicated animals to housing that had previously contained medicated animals. Drug-withdrawal studies were also carried out on medicated animals. Distinction between treated animals and those exposed to QCA might be required by competent national authorities to determine whether a positive result for QCA in tissue is truly 'violative'. Comparison of the ratio concentrations of QCA in tissues and body fluids was made to determine if they could be used as criteria for discrimination between illegally treated animals and environmental contamination.  相似文献

Dexamethasone is one of a number of synthetic corticosteroids illegally used to promote growth in food-producing animals. Since these low-level drug cocktails evade detection by currently available chemical methods, simple biological indicators that can aid in laboratory analysis are needed. In an attempt to devise an accurate biological method that could detect illicit drug treatment in food-producing animals, we characterized microscopic morphologic alterations of the thymus in veal calves administered low-dose dexamethasone versus control animals. For this purpose, 122 male calves were farmed for 6 months in controlled condition: 81 animals were orally administered dexamethasone (0.4?mg?day?1) for 20 days during the sixth month and the remaining 41 were kept as control. Urine samples were collected systematically during the treatment period, the suspension period and at the slaughterhouse. All animals were slaughtered 10 per day starting from 10 days after the last dexamethasone administration and the thymus was sampled for histological examination. The difference between the two animal groups was evaluated by means of a non-parametric test of hypothesis. No residues were detected in the urines collected since the third day after the last administration, whereas morphometric analysis of the thoracic thymus revealed a significant decrease in the cortex:medulla ratio in the treated animals (p?<?0.0005). We can conclude that this histological approach offers encouraging prospects as a screening method to overcome current limitations in controlling growth promoter abuse.  相似文献

The ouabain/veratridine-dependent neuroblastoma (neuro-2a) cell-based assay (CBA) was applied for the determination of the presence of ciguatoxin (CTX)-like compounds in ciguatera-suspected fish samples caught in the Canary Islands. In order to avoid matrix interferences the maximal concentration of wet weight fish tissue exposed to the neuro-2a cells was set at 20?mg tissue equivalent?(TE)?ml?1 according to the sample preparation procedure applied. In the present study, the limit of quantification (LOQ) of CTX1B equivalents in fish extract was set at the limit of detection (LOD), being defined as the concentration of CTX1B equivalents inhibiting 20% cell viability (IC20). The LOQ was estimated as 0.0096?ng CTX1B eq.?g?TE?1 with 23–31% variability between experiments. These values were deemed sufficient even though quantification given at the IC50 (the concentration of CTX1B equivalents inhibiting 50% cell viability) is more accurate with a variability of 17–19% between experiments. Among the 13 fish samples tested, four fish samples were toxic to the neuro-2a cells with estimations of the content in CTX1B?g?1 of TE ranging from 0.058 (±0.012) to 6.23 (±0.713) ng CTX1B eq.?g?TE?1. The high sensitivity and specificity of the assay for CTX1B confirmed its suitability as a screening tool of CTX-like compounds in fish extracts at levels that may cause ciguatera fish poisoning. Species identification of fish samples by DNA sequence analysis was conducted in order to confirm tentatively the identity of ciguatera risk species and it revealed some evidence of inadvertent misidentification. Results presented in this study are a contribution to the standardisation of the neuro-2a CBA and to the risk analysis for ciguatera in the Canary Islands.  相似文献

An immunoassay-based lateral flow device for the quantitative determination of four major aflatoxins in maize has been developed. The one-step assay has performance comparably with that of other screening methods, as confirmed by the intra- and the inter-day precision of the data (RSD 10–22%), and can be completed in 10?min. Quantification was obtained by acquiring images of the strip and correlating intensities of the coloured lines with analyte concentration by means of a stored calibration curve carried out by diluting aflatoxins in the extract from a blank maize sample. Limit of detection (1?µg?kg–1) and dynamic range (2–40?µg?kg?1) allows the direct assessment of aflatoxin contamination in maize at all levels of regulatory relevance. All reagents are immobilized on the lateral flow device. In addition, very simple sample preparation, using an aqueous buffered solution, has been demonstrated to allow the quantitative extraction of aflatoxins. Twenty-five maize samples were extracted with the aqueous medium and analyzed by the developed assay. A good correlation was observed (y?=?0.97x?+?0.07, r 2?=?0.980) when data was compared with that obtained through an official method. The developed method is reliable, rapid and allows for application outside the laboratory as a point-of-use test for screening purposes.  相似文献

Lasalocid is a polyether ionophoric coccidiostat used for the prevention of coccidiosis in poultry at a prescribed concentration and during a certain time interval. Due to a public health concern about the presence of coccidiostat residues in poultry, the aim of the present study was to determine the levels of lasalocid residues in the edible tissues of broiler chickens (breast muscle, thigh muscle, heart, liver, gizzard, kidneys and skin/fat) fed commercially produced feed containing 100?mg?kg?1 of lasalocid in complete feed throughout the 5-day withdrawal period (WP). The residues were investigated by liquid chromatography coupled with electrospray ionisation (ESI) tandem mass spectrometry (MS/MS) with triple quadrupole. The limit of detection (LOD) and the limit of quantification (LOQ) of the method were 0.47 and 1.44?µg?kg?1, respectively. The average recovery based on the matrix-fortified calibrations for chicken tissues ranged between 79% and 98%. Lasalocid was found to accumulate in the liver, followed by the heart, skin/fat, kidneys, thigh muscle and gizzard. The lowest concentrations of lasalocid residues were found in the breast muscle. On day 5 of the WP, residue concentrations of lasalocid did not decline below the LOQ of the method, but were far below the maximum residue level (MRL) established for lasalocid in poultry from 20 to 100?µg kg?1 by European Commission Regulation (EU) No. 37/2010. The results confirmed that the WP established for lasalocid is sufficient to ensure the decline of its residues in the tissues of broiler chickens to the safe residue level.  相似文献

A rapid and sensitive enzyme-linked immunosorbent assay (ELISA) was applied for the qualitative screening analysis of dexamethasone, betamethasone, flumethasone, and prednisolone in milk and urine, and dexamethasone, flumethasone and prednisolone in liver samples at levels corresponding to the European Union maximum residue limit (MRL), or at required performance levels (RPLs) for substances for which there is no established MRL. Method validation was performed according to Commission Decision 2002/657/EC criteria established for qualitative screening methods. In this regard, the following parameters were determined: detection capability (CCβ), specificity, limit of detection (LOD), limit of quantitation (LOQ), recovery, within-laboratory reproducibility, linearity and ruggedness. LODs were 0.2, 1.2 and 0.6?µg?kg?1 in milk, urine and liver samples, and LOQ values were 0.3, 1.2 and 1.4?µg?kg?1 in milk, urine and liver, respectively. Recoveries from spiked samples ranged from 68% to 131% for dexamethasone, from 57% to 120% for flumethasone, from 60% to 155% for betamethasone, and from 23% to 32% for prednisolone, with a coefficient of variation (CV) between 1.6% and 21.2%. The CCβ value was below the MRL/RPL for all examined matrices. Moderate variations of some critical factors in the sample pre-treatment for liver and milk samples were deliberately introduced for ruggedness evaluation and did not result in any negative effects on corticosteroid detection. The proposed method is suitable for qualitative screening analysis of corticosteroids in the above-mentioned food in conformity with the current European Union performance requirements.  相似文献

为建立一种白酒香型鉴别方法,以DB-WAX为色谱柱,样品加标后直接采用气相色谱(GC)法进样分析。结果表明,香气成分线性相关系数R2>0.999,检测限<5.11 mg/L,精密度试验结果的相对标准偏差(RSD)<4.11%;平均回收率为80.20%~97.93%。采用偏最小二乘法(PLS)对不同香型白酒数据进行建模和预测,不同香型白酒样品在PLS得分图中能分开,利用训练集和验证集数据对模型进行验证,结果表明预测准确率较高,校正均方差(RMSEE)和预测均方差(RMSEP)值也说明PLS模型对不同白酒香型具有较强的预测性。该方法在判别白酒香型分析中具有较高的准确性。  相似文献

A specific and sensitive LC-MS/MS method was firstly established for the simultaneous extraction and determination of cyadox and its three main metabolites—1,4-bisdesoxycyadox, 4-desoxycyadox, and quinoxaline-2-carboxylic acid—in chicken muscle, liver, kidney, and fat tissues. Samples were subjected to extraction using ethyl acetate and followed by acetonitrile–chloroform (1:4, v/v) and further purified by Oasis mixed mode anion exchange SPE cartridge. Analysis was performed on a C18 column by detection with MS in multiple-reaction monitoring mode. A gradient elution program with 0.1 % formic acid solution, acetonitrile, and 1 % formic acid (adjusted to pH 8 with ammonia) was performed at a flow rate of 0.2 mL min?1. The correlation coefficients (r) for each calibration curve are higher than 0.99 within the experimental concentration range. The recoveries of the four target analytes at three spiking levels of 2.5, 25 and 250 μg kg?1 were between 74.5 and 93.8 %, with relative standard deviations less than 12 %. The decision limits (CCαs) of the four analytes in chicken edible tissues ranged from 0.3 to 1.5 μg kg?1, and the detection capabilities (CCβs) were below 2.3 μg kg?1. The developed method demonstrated a satisfactory applicability in incurred chicken tissue samples.  相似文献
