The high ingestion of oleic (OLA) and linoleic (LNA) acids by Western populations, the presence of inflammatory diseases in these populations, and the importance of neutrophils in the inflammatory process led us to investigate the effects of oral ingestion of unesterified OLA and LNA on rat neutrophil function. Pure OLA and LNA were administered by gavage over 10 days. The doses used (0.11, 0.22 and 0.44 g/kg of body weight) were based on the Western consumption of OLA and LNA. Neither fatty acid affected food, calorie or water intake. The fatty acids were not toxic to neutrophils as evaluated by cytometry using propidium iodide (membrane integrity and DNA fragmentation). Neutrophil migration in response to intraperitoneal injection of glycogen and in the air pouch assay, was elevated after administration of either OLA or LNA. This effect was associated with enhancement of rolling and increased release of the chemokine CINC-2αβ. Both fatty acids elevated l-selectin expression, whereas no effect on β2-integrin expression was observed, as evaluated by flow cytometry. LNA increased the production of proinflammatory cytokines (IL-1β and CINC-2αβ) by neutrophils after 4 h in culture and both fatty acids decreased the release of the same cytokines after 18 h. In conclusion, OLA and LNA modulate several functions of neutrophils and can influence the inflammatory process.  相似文献

Two studies were conducted to determine the effects of dietary cholesterol (CHO) and cholesterol oxides (COPs) on the development of atherosclerosis and the changes in fatty acid and blood characteristics in rabbits. In the first study, forty male New Zealand white rabbits were divided into 5 groups and fed commercial rabbit chow with no added CHO or COPs, 1 g CHO, 0.9 g CHO + 0.1 g COPs, 0.8 g CHO + 0.2 g COPs, or 0.5 g CHO + 0.5 g COPs per kg diet. In the second study, 24 male New Zealand White rabbits were divided into 3 groups and fed a diet containing 2 g CHO, 1.6 g CHO + 0.4 g COPs, or 1.2 g CHO + 0.8 g COPs per kg diet. All diets induced atherosclerotic lesions in the rabbits’ ascending thoracic aorta. The serum CHO and triglyceride levels (p < 0.05) increased significantly with the increased levels of CHO in the diets. Dietary CHO or COPs did not influence high-density lipoprotein CHO levels. The ratio of saturated fatty acid to unsaturated fatty acid increased as the level of dietary CHO and COPs increased.  相似文献

Probiotics are live organisms that are primarily used to improve gastrointestinal disorders such as diarrhea, irritable bowel syndrome, constipation, lactose intolerance, and to inhibit the excessive proliferation of pathogenic intestinal bacteria. However, recent studies have suggested that probiotics could have beneficial effects beyond gastrointestinal health, as they were found to improve certain metabolic disorders such as hypertension. Hypertension is caused by various factors and the predominant causes include an increase in cholesterol levels, incidence of diabetes, inconsistent modulation of renin and imbalanced sexual hormones. This review discusses the antihypertensive roles of probiotics via the improvement and/or treatment of lipid profiles, modulation of insulin resistance and sensitivity, the modulation of renin levels and also the conversion of bioactive phytoestrogens as an alternative replacement of sexual hormones such as estrogen and progesterone.  相似文献

Knowledge about the effects of dietary fats on subclinical inflammation and cardiovascular disease risk are mainly derived from studies conducted in Western populations. Little information is available on South East Asian countries. This current study investigated the chronic effects on serum inflammatory markers, lipids, and lipoproteins of three vegetable oils. Healthy, normolipidemic subjects (n = 41; 33 females, 8 males) completed a randomized, single-blind, crossover study. The subjects consumed high oleic palm olein (HOPO diet: 15% of energy 18:1n-9, 9% of energy 16:0), partially hydrogenated soybean oil (PHSO diet: 7% of energy 18:1n-9, 10% of energy 18:1 trans) and an unhydrogenated palm stearin (PST diet: 11% of energy 18:1n-9, 14% of energy 16:0). Each dietary period lasted 5 weeks with a 7 days washout period. The PHSO diet significantly increased serum concentrations of high sensitivity C-reactive protein compared to HOPO and PST diets (by 26, 23%, respectively; P < 0.05 for both) and significantly decreased interleukin-8 (IL-8) compared to PST diet (by 12%; P < 0.05). In particular PHSO diet, and also PST diet, significantly increased total:HDL cholesterol ratio compared to HOPO diet (by 23, 13%, respectively; P < 0.05), with the PST diet having a lesser effect than the PHSO diet (by 8%; P < 0.05). The use of vegetable oils in their natural state might be preferred over one that undergoes the process of hydrogenation in modulating blood lipids and inflammation.  相似文献

Low high density lipoprotein cholesterol (HDL-C) is a known risk factor of coronary artery disease. Apolipoprotein A1 (APOA1) is the most abundant component of HDL-C. This study aimed at identifying sequence variations (rare and common) in the APOA1 gene and its association with serum HDL-C levels. This study was conducted from April 2012 to February 2013 on 79 Tehranians (participants of Tehran Lipid and Glucose Study) with extremely low HDL-C (within the 5th percentile) and 63 individuals with extremely high HDL-C (within the 95th percentile) levels. After DNA amplification by PCR, DNA sequencing of all three exons and 700 bps of promoter region of the APOA1 gene was performed. Sequence results were analyzed and interpreted using the appropriate software and variants were identified. After sequencing 42 common and rare variants were identified, 11 of which were known variants and the others had been unreported so far. Of the exonic variants, 11 were missense, 6 were synonymous and 1 was nonsense. There was a significant association between serum HDL-C and variant of rs2070665 as well as variants Chr.11:116707788, Chr.11:116708059, Chr.11:116708036, Chr.11:116707729, rs201148448, Chr.11:116707018, Chr.11:116707801, Chr.11:116708530, Chr.11:116708088, rs121912724 and Chr.11:116706966 (p < 0.001). Variants Chr.11:116707018, rs121912724 and 2070665 were independent predictors of the HDL-C level (p < 0.001). SNP Chr.11:116707018 was the strongest predictor of the HDL-C level (OR 7.527, p < 0.001). This study identified 42 variants in APOA1 gene, 31 of which were new variants. Three variants of rs2070665, rs121912724 and Chr.11:116707018 could predict the HDL-C level independently. Variant rs2070665 was protective against low-HDL-C levels while variants rs121912724 and Chr.11:116707018 were risk factors for that in our population.  相似文献

This paper describes the inhibitory activities of cholesterol derivatives such as cholesterol, sodium cholesteryl sulfate, cholesteryl-5α, 6α-epoxide, cholesteryl chloride, cholesteryl bromide, and cholesteryl hemisuccinate (compounds 16, respectively) against DNA polymerase (pol), DNA topoisomerase (topo), and human cancer cell growth. Among the compounds tested, compounds 2 and 6 revealed themselves to be potent inhibitors of animal pols, and the IC50 values for pols were 0.84–11.6 and 2.9–148 μM, respectively. Compounds 2, 3 and 6 inhibited the activity of human topo II, with IC50 values of 5.0, 12.5 and 120 μM, respectively. Compounds 2, 3 and 6 also suppressed human cancer cell (promyelocytic leukemia cell line, HL-60) growth, and LD50 values were 8.8, 20.2 and 72.3 μM, respectively, suggesting that cell growth inhibition had the same tendency as the inhibition of topos rather than pols. Compounds 2 and 6 arrested the cells in S and G2/M phases, compound 3 arrested the cells in the G2/M phase, and these compounds also increased sub-G1 phase in the cell cycle. These results suggested that the effect of cell cycle arrest might be effective on both pols and topos activities. From these findings, the action mode of cholesterol derivatives as anti-cancer compounds is discussed.  相似文献

The aim of this study was to analyze the relationship between statin use along with serum cholesterol levels and prostate cancer (PCa) detection and aggressiveness. Statin users of three years or more and serum cholesterol levels (SC) were assessed in 2408 men scheduled for prostate biopsy. SC was classified as normal (NSC: <200 mg/dL) or high (HSC: >200 mg/dL). High-grade PCa (HGPCa) was considered if the Gleason score was greater than 7. Statin users comprised 30.9% of those studied. The PCa detection rate was 31.2% of men on statins and 37% of non-statin users (p < 0.006). The PCa detection rate was 26.3% in men with NSC and 40.6% in those with HSC (p < 0.001). In the subset of NSC men, the PCa rate was 26.5% for statin users and 26.2% for non-users (p = 0.939), while in men with HSC, the PCa rate was 36.4% for statin users and 42.0% for non-statin users (p = 0.063). The HGPCa rate was 41.8% for statin users and 32.5% for non-users (p = 0.012). NSC men had a 53.8% rate of HGPCa, while the rate was only 27.6% in HSC men (p < 0.001). NSC men on statins had an HGPCa rate of 70.2%, while non-statin users had a rate of 41.2% (p < 0.001). The HGPCa rate for HSC men on statins was 18.8%, while the rate was 30.0% (p = 0.011) for non-users. Logistic regression analysis suggested that serum cholesterol levels could serve as an independent predictor of PCa risk, OR 1.87 (95% CI 1.56–2.24) and HGPCa risk, OR 0.31 (95% CI 0.23–0.44), while statin usage could not. Statin treatment may prevent PCa detection through serum cholesterol-mediated mechanisms. A disturbing increase in the HGPCa rate was observed in statin users who normalized their serum cholesterol.  相似文献

Cholesterol oxidation products (COPs) have been considered as specific in vivo markers of oxidative stress. In this study, an increased oxidative status was induced in Wistar rats by feeding them a high-fat diet (cafeteria diet). Another group of animals received the same diet supplemented with a combination of two different antioxidants, ascorbic acid (100 mg/kg rat/day) and sodium selenite (200 μg/kg rat/day) and a third group fed on a control diet. Total and individual COPs analysis of the different diets showed no differences among them. At the end of the experimental trial, rats were sacrificed and serum cholesterol, triglycerides and COPs were measured. None of the diets induced changes in rats body weight, total cholesterol and triglycerides levels. Serum total COPs in rats fed on the high-fat diet were 1.01 μg/ml, two times the amount of the control rats (0.47 μg/ml). When dietary antioxidant supplementation was given, serum total COPs concentration (0.44 μg/ml) showed the same levels than those of the rats on control diet. 7β-hydroxycholesterol, formed non-enzymatically via cholesterol peroxidation in the presence of reactive oxygen species, showed slightly lower values in the antioxidant-supplemented animals compared to the control ones. This study confirms the importance of dietary antioxidants as protective factors against the formation of oxysterols.  相似文献

Abstract

Variability in the chemical composition of surface properties of various wood fibers (eastern white cedar, jack pine, black spruce, and bark) was investigated using diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) and X-ray photoelectron spectroscopy (XPS). Both DRIFTS and XPS showed high variability in fiber surface composition between species and between fiber types (sapwood, heartwood, and bark). Fiber surface was modified by esterification reaction using a maleic anhydride polyethylene (MAPE) treatment. DRIFTS failed to assess surface modification, whereas XPS results showed that MAPE treatment increased the surface hydrocarbon concentration of jack pine wood fiber, indicated by a decrease in oxygen–carbon ratio and an increase in relative intensity of the C1 component in the C1s signal. Lignin concentration variability on the fiber surface was determined as the major factor that prevents esterification from taking place.  相似文献

The effects of treatment with commercial lecithin followed by bleaching and deodorization processes on cholesterol reduction (removal) of Iranian beef tallow were studied. Steam-rendering extracted fat was subjected to lecithin treatment and the effects of four variables, i.e., ratio of lecithin to tallow (1:5, 1:10, 1:20 and 1:30 w/w), stirring time (0.5, 1.5, 3, 6 and 12 h), stirring rate (200, 500, 1,000 and 1,250 rpm) and ratio of lecithin to water (1:2, 1:5 and 1:10 w/v) were investigated. The results showed that cholesterol removal is increased with increasing the lecithin to tallow ratio and the stirring rate. Increasing stirring time up to 1.5 h increased cholesterol removal; however, longer times did not have any significant effect on it. The ratio of lecithin to water did not have any significant effect on cholesterol removal, either. It was concluded that up to 43% of the tallow cholesterol content could be removed by lecithin. Treatment with lecithin increased tallow acidity and color but did not have any effect on the peroxide value (PV). It is proposed that since tallow, as a slaughterhouse by-product, is not expensive, by this method we can improve its eating quality for use in dietary or industrial applications.  相似文献

Treatment of neonatal rats with U18666A, an inhibitor of desmosterol Δ24-reductase, results in accumulation of desmosterol (Δ5,24) and depletion of cholesterol (Δ5) in various bodily tissues and also causes cataracts. We evaluated the effects of U18666A on the sterol composition, de novo sterol synthesis, and histological structure of the retina. Neonatal Sprague-Dawley rats were injected subcutaneously with U18666A (15 mg/kg, in olive oil) every other day from birth through 3 wk of age; in parallel, control rats received olive oil alone. At 21 d, treated and control groups each were subdivided into two groups: one group of each was injected intravitreally with [3H]acetate; retinas were removed 20 h later and non-saponifiable lipids (NSL) were analyzed by radio-high-performance liquid chromatography. The other group was injected intravitreally with [3H]leucine; 4 d later, one eye of each animal was evaluated by light and electron microscopy and light microscopic autoradiography, while contralateral retinas and rod outer segment (ROS) membranes prepared thereform were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis/fluorography. In the treated group, the Δ55,24 mole ratio of retinas was ca. 1.0, and >88% of the NSL radioactivity was in Δ5,24; in contrast, control retinas had Δ55,24 >170, with >80% of the NSL radioactivity in Δ5. Retinal histology, ultrastructure, ROS renewal rates, and rhodopsin synthesis and intracellular trafficking were comparable in both treated and control animals. These results suggest that desmosterol can either substitute functionally for cholesterol in the retina or it can complement subthreshold levels of cholesterol by sterol synergism.  相似文献

MCM-41介孔分子筛先后与苄基氯和氯磺酸接枝反应,得到苄基磺酸介孔分子筛(SBM)。通过X射线多晶衍射(XRD)、氮气吸附脱附、红外光谱(IR)、有机元素分析、扫描电镜(SEM)等方法对所得样品进行了表征。研究结果表明,MCM-41表面硅羟基与苄基氯反应,接枝量w(C)=6.68%;进一步磺化,接枝率w(S)=8.46%。XRD等分析结果表明,经过两步接枝反应,SBM仍然具有介孔结构特征,比表面积和孔容分别为366.9 m2/g、0.349 cm3/g。以SBM为催化剂,催化合成柠檬酸三丁酯;在n(酸)∶n(醇)=1.0∶9.0,w(催化剂)=3%,反应温度110℃,回流时间7 h的条件下,柠檬酸转化率可达93.2%;催化剂重复使用4次柠檬酸转化率为76.3%。  相似文献

针对蛋白质离子交换层析分离过程预测的困难,选择zeta电位作为中间参数,建立zeta电位与层析容量因子之间的关联.选择牛血清白蛋白(BSA)作为模型蛋白质,考察了pH、离子浓度、离子种类等对蛋白质zeta电位的影响,证实了zeta电位作为蛋白质表面电荷特性表征参数的可行性.以阴离子交换Sepharose FF DEAE为模型层析介质,测定了BSA在不同pH和盐浓度下的容量因子.综合考虑蛋白质和介质两方面因素,选择两者zeta电位乘积(-ζ1×ζ2)作为中间参数,对于不同pH和盐浓度的流动相,(-ζ1×ζ2)与容量因子的对数呈现良好的线性关系,实现了由简便的zeta电位分析预估宏观离子交换层析分离行为的目的,该关联关系的建立将有助于层析分离过程的优化设计.  相似文献

This study aimed to investigate the associations of the serial changes of serum levels of various growth factors with liver regeneration after hepatectomy in healthy liver donors. Sixteen healthy liver donors who underwent conventional liver resection were included. Serum levels of various growth factors before hepatectomy and on postoperative day (POD) 1, 3, 5 and 7 were measured. Liver volume data calculated by multi-detector computed tomography using workstation. The ratio of remnant liver volume on POD 0 to liver volume before the operation was 51% ± 20%. The ratio of liver volume on POD 14 to liver volume on POD 0 were inversely correlated with remnant liver volume on POD 0 (r = −0.91). The ratio of liver volume on POD 14 to liver volume on POD 0 were significantly correlated with serum hepatocyte growth factor (HGF) levels on POD 1 (r = 0.54), serum leptin levels on POD 1 (r = 0.54), and serum macrophage colony-stimulating factor (M-CSF) levels on POD 5 (r = 0.76) and POD 7 (r = 0.80). These results suggest that early-phase elevation of serum levels of HGF, leptin and M-CSF may be associated with the acceleration of liver regeneration after hepatectomy in humans.  相似文献

Oxidized LDL (oxLDL) has been shown to activate the sphingomyelinase pathway producing ceramide in vascular smooth muscle cells. Therefore ceramide, which is a biologically active lipid causing apoptosis in a variety of cells, may be involved in the apoptotic action of oxLDL. In this study, we examined whether cholesterol enriched diets affected ceramide metabolism and oxidation product of LDL, represented by degradation of apolipoprotein B-100 (apoB) in apoE-deficient (apoE−/−) mice. ApoE−/− and wild type mice were fed a standard (AIN-76) diet or 1% cholesterol-enriched diet for 8 weeks. Tissue ceramide levels were analyzed using electrospray tandem mass spectrometry (LC-MS/MS). Ceramide levels in the plasma and the liver of apoE−/− mice were intrinsically higher than those of the wild type. In apoE−/− mice, dietary cholesterol significantly increased several ceramides and degradation products of apoB in plasma compared to those fed the control diet. Dietary cholesterol did not affect tissue ceramide levels in the wild type mice. Based on these results, plasma ceramides possibly correlate with the increase in LDL oxidation and are a risk factor for atherosclerosis.  相似文献

Exogenous and endogenous cholesterol oxidation products (COPs) perturb various metabolic processes, and thereby they may induce various homeostasis-related disorders. Here, we observed that procyanidin-rich dietary apple polyphenol (APP) from unripe apples alleviates the perturbation of lipid metabolism by decreasing the exogenous COP levels in rats. Dietary COPs may be the greatest source of COPs found in the human body. Rats (4 weeks of age) were fed AIN-purified diets containing 0.3% COPs supplemented with 0.5 or 2.5% APP for 3 weeks. Dietary APP alleviated the growth inhibition action of the exogenous COPs. The modulations of the liver lipid profile by COPs remained unchanged. However, serum total cholesterol, high-density lipoprotein cholesterol, and triglyceride levels increased following the intake of dietary APP. Further, dietary APP inhibited the increase in lipid peroxide levels in the liver and serum by COPs. The activity of hepatic Δ6 desaturase was lowered by dietary APP in a dose-dependent manner, although exogenous COPs generally increased the activity of this enzyme. In keeping with this observation, Δ6 desaturation indices in the phospholipids and cholesteryl esters of the liver and serum lipids were lower in the APP-fed groups than those in the control group. Dietary APP also promoted the excretion of exogenous COPs, cholesterol, and acidic steroids in feces. Therefore, the inhibition of intestinal absorption of COPs may partly contribute to the alleviation of the perturbation of lipid metabolism and lipid peroxidation levels. Thus, APP may be an important removal agent of exogenous toxic material such as COPs contained in processed or fast foods.  相似文献

The aim of the present study was to investigate the effect of sandalwood seed oil on fatty acid (FA) profiles and inflammatory factors in rats. Fifty male Sprague–Dawley rats were randomly divided into five different dietary groups: 10 % soybean oil (SO), 10 % olive oil (OO), 10 % safflower oil (SFO), 10 % linseed oil (LSO) and 8 % sandalwood seed oil blended with 2 % SO (SWSO) for 8 weeks. The SWSO group had a higher total n-3 polyunsaturated fatty acids (PUFA) levels but lower n-6:n-3 PUFA ratios in both adipose tissue and liver than those in the SO, OO and SFO groups (p < 0.05). Although the SWSO group had a much lower 18:3n-3 level (4.51 %) in their dietary lipids than the LSO group (58.88 %), the levels of docosahexaenoic acid (DHA: 22:6n-3) in liver lipids and phospholipids of the SWSO group (7.52 and 11.77 %) were comparable to those of the LSO group (7.07 and 13.16 %). Ximenynic acid, a predominant acetylenic FA in sandalwood seed oil, was found to be highly incorporated into adipose tissue (13.73 %), but relatively lower in liver (0.51 %) in the SWSO group. The levels of prostaglandin F, prostaglandin E2, thromboxane B2, leukotriene B4, tumor necrosis factor-α and interleukin-1β in both liver and plasma were positively correlated with the n-6:n-3 ratios, suggesting that increased n-6 PUFA appear to increase the formation of pro-inflammatory cytokines, whereas n-3 PUFA exhibit anti-inflammatory activity. The present results suggest that sandalwood seed oil could increase tissue levels of n-3 PUFA, DHA and reduce the n-6:n-3 ratio, and may increase the anti-inflammatory activity in rats.  相似文献
