Eight major mulberry cultivars [Nakhonratchasima 60 (NS 60), Buriram 60 (BR 60), Chumphon (CP), Wavee (WV), Chaingmai (CM), Pikultong (PT), Kamphaengsaen (KS) and Kamnanchul (KJ)] cultivated in Thailand were assessed for their flavonoid and phenolic acid composition using HPLC and tested for antioxidant potential using 2,2‐diphenyl‐1‐picrylhydrazyl hydrate (DPPH) assay. The total phenolic content (TPC) ranged from 104.78 to 213.53 mg GAE/100 g DW, and total flavonoid content (TFC) ranged from 69.58 to 211.01 mg CE/100 g DW. The major flavonoid compounds in mulberry fruit cultivars were (+)‐catechin (309.26–750.01 mg/100 g DW), procyanidin B1 (62.59–224.41 mg/100 g DW), quercetin (5.36–58.42 mg/100 g DW), rutin (18.73–26.90 mg/100 g DW) and (?)‐epicatechin (8.47–29.21 mg/100 g DW). Gallic acid, cinnamic acid and p‐hydroxybenzoic acid were found to be the major phenolic acids in mulberry fruit cultivars. The gallic acid and cinnamic acid contents ranged from 7.33 to 23.90 mg/100 g DW and from 11.64 to 15.05 mg/100 g DW, respectively. p‐Hydroxybenzoic acid content ranged from 1.77 mg/100 g DW (PT) to 7.13 mg/100 g DW (KJ). DPPH‐scavenging ability was excellent for ethanolic extract of NS 60, and EC50 value of NS 60 (241.83 μg mL?1) was significantly lower than those of the others (< 0.05). TPC and TFC of the mulberry fruit were positively correlated with the DPPH‐scavenging ability.  相似文献

Nine phenolic acids were identified and quantified by HPLC–MS in leaves and 10 in seeds of kale (black cabbage). The free, ester (methanol-soluble), glycoside and ester-bound (methanol-insoluble) phenolic acid contents of the leaves were 487, 532, 4989 and 6402 ng/g fresh weight, respectively. Ferulic and caffeic acids (total contents; 4269 and 4887 ng/g, respectively) were the most abundant. The seed contents of these fractions were 1993, 1477, 1231 and 4909 ng/g dry weight (DW), respectively, and sinapic acid was the most abundant (5037 ng/g DW). The fractions’ total phenolic contents, determined colorimetrically, were highly correlated with their DPPH scavenging capacity, and in antimicrobial activity assays, with nine test organisms representing a wide array of taxa, all of the fractions were effective against Staphylococcus aureus, Enterococcus faecalis, Bacillus subtilis and (most strongly) Moraxella catarrhalis. Antimicrobial and antioxidant activities of kale phenolics in free and conjugated forms are discussed.  相似文献

The aim of this study was to assess the level of some phytochemicals in 19 raspberry cultivars grown in Lithuania. The content of total ellagic acid measured after 20 h acidic hydrolysis of investigated raspberry cultivars, varied from 119.8 (cv. ‘Pokusa’) to 323.5 mg/100 g (cv. ‘Bristol’). The content of total phenolics ranged from 278.6 (cv. ‘Pokusa’) to 714.7 mg/100 g (cv. ‘Bristol’). The total anthocyanins content varied from 2.1 (yellow cv. ‘Beglianka’) to 325.5 mg/100 g (black cv. ‘Bristol’). The radical scavenging capacity of the tested raspberry cultivars highly correlating with their total phenolics and total ellagic acid content (r = 0.90 and 0.92, respectively). The results of this study expand the knowledge about variation in the content of valuable bioactive compounds in raspberries and may help for the selection and validation of the most productive cultivars.  相似文献

In this study, the chemical composition of white (Morus alba L.), red (Morus rubra L.) and black (Morus nigra L.) mulberry fruits grown in the East Anatolia Region of Turkey was investigated. The highest total phenolic and flavonoid contents were observed in black mulberry (1422 mg gallic acid equivalents/100 g fresh matter and 276 mg quercetin equivalents/100 g fresh matter). M. alba had the highest total fat content (1.10%), followed by M. nigra (0.95%) and M. rubra (0.85%), respectively. The major fatty acids in mulberry fruits were linoleic acid (54.2%), palmitic acid (19.8%) and oleic acid (8.41%), respectively. The total soluble solids content of mulberry species varied between 15.9% (M. rubra L.) and 20.4% (M. alba L.), acidity between 0.25% (M. alba L.) and 1.40% (M. nigra L.), pH between 3.52 (M. nigra L.) and 5.60 (M. alba L.), ascorbic acid 19.4 mg/100 g (M. rubra L.) and 22.4 mg/100 g (M. alba L.), respectively. Mineral compositions of the mulberry species were 0.83% N, 235 mg/100 g P, 1141 mg/100 g K, 139 mg/100 g Ca, 109 mg/100 g Mg, 60 mg/100 g Na, 4.3 mg/100 g Fe, 0.4 mg/100 g Cu, 4.0 mg/100 g Mn and 3.1 mg/100 g Zn, respectively.  相似文献

Five different varieties of cauliflower (Brassica oleracea L. ssp. botrytis); two white (cv. ‘Aviso’, ‘Dania’), one purple (cv. ‘Grafitti’), one green (cv. ‘Emeraude’) and one romanesco/green pyramidal (cv. ‘Celio’) cultivar have been studied. All samples were thermally processed and the effects on the levels of glucosinolates (GLS), total phenols (TP), total monomeric anthocyanins (TMA), l-ascorbic acid (l-AA) and antioxidant capacities (FRAP and ORAC) were investigated. Processing methods applied were: blanching (3 min), boiling (10 min) and steaming (10 min). Total GLS were significantly (p < 0.05) affected by processing with the highest losses, 55 and 42% on average, occurring for boiled and blanched samples, respectively. Significant effects were also noted for steaming, but to a lesser extent, i.e. 19% average reduction. Antioxidant-related parameters were similarly affected with average losses of 27, 33, 36 and 46% in boiled cauliflower and 16, 21, 22 and 28% in blanched for TP, FRAP, l-AA and ORAC, respectively. Blanching and boiling reduced TMA in purple cauliflower by 38 and 53%, respectively. Steaming affected the antioxidant-related parameters the least for all cultivars. l-AA was significantly reduced by 14% in all cultivars by steaming. Some differences in behaviour between cultivars were noted, especially between white and coloured cultivars for TP, FRAP and l-AA, but also for some GLS. The main losses were caused by leaching into the processing water.  相似文献

Inactivation kinetics of peroxidase and polyphenol oxidase in fresh Rabdosia serra leaf were determined by hot water and steam blanching. Activation energy (52.30 kJ mol−1) of polyphenol oxidase inactivation was higher than that (20.15 kJ mol−1) of peroxidase. Water blanching at 90 °C or steam blanching at 100 °C for 90 s was recommended as the preliminary treatment for the retention of phenolics. Moreover, comparative evaluation of drying methods on the phenolics profiles and bioactivities of R. serra leaf were conducted. The results indicated that only intact leaf after freeze drying retained the initial quality. The sun- and air-dried leaves possessed identical phenolic profiles. The homogenised leaf (after freeze-drying) possessed a lower level of phenolics due to enzymatic degradation. Good antioxidant activities were detected for the sun- and air-dried leaves. There was insignificant difference in anti-tyrosinase and anti-α-glucosidase activities among sun-, air-, and freeze-dried leaves.  相似文献

The effect of blanching and treatment with white vinegar containing acetic acid on dietary fibre, low-molecular-weight carbohydrates and glucosinolates was studied in two cultivars of white cabbage (Heckla and Predikant). The total content of dietary fibre and low-molecular-weight carbohydrates was similar in both cultivars (24 and 60 g/100 g DM, respectively), while the distribution between soluble and insoluble fibre differed (19% was soluble in Heckla versus 26% in Predikant, P < 0.01). Further, Heckla contained higher amounts of glucose and sucrose, while the content of fructose and total glucosinolates was lower than in Predikant. The content of individual glucosinolates differed between the two cultivars. During blanching there was a loss of dry substance (30–34 g/100 g DM), where low-molecular-weight carbohydrates primarily explained the loss (82–90%), but some of the loss was also dietary fibre (about 8%), both soluble fibre containing uronic acids (mainly Predikant) and insoluble ones containing glucose (mainly Heckla). The glucosinolate levels decreased substantially in both cultivars, although the total loss was higher in Predikant (74%) than in Heckla (50%). The individual glucosinolates were affected to different degrees (15–91%). During souring with acetic acid, the content of dietary fibre (primarily insoluble ones) decreased further, while the content of low-molecular-weight carbohydrates was less affected. The total content of glucosinolates was not affected in Heckla but was further reduced in Predikant. There was, however, a substantial increase in 4-methoxyglucobrassicin in both cultivars. It is concluded that blanching and souring decrease the content of carbohydrates and glucosinolates to a great extent and both cultivars behaved similarly. However, individual components were affected differently in the two cultivars.  相似文献

The mulberry (Morus alba L.) leaf is a promising dietary source of antioxidants such as quercetin due to its relatively high content of that compound. We investigated effects of an air-drying process on the antioxidant capacity and stability of antioxidant polyphenolic compounds in mulberry leaves. Main compounds playing a central role in antioxidant activities in mulberry leaves are quercetin glycosides and chlorogenic acid. Raw mulberry leaves were air-dried at various temperatures, and antioxidant activity using DPPH radical scavenging assay and levels of antioxidant compounds were measured. DPPH radical scavenging activity and levels of polyphenolic compounds in mulberry leaves air-dried at 60 °C or below were not significantly different from those of freeze-dried mulberry leaves, whereas both values in mulberry leaves air-dried at 70 °C and over decreased significantly. These results indicate that strict temperature control is important in the production of mulberry leaf products to maintain antioxidant activity and levels of polyphenolic compounds.  相似文献

Coumarin is a natural product well-known for its pleasant sweet-herbaceous and cherry flower-like odour. Despite coumarin being widely found in the plant kingdom, its occurrence in tea leaves is very poorly characterised. In this work, green tea made from the cultivars “Shizu-7132”, “Koushun” and “Tsuyuhikari” were found to have sweet-herbaceous odour. Application of the stable isotope dilution assay for the quantification of coumarin revealed that its levels in these Japanese green tea products ranged from 0.26 to 0.88 μg/g of green tea product, whereas concentrations were generally below 0.2 μg/g in common green tea products. In contrast to the leaf, the stem part contained much less coumarin. During the manufacturing process of the tea (Shizu-7132), the steaming time and drying temperature influenced the content of coumarin in the final product. Due to hydrolysis of glycosidically bound coumarin precursors, in fresh tea leaves most coumarin occurred in its free form. Tea leaves also contained small amounts of bound coumarin precursors, such as primeveroside.  相似文献

There is no published information on the genotypic variation of major biochemical constituents in mango fruit endemic to Sri Lanka. Accordingly, non-structural carbohydrates, non-volatile organic acids and total phenolics were determined from the peel and pulp of pre-climacteric Sri Lankan mango cultivars (viz. Willard, Karutha Colomban, Vellai Colomban, Ampalavi, and Malgova) at three different maturity stages. Principal components analysis revealed distinct clustering of samples according to their biochemical profiles of peel and pulp at three maturity stages. Sugar concentrations generally declined with maturity in both peel and pulp except for cv. Willard. Fructose was the predominant sugar in both peel (56.2–106 mg/g dry weight (DW)) and pulp (67.4–141 mg/g DW), followed by glucose and sucrose. Starch concentration increased with maturity and was higher in pulp (26.0–55.0% DW) than peel (18.2–38.9% DW) at full mature stage. Dry matter as a proportion of fresh weight (FW) increased with maturity.  相似文献

Phenolic acids in potherb mustard (Brassica juncea, Coss.) were determined and the effects of pickling methods on the contents of total free phenolic acids, total phenolic acids, total phenolics, and antioxidant activities were investigated. Gallic acid, protocatechuic acid, p-hydroxybenzoic acid, vanillic acid, caffeic acid, p-coumaric acid, ferulic acid, and sinapic acid were identified in the present study. The contents of total free phenolic acids, total phenolic acids and total phenolics in fresh potherb mustard were 84.8 ± 0.58 μg/g dry weight (DW), 539 ± 1.36 μg/g DW, and 7.95 ± 0.28 mg/g DW, respectively. The total free phenolic acids increased during the pickling processes, but the total phenolic acids, total phenolics, and antioxidant activities decreased. However, after 5 weeks of fermentation, all the pickling methods retained over 70% of total phenolic contents and above 65% of antioxidant capacities. The results indicated that pickling processes were relatively good methods for the preservation of phenolic acids and antioxidants for potherb mustard.  相似文献

为研究大曲-多菌种耦合-发酵桑叶茶的品质,以大曲接种量、初始糖度、发酵温度和叶子含水量为工艺指标,研究不同工艺参数对桑叶茶黄酮、氨基酸、总酚和感官品质的影响,通过响应面-主成分分析法确定最佳工艺。实验结果表明: 发酵桑叶茶的最佳工艺条件为:叶子含水量65%,大曲接种量8%,初始糖度6%,发酵温度30 ℃,规范化综合得分为0.996±0.02,与理论规范化综合得分1.0105±0.01接近。在此条件下,得到发酵桑叶茶黄酮含量为35.81 mg/g,氨基酸含量为1.38 mg/g,总酚含量为16.68 mg/g。本实验通过响应面-主成分分析法优化桑叶茶发酵工艺, 为桑叶茶的开发利用提供一定的数据支持。  相似文献

Vitamin C and polyphenol content as well as total antioxidative activity were investigated in fresh leaves of kale; in leaves after blanching or cooking; and in frozen and canned leaves. In 100 g fresh matter, kale leaves contained 384.9 mg polyphenols and 112.1 mg vitamin C, with a Trolox equivalent antioxidant capacity (TEAC) level of 1175 μM Trolox. Of the polyphenols identified in kale leaves, ferulic acid occurred in the highest amount (240.44 mg/100 g, constituting 62% of total polyphenols). Freezing was a better method of preserving kale leaves since the loss of nutritive constituents was lower than in the case of canning. Depending on preliminary processing and storage temperature, after one-year storage frozen leaves contained 82.9–171.3 mg polyphenols and 39.3–65.4 mg vitamin C, with TEAC at the level of 501–681 μM Trolox in 100 g. In canned leaves these values were: 91.3–94.1 mg polyphenols, 16.1–19.3 mg vitamin C and 268–293 μM Trolox.  相似文献

The effect of different extraction set-ups that influence the extraction efficiency of catechins and caffeine from green tea leaves (variety Fanning Belas, China) were studied using different aqueous and pure solvents (acetone, ethanol, methanol, acetonitrile, water), different temperatures (60, 80, 95 and 100 °C) and times (5–240 min). Raw extracts were analysed for contents of major catechins (EC, EGC, ECG, EGCG), caffeine, proanthocyanidins and flavonols (myricetin, caempherol, quercetin). Starting material was found to contain 191 g major catechins/kg material, 36 g caffeine/kg material and 5.2 g flavonols/kg material on a dry mass basis. The content of major catechins in green tea extracts varied from approximately 280–580 g/kg dry extract, with extraction efficiencies of major catechins varying from 61% to almost 100%. Content of caffeine in extract was in the range of 75 g/kg, where its extraction efficiency varied from 62% to 76%. Average extraction yield was 30% with exceptions when using pure acetone and acetonitrile, where extraction yield was about 3%. Contents of flavonols and proanthocyanidins were in the ranges 6–20 and 12–19 g/kg, respectively. Different extraction procedures with water were also investigated and optimal conditions determined: maximum achieved extraction efficiency of catechins with water was obtained at 80 °C after 20 min (97%) and at 95 °C after 10 min of extraction (90%). Degradation of catechins was observed at higher extraction temperatures and with prolonged extraction times. Using a lower ratio of solvent to material, extraction efficiencies were increased by applying a multi-step extraction procedure. Optimal extraction procedure was then performed using decaffeinated green tea leaves, which were obtained by high-pressure extraction with CO2, when 98% of caffeine was selectively isolated without significant impact on valuable catechins.  相似文献

目的:桑树叶片在传统医学中被用作降糖和降血压的药物,其作用机理与活性成分含量有关。评价桑叶提取物不同生物活性,包括生物活性物质含量、抗氧化活性和抗α-淀粉酶活性。方法:采用SPE(固相萃取技术)将多酚粗提液分离纯化为花色苷、非花色苷多酚和水层三个组分,采用HPLC-PDA和HPLC-ESI/MS2对桑叶中的多酚物质进行系统地定性和定量。利用DPPH(1,1-二苯基-2-三硝基苯肼自由基清除能力)、ABTS(2,2’-联氮-双(3-乙基苯并噻唑啉-6-磺酸))和FRAP(铁离子还原能力)三种方法测定不同提取物的抗氧化活性,并采用福林酚法、pH差异法和比色法测定桑叶提取物总酚、总花色苷和总原花青素含量。结果:桑叶中共鉴定出6种花色苷、42种非花色苷多酚,其中,矮牵牛素-3-葡萄糖苷和原花青素衍生物含量很高;桑叶中总酚、总花色苷和总原花青素的含量分别为14.09 mg GAE/g DW、0.17 mg C3G/g DW和17.24 μg PA2/g DW;多酚粗提液、花色苷、非花色苷多酚和水层对α-淀粉酶活性的IC50(半抑制浓度)分别为8.31、13.70、0.25和12.00 mg/mL,由此看出非花色苷多酚的抗α-淀粉酶活性最高。结论:桑叶具有较强的抗氧化活性,其中花色苷显示出最高的抗氧化活性。这些数据可以为蒙古桑叶用作功能食品提供重要参考。  相似文献

Five tea clones of the Camellia assamica variety grown in Wushwush tea plantation farms, Ethiopia, were analyzed for their contents of essential, non-essential and toxic metals (K, Ca, Mg, Fe, Mn, Cu, Zn, Na, Cd and Pb) by atomic absorption flame emission spectroscopy. Both the tea leaves and the soils of the study farms showed similar accumulation patterns in their contents of the studied macronutrients. Among the macronutrient metals, K was the most abundant element in the tea leaves (17.7–24.8 mg/g) and the soils (7.14–9.73 mg/g). Mn was the predominant micronutrient heavy metal in the tea leaf tissues ranging between 501 and 1281 mg/kg. Level of Fe (29.6–100 mg/kg) in the leaf tissue was found to be the second most abundant micronutrient next to Mn whereas concentrations of Cu and Co were relatively lower both in the soil and tea samples. The toxic heavy metals Pb and Cd in the leaf tissues were present at levels too low to be detected by the analytical technique used in this study. The soils were found to be acidic (pH 5.04–5.49) with high organic matter (5.48–6.02%). Fe was the most abundant metal followed by Mn, Na and Zn in the soils. Unlike the tea leaves, the soils were found to contain traces of the toxic metal, Cd (0.02–1.10 mg/kg). The levels of most of the metals determined in this study compared well with those reported for tea leaves from some other parts of the world.  相似文献

The variation in contents of seven phenolic compounds in pigeon pea seedlings during growth and storage was investigated. Maximum contents of vitexin, isovitexin and orientin were found in leaves growing 40 days, which were 0.99 ± 0.06, 6.63 ± 0.35 and 30.89 ± 1.92 mg/g DW. Apigenin and luteolin were extensively distributed in leaves, stems and roots. Pinostrobin and cajaninstilbene acid were mainly accumulated in leaves, the peak values 3.53 ± 0.18 and 2.49 ± 0.13 mg/g DW appeared at the 60th day. Slight and steady increases of seven phenolic compounds were found in room temperature (25 °C) stored pigeon pea leaves up to 120 days. The highest accumulation of seven phenolic compounds at chilling temperature (4 °C) was observed at the 45th day, after which the contents decreased sharply. The stems extracts exhibited more efficient DPPH radical-scavenging ability while the roots extracts demonstrated the strongest lipid peroxidation inhibitory activity.  相似文献

Phenolic compounds in fruits of 27 cultivars of strawberry (Fragaria x ananassa Duch.) grown in Norway were characterised and quantified by HPLC-DAD-MSn. Total phenolic content, calculated as the sum of the individual compounds, varied 2.3-fold among cultivars, i.e., from 57 to 133 mg/100 g of fw. There were significant differences among cultivars in concentration of all phenolic compounds. The highest variation between cultivars was found for cinnamoyl glucose (0.6-24.9 mg/100 g of fw). Concentration of anthocyanins, the most abundant class of phenolic compounds in the majority of the cultivars, varied from 8.5 to 65.9 mg/100 g of fw. Flavan-3-ols (11-45 mg/100 g of fw) and ellagitannins (7.7-18.2 mg/100 g of fw) contributed on average 28% and 14% to total phenolic contents in the strawberry cultivars, respectively. In three cultivars harvested at three stages of ripeness, anthocyanins and cinnamic acid conjugates were the compounds most affected by ripening. The anthocyanin profile for the individual cultivars was only slightly affected by ripening and growing conditions.  相似文献

Mulberry 1-deoxynojirimycin (DNJ), a potent α-glucosidase inhibitor, suppresses postprandial blood glucose, thereby possibly preventing diabetes mellitus. At present, mulberry dry teas are commercially supplied as functional foods in many countries, but these products may not provide an effective dose (6 mg DNJ/60 kg human wt) due to their low DNJ content (about 100 mg/100 g of dry wt). Therefore, development of tea with higher DNJ content is desirable. To do this, we investigated distribution of DNJ content and α-glucosidase inhibitory activity in 35 Thai mulberry varieties. DNJ content in young leaves varied among mulberry varieties from 30 to 170 mg/100 g of dry leaves. Varieties having highest DNJ content were Kam, Burirum 60 and Burirum 51. Leaf position affected DNJ content: shoots > young leaves > mature leaves. DNJ concentration and α-glucosidase inhibitory activity were highly correlated (r = 0.84), suggesting that α-glucosidase inhibitory activity of mulberry leaves is mainly due to DNJ. Consequently, high DNJ content mulberry tea was produced from shoots of varieties such as Burirum 60, which contains 300 mg/100 g of dry wt. Tea-making conditions were optimized for highest DNJ extraction using response surface methodology. Approximate 95% of total DNJ in high DNJ content dry tea was extracted when temperature was maintained at 98 °C for 400 s; these conditions could be applicable for preparation of commercial products with high DNJ content. One cup (230 ml, a normal serving) of DNJ-enriched mulberry tea contained enough DNJ (6.5 mg) to effectively suppress postprandial blood glucose.  相似文献
