In this study, ascorbic acid, total polyphenols and proanthocyanidins of fresh-cut cashew apple were quantified. Antioxidant capacity was determined in whole juice and in polyphenols extracts by three methods: 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP) and ??-carotene bleaching. Effect of cutting and storage for 24 h at 2 °C, 27 °C and 40 °C on these compounds were also evaluated. Cashew apple presented 163 mg of ascorbic acid per 100 g of fresh weight (FW). Soluble and hydrolysable polyphenols contents were 12.79 mg GAE/100 g FW and 18.53 mg GAE/100 g FW and proanthocyanidins were 9.27 mg/100 g FW. Antioxidant capacity of juice and polyphenols extract was high for DPPH method. Storage temperatures affected bioactive compounds on cut cashew apple. The content of ascorbic acid decreased in all temperatures. Proanthocyanidins were more sensitive to 40 °C than to other temperatures. The content of polyphenols and antioxidant capacity of juice by DPPH assay did not change. However, the reducing power was lower in samples kept at high temperatures. A strong positive correlation between ascorbic acid and FRAP (r = 0.99) and a negative correlation between DPPH and FRAP (r = − 0.79) were observed. No correlations were found between polyphenols and antioxidant capacity indicating the importance of phenolic composition in the extracts. The results confirm the importance of temperature and injury on the quality of cashew apple.  相似文献

Total phenolic content (TPC) and total antioxidant capacity (TAC) of four onion varieties (red, white, yellow and sweet) and shallot from selected locations (Washington, Idaho, Oregon, Texas and Georgia) were determined using Fourier transform infrared (FT-IR) spectroscopy (4000–400 cm−1). The Folin–Ciocalteu (F–C) assay was used to quantify TPC and three assays were used to determine TAC, including 2,2-diphenyl-picrylhydrazyl (DPPH) assay, Trolox equivalent antioxidant capacity (TEAC) assay and ferric reducing antioxidant power (FRAP) assay. Partial least squares regression (PLSR) with cross-validation (leave-one-out) was conducted on onion and shallot extracts (n = 200) and their corresponding F–C, DPPH, TEAC and FRAP values were employed to obtain four independent calibration models for predicting TPC and TAC for the extracts. Spectra from an extra 19 independent extracts were used as an external validation set for prediction. A correlation of r > 0.95 was obtained between FT-IR predicted and reference values (by F–C, DPPH, TEAC and FRAP assay) with standard errors of calibration (SEC) and standard errors of cross-validation (SECV) less than 2.85, 0.35 and 0.45 μmol Trolox/g FW of extracts for TEAC, FRAP and DPPH assay, respectively; and 0.36 mg gallic acid/g FW of extracts for the F–C assay. In addition, cluster analysis (principal component analysis (PCA)) and discriminant function analysis (DFA) could differentiate varieties of onions and shallot based upon infrared spectral features. Loading plots for the various chemometrics models indicated that hydroxyl and phenolic functional groups were most closely correlated with antioxidant capacity. The use of mid-infrared spectroscopy to predict the total antioxidant capacity of vegetables provides a rapid and precise alternative to traditional wet chemistry analysis.  相似文献

Pomegranate juice is consumed widely for its possible health benefits. The aril juice from 15 pomegranate cultivars grown in Georgia were analysed for juice yield based on fresh weight (FW) and physico-chemical properties, using blender and mechanical press extraction. Blender had a higher juice yield (42.04% FW) compared to mechanical press (38.05% FW). Total phenolics and antioxidant capacity was determined by Folin–Ciocalteau method and ferric reducing antioxidant power (FRAP), Trolox equivalent antioxidant capacity (TEAC), and oxygen radical absorbance capacity (ORAC) assays, respectively. Total monomeric anthocyanins were determined by pH differential method and RP-HPLC. The major anthocyanin was delphinidin 3-glucoside. High negative and significant (p ? 0.05) correlations were found between pH and titratable acidity (TA). The total soluble solids content (TSS) averaged 15.59 in blender and 14.94 °Brix in mechanical press. Chemical analysis of juice showed significant differences among cultivars and extraction methods. Overall, blender was more efficient than mechanical press juice extraction.  相似文献

The total polyphenol content and the antioxidant activity of kudingcha made from Ilex kudingcha C.J. Tseng were determined by Folin–Ciocalteu reagent, and DPPH, FRAP, and TEAC methods, respectively. The crude extract (CE) of kudingcha and its four fractions of chloroform (CfF), ethyl acetate (EaF), n-butanol (nBF), and water (WtF) were prepared and subjected to antioxidant evaluation and analysis by high performance liquid chromatography. The extracts of kudingcha contained large amounts of caffeoylquinic acid (CQA) derivatives, including 3-CQA, 5-CQA, 3,4-diCQA, 3,5-diCQA, and 4,5-diCQA, and showed potent antioxidant activity. The antioxidant activities of CE and its fractions decreased in the order of EaF > nBF > CE > WtF > CfF, according to the DPPH assay and FRAP assay, which were the same, with the exception of the rank order of CfF and WtF, as the TEAC assay. Furthermore, a satisfactory correlation between total polyphenol content and antioxidant activity was observed.  相似文献

The methanol extract from mature fruits of Aristotelia chilensis (Mol) Stuntz (Elaeocarpaceae) showed antioxidant activities and cardioprotective effects on acute ischemia/reperfusion performed in rat heart in vivo. This extract protected animals from heart damage by the incidence of reperfusion dysrythmias, and the no-recovery of sinus rhythm. On the other hand, the MeOH extract of the fruit was able to prevent these harmful events in the animal’s heart by diminishing lipid oxidation and reducing the concentration of thiobarbituric acid reactive substances (TBARS), a lipid peroxidation index. In addition, MeOH extract of A. chilensis was evaluated for DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging, crocin radical scavenging, oxygen radical absorption capacity (ORAC), ferric reducing antioxidant power (FRAP), an estimation of lipid peroxidation in liposomes through the inhibition of formation of TBARS. MeOH extract was found to have IC50 of 1.62 ppm against DPPH and 2.51 ppm against TBARS, compared with the juice, whose IC50 was 12.1 ppm and 9.58 ppm against DPPH and TBARS formation, respectively. Antioxidant activities of MeOH extract were strongly correlated with total polyphenol content. Consistent with this finding, MeOH had the greatest ORAC and FRAP values as percentage of activity. These results show that these fruits could be useful as antioxidant, cardioprotective and nutraceutical sources.  相似文献

The aim of this study was to examine the contribution of sugar, organic acid, neutral phenol, and anthocyanin fractions and added ascorbic acid to grape and pomegranate‐nectarine juice total phenol, oxygen radical absorbance capacity (ORAC), ferric reducing antioxidant power (FRAP) and 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) values. Neutral phenol and anthocyanin fractions contributed ≥75% of the total antioxidant capacity (TAC) for both juices. Intrinsic synergy and antagonism among the fractionated constituents occurred inconsistently in each assay. Sugars and organic acids antagonised pomegranate juice neutral phenols and anthocyanins in the DPPH assay by 50% and the grape juice ORAC value by 21%, but were synergistic to the grape juice FRAP value. The added ascorbic acid was dose‐dependently synergistic with pomegranate and grape juice total phenol, DPPH and FRAP assays, but less so in the ORAC assay. Thus, the interactions between grape and pomegranate juice constituents determine TAC and total phenol values, and synergy in these assays could not be attributed solely to polyphenols.  相似文献

The content of antioxidant compounds and antioxidant capacity of whole and fresh-cut mango, stored for 10 days at 12 °C and 5 °C, respectively and their influence on serum antioxidant capacity and lipid profile of normolipidemic humans were studied. Whole mango (WM) had a higher content of flavonoids, ??-carotene and antioxidant capacity, determined by oxygen radical scavenging capacity, (ORAC) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assays, than the fresh-cut fruit (FCM). FCM presented higher amounts of total phenols. Thirty normolipidemic volunteers, ages 20-50 years, were randomly divided into two groups (WM and FCM) 15 persons each. During 30 consecutive days volunteers from groups 1 and 2, received daily 200 g of WM or FCM, respectively. Lipid levels and antioxidant capacity in plasma were determined at 0, 15 and 30 days of the experiment. Serum triglycerides were significantly reduced after 30 days of supplementation with WM and FCM (37 and 38% respectively); VLDL levels were reduced in a similar proportion. No significant changes in other plasma lipid levels were observed. Both treatments increased plasma antioxidant capacity measured by ORAC and TEAC methods. According to the results obtained in this study, we suggest that addition of mango fruit to generally accepted healthy diets could have a beneficial effect preventing hypertryglyceridemia, and that fresh-cut processing does not affect the beneficial properties of mango.  相似文献

The stability of antioxidants in an apple polyphenol–milk model system was examined. The model system consisted of skim milk fortified with pH-neutralised apple polyphenols (AP, 0–200 mg per 100 ml milk), with or without ascorbic acid (100 mg per 100 ml milk). Physical and chemical changes were evaluated after thermal treatment (120 °C, 5 min) and oxidative storage (20 °C and 38 °C, up to 12 weeks). Antioxidant capacity was determined using both oxygen radical absorbance capacity (ORAC) assay and ferric reducing antioxidant power (FRAP) assay. Significant antioxidant capacity was detected in the presence of milk. Antioxidant capacity was retained during thermal treatment but decreased slowly during storage.  相似文献

The changes in total phenol content and antioxidant capacity were monitored in six industrial dark fruit juices during 29-day refrigerated storage. The initial total phenol values ranged from 1302.1 mg/L GAE (strawberry) to 1919.8 mg/L GAE (black currant) with a mean of 1573.3 mg/L GAE. All juices exhibited fluctuations in TP values with a marked increase after 48 hours in refrigerated storage, and a greater overall TP content in 5/6 studied juices after 29 days. Antioxidant capacity was evaluated using the DPPH radical scavenging assay and cyclic voltammetry (CV) and expressed as Trolox equivalent antioxidant capacity (TEAC). Black currant juice exhibited the highest TEAC values according to both CV (2.42 mM Trolox) and the DPPH assay (5.68 mM Trolox), while cranberry juice antioxidants exhibited the greatest storage stability and the smallest antioxidant capacity decrease on day 29, 20% (CV) and 15% (DPPH assay). At the end of 29-day storage 5/6 juices exhibited a significant loss in antiradical activity and all 6 juices exhibited a significant loss in TEAC derived from CV measurements. Significant linear correlation was observed between the results of CV measurements and the DPPH antiradical activity (r2 = 0.62).  相似文献

Fruit of eight Actinidia genotypes were evaluated for antioxidant potential by several assays (DPPH, ABTS, ORAC, FRAP, SASR and MCC) and tested for their polyphenol composition and vitamin C contents. The significance analysis demonstrated that the antioxidant capacity of Actinidia eriantha and Actinidia latifolia fruits were significantly higher than that of other genotypes, which was about 3.3–8.7-fold higher than the Actinidia deliciosa cv. Hayward assayed in ABTS, DPPH, ORAC and FRAP methods. The total polyphenols and vitamin C contents showed a great variety amongst Actinidia genotypes and highly correlation with the total antioxidant capacity. It is concluded that significant genotypic difference exists in the total antioxidant capacity of Actinidia fruits. The wild A. eriantha and A. latifolia species have significantly higher antioxidant capacity than the cultivars of A. chinensis and A. deliciosa. Both total polyphenols and vitamin C are major contributors to the total antioxidant capacity in Actinidia fruit.  相似文献

The physicochemicals associated with fruit quality, antioxidant activities and changes in polyphenol composition during Fuji apple growth were investigated from the 25th to 105th day after full bloom day (DAFB). Three kinds of antioxidant assays, including oxygen radical absorbing capacity (ORAC) were executed, and polyphenol composition were examined using HPLC. The change in weight of the Fuji apples during fruit growth showed a typical sigmoidal curve. Both the rise of the soluble solid content and the drop of the titratable acidity appeared to be more significant (p < 0.05) after the 85th DAFB. Concurrently, the antioxidant activities and the polyphenol content decreased (p < 0.05) rapidly after the same date. Accordingly, a profitable thinning date can be chosen around the 85th DAFB to utilise dropped unripe apples as resources of antioxidants. Furthermore, the antioxidant activities of Fuji apples may be highly affected by the chlorogenic acid content.  相似文献

The effects of varying doses of UV and ultrasound (US), and incubation times at 25 °C on total phenolics (TP), antioxidant capacity by two methods, trolox equivalent antioxidant capacity (TEAC) and oxygen radical absorbance capacity (ORAC) and sensory overall acceptance (OA) of treated peanuts were investigated. Optimum process parameters for UV and US were determined using response surface methodology. UV and US increased TP, TEAC and ORAC total antioxidant capacity (TAC) of most treated peanuts, whereas OA decreased in all treatments compared to controls. US was more effective than UV in increasing total antioxidants. Optimum US process parameters at 36 h incubation time will result in products with TAC ≥ 68 μM TE/g, TP ≥ 1.51 mg GAE/g, TEAC ≥ 2.76 μM TE/g, and OA ≥ 5 or neither like nor dislike.  相似文献

Avocado (Persea americana) is an important tropical fruit and a good source of lipophilic phytochemicals such as monounsaturated fatty acids, carotenoids, vitamin E and sterols that have been inversely related to cardiovascular diseases. However, their antioxidant capacities have received far less attention compared with hydrophilic phytochemicals in this fruit. In this context, this study evaluated the effect of the stage of ripeness of ‘Hass’ avocado on the content of lipophilic and hydrophilic phytochemicals and their correlation with the antioxidant capacity. In every ripeness stage the fatty acids, total phenolic and flavonoid content as well as the antioxidant capacity were evaluated. Physiological and physico-chemical analysis were also performed including respiration rate, ethylene production, firmness, color (L*, °Hue, and Chroma), dry matter and oil content. In general, total phenols increased during ripening, while flavonoids slightly decreased. The main fatty acid identified was oleic acid (about 67-70% of total content). In general, a significant increase in monounsaturated and saturated fatty acids was observed during avocado ripening while polyunsaturated fatty acid content decreased (p < 0.05). Lipophilic extracts showed higher values of antioxidant capacity than hydrophilic, however, both extracts had similar trends for DPPH, TEAC and ORAC assays. DPPH and TEAC assays had a positive correlation with some unsaturated fatty acids. Avocado at different ripeness stages showed a better antioxidant capacity in the lipophilic fraction, which correlated with the fatty acid content evaluated.  相似文献

The antioxidant properties of 25 edible tropical plants, expressed as Trolox equivalent antioxidant capacity (TEAC), were studied using DPPH (1,1-diphenyl-2-picrylhydrazyl free radical) scavenging and reducing ferric ion antioxidant potential (FRAP) assays. Their cupric ion chelating activities (CCA) and total polyphenol contents (TPC) were also determined. A strong correlation between TEAC values obtained for the DPPH assay (TEACDPPH) and those for the FRAP assay (TEACFRAP) implied that compounds in the extracts were capable of scavenging the DPPH free radical and reducing ferric ions. A satisfactory correlation of TPC with TEACDPPH and TEACFRAP suggested that polyphenols in the extracts were partly responsible for the antioxidant activities while its correlation with CCA was poor, indicating that polyphenols might not be the main cupric ion chelators. Principal component analysis (PCA) indicated that TEACDPPH, TEACFRAP and TPC contributed to the total variation in the antioxidant activities of the plants.  相似文献

Microwave-assisted extraction (MAE) was investigated for extraction of phenolic compounds from tomato with maximised antioxidant activities using response surface methodology (RSM) coupled with a central composite design, and in vitro antioxidant assays (FRAP and ORAC). MAE was more efficient for greater antioxidant activities and higher total phenolic contents than solvent extraction. The optimal MAE processing parameters were 96.5 °C, 2.06 min, 66.2% ethanol for FRAP, and 96.5 °C, 1.66 min, 61.1% ethanol for ORAC. The models were successfully applied to 20 tomato cultivars, whose total phenolic contents (TPC) and indexes (TPI) were 489.30–997.45 mg gallic acid equivalent/100 g dry weight (DW) and 281.34–468.52 mg/100 g DW, respectively. Eight phenolic compounds were identified. Individual phenolics were 6.10–42.73 mg/100 g DW. The FRAP, but not the ORAC value showed good correlation with the TPC or TPI. The methodologies developed and the knowledge acquired in this study will provide useful information to tomato breeders and food processors.  相似文献

Arils from six pomegranate (Punica granatum L.) cultivars obtained from various sites from the Mediterranean region of Turkey were evaluated for their chemical and antioxidant properties. These properties included total phenolics (TP), total monomeric anthocyanins (TMA), soluble solids (TSS), titratable acidity (TA), individual sugars and organic acids. Antioxidant capacities of arils were determined by both the ferric reducing antioxidant power (FRAP) and trolox equivalent antioxidant capacity (TEAC) assays. The antioxidant capacities averaged 5.60 and 7.35 mmol TE/l by the TEAC and FRAP methods. Variability among cultivars was greatest for TMA content (CV 132%); individuals ranged from 6.1 to 219 mg cy3-Gluc l−1. TP means averaged 1507 mg GAE/l. Levels of FRAP, TEAC, TP, and TMA were strongly correlated (r = 0.82–0.96). The major sugars were fructose (6.4 g/100 ml) and glucose (6.8 g/100 ml), the major acids were citric (1.78 g/100 ml) and malic (0.12 g/100 ml).  相似文献

In order to investigate the antioxidant capacity of sedum (dolnamul, Sedum sarmentosum) and to offer a scientific basis for the medicinal use of it, lyophilized sedum leaves were extracted with n-hexane, n-butanol, ethyl acetate, and methanol, respectively. The methanol extract (ME) showed the highest extraction yield, but no significant differences in the extraction yield were observed. The total polyphenol content (TPC) of the ethyl acetate extract (EAE) and the ME was significantly higher than that of the non-polar solvent extracts. The EAE and ME showed the superior antioxidant activities in the DPPH, oxygen radical absorbance capacity (ORAC), and ferric reducing ability of plasma (FRAP) assays. Lipid peroxidation was efficiently inhibited by the addition of the sedum extracts in the ferric thiocyanate (FTC) and thiobarbituric acid reactive substances (TBARS) assays, but significant differences among the tested extracts were not observed. The ME showed the potent lipid peroxidation inhibitory activity comparable to butylated hydroxytoluene (BHT). The correlations between the TPC and the antioxidant activities based on the DPPH, ORAC, and FRAP assays were highly positive (R 2>0.899).  相似文献

Oxygen radical antioxidant capacity (ORAC) and trolox equivalent antioxidant capacity (TEAC) assays were compared to estimate the total antioxidant capacity (TAC) of orange juice, milk, and an orange juice-milk beverage. When the TEAC method was used with this beverage, an increase in the concentration of orange juice corresponded to an increase in TAC, but increasing the percentage of milk did not increase the TAC value. When the ORAC method was applied, it was seen that increased concentrations of juice or milk corresponded to greater antioxidant capacity. An evaluation was also made of the influence of certain compounds (ascorbic acid, gallic acid, β-carotene, lutein, zeaxanthin and albumin) with antioxidant capacity that were present in the samples studied.  相似文献

Eleven different cider apple pomaces (six single-cultivar and five from the cider-making industry) have been analysed for low molecular phenolic profiles and antioxidant capacity. The Folin index ranged between 2.3 and 15.1 g gallic acid per kg of dry matter. Major phenols were flavanols, dihydrochalcones (phloridzin and phloretin-2′-xyloglucoside), flavonols and cinnamic acids (chlorogenic and caffeic acids). The group of single-cultivar pomaces had higher contents of chlorogenic acid, (-)-epicatechin, procyanidin B2 and dihydrochalcones, whereas the industrial samples presented higher amounts of up to four unknown compounds, with absorption maxima between 256 and 284 nm. The antioxidant capacity of apple pomace, as determined by the DPPH and FRAP assays, was between 4.4 and 16.0 g ascorbic acid per kg of dry matter, thus confirming that apple pomace is a valuable source of antioxidants. PLSR analysis gave reliable mathematical models which allowed to predict the antioxidant activity of apple pomace as a function of the phenolic profile. The variables with the higher modelling power were phloridzin > procyanidin B2 > rutin + isoquercitrin > protocatechuic acid > hyperin.  相似文献

Physical and biochemical properties of pressurised and pasteurised longan juices with various xanthan additions, such as viscoelastic behaviour, colour L (lightness), −a (greenness), b (yellowness), ΔE (total different colours) and BI (Browning Index) parameters, polyphenol oxidase (PPO) activity, ascorbic acid, gallic acid, ellagic acid, total phenols and antioxidant capacity (DPPH assay) were studied. Viscoelastic determination indicated that longan juice with 0.15% xanthan addition was optimal for a fruit drink. Colour parameters showed pressurised longan juice at 500 MPa was brighter and more transparent than fresh and other processed juices. PPO was completely inactivated in pasteurised juices, whereas in pressurised juices at 300 and 500 MPa, the activities were more than 100% and 95–99%, respectively. Bioactive components including ascorbic acid were significantly reduced according to treatment severities, whereas gallic and ellagic acids were relatively stable in all processed juices. Total phenols and DPPH radical-scavenging activity decreased significantly on pasteurisation, but were stable on pressurisation.  相似文献
