化妆品中所添加的活性成分往往存在光、热稳定性差,溶解性差等原因,需通过包载技术提高其生物利用度。在各种包载体系中,脂质体由于结构与人体皮肤细胞相似,且具有良好的生物相容性,是一种有效的包载方法。然而传统脂质体由于稳定性、可变形性差等原因难以通过皮肤屏障,达到促渗效果。如今,通过改变传统脂质体磷脂双层膜的结构获得了各种新型脂质体,如醇质体、类脂质体、传递体、磷脂复合物、甘油体等,新型脂质体由于稳定性良好、具有可变形性、可改变角质层脂质结构等特点,能够改善药物经皮输送效果。文章综述了几种新型脂质体,并对其透皮效果进行分析,旨在为化妆品包载领域提供借鉴和思路。 相似文献

Transdermal delivery offers several advantages in drug distribution, including convenience, painless administration, avoidance of first-pass metabolism, and ease of termination. However, the natural protective barriers of the skin, such as the stratum corneum, the topmost layer of skin, limit the systemic absorption of external therapeutics via transdermal delivery. Therefore, extensive application of transdermal delivery in medical treatment has been limited. Over the past few years, many formulation strategies and physical technologies, therefore, have been developed to enhance transdermal delivery. The separable microneedles (MNs), consisted of upper metformin-loaded PCL arrowheads and lower PVA/PCL supporting array, have been fabricated by separately molding method. The metformin-loaded and thermal ablation MNs arrowheads were capped on dissolving solid supporting array. The as-fabricated MNs exhibited an excellent thermal ablation change due to the low melting point of PCL. After insertion into the skin, the upper separable MNs arrowheads could be embedded into the skin. When the separable MNs arrowheads were exposed to thermal stimulus, causing the PCL MNs arrowheads to melt and enabling the release of encapsulated metformin to be thermally modulated. 相似文献

Two different types of polyurethanes (PUs) were prepared with castor oil, ethylene glycol, isophorene diisocyanate and castor oil, and isophoren diisocyanate and poly‐(ethylene glycol) (400 or 600). PU films were prepared and characterized by Fourier transform infrared spectroscopy, differential scanning calorimetry, and gel permeation chromatography. We prepared transdermal patches by loading different amounts of drug, plasticizer, and penetration enhancer. In vitro drug permeability through the castor‐oil‐based aliphatic PU patches was examined with a Keshary–Chien diffusion cell. The effect of castor oil on the film‐forming properties and the effect of penetration enhancers on diffusion characteristics of indomethacin (IDM) drug through the castor‐oil‐based PU were investigated. Prolonged release of IDM was observed from the prepared PU patches. In vitro drug diffusion revealed that slow and prolonged release of IDM was achieved in the absence of penetration enhancers. 为了提高辅酶Q10(CoQ10)的透皮吸收效果,以辛酸癸酸甘油三酯(GTCC)为液体脂质,鲸蜡醇棕榈酸酯(CP)为固体脂质,制备得到了不同浓度固体脂质的CoQ10-脂质纳米囊,并对其稳定性及透皮性能进行了研究。结果表明,固体脂质的添加未对CoQ10-脂质纳米囊的粒径及储藏稳定性产生影响,粒径均为52nm左右,储藏180d后其粒径及外观均无明显变化。FT-IR结果表明,CoQ10与脂质纳米囊之间未发生化学反应且能被很好的包埋在脂质纳米囊中。通过闭合效应、体外透皮实验及激光共聚焦的观察发现,当CP质量浓度增加至70%和95%(以固液总脂质为基准,下同)时,CoQ10-脂质纳米囊能显著减少皮肤水分的蒸发,并更好的促进CoQ10的透皮吸收,使表皮层和真皮层的荧光强度明显增强。 相似文献
