The natural co-occurrence of fungal metabolites in maize samples from the South region of Brazil was studied using an LC-MS/MS based multi-mycotoxin method. All maize samples (n = 148) were contaminated with fumonisin B1 (FB1) and fumonisin B2 (FB2). Aflatoxin B1 (AFB1) and aflatoxin G1 (AFG1) were detected in 38 and 11 samples, respectively, while zearalenone (ZEN) and deoxynivalenol (DON), which were first regulated in 2014, were found in 110 and 71 samples, respectively. Apart from regulated mycotoxins, a broad range of non-regulated metabolites, from Aspergillus, Fusarium, Alternaria, Penicillium and other microbes, were also detected in maize sample. Fusarin C, a possible carcinogenic compound to humans, produced by Fusarium species and not addressed by Brazilian legislation, was detected in 54.2% of maize samples. All analysed maize samples were found to be contaminated by at least ten different metabolites, with the largest number of metabolites found in the same sample being 51.  相似文献

Mycotoxins are secondary metabolites produced by filamentous fungi that usually contaminate food products. Coffee is a natural product susceptible to mycotoxin contamination. The present study evaluates the presence of nivalenol, deoxynivalenol, T-2 and HT-2 Toxin, diacetoxyscirpenol, aflatoxin B1, aflatoxin B2, aflatoxin G1, aflatoxin G2, fumonisin B1, fumonisin B2, ochratoxin A, zearalenone, enniatin A, enniatin A1, enniatin B, enniatin B1, and beauvericin in coffee samples, using liquid chromatography tandem mass spectrometry (LC-MS/MS). The results show that zearalenone was not present in any sample. In the positive samples the contents of fumonisins ranged from 58.62 to 537.45 μg/kg, emerging mycotoxins ranged from 0.10 to 3569.92 μg/kg, aflatoxins ranged from 0.25 to 13.12 μg/kg, and trichothecenes, excepting nivalenol, ranged from 5.70 to 325.68 μg/kg. Nivalenol presented the highest concentrations, from 0.40 to 25.86 mg/kg. Ochratoxin A ranged from 1.56 to 32.40 μg/kg, and five samples exceeded the maximum limit established by the European Commission.  相似文献

The natural occurrence of mycotoxins in maize in Argentina, from 1999 to 2010 was analysed. Total aflatoxins, deoxynivalenol and zearalenone, were detected and quantified by TLC. Each aflatoxins and fumonisins was quantified by HPLC. A total of 3246 samples for freshly harvested (1655) and storage (1591) was obtained from different regions, from 1999 to 2010. Except for 2003 year in harvest, and for 2007 in storage, aflatoxins levels were low. Average values of aflatoxin B1 for freshly harvested samples were between 0.38 and 2.54 μg kg−1 and for storage samples were between 0.22 and 4.5 μg kg−1. The average values and frequency of contamination for zearalenone and deoxynivalenol, were low for all years. The average zearalenone contamination in samples of freshly harvested, showed values from no detected up to 83 μg kg−1, and in storage samples, from no detected to 17 μg kg−1. The average deoxynivalenol contamination of freshly harvested, showed values from no detected up to 140 μg kg−1, and for storage samples showed values from no detected to 14 μg kg−1. The percentage of maize samples contaminated by fumonisins was between 90 and 100% for all years studied; the average levels were from 1773 to 9093 μg kg−1 for freshly harvested maize and for storage from 2525 to 11,528 μg kg−1.Co-occurrence of aflatoxins and fumonisin was the most frequent (8.4%), followed by zearalenone and fumonisins (2%).  相似文献

Aflatoxins are a group of carcinogenic compounds produced by Aspergillus fungi that can grow on different agricultural crops. Both acute and chronic exposure to these mycotoxins can cause serious illness. Due to the high occurrence of aflatoxins in crops worldwide fast and cost-effective analytical methods are required for the identification of contaminated agricultural commodities before they are processed into final products and placed on the market. In order to provide new tools for aflatoxin screening two prototype fast ELISA methods: one for the detection of aflatoxin B1 and the other for total aflatoxins were developed. Seven monoclonal antibodies with unique high sensitivity and at the same time good cross-reactivity profiles were produced. The monoclonal antibodies were characterized and two antibodies showing IC50 of 0.037 ng/mL and 0.031 ng/mL for aflatoxin B1 were applied in simple and fast direct competitive ELISA tests. The methods were validated for peanut matrix as this crop is one of the most affected by aflatoxin contamination. The detection capabilities of aflatoxin B1 and total aflatoxins ELISAs were 0.4 μg/kg and 0.3 μg/kg for aflatoxin B1, respectively, which are one of the lowest reported values. Total aflatoxins ELISA was also validated for the detection of aflatoxins B2, G1 and G2. The application of the developed tests was demonstrated by screening 32 peanut samples collected from the UK retailers. Total aflatoxins ELISA was further applied to analyse naturally contaminated maize porridge and distiller's dried grain with solubles samples and the results were correlated with these obtained by UHPLC-MS/MS method.  相似文献

In this study, the co-occurrence of multiple mycotoxins in maize kernels collected from 300 households' stores in three agro-ecological zones in Tanzania was evaluated by using ultra high performance liquid chromatography/time-of-flight mass spectrometry (TOFMS) with a QuEChERS-based procedure as sample treatment. This method was validated for the analysis of the main eleven mycotoxins of health concern that can occur in maize: aflatoxin B1 (AFB1), aflatoxin B2 (AFB2), aflatoxin G1 (AFG1), aflatoxin G2 (AFG2), ochratoxin A (OTA), deoxynivalenol (DON), fumonisin B1 (FB1), fumonisin B2 (FB2), HT-2 toxin, T-2 toxin and zearalenone (ZEN). From each zone one major maize producing district for home consumption was chosen and 20 villages for each district were randomly selected for sampling. All mycotoxins of health concern, except for T-2 toxin, were detected in the maize samples. Particularly high levels of AFB1 (50%; 3–1,081 μg kg−1), FB1 (73%; 16–18,184 μg kg−1), FB2 (48%; 178–38,217 μg kg−1) and DON (63%; 68–2,196 μg kg−1) were observed. Some samples exceeded the maximum limits set in Tanzania for aflatoxins or in European regulations for other mycotoxins in unprocessed maize. Eighty seven percent of samples were contaminated with more than one mycotoxin, with 45% of samples co-contaminated by carcinogenic mycotoxins, aflatoxins and fumonisins. Significant differences in contamination pattern were observed among the three agro-ecological zones. The high incidence and at high levels (for some) of these mycotoxins in maize may have serious implications on the health of the consumers since maize constitute the staple food of most Tanzanian population. Effective strategies targeting more than one mycotoxin are encouraged to reduce contamination of maize with mycotoxins.  相似文献

A total of 209 samples of different groups of foods widely consumed by the Tunisian population were collected during 2004–2005 years. Samples were analyzed for contamination with aflatoxins, ochratoxin A and zearalenone, using competitive enzyme-linked immunosorbent assay (ELISA). The predominant mycotoxin was ochratoxin A with a mean level of 3.5 ± 5.3 ng g−1 in 59.8% of studied samples. Furthermore, Aflatoxins were detected in all analyzed commodities with a contamination frequency of 50.5%. In addition, aflatoxin B1 was found in 37% of the samples. The zearalenone was detected around 15% with a mean level of 10.4 ± 11.8 ng g−1. Species, dried fruits and sorghum were the most contaminated samples by aflatoxin and ochratoxin mycotoxins, whereas Rice was the least contaminated commodity. The most frequent mycotoxins co-occurrence included aflatoxins and ochratoxin A, which have been detected in 33.8% of analyzed samples. Furthermore, the simultaneous contamination by aflatoxins, ochratoxin A and zearalenone was observed in 7.2% of studied samples.  相似文献

An ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) method is described for simultaneous determination of aflatoxins (AFB1, AFB2, AFG1 and AFG2), ochratoxin A (OTA), zearalenone (ZEA), deoxynivalenol (DON), fumonisins (FB1 and FB2), T-2 and HT-2 toxins in cereals. Mycotoxins were separated by reverse phase liquid chromatography (RP-LC) and detected by tandem mass spectrometry using an electro spray-ionization interface (ESI) in both positive- and negative- ion modes. The mean recoveries of mycotoxins from spiked cereals ranged from 83.5% to 107.3%, whereas the limit of detection (LOD) and limit of quantification (LOQ) ranged from 0.01 to 25 ng/g and 0.02-40 ng/g, respectively. The multi-mycotoxin method developed in this work was applied for the simultaneous determination of mycotoxins in 80 cereal samples collected from Malaysian markets. A total of 60 cereal samples (75%) were contaminated with at least one of these mycotoxins at levels greater than the LOD. Only one maize sample and two rice samples were contaminated at levels exceeding the European regulatory limits for aflatoxins and OTA (4 and 5 ng/g, respectively). The rates of the occurrence of mycotoxins in the commercial cereal samples were 50, 30, 19, 30, 16, 14, 14 and 12% for the aflatoxins (the total amount of AFB1, AFB2, AFG1 and AFG2), OTA, ZEA, DON, FB1, FB2, T-2 and HT-2 toxins, respectively. The results demonstrated that the procedure was suitable for the simultaneous determination of these mycotoxins in cereals and could be performed for their routine analysis in mycotoxin laboratories.  相似文献

An accurate and rapid LC-ESI-MS/MS analytical method was developed and validated for the simultaneous determination of aflatoxin B1, B2, G1 and G2 in lotus seeds. The samples were firstly extracted with methanol-water solution (80:20, v/v), and then cleaned up by immunoaffinity columns. The mass spectrometer was operated in the positive ionization electrospray (ESI+) mode using multiple reaction monitoring (MRM) for analysis of four aflatoxins. The transitions of m/z 313 → 285 (AFB1, CE 33 eV), m/z 315 → 259 (AFB2, CE 37 eV), m/z 329 → 243 (AFG1, CE 37 eV) and m/z 331 → 257 (AFG2, CE 37 eV) were used to quantify these four aflatoxins, respectively. The limits of detection (LODs) of aflatoxin B1, B2, G1 and G2 were 0.007, 0.005, 0.003 and 0.005 μg kg?1 based on a signal-to-noise ratio of 3:1, respectively. The limits of quantification (LOQs) of aflatoxin B1, B2, G1 and G2 were 0.02, 0.015, 0.01 and 0.015 μg kg?1 based on a signal-to-noise ratio of 10:1, respectively. Recoveries for samples of spiked lotus seeds were all above 66% with relative standard deviation all below 15% for all compounds. Nineteen out of twenty batches of lotus seeds collected from different drug stores or markets in China were found to be contaminated with aflatoxins at different levels ranging from 0.02 to 688.4 μg kg?1.  相似文献

The aim of this study was to optimize and validate a powerful method for the simultaneous analysis of aflatoxins B1 (AFB1), B2 (AFB2), G1 (AFG1), G2 (AFG2) and ochratoxin A (OTA) in ginger and related products collected from local markets in Beijing, China. The optimized analytical procedure was based on immunoaffinity column (IAC) clean-up, followed by ultra-high performance liquid chromatography with fluorescence (UHPLC-FLR) detection. Limits of detection (LOD) and quantification (LOQ) for the five mycotoxins were 0.005–0.2 and 0.0125–0.5 μg kg−1, respectively. The average recoveries ranged from 84.2 to 97.3% with relative standard deviations (RSDs) from 0.63 to 7.86% at three spiking levels. Good linearity was observed for the analytes with correlation coefficients all higher than 0.9995. The established method was applied to 30 samples of 10 different species of ginger and related products, and all positive samples were confirmed by liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). The results showed that 5 samples of ginger products were contaminated with AFB1 at 0.13–1.38 μg kg−1, while 3 samples of ginger and 2 samples of ginger products were contaminated with OTA at 0.31–5.17 μg kg−1. Gamma ray was applied to reduce mycotoxins, i.e. ochratoxin A (OTA) and aflatoxins B1, B2, G1 and G2 (AFB1, AFB2, AFG1 and AFG2) in black pepper. Response surface methodology (RSM) was applied to evaluate the effect of dose of gamma ray ranging from 0 to 60 kGy and mycotoxin concentration ranging from 10 to 100 ng g−1 on the mycotoxin reduction. The maximum reduction was found at 60 kGy which was 52%, 43%, 24%, 40% and 36% for OTA, AFB1, AFB2, AFG1 and AFG2, respectively. Results showed the gamma rays even at 60 kGy were not effective in completely destroying of ochratoxin and aflatoxins.  相似文献

Forty-four pre- and post-nature drying maize kernels were collected from North China Plain and assessed for fungi infection and mycotoxins contamination. The percentage of fungi infection was significantly higher in pre-nature drying samples than post-nature drying samples except for Fusariuim graminearum, which increases from 6.06% to 24.09%. Fusarium, Aspergillus, Alternaria and Trichoderma were main genera. Fusarium verticillioides (24.77%) and F. graminearum (15.08%) were predominant species, followed by Aspergillus niger (7.51%) and Aspergillus flavus (4.93%). FB1 and DON were the major mycotoxins presented in the samples, followed by ZEN. All samples showed FB1 ranging from 16.5 to 315.9 μg/kg. All post-nature drying maize kernels showed DON ranging from 5.8 to 9843.3 μg/kg, while 7 of 22 pre-nature drying samples contaminated with DON ranging from 50.7 to 776.6 μg/kg. The samples contaminated with ZEN in pre- and post-nature drying maize were 3, with the content ranging from 60.5 to 147.6 μg/kg and from 40.7 to 1056.8 μg/kg, respectively. Only 1 sample contaminated with AFB1 of 148.4 μg/kg. The occurrence of mycotoxins is highly in accordance with the incidence of the corresponding mycotoxin-producing fungi. This is the first comprehensive comparison of fungi infection and mycotoxins contamination between pre- and post-nature drying maize kernels.  相似文献

Fourteen spice samples were collected from local markets in Doha, Qatar, during 2012, and were surveyed for the presence of potentially harmful mycoflora and for contamination with aflatoxins B1, B2, G1, and G2 by high-performance liquid chromatography (HPLC). Among the tested spice samples, chili powder showed the highest presence of fungal propagules, while ginger, curry and garlic samples did not present any fungal contamination. A total of 120 isolates, mostly belonging to Aspergillus and Penicillium genera, were collected and 33 representative species were identified by amplification and sequencing of the internal transcribed spacer (ITS) region. Aspergillus flavus, Aspergillus nomius and Aspergillus niger were the most dominant. Thirty-seven Aspergillus strains were screened for their potential to produce aflatoxins using biochemical and molecular tools: only 9 A. flavus strains showed both fluorescence and amplification with all the three primers targeting aflP, aflM and aflR genes. Aflatoxins were detected in five spices (black pepper, chili, tandoori masala. turmeric and garam masala), and with the exception of garam masala, the tested samples of turmeric, black pepper, tandoori masala and chili powder exceeded B1 and/or total aflatoxin maximum levels. Our results demonstrate the potential for mycotoxin biosynthesis by fungi contaminating imported spice products.  相似文献

Studies conducted in Benin, in which the main staple foods are maize, cassava, groundnuts and yams, showed high levels of aflatoxin residues in blood of the exposed population. The natural contamination with fungi and aflatoxins in cassava chips sold at markets in Benin, West Africa was investigated. A total of sixty samples were sampled from open markets in 11 districts of 3 agroecological zones and analyzed for the presence of mycoflora and aflatoxin B1, B2, G1 and G2. Fourteen genera of fungi were associated with marketed dried cassava chips. Within these, twenty- two isolates were identified to species level, whereas four were identified only to genus. The dominating fungal species isolated were Rhizopus oryzae, Nigrospora oryzae, Chrysonilia sitophila, Cladosporium resinae, Cladosporium herbarum, Apergillus niger and Aspergillus flavus. Fifty-four out of sixty samples were contaminated with A. flavus. The rate of occurrence in CFU/g of A. flavus fungi was lower than for all other fungal species together. Aflatoxin was not detected in any of the samples analyzed using HPLC with post-column photochemical derivatization and fluorescence detection. The limit of detection (LOD) was 0.1 μg/kg. Results from this study suggest cassava chips are unlikely to be a source of aflatoxin in Benin, and that other staples such as maize and groundnuts are more important in aflatoxin exposure. Therefore it can be speculated that staples like maize and groundnut are more important in aflatoxin exposure.  相似文献

Sixty samples of cereals (20 of corn, 20 of barley, and 20 of wheat) and 55 samples of spices (14 of paprika, 12 of ginger, 14 of cumin, and 15 of pepper) purchased from popular markets of Rabat and Salé in Morocco were analyzed for mycotoxins.Cereals samples were all analyzed for ochratoxin A (OTA). The average levels of contamination were 1.08, 0.42, and 0.17 μg/kg for corn, wheat, and barley, respectively. Samples of corn were also analyzed for zearalenone (ZEA) and fumonisin B1 (FB1) the average contaminations were 14 and 1930 μg/kg, respectively. Co-occurrence of OTA, FB1, and ZEA was also checked. Spices samples were analyzed only for aflatoxins (AFs) and the average contaminations found for aflatoxin B1 (AFB1) were 0.09, 0.63, 2.88 and 0.03 μg/kg for black pepper, ginger, red paprika and cumin, respectively. The higher level of contamination was found in red paprika (9.68 μg/kg).The present report is the first one ever drafted on the natural co-occurrence of OTA, FB1 and ZEA in cereals and on the occurrence of AFs in spices from Morocco.  相似文献

Edible nuts imported in Qatar from June 1997 to December 1998 were anlaysed for aflatoxins. Eighty-one nut samples were analysed in the second half of 1997 and contamination was detected in 19 samples with total aflatoxin level varied from a low of about 0.53 to a high of 289 μg/kg. Aflatoxin contamination was detected in pistachios and peanuts, while other nuts such as almond, cashew nut, walnut and hazel nut were found free from aflatoxins. During 1998 testing was carried out only for pistachios and about 101 samples were analysed; contamination was detected in 48 samples with total aflatoxin level in the range of 1.2–275 μg/kg. In pistachios without shell level of contamination was very high (total aflatoxin 8.3–275 μg/kg) compared to pistachio with shell (total aflatoxin 1.2–75 μg/kg). Aflatoxin B1 and B2 were detected in all the contaminated samples of pistachios, whereas aflatoxin G1 and G2 were detected only in three samples of pistachios at the level of 0.8–1.9 μg/kg for aflatoxin G1 and 0.4–1.3 μg/kg for aflatoxin G2.  相似文献

Cereals products enriched with leguminous or oleaginous are used in Burkina Faso as food complement or to avoid infant malnutrition. Infant formulas from cereals are consumed as food supplements after the weaning age. They are produced in Burkina Faso with a mixture of several cereals (maize, millet, rice, or sorghum), leguminous (bean), oleaginous (peanut, soya or sesame), sugar or salt and sometimes milk powder. The production is artisanal or semi-industrial. These infant foods from cereals should be free from mycotoxins and pathogenic bacteria. The objective of this work was to determine the occurrence of mycotoxins like aflatoxins, ochratoxin A and fumonisins in infant formulas and grains in Ouagadougou (Burkina Faso). The mycotoxins (aflatoxins, ochratoxin A and fumonisins) were determined by HPLC-FLD in 248 samples of infant formulas produced by the Recovery and Nutrition Education Centers (CRENs) and sold on the market places in Burkina Faso.Results showed that the majority of samples of infant formulas presented high levels of mycotoxins. The frequency of contaminated samples by aflatoxin B1, ochratoxin A and fumonisins in analyzed samples were 83.9% (167/199), 7.5% (15/199) and 1.5% (3/199), respectively. The highest values registered in infant formulas were 900, 6 and 3 times higher for aflatoxin B1 (EU limit: 0.1 μg/kg), ochratoxin A (EU limit: 0.5 μg/kg) and fumonisins (EU limit: 200 μg/kg), respectively, than the EU regulation limits (1881/2006). This study presents the first results concerning the safety assessment of infant formulas regarding mycotoxins in Burkina Faso.  相似文献

The presence of aflatoxins, Aspergillus flavus and Aspergillus parasiticus in dried fruits was investigated. A total of 62 dried fruit samples were analyzed (24 black sultanas, 19 white sultanas and 19 dried figs). A total of 10 A. flavus isolates were found, nine in one white sultana sample (corresponding to 18% infection) and one isolate in dried figs (2%), and all of them were aflatoxin B1 and B2 producers. A. parasiticus was not found. Aflatoxins were detected in 3 of 19 (16%) white sultana samples analyzed and, the limits were not higher than 2.0 μg/kg. In dried figs 11 of 19 (58%) samples were contaminated with aflatoxins and, with exception of one sample that was contaminated with 1500 μg/kg of B1 aflatoxin, the others had less than 2.0 μg/kg. Neither aflatoxigenic or aflatoxins contaminated black sultanas.  相似文献

A survey in African snacks was carried out in order to evaluate the intake of 23 mycotoxins. The African snack samples were purchased from street vendors within Lagos metropolis (Nigeria) and evaluated for the presence of 23 mycotoxins using a modified QuEChERS procedure coupled with liquid chromatography-triple quadrupole linear ion trap mass spectrometer. The snacks included akara, baked coconut, coconut candy, donkwa, groundnut cake (kulikuli), lafun, milk curd (wara), fresh and dried tiger-nuts, and yam flour. Only three mycotoxins were detected in 23.8% of the studied snacks, and at concentrations ranging from 6 to 54 μg kg−1. The concentrations of aflatoxin B1 (AFB1) and AFB2 reached 23 μg kg−1 and 3 μg kg−1, respectively. Moreover a sample of baked coconut contained α-zearalenol (α-ZOL), which was up to 54 μg kg−1 in coconut candy. As considers prevalence, aflatoxins and α-ZOL were not detected in lafun and groundnut-based snacks (donkwa and kulikuli), whereas each of the three mycotoxins contaminated 12.5% (1/8) of the coconut-based samples. This is the first report of α-ZOL in cassava and coconut, and their products. AFB1 and total aflatoxins (TAFs) concentrations exceeded the maximum allowable limit recommended by National Agency for Food and Drug Administration and Control Nigeria (NAFDAC) in one sample of baked coconut (AFB1 = 23 μg kg−1 and TAFs = 26 μg kg−1) and donkwa (AFB1 = 19 μg kg−1 and TAFs = 21 μg kg−1).  相似文献

Residents of certain areas of Tanzania are exposed to mycotoxins through the consumption of contaminated maize based foods. In this study, 101 maize based porridge samples were collected from villages of Nyabula, Kikelelwa and Kigwa located in different agro-ecological zones of Tanzania. The samples were collected at three time points (time point 1, during maize harvest; time point 2, 6 months after harvest; time point 3, 12 months after harvest) over a 1-year period. Ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) was used to detect and quantify 9 mycotoxins: aflatoxin B1 (AFB1), aflatoxin B2 (AFB2), aflatoxin G1 (AFG1), aflatoxin G2 (AFG2), fumonisin B1 (FB1), fumonisin B2 (FB2), deoxynivalenol (DON), ochratoxin A (OTA) and zearaleneone (ZEN) in the samples following a QuEChERS extraction method. Eighty two percent of samples were co-contaminated with more than one group of mycotoxins. Fumonisins (FB1 + FB2) had the highest percentage occurrence in all 101 samples (100%) whereas OTA had the lowest (5%). For all three villages the mean concentration of FB1 was lowest in samples taken from time point 2. Conversely, In Kigwa village there was a distinct trend that AFB1 mean concentration was highest in samples taken from time point 2. DON concentration did not differ greatly between time points but the percentage occurrence varied between villages, most notably in Kigwa where 0% of samples tested positive. ZEN occurrence and mean concentration was highest in Kikelelwa. The results suggest that mycotoxin contamination in maize can vary based on season and agro-ecological zones. The high occurrence of multiple mycotoxins found in maize porridge, a common weaning food in Tanzania, presents a potential increase in the risk of exposure and significant health implications in children.  相似文献
