Several theorists have proposed that the sex steroid testosterone acts on the fetal brain during a critical period of development to influence cerebral lateralization (N. Geschwind & A. M. Galaburda, 1987; M. Hines & C. Shipley, see PA, Vol 71:8996; S. F. Witelson, see PA, Vol 79:26441. In the present study. relations were examined between prenatal testosterone levels in 2nd trimester amniotic fluid and lateralization of speech, affect, and handedness at age 10. Girls with higher prenatal testosterone levels were more strongly right-handed and had stronger left-hemisphere speech representation. Boys with higher prenatal testosterone levels had stronger right-hemisphere specialization for the recognition of emotion. This pattern of results is most consistent with Witelson's (1991) claim that prenatal testosterone leads to greater lateralization of function. Intellectually challenged adults, with and without Down's syndrome (DS), pointed to drawings of animals following the dichotic presentation of animal names. Although there were no reliable ear differences between the 2 groups, there was tremendous between-persons variability within the group of participants with DS. Moreover, left-ear advantages in persons with DS were associated with more speech production errors when participants either read or repeated a string of 1 syllable words. This was not true for control participants. The relationship between ear advantage and speech errors in persons with DS could reflect their unique pattern of cerebral specialization and brain development. This article examines caregiver speech to young children. The authors obtained several measures of the speech used to children during early language development (14-30 months). For all measures, they found substantial variation across individuals and subgroups. Speech patterns vary with caregiver education, and the differences are maintained over time. While there are distinct levels of complexity for different caregivers, there is a common pattern of increase across age within the range that characterizes each educational group. Thus, caregiver speech exhibits both long-standing patterns of linguistic behavior and adjustment for the interlocutor. This information about the variability of speech by individual caregivers provides a framework for systematic study of the role of input in language acquisition. BACKGROUND: In previous studies on the use of natriuretic peptides to detect left-ventricular systolic dysfunction, a higher rate of cardiac disorders in the control groups than in the study groups could have led to bias. We investigated the effectiveness of plasma N-terminal atrial natriuretic peptide (NT-ANP) and brain natriuretic peptide (BNP) concentrations to show left-ventricular systolic dysfunction in a random sample of the general population. METHODS: We randomly selected 2000 participants aged 25-74 years from family physicians' lists in Glasgow, UK. We sent all participants questionnaires. 1653 respondents underwent echocardiography and electrocardiography. We took a left-ventricular ejection fraction of 30% or less to show left-ventricular systolic dysfunction. NT-ANP and BNP were measured in plasma by RIAs. FINDINGS: 1252 participants had analysable electrocardiograms and echocardiograms, completed questionnaires, and available blood samples. Median concentrations of NT-ANP and BNP were significantly higher in participants with left-ventricular systolic dysfunction (2.8 ng/mL [IQR 1.8-4.6] and 24.0 pg/mL [18.0-33.0]) than in those without (1.3 ng/mL [0.9-1.8] and 7.7 pg/mL [3.4-13.0]; each p < 0.001). Among participants with left-ventricular systolic dysfunction, both symptomatic and asymptomatic subgroups had raised NT-ANP and BNP concentrations. A BNP concentration of 17.9 pg/mL or more gave a sensitivity of 77% and specificity of 87% in all participants, and 92% and 72% in participants aged 55 years or older. INTERPRETATION: Measurement of BNP could be a cost-effective method of screening for left-ventricular systolic dysfunction in the general population, especially if its use were targeted to individuals at high risk.  相似文献

In examining the prediction that left cerebral dominance, as indexed by hand and sighting preference, should be a positive accompaniment of speech learning in individuals whose cerebral speech areas are likely to be in the left hemisphere, it was found that over a 10-12 year period of formal education right-handed-right-sighting deaf students consistently earned higher speech grades than their right-handed-left sighting and right-handed-mixed-sighting counterparts matched on the basis of hearing loss. This finding supports the hypothesis that the cerebral mechanisms relevant to speech acquisition are simplified, facilitated, and/or less prone to interference when control of speech, hand, and eye is localized primarily in 1 hemisphere of the brain. Objective: To examine cardiovascular reactivity and recovery to laboratory stress among a naturalistic sample of individuals diagnosed with major depressive disorder (MDD) and healthy control participants. Prospective evidence suggests that MDD confers risk for cardiovascular disease equal to or greater than the risk associated with depressed mood. Enhanced cardiovascular reactivity has been proposed as a mechanism explaining increased risk, but data are inconsistent as to whether depressed individuals exhibit enhanced or attenuated reactivity. Further, few studies have examined appraisal and recovery differences. Design: Participants diagnosed with MDD (N = 25) and healthy control participants (N = 25) engaged in a cardiovascular reactivity protocol including 2 tasks, each followed by a brief recovery period. Main outcome measures: Blood pressure, heart rate, pre-ejection period, cardiac output and total peripheral resistance were assessed. Appraisals of tasks were assessed prior to each task. Results: Depressed participants exhibited significantly less systolic blood pressure, heart rate, and cardiac output reactivity during speech, less heart rate reactivity during mirror tracing, and less heart rate recovery after speech and mirror tracing than controls. Depressed participants appraised the tasks as more demanding, threatening, and stressful and reported being less able to cope than controls. Appraisals were related to heart rate reactivity, but appraisals did not mediate the relationship between depression group and reactivity. Conclusion: Impaired recovery rather than exaggerated cardiovascular reactivity may partially explain the increased prospective cardiovascular disease risk in depressed individuals. Monkeys and children show sex differences on tasks that depend on the orbital prefrontal cortex. To determine whether similar sex differences exist across the life span, adults were tested on an orbital-dependent decision-making task, the Iowa Card Task, as well as on a control task, the California Weather Task. In addition, estradiol, progesterone, and testosterone were assayed. The 6 groups of participants were college-age men, older men, young low-hormone (menstruating) women, young high-hormone (midluteal) women, older postmenopausal women on estrogen replacement therapy (ERT), and older postmenopausal women not on ERT. Results showed a male superiority on the Iowa Card Task. Among college-age men there was a negative correlation between performance and testosterone levels. There were no significant differences among groups of women on the card task. There were no significant sex differences or hormone correlations on the California Weather Task. To examine brain patterns of metabolic and functional activity, the distribution of cytochrome oxidase, a mitochondrial enzyme marker for neuronal functional activity, was mapped throughout the rat brain. Mapping was done qualitatively by enzyme histochemistry of brain sections cut in three planes (coronal, sagittal and horizontal), and quantitatively by optical densitometry of stained sections and by biochemical assays of brain tissue homogenates. Activity of the enzyme was distributed in characteristic patterns and amounts that differed among various neural pathways, brain nuclei, cerebral cortical areas and layers, and neuron types. Gray matter essentially always had higher enzyme activity than did white matter, by a factor of eight- to 12-fold. Among different neural pathways, cytochrome oxidase activity was relatively high in special sensory, somatosensory and motor systems, and was relatively low in associative, limbic, autonomic and visceral regulatory systems (though exceptional areas were present). Among 11 different neuron types, nearly a two-fold range of histochemical staining intensities was observed, with the darkest staining in neurons of the mesencephalic trigeminal nucleus. The observed patterns of cytochrome oxidase activity were mostly similar to the patterns of 2-deoxyglucose uptake seen previously [Schwartz W. J. and Sharp F. R. (1978) J. comp. Neurol. 177, 335-360; Sokoloff L. et al. (1977) J. Neurochem. 28, 897-916] in conscious, "resting" animals, though some differences were found. For example, whereas 2-deoxyglucose uptake was about three-fold higher in gray matter than in white matter [Sokoloff L. et al. (1977) J. Neurochem. 28, 897-916], cytochrome oxidase activity was about eight- to 12-fold higher. This and other discrepancies probably reflect basic technical differences between these two methods. Compared to 2-deoxyglucose, cytochrome oxidase is more specific for oxidative rather than glycolytic metabolism, and more reflective of overall neuronal functional activity occurring over longer time periods lasting hours to weeks, rather than minutes. The anatomical resolution of cytochrome oxidase histochemistry is also finer than that of 2-deoxyglucose autoradiography, extending to the electron microscopic level. The metabolic map of cytochrome oxidase activity reveals patterns of normal brain function, and may be useful as a baseline for comparison in studies of brain disease, development, ageing and plasticity.  相似文献

Sexual differentiation of rodent brain is dependent upon hormonal exposure during a "critical period" beginning in late gestation and ending in early neonatal life. Steroid hormone action at this time results in anatomical and physiological sexual dimorphisms in adult brain, but the mechanism mediating these changes is essentially unknown. The inhibitory neurotransmitter, GABA, is involved in regulation of sexually dimorphic patterns of behavior and gonadotropin secretion in the adult. Recent evidence suggests that during development GABA is excitatory and provides critical neurotrophic and neuromodulatory influences. We hypothesized that steroid-induced changes in GABAergic neurotransmission during this critical period are important mediators of sexual differentiation in brain. Therefore, we quantified levels of mRNA for GAD, the rate-limiting enzyme in GABA synthesis. On Postnatal Day 1, males had significantly higher levels of GAD mRNA in the dorsomedial nucleus, arcuate nucleus, and CA1 region of hippocampus. On Postnatal Day 15, after the critical period for sexual differentiation has ended, these differences were no longer present. We examined the role of gonadal steroids in regulating GAD by removing testes of males and administering testosterone to females at birth. Exposure to testosterone was correlated with increased GAD mRNA in the dorsomedial nucleus. A sex difference in GAD mRNA was also observed in the medial preoptic area, but the influence of testosterone was inconclusive. We conclude that sex differences in the GABAergic system during development are partially hormonally mediated, and that these differences may contribute to the development of sexually dimorphic characteristics in adult brain.  相似文献

In this study, the psychological symptom patterns of individuals with chronic pain on the Brief Symptom Inventory (BSI) were cluster analyzed. Cluster analysis was initially performed on a sample of 1,489 people with chronic pain and then cross-validated on a smaller sample of 244 people. A 2-cluster solution was deemed most appropriate. The clusters reflected low- and high-profile elevations on all BSI subscales. As part of their clinical evaluation, participants also completed the Pain Disability Index, the adjective list from the McGill Pain Questionnaire, and a modified version of the Posttraumatic Chronic Pain Test. Among persons in the smaller sample, high-profile participants were more likely to be involved in litigation, report a higher frequency of posttraumatic stress symptoms, and display poorer psychosocial functioning. The results suggest that the BSI subgroups are associated with psychosocial characteristics that may be important in terms of treatment planning and outcome. Affective startle eyeblink modulation by unipolar depressed and nondepressed participants was assessed during the anticipation and viewing of emotional pictures. Anticipatory startle probes were presented at 2,000 ms and 750 ms before picture onset. Startle probes during picture viewing were presented at 300 ms and 3,500-4,500 ms after picture onset. Although nondepressed participants demonstrated the predicted quadratic and linear patterns of responding in the 2,000-ms anticipatory and 3,500-4,500-ms viewing conditions, respectively, depressed participants were not significantly responsive to differences among picture valence categories at these probe conditions. There were no between-groups differences in startle modulation at the other two probe intervals, in picture ratings, or in behavioral responses to pictures. There was also little evidence of hyperresponsivity to negatively valenced stimuli in the depressed group. These results indicate that depression-related affective hyporesponsivity extended to startle modulation but that the nature and magnitude of the differences between depressed and nondepressed individuals were conditional on the specific cognitive and motivational processes recruited at different points in time. In the human prostate, various androgen-metabolizing enzymes are present. Among these enzymes, testosterone 5 alpha-reductase seems to be dominant. However, androstenedione is also a potential substrate of the prostatic 5 alpha-reductase. To address the question of to what extent the reduction of androstenedione to androstanedione occurs, the present study describes in detail the kinetic characteristics (Km and Vmax) and possible age-dependent alterations of this enzymatic step in epithelium and stroma of the human prostate. In normal prostate (NPR), the mean Km (nM) and Vmax (pmol/mg protein.h) were about twofold higher in stroma (Km, 211; Vmax, 130) than in epithelium (Km, 120; Vmax, 56), whereas in the benign prostatic hyperplasia (BPH), the mean Km (nM; mean +/- SEM) and Vmax (pmol/mg protein.h; mean +/- SEM) were about sixfold higher in stroma (Km, 668 +/- 121; Vmax, 415 +/- 73) than in epithelium (Km, 120 +/- 10; Vmax, 73 +/- 8). In BPH, those differences between epithelium and stroma were highly significant (p < 0.001). However, the efficiency ratios (Vmax/Km) of neither BPH nor NPR showed any significant differences between epithelium (NPR, 0.47; BPH, 0.62 +/- 0.06) and stroma (NPR, 0.70; BPH, 0.63 +/- 0.05). With respect to age-related changes, only stroma showed a significant increase of Km (p < 0.01) and Vmax (p < 0.05) with age. In summary, in both epithelium and stroma of the human prostate, a 5 alpha-reductase converts in measurable amounts androstenedione to androstanedione. The kinetic data were, in part, different between epithelium and stroma; the reason for this difference remains unclear. In comparison to other metabolic conversions, such as testosterone to dihydrotestosterone and androstenedione to testosterone, it is unlikely that, in the human prostate, the adrenal androgen androstenedione contributes significantly to the formation of testosterone and, further, of dihydrotestosterone.  相似文献

Adaptive linear filtering can improve effective speech-to-noise ratios by attenuating spectral regions with intense noise components to reduce the noise's spread of masking onto speech in neighboring regions. This mechanism was examined in static listening conditions for seven individuals with sensorineural hearing loss. Subjects were presented with nonsense syllables in an intense octave-band noise centered on 0.5, 1, or 2 kHz. The nonsense syllables were amplified to maximize. the articulation index; the noises were the same for all subjects. The processing consisted of applying frequency-selective attenuation to the speech-plus-noise with the goal of attenuating the frequency region containing the noise by various amounts. Consonant recognition scores and noise masking patterns were collected in all listening conditions. When compared with masking patterns obtained from normal-hearing subjects, all hearing-impaired subjects had higher masked thresholds at frequencies below, within, and above the masker band except for one subject who demonstrated additional masking above the masker only. Frequency-selective attenuation resulted in both increases and decreases in consonant recognition scores. Increases were associated with a release from upward spread of masking. Decreases were associated with applying too much attenuation such that speech energy within the masker band that was audible before processing was partially below threshold after processing. Fletcher's [Speech and Hearing in Communication (Van Nostrand, New York, 1953)] version of articulation theory (without modification) accounted for individual subject differences within the range of variability associated with the consonant recognition test in almost every instance. Hence, primary factors influencing speech reception benefits are characterized by articulation theory. Fletcher's theory appears well-suited to guide the design of control algorithms that will maximize speech recognition for individual listeners.  相似文献

High-span individuals (as measured by the operation span [OSPAN] technique) are less likely than low-span individuals to notice their own names in an unattended auditory stream (A. R. A. Conway, N. Cowan, & M. F. Bunting, 2001). The possibility that OSPAN accounts for individual differences in auditory distraction on an immediate recall test was examined. There was no evidence that high-OSPAN participants were more resistant to the disruption caused by irrelevant speech in serial or in free recall. Low-OSPAN participants did, however, make more semantically related intrusion errors from the irrelevant sound stream in a free recall test (Experiment 4). Results suggest that OSPAN mediates semantic components of auditory distraction dissociable from other aspects of the irrelevant sound effect. This study examined whether highly cynical individuals benefit less from social support during an acute stressor than individuals low in cynicism. College students (52 men, 52 women) performed a stressful speech task alone or in the presence of a supportive confederate. There was an interactive effect of social support and cynicism on cardiovascular reactivity: Low cynicism participants who received support had smaller increases in blood pressure during the speech than low cynicism participants without support and high cynicism participants with or without support. Participants' psychological stress appeared to mediate the main effects of support on blood pressure reactivity, but not the Support?×?Cynicism interaction. This study aimed to correlate testosterone levels with natural written language in 2 people undergoing testosterone therapy. Two participants, a man receiving treatment for loss of upper-body strength and a female-to-male transgendered individual, supplied records of injections over 1-2 years along with e-mails or journal entries as writing samples. Results showed that higher testosterone levels correlated with reduced use of words related to social connections. Language relating to anger, sexuality, and achievement was unrelated to testosterone levels. It appears that testosterone steers attention away from social connections but not necessarily toward concerns with aggression or sexual activity. Why do some people strive for high status, whereas others actively avoid it? In the present studies, the authors examined the psychological and physiological consequences of a mismatch between baseline testosterone and a person's current level of status. The authors tested this mismatch effect by placing high and low testosterone individuals into high or low status positions using a rigged competition. In Study 1, low testosterone participants reported greater emotional arousal, focused more on their status, and showed worse cognitive functioning in a high status position. High testosterone participants showed this pattern in a low status position. In Study 2, the emotional arousal findings were replicated with heart rate, and the cognitive findings were replicated using a math test. In Study 3, the authors demonstrate that testosterone is a better predictor of behavior than self-report measures of the need for dominance. This investigation determined the influence of testosterone and neurobehavioral disinhibition (ND) on risk for substance use disorder (SUD). Testosterone level during puberty was hypothesized to promote social dominance associated with norm-violating behavior that, in turn, predisposes individuals to use of illicit drugs and, subsequently, SUD. Using a prospective paradigm, the authors recruited 179 boys (mean age=11.62 years, SD=0.88) and followed up when participants were ages 12-14, 16, 19, and 22. Results indicated that social dominance/norm-violating behavior (SD/NVB) at age 16 mediated the association between testosterone level (ages 12-14) and SUD (age 22). In addition, SD/NVB mediated the association between ND and SUD. These findings suggest that development of SUD is influenced by androgen-dependent and neurobehavioral processes via a social motivational style characterized by SD/NVB. Specific brain receptors for oxytocin have been described in several mammalian species. The distribution of these receptors differs greatly across species and in the rat, receptor binding in specific brain regions appears to depend upon gonadal steroids. This study used in vitro receptor autoradiography to examine the effects of testosterone on oxytocin receptor binding in the mouse forebrain. Three groups of male mice were compared: castrates treated with blank capsules, castrates treated with testosterone filled capsules, and intact males. Irrespective of steroid treatment, the distribution of oxytocin receptors in mouse forebrain differed markedly from patterns previously described in the rat. In addition to these species differences in receptor distribution, testosterone had effects in the mouse which differed from the induction of receptors previously reported in the rat. In the mouse ventromedial nucleus of the hypothalamus, binding in the untreated castrate males was approximately double that observed in either the intact or the testosterone-treated castrates. In other regions of the mouse brain, such as the intermediate zone of the lateral septum, binding to oxytocin receptors was increased with testosterone treatment. These results suggest that the brain oxytocin receptor varies across species not only in its distribution but also in its regional regulation by gonadal steroids. These apparently paradoxical changes in oxytocin receptor binding may result from either direct or indirect effects of gonadal steroids in mouse brain.  相似文献
