This study was aimed at assessing the prevalence of pathogenic Escherichia coli strains, relative to the total count of E. coli, faecal coliforms and other heterotrophic mesophilic aerobic bacteria (HMAB) isolated in groundwater in the equatorial region of Cameroon (Central Africa). Bacteria were isolated using standard methods. Pathogenic E. coli strains were then identified using haemagglutination and antisera tests. The maximum abundance of HMAB, faecal coliforms and E. coli strains were 4.9 × 106, 5.6 × 103 and 1 × 103 colony‐forming units (CFU)/100 mL, respectively. The count of pathogenic E. coli strains reached 3 CFU/100 mL. The counts of commensal and pathogenic E. coli strains underwent temporal and spatial fluctuations. In 21% of sampling sites, the abundance of faecal coliforms was significantly correlated to that of E. coli (P < 0.05). However, the isolated bacterial count was not significantly correlated to that of the pathogenic E. coli strains (P > 0.05). The bacteria abundance dynamics may be impacted by many interacting factors.  相似文献

An investigation of the tidal factors controlling the abundance and distribution of coliform bacteria in the sediments of two intertidal sites in the River Mersey (U.K.) was made during 1976. Samples were collected, at approximately weekly intervals, from September to December. Counts of the total coliform, faecal coliform and E. coli type I, were made in order to relate the numbers of bacteria to the salinity conditions of the tidal waters covering the sites. The use of faecal coliform to faecal streptococci ratios as indicators of the source of faecal pollution has become popular. However, this methodology has been attacked on the grounds that the differential die-away rates of FC and FS invalidate the use of the ratio. Data is presented here to show that this differential die-away can in fact strengthen the value of the FC:FS ratio as a means of distinguishing human from non-human pollution. It is concluded that initially high FC:FS values which fall indicate human contamination, whereas initially low ratios which subsequently rise suggest a non-human faecal source.  相似文献

The aim of this study was to investigate the degree of contamination of sewage sludge with ESBL-producing Escherichia coli strains and the effectiveness of different sewage sludge treatment methods.Monthly sewage sludge samples were collected between January and September 2009 in 5 different sewage treatment plants and tested for the presence of ESBL E. coli. In addition, the number of colony forming units (CFU) of E. coli and coliform bacteria before and after the different sludge treatment methods (aerobic/anaerobic digestion, lime stabilization, and thermal treatment) was investigated.Of the 72 sewage sludge samples investigated, ESBL-positive E. coli were found in 44 (61.1%) sewage sludge samples. The classification of β-lactamase groups was carried out in 15 strains resulting in the detection of 2 different groups (CTX-M and TEM) of bla genes. All 15 of them had a CTX-M gene and 4 of these strains furthermore carried a TEM gene.With regard to the CFU of E. coli and coliform bacteria, thermal treatment and lime stabilization following dehydration sufficiently reduced pathogen concentrations. The plants using merely stabilization and dehydration showed an increase of E. coli and coliform bacteria and thus also an increase in ESBL-producing E. coli.  相似文献

The present study was conducted with the aim of improving faecal coliform (FC) and faecal streptococcus (FS) removal efficiencies in tertiary maturation stages of a sewage treatment plant in Southern England, where climatic conditions are sub-optimal. The research used intensive field assessments (bacteriological, general quality and hydraulic) to identify the parameters that affect the bacteriological quality of the effluent from three parallel maturation ponds (North, Central and South) of similar geometry and dimensions. An engineering intervention was carried out to convert the South pond to three channels to increase the L/W ratio from 9:1 to 79:1. Hydraulic tracer studies in the South pond with Rhodamine WT showed that the dispersion number 'd' was reduced from 0.37 (dispersed flow) to 0.074 by this intervention under similar flow conditions (4.5l/s). Hydraulic retention time was thus increased by 5h, delay in jet flow short-circuiting was increased from 2.5 to 17.5h thus increasing the exposure times for all elements. As a result of the intervention FC removal increased substantially. Maximum channel-lagoon efficiency of 99.84% was obtained at 4.5l/s and 19 degrees C, when exposure to sunlight was 17 h in summer. It is concluded that the channel configuration produces a higher hydraulic efficiency than conventional maturation ponds. It is therefore recommended as a viable engineering solution which permits a low-cost upgrading of plant performance, requiring no additional land, and with minimal maintenance costs.  相似文献

The concentrations of coliform and Escherichia coli bacteria were determined in inshore seawater from 15 different areas, embracing more than three quarters of the coastline of Penang Island. Each area included from 3 to 20 sampling sites, each of which was sampled on at least two occasions, once at high tide and once at low tide. Samples were collected from water 100–150 mm below the surface and where, in most cases, the total water depth was about 0.6 m.Logarithmic mean concentrations in samples from areas with hinterlands having small or large human (and/or domestic animal) populations ranged from 493 to 712,000 coliform bacteria and from 72 to 234,700 E. coli bacteria per 100 ml.Samples from an area with a hinterland of natural forest and very few dwellings contained logarithmic mean concentrations of 31 coliform and less than 7 E. coli bacteria per 100 ml. The highest concentrations found in any sample from this area were 1100 coliform and 93 E. coli bacteria per 100 ml. It is suggested that the bacterial concentrations in seawater from this area may approximate to ‘base-line’ contamination levels.The overall results are considered in relation to standards applied elsewhere to bathing waters and to those applied to waters for shellfish culture.  相似文献

Incomplete elimination of bacteria and pharmaceutical drugs during wastewater treatment results in the entry of antibiotics and antibiotic-resistant bacteria into receiving streams with effluent inputs. In Mud Creek in Fayetteville, AR, ofloxacin, trimethoprim, and sulfamethoxazole have been detected in water and sediment, and tetracycline has been detected in sediment downstream of treated effluent input. These antibiotics have been measured repeatedly, but at low concentrations (< 1 μg/L) in the stream. To determine if effluent input results in detectable and stable changes in antibiotic resistances downstream of effluent input, antibiotic resistance in Escherichia coli and total coliform bacteria in Mud Creek stream water and sediment were determined using a culture-based method. Isolated E. coli colonies were characterized for multiple antibiotic resistance (MAR) patterns on solid media and to evaluate E. coli isolate richness by amplification of a partial uidA gene followed by denaturant gradient gel electrophoresis (DGGE). Despite temporal variability, proportions of antibiotic-resistant E. coli were generally high in effluent and 640 m downstream. The MAR pattern ampicillin-trimethoprim-sulfamethoxazole was associated with a DGGE profile that was detected in effluent and downstream E. coli isolates, but not upstream. Percent resistance among coliform bacteria to trimethoprim and sulfamethoxazole was higher 640 m downstream compared to upstream sediment and water (with one exception). Resistance to ofloxacin was too low to analyze statistically and tetracycline resistance was fairly constant across sites. Resistances changed from 640 m to 2000 m downstream, although dissolved nutrient concentrations within that stream stretch resembled effluent. Antibiotic resistant bacteria are entering the stream, but resistances change within a short distance of effluent inputs, more quickly than indicated based on chemical water properties. Results illustrate the difficulty in tracking the input and fate of antibiotic resistance and in relating the presence of low antibiotic concentrations to selection or persistence of antibiotic resistances.  相似文献

Bacteriological failure investigations are crucial in the provision of safe, clean drinking water as part of a process of quality assurance and continual improvement. However, the financial implications of investigating coliform and Escherichia coli failures during routine water quality monitoring are poorly understood in the industry. The investigations for 737 coliform and E. coli failures across five UK water companies were analysed in this paper. The principal components of investigation costs were staff hours worked, re-samples collected, transportation, and special investigatory activities related to the sample collection location. The average investigation costs ranged from £575 for a customer tap failure to £4,775 for a water treatment works finished water failure. These costs were compared to predictions for US utilities under the Revised Total Coliform Rule. Improved understanding of the financial and staffing implications of investigating bacteriological failures can be used to budget operational expenditures and justify increased funding for preventive strategies.  相似文献

The occurrence of faecal microorganisms such as total coliforms (TCs), Escherichia coli (E. coli), coliphages (somatic and F+ specific) and the genomes of faecal viruses: human adenovirus (HAdV), norovirus (NoV) genogroup I (GI) and II (GII), enterovirus (EV), aichi virus (AiV) and Pepper mild mottle virus (PMMoV), was explored in submarine groundwater discharges (SGDs) in the Mexican Caribbean. Ultra-filtrated water samples were subjected to nucleic acid extraction and real-time quantitative polymerase chain reaction (PCR). Results showed the presence of TC bacteria at all sites, whereas the detection of E. coli occurred only during the dry season. Coliphages occurred during the rainy and dry seasons, with concentrations ranging from 10 to 2560 plaque forming units (PFU/100 ml). HAdV and AiV were detected in 50% and 41% of the samples, at concentrations ranging from 102 to 104 genome copies per litre (GC/L) and from 102 to 106 GC/L, respectively. NoV GI and II were detected in 25% and 16% of the samples, at concentrations of 101 GC/L and 102–103 GC/L, respectively. PMMoV was detected in 50% of the samples at concentrations ranging from 101 to 103 GC/L. The EV genomes were not detected. These findings demonstrate that faecal microorganisms can be transported through SGDs in the Mexican Caribbean, potentially contributing to human health risks for recreation.  相似文献

Rainwater catchment systems (RWCS) in Micronesia were sampled to assess their bacteriological water quality and to determine which RWCS characteristics had a significant impact on water quality. Total and fecal coliform bacteria tests were used to evaluate 203 catchments on 10 islands. Fifty-seven percent of the RWCS had no fecal coliform bacteria and 61% had fewer than 10 total coliform bacteria per 100 ml. Catchment characteristics were found to have a statistically significant effect on total coliform bacteria levels but they did not affect faecal coliform bacteria concentrations. Rainwater catchment systems were found to provide acceptable water in most cases but disinfection prior to consumption is still highly recommended.  相似文献

The inactivation effects of lime, when applied to faeces on faecal coliform bacteria and Ascaris lumbricoides ova, were investigated in the laboratory. A faecal coliform inactivation of about 5 orders of magnitude could be achieved at 19 g CaO l−1 (or initial sample pH of 12) and contact times over a 3 h period, while negligible reduction of faecal coliform bacteria was obtained at 5.7 g CaO l−1 (or initial sample pH of 9) with contact times from 3 to 48 h. However, only a 26.5% reduction of Ascaris ova was observed with 19 g CaO l−1 and a contact time of 48 h. Microscopic examination of the treated samples suggested the ovicidal effects of lime upon the Ascaris ova was of a low efficiency.  相似文献

This paper explores ways in which the analysis of microbial data from routine compliance monitoring, in combination with basic environmental data, can provide insight into the factors affecting faecal-indicator organism concentrations in coastal waters. In the case study presented, eight designated bathing waters on the Fylde coast are continuing to exhibit unreliable compliance with the Imperative standards for total coliform (TC) and faecal coliform (FC) concentrations specified in the EU Bathing Water Directive (76/160/EEC), despite significant reductions in geometric mean concentrations following recent major investment in the sewerage infrastructure. Faecal streptococci (FS) concentrations have remained high and have not been improved by the new sewerage schemes. The results suggest that, prior to the schemes, higher bacterial concentrations were strongly associated with rainfall; and sewage sources were important for TC and FC, but less important for FS, which may have been more strongly affected by diffuse catchment sources. In the post-schemes period, catchment sources appear to be of greater significance; rainfall remains as a significant, though less important, predictor; and tide height at time of sampling, together with variables such as sunshine and the proportion of onshore winds (which affect the survival and movement of bacteria that have already entered the coastal waters), assume greater significance. The approach used here provides a cost-effective management tool for the exploratory investigation of any monitoring point that is failing to meet recreational water quality standards.  相似文献

Escherichia coli and Vibrio cholerae strains were inoculated separately, and together, to aquatic microcosms containing water of 7–25‰ salinities. V. cholerae could be recovered for up to 56 days if sediment was stirred before sampling, but for only 14 days from the unstirred water column. E. coli could be recovered for no more than 7 days, and lost its metallic sheen characteristic on EMB or Endo agar after 24 h, and the characteristic blue colony color on M-FC agar was lost by fecal strains after exposure to salinity for 48–72 h. Results reported indicate that the coliform criteria for harvest of shellfish in estuarine waters needs critical re-evaluation.  相似文献

The objectives of this study were (i) to apportion sources of faecal coliforms (FC) and faecal streptococci (FS) loads from a dairy farm to a stream in the Irvine catchment; (ii) to assess efficacy of pollution mitigation measures installed on this farm; (iii) to assess frequency with which observed FC loads contribute to high risk of failure of coastal bathing water to meet microbial standards. FC and FS loads in a primary stream running through the farm were estimated at up to four stations (above the farm, above the steading, below the farm and below a pre-existing 0.6 ha area of open water/wetland), in summer 2004 and 2005. During this period, steading and field mitigation measures were being installed. We estimated that farm FC loads likely to cause bathing water failure were 8.9 x 10(8) colony-forming units (cfu) ha(-1)d(-1) (guideline standards) and 1.7 x 10(10)cfu ha(-1)d(-1) (mandatory standards). In 2005, the guideline exceedance risk (42%) was associated about equally with field and steading sources, whereas the mandatory exceedance risk (11%) was mainly due to steading sources. The pond/wetland below the farm reduced these exceedance risks to 20% and <1%, respectively, but was a source of FS at high discharge. The exceedence curves below the farm were similar in 2004 and 2005, despite lower flows and installation of pollution mitigation measures. The results suggest that further mitigation efforts on this farm should be focused on improving management of the steading FC sources and that wetlands to receive steading runoff can be cost:effective mitigation tools for faecal indicator pollution.  相似文献

Contamination of recreational waters with Escherichia coli and Enterococcus sp. is a widespread problem resulting in beach closures and loss of recreational activity. While E. coli is frequently used as an indicator of fecal contamination, and has been extensively measured in waterways, few studies have examined the presence of potentially pathogenic E. coli strains in beach waters. In this study, a combination of high-throughput, robot-assisted colony hybridization and PCR-based analyses were used to determine the genomic composition and frequency of virulence genes present in E. coli isolated from beach water in Avalon Bay, Santa Catalina Island, CA. A total of 24,493 E. coli isolates were collected from two sites at a popular swimming beach between August through September 2007 and from July through August 2008. All isolates were examined for the presence of shiga-like toxins (stx1/stx2), intimin (eaeA), and enterotoxins (ST/LT). Of the 24,493 isolates examined, 3.6% contained the eaeA gene, indicating that these isolates were potential EPEC strains. On five dates, however, greater than 10% of the strains were potential EPEC, suggesting that incidence of virulence genes at this beach has a strong temporal component. No STEC or ETEC isolates were detected, and only eight (<1.0%) of the potential EPEC isolates were found to carry the EAF plasmid. The potential EPEC isolates mainly belonged to E. coli phylogenetic groups B1 or B2, and carried the β intimin subtype. DNA fingerprint analyses of the potential EPEC strains indicated that the isolates belonged to several genetically diverse groups, although clonal isolates were frequently detected. While the presence of virulence genes alone cannot be used to determine the pathogenicity of strains, results from this study show that potential EPEC strains can be found in marine beach water and their presence needs to be considered as one of the factors used in decisions concerning beach closures.  相似文献

Domestic wastewater and 0.22 μm pore size wastewater filtrates in a small-scale laboratory wastewater treatment system were inoculated with qual numbers of Eschericia coli and of Bdellovibrio cells. Over a 9-day period, cell counts were made for Bdellovibrio and coliforms. A 3 log reduction within a 72 h incubation period was recorded for both Bdellovibrio and the coliform population inoculated into wastewater. A significant difference (P < 0.05) was observed between the Bdellovibrio populations in wastewater and in wastewater filtrates within an incubation period of 24h and between the coliform populations within an incubation period of 48 h. The data from the study indicate that steady-state conditions appear to exist concerning the interaction between Bdellovibrio and E. coli in waste-water and that Bdellovibrio has only a minor role in the elimination of E. coli from wastewater.  相似文献

A monitoring program was undertaken to assess the microbial quality of greywater collected from 93 typical households in Melbourne, Australia. A total of 185 samples, comprising 75 washing machine wash, 74 washing machine rinse and 36 bathroom samples were analysed for the faecal indicator Escherichia coli. Of these, 104 were also analysed for genetic markers of pathogenic E coli and 111 for norovirus (genogroups GI and GII), enterovirus and rotavirus using RT-PCR. Enteric viruses were detected in 20 out of the 111 (18%) samples comprising 16 washing machine wash water and 4 bathroom samples. Eight (7%) samples were positive for enterovirus, twelve (11%) for norovirus genogroup GI, one (1%) for norovirus genogroup GII and another (1%) for rotavirus. Two washing machine samples contained more than one virus. Typical pathogenic E. coli were detected in 3 out of 104 (3%) samples and atypical enteropathogenic E. coli in 11 (11%) of samples. Levels of indicator E. coli were highly variable and the presence of E. coli was not associated with the presence of human enteric viruses in greywater. There was also little correlation between reported gastrointestinal illness in households and detection of pathogens in greywater.  相似文献

Escherichia coli strains were isolated from surface water collected from three sampling points and cultured on CHROMAgar E. coli. The zinc and copper concentrations of the water samples were determined using Atomic Absorption Spectrophotometer. Colonies of Escherichia coli were subjected to increasing concentration (0.1–1.0 mM) of copper and zinc in the isolation medium. Isolates showing resistance to both metals at 1 mM were identified using conventional methods and by targeting the uidA gene, while PCR amplification of the copA and zntA genes was carried out using specific primers. The selected bacteria were subjected to 10 classes of antibiotics using the Kirby–Bauer disc diffusion technique. Five E. coli showing resistance to copper and zinc and possessing uidA gene were obtained. Of the five isolates, three possessed both zntA and copA genes, while zntA was absent in the remaining two. All the isolates peculiarly showed resistance to ampicillin and doxycycline, while three were multi-drug resistant (MDR).  相似文献

In this study, bacterial removal efficiencies of planted and unplanted subsurface vertical flow constructed wetlands are compared. Indicator organisms such as faecal coliforms (Escherichia coli, total coliforms) and enterococci, and a number of heterotrophic bacteria (heterotrophic plate counts) have been analysed from the influent and effluent of the constructed wetlands as well as at different depths (water and substrate samples). Furthermore dry matter content and total organic carbon (TOC) have been analysed and correlated. The investigated systems show a high removal rate for indicator organisms (a log removal rate of 2.85 for HPC, 4.35 for E. coli, 4.31 for total coliforms and 4.80 for enterococci was observed). In general no significant difference in the removal efficiency of planted and unplanted vertical flow beds could be measured. Only enterococci measured in the substrate samples of the main layer of the filter could a statistically significant difference be observed.  相似文献

Water pollution by microorganisms of fecal origin is a current world-wide public health concern. Total coliforms, fecal coliforms (Escherichia coli) and enterococci are indicators commonly used to assess the microbiological safety of water resources. In this study, influent water samples and treated water were collected seasonally from a water treatment plant and two major water wells in a Black Belt county of Alabama and evaluated for water quality indicator bacteria. Influent river water samples serving the treatment plant were positive for total coliforms, fecal coliforms (E. coli), and enterococci. The highest number of total coliform most probable number (MPN) was observed in the winter (847.5 MPN/100 mL) and the lowest number in the summer (385.6 MPN/100 mL). Similarly E. coli MPN was substantially higher in the winter (62.25 MPN/100 mL). Seasonal variation of E. coli MPN in influent river water samples was strongly correlated with color (R2 = 0.998) and turbidity (R2 = 0.992). Neither E. coli nor other coliform type bacteria were detected in effluent potable water from the treatment plant. The MPN of enterococci was the highest in the fall and the lowest in the winter. Approximately 99.7 and 51.5 enterococci MPN/100 mL were recorded in fall and winter seasons respectively. One-way ANOVA tests revealed significant differences in seasonal variation of total coliforms (P < 0.05), fecal coliforms (P < 0.01) and enterococci (P < 0.01). Treated effluent river water samples and well water samples revealed no enterococci contamination. Representative coliform bacteria selected by differential screening on Coliscan Easygel were identified by 16S ribosomal RNA gene sequence analysis. E. coli isolates were sensitive to gentamicin, trimethoprim/sulfamethazole, ciprofloxacin, vancomycin, tetracycline, ampicillin, cefixime, and nitrofurantoin. Nonetheless, isolate BO-54 displayed decreased sensitivity compared to other E. coli isolates. Antibiotic sensitivity pattern can be employed in microbial source tracking.  相似文献
