This study was conducted to obtain annatto extracts with both high antioxidant capacity and colour potential using solvents of different polarities (water, ethanol/water, ethanol, ethanol/ethyl acetate and ethyl acetate). The highest levels of total phenolic compounds were found in the water, ethanol/water and ethanol extracts (0.5 mg GAE mL?1), and the highest level of bixin was found in the ethanol/ethyl acetate extract (5.2 mg mL?1), which was characterised as the reddest and the most vivid one (a* = 40.5, = 46.1, C* = 58.4). The ethanol/ethyl acetate extract also showed the highest antioxidant activity (4.7 μm TEAC mL?1) and the highest percentage of tryptophan protection against singlet oxygen (63.6%). On the other hand, ethyl acetate and ethanol/water were the least effective solvents for the extraction of phenolic compounds and bixin, respectively. According to the multivariate statistical analysis, ethanol/ethyl acetate and ethyl acetate were the most promising solvents to obtain annatto extracts with both antioxidant and colour properties.  相似文献

Ethanol and hexane mixture agent microwave‐assisted extraction (MAE) method was conducted to extract peony (Paeonia suffruticosa Andr.) seed oil (PSO). The aim of the study was to optimise the extraction for both yield and energy consumption in mixture agent MAE. The highest oil yield (34.49%) and lowest unit energy consumption (14 125.4 J g?1) were obtained under optimum extraction condition: solid‐liquid ratio 0.37 g mL?1, extraction time 3.72 min, extraction temperature 80.92 °C, ethanol ratio 20.00%. GC–MS results showed that unsaturated fatty acids (UFAs) accounted for 88.60% of total fatty acids in PSO. Moreover, linolenic acid content of 37.35% was the highest UFA and caused PSO to possess good nutrition. PSO in DPPH radical scavenging experiment showed that IC50 value of 28.80 ± 2.13 mg mL?1 exhibited strong antioxidant property. All experiments proved that mixed solvent MAE is an efficient and promising method to extract PSO. This method can effectively reduce the energy consumption and extraction time.  相似文献

Extraction, physicochemical properties and fatty acid composition analysis of coconut testa oil (CTO), antioxidant activity and the protective effect on oxidative damage to human serum albumin (HSA) of coconut testa oil extract (CTOE) were investigated. Results showed that the optimal extract condition of CTO was B3A2C2 (temperature of 60 °C, material‐to‐solvent of 1:4 g mL?1 and extraction time of 3 h) with the maximum oil yield (76.83 ± 0.53%). The obtained CTO was nondrying oil with iodine value of 14.69 g per 100 g, and lauric acid was the main component of 42.28%. Hydrogen peroxide scavenging activity of CTOE can reach to 49.81% at 2.5 mg mL?1, while antioxidant activity (AA) on the oxidation of linoleic acid dropped from 56.82% to 31.70% during the first 80 min. CTOE could prevent HSA from oxidative damage induced by hydrogen peroxide via inhibiting the formation of protein carbonyl and increase of hydroperoxides content effectively. Total phenolic content was 68 mg g?1, and the epicatechin and catechin were 2.74 and 2.26 mg g?1 in its phenolic compositions, respectively. These all suggested CTO and CTOE might be new worthy exploiting functional sources.  相似文献

The antioxidant activities and xanthine oxidase inhibition effects of walnut fruit, stem and leaf were studied in this work. The total phenolic contents were in a descending sequence: shell extract > leaf extract > stem extract > defatted walnut kernel (DWK) extract > green husk extract (GHE). The sequence of total phenolic contents was in agreement with the sequence of antioxidant capacities evaluated by ferric reducing antioxidant power and oxygen radical absorbance capacity (ORAC) assays except for DWK extract. The walnut shell extract showed the highest phenolic content (14.81 gallic acid equivalent g/100 g dry extract) and the best antioxidant activity (the ORAC value was 3423.44 ± 142.52 μmol Trolox equivalents g?1). All tested samples possessed xanthine oxidase inhibitory (XOI) effects, the inhibition percentages of which were >50% at 2 mg mL?1 except for stem and DWK extracts. The contents of the major compound (hydrojuglone) in all extracts were in a descending order: GHE > walnut stem extract > walnut shell extract > walnut leaf extract > DWK extract.  相似文献

Pressurised liquid extraction (PLE) of antioxidant compounds from bitter gourd fruits (Momordica charantia) in aqueous ethanolic solvent was investigated using response surface methodology at laboratory scale to understand key impact of extraction variables. Extraction efficiency was optimised by measuring the yield of extraction, total phenolic content (TPC), total flavonoid content (TFC), ferric reducing/antioxidant power assay (FRAP) and radical scavenging activity (RSA). The optimal extraction conditions were reached at 80% ethanol concentration, 10‐min extraction time and at 160 °C. Under these extraction conditions, values of TPC (5.40 ± 0.30 g GAE per 100 g), TFC (1.50 ± 0.10 g QE per 100 g), FRAP (778.55 ± 10 μmol eq Fe (II) g?1), yield (178.50 ± 5.50 mg g?1 dc) and RSA (75.50 ± 4.50%) were achieved. Furthermore, statistical analysis revealed that antioxidative attributes of bitter gourd extract were strongly and positively correlated with extraction temperature and ethanol concentration rather than processing time. This study illustrated that PLE has the potential to extract antioxidant compounds from tropical fruit vegetables in an accelerated manner. Furthermore, influential parameters affecting the process could be optimised for further industrial intake.  相似文献

A systematic study about the solvent effect on the mulberry polyphenol (MP) profile, antioxidant and antiproliferation capacities against human hepatoma HepG2 cells was conducted. Results indicated that MP profiles showed significant differences as a function of solvents including AA/W (acetic acid/water), MeOH/AA/W (methanol/ acetic acid/water), EtOH/AA/W (ethanol/ acetic acid/water) and ME2CO/AA/W (acetone/ acetic acid/water). Among the solvents, EtOH/AA/W took advantage in obtaining MP with the highest yield (49.81 mg GAE g?1 dw), and cellular antioxidant capacity in the PBS no‐wash protocol (63.2 μmolQE/100 g), as well as low cell cytotoxicity (≥50.0 mg mL?1). ME2CO/AA/W was the best choice for extracting MP with the most various compositions (two phenolic acids, four anthocyanins and four flavonols), greatest potential in inhibiting proliferation of HepG2 cells (EC50 = 28.2 mg mL?1) and highest intracellular antioxidant activity (38.0 μmolQE/100 g). Results provide baseline information for efficient solvent extraction to obtain MP that is promising as natural antioxidants and antitumor agents.  相似文献

The processing of peaches to produce fruit pulp generates solid and liquid wastes rich in phytochemicals, such as carotenoids; thus, the objective of this work was to study the use of this waste for carotenoid extraction based on a complete experimental design and using response surface methodology. The parameters studied were the amount of solvent (20–50 mL), the number of extractions (1–5) and the extraction time (10–30 min). The extracts were analysed by spectrophotometry and the optimised conditions by HPLC. The optimised results were four extractions of 10 min using 38.5 mL of ethanol, which presented a yield of 168.59 μg g?1 DW of total carotenoids of which 67.55 μg g?1 corresponds to β‐carotene, 86.75 μg g?1 to cryptoxanthin, 12.08 μg g?1 to zeaxanthin and 2.2 μg g?1 to lutein, which representing 66% of extraction pigments relative to the total content of carotenoids present in the peach waste.  相似文献

The aim of this study was to investigate the antioxidant activities of persimmon seed extracts (PSE) using different solvents such as ethanol, methanol, acetone, and their aqueous 80% solvents. The EC50 values of the extracts from absolute ethanol (EE) and methanol (ME) in 2, 2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical–scavenging assay were 49.71 and 51.15 μg mL?1, respectively, while the EC50 of butylated hydroxyanisole (BHA) was 70.82 μg mL?1. However, the EC50 value of reducing power for the absolute acetone extract (AE) was higher (210.06 μg mL?1) than that of BHA (212.67 μg mL?1). Although the absolute ME had the highest antioxidant activity, it exhibited the lowest total phenolics and flavonoids. In contrast, the antioxidant activities of the aqueous solvent extracts showed a good correlation with total phenolics and flavonoids when compared to the absolute solvent extracts. The results showed that PSE could potentially be used as an inexpensive source of natural antioxidant in food and pharmaceutical industries.  相似文献

Supercritical fluid extraction (SFE) was applied for the extraction of valuable compounds from grape (Vitis labrusca B.) peel. Extraction was carried out according to an orthogonal array design (OAD) and independent variables selected were temperature, pressure and modifier concentration. SFE process was optimized by using response surface methodology (RSM) for the extract yield, total phenols, antioxidants and total anthocyanins from grape peel. Effects of extraction temperature and pressure were found to be significant on all responses. Optimal SFE conditions were identified as 45–46 °C temperature, 160–165 kg cm? 2 pressure and 6–7% ethanol as modifier for maximum extract yield (12.31%), total phenols (2.156 mg GAE/100 mL), antioxidants (1.628 mg/mL) and total anthocyanins (1.176 mg/mL). Experimental values for response variables at these optimal conditions match well with the predicted values. Grape peel extracts obtained by SFE showed more than 93% DPPH radical scavenging activities.Industrial relevanceThis study describes the response surface optimization of supercritical fluid extraction (SFE) process for the enhanced recovery of total phenols, antioxidant and anthocyanins from grape peel. SFE uses CO2 as supercritical fluid which is environment friendly solvent; allows extraction at lower temperature and the extracts obtained possess higher quality and safety. The extract yields obtained by three methods in different solvents from chilli were compared. The antioxidant activities of chilli extracts vs. butylated hydroxytoluene (BHT) was determined through the peroxide value (POV), the beta‐carotene bleaching assay and the 2,2‐diphenyl‐1‐picrylhydrazyl radical scavenging method. The results showed that the extract yields by three extract methods, namely, Soxhlet, ultrasonic wave and saturation, were 16.32%, 15.78% and 11.62%, respectively, when 85% ethanol acted as solvent. The extracting efficiency of solvent from high to low was 95% ethanol > ethyl acetate > anhydrous ethanol > distilled water. The total phenol content of chilli extracts from ultrasonic wave with 85% ethanol as solvent was 73.91 mg gallic acid equivalents (GAE) per gram extracts and the value of inhibitory capacity IC50 on free radical (DPPH) was 38.99 mg mL?1. The POV of chilli extracts was close to that of BHT. The ultrasonic wave was more helpful for the extraction of antioxidant substance from chilli than other methods. In addition, the composition of chilli extracts was analysed by gas chromatography‐mass spectrum (GC‐MS).  相似文献

The peel of Gac fruit is regarded as waste product in the processing of Gac although it contains high level of carotenoids and possesses a significant antioxidant capacity. This study optimised the extraction yields of carotenoids and antioxidant capacity from Gac peel. Different organic solvents were examined to determine the most suitable solvent for the extraction. The extraction conditions including time, temperature and solvent–solid ratio were then optimised for maximising extraction yields of carotenoids and antioxidant capacity from Gac peel using response surface methodology. Ethyl acetate was identified as the most suitable solvent. The optimal extraction time, temperature and solvent–solid ratio were 150 min, 40.7 °C and 80 mL g?1, respectively. The carotenoid extraction yield and the antioxidant capacity extraction yield were 271 mg/100 g DW and 737 μm TE/100 g DW, respectively. Thus, the extraction using ethyl acetate with the ratio of 80:1 (mL solvent per g Gac peel) for 150 min at 40.7 °C is suggested for recovering carotenoids and antioxidant capacity from Gac peel.  相似文献

The aim of the present study was to evaluate and compare the phenolic content and antioxidant and antimicrobial properties of olive leaf extracts obtained by ultrasound-assisted extraction (UAE) and conventional extraction (CSE). UAE of olive leaf extracts yielded a higher total phenolic content (TPC) and total flavonoid content (TFC) of 14.31% and 19.50%, respectively. Higher antioxidant activities were found from the extracts prepared with UAE (for 18.5%, 12.5%, 10.9% and 17.6% higher determined by DPPH, ABTS, FRAP and CUPRAC methods, respectively). Good antibacterial inhibitory activity (as MIC and MBC) was observed against both Y. enterocolitica and S. aureus (1.40 ± 0.40 mg mL−1 and 4.00 ± 1.60 mg mL−1, respectively) with the extract prepared with UAE. In conclusion, olive leaf extracts prepared with UAE exhibited higher antioxidant and antimicrobial activities against common food-borne pathogens than CSE extracts and thus could be beneficial in ensuring food quality and food safety.  相似文献

This study aimed to optimise aqueous extraction conditions for total phenolic compounds (TPC) from Davidson's plum (Davidsonia pruriens F. Muell) and to assess the physicochemical and antioxidant properties of the phenolic‐enriched extract. The results showed that temperature, time and ratio significantly affected the extraction of TPC. Optimization of extraction conditions was performed using response surface methodology (RSM) utilising a Box–Behnken design. Optimal extraction conditions were determined to be temperature: 90 °C, extraction time: 30 min and solvent to mass ratio: 20:1 mL g?1. The extracted solid obtained under these conditions had low‐moisture content, high water solubility and contained 45 mg GAE g?1 of TPC, 22 mg RUE g?1 of flavonoids, 3.2 mg CAE g?1 of proanthocyanidins, 2 mg CGE g?1 of anthocyanidins and 56 mg ACE g?1 vitamin C. The extract possessed potent antioxidant capacity, but was comparatively lower than those of vitamin E and BHT. Thus, Davidson's plum should be further investigated for its potential health promoting benefits and utilisation in the nutraceutical and food industries.  相似文献

Response surface methodology was used to predict optimum conditions for hot air roasting of barley grains (temperature, time, and amount). Antioxidant capacity in the grains was highest under optimum conditions of 250 °C, 63.5 min and 42 g (one and a half layers). A correlation of R 2 = 0.74 (p < 0.05) was found between 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and total phenolic contents. Ethanol and aqueous extracts were prepared from grains roasted under optimum conditions and assessed for antioxidant capacity. Antioxidative compounds in the extracts were then identified using GC–MS. The IC50 value of ethanol extract was significantly lower (11.45 μg mL–1) than that of aqueous extract (33.54 μg mL–1) and α-tocopherol (12.6 μg mL–1) but higher than BHT (9.59 μg mL–1). The same trend was observed in linoleic acid assay. In reducing power, the ethanol extract and α-tocopherol were not significantly different. Phenolic acids p-hydroxybenzaldehyde, vallinic and gallic acids were identified as the major compounds in the extracts. The results obtained from this study show that it is possible to optimize antioxidant capacity in barley grains during roasting.  相似文献

This study aimed to identify the least aggressive and highest yield extraction method to obtain bioactive compounds from Inga marginata Willd fruits, determine the chemical components, and evaluate the extracts’' antimicrobial and cytotoxic activity. The extraction efficiency was expressed by the total phenolic and total flavonoid content and antioxidant activity (DPPH, IC50, and ORAC) using conventional, ultrasound, and microwave-assisted extraction. The highest bioactive compound content was achieved using 5 min at 60 °C for total phenolic content (214.98 mg GAE g−1), total flavonoid content (22.90 mg EQ g−1), DPPH (45.98 μmol TEAC g−1), inhibitory capacity (0.80 mg mL−1), and ORAC (167.25 μmol Trolox g−1) using ultrasonic extraction, and the extract inhibited the growth of all microorganisms tested. Thirteen chemical compounds were determined by ESI-ToF-MS, confirming the high phytochemical capacity of the extract. Lastly, the Inga extract showed no cytotoxicity at the concentrations used.  相似文献

Preheated solvent extraction (PSE) was evaluated via the analysis of the extraction kinetics, microstructure of extracted samples and energy consumption as alternative to microwave‐assisted extraction (MAE). Cabbage outer leaves and ethanol were used as test material and extraction solvent, respectively. MAE was first optimised in terms of glucosinolates and phenolics yields; total antioxidant activity of the extracts was also assessed. MAE at a specific absorbed power of 0.37 W g?1 for 9 min was selected as optimum condition to extract glucosinolates and phenolics, while PSE was optimised at 6 min of the extraction. The highest normalised total glucosinolates and phenolic contents as well as antioxidant activities of the extracts obtained via MAE were not significantly different from those obtained via PSE. Confocal laser scanning microscopy revealed no significant differences in cabbage cell damages rendered by MAE and PSE. PSE nevertheless exhibited slightly higher specific energy consumption than MAE.  相似文献

Bioactive compounds from Brazilian hop (Humulus lupulus L.) cultivars were extracted by ultrasound and their phenolic profile compared with commercial hop from the USA. The most effective extraction conditions (solution of ethanol 49%, at 52 °C and a solid/liquid ratio of 1 g per 34 mL) for the total phenolic compounds (TPC) were determined using a Central Composite Rotatable Design. The Brazilian hop showed higher content of TPC (33.93 ± 0.67 mg GAE g−1), total flavonoids (54.47 ± 0.10 mg QE g−1) and higher antioxidant activity (ABTS: EC50 21.29 ± 1.36 μL mL−1; DPPH: EC50 3.91 ± 0.17 μL mL−1) when compared with the USA hop. The main phenolic compounds present in the extracts were the flavonoids isoquercitrin and quercetin. The antioxidant properties of the Brazilian hop extract had not been reported yet in the literature for this raw material, thus showing potential to be incorporated in polymeric films used as active packaging.  相似文献

Grape juices made using small‐scale production techniques are widely consumed. The extraction procedures employed to produce them, however, can affect bioactive compounds and antioxidant activity in the final product. In this study, juices prepared using four extraction methods (steam, extractor, juicer and blender) were evaluated for soluble and hydrolysable polyphenol content, total anthocyanin content, antioxidant capacity, physicochemical characteristics and colour. Acceptance of steam‐extracted juices and their stability during storage were also evaluated. Steam extraction resulted in a higher soluble phenolic (1073 ± 58 mg gallic acid L?1) and anthocyanin content (138 ± 22 mg cyanidin L?1), as well as a higher antioxidant capacity when compared to juices prepared using other methods. Although steam‐extracted juice remained microbiologically stable during 24 months of storage, changes in phytochemical compounds and antioxidant capacity did occur. Our results indicate that steam‐extracted grape juices have high commercial potential.  相似文献

Epidemiological evidence supports that polyphenol‐rich foods with high antioxidant potency promote health and may influence various metabolic diseases' development. Often beverage products claim to have antioxidant potency based on the perceived polyphenol content; however, few studies have examined the antioxidant potency of beverages with none reporting on commonly consumed youth beverages. This study's purpose was to measure total antioxidant capacity (TAC), expressed in Trolox equivalents (μmol mL?1), in a variety of youth beverages, including juices, vitamin enhanced waters, sport drinks, energy drinks and various milk products. Significantly higher TAC (3.8–6.7 μmol mL?1) was measured in 100% juices, while vitamin enhanced waters varied by brand (0.2–3.1 μmol mL?1). Soda, energy and sports drinks measured the least activity (0.0–0.3 μmol mL?1), whereas other milk types (strawberry, organic, regular and soya) measured no activity, except chocolate milk (3.0 μmol mL?1). These results may improve beverage selection practices for youth, parents and dietitians to increase TAC of the paediatric diet.  相似文献

The in vitro inhibitory activities of different seed extracts prepared from cranberry bean mutant SA‐05 and its wild‐type variety Hwachia against aldose reductase, α‐glucosidase and α‐amylase were examined. The results indicated that the polyphenolics‐rich extracts obtained using 800 g kg?1 methanol and 500 g kg?1 ethanol demonstrated inhibitory activities against aldose reductase (IC50 of 0.36–0.46 mg mL?1) and α‐glucosidase (IC50 of 1.32–1.94 mg mL?1). The 500 g kg?1 ethanol extracts also showed α‐amylase inhibitory activities (IC50 of 70.11–80.22 μg mL?1). Subsequent extracts, prepared further with NaCl and H2O from precipitates of 800 g kg?1 methanol or 500 g kg?1 ethanol extracts, exhibited potent α‐amylase inhibitory activities (IC50 of 17.68–38.68 μg mL?1). A combination of 500 g kg?1 ethanol extraction plus a subsequent H2O extraction produced highest polyphenolics and α‐amylase inhibitors. The SA‐05 α‐amylase inhibitor extracts showed greater inhibitory activities than that of Hwachia. Thus, cranberry bean mutant SA‐05 is an advantageous choice for producing anti‐hyperglycaemic compounds.  相似文献
