Dry-cured Iberian ham slices were stored under vacuum and under four different modified atmospheres (60/40 = 60%N2 + 40%CO2; 70/30 = 70%N2 + 30%CO2; 80/20 = 80%N2 + 20%CO2; argon = 70%argon + 30%CO2) at 4 ± 1 °C during 120 days. Gas composition, moisture content, pH, colour, pigment content, and lipid stability were measured, as well as sensory and microbial analysis were carried out throughout storage. A loss of intensity of red colour (a*-values) was observed during storage in ham slices (P < 0.05). Consistently, MbFe(II)NO content also decreased throughout storage (P > 0.05). Slices of ham packed in 40%CO2 (60/40) and 30%CO2 (70/30) showed lower a*-values than the rest of the batches after 60 days (P < 0.05), though differences were not evident after 120 days (P > 0.05). TBARs values showed an upward trend during the storage of packaged slices (P < 0.05). Vacuum-packed slices showed the lowest TBARs values and those packed with 40%CO2, the highest. Sensory attributes did not vary significantly (P > 0.05) throughout storage under refrigeration and packed either in vacuum or in modified atmospheres. No safety problems were detected in relation to the microbial quality in any case.  相似文献

Different derivatization procedures were assayed to simultaneously analyse iminosugars such as deoxynojirimycin (DNJ) or fagomine and other carbohydrates of low molecular weight by gas chromatography coupled to mass spectrometry (GC–MS) in Morus sp. Both oximation + trimethylsilylation and oximation + acetylation allowed the separation of target compounds, whereas trimethylsilyl (TMS) and acetylated derivatives showed several coelutions. Nevertheless, oximation + acetylation were discarded for giving inaccurate results for ketoses due to their incomplete derivatization. Different conditions for the conversion into trimethylsilyl oximes (TMSO) were assayed, the best results being achieved using hexamethyldisilazane with trifluoroacetic acid as silylation agent. Contents of iminosugars (DNJ, fagomine and pipecolic acid derivatives) and other carbohydrates such as mono and disaccharides, myo-inositol and galactinol isomers in mulberry extracts (fruits, leaves and branches) were determined by GC–MS using the TMSO procedure.  相似文献

A method for the multi-elemental determination of metals (Al, Ba, Ca, Cu, Fe, K, Mg, Mn, Sr and Zn), metalloids (B and Si), and non-metals (Cl, P and S) in the babassu nut and mesocarp, sapucaia nut, coconut pulp, cupuassu pulp and seed, and cashew nut by axially viewed inductively coupled plasma optical emission spectrometry is presented. A diluted oxidant mixture (2 ml HNO3 + 1 ml H2O2 + 3 ml H2O) was used to achieve the complete decomposition of the organic matrix in a closed-vessel microwave oven. The accuracy of the entire proposed method was confirmed by standard reference material analysis (peach leaves-NIST SRM1547). The certified values showed a good agreement at a 95% confidence limit (Student’s t-test). The average RSD for repeatability of calibration solutions measurements were in the range of 1.1–6.7%. Limits of quantification (LOQ = 10 × LOD) were in the level of 0.00072–0.0532 mg/l. The macro and micronutrient ranges in the different nuts and seeds did not exceed the dietary reference intake (DRI), except for Mn in the babassu nut.  相似文献

Lipid and protein structural characteristics of frankfurter formulated with olive oil-in-water emulsion stabilized with soy protein isolate (SPI) as pork backfat replacer were investigated using Fourier transform infrared spectroscopy (FT-IR). Proximate composition and textural properties were also evaluated. Different frankfurters were reformulated: F/PF with pork backfat, F/SPI with oil-in-water emulsion stabilized with SPI and F/SPI + SC + MTG with emulsion stabilized with a combination of SPI, sodium caseinate (SC) and microbial transglutaminase (MTG). Replacement of pork backfat with these emulsions produced an increase (P < 0.05) of hardness, springiness, cohesiveness and chewiness but a reduction (P < 0.05) of adhesiveness. F/SPI and F/SPI + SC + MTG frankfurters showed the lowest (P < 0.05) half-bandwidth in the 2922 cm−1 band, which could be related to lipid chains were more ordered than in F/PF. Modifications in the amide I band profile revealed a higher concentration of aggregated intermolecular β-sheets in F/SPI + SC + MTG samples. Lipid and protein structural characteristics could be associated with specific textural properties of healthier frankfurters.  相似文献

A Taylor dispersion method is used to measure ternary mutual diffusion coefficients (D11, D22, D12 and D21) for aqueous solutions of CuCl2 (1) + caffeine (2) + H2O-(3) at 25 °C and 37 °C at carrier concentrations from (0.000 to 0.020 mol dm−3), for each solute, respectively. The results are compared with these obtained for the binary systems, that is, CuCl2 (1) + H2O (2) and caffeine (1) + H2O (2), also reported here. From these data it is possible to make conclusions about the influence of the caffeine solutes in diffusion of copper chloride.  相似文献

l-Glutamate (Glu) is a major excitatory neurotransmitter responsible for neurotransmission in the vertebrate central nervous system. Vesicular Glu transporters VGLUT1 and VGLUT2 concentrate (50 mM) Glu [Michaelis constant (measuring affinity), or Km, = 1 to 4 mM] into synaptic vesicles (SV) for subsequent release into the synaptic cleft of glutamatergic neurons. Vesicular Glu transporter activity is dependent on vacuolar H+-ATPase function. Previous research has shown that ergopeptines contained in endophyte-infected tall fescue interact with dopaminergic and serotoninergic receptors, thereby affecting physiology regulated by these neuron types. To test the hypothesis that ergopeptine alkaloids inhibit VGLUT activity of bovine cerebral SV, SV were isolated from cerebral tissue of Angus-cross steers that were naive to ergot alkaloids. Immunoblot analysis validated the enrichment of VGLUT1, VGLUT2, synaptophysin 1, and vacuolar H+-ATPase in purified SV. Glutamate uptake assays demonstrated the dependence of SV VGLUT-like activity on the presence of ATP, H+-gradients, and H+-ATPase function. The effect of ergopeptines on VGLUT activity was evaluated by ANOVA. Inhibitory competition (IC50) experiments revealed that VGLUT-mediated Glu uptake (n = 9) was inhibited by ergopeptine alkaloids: bromocriptine (2.83 ± 0.59 μM) < ergotamine (20.5 ± 2.77 μM) < ergocornine (114 ± 23.1 μM) < ergovaline (137 ± 6.55 μM). Subsequent ergovaline kinetic inhibition analysis (n = 9; Glu = 0.05, 0.10, 0.50, 1, 2, 4, 5 mM) demonstrated no change in apparent Km. However, the maximum reaction rate (Vmax) of Glu uptake was decreased when evaluated in the presence of 50, 100, and 200 μM ergovaline, suggesting that ergovaline inhibited SV VGLUT activity through a noncompetitive mechanism. The findings of this study suggest cattle with fescue toxicosis may have a decreased glutamatergic neurotransmission capacity due to consumption of ergopeptine alkaloids.  相似文献

Forty-nine Manchega breed male suckling lambs were used in this experiment. The effect of CO2 concentration and exposure time at stunning [80% CO2 for 90 s (G1); 90% CO2 for 90 s (G2); 90% CO2 for 60 s (G3); 80% CO2 for 60 s (G4)] plus an electrically stunned control group (G5) was assessed for pH, colour (L, a, b, C and h), water holding capacity (WHC), drip loss (DL), cooking loss (CL) and shear force (SF) in samples packed under two different types of modified atmospheres (MA: MA A: 70%O2 + 30%CO2; MA B: 69.3%N2 + 30%CO2 + 0.7%CO) at 7, 14 and 21 d post-packaging. The lowest pH was found in G4 and in G5. The highest WHC and the lowest CL were found in G2 and G3 groups (P < 0.05). Modified atmospheres did not affect on pH, WHC, CL and DL, although a significant effect (P < 0.001) on colour was found at all the analysis times. Both the type of stunning and the modified atmosphere affected SF values.  相似文献

‘Pink Lady®’ apples (Malus domestica) fruit were harvested at commercial maturity treated with three different agrochemical products, and stored at 1 °C under either air or controlled atmosphere conditions (2.5 kPa O2 + 3 kPa CO2 and 1 kPa O2 + 2 kPa CO2) for 15 and 28 weeks. Diphenylamine, folpet and imazalil contents in both skin and flesh were simultaneously determined after cold storage plus a simulated marketing period of 1 or 7 days at 20 °C. Results showed that apples stored in 2.5 kPa O2 + 3 kPa CO2 retained higher contents of diphenylamine residues in comparison with those stored in 1 kPa O2 + 2 kPa CO2 or refrigerated air. Significant differences in imazalil skin contents were found throughout the simulated marketing period at 20 °C after storage for 28 weeks in controlled atmospheres.  相似文献

The present research evaluated the effects of tea polyphenol (TP) combined with ozone water (O3) on the quality of black sea bream (Sparus macrocephalus) over a period of 15 days storage at 4 °C. A solution of TP (0.2%, w/v) was used to coat the fish after washing with ozone water (1 mg/L). Fish physicochemical (pH, K value, peroxide value, thiobarbituric acid, total volatile basic nitrogen, trimethylamine, texture, and colour), sensory, and bacteriological characteristics were all analysed. TP + O3 treatment effectively reduced nucleotide breakdown, lipid oxidation, protein decomposition, and microbial growth, and maintained better characteristics of texture, colour, and sensory compared with the control. The efficiency of TP + O3 treatment was also better than that of TP treatment or O3 treatment alone. Therefore, tea polyphenol coating combined with ozone water prewashing may be a promising method of maintaining the storage quality of black sea bream and of extending fish post-mortem shelf-life during 4 °C storage.  相似文献

Effects of lyophilized Urtica dioica L. water extract (LUWE) and modified atmosphere packaging (MAP) on the quality and shelf life of ground beef were investigated. Ground beef was stored as aerobic control, MAP (80%O2 + 20% CO2), MAP + 250 ppm LUWE and MAP and 500 ppm LUWE at 2 ± 0.5 °C for 14 days. MAP and LUWE had significant effects on mesophilic, psychrotrophic and lactic acid bacteria and Pseudomonas counts. Depending on the level of LUWE, Pseudomonas and psychrotrophic counts decreased. Treatment with 500 ppm LUWE + MAP showed the lowest TBARS values compared to other groups during storage. 80% O2-MAP increased TBARS values. Treatment had no significant effect on L* and b* values of the exterior of the ground beef, but had significant effects on the color of interior sections.  相似文献

To investigate the effects of low O2 and high CO2 atmospheres on the berry drop of ‘Kyoho’ grapes (Vitis vinifera X V. labrusca), changes of fruit detachment force (FDF) and berry abscission and enzyme activities in the abscission zone were examined during 60 days of storage in air (control), 4%O2 + 9%CO2 or 4%O2 + 30%CO2 at 0 °C and 95%relative humidity. There was a high negative correlation between FDF and berry drop. Cellulase activity increased over time and was closely correlated with berry abscission. Polygalacturonase (PG) activity rose markedly for the first 30 days and then remained relatively constant. Peroxidase (POD) activity dropped significantly for the first 15 days and subsequently rose sharply (P < 0.05). Cellulase, PG and POD activities were the lowest in fruits in 4%O2 + 30%CO2, followed by 4%O2 + 9%CO2 and air storage. Pectinesterase (PE) maintained a basal level of activity and there were no significant differences among three treatments (P < 0.05). Compared to air control, the combined effects of the lower level of O2 and the higher level of CO2 suppressed the activities of cellulase, PG and POD, maintained greater FDF, and reduced berry abscission during storage.  相似文献

The present study evaluated the sensory characteristics of low temperature long time (LTLT) treated Semitendinosus from pork and beef and Pectoralis profundus from chicken. Semitendinosus and Pectoralis profundus muscles were heat treated at 53°C and 58°C for Tc + 6 h, Tc + 17 h, and Tc + 30 h (only Semitendinosus from pork and beef). Tc was the time for the samples to equalize with the temperature in the water bath. Tenderness increased with increasing heating temperature and time in pork and beef, but not in chicken. Juiciness decreased with increasing heating temperature and time in all species, and cooking loss increased with increasing temperature. A done appearance was developed with increasing heating time at 58 °C in pork and beef, while in chicken the done appearance was only affected by temperature. Flavor attributes were less affected by the LTLT treatment for all species.  相似文献

The effects of a reduced-oxygen atmosphere (ROA) ([O2] < 4.5%) during part or the whole of dry-cured ham processing on microbiological and physico-chemical parameters and mite growth were investigated in two independent experiments. In Experiment 1, six hams were processed in ROA and six in air for 275 days; in Experiment 2, where lower RH was used, six hams were processed in ROA for 289 days, six for 214 days in air + 75 days in ROA, and six in air for 289 days. Microbiological analyses during the process and physicochemical analyses in final products were carried out. The use of ROA during the whole process increased the L∗ colour parameter in the subcutaneous fat and proteolysis index and decreased b∗ in the external part of the subcutaneous fat and cholesterol oxide concentration. The use of ROA combined with low RH retarded microbial growth and prevented mite growth.  相似文献

Selection of informative wavelengths is a crucial step for online spectral imaging applications. In this paper, four cultivars of apples were utilized to select effective wavelengths for bruise detection within 380-1000 nm. Each of the wavelength variables was considered as an independent classifier for bruised/normal classification, and all classifiers were evaluated and compared by receiver operating characteristic (ROC) analysis. According to the area under the ROC curve (AUC), the reflectance difference between two wavelengths (R(λ1) − R(λ2)), was determined as the best wavelength pair for the Fuji, Jonagold, Orin and Sinano Gold cultivars as follows: R(808 nm) − R(760 nm), R(832 nm) − R(772 nm), R(834 nm) − R(762 nm) and R(788 nm) − R(742 nm), respectively. The performance of the wavelengths selected in this paper was measured by comparing the predicted sensitivity, predicted specificity and predicted classification accuracy with those of the model proposed by partial least squares discriminant analysis (PLSDA), which used all of the wavelength variables. The results showed that the predictive ability of both methods was generally on the same level.  相似文献

Polyphenoloxidase (PPO) was partially purified from chufa corms through ammonium sulphate precipitation and dialysis. Biochemical properties of chufa PPO were analysed using exogenous substrate catechol. Optimal pH and temperature for PPO activity were 5 and 45 °C. Ethylenediaminetetraacetic acid disodium salt and l-cysteine could not inhibit the PPO activity. However, sodium thiosulphate pentahydrate exhibited the strongest inhibiting effect, followed by ascorbic acid and anhydrous sodium sulphite. Except for K+, other metal ions such as Zn2+, Cu2+, Fe3+, Ca2+, Fe2+ and Na+ accelerated the enzymatic reaction between catechol and PPO. Kinetic analysis showed that the apparent Km and Vmax values were around 10.77 mM and 82 units/ml min. In addition, (−)-gallocatechin gallate, (−)-epicatechin gallate and (+)-catechin gallate isolated and identified from chufa corms were supposed to be the potential endogenous PPO substrates due to their ortho-diphenolic or pyrogallolic structures. These polyphenols might be catalysed by PPO, resulting in the browning of chufa corms after fresh-cut processing.  相似文献

We examined the influence of chitosan on lipid oxidation and color stability of ground beef stored in different modified atmosphere packaging (MAP) systems. Ground beef patties with chitosan (1%) or without chitosan (control) were packaged either in high-oxygen MAP (HIOX; 80% O2 + 20% CO2), carbon monoxide MAP (CO; 0.4% CO + 19.6% CO2 + 80% N2), vacuum (VP), or aerobic packaging (PVC) and stored at 1 °C. Chitosan increased (P < 0.05) redness of patties stored in PVC and CO, whereas it had no effect (P > 0.05) in HIOX. Chitosan patties demonstrated lower (P < 0.05) lipid oxidation than controls in all packaging. Control patties in PVC and HIOX exhibited greater (P < 0.05) lipid oxidation than those in VP and CO, whereas chitosan patties in different packaging systems were not different (P > 0.05) from each other. Our findings suggested that antioxidant effects of chitosan on ground beef are packaging-specific.  相似文献

Yellow peaches (Prunus persicu L. Batsch.) were stored under controlled atmospheres (CA) of 2% O2 + 5% CO2, 5% O2 + 10% CO2, 2% O2 + 10% CO2, 5% O2 + 5% CO2, and normal atmosphere at 2 °C, to investigate the effects of different concentrations of O2 and CO2 on the structure of a single sodium carbonate-soluble pectin (SSP) molecule. The microstructure changes, including aggregates and branches, were studied by atomic force microscopy (AFM) at initially, on the 15th and 45th day. The microstructure of SSP molecules and polymers showed that aggregates separated gradually with the storage time. The degradation took place in the linear backbone as well as in side chains. The degradation of SSP molecules was inhibited by lower O2 and higher CO2 concentrations. Almost all of the chains were composed of four basic units with widths of 11.719, 15.625, 19.531 and 17.578 nm, which can be visualized and calculated exactly by AFM. This indicated that parallel linkage or intertwist between the basic units was a fundamental characteristic of SSP molecules.  相似文献

In this study, we evaluated the antioxidant activity (diphenylpicrylhydrazyl (DPPH)-radical scavenging activity) of the low-molecular weight fraction of water-soluble Susabinori (Porphyra yezoensis) extract (LMF). DPPH-radical scavenging activity of LMF (65.5 μmol-Trolox eq/ml) was enhanced at temperatures over 100 °C (1.4, 2.0 and 2.4 times higher at 100, 110 and 120 °C, respectively). As a result of HPLC separations, a newly produced peak was observed in heat treated LMF, while a marked decrease of another peak was found after heat treatment of LMF. The decreased peak was identified as porphyra-334, which is a typical mycosporine-like amino acid, by ESI/MSn analysis (protonated molecule [M + H]+ at m/z 347.1 and its fragment ion 303.1). The newly produced compound with [M + H]+ at m/z 329.1 was found to be a dehydrated compound of porhyra-334. The compound exhibited an extremely high-IC50 value of 10.1 μg/ml in DPPH-radical scavenging activity, compared to porphyra-334 (>1000 μg/ml).  相似文献

Aerosolization was investigated as a potential way to apply allyl isothiocyanate (AIT), hydrogen peroxide (H2O2), acetic acid (AA) and lactic acid (LA) on fresh baby spinach to control Escherichia coli O157:H7 during refrigeration storage. In this study, baby spinach leaves were dip-inoculated with E. coli O157:H7 to a level of 6 log CFU/g and stored at 4 °C for 24 h before treatment. Antimicrobials were atomized into fog-like micro-particles by an ultrasonic nebulizer and routed into a jar and a scale-up model system where samples were treated. Samples were stored at 4 °C for up to 10 days before the survival of the cells was determined. A 2-min treatment with 5% AIT resulted in a > 5-log reduction of E. coli O157:H7 on spinach after 2 days refrigeration regardless if the samples were pre-washed or not; however, this treatment impaired the sensory quality of leaves. Addition of LA to AIT improved the antimicrobial efficacy of AIT. In the jar system, washing with 3% H2O2 followed by a 2-min treatment of 2.5% LA + 1% AIT or 2.5% LA + 2% AIT reduced E. coli O157:H7 population by 4.7 and > 5 log CFU/g, respectively, after 10 days refrigeration. In the scale-up system, up to 4-log reduction of bacterial population was achieved for the same treatments without causing noticeable adverse effect on the appearance of leaves. Thus, this study demonstrates the potential of aerosolized AIT + LA as a new post-washing intervention strategy to control E. coli O157:H7 on baby spinach during refrigeration storage.  相似文献
