Natural products represent a rich source of biologically active compounds and are an example of molecular diversity, with recognised potential in drug discovery. Herein, the methanol extract of Matricaria recutita L. (German chamomile) and its decoction and infusion (the most consumed preparations of this herb) were submitted to an analysis of phytochemicals and bioactivity evaluation. The antioxidant activity was determined by free radicals scavenging activity, reducing power and inhibition of lipid peroxidation; the antitumour potential was tested in human tumour cell lines (breast, lung, colon, cervical and hepatocellular carcinomas), and the hepatotoxicity was evaluated using a porcine liver primary cell culture (non-tumour cells). All the samples revealed antioxidant properties. The decoction exhibited no antitumour activity (GI50 > 400 μg/mL) which could indicate that this bioactivity might be related to compounds (including phenolic compounds) that were not extracted or that were affected by the decoction procedure. Both plant methanol extract and infusion showed inhibitory activity to the growth of HCT-15 (GI50 250.24 and 298.23 μg/mL, respectively) and HeLa (GI50 259.36 and 277.67 μg/mL, respectively) cell lines, without hepatotoxicity (GI50 > 400 μg/mL). Infusion and decoction gave higher contents of organic acids (24.42 and 23.35 g/100 g dw). Otherwise, the plant methanol extract contained the highest amounts of both phenolic acids (3.99 g/100 g dw) and flavonoids (2.59 g/100 g dw). The major compound found in all the preparations was luteolin O-acylhexoside. Overall, German chamomile contains important phytochemicals with bioactive properties (mainly antitumour potential selective to colon and cervical carcinoma cell lines) to be explored in the pharmaceutical, food and cosmetics industries.  相似文献

A comparative study of the organic acids and phenolics composition and of the total alkaloids content of entire wild edible mushrooms (Russula cyanoxantha, Amanita rubescens, Suillus granulatus and Boletus edulis) and correspondent caps and stipes was performed. All species presented oxalic, citric, malic and fumaric acids, with A. rubescens exhibiting the highest total organic acids content. Organic acids were preferably fixed in the cap. Among phenolics, only p-hydroxybenzoic acid was found in A. rubescens and S. granulatus, in very low amounts. B. edulis was the species that presented the highest total alkaloid amounts. Except for this species, alkaloids mainly accumulated in the cap. All of the species exhibited a concentration-dependent scavenging ability against DPPH·. B. edulis revealed the highest antioxidant capacity. The cap seemed to be the part with highest antioxidant potential. Some relationships between chemical composition and antioxidant capacity were considered.  相似文献

Leaves and products made from them are the major focus of research on bioactive compounds of mate (Ilex paraguariensis). However, studies on other parts of the plant, are scarce. The aim of this study was to determine the methylxanthines, phenolic compounds, and antioxidant activity of mate bark (residual biomass) and compare with those of mate leaves. The high antioxidant activity of mate bark and its high concentration of total polyphenols were apparent in both the aqueous and the methanolic extracts, the values of which were greater than those detected in the leaves. Of the phenolic acids identified, the levels of chlorogenic acid and 4,5-dicaffeoylquinic acid in the samples were significantly higher in the methanolic bark extract. With regard to methylxanthines, considerable concentrations were detected in the samples. The results obtained contribute to the viability of exploiting this residue, broadening the use of the mate plant.  相似文献

An extract of Achillea millefolium herb (YE) was investigated for antioxidant activity using chemical and biological assays. Qualitative and quantitative analysis of some major phenolics was carried out by HPLC. An on-line HPLC-DPPH assay showed that YE possesses significant antiradical activity which is due to the presence of active components amongst phenolic compounds. Furthermore, direct effects of YE and a mixture of its identified phenolic compounds (MPC) on isolated rat heart mitochondrial function were investigated. We found that YE in concentration-dependent manner induce a decrease in State 3 respiration rate without any changes in the integrity of inner mitochondrial membrane. Thus, pyruvate oxidation was affected only by the highest used YE concentrations; meanwhile succinate oxidation was reduced even at lower YE concentrations. MPC had no effect on mitochondrial State 3 respiration rate. Fluorimetric measurements demonstrated that YE at concentrations that had no effect on the State 3 respiration rate significantly decreased H2O2 production in mitochondria.  相似文献

Plants are a source of compounds that may be used as pharmacologically active products. Cytisus multiflorus, Filipendula ulmaria and Sambucus nigra have been used as important medicinal plants in the Iberian Peninsula for many years, and are claimed to have various health benefits. Herein, the phytochemical composition and antioxidant activity of the mentioned wild medicinal plants were evaluated in vitro, based on chemical, biochemical and electrochemical methods. F. ulmaria was found to be richest in antioxidant phytochemicals, such as phenolics (228 mg GAE/g DW), flavonoids (62 mg CE/g DW), ascorbic acid (2700 μg/g DW) and tocopherols (497 μg/g DW). The antioxidant activity was found to vary in the order: F. ulmaria S. nigra > C. multiflorus, irrespective of the analysis method. Electrochemical methods have proven to be rapid and inexpensive techniques to characterise the antioxidant activity of plant extracts.  相似文献

Honey possesses antioxidant and antibacterial activities; however the compounds that are responsible for these activities are still under investigation. To determine whether there are common features/compounds that underlie the antioxidant and antibacterial activities, 20 Canadian honeys were analysed using the oxygen radical absorbance capacity (ORAC) and the broth micro-dilutions assay, respectively. Principal component analysis revealed the highest correlation between ORAC and Maillard reaction-like products (MRLP) (R = 0.938, p < 0.0001). In addition, the extremely significant correlations between ORAC, MRLPs, phenolic content and honey colour may suggest that these compounds represent the same chemical entity and exert their antioxidant activity while being part of a higher molecular weight structure. Moreover, the antioxidant activity and MRLPs were also the dominant variables contributing to the antibacterial activity against Escherichia coli (R = 0.737, p < 0.0001 and R = 0.65, p < 0.002, respectively) however no such correlation against Bacillus subtilis. In conclusion, MRLPs in honey appear to underlie the antioxidant and partially the antibacterial activity against E. coli.  相似文献

Mushrooms are widely appreciated for their organoleptic qualities, being also recognized as good sources of bioactive compounds that provide antioxidant, antimicrobial and cytotoxic activities. Polysaccharides (including glucans) are often pointed out as the most bioactive compounds isolated from mushrooms, but other molecules such as triterpenoids, might also be highlighted for their bioactivity. In scientific research, when isolated compounds are used, potential synergistic effects might be lost. Accordingly, the bioactivity of Phellinus linteus was evaluated in selected fractions (polysaccharides, glucans and triterpenoids), as well as in the methanolic and ethanolic extracts. The best antioxidant and antibacterial activities were obtained with methanolic extract, while glucan and triterpenoid fractions gave the strongest antifungal activity. In contrast, ethanolic extract gave the best results in cytotoxic activity, indicating that the bioactive compounds present might act synergistically. The differentiated activity of P. linteus fractions and extracts could be useful to find antimicrobial, antioxidant and cytotoxic agents as alternatives to synthetic chemicals with application in agriculture, food industry or pharmacy.  相似文献

In this study, polysaccharides from Tricholoma matsutake (TM-P) were purified using a DEAE-Sepharose fast flow column and three polysaccharide fractions (TM-P1, TM-P2 and TM-P3) were obtained. The chemical composition and structural characteristics of TM-P2 were quite different from those of TM-P1 and TM-P3. TM-P2 consisted of glucose, galactose and mannose with a molar ratio of 5.9:1.1:1.0. The glycosidic linkages were mainly composed of 1,6- and 1-linked glucose. Furthermore, TM-P2 showed the strongest in vitro antioxidant and antitumour activities. The oxygen radical absorbance capacity (ORAC) of TM-P2 was 2100.44 μmol Trolox/g. The antiproliferative activities of TM-P2 (4.0 mg/ml) on the growth of HepG2 and A549 cells were 67.98% and 59.04%, respectively.  相似文献

Hydrophilic phytochemicals and antioxidant capacities of eight commercially grown native Australian fruits were determined. Kakadu plum (Terminalia ferdinandiana) contained a 6-fold higher level of total phenolic compounds and quandong (Santalum acuminatum) a 1.9-fold higher level of total phenolic compounds (TP, Folin–Ciocalteu assay) than blueberry (Vaccinum sp., cv. Biloxi). Both fruits displayed superior oxygen radical-scavenging capacity (ORAC-H assay) that was, respectively 4.1-fold and 6.5-fold of that of blueberry. The total reducing capacity (TRC; ferric reducing antioxidant power (FRAP) assay) of Kakadu plum and quandong exceeded the TRC of blueberry, respectively, 13.1- and 2.3-times. The primary sources of antioxidant capacities in the evaluated fruits were phenolic acids (benzoic and cinnamic) and flavonoids (flavonols, flavanones and anthocyanins) tentatively detected by liquid chromatography–mass spectrometry (LC–PDA–MS/MS). A high level of vitamin C was recorded for Kakadu plum and Australian citrus fruits. The major organic acids detected were citric and malic acid.  相似文献

Limoniastrum monopetalum is a traditional medicinal species which leaf infusion exhibits antidysenteric properties against infectious diseases. In this study, ten kinds of leaf extracts were used to examine the effect of extraction solvent system with varying polarities on polyphenol contents and DPPH scavenging activity. Then the superoxide scavenging activity and the reducing power of the most promising solvent extracts were evaluated too. Moreover, the efficiency of the best leaf extract has been investigated against pathogenic bacteria and yeast. Eventually leaf extract was hydrolyzed by acid and the phenolics identified by RP-HPLC. Results showed that phenolic contents and antioxidant activities varied considerably as function of solvent polarity. Leaf extract using pure methanol showed the highest polyphenol content (15.85 mg GAE/g DW). Moreover, antiradical capacities against DPPH and superoxide, and reducing power were maxima in acetone/water (8:2) of leaf extract. However, the latter showed a slight antimicrobial activity against human pathogen strains such as Staphylococcus aureus, Micrococcus luteus and Candida holmii. The HPLC analysis revealed several phenolic compounds in L. monopetalum leaf including vanillic and gallic acids as major phenolics. Our findings identified the appropriate solvent for extracting halophyte phenolics which might provide a rich and novel source of natural antioxidants as food additives replacing synthetic ones in food industry.  相似文献

Phenolic compounds make up the major secondary metabolites with high pharmaceutical potential. Microalgae were reported to contain low amounts of phenolic compounds. The present study aimed to investigate the hepatoprotective potential of biomass of Spirulina platensis enriched in phenolic compounds. The protective effects of the biomass of S. platensis with low amounts of phenolics (SP1) and with high amounts of phenolics (SP2) against CCl4-induced acute hepatotoxicity were evaluated in rats. The increased levels of ALT, AST and MDA along with decreased activities of SOD and CAT were significantly (< 0.01) ameliorated by SP2. Histological examinations revealed that SP2 was more potent than SP1 in protecting the liver from toxic injury of CCl4 and preserving the hepatocyte ultrastructure. The lesions including necrosis, lymphocyte infiltration, ballooning degeneration and hepatocyte injury as irregular lamellar organisation, dilations in endoplasmic reticulums and the presence of great number of cytoplasmic vacuolization were healed by SP2.  相似文献

Leaves and fruits (peel and flesh) of six improved cultivars (‘Mizuho’, ‘Néctar de Cristal’, ‘Mizauto’, ‘Mizumo’, ‘Centenária’ and ‘NE-3’) of loquat (Eriobotrya japonica Lindl.) were studied for their phenolic composition and antioxidant capacity. The analysis by HPLC-DAD-ESI-MS/MS allowed the identification of 18 compounds (8 hydroxycinnamic acid derivatives and 10 flavonoid glycosides). The quantification of the identified compounds revealed distinct profiles amongst the three analysed materials. Loquat leaves exhibited the lowest amounts of phenolics. 3- And 5-caffeoylquinic, and 5-feruloylquinic acids were the major compounds. Generally, ‘Mizauto’ cultivar presented the highest phenolic content. All loquat materials exhibited DDPH scavenging capacity, in a concentration-dependent manner, the leaves being the most active one. This effect seems to be related to the flavonoid content.  相似文献

Fatty acids (FA), phytosterols, tocopherols, phenolic compounds, total carotenoids and hydrophilic and lipophilic ORAC antioxidant capacities were evaluated in 16 cultivars of Sacha inchi (SI) seeds with the aim to valorise them and offer more information on the functional properties of SI seeds. A high α linolenic (α-Ln) fatty acid content was found in all cultivars (ω3, 12.8–16.0 g/100 g seed), followed by linoleic (L) fatty acid (ω6, 12.4–14.1 g/100 g seed). The ratio ω6/ω3 was within the 0.83–1.09 range. γ- and δ-tocopherols were the most important tocopherols, whereas the most representative phytosterols were β-sitosterol and stigmasterol. Contents of total phenolics, total carotenoids and hydrophilic and lipophilic antioxidant capacities ranged from 64.6 to 80 mg of gallic acid equivalent/100 g seed; from 0.07 to 0.09 mg of β-carotene equivalent/100 g of seed; from 4.3 to 7.3 and, from 1.0 to 2.8 μmol of Trolox equivalent/g of seed, respectively, among the evaluated SI cultivars. Results showed significant differences (p < 0.05) among the evaluated SI cultivars in the contents of ω3, ω6, antioxidant capacities and other evaluated phytochemicals. SI seeds should be considered as an important dietary source of health promoting phytochemicals.  相似文献

The antioxidant capability and total phenolic contents of methanolic extracts of Agaricusbrasiliensis in two stages of maturity, young (YB) and mature (MB), were evaluated in this work. Four complementary assays, reducing power, radical scavenging capacity, inhibition of lipid peroxidation and chelating ability for ferrous ions were used to screen the antioxidant properties of extracts. Minor differences in the composition of phenolic compounds were detected, but the extracts showed similar antioxidant activities, except for the chelating ability for ferrous ions, higher in MB than in YB. Our results support the use of both young and mature fruiting bodies of Agaricus blazei as sources of antioxidant compounds.  相似文献

The antioxidative and hepatoprotective potential of Solidago microglossa D.C, a widely used medicinal plant from Brazil was investigated. The leaf extract showed inhibition against thiobarbituric acid reactive species (TBARS) induced by different prooxidants (10 μM FeSO4 and 5 μM sodium nitroprusside SNP) in rat liver, brain and phospholipid homogenates from egg yolk. Moreover, the free radical scavenging activities of the extract was evaluated by the scavenging of 2,2-diphenyl-1-picrylhydrazyl (DPPH) (IC50, 3.8 ± 0.5 μg/ml) and hydroxyl radical on benzoic acid hydroxylation (IC50, 32.3 ± 1.3 μg/ml) and deoxyribose (IC50, 39.1 ± 2.4 μg/ml) assays. The ethanolic extract showed significant hepatoprotective activity against paracetamol (250 mg/kg) induced liver damage in mice in a dose dependent manner. The phenolic composition and their quantification by high performance liquid chromatography (HPLC) resulted in the identification of gallic acid and flavonoids: quercetrin (quercetin-3-O-rhamnoside), rutin (quercetin-3-O-rutinoside) and quercetin.  相似文献
