Phenylpropanoid amides of octopamine (OA) 1a1e and dopamine (DA) 2a2e were synthesised and the structure–activity relationships (SARs) for antioxidant and tyrosinase inhibition activities were analysed. Among synthesised compounds, 2c, which contains two catechol moieties, exhibited the most DPPH radical-scavenging activity (EC50 = 16.2 ± 2.4 μM), and 1d exhibited significant tyrosinase inhibitory activity (IC50 = 5.3 ± 1.8 μM). Interestingly, with the same acid moiety, OA derivatives showed more inhibitory effect on tyrosinase than did compounds derived from DA, whereas DA derivatives were found to have higher antioxidant activity than compounds derived from OA. The relationship between their structures and their potencies, demonstrated in the current study, will be useful for the design of optimal agents.  相似文献

The fruits of Persea americana (Avocado) are nowadays used as healthy fruits in the world. Bioassay-guided fractionation of the active ethyl acetate soluble fraction has led to the isolation of five new fatty alcohol derivatives, avocadenols A–D (14) and avocadoin (5) from the unripe pulp of P. americana, along with 12 known compounds (617). These structures were elucidated by spectroscopic analysis. Among the isolates, avocadenol A (1), avocadenol B (2), (2R,4R)-1,2,4-trihydroxynonadecane (6), and (2R,4R)-1,2,4-trihydroxyheptadec-16-ene (7) showed antimycobacterial activity against Mycobacterium tuberculosis H37RVin vitro, with MIC values of 24.0, 33.8, 24.9, and 35.7 μg/ml, respectively.  相似文献

A phytochemical study on the whole plant of Sonchus arvensis and its antioxidant activity has been carried out. Three quinic acid derivatives (13), the rarely naturally occurring (p-hydroxyphenylacetyl) quinic acids, and two eudesmanolides (4 and 5) were newly found. Four known eudesmanolides (69) were isolated from the plant for the first time. Their structures were characterized by HRESIMS, IR, UV, 1D NMR, and 2D NMR. 1,1-Diphenyl-2-picrylhydrazyl (DPPH·)-scavenging activity was evaluated for each of the above 9 compounds (19) in comparison to standard antioxidants (caffeic acid and ascorbic acid). However, none proved to have a positive activity. The absence of antioxidant activity could be caused by the absence of ortho or para-diphenolic groups in all detected compounds, that are responsible of the activity against free radicals by an electron transfer reaction.  相似文献

Two new sesquiterpene glucosides, litchiosides A (1) and B (2), were obtained along with pumilaside A (3) and funingensin A (4) from lychee (Litchi chinensis Sonn.) seed. Their structures were elucidated on the basis of spectroscopic and chemical evidence. The in vitro antitumour activity of compounds 14 against A549, LAC, Hela, and Hep-G2 cell lines was determined using the MTT colourimetric assay. Pumilaside A (3) exhibited significant activity against all the tested cell lines with IC50 values of 0.012–6.29 μM, and funingensin A (4) showed moderate activity against Hep-G2 cells (IC50 = 39.27 μM), while litchiosides A (1) and B (2) were found to be inactive.  相似文献

Damascenone is well-known for its potent flavour with an extremely low odour threshold. Several glycosidically bound precursors of damascenone have been isolated from several plants, but little is known about their occurrences in green tea infusions. In this work, three major glycosidic precursors of damascenone, 9-O-β-d-glucopyranosyl-megastigma-6,7-dien-3,5,9-triol (1a), 9-O-β-d-glucopyranosyl-3-hydroxy-7,8-didehydro-β-ionol (2a), and 3-O-β-d-glucopyranosyl-3-hydroxy-7,8-didehydro-β-ionol (2b) were isolated and identified in green tea infusions, and the stereochemistries at C-3 and C-9 positions of aglycone parts of the three glycosidic precursors were determined as (3S, 9R)-1a, (3R, 9R)-2a, and (3R, 9R)-2b, respectively. Compounds 1a, 2a, and 2b as well as 3-O-β-d-glucopyranosyl-megastigma-6,7-dien-3,5,9-triol (1b) were hydrolysed to form damascenone in a model system with strong acidic conditions (pH 2.0) and at high temperature (90 °C). In contrast to hydrolysis of 2a and 2b, more damascenone was transformed from 1a and 1b. Furthermore, the β-d-glucosyl moiety at the C-3 position gave a higher dehydration rate from megastigma-6,7-dien-3,5,9-triol to 3-hydroxy-7,8-didehydro-β-ionol than compound 1a carrying the sugar residue at C-9 position. Interestingly, the four glycosidic precursors of damascenone were not hydrolysed to give damascenone under slightly acidic conditions (pH 5.4 and 120 °C for 10 min), but they could be transformed to damascenone in the presence of green tea infusions even under the equal conditions.  相似文献

The fruiting bodies or mycelia of mushrooms have been used as food and food-flavoring material for centuries due to their nutritional and medicinal value and the diversity of their bioactive components. The present research is the first to investigate the bioactive secondary metabolites from the solid culture of the edible mushroom Neolentinus lepideus. Two new secondary metabolites, 5-methoxyisobenzofuran-4,7(1H,3H)-dione (1) and 1,3-dihydroisobenzofuran-4,6-diol (2), as well as seven known compounds including one benzoquinone derivative (3) and six cinnamic acid derivatives (49) were obtained. Their structures were established by means of spectroscopic methods, including 1D and 2D NMR. The bioactivity on the nitric oxide production in lipopolysaccharide-induced macrophages was evaluated for all metabolites (19) isolated. Compound 1 showed strong NO inhibitory activity with the IC50 value of 6.2 μM. Compound 2 displayed moderate NO inhibitory activity with the IC50 value of 88.8 μM. In the DPPH scavenging assay, compound 2 displayed antioxidant activity with IC50 of 68.6 μM. The discovery of new NO production inhibitors from N. lepideus expands its usage as a functional food.  相似文献

In an effort to find a new whitening agent, we have found that the methanol extract of the dried skin of Allium cepa showed inhibition of melanin formation. Bioassay-guided fractionation led to the isolation of quercetin (1) and quercetin 4’-O-β-glucoside (3) from A. cepa as the inhibitors of melanin formation in B16 melanoma cells with IC50 values of 26.5 and 131 μM, respectively. In addition, we evaluated the effect of some quercetin derivatives, such as isoquercitrin (2), quercetin 3,4’-O-diglucoside (4), rutin (5) and hyperin (6) on B16 melanoma cells. These quercetin derivatives did not show any inhibition of melanin formation. Furthermore, the ORAC values of compounds 16 were 7.64, 8.65, 4.82, 4.32, 8.17 and 9.34 μmol trolox equivalents/μmol, respectively. Dried skin of red onion showed inhibitory activity against melanin formation in B16 melanoma cells, as well as antioxidant properties.  相似文献

A phytochemical investigation of the seeds of Persian leek afforded the isolation of two new spirostane glycosides, persicosides A (1) and B (2), four new furostane glycosides, isolated as a couple of inseparable mixture, persicosides C1/C2 (3a/3b) and D1/D2 (4a/4b), one cholestane glycoside, persicoside E (5), together with the furostane glycosides ceposides A1/A2 and C1/C2 (6a/6b and 7a/7b), tropeosides A1/A2 and B1/B2 (8a/8b and 9a/9b), and ascalonicoside A1/A2 (10a/10b), already described in white onion, red Tropea onion, and shallot, respectively. Structure elucidation of the compounds was carried out by comprehensive spectroscopic analyses, including 2D NMR spectroscopy and MS spectrometry, and by chemical evidences. The chemical structure of new compounds were identified as (25S)-spirostan-2α,3β,6β-triol 3-O-[β-d-glucopyranosyl-(1 → 3)] [β-d-xylopyranosyl-(1 → 2)]-β-d-glucopyranosyl-(1 → 4)-β-d-galactopyranoside (1), (25S)-spirostan-2α,3β,6β-triol 3-O-[β-d-xylopyranosyl-(1 → 3)] [α-l-rhamnopyranosyl-(1 → 2)]-β-d-glucopyranosyl-(1 → 4)-O-β-d-galactopyranoside (2), furosta-1β,3β,22ξ,26-tetraol 5-en 1-O-β-d-glucopyranosyl (1 → 3)-β-d-glucopyranosyl (1 → 2)-β-d-galactopyranosyl 26-O-α-l-rhamnopyranosyl (1 → 2)-β-d-galactopyranoside (3a,3b), furosta-2α,3β,22ξ,26-tetraol 3-O-β-d-glucopyranosyl (1 → 3)-β-d-glucopyranosyl (1 → 2)-β-d-galactopyranosyl 26-O-β-d-glucopyranoside (4a,4b), (22S)-cholesta-1β,3β,16β,22β-tetraol 5-en 1-O-α-l-rhamnopyranosyl 16-O-α-l-rhamnopyranosyl (1 → 2)-β-d-galactopyranoside (5).  相似文献

Imperatorin (IMP) is a major constituent of many herbal medicines and possesses anti-osteoporosis activity. The present research work aimed to study the biotransformation processes of IMP and evaluated the anti-osteoporosis activity of the transformed metabolites. Among 18 strains of filamentous fungi screened, Penicillium janthinellum AS 3.510 exhibited good capability to metabolise IMP to the new derivatives. Ten transformed products were isolated and purified, and their structures were identified accurately based on spectroscopic data. Eight metabolites (2–8 and 10) were novel and previously unreported. The major biotransformation reactions involved hydroxylation of the prenyloxy side-chain and the lactone ring-opening reaction of furocoumarin skeleton. In addition, anti-osteoporosis activities of all products (1–10) were evaluated using MC3T3-E1 cells. The results showed that products 5 and 8 had the best bioactivities in increasing MC3T3-E1 cell growth. These products could be used in future therapeutic regimens for treating osteoporosis.  相似文献

Tyrosinase can catalyze the oxidation of o-diphenols to o-quinones. In this paper, some o-diphenols were used as mushroom tyrosinase substrates to study the catalyzed specificity of the enzyme. The enzyme kinetic analysis of substrate specificities and the substrate analogues towards mushroom tyrosinase has been investigated. Taking l-3,4-dihydroxyphenylalanine (I), 3,4-dihydroxyhydrocinnamic acid (II), 3,4-dihydroxycinnamic acid (III) and 1,2,4-benzenetriol (IV) as substrates, the results of specificity studies showed that the oxidation reaction of tested o-diphenols by mushroom tyrosinase followed Michaelis–Menten kinetics. The Michaelis–Menten constants for these four substrates were determined to be 0.615, 1.238, 0.331 and 1.886 mM, respectively. The values of Vm/Km, which denotes the affinity of the enzyme to the substrate, were determined and compared, and the results showed that the affinity of the enzyme to these substrates followed the order: compound IV > III > I > II. Furthermore, mushroom tyrosinase cannot catalyze the oxidation of 3,4-dihydroxybenzonitrile (a), 3,4-dihydroxybenzaldehyde (b), 3,4-dihydroxybenzoic acid (c) and 2,3-dihydroxybenzoic acid (d). On the contrary, compounds a, b and c can inhibit the activity of tyrosinase for the oxidation of DOPA, while compound d had no effects on enzyme activity. The results show that compounds a and b are reversible non-competitive inhibitors.  相似文献

Three new flavonol glycosides, 3′,5′ dimethoxymyricetin-4″-O-α-l-rhamnopyranosyl (1–4) β-d-glucopyranoside (1), 3′-methoxyquercetin-4″-O-α-l-rhamnopyranosyl (1–4) β-d-glucopyranoside (2) and 3′-methoxyqurecetin-6″-O-α-l-rhamnopyranosyl (1–6) β-d-glucopyranoside (3), have been isolated from the aerial part of Barbeya oleoides Schweinf., along with twelve known compounds, uvaol (4), ursolic acid (5), corosolic acid (6), arjunolic acid (7), β-sitosterol-3-O-β-d-glucoside (8), (+)–catechin (9), (-)-epicatechin (10), isorhamnetin-4′-O-glucoside (11), arjunglucoside I (12), d-(-)-bornesitol (13), gallocatechin (14) and epigallocatechin (15). Compounds 415 were isolated for the first time from Barbeyaceae. Structure elucidation of compounds 1–3 was based on MS and NMR data. The ethyl acetate extract of the stems as well as compounds 5, 6, 14 and 15 showed significant antimicrobial activity, while the ethanol extracts of leaves, stems and compounds 4, 7, 8, 13–15 have dose-dependent spasmolytic action.  相似文献

Zanthoxylum nitidum is commercially used in healthcare products for the improvement of tooth condition. Its fruit is also used as a spice. Three new benzenoids, (E)-4-(4-hydroxy-3-methylbut-2-enyloxy)benzaldehyde (1), (E)-methyl 3-(4-((E)-4-hydroxy-3-methylbut-2-enyloxy)phenyl)acrylate (2), and (Z)-methyl 3-(4-((E)-4-hydroxy-3-methylbut-2-enyloxy)phenyl)acrylate (3), have been isolated from the stem wood of Z. nitidum, together with 17 known compounds (420). The structures of these new compounds were determined through spectroscopic and MS analyses. Compounds 810, 13, 14, 17, and 18 exhibited inhibition (IC50 ? 7.95 μg/ml) of superoxide anion generation by human neutrophils in response to formyl-l-methionyl-l-leucyl-l-phenylalanine/cytochalasin B (fMLP/CB). Compounds 79, 13, 14, and 17 inhibited fMLP/CB-induced elastase release with IC50 values ?7.45 μg/ml.  相似文献

In this study, we investigated the antioxidative activities of (−)-catechin (1), (−)-catechin-7-O-β-d-apiofuranoside (2) and (−)-catechin-7-O-β-d-xylopyranoside (3) in terms of their abilities to promote metal chelation, prevent lipid peroxidation, and inhibit DNA cleavage. Seven new (17) and three known (810) abietane diterpenoids were isolated from the methanolic extract of the aerial parts of Isodon lophanthoides var. graciliflorus (Lamiaceae), a folk Chinese medicine and an herb for functional beverages. They were identified as 16-acetoxylsugiol (1), graciliflorin E (2), graciliflorin F (3), 15-O-methylgraciliflorin F (4), 15-hydroxy-20-deoxocarnosol (5), 3β-hydroxysempervirol (6), 15-hydroxy-1-oxosalvibretol (7), abieta-8,11,13-triene-14,19-diol (8), 6,12,15-trihydroxy-5,8,11,13-abietatetraen-7-one (9), and 3α-hinokiol (10) based on the spectroscopic data including COSY (correlated spectroscopy), HMBC (heteronuclear multiple bond correlation), and HR-ESI-MS (high-resolution electrospray ionization mass spectrometry). All the compounds except 10 were obtained from I. lophanthoides for the first time. Compounds 1, 2, 5, 6, 8, and 9 exhibited in vitro cytotoxicity against A549 (human lung adenocarcinoma), MCF-7 (human breast adenocarcinoma), and HeLa (human cervical carcinoma) cell lines with the IC50 values of 1.79–52.67 μM.  相似文献

In this study, bioassay-guided fractionation of extracts from the leaves of Morus alba L. led to the isolation of 15 bioactive constituents with α-glucosidase and tyrosinase inhibitory activities, among which prenylated stilbenes were proved to be a new group of α-glucosidase inhibitors apart from iminosugars derived from Morus alba. Their structures were identified on the basis of extensive spectroscopic analysis and chemical evidence, as well as comparing with data from the literature. Among them, compounds (2R)/(2S)-Euchrenone a7 (6a/6b), Chalcomoracin (7), Moracin C (8), Moracin D (9) and Moracin N (10) exhibited a significant degree of α-glucosidase inhibitory activity with IC50 of 6.28, 2.59, 4.04, 2.54 and 2.76 μM, respectively, while (2R)/(2S)-Euchrenone a7 (6a/6b), Moracin N (10), Quercetin (13), Norartocarpetin (14), the interconvertible epimeric mixture of (2R)/(2S)-7-methoxyl-8-ethyl-2′,4′-dihydroxylflavane-2″-O-β-d-glucopyranoside (1a/1b) and the interconvertible enantiomers of (2R)/(2S)-7-methoxyl-8-hydroxyethyl-2′,4′-dihydroxylflavane (5a/5b) displayed a potent tyrosinase inhibitory effect with IC50 of 0.260, 0.924, 0.523, 0.0824, 0.616 and 0.528 μM, respectively. Especially, (2R)-7-methoxyl-8-ethyl-2′,4′-dihydroxylflavane-2″-O-β-d-glucopyranoside (1a), (2S)-7-methoxyl-8-ethyl-2′,4′-dihydroxylflavane-2″-O-β-d-glucopyranoside (1b), (2S)-8-hydroxyethyl-7,4′-dimethoxylflavane-2′-O-β-d-glucopyranoside (2), (2R)-7-methoxyl-8-hydroxyethyl-2′,4′-dihydroxylflavane (5a) and (2S)-7-methoxyl-8-hydroxyethyl-2′,4′-dihydroxylflavane (5b) were identified as new compounds.  相似文献

The root of Scorzonera austriaca has been used in indigenous cuisines as a delicious food and in the Tibetan traditional medicine in northwestern China. Two novel dimeric guaianolides linked by a carbon–carbon bond with a rare carbon skeleton, termed biguaiascorzolides A (1) and B (2), respectively, have been isolated from roots of S. austriaca. Acetylation of 1 gave 1a. The structures of 1, 1a and 2 were characterised by HR-ESI-MS, EI-MS, UV, IR, and 1D- and 2D-NMR techniques (1H and 13C NMR, 1H-1H COSY, HMQC, HMBC, and NOESY experiments). The cytotoxicity of 1a was assayed against selected cancer cell lines, including the human erythroleukaemia adriamycin-resistant subline (K562/ADM) and human stomach carcinoma (MGC-803) cell lines. Compound 1a exhibits a moderate activity against K562/ADM cell lines (IC50 39.8 μm) and is inactive towards MGC-803 cell lines.  相似文献

A phytochemical investigation of the fresh bulbs of Allium nigrum L. led to the isolation of new spirostane-type glycosides as two inseparable isomer mixtures, nigrosides A1/A2 (1a/1b) and nigrosides B1/B2 (2a/2b), two new cholestane-type glycosides, nigrosides C and D (3 and 4), together with the known compounds, 25(R,S)-5α-spirostan-2α,3β,6β-trio1-3-O-β-d-glucopyranosyl-(1 → 2)-O-[β-d-xylopyranosyl-(1 → 3)]-O-β-d-glucopyranosyl-(1 → 4)-β-d-galactopyranoside (5a/5b) and 25(R,S)-5α-spirostan-2α,3β,6β-trio1 3-O-β-d-glucopyranosyl-(1 → 2)-O-[4-O-(3S)-3-hydroxy-3-methylglutaryl-β-d-xylopyranosyl-(1 → 3)]-O-β-d-glucopyranosyl-(1 → 4)-β-d-galactopyranoside (6a/6b), isolated from this plant for the first time. All structures were elucidated mainly by spectroscopic analysis (1D and 2D NMR experiments, FABMS, HRESIMS) and by comparison with literature data. Cytotoxicity of the isolated compounds was assessed against human colon carcinoma (HT-29 and HCT 116) cell lines. Compounds 5a/5b and 6a/6b were found to be the most active with IC50 values 1.09 and 2.82 μM against HT-29 and 1.59 and 3.45 μM against HCT 116, respectively.  相似文献

Five previously uncharacterised polyphenols, ethyl gallate (2), 1-β-O-galloyl-d-glucopyranose (3), methyl brevifolin carboxylate (4), brevifolin (5) and 4-O-α-l-rhamnopyranosyl-ellagic acid (8), and three previously identified polyphenols, gallic acid (1), corilagin (6) and ellagic acid (7), were isolated from longan seeds. Their structures were identified by spectroscopic and chemical methods including HRESIMS and NMR. Eight polyphenols exhibited scavenging activity towards DPPH radicals with SC50 values of 0.80–5.91 μg/ml and towards superoxide radicals with SC50 values of 1.04–7.03 μg/ml. The radical-scavenging activity of the newly characterised polyphenols was comparable to that of gallic acid, corilagin and ellagic acid.  相似文献

Ganoderma resinaceum Boud. (Polyporeseae) has long been used for antioxidant, immunoregulation and liver protection. From the fruiting bodies of G. resinaceum, eight new lanostanoids, lucidones D–G (14), 7-oxo-ganoderic acid Z2 (5), 7-oxo-ganoderic acid Z3 (6), ganoderesin A (7), and ganoderesin B (8), together with six known lanostanoids (914) were isolated. The structures of new compounds were elucidated through extensive spectroscopic analysis. In an in vitro model, ganoderesin B (8), ganoderol B (10) and lucidone A (11) showed inhibitory effects against the increase of ALT and AST levels in HepG2 cells induced by H2O2 compared to a control group in the range of their maximum non-toxic concentration (MNTC). However, compounds 8, 10 and 11 displayed no anti-oxidant activities by DPPH assay. Meanwhile, activation for PXR (Pregnane X Receptor) of ganoderesin B (8), ganoderol B (10) and lucidone A (11) was evaluated; ganoderol (10) exhibited a vital activation for PXR-induced CYP3A4 expression. These results suggested that GTs (Ganoderma triterpenoids) exhibited hepatoprotective activities by lowering ALT and AST levels.  相似文献
