目的 采用超高效液相色谱-高分辨质谱法(ultra performance liquid chromatography-high resolution mass spectrometry,UPLC-HRMS)对咖啡及其掺假物建立高分辨质谱数据库,建立一种基于组学技术对咖啡掺假进行鉴别的方法。方法 咖啡及其掺假物经甲醇-水(7:3,V:V)超声提取后,经BEH C18色谱柱(2.1 mm×100 mm,1.7μm)分离,电喷雾电离(electrospray ionization,ESI)离子源,在高分辨质谱Full MS/dd MS2扫描模式下采集数据,建立咖啡及掺假物黑玉米、黑豆、大麦的高分辨质谱数据。采用Compound Discoverer 3.3 (CD 3.3)组学软件对UPLC-HRMS数据进行主成分分析(principal component analysis,PCA)和火山图差异分析,根据咖啡及掺假物中差异化合物的精准分子质量数、母离子同位素组成及碎片离子信息质量信息检索mzCloud和Chem Spider在线数据库,对差异成分进行匹配鉴定。结果 PCA结果表明咖啡与掺... 相似文献

高速逆流色谱分离制备胡椒中的胡椒碱 总被引:1,自引:0,他引:1
采用高速逆流色谱(high-speed countercurrent chromatography,HSCCC)法从胡椒中分离制备胡椒碱。HSCCC的溶剂系统条件为正己烷-乙酸乙酯-甲醇-水(1:1:1:1,V/V)。从5g粗提物中可一次分离得到纯度为98.72%的胡椒碱单体1.58g,分离得到的胡椒碱结构经电喷雾质谱以及核磁共振氢谱(1H NMR)和碳谱(13C NMR)进行鉴定。该法制备量大、分离效率高,对胡椒碱在食品医药领域的应用具有重要意义。 相似文献

Effect of Gamma-irradiation on Color, Pungency, and Volatiles of Korean Red Pepper Powder 总被引:1,自引:0,他引:1
ABSTRACT: Effect of gamma-irradiation on color, pungency, and volatiles of Korean red pepper powder ( Capsicum annuum L.) was investigated. Red pepper powder, vacuum-packaged in a polyethylene/polypropylene bag, was gamma-irradiated up to 7 kGy. An irradiation dose of 7 kGy reduced the population of mesophilic bacteria and fungi effectively without affecting major quality factors. Pungency of irradiated red pepper powder was not changed based on the amount of capsanoids by high-performance liquid chromatography (HPLC) and the Scoville sensory score. The red color of irradiated pepper powder was not significantly different from that of the control, judged from the capsanthin content by HPLC and color assessment using spectrophotpmetric (American Spice Trade Assn. units) and colorimetric measurements (Hunter a values). Further, the sensory evaluation showed no significant difference in pungent odor and off-odor between nonirradiated control and irradiated red pepper powder. However, when headspace volatiles of gamma-irradiated red pepper powder were evaluated by gas chromatography/ mass spectrometry with solid-phase microextraction and electronic nose with metal oxide sensors, the profiles of odor were classified into irradiated dose levels of 0, 3, 5, and 7 kGy by principal component analysis and multivariate analysis of variance. Such a difference of odor might result from the disappearance of some volatiles, such as hexanoic acid and tetramethyl-pyrazine, and the appearance of 1,3-di-tert-butylbenzene during irradiation. Moreover, it appears that the irradiation of packaging material induced a formation of 1,3-di-tertbutylbenzene, which migrated into the red pepper powder. 相似文献

建立了胡椒中胡椒碱含量测定的超高效液相色谱(UPLC)测定法。样品经95%乙醇回流提取,以甲醇/水(70/30,V/V)为流动相,流速0.25mL/min,343nm波长进行检测。结果表明:胡椒碱保留时间为2.585min,测定线性范围为0.05~10.0μg/mL,r=0.9998,当添加量为0.4~4.0g/100g时,回收率为90.5%~95.8%,相对标准偏差为0.56%~1.81%,检测限为0.008g/100g。该方法操作简单,灵敏度高,重现性好,省时,适合于胡椒碱含量的测定。 相似文献

普通乳粉经回潮、成型、干燥等工艺制成成型乳粉,其感官指标、理化指标及微生物指标与普通乳粉无显著差异(P>0.05),但其表观密度却由0.5~0.6g/mL增加到1.1g/mL,体积减小一半。因保藏期间吸收水分的能力降低,故其保藏期增加。试验表明,在45℃的饱和湿空气中,回潮乳粉含水量(X,%)、基粉含水量(a,%)和回潮时间(Y,min)之间存在着以下关系:Y=31.49(x—a)~(0.2696)(r=0.9956~(**))。在试验室条件下,回潮乳粉含水量以4.5%,成型压力以470.4kPa为宜。成型后干燥条件:温度60℃,时间2h。 相似文献

本文分别对四种加工方式(直接日晒、热烫处理后日晒、日晒1 d轻度发酵、日晒2 d轻度发酵)过程中的黑胡椒风味成分进行分析。采用水蒸气蒸馏法提取胡椒精油,四种产品中胡椒精油的含量分别为1.10、1.09、1.16、0.89 m L/100 g,发现四种加工过程中胡椒精油均有较大程度的损失,损失率范围63%71%。经GC-MS检测四种加工方式所得产品中的精油分别鉴定出19、17、23、22种化合物,其总检出量在95%以上;其中α-蒎烯、β-蒎烯、3-蒈烯、D-柠檬烯、蛇麻烯、反式-石竹烯、β-甜没药烯在四种产品中的含量较高,最高的为反式-石竹烯,相对含量范围43.47%56.16%。利用HPLC法检测胡椒碱含量,四种加工产品中胡椒碱含量分别为3.64、3.53、3.32、3.58 g/100 g,胡椒碱在加工过程中的损失率范围为9%17%。 相似文献
